Twenty prepubertal Holstein heifers were utilized to assess plasma 13, 14-dihydro-15-keto-prostaglandin F(2)alpha (PGFM), serum progesterone (P(4)) and estradiol-17beta (E(2)) concentrations as well as the E(2):P(4) ratio during the onset of puberty in cattle. All animals were maintained as a group along with a sterile marker bull to assist in the detection of estrus. Upon detection of the first estrus (Day=O), daily blood samples were collected from a jugular vein until the heifers had completed 3 estrous cycles. The average body weight and age at first estrus were 247.6+/-4.8 kg and 304.0+/-7.5 days, respectively. Frequency of abnormal length estrous cycles was greater (P<0.02) during the first (40%) and second (35%) cycles than during the third estrous cycle (0%). All heifers had normal cycle lengths (18 to 24 days) by the third estrous cycle. Serum P(4) was greater during the third cycle (P<0.05) from Day 10 to Day 4 before the next estrus compared with the same period of the first estrous cycle. Serum E(2) did not peak until the day of estrus in the first cycle, whereas E(2) reached a maximal level 2 days before estrus in the third estrous cycle. Serum E(2) was higher (P<0.0001) 2 days before estrus in the third cycle than in the first estrous cycle. Plasma PGFM reached maximum concentrations 3 days before estrus in the third cycle compared with 1 day before estrus at the end of first estrous cycle. As estrus approached during the third cycle, PGFM rose 1 day before E(2) rose and P(4) declined, while the rise in PGFM and E(2) occurred simultaneously, with P(4) declining at the end of the first estrous cycle. During diestrus, the E(2):P(4) ratio was lower (P<0.07) in the third cycle than in the first, but it was higher (P<0.04) at estrus and 1 day before in the third estrous cycle. These data reveal a high incidence of abnormal length estrous cycles during the first two estrous cycles of the peripubertal period, and demonstrate anomalies in uterine and ovarian endocrine activity during the peripubertal period in cattle.  相似文献

To assess endocrine and morphological responses of ovaries to total weaning at parturition, 6 Zebu (Bos indicus) cows 5 years or older were investigated. Following parturition, blood samples were collected daily during the first month and twice weekly thereafter until day 60 to determine concentrations of progesterone (P4) and prostaglandin F2α metabolite. It took between 25 to 32 days to complete uterine involution.The prostaglandin metabolite remained elevated for a mean period of 14.2 days (range, 4-21) postpartum. Five of the animals resumed cyclicity with a short estrous cycle starting between days 7 to 34 and lasting between 7 and 14 days. No estrous behavior was recorded prior to the short estrous cycles, but subsequent normal-length estrous cycles were all preceded by signs of estrus. In the 1 animal that resumed cyclicity with an estrous cycle of normal length on day 37 (length 20 days), the cycle was preceded by estrous behavior. Progesterone concentrations reached a mean maximum of 4.8 nmol liter−1 during the short estrous cycles, and prostaglandin metabolite concentrations peaked while P4 concentrations were decreasing. P4 concentrations reached a mean maximum of 12.2 nmol liter−1 during the estrous cycles of normal length. The interval from parturition to the first estrous cycle of normal length varied between 16 and 48 days, and the length of the cycle was 18 to 22 days. Starting 2 days postpartum, ovaries from 5 of the cows were scanned by ultrasonography every second day until day 30 postpartum. Medium-sized follicles were detected between days 4 to 7 postpartum in 4 of the scanned cows that later had short estrous cycles. The time between parturition and the appearance of the first dominant follicle was 7.6 days (range 6-10 days). The interval between parturition and the appearance of the first ovulatory-sized follicle was 10.2 days (range 8-13 days). In 3 of the scanned cows this ovulatory-sized follicle ovulated. We conclude that cyclic ovarian activity in Zebu cows can start early in the postpartum period in the absence of offspring, and that short luteal phases, not preceded by estrous behavior, may play an important role in establishing normal postpartum ovarian activity.  相似文献

Studies were conducted to determine: (1) if fecal hormone metabolite concentrations correlated with serum estrogen and progesterone concentrations, follicular activity and reproductive behavior in the black rhinoceros (Diceros bicornis) and (2) if threshold values of respective fecal metabolite concentrations correlated with pregnancy. Blood and fecal samples were collected, in conjunction with transrectal ultrasound and behavior observations, for an 18-month period from one black rhinoceros female. Subsequently, serial fecal samples were collected from 13 females in 10 zoos. Quantitative analysis of serum progesterone (P4) and estradiol (E2) was performed by radioimmunoassay (RIA): analysis of fecal estrogen metabolites (E) and fecal progesterone metabolites (P) were performed by enzyme immunoassay (EIA). Serum P2 concentrations identified two luteal phase patterns and two nadirs which corresponded with behavioral estrus. Fecal E patterns indicated a sharp peak which corresponded with breeding. concentrations of fecal P illustrated identifiable nadirs and several peaks which corresponded to serum P4 nadirs and luteal phases. Serum P4 concentrations were not different between the luteal phase and pregnancy. Fecal P concentrations started to rise above luteal phase concentrations approximately 150 days postbreeding and remained elevated until immediately before parturition. Serum E2 and fecal E concentrations rose and subsequently declined after parturition. In the fecal samples from seven pregnant females, fecal P concentrations were similarly elevated compared to six nonpregnant females. Results indicated that fecal steroid metabolites accurately reflected serum steroid hormone concentrations and that the measurement of P and E concentrations permitted the characterization of the estrous cycle, the diagnosis of pregnancy, and the onset of parturition. Serum progesterone and urinary total estrogen concentrations were determined weekly to bi-weekly in 2 female Asian elephants for 96 weeks. The mean estrous interval was approximately 16 weeks in the nonpregnant animal. A total of 5 cycles were observed in the 96 week study period. The serum progesterone concentration ranged from 150 pg/ml to greater than 350 pg/ml during the luteal phase of the estrous cycle. The serum progesterone was elevated for 8–12 week weeks of the 16 week estrous cycle. The urinary total estrogen concentration ranged from less than 10 to greater than 300 pg/μg creatinine. The second animal was pregnant at the beginning of the study period. The serum progesterone concentration was elevated (> 100 pg/ml) in the pregnant animal until parturition. The urinary total estrogens increased from approximately 50 pg/μg creatinine to greater than 400 pg/μg creatinine during the first year of pregnancy and remained elevated until parturition. Estrous cycling had not resumed by 3 months post partum.  相似文献

Serum samples were collected 1–3 times weekly from two Baird's tapirs (Tapirus bairdii) for 6 months in 1987–1988, and for more than 3 consecutive years beginning in 1989 to characterize hormone patterns during the estrous cycle and pregnancy. Based on serum progesterone concentrations, mean (±SEM) duration of the estrous cycle (n = 20) was 30.8 ± 2.6 days (range, 25–38 days) with a luteal phase length of 18.1 ± 0.4 days (range, 15–20 days). Mean peak serum progesterone concentrations during the luteal phase were 1.35 ± 0.16 ng/ml, and nadir concentrations were 0.19 ± 0.03 ng/ml during the interluteal period. Distinct surges of estradiol preceded luteal phase progesterone increases in most (14/20) cycles. Gestation length was 392 ± 4 days for three complete pregnancies. Mean serum progesterone concentrations increased throughout gestation and were 1.83 ± 0.13, 2.73 ± 0.13, and 4.30 ± 0.16 ng/ml during early, mid- and late gestation, respectively. Serum estradiol concentrations began to rise during mid-gestation, increasing dramatically during the last week of pregnancy. Patterns of serum estriol and estrone secretion during pregnancy were similar to that observed for estradiol. In contrast to progesterone and estrogens, serum cortisol concentrations were unchanged during pregnancy or parturition. Females resumed cycling 16.2 ± 2.0 days after parturition (n = 4) and, on two occasions, females became pregnant during the first postpartum estrus. These data suggest that the tapir cycles at approximately monthly intervals and that increases in serum progesterone are indicative of luteal activity. The interluteal period is relatively long, comprising approximately 40% of the estrous cycle. During gestation, progesterone concentrations are increased above luteal phase levels, and there is evidence of increased estrogen production during late gestation. The absence of increased cortisol secretion at the end of gestation suggests that this steroid does not play a major role in initiating parturition in this species. © 1994 Wiley-Liss, Inc.  相似文献

It was hypothesized that prolonged elevation in 17beta-estradiol (E(2)) preceding ovulation as a result of a persistent ovarian follicle would have a detrimental effect on pregnancy rate after Day 7 (behavioral estrus = Day 0) of the estrous cycle. Cows were either treated with exogenous progesterone (P(4)) for 10 d or remained untreated (CON; n = 76). Cows were treated with 1 of 2 doses of P(4) from Day 6 to 16 which was intended to result in either elevated E(2) (EE(2); n = 76) or normal E(2) (NE(2); n = 76) concentration in the circulation. At the initiation of P(4) treatment, cows received prostaglandin F(2alpha) (PGF(2alpha)) to eliminate the endogenous source of P(4). On Day 16, the exogenous source of P(4) was removed from treated cows, while cows in the CON group received PGF(2alpha). A single embryo was transferred into each cow 7 days after observation of behavioral estrus. Blood samples were taken on alternating days during the treatment period to determine concentrations of P(4) and E(2). The pregnancy rate was determined by ultrasonographic examination 25 to 32 d after embryo transfer. There was a treatment-by-day interaction (P < 0.0001) on E(2) concentrations in the plasma during the 10-d treatment period. Cows in the EE(2) group had a higher concentration of E(2) by Day 8 (6.1 +/- 0.5 pg/ml) and this concentration remained elevated until PRID removal compared with that of cows in the NE(2) (2 +/- 0.2 pg/ml) and CON (2.0 +/- 0.3 pg/ml) groups, which had concentrations of E(2) similar to those at the initiation of treatment. Pregnancy rates after embryo transfer did not differ (P = 0.56; X(2) = 1.1) among cows in the EE(2) (30.7%), NE(2) (36.2%) and CON (32.9%) groups. Prolonged elevation of E(2) concentrations associated with the development of a persistent ovarian follicle preceding ovulation did not affect the pregnancy rate to embryo transfer after Day 7 of the estrous cycle in cows.  相似文献

Two trials were designed to test whether a single treatment with a microsphere formulation of progesterone (P) could simulate the luteal phase of the estrous cycle and lead to estrus and subsequent luteal development. The first experiment was to characterize the pattern of serum P concentrations and estrus in cows treated with a microsphere formulation (P + E) that contained 625 mg P and 50 mg estradiol (E). Four cows with palpable corpora lutea were treated with 25 mg prostaglandin F2 m. Each cow was given P + E (i.m.) 12 h later. Tail vein blood samples were taken on Days 1 and 2 following P + E treatment and then three times weekly for 24 days. Serum P increased from 0.8 +/- 0.1 ng/ml at P + E treatment to 4.7 +/- 0.6 ng/ml on Day 1, declined gradually to 4.1 +/- 0.3 ng/ml on Day 7 and then declined more rapidly to 0.6 +/- 0.1 ng/ml on Day 13. Treated cows showed estrus 16.25 +/- 0.7 days after P + E treatment. Thereafter, serum P increased beginning on Day 20 after P + E treatment, as expected following estrus. In Experiment 2, Angus and Simmental heifers (10.5-11.5 months of age) were administered i.m. either the vehicle (controls), E (50 mg), P (625 mg) or P + E (n = 13 per group). While treatment with E resulted in behavioral estrus (1-2 days after treatment) in each treated heifer, it did not (P > 0.5) initiate estrous cycles as indicated by subsequent increased serum P. In contrast, the P and P + E treatments increased (P < 0.05) the proportion (11/13) of heifers that showed estrus by 21 days after treatment followed by elevated serum P. We conclude that the microsphere formulation of P simulated the pattern of serum P concentrations during the luteal phase of the estrous cycle and initiated estrous cycles in peripubertal heifers with or without E.  相似文献

Direct radioimmunoassays (RIA) for urinary oestrone conjugates and pregnanediol-3 alpha-glucuronide (PdG) were used to study ovarian activity patterns and pregnancy in Eld's deer. In 2 does, urinary metabolite patterns were compared to temporal patterns of plasma LH, oestradiol-17 beta and progesterone. Preovulatory LH peaks occurred coincident with behavioural oestrus, and plasma progesterone secretion paralleled PdG excretion. Although plasma oestradiol-17 beta levels fluctuated between 5 and 10 pg/ml throughout the oestrous cycle, no preovulatory oestrogen surge was observed. Based on PdG excretion, non-conception oestrous cycles averaged 21.5 +/- 2.1 days (+/- s.e.m., n = 65); however, 2 of 13 does exhibited prolonged oestrous cycles (30.1 +/- 4.4 days; range 14-62 days, n = 14) characterized by sustained PdG excretion. Excluding these 2 females, the mean oestrous cycle was 18.5 +/- 0.3 days (range 14-23 days, n = 51). Behavioural oestrus (12-24 h duration) was observed in 42 of 65 cycles (64.6%), and always corresponded with intercyclic troughs in PdG excretion (2-5 days duration). Mean gestation duration (n = 10) was 33.5 +/- 0.4 weeks. PdG concentrations increased (P less than 0.05) by Week -32 (3rd week of gestation), plateaued between Weeks -31 and -25, increased (P less than 0.05) markedly by Week -22 and then rose steadily until parturition, declining (P less than 0.05) rapidly thereafter. Mean excretion of oestrone conjugates remained low until Week -30, increased (P less than 0.05) steadily to Week -24 (P less than 0.05) and then returned to baseline by Week -17. Increased (P less than 0.05) oestrone conjugates concentrations were detected again by Week -4 followed by a rapid increase to peak pregnancy levels by Week -1, declining (P less than 0.05) precipitously after parturition. The results confirm that the Eld's deer is seasonally polyoestrous with onset (January-March) and cessation (August-October) of regular, cyclic ovarian activity coinciding with increasing and decreasing daylengths, respectively. Urinary PdG excretion accurately reflects cyclic ovarian activity and markedly elevated concentrations of this metabolite provide an accurate index of pregnancy. The simultaneous monitoring of oestrone conjugates appears useful for estimating the stage of pregnancy and predicting parturition onset.  相似文献

Serum luteinizing hormone (LH), estrone, estradiol-17β, and progesterone were measured during the estrous cycle, pregnancy and parturition in seven adult pregnant Labrador bitches and during the estrous cycle and one gestation length equivalent in six adult pseudopregnant bitches. Although the duration of proestrus was similar in both groups, the duration of estrus was longer in the bitches that subsequently became pregnant. Mean serum LH concentrations were similar in both groups during most of the study. However, during Weeks 6 to 9 after the preovulatory LH peak, serum LH concentration was higher in both pseudopregnant and pregnant groups of animals and declined to basal levels thereafter. Mean serum estrone concentrations in the pregnant animals were higher than those of pseudopregnant animals and remained elevated throughout gestation, followed by a decline at whelping. Serum estradiol-17β levels were higher during the 4 wkimmediately following ovulation in the pseudopregnant group compared with those observed in pregnant animals. Serum progesterone concentrations generally remained higher during pseudopregnancy compared with those of the pregnant animals during gestation.In conclusion, a major difference between pregnant and pseudopregnant bitches is a pregnancy-specific elevation in estrone levels. The placenta may be a likely source of estrone during pregnancy.  相似文献

Serum inhibin levels were measured by heterologous RIA during pregnancy, lactation, and the post-weaning estrous cycle in the rat and correlated with changes in serum FSH and LH and prolactin. Blood was serially collected by cardiac puncture under light ether anesthesia from adult Sprague-Dawley rats on alternate days throughout the experimental period. For the first 8 days of pregnancy, immunoreactive inhibin levels remained high, then gradually decreased to reach a nadir at Day 16, and subsequently rose steeply until parturition. The pattern of serum immunoreactive inhibin levels during early pregnancy does not support a corpus luteum source and the dramatic rise from Day 16 to Day 22 correlates with the recommencement of follicular development in the ovary. Inhibin levels decreased rapidly on the day after birth and were suppressed until Day 8 of lactation, slowly increasing thereafter to reach a plateau from Day 14 until weaning (Day 22.5 of lactation). These changes in inhibin levels positively correlated with LH and FSH and negatively with prolactin, and are consistent with an ovarian source for inhibin associated with the recommencement of follicular development resulting from the diminution of the suckling stimulus. Immediately after weaning, serum immunoreactive inhibin levels showed a 4-day cyclic pattern corresponding to the estrous cycle identified by vaginal smear. Inhibin levels peaked on the day of proestrus, reached a nadir on the day of estrus, and rose slowly during metestrus and diestrus to a new peak at proestrus. Serum FSH levels showed an inverse correlation to inhibin levels consistent with a feedback relationship with inhibin.  相似文献

In the present study, pregnancy and the estrous cycle were monitored in captive brown brocket deer (Mazama gouazoubira) by measuring fecal progestagens with a commercial enzyme immunoassay (EIA), along with behavioral data. Fecal samples were collected twice a week during pregnancy and daily during the estrous cycle and post-partum period. It was possible to distinguish between inter-luteal and luteal phases of the estrous cycle. Behavioral estrus corresponded with low concentrations of fecal progestagens. Samples from two consecutive cycles were available from five hinds, and the mean estrous cycle (n=10) was 26.9+/-1.7 d (mean+/-S.E.M.). However, when two extreme cycles (34 and 37 d) were deleted, the mean estrous cycle was 24.7+/-1.2 d. Three animals became pregnant (gestation ranged from 208 to 215 d). After fertile breeding, progestagen concentration in these hinds remained among luteal phase concentrations throughout pregnancy, with the exception of a few peaks. Within 4 d post-partum, two hinds reached interluteal phase values, while one hind maintained luteal concentrations for at least 1 week.  相似文献

Urinary total polyamines; putrescine, spermidine and cadaverine, levels were determined in the estrous cycle and during the pregnancy in female Wistar rats. Urinary total polyamines level in 12 estrous cycles in 4 female rats revealed the definite cyclic changes, showing high levels at estrous and low levels at diestrous stage. After the ovariectomy those cyclic changes disappeared. Urinary total polyamines levels were constantly low during the first half of the pregnancy of 8 female rats, whereas the levels increase abruptly from 10 days before parturition. After fetusectomy at 15th and 17th days of the pregnancy, those levels decreased gradually. These data suggested that the urinary total polyamines levels were closely related with the pituitary-ovarian function and the growth of fetus of rats.  相似文献

Effects of prostaglandin F(2alpha) (PGF(2alpha)), administered during the mid-luteal phase of the estrous cycle, were examined in ewes exhibiting estrous cycles classified as short (< or =16.5 days, short-cycle ewes, n = 10) or long (> or =18 days, long-cycle ewes, n = 9) based on the durations of two estrous cycles (cycles -2 and -1) before treatment. The ewes received (i.m.) 20mg of PGF(2alpha) on day 10 of the third estrous cycle (cycle 0) followed, 36 h later, by 25 microg of gonadotropin releasing hormone (GnRH) to time the events of ovulation. Duration of subsequent estrous cycles +1 and +2 were recorded, and then the ewes were treated with the same combination of PGF(2alpha) and GnRH beginning on day 10 of estrous cycle +3. Ovaries were recovered 6h after GnRH administration to assess development of pre-ovulatory follicles. The proportion of ewes that exhibited estrus after PGF(2alpha) and GnRH treatment on cycle 0 was not different (P > 0.05) between short- and long-cycle ewes. Onset of estrus occurred sooner (P < 0.05) after PGF(2alpha) injection in short-cycle ewes than in long-cycle ewes (1.9 +/- 0.1 days and 2.3 +/- 0.1 days, duration of cycle 0 was 11.9 and 12.3 days, respectively). Duration of estrous cycle +1 was 1.2 days longer (P < 0.01) than cycle -1 in short-cycle ewes. However, duration of estrous cycle +1 did not change (P > 0.05) after PGF(2alpha) and GnRH administration in ewes having long cycles. Pre-ovulatory follicles did not differ (P > 0.05) in numbers, diameter, layers of granulosa cells nor concentrations of progesterone and estradiol-17beta in follicular fluid between short- and long-cycle ewes after PGF(2alpha) and GnRH treatment. In conclusion, ewes having short or long estrous cycles responded differently to PGF(2alpha) and GnRH treatment with respect to the interval to onset of estrus and duration of the subsequent estrous cycle.  相似文献

Oviducts from 22 crossbred heifers were examined for the presence of nuclear estrogen (ERalpha) and progesterone (PR) receptors at different phases (estrus, metaestrus and diestrus) of naturally occurring estrous cycles and estrous cycles during which superovulation was induced. Receptors were detected by immunohistochemistry in the epithelial cells, connective tissue and muscular layer of oviductal infundibulum, ampulla, ampullary/isthmic transition and isthmus. Epithelial ERalpha was found along the entire oviduct regardless of the cycle phase and of the circulating concentrations of 17beta-estradiol (E(2)) and progesterone (P(4)). Epithelial PR was found mainly at the ampullary-isthmic transition and isthmus and more intensely at the estrus phase but their amount was not correlated with P(4) concentrations. ERalpha in the connective tissue was more abundant at the infundibulum and ampulla, regardless of the phase of the estrous cycle and of E(2) and P(4) circulating concentrations. PR in the connective tissue was found mostly at the ampulla, regardless of the estrous cycle phase but no correlations were found between amount and P(4) concentrations. ERalpha staining intensity in the muscular layer was similar at all phases of the estrous cycle and at all anatomical segments of the oviducts. However, PR staining was more intense at the isthmus during the metaestrus phase and it was negatively correlated with P(4) concentrations. In general, data from the present research suggest that P(4) exerts an inhibitory role upon ERalpha and PR. Also, no differences were found between animals subjected or not to superovulation.  相似文献

The endocrinology was studied throughout pregnancy in reindeer (Rangifer tarandus tarandus) located in Oulu, Finland (65 degrees N, 25 degrees E) with 13 captive, semi domestic adult females. Blood samples were analyzed for plasma progesterone (P4), estradiol (E2) and estrone sulphate (E1SO4), 15-ketodihydro-PGF2alpha (PG-metabolite) and pregnancy associated glycoproteins (PAG). The mean plasma P4 concentration peaked twice during gestation: at around 24 and three weeks prior to calving. In pregnant females the plasma PAG concentration increased over basal concentrations 21-30 days after the estimated day of conception and peaked at the time of calving. The concentrations of E2 and E1SO4 remained low until 60 days before calving when a rapid increase was found for both hormones. The mean plasma concentration of PG-metabolite increased throughout pregnancy to a maximum at parturition. The estimated mean (range) gestation length was 216 (212-220) days. Judged from measures on reproductive organs collected from 86 free-ranging, semi-domestic female reindeer of unknown age presented for slaughter at Roros, Norway (63 degrees N, 11 degrees E) in the second week of December 1999, it was concluded that the breeding season lasted from early September until the end of November. The results also showed that plasma PAG concentration could provide a tool for detection of pregnancy in reindeer.  相似文献

Thirty-six crossbred gilts were maintained in outside dirt lots for 2 or more estrous periods. Eighteen of the gilts were moved to another outside dirt lot (non-confined) and the other eighteen were moved to a single pen in a confinement finishing building (confined). A blood sample was collected on the day of movement and every 7 days thereafter for 12 weeks and quantitated for progesterone to verify the occurrence of each ovulation. Any estrous cycle shorter than 18 days or longer than 23 days was classified as abnormal. The duration of estrous cycles of non-confined gilts were not different before, during or after movement, whereas the duration of estrous cycles were greater (P=.005) during movement than before movement in the confined gilts. The duration of estrous cycles of confined gilts were greater (P=.020) than those of non-confined gilts during movement. The percentage of abnormal estrous cycles were greater during movement than before (P=.007) or after (P=.052) movement for the confined gilts, whereas the percentage age of abnormal estrous cycles were greater (P=.050) during movement than before movement in non-confined gilts. Therefore, the greatest percentage of abnormal estrous cycles occurred during movement in both groups. A single incident of silent estrus followed by normal estrous cycles was observed. In addition, an estrus occurred while plasma progesterone concentration was elevated (51 ng/ml) and was followed by normal estrous cycles.  相似文献

Changes in plasma progesterone levels during the estrous cycle and pregnancy were determined in group-housed IVCS strain mice showing regular 4-day cycles in the absence of a male. The pattern of progesterone secretion during the estrous cycle consisted of a large peak and a small one, the former during the late afternoon of proestrus and the latter on the afternoon of metestrus. From Day 0 to Day 2 of pregnancy, a sharp rise was observed, and then this level reached a plateau. A marked increase in progesterone was shown from Day 13 until the day before parturition.  相似文献

Plasma concentrations of progesterone (P4), estradiol-17β (E2), estrone (E1) and estrone sulfate (E1S) were measured during gestation in eight guanacos kept in captivity. Gestational length was 346.1 ± 9.8 days. P4 plasma concentrations increased after ovulation and remained elevated until parturition. However, during the last 4 weeks of gestation, a gradual decrease from 4.17 × 1.17±1 nmol/L to 2.02 × 1.95±1 nmol/L on day 5 before parturition was observed, followed by a more abrupt final decline to baseline concentrations which were reached on the day after parturition. Mean E2 plasma concentrations started to increase during the eighth month of gestation, and were significantly elevated up to maximum concentrations of 484.7 × 1.21±1 pmol/L during the last 2 months of pregnancy. Concentrations returned to baseline during the last 2 days of gestation. An increase of E1S concentrations (p < 0.01) was observed in the eleventh month of gestation. Mean E1S concentrations remained rather constant during the last 3 weeks of gestation between 4 to 8 nmol/L until parturition, when a steep precipitous decline was observed. E1 concentrations were slightly elevated during the last 4 weeks of gestation, however, maximum concentrations did not exceed 1.5 nmol/L. The results show distinct species specific features of gestational steroid hormone profiles in the guanaco in comparison to domestic South American camelids, such as a more pronounced gradual prepartal decrease of P4 concentrations prior to the final decline to baseline, and clearly lesser E1S concentrations during the last 4 weeks of gestation, which lack a continuous prepartal increase.  相似文献

The objectives of this experiment were to determine if subnormal levels of progesterone (P4) indicative of luteal insufficiency influence (1) pulsatile release of luteinizing hormone (LH), (2) the interval to the preovulatory surge of LH after removal of P4, and (3) the secretion of P4 during the estrous cycle subsequent to administration of subnormal levels of P4. On Day 5 (Day = 0 day of estrus) of the estrous cycle, cows received P4-releasing intravaginal devices (PRID) to produce normal (2 PRIDs; n = 7) or subnormal (0.5 PRID; n = 6) concentrations of P4. Five cows served as controls. On Day 10, serial blood samples were collected from all cows. Collection of blood samples was again initiated on Day 17 in cows receiving PRIDs. The PRIDs were removed and blood collection continued for 78 h. Daily blood samples were collected from all animals for 42 days subsequent to estrus (estrous cycles 1 and 2, respectively). During estrous cycle 1, mean concentration of P4 was lower (p less than 0.05) and frequency of pulses of LH was higher (p less than 0.05) in cows receiving subnormal P4 than in cows receiving normal P4 and control cows. Plasma concentrations of estradiol (E2) were higher (p less than 0.05) on Days 9-16 of estrous cycle 1 in cows receiving subnormal P4 than in cows receiving normal P4 or in control cows. Mature female guinea pigs that had been observed to undergo three consecutive periods of estrus at approximately 16-day intervals were immunized with either 100 μg gonadotropin releasing hormone (GnRH) conjugated to 100 μg bovine serum albumin (BSA) or 100 μg BSA alone during diestrus (day 5–10) of the fourth cycle. Booster immunizations were administered 32 days after the first injection. Animals were bled by cardiac puncture at the time of first injection and at 16, 32, 48 and 64 days. Animals were necropsied at 64 days after first treatment.Daily observation indicated that vaginal manifestation of estrus was not apparent after a period equal to one estrous cycle in seven of ten GnRH immunized guinea pigs and after two cycles in the remaining three GnRH immunized guinea pigs. Estrous cycles persisted in BSA treated females throughout the experiment.Serum luteinizing hormone (LH) declined significantly by 32 days after the first immunization against GnRH and remained lower than both pretreatment values and levels in control animals at the same bleeding times throughout the experiment. Serum progesterone levels were significantly lower in the GnRH immunized group than in the control group at 48 and 64 days.At necropsy the weight of the ovaries of GnRH immunized guinea pigs was significantly lower than that of controls. Corpora lutea and antral follicles were present in both GnRH treated and control females. The presence of serum progesterone levels and of antral follicles in the GnRH immunized females suggests that a low level of gonadotropic support may have persisted to 64 days after initiation of treatment.Results indicate that immunization against GnRH can reduce LH and progesterone levels and induce cessation of estrous cycles in the guinea pig.  相似文献
