细菌视紫红质的两种光电微分响应及其机制   总被引:3,自引:1,他引:2  
利用MATLAB软件对细菌视紫红质 (BR)膜光电器件的脉冲响应实验数据进行拟合 ,得到器件的冲激响应函数。据此用SIMULINK模块构造出了反映BR光电器件特性的仿真系统。利用此系统对不同间断光入射BR光电器件时的输出响应信号进行了仿真计算。通过分析得出结论 :以前所描述的微分响应 (发生在毫秒到秒的时间量级 ,在光打开时产生一个正脉冲 ,在光关掉时产生一个负脉冲 )并非BR分子固有的特性 ,部分是由于测量电路引起的。BR分子本身特性引起的微分响应是发生在微秒时间量级 ,而且在光打开时产生一个负脉冲 ,在光关掉时产生一个正脉冲。对这两种微分响应产生的机制分别进行了探讨  相似文献

在长方形光脉冲光照下,菌紫质(bacteriorhodopsin,BR)干膜组装成夹层光电池具有微分光电流响应.在氧化铟锡(ITO)导电玻璃/BR膜/封口膜/不锈钢形成的干膜电池下可观察到整流特性,而在不锈钢/BR膜/封口膜/ITO导电玻璃形成的干膜电池下则观察不到整流特性,这说明是电极介导的整流.平衡电压测定表明:工作电极/BR膜表面与对电极/BR膜表面有不同的性质,电极的界面效应控制了BR的取向.酸与碱产生的瞬间电流极性也证实了电极整流行为的存在.这些结果将有助于了解BR膜的微分光电响应.  相似文献

RH57%的APMLBF构制的Cu/Parafilm/APMLBF/ITO型BR光电池具有的光电响应能力随APMLBF暗适应程度增加而增大,光适应后趋于稳定。光电响应信号的大小随被光照的截面减小而变小;不同引线输出的信号极性与质子泵运相一致。结果认为这种光电池可用于进一步构制光电池组以模拟视网膜的某些功能。  相似文献

在ITO导电玻璃上制备定向细菌视紫红质 (BR)电泳沉积膜或LB膜组成光电池系统 ,在短脉冲激光照射下 ,测定其脉冲响应光电压 ;在间断光照射下 ,测定其对光强变化产生的微分响应信号。对脉冲光电响应和微分响应的机理及其关系进行理论分析和解释 ,认为脉冲响应是BR分子内部生色团快速光极化引起的电荷分离和希夫碱及其周围氨基酸去质子化和再质子化过程引起的质子定向运输产生的位移电流 ,是一个快反应过程 ,是微分响应的早期反应和基础。微分响应则是由于菌紫质的光驱动质子泵产生的连续质子流在光开和光关瞬间引起光电池系统充放电以及测量电路的耦合特性引起的 ,是一个慢变化过程  相似文献

A device for quickly and accurately measuring the population density of a suspension of microorganisms, permitting the preparation of yeast suspensions of known density to within 1 per cent error, was constructed with two Visitron photoelectric cells, a single light of high intensity and a good Wheatstone bridge for balancing the currents from the two photoelectric cells. A large Pyrex milk culture tube holding the suspension is placed in the path of one beam of light coming through a small longitudinal slit and thence to one photocell; a second similar slit directs another beam of light upon the second photocell, thus causing dissimilar currents to flow, the ratio of whose magnitudes may be measured by the bridge resistances. A relation between these currents and the relative light intensities is shown, and the one significant unmeasurable variable (the characteristic constant of a photocell) is practically eliminated by the use of a method of ratios. After careful standardization of technique the apparatus proved more accurate than other methods available for the purpose indicated. In rapid use its accuracy may be put safely at 1 per cent for measuring the densities of cultures of approximately the same age and composed of cells having comparable optical characteristics.  相似文献

Light exposure was measured in six day and six night watches (working 12-hour shifts five days in a row) during 48 h on work days and 48 h on days off using a photocell with a sensitivity corresponding to photopic vision. The photocell was mounted on a frame of spectacles, thus measuring in viewing direction. Light exposure was low both in night and day watches; however, in night watches exposures were significantly lower: On work days, night watches spent a mean of 13 min above 1,500 lx, day watches 52 min; on days off, night watches spent 3 min above 1,500 lx but day watches 89 min. Unexpectedly, night watches had no higher exposure during days off. We suspect that this is due to a light avoidance tendency in permanent night workers. High negative correlations between the acrophases of subjective state (e.g., alertness and mood) and light exposure in night watches indicate that bright light would probably increase desynchroniza-tion between subjective state, sleep, and activity.  相似文献

The photoelectric response of a detector using dried bacteriorhodopsin (bR) film as the light sensing material is mathematically modeled and experimentally verified in this paper. The photocycle and proton transfer kinetics of dried bR film differ dramatically from the more commonly studied aqueous bR material because of the dehydration process. The photoelectric response of the dried film is generated by charge displacement and recombination instead of transferring a proton from the cytoplasmic side to the extracellular side of the cell membrane. In this work, the wild-type bR samples are electrophoretically deposited onto an indium tin oxide (ITO) electrode to construct a simple multiple layered photo-detector with high sensitivity to small changes in incident illumination. The light absorption characteristics of the thin bR film are mathematically represented using the kinetics of the bR photocycle and the charge displacement theorem. An electrically equivalent RC circuit is used to describe the intrinsic photoelectric properties of the film and external measurement circuitry to analyze the detector's response characteristics. Simulated studies and experimental results show that the resistance of the dried bR film is in the order of 10(11) Omega. When the input impedance of the measurement circuitry is one order of magnitude smaller than the dried film, the detector exhibits a strong differential response to the original time-varying light signal. An analytical solution of the equivalent circuit also reveals that the resistance and capacitance values exhibited by the dried bR film, in the absence of incident light, are almost twice as large as the values obtained while the material is under direct illumination. Experimental observations and a predictive model both support the notion that dried bR film can be used in simple highly sensitive photo-detector designs.  相似文献

The actinic light effect on the bacteriorhodopsin (BR) photocycle kinetics led to the assumption of a cooperative interaction between the photocycling BR molecules. In this paper we report the results of the actinic light effect and pH on the proton release and uptake kinetics. An electrical method is applied to detect proton release and uptake during the photocycle [E. Papp, G. Fricsovszky, J. Photochem. Photobiol. B: Biol. 5 (1990) 321]. The BR photocycle kinetics was also studied by absorption kinetics measurements at 410 nm and the data were analyzed by the local analysis of the M state kinetics [E. Papp, V.H. Ha, Biophys. Chem. 57 (1996) 155]. While at high pH and ionic strength, we found a similar behavior as reported earlier, at low ionic strength the light effect proved to be more complex. The main conclusions are the following: Though the number of BR excited to the photocycle (fraction cycling, fc) goes to saturation with increasing laser pulse energy, the absorbed energy by BR increases linearly with pulse energy. From the local analysis we conclude that the light effect changes the kinetics much earlier, already at the L intermediate state decay. The transient electric signal, caused by proton release and uptake, can be decomposed into two components similarly to the absorption kinetic data of the M intermediate state. The actinic light energy affects mainly the ratio of the two components and the proton movements inside BR while pH has an effect on the kinetics of the proton release and uptake groups at the membrane surface.  相似文献

The deposition behavior and photoelectric response characteristics of chlorophylla (Chla) monolayers and multilayers were investigated under various film fabrication conditions. Chla LB films were deposited onto quartz and pretreated ITO glass substrates under several fabrication conditions, including surface pressure and number of layers. The absorption spectra of Chla in a solution state and solid-like state (LB films) were fairly consistent with each other, and two absorption peaks were found at 678 and 438 nm, respectively. The prepared Chla LB films were set into an electrochemistry cell equipped with a Pt plate as the counter electrode, and the photoelectric response characteristics were obtained and analyzed relative to the light illumination. By considering the resulting photocurrents, the optimal fabrication conditions for Chla LB films were determined as 20 mN/m of surface pressure and 20 layers. The action spectrum of the Chla LB films was obtained in the visible region, and was found to be in good agreement with the absorption spectrum. The possible application of the proposed system as a constituent of an artificial color recognition device was suggested based on combining with the photoelectric conversion property of another lightsensitive biological pigment.  相似文献

Photochromic ligands, molecules that can be induced to change their physical properties through applied light, are currently the topic of much chemical biology research. This specialized class of small organic structures are, surprisingly to many, fairly common in nature. At the core of a number of natural biological processes lies a small molecule that changes shape or some other measurable property in response to light absorption. For instance, conformational changes invoked by reversible photoisomerization of a retinoid small molecule found in the photoreceptors of the human eye leads to vision. In plants, photoisomerization of a cinnamate moiety leads to altered gene expression. The photosensitive molecule can be viewed simply as a nanosensor of light, much like a photosensitive electrical component might be added to a circuit to sense day versus night to turn an electrical circuit on or off. Synthetic organic chemists and chemical biologists have been, for at least the last 15 years, trying to either mimic or exploit the native photochromism found in nature. Here, we describe the design process to develop a photochromic molecule to be used in neurobiology.  相似文献

Although the physiological functions of phytochrome A (PhyA) are now known, the distribution of endogenous PhyA has not been examined. We have visualized endogenous PhyA apoprotein (PHYA) by immunolabeling cryosections of pea tissue, using PHYA-deficient mutants as negative controls. By this method, we examined the distribution of PHYA in different tissues and changes in its intracellular distribution in response to light. In apical hook cells of etiolated seedlings, PHYA immunolabeling was distributed diffusely in the cytosol. Exposure to continuous far-red (cFR) light caused a redistribution of the immunolabeling to the nucleus, first detectable after 1.5 hr and greatest at 4.5 hr. During this time, the amounts of spectrally active phytochrome and PHYA did not decline substantially. Exposure to continuous red (cR) light or to a brief pulse of red light also resulted in redistribution of immunolabeling to the nucleus, but this occurred much more rapidly and with a different pattern of intranuclear distribution than it did in response to cFR light. Exposures to cR light resulted in loss of immunolabeling, which was associated with PHYA degradation. These results indicate that the light-induced intracellular location of PHYA is wavelength dependent and imply that this is important for PhyA activity.  相似文献

The initial photoinduced charge separation in bacteriorhodopsin is shown to occur in <5 ps. This result is obtained by measuring the photovoltage rise time in an oriented film of bacteriorhodopsin (BR). A dye laser syncronously pumped by an Argon ion cw mode locked laser is used to produce 3-ps light pulses which, after passing through a dye amplifier chain, photoexcite the BR sample. The photovoltage transient is detected by an ultra-fast Josephson junction digital sampling oscilloscope with liquid-helium-cooled input circuitry.  相似文献

When a light pulse of 1 h duration was given 3 h after lights off in a photoperiod of 11 h light : 13 h dark (LD 11 : 13) at 20°C, the phase of the major peak of locomotor activity rhythm in Delia antiqua was delayed for approximately 0.6 h. In contrast, it was advanced by approximately 0.6 h by a light pulse given 9 h after lights off. It is suggested that in the circadian clock, a pulse falling in the early scotophase is taken as a new dusk and a pulse falling in the late scotophase is taken as a new dawn. Although a sharply defined critical photoperiod did not exist in the diapause response to photoperiod in D. antiqua, the percentage of pupal diapause decreased by these pulses in LD 11 : 13 at 20°C. The effect of a 15 min light pulse on both locomotor activity rhythm and pupal diapause induction was stronger at 3 h than at 9 h after lights off, while a 1 min light pulse was ineffective at both times. The parallel effects of light pulse on locomotor activity rhythm and diapause response might be based on the same chronobiological functions.  相似文献

A method is described for measuring small differences in the acoustic sensitivity of protocerebral interneurons on one side of the noctuid brain when ultrasonic pulses are directed first at one tympanic organ and then at the other. In 23 preparations ipsilateral sensitivity of the brain interneurons was consistently greater by 3 to 4 dB (range 0–7 dB). The spike response of protocerebral interneurons to tympanic stimulation was accompanied by a negative potential having a time course of 40 to 50 msec in response to a 10 msec stimulus pulse. The consistent positive ipsilateral bias in the sensitivity of brain interneurons is much less than the increased sensitivity of the tympanic organ when sounds originate on the ipsilateral side as compared with its sensitivity to sounds directed at the contralateral side. A possible neural mechanism and the behavioural significance of this arrangement are discussed.  相似文献

Chemically modified bacteriorhodopsin (BR) films embedded in gelatin matrix were fabricated. It was found that chemically modified BR/gelatin film remained the original conformation of BR and possessed homogeneity. Then, the photochemical conversion from all-trans O state to 9-cis P state in the chemically modified BR film/gelatin was investigated. For comparison purpose, the same conversion in the wild-type BR (BRwt) film was also presented here. To our know, it is the first report to show that the absorption changes of the P state in the chemically modified BR film were larger than in the BRwt film upon illumination with red light at the low actinic power density.  相似文献

The present study investigated thermoelectric phenomena in the cuticle of the Oriental hornet Vespa orientalis (Hymenoptera, Vespinae). This was done in dependence on the pigment extant at various cuticular region, that is, the brown cuticle in which the primary pigment is melanin and embedded within the cuticle, and the yellow stripes in which the yellow pigment is comprised of purines and pteridines that are located in special pockets between the upper part of the cuticle and the basement membrane. The yellow pigment could be separated from the cuticle proper, but the brown pigment was not thus separable. We found that all cuticular regions of the gaster evinced a thermoelectric response, in that with rise in temperature there was a rise in the thermoelectric current, and vice versa. Additionally, the intact hornet displayed a negative photoelectric response in each of its yellow segments, so that upon illumination with UV light, the maximal current dropped by about 40-50%. Measurements taken on individual stripes in the gaster segments revealed that the photoelectric response is elicited only in the yellow stripes. In all the latter the photoelectric response persists but the maximal current level is lower than in the intact whole hornet. If the yellow pigment is detached mechanically or by bacterial incubation, the photoelectric property of the cuticle is abrogated. Likewise the photoelectric property is abrogated upon immersion of the cuticle in alcohol, even though the yellow pigment is still retained. The specific heat of the yellow stripes in the cuticle is about twice as high as that of the same stripes that had been depleted of their yellow pigment, amounting to 1.8-1.9 J/g.K vs. 0.8 J/g.K.  相似文献

The transpiration response of excised primary Avena leaves was studied when pulse perturbations were given to the water regulatory system. Repeated light pulses given to the leaf caused regularly alternating transpiration responses, i.e. the magnitude alternated regularly between a high and a low value. This effect, denoted alternating pulse response, could be recorded under quite different light pulse conditions but was not found when the pulse interval was too long or too short (longer than about 60 min. shorter than about 15 min). Sodium chloride given to the transpiration stream induced and increased the effect. Alternating pulse response could also be recorded when mannitol pulses were given to the root system of intact plants.  相似文献
