The highest yield of protoplasts in the strainAspergillus niger K 10 was obtained from young, freshly harvested hyphae, grown on simple medium of sucrose-asparagine type on a rotary shaker. The residual cultivation medium has to be washed from the mycelial suspension with a solution of high osmotic pressure. Lyophilized snail digestive juice in concentration of 1 %, temperature 31° C, and incubation in Erlenmayer flasks on reciprocal shaker were optimal for the release of protoplasts. Good stabilizers of released protoplasts (in combination with CaCl2) were for example galactose, mannitol, inositol and mixture of NaCl with glucose, sucrose or lactose.  相似文献

Quantitation of exact total protein content is often a key step and is common to many applications in general biochemistry research and routine clinical laboratory practice. Before embarking on any type of protein analysis, particularly comparative techniques, it is important to accurately quantitate the amount of protein in the sample. In order to assess the quality of total protein estimation results, five methods were tested and were applied to the same pooled plasma sample. For this aim, Bradford (Coomassie Brilliant Blue), Lowry (Folin-Ciocalteau), Biüret, Pesce and Strande (Ponceau-S/TCA), and modified method of Schaffner-Weismann (Amido Black 10B) were used. The last two methods employ simultaneous precipitation of proteins with the acid containing dye solutions followed by dissolution of precipitate in a NaOH solution. It is shown that each assay has advantages and disadvantages relative to sensitivity, ease of performance, acceptance in literature, accuracy and reproducibility/coefficient of variation. All of the methods tested show a CV % < 6. Besides pooled plasma, a known concentration of human serum albumin was also analyzed and discussed by means of standardization of plasma total protein content.  相似文献

<正> 本文对水解蛋白注射液中色氨酸比色测定的方法进行了比较研究,结果表明用N-1-萘基乙二胺方法比对二甲氨基苯甲醛法具有灵敏度高(约相当于20倍),精密度好(前法的变异系数为1.7%,后法为13.1%)等优点,两法的平均回收率均为97%。另外,还测定了上述二法的吸收曲线和应用两法对武汉地区用不同工艺生产的水解蛋白注射液进行了实际考察。结果表明用酶法生产的水解蛋白,其色氨酸含量可达550μg/ml。  相似文献

Several methods for ergosterol determination in two different Saccharomyces cerevisiae strains were tested. The best results were obtained by a simple method including saponification with 8 M KOH, extraction of non-saponifiable lipids and ergosterol determination by reversed phase HPLC. The methods in which the samples were treated with 0.1 M HCl prior to hydrolysis with 2.5 or 4.5 M water-ethanolic KOH underestimated the ergosterol contents.  相似文献

The effects of nine antimycotics on the biosynthesis of cellular macromolecules were analyzed using the regenerating system of protoplats ofAspergillus niger. The incorporation of several specific radioactive precursors into major cellular components were measured in the presence or in the absence of respective agents. Miconazole, ketoconazole, and tolnaftate inhibited the lipid synthesis. 5-Fluorocytosine strongly inhibited the DNA and protein syntheses. Griseofulvin, however, specifically inhibited the synthesis of cell wall polysaccharides, i.e. chitin and glucan. Other agents showed non-specific inhibition effects. Conidia of Aspergillus niger 20 Osm producing extracellular inulinase were immobilized on pumice stones or polyurethane sponge and used in repeated-batch processes. Some factors affecting inulinase biosynthesis by the mycelium A. niger immobilized on pumice stones were investigated. Maximal inulinase production occurred in 50 ml of medium containing 0.5 g of carrier at 30 °C, pH 6.0 and at an agitation speed of 200 rpm. This procedure enabled repeated-batch enzyme production and as many as six subsequent 24 h batches could be fermented by using the same carrier. This is the first report on inulinase biosynthesis by mycelium of A. niger immobilized on polyurethane sponge using unconventional oxygenation of culture which ensures that the dissolved oxygen concentration remains constant.  相似文献

Summary A comparison of citric acid fermentations in manganese-deficient and manganese-containing media showed that manganese strongly influences idiophase metabolism. In the presence of manganese, cell growth increases, sugar consumption is diminished and acidogenesis decreases drastically. An investigation of the key enzymes of glycolysis, the pentosephosphate pathway, TCA-cycle, nitrogen metabolism, and gluconeogenesis indicated that manganese deficiency was accompanied by a repression of anabolic and TCA-cycle-enzymes with the exception of citrate synthase. The activity of this enzyme and the enzymes of glycolysis paralleled the sugar consumption rate. In the presence of manganese, no repression of enzyme synthesis was observed. Activities of 2-oxoglutarate dehydrogenase and isocitrate lyase could not be detected in either case. The results support the hypothesis that manganese deficiency mainly affects the operation of biosynthetic reactions inAspergillus niger, thus leading to an overflow of citric acid as an end product of glycolysis.  相似文献

Mycelial fungi play a central role in element cycling in nature by degrading dead organic material such as wood. Fungal colonization of a substrate starts with the invasion of exploring hyphae. These hyphae secrete enzymes that convert the organic material into small molecules that can be taken up by the fungus to serve as nutrients. Using green fluorescent protein (GFP) as a reporter, we show for the first time that exploring hyphae of Aspergillus niger differentiate with respect to enzyme secretion; some strongly express the glucoamylase gene glaA, while others hardly express it at all. When a cytoplasmic GFP was used, 27% of the exploring hyphae of a 5-day-old colony belonged to the low expressing hyphae. By fusing GFP to glucoamylase and by introducing an ER retention signal, this number increased to 50%. This difference is due to cytoplasmic streaming of the reporter in the former case, as was shown by using a photo-activatable GFP. Our findings indicate that a fungal mycelium is highly differentiated, especially when taking into account that hyphae in the exploration zone were exposed to the same nutritional conditions.  相似文献

Fungal mycelia are exposed to heterogenic substrates. The substrate in the central part of the colony has been (partly) degraded, whereas it is still unexplored at the periphery of the mycelium. We here assessed whether substrate heterogeneity is a main determinant of spatial gene expression in colonies of Aspergillus niger. This question was addressed by analyzing whole-genome gene expression in five concentric zones of 7-day-old maltose- and xylose-grown colonies. Expression profiles at the periphery and the center were clearly different. More than 25% of the active genes showed twofold differences in expression between the inner and outermost zones of the colony. Moreover, 9% of the genes were expressed in only one of the five concentric zones, showing that a considerable part of the genome is active in a restricted part of the colony only. Statistical analysis of expression profiles of colonies that had either been or not been transferred to fresh xylose-containing medium showed that differential expression in a colony is due to the heterogeneity of the medium (e.g., genes involved in secretion, genes encoding proteases, and genes involved in xylose metabolism) as well as to medium-independent mechanisms (e.g., genes involved in nitrate metabolism and genes involved in cell wall synthesis and modification). Thus, we conclude that the mycelia of 7-day-old colonies of A. niger are highly differentiated. This conclusion is also indicated by the fact that distinct zones of the colony grow and secrete proteins, even after transfer to fresh medium.  相似文献

The effect of different carbon sources on the sugar concentration and composition of theAspergillus niger mycelium has been studied at different days of incubation. No positive correlation could be drawn among dry weight, growth rate, and concentration of sugar and with substrate and sugar composition of mycelium. Glucose was the predominant sugar in all the cases.

A simple method is described for the immobilization of Aspergillus niger GIV-10 which produces an extracellular glucose oxidase. A. niger conidia were immobilized on sintered glass Raschig rings, pumice stones or polyurethane foam. Mycella growing out from the spores produced extracellular glucose oxidase: the highest production was with the pumice stone carrlers. This technique facilitates the growth of the filamentous cultures in the spongy structure of a support with continuous accumulation of biomass. After 24 to 36 h, a culture liquid with 2.7 to 3.1 U of glucose oxidase/ml was obtained. This procedure also made possible repeated batch enzyme production and as many as 25 subsequent 24-h batches could be fermented by using the same carrier with only a small loss of glucose oxidase activity.The authors are with the Institute of Microbiology, M. Lytic enzyme activity in autolysing mycelium of Aspergillus niger   总被引:1,自引:0,他引:1  
We have studied changes in the activity of some lytic enzymes contained in mycelium of Aspergillus niger in cultures relative to the autolytic phase of growth. Acid phosphatase, polygalacturonidase and alpha-amylase activity reached its highest level (40.7, and 8 U/sample, respectively) at the initiation of the autolytic phase of growth. 1.3-beta-Glucanase and beta-N-acetylglucosaminidase reached its highest level (3.5 and 2 U/sample, respectively) during the first days of autolysis. Alkaline phosphatase, cellulase, invertase, esterase, chitinase and proteolytic activity is also present in autolysing mycelium of A. niger, though comparatively low. Their maximum activity coincided with the beginning of the autolytic phase of growth. In all enzymes studied here, as autolyis proceeded, enzyme activity decreased by about 90%. Only esterase activity remained nearly constant throughout the whole period of autolysis described here.  相似文献

J. FIEDUREK, J. SZCZODRAK AND J. ROGALSKI. 1995. A simple method for the immobilization of Aspergillus niger mycelium producing polygalacturonase (PG) and pectinesterase (PE) is described. Fungal conidia were immobilized on wheat, rye, barley, peas, buckwheat and mustards seeds. Spongy mycelia overgrowing the seed surfaces on mineral medium with pectin produced extracellular PG and PE; the highest production was reached on the wheat carrier. Some of the variables influencing the enzymatic activity have been optimized. After every 24 h, a culture liquid with 6˙8–7˙8 U of PG ml−1 and 7˙0–10˙1 U of PE ml−1 was obtained. This procedure also made possible repeated batch enzyme production and, as many as eight subsequent 24-h batches could be fermented by using the same carrier without any loss of PG activity. The addition of sodium orthovanadate (1 mmol) into the medium with pectin caused a significant increase in PG and PE activity produced by free cells of A. niger (by 1˙59-fold and 1˙67-fold respectively), and only 0˙47-fold of PG activity in case of the immobilized mycelium.  相似文献

Summary A pectin methylesterase-encoding gene (pmeA)_has been cloned and transformed intoA. niger wild-type NRRL3. Transformants produced 20-fold more PME than the host strain. For studying the effects of different promoters on thepmeA expression two novel plasmids were constructed, in which thepmeA promoter was replaced by efficient promoters such as theA. nidulans glyceraldehyde-3-phosphate dehydrogenase (pK45) or theA. oryzae -amylase (pK61) promoter. Abstract. The measurement of diversity, one of the most important concepts in present-day ecology, can be improved by methods of diversity ordering which have recently been developed. This ordering is achieved by a D(α) diversity index family. Indices of this family show varying sensitivities to the rare and abundant species as the scale parameter, α, changes. The aim of this paper is to review and assess 12 methods of diversity ordering and discuss their relationships in detail. Two of the methods are new to the ecological literature. The diversity ordering methods are compared as to their effectiveness in graphically displaying the differences of community structure and demonstrating the (non-)comparability of communities. Small, medium and large data sets were used to evaluate the methods. A small artificial data set (five to seven species) and a large semi-artificial data set (31 — 141 species) are used in this paper. The results suggest that Rényi's diversity index family and Logarithmic dominance ordering are the most useful methods for diversity ordering of communities of all sizes. Right-tailsum diversity ordering performs well for small communities.  相似文献
