Cell surface antigens of normal and anemic (WW) mouse erythroid cells have been examined in cytotoxicity assays with two rat antisera. When tested on fetal liver cells, a rat anti-erythroblast serum recognized antigen(s) present on erythroid cells early in development, while rat anti-adult red blood cell serum recognized antigen(s) present on mature erythroid cells. Each of these sera had different activity on normal (+/+ or W+) as compared to anemic (WW) erythroid cells.  相似文献

The small intestines of 31 mature crossbred gilts were infused with Escherichiacoli endotoxin through permanent jejunostomies. A total of 86 infusions were done. The endotoxin was absorbed after 4 infusions. Evidence of absorption included variations in body temperatures, decline in white blood cell counts, detection of the endotoxin in circulating blood plasma samples by a limulus amebocyte lysate gelation test, and abnormal clinical signs. E.coli endotoxin placed in the small intestines of normal, healthy, mature gilts does not readily cause illness.  相似文献

Trimethyloxonium ion inactivates acetylcholinesterase from the electric eel and acetylcholinesterase on the surface of human red blood cells. Tetramethylammonium ion, which is a competitive inhibitor of acetylcholinesterase, protects against this inactivation. Trimethyloxonium ion does not inactivate the system that transports choline into the red blood cell. We conclude that trimethyloxonium ion is an affinity-labeling reagent for acetylcholinesterase and that red blood cell acetylcholinesterase is probably not a component of the choline transport system.  相似文献

Depletion of energy stores of human red cells decreases the maximum transport capacity, Jm, for glucose transport to a value one-third or less of that found in red cells from freshly drawn blood. There is no change in Km. Hemolysis and resealing of red cells with ATP or ADP reverses the decrease in Jm. The maximum effect occurs at concentrations of ATP in the normal range for red cells, however, there is little effect from ADP concentrations in its normal range in freshly drawn red cells. Hemolysis and resealing with ATP gives an increase in Jm and an increase in differential labeling by photolytic labeling with tritiated cytochalasin B. Most of the activation is lost after a second hemolysis-reseal without ATP but about 25% of the activation remains.  相似文献

Trichloroacetic acid extracts of red cell often produce an iron-ATP complex after ion exchange chromatography of the extract amounting to about 13 of the total ATP. In the present work the presence of 14–50% of iron-ATP in such extracts from human and Rhesus monkey blood has been shown.Experiments designed to clarify the possible role and origin of iron-ATP revealed that non-acid treatment of human blood or red cells, as in the freeze-thaw process, followed by separation on a Sephadex column did not produce an iron-ATP fraction. In addition, purified hemoglobin and ATP were combined and incubated at pH 7.4. After Sephadex chromatography, there was no evidence of an iron-ATP fraction. However, similar combinations of incubated hemoglobin and ATP treated with trichloroacetic and separated by ion exchange chromatography did produce an iron-ATP fraction similar to that obtained from acid-extracted blood.It appears that iron-ATP in quantities found in acid-extracted blood is the result of iron release from hemoglobin and the subsequent complexing of such iron with available ATP.  相似文献

Adjuvant induced polyarthritis is accompanied by severe histopathological and biochemical changes. It was learned from this study that red blood cells from arthritic rats exhibit an enhanced resistance to lysis. This obtained whether lysis was provoked by hypotinicity, heat or mechanical means. Plasma protein alterations, while prominent, did not appear to influence the observed change. The onset of the change was delayed and required relatively large amounts of M. butyricum.  相似文献

Monoamine oxidase, an intrinsic protein of outer mitochondrial membrane, was purified from bovine liver and rabbit antibody against the enzyme was prepared. The antibody could react with the monoamine oxidase of rat liver mitochondria. When rat liver RNA was translated invitro using rabbit reticulocyte lysate and monoamine oxidase peptide in the translation products was immunoprecipitated by the antibody, the peptide was detected in the products programmed by the messenger RNA's from total and free polysomes but not that from bound polysomes. The enzyme synthesized invitro had the same apparent molecular size as the mature protein in outer mitochondrial membrane.  相似文献

It has been suggested that the human red cell anion transport protein, band 3, is the site not only of the cation leak induced in human red cells by treatment with the sulfhydryl reagent pCMBS (p-chloromercuribenzene sulfonate) but is also the site for the inhibition of water flux induced by the same reagent. Our experiments indicate that N-ethylmaleimide, a sulfhydryl reagent that does not inhibit water transport, also does not induce a cation leak. We have found that the profile of inhibition of water transport by mercurial sulfhydryl reagents is closely mirrored by the effect of these same reagents on the induction of the cation leak. In order to determine whether these effects are caused by band 3 we have reconstituted phosphatidylcholine vesicles containing only purified band 3. Control experiments indicate that these band 3 vesicles do not contain (Na+ + K+)-ATPase as measured by ATP dephosphorylation. pCMBS treatment caused a significant increase in the cation leak in this preparation, consistent with the view that the pCMBS-induced cation leak in whole red cells is mediated by band 3.  相似文献

Brief exposure to the protein neurotoxin, β-bungarotoxin, is known to disrupt neuromuscular transmission irreversibly by blocking the release of transmitter from the nerve terminal. This neurotoxin also has a phospholipase A2 activity, although phospholipases in general are not very toxic. To determine if the toxicity of this molecule might result from specific binding to neural tissue, we have looked for high affinity, saturable binding using 125I-labeled toxin. At low membrane protein concentration 125I-labeled toxin binding was directly proportional to the amount of membrane; at fixed membrane concentration 125I-labeled toxin showed saturable binding. It was unlikely that iodination markedly changed the toxin's properties since the iodinated toxin had a comparable binding affinity to that of native toxin as judged by competition experiments. Comparison of toxin binding to brain, liver and red blood cell membranes showed that all had high affinity binding sites with dissociation constants between one and two nanomolar. This is comparable to the concentrations previously shown to inhibit mitochondrial function. However, the density of these sites showed marked variation such that the density of sites was 13.0 pmol/mg protein for a brain membrane preparation, 2.4 pmol/mg for liver and 0.25 pmol/mg for red blood cell membranes.In earlier work we had shown that calcium uptake by brain mitochondria is inhibited at much lower toxin concentrations than is liver mitochondrial uptake. Both liver and brain mitochondria bind toxin specifically, but the density of 125I-labeled toxin binding sites on brain mitochondrial prepartions (3.3 ± 0.3 pmol/mg) exceeded by a factor of ten the density on liver mitochondrial preparations (0.3 ± 0.05 pmol/mg). It is also shown that the labeled toxin does not cross synaptosomal membranes, suggesting that mitochondria may not be the site of action of the toxin in vivo. We conclude the β-bungarotoxin is an enzyme which can bind specifically with high affinity to cell membranes.  相似文献

We report the isolation and some properties of a flavodoxin from a eukaryotic organism, the naturally occurring red alga Chondrus crispus. Unlike the situation with most other organisms the flavodoxin, under normal growth conditions, is the predominantly formed low-potential electron carrier, an accompanying ferredoxin occurring in only very small amounts. The flavodoxin is of molecular weight 21000 and one mole of FMN is present per mole of protein. Reduction of the flavoprotein proceeds via a blue flavosemiquinone radical. The flavodoxin is active both in photosynthetic NADP reduction by broken chloroplasts, and in phosphoroclastic cleavage of pyruvate by cell-free extracts of Clostridium pasteurianum.  相似文献

Eleven cyclic Hereford heifers were synchronized to estrus with a commercial luteolytic agent. Eight heifers had been inoculated with Anaplasmamarginale. Six of the eight inoculated heifers became anemic, i.e. packed cell volume, red blood cell counts and hemoglobin decreased. White blood cell counts were elevated during severe anemia. Anemic heifers did not show behavioral estrus or ovulate, and during this stage, serum concentrations of progesterone and estradiol-17β were significantly depressed. Mature beef heifers that have acute anaplasmosis suffer from inhibited ovarian function and anestrus.  相似文献

The presence or absence of calcium ions during the isolation of nuclei from SV40-infected African green monkey kidney cells significantly affects the size of SV40 DNA synthesized in vitro. When Ca++ is present during the nuclear isolation procedure, the 3–7S fragments of SV40 DNA synthesized in vitro mature into long chains; in the absence of Ca++ they do not.  相似文献

Previous studies have shown that the assembly of the precursor shell (prohead) of bacteriophage P22 requires the copolymerization of the gene 5 coat protein with the gene 8 scaffolding protein. Removal of the scaffolding protein by mutation prevents efficient coat protein assembly, but some aberrant particles do form. We have now isolated these structures and characterized them with respect to morphology, protein composition, and small-angle X-ray scattering properties.The aberrant particles fall into three morphological classes, i.e. complex spirals and closed shells of two sizes. Small-angle X-ray scattering studies confirm that the larger particles are hollow shells with the radius of proheads (r = 260 A?), and not of the mature virus (r = 285 A?). These structures lack the inner shell of scaffolding protein found in proheads. The small particles have a radius of 195 Å, smaller than proheads, and appear to contain material, not scaffolding protein, within the outer shell.The aberrant particles contain two minor protein species, the gene 9 tail-spike protein, and an unidentified 67,000 molecular weight polypeptide, probably from the host. Neither is found in normal proheads. Removal of gene.9 product by mutation did not affect the formation of the aggregates. Fractionation of the morphological classes of particles revealed that the 67,000 molecular weight band was associated with the closed shells. It may be serving as a pseudo-initiator.Earlier studies had shown that treatment of proheads with sodium dodecyl sulfate in vitro resulted in loss of the scaffolding protein, and expansion of the shell to the mature radius of 285 Å. When the 8? prohead-sized shells were treated similarly, they also expanded to the mature-sized shell. These results support the idea that there are at least two stable states of the coat protein, one of which, the prohead form, is an obligatory precursor of the mature form.  相似文献

31P NMR and column fractionation studies do not substantiate the existence in solution of a complex of phosphoglycerate kinase and phosphoglycerate mutase or of one involving these enzymes and glyceraldehyde-3-phosphate dehydrogenase. It is shown that the small shifts (<3.2Hz) in the 31P resonances of 2,3-bisphosphoglycerate which were interpreted to indicate the existence of such complexes (Fossel and Solomon (1977) BBA 464, 82–92) probably result from very small variations in pH (<0.1 unit). Further, no significant resonance shifts are detected in the presence of ouabain in glucose-depleted human red blood cells. An error analysis of the NMR data indicates that previously reported ouabain-induced shifts are within the noise level of the measurement and do not indicate the presence of enzyme complexes in the red cell.  相似文献

Cytoplasmic free and bound polysomes were isolated from bovine adrenal cortex, and used to program invitro protein synthesis in rat liver cell sap and wheat germ lysate systems. Synthesis of adrenodoxin(Ad) and adrenodoxin reductase(AdR) in the cell-free systems was determined by immunoprecipitation using monospecific antibodies, and the sizes of the invitro products were analyzed by SDS-polyacrylamide gel electrophoresis. Ad was synthesized by both free and bound polysomes as a putative large precursor having molecular weight of approximately 20,000 daltons, which was processed to mature size Ad (MW 12,000 daltons) by invitro incubation with adrenal cortex mitochondria. On the other hand, AdR was synthesized only by free polysomes apparently as the mature size product.  相似文献

Substances with the human blood group Sda character occur on red cells and are also present in secretions. In urine Sda activity is associated with the Tamm and Horsfall (T-H) glycoprotein. About 4% of Caucasian individuals, whose red cells are Sda negative, have a T-H glycoprotein that is without Sda activity. The two immunologically distinct forms of T-H glycoprotein have almost identical qualitative and quantitative amino acid and carbohydrate compositions. The only exception is the N-acetylgalactosamine content which falls in the range of 1–2% for preparations from Sd(a+) individuals whereas the level is negligible in the Sda inactive preparations. These results strongly indicate that N-acetylgalactosamine makes an important contribution to the Sda determinant structure in the T-H glycoprotein.  相似文献

Compound 4880, a condensation product of N-methyl-p-methoxyphenethylamine with formaldehyde, is composed of a family of cationic amphiphiles differing in the degree of polymerization. Compound 4880 was found to be a potent inhibitor of the calmodulin-activated fraction of brain phosphodiesterase and red blood cell Ca2+-transport ATPase, with IC50 values of 0.3 and 0.85 μg/ml, respectively. However, the basal activity of both enzymes is not at all suppressed by the drug at concentrations up to 300 μg/ml. Inhibition of Ca2+ transport into inside-out red blood cell vesicles by compound 4880 follows a similar pattern in that basal, calmodulin-independent, transport is also not affected by the drug. Kinetic analysis revealed that the stimulation of Ca2+-transport ATPase induced by calmodulin is inhibited by compound 4880 according to a competitive mechanism. It was demonstrated that the inhibitory constituents of compound 4880 bind to calmodulin in a Ca2+-dependent fashion. Comparison of the specificity of several anti-calmodulin drugs showed that compound 4880 is the most specific inhibitor of the calmodulin-dependent fraction of red blood cell Ca2+-transport ATPase that has been described hitherto. In addition, compound 4880 was found to be a rather specific inhibitor of the calmodulin-induced activation of Ca2+-transport ATPase when compared with the stimulation induced by an anionic amphiphile or by limited proteolysis. Half-maximal inhibition of the activity stimulated by oleic acid or mild tryptic digestion required 8- and 32-times higher concentrations of compound 4880, respectively, compared with the calmodulin-dependent fraction of the ATPase activity. Moreover, calmodulin-independent systems as rabbit skeletal muscle sarcoplasmic reticulum Ca2+-transport ATPase or calf cardiac sarcolemma (Na+ + K+)-transport ATPase are far less influenced by compound 4880 as compared with trifluoperazine and calmidazolium. Because of its high specificity compound 4880 is proposed to be a promising tool for studying calmodulin-dependent processes.  相似文献

This paper presents the first unambiguous demonstration that a unique protein isolated from the hemolysate of human erythrocytes is responsible for increasing both the apparent Ca2+ ion affinity and maximum rate of ATP hydrolysis of the membrane-bound Ca2+Mg2+ ATPase. Unlike previous reports where an unpurified extract from red blood cells was used to activate the ATPase, our results clearly demonstrate that a single protein species, whether initially associated with or added back to the membrane is responsible for the observed changes in ATPase activity.  相似文献

When human red cells are treated with the mercurial sulfhydryl reagent, p-chloromercuribenzene sulfonate, osmotic water permeability is suppressed and only diffusional water permeability remains (Macey, R.I. and Farmer, R.E.L. (1970) Biochim. Biophys. Acta 211, 104–106). It has been suggested that the route for the remaining water permeation is by diffusion through the membrane lipids. However, after making allowance for the relative lipid area of the membrane, the water diffusion coefficient through lipid bilayers which contain cholesterol is too small by a factor of two or more. We have measured the permeability coefficient of normal human red cells by proton T1 NMR and obtained a value of 4.0 · 10?3 cm · s?1, in good agreement with published values. In order to study permeation-through red cell lipids we have perturbed extracted red cell lipids with the lipophilic anesthetic, halothane, and found that halothane increases water permeability. The same concentration of halothane has no effect on the water permeability of human red cells, after maximal pCMBS inhibition. In order to compare halothane mobility in extracted red cell membrane lipids with that in red cell ghost membranes, we have studied halothane quenching of N-phenyl-1-naphthylamine by equilibrium fluorescence and fluorescence lifetime methods. Since halothane mobility is similar in these two preparations, we have concluded that the primary route of water diffusion in pCMBS-treated red cells is not through membrane lipids, but rather through a membrane protein channel.  相似文献
