We examined how dominance (% canopy cover) and invasion history of common reed, Phragmites australis, affected benthic macroinvertebrate diversity and density in 8 marshes along Lake Erie’s southern shoreline. We also compared macroinvertebrate densities among patches (0.25 m2) of reed, cattail (Typha spp.), and native flora (e.g., Sagittaria, Sparganium) and epiphytic algal communities on submerged stems of reed and cattail. Narrow-leaf cattail (T. angustifolia) is also a common invasive plant to these wetlands, but does not greatly change plant community composition or ecosystem conditions like reed. Macroinvertebrate diversity (Shannon–Weaver H′) was positively related to reed cover and was highest (4.6) in two marshes with ~35- and 5-year invasion histories. Shading from high reed cover increased H′-diversity, in part, by reducing the abundance of floating duckweed, which harbored many Hyalella azteca amphipods. Percent Ephemeroptera, Odonata, and Trichoptera was low to moderate across marshes, regardless of reed cover and invasion history. Macroinvertebrate density was not affected by reed cover or average plant stem density, and did not differ among plant types. However, epiphyton densities and % diatoms were greater on reed than on cattail, suggesting reed provides a better feeding habitat for microalgal grazers than Typha. Abundance rankings of common species in these diatom-dominated communities were also typically dissimilar between these plant types. Although % grazers was unrelated to epiphyton densities and % diatoms, grazer identity (snails) differed between natural and diked marshes, which had different microalgal food supplies. Our findings suggest that Phragmites does not necessarily adversely affect macroinvertebrate community structure and diversity and that invasion history alone has little effect on the H′-diversity–reed dominance relationship.  相似文献

Azospirillum isolates were obtained from rhizosphere soil and roots of three cactaceae species growing under arid conditions. All Azospirillum isolates from rhizosphere and roots ofStenocereus pruinosus andStenocereus stellatus were identified asA. brasilense; isolates of surface-sterilized roots fromOpuntia ficus-indica were bothA. brasilense andA. lipoferum. Azospirilla per g of fresh root in the three species ranged from 70×103 to 11×103. The most active strains in terms of C2H2 reduction (25–49.6 nmol/h·ml) and indoleacetic acid (IAA) production (36.5–77 μg/ml) were those identified asA. brasilense and isolated from Stenocereus roots.A. lipoferum isolated from Opuntia roots produced low amounts of IAA (6.5–17.5 μg/ml) and low C2H2-reduction activity (17.8–21.2 nmol/h·ml).  相似文献

A total of 360 bacteria, isolated from the rhizospheres of a system of rice intensification (SRI) fields, were characterized for the production of siderophore, fluorescence, indole acetic acid (IAA), hydrocyanic acid (HCN) and solubilization of phosphorus. Of them, seven most promising isolates (SRI-156, -158, -178, -211, -229, -305 and -360) were screened for their antagonistic potential against Macrophomina phaseolina (causes charcoal rot in sorghum) by dual culture assay, blotter paper assay and in greenhouse. All the seven isolates inhibited M. phaseolina in dual culture assay, whereas six isolates solubilized phosphorous (except SRI-360), all seven produced siderophore, four produced fluorescence (except SRI-178, -229 and -305), six produced IAA (except SRI-305) and five produced HCN (except SRI-158 and -305). In the blotter paper assay, no charcoal rot infection was observed in SRI-156-treated sorghum roots, indicating complete inhibition of the pathogen, while the roots treated with the other isolates showed 49–76% lesser charcoal rot infection compared to the control. In the antifungal activity test (in green house on sorghum), all the isolates increased shoot dry mass by 15–23% and root dry mass by 15–20% (except SRI-158 and -360), over the control. In order to confirm the plant growth-promoting (PGP) traits of the isolates, the green house experiment was repeated but, in the absence of M. phaseolina. The results further confirmed the PGP traits of the isolates as evidenced by increases in shoot and root dry mass, 22–100% and 5–20%, respectively, over the control. The sequences of 16S rDNA gene of the isolates SRI-156, -158, -178, -211, -229, -305 and -360 were matched with Pseudomonas plecoglossicida, Brevibacterium antiquum, Bacillus altitudinis, Enterobacter ludwigii, E. ludwigii, Acinetobacter tandoii and P. monteilii, respectively in BLAST analysis. This study indicates that the selected bacterial isolates have the potential for PGP and control of charcoal rot disease in sorghum.  相似文献

Summary Different ecotypes of reed (Phragmites communis Trinius) provide an ideal resource for studies on plant environmental adaptations and presence of genes relating to stress resistance. Dune reed is a drought-tolerant reed ecotype growing in the desert regions of north-west China. In this work, in vitro culture systems of dune reed and local swamp reed (as control) were established by optimizing the culture conditions for each of them. Bright yellow calluses were induced on a Murashige and Skoog medium containing 4.5 μM 2,4-dichlorophenoxyacetic acid (2,4-D), 5.4 μM naphthaleneacetic acid and 2.2μM benzyladenine. Benzyladenine promoted callus induction, but was not required for callus maintenance. Four types of callus have been identified from each of the reed ecotypes. Two types of callus, i.e. type A (formed normal green shoots) and type C (formed albino plants), were both found as embryogenic calluses. The optimal concentrations of 2,4-D to maintain embryogenic callus were 2.3–4.5 μM for dune reed and 9.0–13.5 μM for swap reed. Plant regeneration was achieved from types A and C callus in a hormone-free medium. The embryogenic calluses of swamp reed have been maintained for over 2 yr and still retain their strong embryogenic potential; however, those of dune reed gradually lost their embryogenic potential after only 7 mo. of culture. Regenerated plants from the two reed ecotypes showed, after a growth season, similar morphology and the same chromosome number (2n=8x=96, octoploid) as the wild plants.  相似文献

The prevalence of ten antibiotic resistance genes (ARGs) was evaluated in a total of 616 Escherichia coli isolates from swine manure, swine lagoon effluent, and from soils that received lagoon effluent on a commercial swine farm site in Sampson County, North Carolina (USA). Isolates with ARGs coding for streptomycin/spectinomycin (aadA/strA and strB), tetracycline (tetA and tetB), and sulfonamide (sul1) occurred most frequently (60.6–91.3%). The occurrence of E. coli isolates that carried aadA, tetA, tetB, and tetC genes was significantly more frequent in soil samples (34.097.2%) than in isolates from lagoon samples (20.9–90.6%). Furthermore, the frequency of isolates that contain genes coding for aadA and tetB was significantly greater in soil samples (82.6–97.2%) when compared to swine manure (16.8–86.1%). Isolates from the lagoon that carried tetA, tetC, and sul3 genes were significantly more prevalent during spring (63.3–96.7%) than during winter (13.1–67.8%). The prevalence of isolates from the lagoon that possessed the strA, strB, and sul1 resistance genes was significantly more frequent during the summer (90.0–100%) than during spring (66.6–80.0%). The data suggest that conditions in the lagoon, soil, and manure may have an impact on the occurrence of E. coli isolates with specific ARGs. Seasonal variables seem to impact the recovery isolates with ARGs; however, ARG distribution may be associated with mobile genetic elements or a reflection of the initial numbers of resistant isolates shed by the animals.  相似文献

 Strips of horticultural film (16–32 cm2) were used to trap extraradical hyphae emanating from roots of sudangrass [Sorghum sudanense (Piper) Staph] enclosed in 40-μm mesh bags and colonized by Gigaspora rosea FL 224-1, Glomus intraradices EY 113/114, or Glomus caledonium UK 301-1. Strips of film were placed at opposite sides of 17–21 replicate sand culture pots for each isolate and were removed after 12–14 weeks of plant growth. To extract glomalin, a strip was cut into small pieces and submerged in 2 ml of 20 mM citrate, pH 7.0 and then autoclaved for 60 min. A quantitative enzyme-linked immunosorbent assay (ELISA) detected 0.005–0.04 μg glomalin in the volume of extract tested. The Bradford protein assay detected 1.25–5 μg of protein in the volume of extract tested. Both assays gave results ranging from 5–40 μg glomalin/cm2 of film. Protein assay values were correlated with ELISA values (r=0.6091, P≤0.001, n=118). Analysis of variance indicated that isolates differed in Bradford protein values (P=0.001), but not ELISA values (P=0.154). Spatial variability of glomalin deposition ca. 7 cm from roots on opposite sides of pots was indicated by significant paired T tests (P<0.05) for protein values for each of the three isolates and ELISA for two isolates. These results indicate that hyphal traps, Bradford protein assay and ELISA are useful to assess hyphal activity over a growing season. Accepted: 11 October 1998  相似文献

Fungi with dematiaceous septate hyphae, termed dark septate endophytes (DSE), are common in plant roots, particularly in cold-stressed habitats, but their effects on their host plants remain obscure. Here, we report a study that assessed the effects of six DSE on the growth and nutrient balance of Deschampsia antarctica when plants were supplied with the same amount of nitrogen in organic (casein hydrolysate) or inorganic (ammonium sulphate) form under controlled conditions. After 60 days, the DSE, that had each been isolated from D. antarctica and which analyses of internal transcribed spacer and large subunit regions indicated were similar to members of the Helotiales (Oculimacula yallundae, Mollisia and Tapesia spp.) and unassigned anamorphic ascomycetes, typically had no effect on, or reduced by 33–71%, shoot and root dry weights relative to uninoculated controls when plants had been supplied with nitrogen in inorganic form. In contrast, the DSE usually enhanced shoot and root dry weights by 51–247% when plants had been supplied with organic nitrogen. In the presence of inorganic nitrogen, only sporadic effects of DSE were recorded on shoot and root nitrogen or phosphorus concentrations, whereas in the presence of organic nitrogen, three to six of the DSE isolates increased shoot and root nitrogen and phosphorus contents. Most of the isolates decreased the phosphorus concentrations of shoots and roots when plants had been supplied with nitrogen in organic form. Our data suggest that DSE are able to mineralise peptides and amino acids in the rhizosphere, making nitrogen more freely available to roots.  相似文献

Development of microbial inoculants from rhizobacterial isolates with potential for plant growth promotion and root disease suppression require rigorous screening. Fifty-four (54) fluorescent pseudomonads, out of a large collection of rhizobacteria from broad bean fields of 20 different locations within Imphal valley of Manipur, were initially screened for antifungal activity against Macrophomina phaseolina and Rhizoctonia solani, of diseased roots of broad bean and also three other reference fungal pathogens of plant roots. Fifteen fluorescent pseudomonas isolates produced inhibition zone (8–29 mm) of the fungal growth in dual plate assay and IAA like substances (24.1–66.7 μg/ml) and soluble P (12.7–56.80 μg/ml) in broth culture. Among the isolates, RFP 36 caused a marked increase in seed germination, seedling biomass and control of the root borne pathogens of broad bean. PCR–RAPD analysis of these isolates along with five MTCC reference fluorescent pseudomonas strains indicated that the RFP-36 belonged to a distinct cluster and the PCR of its genomic DNA with antibiotic specific primers Phenazine-1-carboxylic acid and 2, 4-diacetyl phloroglucinol suggested possible occurrence of gene for the potent antibiotics. Overall, the result of the study indicated the potential of the isolate RFP 36 as a microbial inoculant with multiple functions for broad bean.  相似文献

Bark beetles are well known vectors of ophiostomatoid fungi including species of Ophiostoma, Grosmannia and Ceratocystis. In this study, the most common ophiostomatoid fungi associated with the lodgepole pine beetle, Dendroctonus murrayanae, were characterized. Pre-emergent and post-attack adult beetles were collected from lodgepole pines at four sites in British Columbia, Canada. Fungi were isolated from these beetles and identified using a combination of morphology and DNA sequence comparisons of five gene regions. In all four populations, Grosmannia aurea was the most common associate (74–100% of all beetles) followed closely by Ophiostoma abietinum (29–75%). Other fungi isolated, in order of their relative prevalence with individual beetles were an undescribed Leptographium sp. (0–13%), Ophiostoma ips (0–15%), Ophiostoma piliferum (0–11%), a Pesotum sp. (0–11%) and Ophiostoma floccosum (0–1%). Comparisons of the DNA sequences of Leptographium strains isolated in this study, with ex-type isolates of G. aurea, Grosmannia robusta, Leptographium longiclavatum, and Leptographium terebrantis, as well as with sequences from GenBank, revealed a novel lineage within the Grosmannia clavigera complex. This lineage included some of the D. murrayane isolates as well as several isolates from previous studies referred to as L. terebrantis. However, the monophyly of this lineage is not well supported and a more comprehensive study will be needed to resolve its taxonomic status as one or more novel taxa.  相似文献

We describe endorhizal fungi associated with Equisetum species collected from Ellesmere Island (82°N), Axel Heiberg Island (80°N), and from sites in Yukon Territory and the Prairie Provinces (51–67°N). Fungal colonization was assessed using a multiple quantitation microintersect method for lactofuchsin-stained roots examined with wide-field and confocal epifluorescence microscopy. Equisetum roots host abundant and diverse endorhizal fungal associates. For 85 specimens from 14 sites, total colonization averaged 30 ± 3%, range 0–97%. Colonization rates for wide aseptate hyphae characteristic of arbuscular mycorrhizae (5 ± 1%) was significantly less than for fine endophytes (20 ± 3%) or septate endophytes (17 ± 2%). Equisetum spp. are abundant in tundra and an important understory plant in boreal forests, where they are particularly common on burned or disturbed sites. Endorhizal fungi associated with Equisetum may have broad ecological relevance.  相似文献

Abstract The dissimilatory nitrate-reducing processes, denitrification, and dissimilatory nitrate-reduction to ammonium were studied in freshwater lake sediments within healthy and degrading Phragmites australis (reed) stands. The samples from the healthy vegetation site contained roots and rhizomes. Cores were supplied with 1.9–5.2 μg 15N-NO3 g−1 dry sediment in the laboratory and subsequently incubated for 8 h at 20°C, in the dark. The 15N compounds were determined before (natural percentage of 15N) and after 1 and 8 h of incubation. The uptake of 15N by the roots and rhizomes in the healthy vegetation was 61%. Nitrogen losses, interpreted as denitrification, accounted for 25 and 84% of the added 15N-NO3 in sediment from the healthy and degrading vegetation sites, respectively. The percentages of nitrate reduced to ammonium were 4 and 9% in sediment from the healthy vegetation and degrading vegetation sites, respectively. The percentage of 15N–total N in the sediment of the healthy vegetation site was 10%, whereas for the degrading vegetation site this percentage was 7%. The percentage of nitrate reduced to ammonium could be potentially underestimated by the percentage of 15N measured in the sediment. In this case, in healthy and degenerating P. australis stands, the percentage of produced ammonium accounted for 14–16%. The nitrate reduction rates were calculated based on an incubation period of one hour. The denitrification rate in sediment from the degrading vegetation site was higher than from the healthy vegetation site. The rate of dissimilatory nitrate reduction to ammonium was almost tenfold higher in sediment from the degrading vegetation site compared to sediment from the healthy vegetation site. The significantly lower percentages of dissimilatory nitrate reduction to ammonium and denitrification in the healthy stand compared to the degrading stand was probably due to the presence of roots and rhizomes. To investigate the biocontrol effectiveness of the antibiotic producing bacterium, Pseudomonas aureofaciens 63–28 against the phytopathogen Rhizoctonia solani AG-4 on Petri plates and in soybean roots, growth response and induction of PR-proteins were estimated after inoculation with P. aureofaciens 63–28 (P), with R. solani AG-4 (R), or with P. aureofaciens 63–28 + R. solani AG-4 (P + R). P. aureofaciens 63–28 showed strong antifungal activity against R. solani AG-4 pathogens in Petri plates. Treatment with P. aureofaciens 63–28 alone increased the emergence rate, shoot fresh weight, shoot dry weight and root fresh weight at 7 days after inoculation, when compared to R. solani AG-4; P + R treatment showed similar effects. Peroxidase (POD) and β-1,3-glucanase activity of P. aureofaciens 63–28 treated roots increased by 41.1 and 49.9%, respectively, compared to control roots. POD was 26% greater in P + R treated roots than R. solani treated roots. Two POD isozymes (59 and 27 kDa) were strongly induced in P + R treated roots. The apparent molecular weight of chitinase from treated roots, as determined through SDS-PAGE separation and comparison with standards, was about 29 kDa. Five β-1,3-glucanase isozymes (80, 70, 50, 46 and 19 kDa) were observed in all treatments. These results suggest that inoculation of soybean plants with P. aureofaciens 63–28 elevates plant growth inhibition by R. solani AG-4 and activates PR-proteins, potentially through induction of systemic resistance mechanisms.  相似文献

The aim of this study was to investigate the effects of NaCl-salinity on the physiological attributes in common reed, Phragmites australis (Cav.) Trin. ex Steudel. Plants grew optimally under salinity treatment with standard nutrient solution without added salt and at NaCl concentrations up to 100 mM. Applied for 21 days, NaCl-salinity (300 and 500 mM) caused a significant reduction in growth allocation of all different tissues of P. australis. Shoot growth of reed plants displayed a highly significant correlation with plant–water relations and photosynthetic parameters. The net photosynthetic rate and stomatal conductance of reed plants treated with NaCl-salinity at varying osmotic potential (ψπ) of nutrient solutions were positively correlated, and the former variable also had a strong positive relationship with transpiration rate. Leaf water potential and ψπ followed similar trends and declined significantly as ψπ of watering solutions was lowered. The increase in total inorganic nutrients resulting from increased Na+ and Cl in all tissues and K+, Ca2+ and Mg2+ concentrations were maintained even at the most extreme salt concentration. Common reed exhibited high K+/Na+ and Ca2+/Na+ selectivity ratios over a wide range of salinities under NaCl-salinity. These findings suggest that reed plants were able to adapt well to high salinities by lowering their leaf ψπ and the adjustment of osmotically active solutes in the leaves.  相似文献

The aim of this study was to assess the diagnostic properties of the two selective plating media and a chromogenic medium for identification of Bacillus cereus. The 324 isolates were B. cereus (37%), Bacillus weihenstephanensis (45%) or Bacillus thuringiensis (18%), as identified by a new combination of techniques. All isolates were growing on mannitol–egg yolk–polymyxin agar (MYP), and they did not form acid from mannitol. However, a significant lower number of B. thuringiensis isolates did not show lecithinase activity. All isolates were also growing on polymyxin–egg yolk–mannitol–bromothymol blue agar (PEMBA); however, 11% isolates indicated that they did produce acid from mannitol, and 15% isolates did not show any lecithinase activity. Five of the isolates did not grow at all on the chromogenic agar, and 14 of the growing isolates were β-glucosidase negative. It is concluded that the two recommended selective plating media MYP and PEMBA for detection of B. cereus group bacteria both have their limitations for identification of some B. cereus, B. weihenstephanensis or B. thuringiensis. However, MYP is preferable compared to PEMBA. The chromogenic medium has its own advantages and limitations, and some of the limitations seem to be solved by incubation at 30°C instead of the recommended 37°C.  相似文献

Three novel endophytic rhizobial strains (RRE3, RRE5, and RRE6) were isolated from naturally growing surface-sterilized rice roots. These isolates had the ability to nodulate common bean (Phaseolus vulgaris). Polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) and sequencing of 16S rDNA of these isolates revealed that RRE3 and RRE5 are phylogenetically very close to Burkholderia cepacia complex, whereas RRE6 has affinity with Rhizobium leguminosarum bv. phaseoli. Plant infection test using gusA reporter gene–tagged construct of these isolates indicated that bacterial cells can go inside and colonize the rice root interiors. A significant increase in biomass and grain yield was also recorded in greenhouse-grown rice plants inoculated with these isolates.  相似文献

Enrichment-based methods targeted at uranium-tolerant populations among the culturable, aerobic, chemo-heterotrophic bacteria from the subsurface soils of Domiasiat (India’s largest sandstone-type uranium deposits, containing an average ore grade of 0.1 % U3O8), indicated a wide occurrence of Serratia marcescens. Five representative S. marcescens isolates were characterized by a polyphasic taxonomic approach. The phylogenetic analyses of 16S rRNA gene sequences showed their relatedness to S. marcescens ATCC 13880 (≥99.4% similarity). Biochemical characteristics and random amplified polymorphic DNA profiles revealed significant differences among the representative isolates and the type strain as well. The minimum inhibitory concentration for uranium U(VI) exhibited by these natural isolates was found to range from 3.5–4.0 mM. On evaluation for their uranyl adsorption properties, it was found that all these isolates were able to remove nearly 90–92% (21–22 mg/L) and 60–70% (285–335 mg/L) of U(VI) on being challenged with 100 μM (23.8 mg/L) and 2 mM (476 mg/L) uranyl nitrate solutions, respectively, at pH 3.5 within 10 min of exposure. his capacity was retained by the isolates even after 24 h of incubation. Viability tests confirmed the tolerance of these isolates to toxic concentrations of soluble uranium U(VI) at pH 3.5. This is among the first studies to report uranium-tolerant aerobic chemoheterotrophs obtained from the pristine uranium ore-bearing site of Domiasiat.  相似文献

Embryogenic cultures of the common reed [Phragmites australis (Cav.) Trin. Ex. Steud.] were induced on Murashige and Skoog (1962)-based medium with 2% (w/v) sucrose, B5 vitamins and 4.5 μM 2,4-dichlorphenoxyacetic acid. Four independent culture lines, two initiated from stem nodes and two from roots, were established. These cultures underwent somatic embryogenesis. In one line of stem node origin, the somatic embryos germinated and developed into plants, following transfer of embryogenic cultures to Murashige and Skoog (1962)-based medium lacking growth regulators, with 108 ± 17 plants being recovered per 100 mg fresh weight of culture. In other lines, the somatic embryos developed roots, but not shoots. Shoot regeneration via somatic embryogenesis offers potential as anin vitro system for physiological studies, including assessments of the response of common reed to environmental pollutants. In a survey conducted in 1999–2001, the carriage of thermotolerant Campylobacter s in cattle was investigated, and the genetic diversity of C. jejuni within one herd was examined and compared with human isolates. C. jejuni, C. coli and other thermotolerant Campylobacter spp. were isolated from intestinal contents from 26%, 3% and 2% of 804 cattle, respectively. The carriage rate was higher in calves (46%) than in adults (29%). Twenty-nine C. jejuni isolates from one herd and 31 human isolates from the study area were genotyped with amplified-fragment length polymorphism (AFLP). Eighty-three % of the bovine isolates fell into three distinct clusters with 95–100% similarity, persistent in the herd for 5–10 months. Among human isolates, 58% showed >90% similarity with bovine isolates. The results show that cattle are a significant and stable reservoir for C. jejuni in the study area. Transmission between individuals within the herd may be sufficient to maintain a steady C. jejuni population independent of environmental influx. The results of this study have provided new information on C. jejuni and C. coli transmission, and also on the carriage in cattle, genotypes stability and similarity between bovine and human isolates.  相似文献

We examined the effects of genetic transformation by Agrobacterium rhizogenes on the production of tylophorine, a phenanthroindolizidine alkaloid, in the Indian medicinal plant, Tylophora indica. Transformed roots induced by the bacterium grew in axenic culture and produced shoots or embryogenic calli in the absence of hormone treatments. However, hormonal treatment was required to regenerate shoots in root explants of wild type control plants. Transformed plants showed morphological features typically seen in transgenic plants produced by A. rhizogenes, which include, short internodes, small and wrinkled leaves, more branches and numerous plagiotropic roots. Plants regenerated from transformed roots showed increased biomass accumulation (350–510% in the roots and 200–320% in the whole plants) and augmented tylophorine content (20–60%) in the shoots, resulting in a 160–280% increase in tylophorine production in different clones grown in vitro.  相似文献

Phragmites mauritianus is the dominant herbaceous species colonizing riverine habitats in the Kruger National Park, South Africa. These perennial systems are characterized by high annual and seasonal flow variability and a complex mosaic of patches of reeds, sand, water, rock and other vegetation. Patterns of increase and decrease in reed cover in the Letaba River were determined from aerial photographs covering a 54-year period. An initial period of reed expansion (1942–1965) was followed by a period of reed loss (1965–1977) and subsequent gradual re-establishment (1977–1996). A spatially explicit analysis of changes in reed distribution over an 8-year period (1988–1996) showed that patches of reed vegetation are, in the short term, highly dynamic elements within the river landscape. Analyzing short-term, small-scale change provides information which is not obtainable from long-term, large-scale studies. We propose that causes of reed expansion or decline cannot be determined without an understanding of both long- and short-term patterns of change.  相似文献
