In strictly anaerobic conditions in a culture medium adjusted to pH 5·2 with HCl and incubated at 30°C, inocula containing < 10 vegetative bacteria of Clostridium botulinum ZK3 (type A) multiplied to give > 108 bacteria per ml in 3 d. Growth from an inoculum of between 10 and 100 spores occurred after a delay of 10–20 weeks. Citric acid concentrations of 10–50 mmol/l at pH 5·2 inhibited growth from both vegetative bacteria and spore inocula, a concentration of 50 mmol/l increasing the number of vegetative bacteria or of spores required to produce growth by a factor of approximately 106. The citric acid also reduced the concentration of free Ca2+ in the medium. The inhibitory effect of citric acid on vegetative bacteria at pH 5·2 could be prevented by the addition of Ca2+ or Mg2+ and greatly reduced by Fe2+ and Mn2+. The addition of Ca2+, but not of the remaining divalent metal ions, restored the concentration of free Ca2+ in the medium to that in the citrate-free medium. The inhibitory effect of citric acid on growth from a spore inoculum was only partially prevented by Ca2+. Citric acid (50 mmol/l) did not inhibit growth of strain ZK3 at pH 6 despite the greater chelating activity of citrate at pH 6 than at pH 5·2. The effect of citric acid and Ca2+ at pH 5·2 on vegetative bacteria of strains VL1 (type A) and 2346 and B6 (proteolytic type B) was similar to that on strain ZK3.  相似文献

Barley ( Hordeum vulgare L. cv. CM 72) was grown for a 28-day period and stressed with treatments of 125 mol m−3 NaCl or KC1 with low Ca2+ (0.4 mol m−3 Ca2+) or high Ca2+ (10 mol m−3 Ca2+). Plants were harvested periodically so that relative growth rate (RGR), net assimilation rate (NAR) and leaf area ratio (LAR) could be calculated using the functional approach to plant growth analysis. Relative growth rate declined with time for all treatments, including controls. Salinity inhibited RGR relative to control values by day 10. High Ca2+ improved the growth of salt-stressed plants in both NaCl-salinity and KCl-salinity. KC1 proved more toxic than NaCl, especially for KCI-salinity plants with low Ca2+, which died by day 28. Net assimilation rate, but not LAR, was highly correlated with RGR for all treatments. This indicates that the photosynthetic-assimilatory machinery was limiting RGR and not the leaf area of the plant.  相似文献

The effects of pH on the growth and the K+ (86Rb) uptake and K+ content of excised rice ( Oryza sativa L. cv. Dunghan Shali) and wheat ( Triticum aestivum L. cv. GK Szeged) roots were investigated. Rice roots responded to H+ stress with an increased K+(86Rb) influx and a decreased K+ content, suggesting an increased exchange between the cytoplasmic K+ pool and the external medium. Under the same experimental conditions wheat did not show any anomalous K+(86Rb) influx. Growth of both rice and wheat was relatively insensitive to pH between 4 to 10.  相似文献

Pectinatus frisingensis , a Gram-negative and strictly anaerobic beer spoilage bacterium is sensitive to nisin. An increase in nisin concentration (0 to 1100 IU ml−1) added to the culture medium prolonged the lag phase, and decreased the growth rate of the bacterium. In addition, late exponential cells of P. frisingensis exposed to low concentrations of nisin lost immediately a part of their intracellular K+. Presence of Mg2+ up to 15 mmol l−1 did not protect P. frisingensis from nisin-induced loss of viability and K+ efflux. Potassium leaks were also measured in P. frisingensis late exponential phase cells exposed to combined effects of nisin addition (100–500 IU ml−1), 10 min mild heat-treatment (50 °C) or rapid cooling (2 °C), and pH (4·0 and 6·2). Net K+ efflux from both starving and glucose-metabolizing cells, was more important at pH 6·2, whatever the temperature treatment and nisin addition. Reincubation at 30 °C of P. frisingensis glucose-metabolizing cells exposed to a preliminary combination of nisin addition and mild heat or cooling down treatment, showed that cells exposed to rapid cooling reaccumulated more K+ than heat-treated cells, whatever the pH conditions. A combination of nisin and mild heat-treatment could thus be of interest to prevent P. frisingensis growth in beers.  相似文献

Abstract: Increasing extracellular pH from 7.4 to 8.5 caused a dramatic increase in the time required to recover from a glutamate (3 µ M , for 15 s)-induced increase in intracellular Ca2+ concentration ([Ca2+]i) in indo-1-loaded cultured cortical neurons. Recovery time in pH 7.4 HEPES-buffered saline solution (HBSS) was 126 ± 30 s, whereas recovery time was 216 ± 19 s when the pH was increased to 8.5. Removal of extracellular Ca2+ did not inhibit the prolongation of recovery caused by increasing pH. Extracellular alkalinization caused rapid intracellular alkalinization following glutamate exposure, suggesting that pH 8.5 HBSS may delay Ca2+ recovery by affecting intraneuronal Ca2+ buffering mechanisms, rather than an exclusively extracellular effect. The effect of pH 8.5 HBSS on Ca2+ recovery was similar to the effect of the mitochondrial uncoupler carbonyl cyanide p -(trifluoromethoxyphenyl)hydrazone (FCCP; 750 n M ). However, pH 8.5 HBSS did not have a quantitative effect on mitochondrial membrane potential comparable to that of FCCP in neurons loaded with a potential-sensitive fluorescent indicator, 5,5',6,6'-tetrachloro-1,1',3,3'-tetraethylbenzimidazolocarbocyanine iodide (JC-1). We found that the effect of pH 8.5 HBSS on Ca2+ recovery was completely inhibited by the mitochondrial Na+/Ca2+ exchange inhibitor CGP-37157 (25 µ M ). This suggests that increased mitochondrial Ca2+ efflux via the mitochondrial Na2+/Ca2+ exchanger is responsible for the prolongation of [Ca2+]i recovery caused by alkaline pH following glutamate exposure.  相似文献

The effects of copper (CuCl2) on active and passive Rb+(86Rb+) influx in roots of winter wheat grown in water culture for 1 week were studied. External copper concentrations in the range of 10–500 μ M in the uptake nutrient solution reduced active Rb+ influx by 20–70%, while passive influx was unaffected (ca 10% of the Rb+ influx in the Cu-free solution). At external Rb+ concentrations of up to 1 m M , Cu exposure (50 μ M decreased Vmax to less than half and increased Km to twice the value of the control. Short Cu exposure reduced the K+ concentration in roots of low K+ status. Pretreatment for 5 min in 50 μ M CuCl2 prior to uptake experiments reduced Rb+ influx by 26%. After 60 min pretreatment with Cu, the corresponding reduction was 63%. Cu in the cultivation solution impeded growth, especially of the roots. The Cu concentration in the roots increased linearly with external Cu concentration (0–100 μ M ) while Cu concentration in the shoots was relatively unchanged. The K+ concentration in both roots and shoots decreased significantly with increased Cu in the cultivation solutions. Possible effects of Cu on membranes and ion transport mechanisms are discussed.  相似文献

Bavaricin MN, a bacteriocin produced by Lactobacillus bavaricus MN, reached titres of 2000 AU ml-1 in APT broth maintained at pH 6.0, 30°C in a batch fermenter. Levels of bavaricin MN at pH 5.5 and 6.5 were lower despite comparable levels of producer cells. The addition of 3.0 g l-1 beef extract to APT broth resulted in increases in both the growth rate of the culture and the production of bavaracin MN. The titre of bavaricin MN in batch fermenters controlled at pH 6.0 in APT broth plus 3 g l-1 beef extract reached 3200 AU ml-1 at 30°C. This level was reduced to 800 AU ml-1 by 76 h. Glucose-limited continuous culture of Lact. bavaricus MN under the same conditions resulted in an increase in the titre of bavaricin MN to 6400 AU ml-1. This level was maintained, independent of growth rate, for 345 h. Growth rates of 0.205, 0.118, 0.169 and 0.058 h-1 were examined.  相似文献

The Na+ uptake mechanism of cardinal tetras Paracheirodon axelrodi displayed specialization for operation in dilute waters of the amazonian Rio Negro. Kinetic analysis revealed low Km and high Jmax values which ensure high rates of uptake even in very dilute waters. In contrast, Na+ uptake of angelfish Pterophyllum scalare did not appear to be specialized for dilute waters at all, with much higher Km and lower Jmax values. Na+ uptake in cardinal tetras was high and completely unaffected down to pH 3·5, while uptake in angelfish was much lower and progressively inhibited by dropping pH; it was completely shut down at pH 3·5. During chronic exposure to pH 4·0 and 3·5, angelfish showed no ability to restore Na+ uptake and at pH 3·5 all individuals died between day 3 and 6 presumably due to small, but steady loss of Na+. At low pH, both species displayed a strong ability to prevent stimulation of diffusive Na+ losses. In angelfish, the ability to control diffusive ion losses at low pH was related to a high branchial affinity for Ca2+. For cardinal tetras, the rate of Na+ loss appeared to be independent of water Ca2+ concentration. The specializations of cardinal tetras are similar to other characid fishes tested, but the results for angelfish reveal a new pattern of ion regulation in acidophilic species from the Rio Negro. The differences between the species may be the result of their different distributions, cardinal tetras are found in the central Rio Negro region while angelfish are more peripheral, or they may reflect phylogenetic differences.  相似文献

The photosynthetic characteristics of the terrestrial cyanobacterium, Nostoc flagelliforme , after complete recovery by rewetting, was investigated to see whether it could use bicarbonate as the external inorganic carbon source when submerged. The photosynthesis–pH relationship and high pH compensation point suggested that the terrestrial alga could use bicarbonate to photosynthesize when submerged. The photosynthetic oxygen evolution rates were significantly inhibited in Na + -free and Na + + Li + media but were not affected by the absence of Cl , implying that the bicarbonate uptake was associated with Na + / HCO3 symport rather than Cl /HCO3 exchange system.  相似文献

We investigated the interaction of Ca2+ and pH on root elongation in Zea mays L. cv. B73 × Missouri 17 and cv. Merit. Seedlings were raised to contain high levels of Ca2+ (HC, imbibed and raised in 10 m M CaCl2) or low levels of Ca2+ (LC, imbibed and raised in distilled water). In HC roots, lowering the pH (5 m M MES/Tris) from 6.5 to 4.5 resulted in strong, long-lasting growth promotion. Surprisingly, increasing the pH from 6.5 to 8.5 also resulted in strong growth promotion. In LC roots acidification of the medium (pH 6.5 to 4.5) resulted in transient growth stimulation followed by a gradual decline in the growth rate toward zero. Exposure of LC roots to high pH (pH shift from 6.5 to 8.5) also promoted growth. Addition of EGTA resulted in strong growth promotion in both LC and HC roots. The ability of EGTA to stimulate growth appeared not to be related to H+ release from EGTA upon Ca2+ chelation since, 1) LC roots showed a strong and prolonged response to EGTA, but only a transient response to acid pH, and 2) promotion of growth by EGTA was observed in strongly buffered solutions. We also examined the pH dependence of the release of 45Ca2+ from roots of 3-day-old seedlings grown from grains imbibed in 45Ca2+. Release of 45Ca2+ from the root into agar blocks placed on the root surface was greater the more acidic the pH of the blocks. The results indicate that Ca2+ may be necessary for the acid growth response in roots.  相似文献

Six cultivars of spring barley ( Hordeum vulgare L. cvs Salve, Nümberg II, Bomi, Risø 1508, Mona and Sv 73 608) were grown in water culture for three weeks with various combinations of mineral supply and differential roots/shoot temperatures during the growth period. Most important for growth and accumulation of N, K+, Ca2+ and Mg2+ was the mineral supply, followed by the root temperature and the choice of cultivar. Treatments with low mineral supply or low root temperature induced a uniform reduction in growth and accumulation of the ions studied. The effects of low mineral supply and low root temperature on growth and N accumulation was additive, which indicates that these factors exert their influence independently of each other.
Roots grown at 10°C were smaller and Rb+(86Rb) influx was higher than in roots grown at 20°C. It is suggested that the control of Rb+(86Rb) influx is affected by the root temperature and the age of the plants. The higher 86Rb+ (86Rb) influx into the low temperature roots could not compensate for the smaller root size. However, the lower total mineral accumulation made up for the needs of the smaller plants and cannot explain the reduction in growth.  相似文献

The effect of 253.7 nm ultraviolet radiation on elongation growth, medium acidification and changes in electric potential difference between vacuole and external medium in cells of maize ( Zea mays L.) coleoptile segments was investigated. It was found that irradiation with 390, 1170, 3900 and 5 850 J m−2 UV-C (ultraviolet radiation 253.7 nm) inhibited elongation growth, whereas at 195 J m−2 stimulation of growth was observed. The administration of IAA (10−5 M ) to the incubation medium of coleoptile segments partially abolished the inhibitory effect of UV-C. The pH of the incubation medium, measured simultaneously with growth, showed that the exposure of the segments to UV-C caused inhibition of H+-extrusion (or stimulation of H+ uptake). The presence of IAA (10−5 M ) in the incubation medium promoted (except after 5850 J m−2 irradiation) H+-extrusion to a level comparable with that produced by IAA in non-irradiated segments. In UV-C irradiated segments the potential difference underwent significant alterations. Irradiation of coleoptile segments with 390 J m−2 caused a transient depolarization, which was fully reversible within 30 min, while at higher doses depolarization was irreversible. The hyperpolarization of the membrane potential (MP) in cells of maize coleoptile induced by IAA was completely nullified by subsequent irradiation with UV-C. It is suggested that UV-C inhibited IAA-induced growth by a mechanism independent of cell wall acidification.  相似文献

N-sufficient cells of Chlorella sorokiniana Shihira and Krauss, strain 211/8k, absorbed NH4+ under light plus CO2 conditions, when growth occurred, but not in darkness or in the absence of CO2, when growth was inhibited. N-sufficient cells subjected to conditions of N-starvation for a 24-h period showed a marked loss of photosynthetic activity. Upon supply of NH4+, N-starved cells sufflated with CO2 air exhibited a time-dependent recovery of photosynthetic activity, both when suspended in light and in darkness. By contrast, growth only occurred in cells suspended in light. N-starved cells absorbed NH4+ in darkness, but at a lower rate than in light. All of these data suggest that dark NH4+ uptake is driven by N assimilation to recover from N-starvation and that the light-dependent NH4+ uptake is driven by growth, being then influenced by conditions that affect recovery or growth. Unlike CO2 conditions, in a CO2-free atmosphere, absorption of NH4+ by N-starved cells occurred at a higher rate in darkness than in light. Accordingly, resumption of photosynthetic potential after NH4+ supply occurred in darkened cells, but not in illuminated cells. Respiratory activity of N-starved cells was enhanced up to 3-fold by NH4+ and 2-fold by methylammonium, with different patterns, suggesting that respiratory enzymes were affected by N-metabolism, especially through short-term control mechanisms triggered by the expenditure of metabolic energy involved in N-metabolism.  相似文献

SUMMARY 1. Unialgal cultures of three species common in the freshwater phytoplankton were used to test limitation of specific growth rate and final yield in defined media of low K+ concentration (range <0.3–6 μmol L−1 or mmol m−3).
2. Growth rate of the diatom Asterionella formosa was independent of K+ concentration above 0.7 μmol L−1. Final yield was dependent on initial concentration when accompanied by K+ depletion below this concentration, but not by lesser depletion with more residual K+. Analyses of particulate K in the biomass indicated a mean final cell content of 2.8 μmol K 10−8 cells, approximately 1.0% of the organic dry weight.
3. Less detailed work with the diatom Diatoma elongatum showed no dependence of growth rate or final yield upon the initial K+ concentration in the range 0.8–3.2 μmol L−1. The phytoflagellate Plagioselmis nannoplanctica suffered net mortality in the lowest concentration tested, 0.8 μmol L−1.
4. Comparison with the range of K+ concentration in natural fresh waters, including a depletion induced by an aquatic macrophyte, suggests that K+ is unlikely to limit growth of phytoplankton. Nevertheless, there can be correlation of K+ with lake trophy.  相似文献

Abstract: The temporal resolution of carbon-fiber microelectrodes has been exploited to examine the plasticity of quantal secretory events at individual adrenal medullary cells. The size of individual quantal events, monitored by amperometric oxidation of released catecholamines, was found to be dependent on the extracellular ionic composition, the secretagogue, and the order of depolarization delivery. Release was observed with either exposure to 60 m M K+ in the presence of Ca2+ or exposure to 3 m M Ba2+ in solutions of different pH, with and without external Ca2+. Ba2+ was demonstrated to induce Ca2+-independent exocytotic release for an extended period of time (>4 min) relative to release induced by K+ (∼30 s), which is Ca2+ dependent. In all cases, simultaneous changes of intracellular divalent cations, monitored by fura-2 fluorescence, accompanied quantal release and had a similar time course. Exocytosis caused by Ba2+ in Ca2+-free medium had a larger mean spike area at pH 8.2 than at pH 7.4. When Ba2+-induced spikes measured at pH 7.4 were compared, the spikes in Ca2+-free medium were found to be broader and shorter but had the same area. Release induced by K+ after exposure to Ba2+ was comprised of larger quantal events when compared with preceding K+ stimulations. Finally, spikes obtained with Ba2+ exposure at an extracellular pH of 5.5 had a different shape than those obtained in more basic solutions. These changes in spike size and shape are consistent with the interactions between catecholamines and other intravesicular components.  相似文献

The hyphal extension rate of Rhizopus oligosporus NRRL 2710 was slowed in the presence of 0·42 and 0·84 mmol NH3 l−1 and inhibited by 1·3 mmol l−1. Sporulation was prevented at NH3 concentrations of 0·42 mmol l−1 andabove. There was no evidence of toxicity due to NH+4 at concentrations up to 300 mmol l−1.Independent of the concentrations of NH3 or NH+4, the lower the pH value, in therange 6·0–9·0, the higher was the rate of hyphal extension. It is suggested that accumulationof toxic levels of NH3 could be responsible for the cessation of mould growth in tempe.  相似文献

The generally rhizotoxic ion Al3+ often enhances root growth at low concentrations. The hypothesis that Al3+ enhances growth by relieving H+ toxicity was tested with wheat seedlings ( Triticum aestivum L.). Growth enhancement by Al3+ only occurred under acidic conditions that reduced root elongation. Al3+ increased cell membrane electrical polarity and stimulated H+ extrusion. Previous investigations have shown that Al3+ decreases solute leakage at low pH and that the alleviation of H+ toxicity by cations appears to be a general phenomenon with effectiveness dependent upon charge (C3+>C2+>Cl+). Alleviation of one cation toxicity by another toxic cation appears to be reciprocal so that Al3+ toxicity is relieved by H+. It has been argued previously that this latter phenomenon accounts for the apparent toxicity of ALOH2+ and Al(OH)+2. Reduction of cell-surface electrical potential by the ameliorative cation may reduce the cell-surface activity of the toxic cation.  相似文献

Abstract. Seedlings of Phaseolus vulgaris were exposed to solutions containing Cd2+ in the range 0 to 1 molm−3. Ethylene formation started following 3 h of exposure to 10−2, 10−1 and 1 mol m−3 Cd2+, peaked at 18 h and returned to a relatively low rate after 24 h. Cadmium-induced ethylene formation depended on the formation of 1-aminocyclopropane-1-carboxylic acid (ACC). Aminoethoxyvinylglycine (AVG, 0.1 mol m−3) inhibited ACC accumulation and ethylene production during exposure to 0.2 mol m−3 Cd2+.
Activity of soluble and ionically-bound peroxidase increased after 18 h of exposure to Cd2+ concentrations above 10−3 mol m−3 due to an increase in activity of cathodic isoperoxidases. Stimulation of soluble and ionically-bound peroxidase by 0.2 mol m−3 Cd2+ was reduced in the presence of 0.1 mol m−3 AVG.
Accumulation of soluble and insoluble ('ligninlike') phenolics was found in plants exposed to Cd2+ (10−2 mol m−3 or above) in the presence or absence of AVG. Deposition of insoluble (autofluorescing) material occurred in cell walls around vessels and was associated with reduced expansion and water content of leaves.  相似文献

A highly proteolytic bacterium isolated from abattoir effluent was identified as a non-pigmented strain of Chromobacterium lividum. Ferrous or ferric ions at concentrations between 1·8 × 10-5 and 9 × 10-4 g ions/1, which is 2–3 orders of magnitude greater than that required for growth, were essential for extracellular proteinase production in aerated but not in static culture. Co2+, Ni2+, Mn2+, Cu2+ or Zn2+ ions could not replace iron. Four proteinases (I-IV) were produced in static culture, but only proteinase I was formed in significant quantities in aerated culture. With both forms of culture amino nitrogen was essential for proteinase production; glucose inhibited formation in aerated, but not static, cultures. Growth occurred over the range 1–33 °C, whereas proteinase production ceased at 27 °C, with maximum activity at 13 °C. Proteinase production appeared to be controlled by an interaction between iron, oxygen tension and glucose.  相似文献

The growth rate of mung bean epicotyls was used for evaluating the effect of brassinolide on cell elongation. Growth above that of control plants was observed at 10−10 M and above. Gibberellic acid showed an additivity relationship with low concentrations (10−9–10−8 M ) of brassinolide in this test system and the two growth promoters may therefore act independently at the cellular level. Because of relative ease of operation, great sensitivity and the short time required for assessing biological activity, this assay could be used in conjunction with the bean second internode bioassay for evaluating the activity of brassinolide and its analogs, as well as of other growth promotors.  相似文献
