杜仲内生真菌的抑菌活性筛选   总被引:15,自引:1,他引:14  
以金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌为测试菌种。对杜仲(Eucommia ulmoides Oliv.)根、茎、叶中分离出的20株内生真菌及其次生代谢物进行抗菌活性筛选。结果表明:有15个菌株至少对1种实验细菌具有抑菌活性,19株的代谢产物至少对1种实验细菌具有抑菌活性,其中有3株内生真菌及其次生代谢产物对测试病原细菌均有较强抑制作用。  相似文献

The beta-glucuronidase activities of bacterial strains isolated from the rat intestinal tract were studied both in vitro in culture media and in vivo in the intestinal contents of gnotobiotic rats. Only 50 of 407 strains tested were found to be positive in vitro. They belonged to the three genera Clostridium, Peptostreptococcus, and Staphylococcus. The in vitro-negative strains were also negative in vivo. The beta-glucuronidase activities of the beta-glucuronidase activities of the positive strains were generally greater in vivo than in vitro. The highest in vivo activities were found in the intact bacterial cells and in the soluble fractions prepared from disrupted pellets. There was a discrepancy between the activities obtained from both conventional and gnotobiotic rats harboring selected positive strains, suggesting that the main beta-glucuronidase-positive strains have not yet been isolated from the intestines of conventional rats.  相似文献

The taxonomic distribution of Streptomyces species capable of producing bioactive compounds was investigated. Nine hundred and six strains were tested for the following four biological activities: antimicrobial, anti-tyrosinase, antioxidant, and hemolytic. Approximately 30% of strains tested showed antimicrobial activities, except for anti-Escherichia coli activity, which was present in only a few strains, while the rates of positivity for the anti-tyrosinase, antioxidant, and hemolytic activities were much lower. The distribution of Streptomyces strains capable of producing bioactive compounds was analyzed by the taxonomy based on 16S rRNA gene sequences. Moreover, the strains of Streptomyces hygroscopicus tested were divided into two clades in the phylogenetic tree, and all of the strains belonging to one clade showed antibacterial and antifungal activities. For detection of polyenes, the UV-visible spectra of metabolic extracts in the strains showing antifungal activities were measured. It was suggested that Streptomyces strains produce universal active compounds under different growth conditions. Further information on the relationship between the microbial taxonomy and the bioactive compounds produced would be useful for the utilization of industrial microorganisms.  相似文献

7株放线菌在辣椒根部定殖及对辣椒叶片PAL与PPO活性的影响   总被引:17,自引:0,他引:17  
采用盆栽接种试验、平皿涂抹法测数及常规酶活测定法研究了7株拮抗性放线菌在辣椒根部的定殖能力及接种24d对辣椒叶片苯丙氨酸解氨酶(PAl。)与多酚氧化酶活性(PPO)的诱导效应。结果表明:(1)供试7株放线菌单独接种均不能在辣椒根内定殖,但与辣椒疫霉P3混合接种时有5株可定殖;供试放线菌在辣椒根部的定殖能力与其体外平皿试验中产生的的拈抗圈大小基本无关;可定殖放线菌的定殖密度随时间延长而降低,至40d时均无活菌检出。(2)在放线菌单接处理中,5株菌接种后可诱导辣椒叶片PAL,活性提高,全部供试菌均能诱导PPO活性提高,其中可使两种酶同步提高的有5株菌;在放线菌+P3混接处理中,有6株接种后可诱导PAL,活性提高,5株菌能诱导PPO活性提高,其中可使两种酶同步提高的有4株菌;在接入放线菌时同时混接辣椒疫霉,能增强2株供试放线菌对辣椒叶片PAL活性及6株供试放线菌对辣椒叶片PPO活性的诱导作用;供试放线菌的定殖能力与辣椒叶片PAL及PPO活性变化无明显规律性关系。  相似文献

6-Phosphofructokinase (6-PFK) activities have been measured in cell extracts from a number of Bacteroides species. Two main types of 6-PFK were found: an ATP-linked 6-PFK and a PPi-linked 6-PFK. In most strains both of these activities were found, although in two strains only ATP-linked 6-PFK was present. The PPi-linked 6-PFK activity was always higher, when both activities were present, and showed Michaelis-Menten kinetics with respect to fructose 6-phosphate. In contrast, the ATP-linked 6-PFK activity usually gave sigmoid kinetics with respect to fructose 6-phosphate, although several strains were found to have activities with Michaelis-Menten kinetics. Conditions for measuring maximum activities of ATP-linked 6-PFK were not identical for different strains, and the activities were rather unstable in extracts. The possible consequences of the observation that most Bacteroides strains possess both an ATP-linked and a PPi-linked 6-PFK are discussed.  相似文献

测定云南八角莲Dysosma aurantiocaulis和川八角莲Dysosma veitchii内生真菌的生物活性,为进一步从内生真菌中寻找活性成分提供指导,也为认识内生真菌相互作用提供有益的参考。从两种八角莲中分离出内生真菌187株,用滤纸片法测抗菌活性,用卤虫模型测抗卤虫活性得知大部分菌株发酵液都有活性,高活性菌株主要集中在镰孢霉属Fusarium、青霉属Penicillium、木霉属Trichoderma、丛梗孢属Monilia、毛壳菌属Chaetomium、无孢类群Mycellia sterilia、小齿梗霉属Rhinotrichum、无梗孢属Trichocladium和胶帚霉属Gliocladium。云南八角莲和川八角莲内生真菌是活性成分筛选的重要来源,并且通过活性分析发现:发酵液的活性强于菌丝体;云南八角莲的抗细菌活性菌株多于川八角莲。  相似文献

目的:研究黑木耳菌株原种胞外酶活分泌特性,为进行大规模生产用种早期判定提供检测手段。方法:以9个黑木耳栽培菌株的原种为实验材料,分别测定了其胞外漆酶、多酚氧化酶、羧甲基纤维素酶、滤纸维素酶的酶活性变化。结果:胞外漆酶、多酚氧化酶、纤维素酶活性的变化趋势大致相似,酶活随培养时间呈规律性的升高、降低,除个别菌株(3、6)外,不同菌株同种酶活性差别不大,酶活相对较高的菌株有2、3、6、9号。不同菌株酶活高峰出现的时间有差异,较早的出现在培养的第10d,较晚的出现在培养的第70d。结论:被测菌株胞外酶活存在一定的规律性,利用该规律性可检测菌种退化、老化和不利变异。  相似文献

从川乌(Radix aconiti)植物的根和茎中分离获得内生真菌61株,经显微形态观察进行内生真菌的分类鉴定,同时采用纸片法进行抗菌活性筛选。结果有29株内生真菌显示抗菌活性,占菌株数的47.54%;毛壳菌属是抗菌活性菌株的优势种属;活性高且抗菌谱广的菌株有3株,主要分布在毛壳菌属和束丝菌属。结果表明:川乌植物内生真菌中广泛分布着有抗菌活性的菌株。  相似文献

从白木香Aquilaria sinensis(Lour.)Gilg木质部的树脂形成部位和健康部位中共分离获得42株内生真菌,经初步鉴定,产孢的33株分属于3目4科7属,其余未产孢的9株暂归为无孢菌群。采用杯碟法和MTT法分别测定了各菌株的发酵上清液对3种病原菌的体外抑菌活性和2种肿瘤细胞的体外细胞毒活性。结果表明,白木香木质部健康部位内生真菌以枝顶孢霉属为优势属,而树脂形成部位的内生真菌种类比健康部位要多,且以青霉属为优势属。其中26株至少能抑制一种指示菌,占总数的61.9%;7株对指示瘤株具有细胞毒活性,占总数的16.7%。抑菌活性菌株主要分布在枝顶孢霉属和青霉属。枝顶孢霉属菌株抑菌活性较强,其抗菌活性成分值得进一步研究。  相似文献

Thirty-five strains of ruminal bacteria belonging to the former Butyrivibrio fibrisolvens species were screened for the presence of site-specific restriction endonuclease and modification methyltransferase activities. Seven strains possessed endonuclease activities detectable in crude cell extracts. The recognition sequences and optimal reaction conditions for seven of them were determined. Five enzymes were found to be isoschizomers of type II endonucleases (EcoRV, NsiI, AseI (2x) and SauI), one was type IIS (FokI) and two remained unknown. The optimal reaction buffer was found to be a low ionic strength buffer and all enzymes possessed sufficient activity at 39 degrees C. The presence of DNA modification among all strains was also determined. Most of the methylation activities correlated with restriction activities, yet some strains possessed unaccompanied modification methyltransferases.  相似文献

The presence of superoxide dismutase was demonstrated in 21 strains of mollicutes, including achuloplas-mas, mycoplasmas and ureaplasmas. Additionally, catalase activities were demonstrated in nearly 50% of the cell lysates. whereas no peroxide activities were detectable. The production of O2-and H2O2 with glucose as substrate was demonstrated for 8 strains of 10 strains tested. Anaerobic mycoplasmas showed the highest amount of radical production, whereas superoxide dismutase and catalase activities were in the range of activities estimated for aerobic mollicutes. Some pathogenic strains additionally released compounds into the culture medium, which stimulated O2-production by PMNs.  相似文献

Sublethal heating of the cells at 55 degrees C resulted in increased sensitivity to 5.5% NaCl. β-Galactosidase (β-gal) and 6-phospho-β-galactosidase (P-β-gal) activities were measured in a total of 34 strains from Lactobacillus acidophilus, Lactobacillus crispatus, Lactobacillus amylovorus, Lactobacillus gasseri and Lactobacillus johnsonii. 为了进一步研究部分正常菌群的生理和病理机制,我们测定了这些菌株的SOD活性和脂质过氧化物(LPO)变化,发现皮肤常住菌的分离株的混合培养时,SOD活性增加,说明它们具有协同作用。此外,对其它菌株SOD活性测定结果可知,类杆菌和双歧杆菌的SOD活性最低,其它菌株也有一定的SOD活性,如乳杆菌的SOD活性为47.47~98.98μg/ml,大肠杆菌的SOD活性为127.12μg/ml肠球菌的SOD活性为172.5μg/ml。  相似文献

Forty-two bifidobacterial strains were screened for alpha-amylase and/or pullulanase activity by investigating their capacities to utilize starch, amylopectin, or pullulan. Of the 42 bifidobacterial strains tested, 19 were capable of degrading potato starch. Of these 19 strains, 11 were able to degrade starch and amylopectin, as well as pullulan. These 11 strains, which were shown to produce extracellular starch-degrading activities, included 5 strains of Bifidobacterium breve, 1 B. dentium strain, 1 B. infantis strain, 3 strains of B. pseudolongum, and 1 strain of B. thermophilum. Quantitative and qualitative enzyme activities were determined by measuring the concentrations of released reducing sugars and by high-performance thin-layer chromatography, respectively. These analyses confirmed both the inducible nature and the extracellular nature of the starch- and pullulan-degrading enzyme activities and showed that the five B. breve strains produced an activity that is consistent with type II pullulanase (amylopullulanase) activity, while the remaining six strains produced an activity with properties that resemble those of type III pullulan hydrolase.  相似文献

2001~2002年从海北高寒草甸生态系统采集土样,用不同方法从中分离放线菌300余株,根据其形态和分类特征,分别归入小单孢菌属(Micromonospora)、诺卡氏菌属(Nocardia)、糖多孢菌属(Saccharopolyspora)、原小单孢菌属(Promicromonospora)和链霉菌属(Streptomyces),并将链霉菌归入7个类群。同时对230株中温菌和110株低温菌的部分酶活性及其对真菌和细菌的拮抗性进行了测定,发现链霉菌不仅具有许多酶活性,而且对真菌和细菌有拮抗性。  相似文献
