The aim of this study was to investigate the microbial community thriving at two shallow hydrothermal vents off Panarea Island (Italy). Physico-chemical characteristics of thermal waters were examined in order to establish the effect of the vents on biodiversity of both Bacteria and Archaea. Water and adjacent sediment samples were collected at different times from two vents, characterised by different depth and temperature, and analysed to evaluate total microbial abundances, sulphur-oxidising and thermophilic aerobic bacteria. Total microbial abundances were on average of the order of 105 cells ml−1, expressed as picoplanktonic size fraction. Picophytoplanktonic cells accounted for 0.77–3.83% of the total picoplanktonic cells. The contribution of bacterial and archaeal taxa to prokaryotic community diversity was investigated by PCR–DGGE fingerprinting method. The number of bands derived from bacterial DNA was highest in the DGGE profiles of water sample from the warmest and deepest site (site 2). In contrast, archaeal richness was highest in the water of the coldest and shallowest site (site 1). Sulphur-oxidising bacteria were detected by both culture-dependent and -independent methods. The primary production at the shallow hydrothermal system of Panarea is supported by a complex microbial community composed by phototrophs and chemolithotrophs.  相似文献

The abundance and diversity of cyanobacteria living in sediments collected from six vents (temperature range 37–130°C) close to the Panarea Island (Italy) were studied. The diversity of cyanobacteria was analyzed by using a PCR-DGGE approach with group-specific primers. The greatest number and diversity of cyanobacteria were registered at the coldest vent. Sequencing of DGGE fragments revealed that most sequences matched with uncultured cyanobacteria deposited in databases and some were distantly affiliated with different species of Synechococcus. New, genetically distant ribotypes of cyanobacteria are the dominant oxygenic phototrophs in shallow hydrothermal system of Panarea Island.  相似文献

Eighty-seven thermophilic, aerobic, spore-forming bacteria were isolated from shallow, marine, thermal vents of the Eolian Islands (Italy) and tested for a broad spectrum of phenotypic characteristics. A numerical taxonomy study was performed on these isolates and 8 thermophilic Bacillus and Geobacillus reference strains by 89 selected features. Results from cluster analysis showed the formation of nine clusters. Most of the isolates (83%) fell into several phenetically well distinguished clusters, loosely related to Geobacillus thermodenitrificans. The remaining isolates grouped together with different reference strains. Eighteen isolates, representative of the different clusters, were selected for subsequent genotypic characterisation, including partial 16S rDNA sequence analysis of 18 strains and almost complete 16S rDNA sequences of 9 strains. Subsequent DNA/DNA reassociation studies and determination of the base composition of DNA identified seven isolates as Geobacillus thermodenitrificans, two isolates as G. thermoleovorans and one isolate as Bacillus pallidus. Four isolates represented two novel species of Bacillus. The remaining four represented novel Geobacillus species, one of which has recently been described as Bacillus vulcani DSMZ 13174 T.  相似文献

Hyperbenthic harpacticoid samples from Japanese hydrothermal vents in the Okinawa Trough and cold seep sites in Sagami Bay were examined and resulted in the discovery of four new species belonging to three new genera of Aegisthidae (Copepoda: Harpacticoida). Females of Nudivorax todai gen. et sp. nov. possess a large area of flexible integument between the cephalosome and the first pedigerous somite which is suggestive of a gorging feeding strategy. Main diagnostic characters separating the new genus from other Aegisthidae are provided by the unusually short caudal rami, the complete lack of intcgumcntal surface lamellae, and the presence in the male of a linear array of pores along the rostral margin which appears to be sensory in function. Scabrantenna yooi gen. et sp. nov. displays several similarities with Aegisthus aculeatus Giesbrecht, 1891 but is highly distinctive in its male morphology which includes extremely atrophied mouthparts and a unique prehensile antenna. Jamstecia terazakii gen. et sp. nov. is only known from a single female caught in the Okinawa Trough. Jamstecia gen. nov. is most closely related to Andromastax Conroy-Dalton & Huys, 1999 but can be distinguished on the basis of the elongate antennules, the antennary morphology, the absence of lateral spinous processes on the cephalosome and swimming legs 2–4, and differences in the mandibular palp and armature of the maxilliped. Andromastax cephaloceratus sp. nov. differs from the type species A. muricatus Conroy-Dalton & Huys, 1998 primarily in the presence of long spinous processes on the cephalosome and the absence of the inner seta on the female P5.  相似文献

Thermophilic microorganisms (4001-4014), described as aerobic or facultatively anaerobic, endospore forming with growth optima temperatures in the range of 60 to 80 degrees C, have been isolated from hot marine springs around Ischia and from hydrothermal vents in the gulf of Naples. Mucous colonies are been selected for the recovery of new strains producing exopolysaccharides (EPS). To induce the biosynthesis of new exopolysaccharides, different sugars were tested as carbon sources in the media. The production of EPS in the strain 4009 reached 60 mg/l using trehalose as carbon source, increasing the yield of about 1000 fold. The 4001-EPS was a mannan with a molecular weight of 380.000 D and with a complex primary structure. In fact, the analysis of the permethylated polysaccharide in GC-MS, showed the presence of mannose, glucose, galactose, mannosamine in the relative ratio of 1:0.1:tr :tr, respectively. Nuclear magnetic resonance spectrum of the exopolysaccharide confirmed the presence of a repetitive unity formed by seven monosaccharides, six with alpha gluco/galacto configuration and one residue with beta conformation.  相似文献

The denitrification potential and density of denitrifying bacteria were determined in suboxidized sediments of Matupi Harbor (Papua New Guinea). The sediments were characterized by low denitrification potentials, not exceeding 0.8 nM/(g h). The maximum density of denitrifying bacteria was recorded in the upper sediment horizons and was 104 cells/g of wet ground.  相似文献

During the BIOACCESS Japanese cruises (1996 & 1998), active hydrothermalism and associated vent fauna were studied on the South-eastern Rift of Manus Basin (South-western Pacific). In the PACMANUS vent field, a conspicuous vent fauna was sampled, including an actinostolid sea anemone (Actiniaria) belonging to an undescribed genus and species. Pacmanactis hashimotoi gen. et spec. nov. is here described, and represents the 9th sea anemone reported from hydrothermal vents.  相似文献

Sediment cores were collected at 5 stations in Lake Varese. They were analyzed for organic matter, N, P, organic C, Cd, Cr, Cu, Hg, Pb, Zn and PAHs. A sedimentological approach has been applied to estimate the ecological risk from identified pollutants.As in other eutrophic lakes in Northern Italy, this lake is also at considerable risk from heavy metal pollution. Cr, Cu and Cd showed the highest enrichment factors for the last 5 years, 23.4, 8.0 and 7.6 respectively. Other metals had enrichment factors ranging from 1 to 3. Fluoranthene was chosen as a representative PAH, derived from combustion products; its average value in surficial sediments ranged from 100 to 220 ng l-1 dw. This compound can be a hazard to human health and aquatic life. An evaluation of radionuclide distributions after the recent Chernobyl accident in the USSR (134Cs, 137C s, 131I, 106Ru) provided useful tracers to follow the cycling of pollutants bound to particulate matter in the aquatic ecosystem.  相似文献

Enteromorpha compressa is the dominant species in coastal areas of northern Chile receiving copper mine wastes. Copper remains as the main heavy metal in these coastal waters and it is accumulated in E. compressa growing at the impacted sites. Algae from these sites showed higher levels of lipoperoxides than from non‐impacted sites, which suggests the occurrence of cellular damage resulting from oxidative stress. The strong activation of ascorbate peroxidase detected in this study probably occurs in order to buffer this oxidative stress. Unexpectedly, the activity of glutathione reductase, normally coupled to ascorbate peroxidase activity, was not affected by the chronic exposure to the mine wastes. Moreover, catalase, dehydroascorbate reductase and glutathione peroxidase, commonly reported to buffer oxidative stress in plants and algae, were not detected in E. compressa from any of the studied sites. Levels of total glutathione and phenolic compounds decreased in algae from mine‐impacted sites. In contrast, high levels of dehydroascorbate were found in algae from impacted sites, whereas ascorbate remained unchanged. Therefore, it is suggested that E. compressa tolerates a copper‐enriched environment, and the accompanying oxidative stress, through the accumulation of copper, activation of ascorbate peroxidase, synthesis of ascorbate (accumulated as dehydroascorbate) and consumption of glutathione and water‐soluble phenolic compounds.  相似文献

Abstract

Onosma echioides is a non-obligate serpentinophytic borage occurring discontinuously on calcareous and serpentine outcrops at the northwest limit of its range. Mean concentrations of Ca, Mg and heavy metals in root and shoot samples of eight populations from the two soil types were first determined. Subsequently, the genetic polymorphism of the same accessions was estimated by means of Amplified Fragment Length Polymorphism (AFLP) fingerprinting technique. Root and shoot samples from serpentine outcrops showed higher levels of Ni, Cr and Mg, and lower Ca/Mg ratios compared with those from calcareous soils. Based on 353 polymorphic AFLP bands, the two edaphic groups of populations showed comparable levels of genetic diversity. A remarkable genetic differentiation between populations and a high level of within-population genetic variance were found. Results of Mantel's test supported a significant correlation between genetic and geographical distances, while no difference in relation to the edaphic factor was detected. Molecular data suggested isolation as the key factor shaping the infraspecific genetic structure of O. echioides, which may be in relation with the short-distance, zoochorous systems of seed dispersal and pollination of this species.  相似文献

Our study aimed to analyse the vegetation of the Borgotrebbia landfill in phytosociological and ecological terms, in order to contribute to the current knowledge of the landfill's vegetation, and to better understand the environmental characteristics of the area, with a view to its restoration. Five vegetation types were identified, all classified into the Stellarietea mediae class that includes annual ruderal communities. Ellenberg's and Landolt's indices shed light on the ecological characteristics of all the vegetation and five typologies. The vegetation of the study area indicates a moderately basic, semi-dry soil, rich in nutrients, low in humus and badly aerated. The high therophyte percentage indicates a stressed environment, the main cause of this stress being a marked edaphic aridity during the dry summer months. In these conditions, annual plants, which complete their life cycle in a short time after the spring precipitations, are favoured. Finally, the species variety within the various typologies of vegetation increases with a decrease in the percentage of species tolerating salts and heavy metals in the soil. As a consequence, this suggests a possible contamination of the soil in some of the areas.  相似文献

Planktonic and benthic bacterial communities hold central roles in the functioning of freshwater ecosystems and mediate key ecosystem services such as primary production and nutrient remineralisation. Although it is clear that such communities vary in composition both within and between lakes, the environmental factors and processes shaping the diversity and composition of freshwater bacteria are still not fully understood.
  2. In order to assess seasonal and spatial variability in lake bacterial communities and identify environmental factors underpinning biogeographical patterns, we performed a large-scale sampling campaign with paired water and sediment sample collection at 18 locations during four seasons in Lake Balihe, a subtropical shallow fish-farming lake in mid-eastern China.
  3. Pelagic and benthic bacterial communities were distinctly different in terms of diversity, taxonomic composition and community structure, with Actinobacteria, Bacteroidetes, Cyanobacteria and Alphaproteobacteria dominating lake water, and Acidobacteria, Bacteroidetes, Chloroflexi, Gammaproteobacteria and Deltaproteobacteria dominating sediment. Nevertheless, these two communities had stronger spatial concordance and overlap in taxa during spring and autumn seasons. Together, the main drivers of both the spatial and temporal variations in Lake Balihe bacterial communities were identified as water temperature, turbidity, nitrogen and phosphorus availability, and thermal stratification controlled by wind-mixing and activity of the dense farmed fish populations. Notably, populations affiliated with Firmicutes, known to be abundant in fish gut microbiome, were especially abundant in the summer season and locations where high fish biomass was found, suggesting a potential link between fish gut microbiome and the pelagic bacterial communities.
  4. Our findings demonstrated seasonal homogenisation of pelagic and benthic bacterial communities linked to marked shifts in a set of seasonally-driven environmental variables including water temperature and nutrient availability.

Large symbiont-hosting snails of the genus Alviniconcha (Gastropoda: Abyssochrysidae) are among the dominant inhabitants of hydrothermal vents in the Western Pacific and Indian oceans. The genus was originally described as monotypic, but unique DNA sequences for mitochondrial genes revealed six distinct evolutionary lineages that we could not distinguish based on external morphology. Subsumed under the name Alviniconcha hessleri Okutani & Ohta, the distinct allopatric and sympatric lineages have been assigned placeholder epithets that complicate scientific communications. Based on the present multi-gene sequence data, we hereby describe five Alviniconcha species (in the order of their discovery) – A. kojimai sp. nov., A. boucheti sp. nov., A. marisindica sp. nov., A. strummeri sp. nov. and A. adamantis sp. nov. Thus, we restrict application of the name A. hessleri to specimens that are genetically similar (≥95% for COI) to those found at localities in the Mariana Trough. Single distinct Alviniconcha species inhabit vent fields along the Central Indian Ridge, the Mariana volcanic arc, and the Mariana back-arc basin, whereas vents in the Manus, Fiji and Lau back-arc basins may host two or three additional species. Formal recognition of these species facilitates future attempts to assess their physiological differences and symbiont associations. Furthermore, their reported distributions have significant biogeographic implications, affecting estimates of the diversity within and overlap among Indo-Pacific vent localities.

Abstract

The distribution, contamination status, and ecological risks of heavy metals in Tahaddart estuary were investigated. 24 surface sediment samples and two cores were collected and analyzed for major (Al and Fe), heavy metals (As, Cd, Cr, Cu, Ni, Pb, and Zn), and grain size composition. The heavy metals assessment was carried out using different environmental indices. The results indicated that the spatial distribution patterns of Al, Fe, and Zn were mainly determined by the distribution of the finer grained fraction (<63?μm) in the sediment. In contrast, As, Cd, Cr, Cu, Ni, and Pb concentrations were controlled by anthropogenic activities (vehicular traffic from Highway Bridge and thermal power plant). The distribution of heavy metals in sediment cores showed an upward enrichment in heavy metals with high concentration found in the uppermost may related to the increasing in human activities. The pollution indexes confirmed that the Tahaddart estuary sediment was considerably to high contaminated by heavy metals near to different anthropogenic inputs. Similarly, the potential ecological risk index and the biological risk index present 21% probability of toxicity posing potential risk to the aquatic organisms. These results provide basic information that can be used to protect and improve the quality of this ecosystem.  相似文献

Metal-contaminated sites can occur naturally in serpentine outcrops or as consequence of anthropogenic activities, such as mining deposits, aerial fallout from smelters and industrial processes. Serpentine outcrops are characterized by high levels of nickel, cobalt and chromium and present a typical vegetation which includes endemisms and plants which also live in uncontaminated soils. These latter metal-tolerant populations provide the opportunity to investigate the first steps in the differentiation of plant populations under severe selection pressure and to select plants to be used in the phytoremediation of industrially contaminated soils. In this report eight populations of Silene paradoxa L. (Caryophyllaceae) growing in copper mine deposits, in serpentine outcrops or in noncontaminated soil in central Italy, were analysed using random amplified polymorphic DNA (RAPD) markers to investigate the pattern of genetic variation. The genetic diversity observed in populations at copper mine deposits was found to be at least as high as that of the neighbouring serpentine populations. Analysis of molecular variance (AMOVA) of the RAPD markers gave high statistical significance to the groupings of populations according: (i) with their geographical location; and (ii) with the metals present in the soil of origin (copper vs. nickel), indicating that RAPD markers detected a polymorphism related to the soil contamination by copper. Finally, two RAPD bands exclusive to copper-tolerant populations were identified.  相似文献

Abstract

The aim of this study was to investigate the extent of heavy metal (HM) pollution and its effect on microorganisms from rhizosphere soil in Baia Mare area (Maramure? County, Romania). Two sites with different contamination degrees were included in the study: one with a long history of mining activities and one within a drinking water safeguard zone. Rhizosphere soil samples were characterized with respect to physico-chemical parameters and the Cd, Cu, Pb and Zn contents. Native bacteria were investigated for HM tolerance and biofilm formation under toxic exposure by the microdilution assay. The most resistant strains were identified and the minimum inhibitory concentrations for HMs were determined. Cd, Cu, Pb and Zn exceeded the intervention threshold in Bozânta tailings site, while Pb content exceeded the intervention level within the area of the drinking water treatment plant. Cd showed a very high potential ecological risk in Bozânta area. The long-term exposure to HMs contributed to the selection of HM-tolerant and weakly adherent strains. Biofouling was significantly reduced under the influence of copper ions. Arthrobacter, Rhodococcus and Acidovorax strains with exceptional resistant profiles were isolated from the tailings site, indicating the important role of native microorganisms in rhizosphere ecosystems of contaminated sites.  相似文献

The phytoplankton succession in Lake Orta, heavy metals and ammonium polluted for decades and subjected to liming intervention in 1989–90, was studied during 1994. The existence of three main succession stages, with different structures of the community emerged. Chlorophyta are the most important taxonomic group in every season. The spring community was dominated by chlorophytes, Dinobryon sertularia and Kephyrion sp., the summer by Synedra sp. and Westella botryoides, the autumn by Rhizosolenia eriensis and W. botryoides, the winter by Synedra sp., R. eriensis and Synechococcus sp. The abundance of Chlorophyta, less important in other deep subalpine lakes, and the dominance of taxa able to thrive in polluted environments, could be related to the presence of trace metals in the sediments.  相似文献
