This work examines the influence of modified gum karaya (MGK) on the oral bioavailability of a poorly water-soluble drug, nimodipine (NM), in comparison with that of gum karaya (GK). A cogrinding method was selected to prepare mixtures of NM and GK or MGK in a 1:9 ratio (NM:GK/MGK). Differential scanning calorimetry (DSC), Fourier transmission infrared (FT-IR) spectroscopy, X-ray diffraction (XRD), solubility studies, and in vitro release studies were performed to characterize the properties of the cogrinding mixtures. No drug-carrier interactions were found, as confirmed by DSC and FT-IR studies. The XRD study revealed that the crystallinity of NM was identical in both the cogrinding mixtures and was decreased when compared to that of physical mixtures or pure NM. The in vitro release rate of NM from both cogrinding mixtures was significantly higher than that of physical mixtures or pure NM. The in vivo study revealed that the bioavailability of NM from pure drug was significantly lower when compared to the cogrinding mixtures. The oral bioavailability was found to be NM powder < cogrinding mixtures of NM and GK < cogrinding mixtures of NM and MGK < NM solution. It can be inferred from the above results that MGK, an economical carrier, could be used for the dissolution enhancement of NM.  相似文献

Male and female albino rats (Wistar strain) were given single and multiple doses of karaya gum suspended either in peanut oil or mixed with basal diet at different concentrations ranging from 0.5 to 8 g gum/kg body weight. The plant gum did not elicit any overt signs of toxicity or death in both sexes of rats. Daily administration of karaya gum mixed with basal diet at different dose levels (0, 5, 20 and 40 g gum/kg diet) for a period of 90 days showed no adverse effects in male and female rats. The body weight, growth pattern, food and water intake were comparable with those of the normal rats. There were no significant biochemical, or morphological alterations in the vital organs of experimental animals.  相似文献

Ten Bacteroides species found in the human colon were surveyed for their ability to ferment mucins and plant polysaccharides ("dietary fiber"). A number of strains fermented mucopolysaccharides (heparin, hyaluronate, and chondroitin sulfate) and ovomucoid. Only 3 of the 188 strains tested fermented beef submaxillary mucin, and none fermented porcine gastric mucin. Many of the Bacteroides strains tested were also able to ferment a variety of plant polysaccharides, including amylose, dextran, pectin, gum tragacanth, gum guar, larch arabinogalactan, alginate, and laminarin. Some plant polysaccharides such as gum arabic, gum karaya, gum ghatti and fucoidan, were not utilized by any of the strains tested. The ability to utilize mucins and plant polysaccharides varied considerably among the Bacteroides species tested.  相似文献

Only three gum exudates are permitted for pharmaceutical and food use by international regulatory authorities, viz. gum tragacanth (Asiatic Astragalus spp.), gum karaya (Sterculia spp.) and gum arabic [Acacia senegal (L.) Willd.], but a wide range of other tree exudates is used for a variety of uses in their countries of origin. This paper presents analytical data for the gum exudates from Atalaya hemiglauca, Cassine aethiopica, Combretum paniculatum, Sclerocarya birrea, and Pseudocedrela kotschyi. These gums may have local technological applications, but are not recommended for addition to foodstuffs.  相似文献

A survey of the inhibitory effects of various plant polysaccharides on PCR amplification of a 974-bp section of rbcL in spinach revealed that most of the polysaccharides tested (arabinogalactan, carrageenan, dextran, gum guar, gum karaya, gum locust bean, inulin, mannan, pectin, starch and xylan) were not inhibitory. In contrast, two of the acidic polysaccharides (dextran sulfate and gum ghatti) were inhibitory. The addition of 0.5% Tween 20 reversed the inhibitory effects of gum ghatti (polysaccharide:DNA ratio of 500:1). The inhibitory effect of dextran sulfate (50:1) could be reversed by the addition of Tween 20 (0.25% or 0.5%), DMSO (5%) or polyethylene glycol 400 (5%), but none of these three additives were effective at a 100:1 ratio of dextran sulfate/DNA.  相似文献

A survey of the inhibition of the amplification of spinach DNA by various plant polysaccharides revealed that neutral polysaccharides (arabinogalactan, dextran, gum guar, gum locust bean, inulin, mannan, and starch) were not inhibitory. In contrast, the acidic polysaccharides (carrageenan, dextran sulfate, gum ghatti, gum karaya, pectin, and xylan)were inhibitory. In the process of preparing random amplified polymorphic DNAs (RAPDs), the loss of large DNA bands appears to be an indicator that the fingerprint pattern has been affected by polysaccharides. The addition of various concentrations of Tween 20, DMSO, or PEG 400 to the PCR reaction mixture resulted in partial restoration of amplification of RAPDs for the acidic polysaccharides. The most effective way to eliminate the effects of polysaccharide inhibition was by diluting the DNA extracts, and thereby diluting the polysaccharide inhibitors.  相似文献

Gum karaya (Sterculia urens gum) was degraded by a fungal isolate, a Cephalosporium sp. The patterns of carbohydrate utilization and enzyme secretion by the organism growing on the purified polysaccharide were determined, together with the ratio of neutral sugars present in the gum. Cephalosporium growth-studies indicate that the gum contains at least three different types of chains. One chain (50% of the total polysaccharide) is postulated to contain repeating units of four galacturonic acid residues containing β-D-galactose branches and having an L-rhamnose residue at the reducing end of the unit. A second chain (17% of the polysaccharide) contained 50% of galacturonic acid, 40% of rhamnose, and 10% of galactose by weight and is postulated to contain an oligorhamnan chain, containing D-galacturonic acid branch-residues, and interrupted occasionally by a D-galactose residue. D-Glucuronic acid is apparently confined to a third type of chain, comprising 33% of the polysaccharide.  相似文献

A panel of monoclonal antibodies, derived for the specific recognition of developmentally regulated epitopes of the plasma membranes and cell walls of plant cells, has been characterized in relation to binding activities with samples of exudate gums. The monoclonal antibodies are shown to recognize specifically structural features present in four exudate gums—arabic, karaya, ghatti and tragacanth. Dialysis of the gums and competitive inhibitor ELISAs indicated that the monoclonal antibodies recognize both high and low molecular weight components of each gum. The spatial relationships, and possible overlap, of the arabinogalactan epitopes on the most antigenic gum, gum arabic, have been explored in more detail by means of an epitope mapping technique.  相似文献

The non-Newtonian behavior and dynamic viscoelasticity of a series of aqueous mixtures of xanthan and guar gum were measured with a rheogoniometer. At a concentration of 0.2% of total gums, gelation did not occur at room temperature but occurred at a low temperature (0°). A much stronger interaction was observed with a mixture of deacetylated xanthan than that with native xanthan. The maximum dynamic modulus was obtained when the ratio of xanthan to guar gum was 2:1. The transition temperatures of dynamic viscoelasticity for mixtures with native and deacetylated xanthan were observed at 25 and 30°, respectively. It was concluded that the side chains of the guar gum molecular prevent an intermolecular interaction with the side chains of the xanthan molecule. An intermolecular interaction between xanthan and guar gum at low temperature might be promoted between the periphery of the side chains of the xanthan molecule and the backbone of the guar gum molecule and dissociation takes place at the transition temperature.  相似文献

The prospective uses of tree gum polysaccharides and their nanostructures in various aspects of food, water, energy, biotechnology, environment and medicine industries, have garnered a great deal of attention recently. In addition to extensive applications of tree gums in food, there are substantial non-food applications of these commercial gums, which have gained widespread attention due to their availability, structural diversity and remarkable properties as ‘green’ bio-based renewable materials. Tree gums are obtainable as natural polysaccharides from various tree genera possessing exceptional properties, including their renewable, biocompatible, biodegradable, and non-toxic nature and their ability to undergo easy chemical modifications. This review focuses on non-food applications of several important commercially available gums (arabic, karaya, tragacanth, ghatti and kondagogu) for the greener synthesis and stabilization of metal/metal oxide NPs, production of electrospun fibers, environmental bioremediation, bio-catalysis, biosensors, coordination complexes of metal–hydrogels, and for antimicrobial and biomedical applications. Furthermore, polysaccharides acquired from botanical, seaweed, animal, and microbial origins are briefly compared with the characteristics of tree gum exudates.  相似文献

The interaction of κ-carrageenan with locust bean gum and dextran has been studied by rheology, differential scanning calorimetry (DSC), and electron spin resonance spectroscopy (ESR). Rheological measurements show that the carrageenan gel characteristics are greatly enhanced in the presence of locust bean gum but not in the presence of dextran. Carrageenan/locust bean gum mixtures show two peaks in the dsc cooling curves. The higher temperature peak corresponds to the temperature of gelation and its intensity increases at the expense of the lower temperature peak as the proportion of locust bean gum in the mixture increases. Furthermore, the DSC heating curves show enhanced broadening when locust bean gum is present, indicating increased aggregation. These results are taken as evidence of carrageenan/locust bean gum association. The gelation process has also been followed by ESR using spin-labeled carrageenan. On cooling carrageenan solutions, an immobile component appears in the ESR spectra signifying a loss of segmental mobility consistent with chain stiffening due to the coil → helix conformational transition and helix aggregation. For carrageenan/locust bean gum mixtures, carrageenan ordering occurs at temperatures corresponding to the higher temperature DSC setting peak and the temperature of gelation. Similar studies using spin-labeled locust bean gum show that its mobility remains virtually unaffected during the gelation process. It is evident, therefore, that carrageenan and locust bean gum interact only weakly. It is proposed that at low carrageenan concentrations the gel network consists of carrageenan helices cross-linked by locust bean gum chains. At high carrageenan concentrations the network is enhanced by the additional self-aggregation of the “excess” carrageenan molecules. For carrageenan/dextran mixtures, only one peak is observed in the dsc cooling curves. The onset of gelation shifts to higher temperatures only at very high (20%) dextran concentrations and this is attributed to volume exclusion effects. Furthermore, there is no enhanced broadening of the peaks in the DSC heating curves as for the carrageenan/locust bean gum systems. It is therefore concluded that carrageenan/dextran association does not occur. The difference in behavior between locust bean gum and dextran is attributed to the greater flexibility of the dextran chains. © 1996 John Wiley & Sons, Inc.  相似文献

Oral administration of antibiotics to treat dental problems mostly yields slow actions due to slow onset and hepatic “first-pass.” Again, commonly used dental paints are generally washed out by saliva within few hours of application. To overcome the challenges, polymeric molds to be placed on an affected tooth (during carries and gum problems) were prepared and evaluated in vitro for sustained drug release for prolonged local action. Here, amoxicillin trihydrate and lidocaine hydrochloride were used as model drugs. Dental molds were prepared using corn zein, carbopol 934 P, gum karaya powder, and poloxamer 407 by mixing and solvent evaporation technique. Different physicochemical evaluation studies such as tooth adhesion test, surface pH, swelling index, and drug-distribution pattern were carried out. Percentage swelling varied from 56% to 93%. Average tooth adhesion strength and mean initial surface pH of the formulations were 50 g and 6.5, respectively. As assessed by scanning electron microscopy, drug distribution was uniform throughout the matrix. Cumulative percentage release of lidocaine hydrochloride and amoxicillin trihydrate in simulated saliva were 98% and 50%, respectively. In vitro drug-release studies revealed the sustained-release patterns of the drugs in simulated saliva at least for 24 h. The stability study shows that the drugs were stable in the formulations following the conditions as per ICH guideline. The formulation is a novel approach to deliver the drug(s) for a prolonged period for local action upon its application on an affected tooth.  相似文献

A method is described for the preparation of purified polysaccharides from strains of Rhizobium in quantities large enough so that with an exhaustive purification scheme enough product is recovered for various characterization purposes. When steps in the purification process are eliminated, much larger amounts of crude gum are obtained.

Organisms were grown in liquid medium and the crude gum was precipitated along with the bacterial cells by a quaternary ammonium complexing agent. This precipitate was dissolved in salt solution, reprecipitated with ethanol or ethanol-acetone mixtures several times, followed by pressure filtration with membrane filters.

The same procedure should be applicable to commercial scale production in large fermentors if a use for the gum could be shown. The method also should be suitable for other organisms with similar growth habits.

Six non-anaesthetized Large White pigs (mean body weight 59 +/- 1.7 kg) were fitted with permanent catheters in the portal vein, the brachiocephalic artery and the right hepatic vein and with electromagnetic flow probes around the portal vein and the hepatic artery. The animals were provided a basal none-fibre diet (diet A) alone or together with 6% guar gum (diet B) or 15% purified cellulose (diet C). The diets were given for 1 week and according to a replicated 3 x 3 latin-square design. On the last day of each adaptation period test meals of 800 g were given prior to blood sampling. The sampling was continued for 8 h. Guar gum strongly reduced the glucose absorption as well as the insulin, gastric inhibitory polypeptide (GIP) and insulin-like growth factor-1 (IGF-1) production. However, the reduction in peripheral blood insulin levels caused by guar gum was not associated with a change in hepatic insulin extraction. IGF-1 appeared to be strongly produced by the gut. The liver had a net uptake of the peptide. Ingestion of guar gum increased the hepatic extraction coefficient of gut produced IGF-1. Guar gum ingestion also appeared to decrease pancreatic glucagon secretion. Cellulose at the level consumed had very little effect on the parameters considered. It is suggested that the modulation of intestinal mechanisms by guar gum was sufficient to mediate the latter internal metabolic effects.  相似文献

Data for the depression of vapour pressure are presented for the following aqueous solutions: NaCl (0.03 to 0.1 molar), KCl (0.03 to 0.1 molar), urea (0.05 to 0.5 molar), sucrose (0.05 to 0.10 molar), lactic and succinic acids, creatine, CaCl2 (0.05 molar), and mixtures of these substances with one another and with certain other solutions (gelatin, gum acacia, sea water, LiCl, etc.). The relation of the depression of vapour pressure of a mixed solution to that of solutions of the individual constituents was investigated in order to ascertain to what extent such studies may be used for the determination of the degree of hydration, or of the state of water, in solutions. Organic substances (urea, sucrose, etc.) showed anomalous results which were markedly affected and unpredictable in mixed solutions. They are, therefore, unsuited for the study of water binding. In the case of solutions of inorganic substances—LiCl and CaCl2—the principle of the additive nature of colligative properties is also only approximately true—except perhaps in very dilute solutions. The limitations of the colligative method for determining the degree of hydration have been defined in accord with the above findings. Studies of the vapour pressures of mixtures of gelatin or gum acacia with NaCl or KCl demonstrated that hydration in gelatin is relatively small at pH = 7 and undetectable in gum acacia solutions. The view, therefore, that hydrophilic colloids are strongly hydrated has not been substantiated. The passage from the sol to the gel state also was not accompanied in gelatin or in blood by any appreciable change in the degree of hydration of the hydrophilic colloids present in these substances.  相似文献

Summary A diallel cross analysis of gum content in barley (Hordeum vulgare) was made using six cultivars of two-rowed spring barley as parents. A Jinks-Hayman analysis of F2 progeny means showed that gum content was controlled by a simple additive-dominance genetic system and that low gum content was strongly dominant. The analysis suggested that gum content was principally controlled by two or three genes showing a high degree of dominance. Some genotype-environment interaction was detected in a comparison between the F2 and F3 generations which were grown in different years and locations. However, the character was found to be highly heritable both within and between generations, suggesting that the selection and breeding of barleys of reduced gum content should not be difficult.  相似文献

This study describes the effects of mixtures of xanthan gum and galactomannan, guar gum, or locust bean gum, on the lipids in plasma and liver in non-diabetic and diabetic rats. Non-diabetic rats were fed cholesterol-free diets with 3% guar gum, locust bean gum, or xanthan gum (3G, 3L, and 3X), or a mixture of xanthan gum and guar gum or locust bean gum (1:2, w/w) (2G1X, 2L1X) for 2 weeks. Rats fed diets not containing these polysaccharides were used as controls. The total cholesterol in plasma and the triacylglycerol in liver were significantly lowered in rats fed the 2G1X diet. The 3G, 3X, 3L, and 2L1X diets showed no significant effect on the total cholesterol and triacylglycerol in plasma and liver. In the streptozotocin-induced (STZ) diabetic rats, the total cholesterol in plasma was lowered in rats fed the 3G, 3X or 2G1X diet for 4 weeks, and the 2G1X diet was more effective than the 3G and 3X diets. The triacylglycerol in plasma in STZ diabetic rats was also significantly lowered by the 2G1X diet. These results showed that a mixture of xanthan gum and guar gum has an improved hypolipidemic effect on non-diabetic and STZ diabetic rats. The effects of the 2G1X diet on the diabetic symptoms in STZ diabetic rats, suppression of food and water intakes, decrease in glucose in urine, and lowering of plasma glucose, were also observed.  相似文献

Summary When paraffin sections of mouse and rat kidney, liver and spleen were fixed in a formalin-type fixative overnight, then preserved in Holt's hypertonic gum-sucrose solution, erythrocytes stained strongly by the McManus periodic acid-Schiff method. The same tissues processed without gum-sucrose did not exhibit such positive staining of red blood cells. Tissues treated with the individual gum-sucrose components, a solution of gum arabic or of sucrose, gave positive staining of erythrocytes only after gum arabic. Powdered gum arabic but not sucrose crystals stained positively. The erythrocyte reaction in formalinfixed tissue treated with gum-sucrose is therefore related to the presence of gum arabic in Holt's solution. Opposing results were encountered with two non-formalin fixatives, glutaraldehyde and acetone, in that staining was consistently positive throughout in the former and consistently negative in the latter, regardless of gum-sucrose treatment.  相似文献
