The vegetative growth of Dactylis glomerata L. in sand was studiedunder controlled light, temperature, and nutritional conditions.Plants were daily supplied with three nutrient solutions ofdifferent nitrate concentrations (10–2, 10–3 and2 x 10–4 mol I–1). For each concentration, growthobeyed an exponential law between the fourth and seventh weeksafter sowing. The time constant of the exponential was the samefor the shoot as for the root, and showed no significant variationwith nitrate concentration. The kinetic results and the strong dependence of the root: shootratio on nitrate concentration are discussed on the basis ofThornley's model. Hypothesizing that the molecular mechanismsof nitrate absorption are independent of the nitrate concentrationof the nutrient solution, we derived a relationship betweenthe root: shoot ratio and nitrate concentration. Bot. 38: 42–52 Oilseed rape (Brassica napus L. cv. Bien venu) was grown for49 d in flowing nutrient solution at pH 6?0 with root temperaturedecrementally reduced from 20?C to 5?C; and then exposed todifferent root temperatures (3, 5, 7, 9, 11, 13,17 or 25?C)held constant for 14 d. The air temperature was 20/15?C day/nightand nitrogen was supplied automatically to maintain 10 mmolm–3 NH4NO3 in solution. Total dry matter production wasexponential with time and similar at all root temperatures givinga specific growth rate of 0?0784 g g–1 d–1. Partitioningof dry matter was influenced by root temperature; shoot: rootratios increased during treatment at 17?C and 25?C but decreasedafter 5 d at 3?C and 5?C. The ratio of shoot specific growthrate: root specific growth rate increased with the ratio ofwater soluble carbohydrates (shoot: root). Concentrations ofwater soluble carbohydrates in shoot and root were inverselyrelated to root temperature; at 3, 5 and 7?C they increasedin stem + petioles throughout treatment, coinciding with a decreasein the weight of tissue water per unit dry matter. These resultssuggest that the accumulation of soluble carbohydrates at lowtemperature is the result of metabolic imbalance and of osmoticadjustment to water stress. The effect of changes in nitrate-nitrogen supply on the absorptionand distribution of copper was examined in grasses grown inflowing solution culture with a maintained concentration ofcopper. Absorption by roots (µg Cu g–1 dry root)decreased markedly when nitrogen had been depleted or was maintainedat 0.1 mg l–1 N, but there was an immediate increase whennitrogen was maintained at 1.0 or 10.0 mg l–1. There werealso large increases in the concentration of copper in the shootsof plants grown with 1.0 and 10.0 mg 1–1 N. The rootsof plants grown with 0.1 or 1.0 mg 1–1 N retained similarproportions of uptake, but a lower proportion was retained whenthe plants were grown with 10.0 mg 1–1. Although a lowerproportion of the copper was associated with cell walls in theplants grown at 10.0 mg 1–1 N this was the result of alower content of cell walls rather than an effect on copperitself. In a longer-term experiment in conventional solutionculture with a range of nitrogen concentration, the concentrationof copper in shoots was largely determined by shoot growth. Decoated seeds of Clitoria ternatea L. germinated on Murashigeand Skoog (Physiologia Plantarum 1962, 15, 473–97) basalmedium (BM) and differentiated callus and bipolar embryoids(two-step method) in low frequency. Calluses developed on lateralroots [BM+KN(0.1 mg 1–1)], on roots and hypocotyls [BM+KN(0.5mg 1–1)], and on roots [BM+KN+IAA (0.5 mg 1–1 ofeach)]. On basal medium with KN (0.5 mg 1–1) and withKN+IAA (0.5 mg1–1 of each), multiple shoot buds and embryoids(one-step method) were differentiated directly on split hypocotylsand roots. In the former, shoot buds developed even on unsplithypocotyls. Rhizogenesis on isolated shoot buds occurred efficientlyin BM+indole butyric acid (IBA 0.1 mg 1–1) and BM+IAA(0.1 mg 1–1 and 0.5 mg 1–1). Profuse direct embryoidsand shoot buds developing on root systems are interesting morphogeneticphenomena rarely reported. Root–shoot interaction in the tea plant, in relation toturnover of root reserves, following pruning and manuring, wereinvestigated by ring barking the roots or severing mature rootsin both unpruned and pruned plants. Barley (Hordeum vulgare L., cvs Golf and Laevigatum) was grownunder nitrogen limitation in solution culture until near maturity.Three different nitrogen addition regimes were used: in the‘HN’ culture, the relative rate of nitrate-N additionwas 0·08 d–1 until day 48 and then stepwise decreasedto, finally, 0·005 d–1 during late grain-filling;the ‘LN’ culture received 45% of the nitrogen addedin HN; the ‘CN’ culture was maintained at RA 0·0375d–1 throughout growth. At four different growth stages(vegetative,anthesis, and twice during grain-filling), 15N-nitrate was fedto the plants. In some cases (‘split root cultures’),label was fed only to one-half of the root system. These wereharvested directly after labelling, whereas ‘standardcultured’ plants were harvested at termination of theexperiment (day 148). Absorption of added nitrate was nearlycomplete in the HN and LN cultures, and translocation of nitrogenwithin the plants could thus be studied independently of differencesin nitrate absorption. Cycling of nitrogen absorbed by vegetativeplants accounted for up to 50% of the nitrogen recovered inthe roots. The sink strength of the roots for cycling nitrogen,however, declined during post-anthesis growth, and net lossof nitrogen from both roots and vegetative shoot tissue occurredconcomitantly with incorporation of labelled 15N-nitrogen. Thenitrogen of the vegetative shoot tissue was substantially lesslabelled than the nitrogen entering the ears, indicating thattranslocation of recently absorbed nitrogen to ears occurs withminor prior exchange with the bulk nitrogen of shoots. In caseswhere the sink strength of the ears was weak, as in LN-culturedLaevigatum (due to high frequency of sterile flowers) and inCN-cultured Golf, nitrogen translocated from roots appearedto be incorporated into the vegetative shoot tissue. There werealso indications that a fraction of the remobilized nitrogenwas actually lost from the plants in these cases. It is concludedthat the root remains efficient in translocation of nitrogento the aerial parts throughout ontogeny and that nitrogen takenup during grain–filling is preferentially directly translocatedto the developing grains. The further translocation of nitrogenreceived by vegetative shoot parts to ears appears mainly relatedto the potential of the ear to accumulate nitrogen. Nitrogenabsorbed/remobilized in excess of the sink strength of the earsis either invested in continued shoot growth, or is irreversiblylost from the plants. Nitrate assimilation was examined in two cultivars (Banner Winterand Herz Freya) of Vicia faba L. supplied with a range of nitrateconcentrations. The distribution between root and shoot wasassessed. The cultivars showed responses to increased applied nitrateconcentration. Total plant dry weight and carbon content remainedconstant while shoot: root dry weight ratio, total plant nitrogen,total plant leaf area and specific leaf area (SLA) all increased.The proportion of total plant nitrate and nitrate reductase(NR) activity found in the shoot of both cultivars increasedwith applied nitrate concentrations as did NO3: Kjeldahl-Nratios of xylem sap. The cultivars differed in that a greaterproportion of total plant NR activity occurred in the shootof cv. Herz Freya at all applied nitrate concentrations, andits xylem sap NO3: Kjeldahl-N ratio and SLA were consistentlygreater. It is concluded that the distribution of nitrate assimilationbetween root and shoot of V. faba varies both with cultivarand with external nitrate concentration. A comparison of plant hydraulic conductances in wheat and lupins   总被引:3,自引:1,他引:2  
Previous studies have shown similar water use for lupins (Lupinusangustifolius L.) and wheat (Triticum aestivum L.), despitea considerably smaller root system in lupins. A field studyand an experiment under controlled conditions using pressure-fluxrelationships were conducted to examine whether higher hydraulicconductances were responsible for the greater water uptake perunit root length in lupins. In the field experiment, the fluxof water and differences in water potential through the soil-plantsystem were measured for both species and used to calculatethe hydraulic conductance through the plant and through theroot and shoot. The hydraulic conductance for the whole plantwas 3–5 times greater in lupins than in wheat. This relativedifference between the species was similar when plant hydraulicconductance was expressed per unit of root length. This occurreddespite the difference in midday water potential between soiland leaves, being consistently greater in wheat (–1.0MPa) than in lupins (–0.7 MPa). When the total plant conductancewas separated into its components, the combined soil and rootconductance and the shoot conductance were 2 and 6 times greater,respectively, in lupins than in wheat. In the experiment undercontrolled conditions, hydraulic conductance for the entireroot system was determined using a pressure chamber. The specificroot hydraulic conductances were 4 times greater in lupins thanin wheat. The results from both field and controlled conditionsexperiments suggest that the greater water uptake per unit rootlength in lupins compared to wheat results from appreciablylarger root and shoot hydraulic conductances. Growth performances of Crotalaria juncea L. and C. sericea Retz.have been compared at two controlled temperatures, 16–20°C, and 28–32 °C, with respect to increase ind. wt and leaf area, relative growth rate, leaf area ratio,specific leaf area, leaf weight ratio, net assimilation rate,the ratio of mean relative growth rate to mean relative rateof leaf area increase () and shoot/root ratios. Both the speciesgrew better at the higher temperature; however the relativegrowth rate was more affected by temperature in C. sericea thanin C. juncea. Further, it was observed to be more dependenton net assimilation rate than on the leaf area ratio. The female gametophyte of Ephedra foliata was used as an explantfor the production of haploids as it is composed of haploidcells, all of the same genotype. The regeneration of roots wasdependent upon the presence of NAA, while BAP had a modifyingeffect. At lower concentrations (0.05 parts 10–6 and 3.5parts 10–6) BAP enhanced the root promotion of NAA (0.05–4.0parts 10–6). At higher concentrations of BAP (1–6parts 10–6), roots and shoot buds were formed. Kinetinat 4.0 parts 10–6 with 0.5 parts 10–6 2, 4-D wasoptimal for shoot bud production in explants at the archegonialstage and 2, 4-D at 2.0 parts 10–6 with 0.5 parts 10–6kinetin was optimal for root formation. Cells of the callusand root tip had the haploid number of chromosomes, n = 7. Meristemoidswere located on the surface or embedded in the callus tissue.The deep seated meristemoids organized only root primordia,but the peripheral ones gave rise to root as well as shoot budprimordia. Upland grasslands are a major component of natural vegetationwithin the UK. Such grasslands support slow growing relativelystable plant communities. The response of native montane grassspecies to elevated atmospheric carbon dioxide concentrationshas received little attention to date. Of such studies, mosthave only focused on short-term (days to weeks) responses, oftenunder favourable controlled environment conditions. In thisstudy Agrostis caplllaris L.5, Festuca vivipara L. and Poa alpinaL. were grown under semi-natural conditions in outdoor open-topchambers at either ambient (340µmol mol–1) or elevated(680µmol mol–1) concentrations of atmospheric carbondioxide (CO2 for periods from 79 to 189 d, with a nutrient availabilitysimilar to that of montane Agrostis-Fescue grassland in Snowdonia,N. Wales. Whole plant dry weight was increased for A. capillarisand P. alpina, but decreased for F. vivipara, at elevated CO2.Major components of relative growth rate (RGR) contributingto this change at elevated CO2 were transient changes in specificleaf area (SLA) and leaf area ratio (LAR). Despite changes ingrowth rate at 680 µmol mol–1 CO2, partitioningof dry weight between shoot and root in plants of A. capillarisand P. alpina was unaltered. There was a significant decreasein shoot relative to root growth at elevated CO2 in F. viviparawhich also showed marked discoloration of the leaves and increasedsenescence of the foliage. The freezing tolerance of many plants, such as pea (Pisum sativum),is increased by exposure to low temperature or abscisic acidtreatment, although the physiological basis of this phenomenonis poorly understood. The freezing tolerance of pea shoot tips,root tips, and epicotyl tissue was tested after cold acclimationat 2C, dehydration/rehydration, applications of 10–4M abscisic acid (ABA), and deacclimation at 25C. Tests wereconducted using the cultivar ‘Alaska’, an ABA-deficientmutant ‘wil’, and its ‘wildtype’. Freezinginjury was determined graphically as the temperature that caused50% injury (T50) from electrical conductivity. Endogenous ABAwas measured using an indirect enzyme-linked immunosorbant assay,and novel proteins were detected using 2-dimensional polyacrylamidegel electrophoresis. The maximum decrease in T50 for root tissuewas 1C for all genotypes, regardless of treatment. For ‘Alaska’shoot tips and epicotyl tissue, exogenous ABA increased thefreezing tolerance by –1.5 to –4.0C, while coldtreatment increased the freezing tolerance by –7.5 to–14.8C. Cold treatment increased the freezing toleranceof shoot tips by –9 and –15C for ‘wil’and ‘wild-type’, respectively. Cold acclimationincreased endogenous ABA concentrations in ‘Alaska’shoot tips and epicotyls 3- to 4-fold. Immunogold labeling increasednoticeably in the nucleus and cytoplasm of the epicotyl after7 d at 2C and was greatest after 30 d at the time of maximumfreezing tolerance and soluble ABA concentration. Maize (Zea mays L., hybrid Cargill 147) seedlings, grown inaseptic conditions, were inoculated with three strains of Azospirillumlipoferum (Al op 33, Al iaa 320, and ATCC 29708) or culturedin different concentrations of indol-3-acetic acid (IAA) orgibberellin A3 (GA3). After 48 h, root length, root surfacearea, root dry weight, and shoot dry weight were measured inall treatments. Gibberellin content was evaluated in selectedroots of control and Azospirillum inoculated seedlings by gaschromatography-mass spectrometry-selected ion monitoring withthe use of deuterated gibberellins as internal standards. Thethree strains of A. lipoferum, IAA (2 ng ml–1), and GA3(40 to 400 pg ml–1) significantly enhanced root growth.Improvement of root hair growth and density was obtained mainlywith A. lipoferum strain Al op 33 and GA3 40 pg ml–1.GA3 was identified by gas chromatography-mass spectrometry (byboth, full scan and selected ion monitoring) in a free acidfraction from roots of the seedlings inoculated with A. lipoferum.In the roots of the non inoculated seedlings GA3 was found afterhydrolysis of a fraction expected to contain glucosyl conjugates. Root growth in chickpea (Cicer arietinum) has been studied fromthe early vegetative phase to the reproductive stage in orderto elucidate its growth and maintenance respiration and to quantifythe translocation of assimilates from shoot to root. A carbonbalance has been drawn for this purpose using the growth andrespiration data. The increase in the sieve tube cross-sectionalarea was also followed simultaneously. Plants growing in a nutrient culture medium were studied todetermine the relative growth rate (RGR) 5–60 d aftergermination. RGR declined from 113 to 41 mg d–1 g–1during the measurement period. Simultaneous with the RGR analysis,respiration rate was also measured using an oxygen electrode.The respiration rate declined as the plants aged and a drasticreduction was recorded following anthesis. The relationshipbetween RGR and respiration rate was used to extrapolate themaintenance respiration (m) and growth respiration (1/YEG).The respiration quotient (r.q.) of the roots was 1.2 and theQ10 in the range 20–25 °C was 2·2. A carbon balance for the roots was constructed by subtractingthe carbon lost during respiration from that gained during growth.The roots were found to respire no less than 80% of the carbontranslocated. The increase in the cross-sectional area composed of sieve tubeswas measured near the root-shoot junction as the plants grew.Chickpea has storied sieve plates which simplifies these measurements.Their cross-sectional area increased during growth mainly becauseof an increase in sieve tube number. The diameter of individualsieve tubes remained constant. Specific mass transfer (SMT) values for seive tubes into theroots have been computed during various stages of growth. SMTvalues were relatively constant before anthesis (approx. 6·5g h–1 cm–2), but decreased following anthesis. Wedid not evaluate possible retranslocation from roots: any suchretranslocation would have the effect of increasing our SMTvalues. The transport of nitrogen from the roots and nodules of chickpea(Cicer anetinum L.), lentil (Lens culinaris Medic), faba bean(Vicia faba L.) and pea (Pisum sativum L.) was examined in glasshouse-grownplants supplied either with nitrate-free nutrients or with nutrientssupplemented with 1,2,4 or 8 mol m-3153N-nitrate. A sixth treatmentcomprised uninoculated plants supplied with 8–0 mol m-31513N-nitrate. For each species, more than 75% of the nitrogenwas exported from the nodules as the amides, asparagine andglutamine. In fully symbiotic plants, the amides also dominatednitrogen transport to the shoot When N2 fixation activity wasdecreased by the addition of nitrate to the rooting medium,the N-composition of xylem exudate and stem solutes changedconsiderably. The relative concentrations of asparagine tendedto increase in the xylem whilst those of glutamine were reduced;the levels of nitrate increased in both xylem exudate and thesoluble nitrogen pool of the stem with a rise in nitrate supply.The changes in relative nitrate contents reflected generallythe contributions of root and shoot to overall nitrate reductaseactivity at the different levels of nitrate used. The relationshipsbetween the relative contents of xylary or stem nitrate andamino nitrogen and the plants' reliance on N2 fixation (determinedby the 15N isotope dilution procedure) were examined. Data suggestthat compositional relationships based on nitrate may be reasonableindicators of symbiotic dependence for all species under studyexcept faba bean when greater than 25% of plant nitrogen wasderived from N2 fixation. To manipulate the ratio of shoot activity to rootactivity, the plants were grown with their shoot base includingthe apical meristem either above (i.e. at 20 °C) or withinthe nutrient solution (i.e. at 20, 16 or 12 °C). In wheat, the ratio of shoot:root dry matter partitioning decreasedat low RZT, whereas the opposite was true for maize. In bothspecies, dry matter partitioning to the shoot was one-sidedlyincreased when the shoot base temperature, and thus shoot activity,were increased at low RZT. The concentrations of non-structuralcarbohydrates (NSC) in the shoots and roots were higher at lowin comparison to high RZT in both species, irrespective of theshoot base temperature. The concentrations of nitrogen (N) inthe shoot and root fresh matter also increased at low RZT withthe exception of maize grown at 12 °C RZT and 20 °Cshoot base temperature. The ratio of NSC:N was increased inboth species at low RZT. However this ratio was negatively correlatedwith the ratio of shoot:root dry matter partitioning in wheat,but positively correlated in maize. It is suggested that dry matter partitioning between shoot androots at low RZT is not causally related to the internal nitrogenor carbohydrate status of the plants. Potato plants (Solanum tuberosum L.) were grown at differentair and soil temperatures to determine the effects of high-temperaturestress on root, tuber, and shoot growth. Cooling the soil (17–27C) at high air temperatures (30–40 C) relieved noneof the visible symptoms of heat stress on shoot growth; norwas the degree of induction to tuberize in leaves increased,as reflected in tuberization of leaf-bud cuttings. Heating thesoil (27–35 C) at cool (17–27 C) air temperatureshad no apparent detrimental effect on shoot growth or inductionof leaves to tuberize. However, in each case hot soil largelyeliminated tuber development. In one experiment stolons grewup out of the hot soil and formed aerial tubers upon reachingthe cool air. When leaf-bud cuttings from induced plants wereused as a model system, high soil temperatures inhibited tuberdevelopment from the buried leaf buds, in the absence of anyroot growth. Apparently the induction of leaves to tuberizeis affected principally by air rather than soil temperature,but expression of the signal to tuberize can be blocked by highsoil temperature. From compartmental analysis of 32P elution measurements, concentrationsand fluxes of phosphate were estimated for root cortical cellsof intact Lolium perenne L. plants, when in complete nutrientsolution containing 0.1 mol m–3 phosphate with and without37 mmol m–3 Al during loading and elution. Failure ofthe data, when plotted as ct. min–1 remaining in the tissueas a function of time, to meet the criteria for first orderkinetics led to a discussion of the relative importance of transportto the shoot, assimilation, and complexation in the vacuole,in causing this discrepancy. It was concluded that complexationwas the most important factor. Transformation of the data tomeet the criteria for first order kinetics gave corrected valuesfor compartmental concentrations and fluxes of phosphate, andestimates of the size of the otherwise unresolved slowly exchangingcompartment within the vacuole equated with condensed phosphate.In the control this was 2.5 mol m–3 P but in Al treatmentsa much larger amount of phosphate was complexed with Al (6.5mol m–3 P). Phosphate transport to the shoot was unaffectedby Al. Instead, levels of Al, common in solution in upland soils,sequestered in the root vacuoles quantities of phosphate significantfor stressing the phosphate economy of nutrient-poor grassland.
