cDNA encoding the bound type trehalase of the European honeybee was cloned. The cDNA (3,001 bp) contained the long 5′ untranslated region (UTR) of 869 bp, and the 3′ UTR of 251 bp including a poly(A) tail, and the open reading frame of 1,881 bp consisting of 626 amino acid residues. The M r of the mature enzyme comprised of 591 amino acids, excluded a signal sequence of 35 amino acid residues, was 69,177. Six peptide sequences analyzed were all found in the deduced amino acid sequence. The amino acid sequence exhibited high identity with trehalases belonging to glycoside hydrolase family 37. A putative transmembrane region similar to trehalase-2 of the silkworm was found in the C-terminal amino acid sequence. Recombinant enzyme of the trehalase was expressed in the methylotrophic yeast Pichia pastoris as host, and displayed properties identical to those of the native enzyme except for higher sugar chain contents. Specialized castes are considered a key reason for the evolutionary and ecological success of the social insect lifestyle. The most essential caste distinction is between the fertile queen and the sterile workers. Honeybee (Apis mellifera) workers and queens are not genetically distinct, rather these different phenotypes are the result of epigenetically regulated divergent developmental pathways. This is an important phenomenon in understanding the evolution of social insect societies. Here, we studied the genomic regulation of the worker and queen developmental pathways, and the robustness of the pathways by transplanting eggs or young larvae to queen cells. Queens could be successfully reared from worker larvae transplanted up to 3 days age, but queens reared from older worker larvae had decreased queen body size and weight compared with queens from transplanted eggs. Gene expression analysis showed that queens raised from worker larvae differed from queens raised from eggs in the expression of genes involved in the immune system, caste differentiation, body development and longevity. DNA methylation levels were also higher in 3‐day‐old queen larvae raised from worker larvae compared with that raised from transplanted eggs identifying a possible mechanism stabilizing the two developmental paths. We propose that environmental (nutrition and space) changes induced by the commercial rearing practice result in a suboptimal queen phenotype via epigenetic processes, which may potentially contribute to the evolution of queen–worker dimorphism. Abstract. For many years, two opposing theories have dominated our ideas of what honeybees see. The earliest proposal based on training experiments was that bees detected only simple attributes or features, irrespective of the actual pattern. The features demonstrated experimentally before 1940 were the disruption of the pattern (related to spatial frequency), the area of black or colour, the length of edge, and the angle of orientation of a bar or grating. Cues discovered recently are the range, and radial and tangential edges, and symmetry, relative to the fixation point, which is usually the reward hole. This theory could not explain why recognition failed when the pattern was moved. In the second theory, proposed in 1969, the bee detected the retinotopic directions of black or coloured areas, and estimated the areas of overlap and nonoverlap on each test pattern with the corresponding positions in the training pattern. This proposal explained the progressive loss of recognition as a test pattern was moved or reduced in size, but required that the bees saw and remembered the layout of every learned pattern and calculated the mismatch with each test image. Even so, the same measure of the mismatch was given by many test patterns and could not detect a pattern uniquely. Moreover, this theory could not explain the abundant evidence of simple feature detectors. Recent work has shown that bees learn one or more of a limited number of simple cues. A newly discovered cue is the position, mainly in the vertical direction, of the common centre (centroid) of black areas combined together. Significantly, however, the trained bees look for the cues mentioned above only in the range of places where they had occurred during the training. These two observations made possible a synthesis of both theories. There is no experimental evidence that the bees detect or re-assemble the layout of patterns in space; instead, they look for a cue in the expected place. With an array of detectors of the known cues, together with their directions, this mechanism would enable bees to recognize each familiar place from the coincidences of cues in different directions around the head. 相似文献

1. The honeybee Apis mellifera is of huge worldwide economic importance in the pollination of crops for human consumption. In recent years, honeybee populations have declined under pressure from diseases and pests. Climate change is increasingly being viewed as an additional threat to honeybees and yet only limited research has been carried out in this area. 2. This paper reports the advance of the first cleansing flight (‘spring cleaning’) of the honeybee in Poznań, Poland, i.e. flights to excrete faeces, over a month in the period 1985–2009. The timing of this flight is advanced not only by higher late winter/spring temperatures but also by higher temperatures in the previous summer and autumn. 3. This earlier activity gives hope that the reported earlier flowering of many native and cultivated species will not cause a pollination synchrony crisis. 相似文献

利用Bac to Bac系统将意大利蜜蜂蜂毒磷脂酶A2(AmPLA2)基因cDNA克隆至转移载体pFastBacHTa中,得到pBacHT-AmPLA2,再将其转化入含穿梭载体Bacmid的受体大肠杆菌DH10Bac中,通过转座作用,得到含AmPLA2基因的重组病毒rBacmid-AmPLA2的DNA。提取其基因组DNA,用脂质体介导转染粉纹夜蛾细胞Tn-5B1-4,得到重组病毒rACV-Bac-AmPLA2。用此重组病毒感染Tn-5B1-4细胞, 在细胞中表达AmPLA2。SDS-PAGE电泳结果显示,与6×His Tag融合表达的产物蛋白分子量约为18 kD左右,表达量约占细胞总蛋白的5.35%。Western blot印迹显示,融合表达产物能与意大利蜜蜂蜂毒AmPLA2抗血清发生免疫反应。生物活性测定显示,含表达产物的细胞蛋白粗提物对底物蛋黄的酶活力约为6.13 μmol·min-1·mg-1。 相似文献

Until recently, African and European subspecies of the honeybee (Apis mellifera L.) had been geographically separated for around 10,000 years. However, human-assisted introductions have caused the mixing of large populations of African and European subspecies in South and Central America, permitting an unprecedented opportunity to study a large-scale hybridization event using molecular analyses. We obtained reference populations from Europe, Africa, and South America and used these to provide baseline information for a microsatellite and mitochondrial analysis of the process of Africanization of the bees of the Yucatan Peninsula, Mexico. The genetic structure of the Yucatecan population has changed dramatically over time. The pre-Africanized Yucatecan population (1985) comprised bees that were most similar to samples from southeastern Europe and northern and western Europe. Three years after the arrival of Africanized bees (1989), substantial paternal gene flow had occurred from feral Africanized drones into the resident European population, but maternal gene flow from the invading Africanized population into the local population was negligible. However by 1998, there was a radical shift with both African nuclear alleles (65%) and African-derived mitochondria (61%) dominating the genomes of domestic colonies. We suggest that although European mitochondria may eventually be driven to extinction in the feral population, stable introgression of European nuclear alleles has occurred. 相似文献

Seminoma constitutes one subtype of human testicular germ cell tumors and is uniformly composed of cells that are morphologically similar to the primordial germ cells and/or the cells in the carcinoma in situ. We performed a genome-wide exploration of the genes that are specifically up-regulated in seminoma by oligonucleotide-based microarray analysis. This revealed 106 genes that are significantly and consistently up-regulated in the seminomas compared to the adjacent normal tissues of the testes. The microarray data were validated by semi-quantitative RT-PCR analysis. Of the 106 genes, 42 mapped to a small number of specific chromosomal regions, namely, 1q21, 2p23, 6p21-22, 7p14-15, 12pll, 12p13, 12q13-14 and 22q12-13. This list of up-regulated genes may be useful in identifying the causative oncogene(s) and/or the origin of seminoma. Furthermore, immunohistochemical analysis revealed that the seminoma cells specifically expressed the six gene products that were selected randomly from the list. These proteins include CCND2 and DNMT3A and may be useful as molecular pathological markers of seminoma. 相似文献

Honeybees (Apis mellifera L.) sampled at sites in Europe, Africa and South America were analysed using a mitochondrial DNA restriction fragment length polymorphism (RFLP) marker. These samples were used to provide baseline information for a detailed analysis of the process of Africanization of bees from the neotropical Yucatan peninsula of Mexico. Radical changes in mitochondrial haplotype (mitotype) frequencies were found to have occurred in the 13-year period studied. Prior to the arrival of Africanized bees (1986) the original inhabitants of the Yucatan peninsula appear to have been essentially of southeastern European origin with a smaller proportion having northwestern European ancestry. Three years after the migration of Africanized bees into the area (1989), only very low levels of maternal gene flow from Africanized populations into the resident European populations had occurred. By 1998, however, there was a sizeable increase in the proportion of African mitotypes in domestic populations (61%) with feral populations having 87% of mitotypes classified as African derived. The results suggest that the early stages of Africanization did not involve a rapid replacement of European with African mitotypes and that earlier studies probably overestimated the prevalence of African mitotypes. 相似文献

东方蜜蜂微孢子虫Nosema ceranae侵染成年蜜蜂导致蜜蜂微孢子虫病。本研究旨在验证东方蜜蜂微孢子虫nce-miR-12220的存在和表达,并检测nce-miR-12220及其靶基因在病原侵染意大利蜜蜂Apis mellifera ligustica (简称意蜂)工蜂过程的表达谱。Stem-loop RT-PCR和Sanger测序结果显示nce-miR-12220真实存在和表达。靶向预测结果显示nce-miR-12220共靶向KRAB-A和γ tubulin等15个基因。上述靶基因可注释到19个GO条目和3条KEGG通路。RT-qPCR结果显示,相较于接种后1 d (1 day post infection,1 dpi),nce-miR-12220在2 dpi上调表达,而在3-12 dpi阶段总体表现出显著下调表达的趋势。类似地,与1 dpi相比,靶基因KRAB-A在2 dpi上调表达,而在3-12 dpi阶段总体呈下调表达的趋势。另外,与1 dpi相比,靶基因γ tubulin在2-12 dpi阶段总体表现出显著下调表达的趋势。上述结果表明nce-miR-12220与KRAB-A和γ tubulin之间存在潜在的靶向结合和正向调控关系;东方蜜蜂微孢子虫通过下调表达nce-miR-12220抑制KRAB-A的表达进而促进增殖;意蜂工蜂可能通过抑制东方蜜蜂微孢子虫的γ tubulin表达抵御病原侵染。研究结果明确了nce-miR-12220及其靶基因KRAB-A和γ tubulin在东方蜜蜂微孢子虫侵染意蜂工蜂过程中的动态表达规律,为深入探究nce-miR-12220在病原侵染中的功能及调控机制提供了理论和实验依据。 相似文献

10-羟基-2-癸烯酸(10-HDA)是蜂王浆中的主要脂肪酸成分,具有抗菌、抗癌、延缓衰老等多种生理活性,但目前关于10-HDA生物合成的分子机制还不清楚。通过克隆蜜蜂NADPH-细胞色素P450还原酶(EC 1.6.2.4,NADPH-cytochrome P450 reductase,CPR),在大肠杆菌中异源表达,并对其酶学特性进行分析。结果表明重组菌经IPTG诱导后表达蛋白的分子量与预期一致,为86.29 kDa,Ni-NTA亲和纯化后测得其比活性为77.33(EU of CPR)/μg。酶学性质分析结果表明蜜蜂CPR酶最适温度与pH分别为40℃和8.0,并对一些金属离子及有机溶剂具有不同程度的耐受性。其对底物细胞色素C的动力学参数Km和kcat分别为76 μM和268/min。以上研究为探究CPR在10-HDA生物合成途径中的功能奠定理论基础。 相似文献

昆虫气味结合蛋白(OBPs)是脂溶性化学物质的载体,参与了嗅觉识别和化学传输等生理过程.本研究利用MEME在线软件对意大利蜜蜂Apis mellifera ligustica OBPs基因家族21条氨基酸序列进行Motif预测分析;采用RT-PCR初步筛选出在意大利蜜蜂头部表达量较高的OBPs基因,然后利用qPCR技术对筛选出的基因进一步分析其在不同日龄内勤蜂头部的表达情况.结果 表明,在预测获得的6个motif中,motif1为21个OBPs所共有,具有极高的保守性,推测该序列元件可能是意大利蜜蜂OBPs与底物结合的区域;表达谱分析结果表明,13个OBPs基因的凝胶电泳条带明显(即在头部的表达量较高),且大多数基因在4日龄时表达量达到最高值,推测这类蛋白可能参与了幼虫信息素和糖类物质的识别过程.本研究结果丰富了蜜蜂OBPs表达特性的研究数据,同时也为探讨昆虫OBPs广泛的生理功能奠定了基础. 相似文献

本研究旨在克隆鉴定西方蜜蜂Apis mellifera发育相关基因AmWnt1,分析其在不同发育时期和刚出房工蜂不同组织的表达特征,为进一步研究Wnt1基因功能提供理论参考。根据NCBI中AmWnt1基因序列信息,利用Primer 6.0设计引物,RT-PCR扩增AmWnt1基因完整的CDS序列,进行生物信息学预测,用推导的氨基酸序列构建系统进化树;利用荧光定量PCR检测该基因在卵(1日龄、2日龄和3日龄)、幼虫(1日龄、3日龄和5日龄)、预蛹(1日龄和3日龄)、蛹(0日龄、2日龄、4日龄、6日龄和8日龄)、刚出房工蜂、哺育蜂和采集蜂以及刚出房工蜂8个组织中相对表达量。克隆获得西方蜜蜂的Wnt1基因CDS序列,命名为AmWnt1,上传NCBI,获得GenBank登录号MT993937。全长1 239 bp,编码412个氨基酸,预测等电点为9.48,相对分子质量为46.40313 kDa。序列比对和系统进化树结果表明:AmWnt1蛋白与其它膜翅目昆虫聚为一类,其中和东方蜜蜂Apis cerana亲缘关系最近,序列相似度为99.50%。时空表达谱结果表明:AmWnt1基因在各个时期中均有表达,且在胚胎后期表达量最高,预蛹期和蛹前期表达量相对较高,其它时期表达量相对较低;AmWnt1基因在刚出房工蜂头、胸、触角表达量高于其它组织。AmWnt1可能参与西方蜜蜂胚胎晚期的神经系统发育和化蛹过程中的四肢发育等关键历程,为进一步研究AmWnt1功能提供了理论参考。 相似文献

RNAi is broadly used as a technique for specific gene silencing in insects but few studies have investigated the factors that can affect its efficiency. Viral infections have the potential to interfere with RNAi through their production of viral suppressors of RNAi (VSRs) and the production of viral small RNAs that can saturate and inactivate the RNAi machinery. In this study, the impact of persistent infection of the RNA viruses Flock house virus (FHV) and Macula-like virus (MLV) on RNAi efficiency was investigated in selected lepidopteran cell lines. Lepidopteran cell lines were found to be readily infected by both viruses without any apparent pathogenic effects, with the exception of Bombyx-derived Bm5 and BmN4 cells, which could not be infected by FHV. Because Sf21 cells were free from both FHV and MLV and Hi5-SF were free from FHV and only contained low levels of MLV, they were tested to evaluate the impact of the presence of the virus. Two types of RNAi reporter assays however did not detect a significant interference with gene silencing in infected Sf21 and Hi5-SF cells when compared to virus-free cells. In Hi5 cells, the presence of FHV could be easily cleared through the expression of an RNA hairpin that targets its VSR gene, confirming that the RNAi mechanism was not inhibited. Sequencing indicated that the B2 RNAi inhibitor gene of FHV and a putative VSR gene from MLV were intact in persistently infected cell lines, indicating that protection against RNAi remains essential for virus survival. It is proposed that infection levels of persistent viruses in the cell lines are too low to have an impact on RNAi efficiency in the lepidopteran cell lines and that encoded VSRs act locally at the sites of viral replication (mitochondrial membranes) without affecting the rest of the cytoplasm. 相似文献

Background
Apis mellifera and Apis cerana are two sibling species of Apidae. Apis cerana is adept at collecting sporadic nectar in mountain and forest region and exhibits stiffer hardiness and acarid resistance as a result of natural selection, whereas Apis mellifera has the advantage of producing royal jelly. To identify differentially expressed genes (DEGs) that affect the development of hypopharyngeal gland (HG) and/or the secretion of royal jelly between these two honeybee species, we performed a digital gene expression (DGE) analysis of the HGs of these two species at three developmental stages (newly emerged worker, nurse and forager).Results
Twelve DGE-tag libraries were constructed and sequenced using the total RNA extracted from the HGs of newly emerged workers, nurses, and foragers of Apis mellifera and Apis cerana. Finally, a total of 1482 genes in Apis mellifera and 1313 in Apis cerana were found to exhibit an expression difference among the three developmental stages. A total of 1417 DEGs were identified between these two species. Of these, 623, 1072, and 462 genes showed an expression difference at the newly emerged worker, nurse, and forager stages, respectively. The nurse stage exhibited the highest number of DEGs between these two species and most of these were found to be up-regulated in Apis mellifera. These results suggest that the higher yield of royal jelly in Apis mellifera may be due to the higher expression level of these DEGs.Conclusions
In this study, we investigated the DEGs between the HGs of two sibling honeybee species (Apis mellifera and Apis cerana). Our results indicated that the gene expression difference was associated with the difference in the royal jelly yield between these two species. These results provide an important clue for clarifying the mechanisms underlying hypopharyngeal gland development and the production of royal jelly.Electronic supplementary material

The recent invasion by self-replicating socially parasitic Cape honeybee workers, Apis mellifera capensis, of colonies of the neighbouring African subspecies Apis mellifera scutellata represents an opportunity to study evolution of intraspecific parasitism in real time. As honeybee workers compete pheromonally for reproductive dominance, and as A. m. capensis workers readily produce queen-like pheromones, we hypothesized that these semiochemicals promoted the evolution of intraspecific social parasitism. Remarkably, the offspring of a single worker became established as a parasite in A. m. scutellata's range. This could have resulted from extreme selection among different clonal parasitic worker lineages. Using pheromonal contest experiments, we show that the selected parasitic lineage dominates in the production of mandibular gland pheromones over all other competitors to which it is exposed. Our results suggest that mandibular gland pheromones played a key role in the evolution of intraspecific social parasitism in the honeybee and in the selection of a single genotype of parasitic workers. 相似文献

Summary Queen attendance behavior of workers from selected honey bee colonies with high and low worker retinue response to synthetic queen mandibular gland pheromone (QMP) was investigated. Antennating, licking, grooming, and feeding of the queen by workers from high and low responding colonies were examined. High and low QMP responding workers did not attend the queen differently. However, workers originating from different colonies antennated and licked the queen more frequently than others, suggesting there may be a genetic basis for queen attendance behavior not necessarily associated with response to QMP. The Bonin (Ogasawara) Islands are oceanic islands located in the northwest Pacific, and have ten native (nine endemic) bee
species, all of which are nonsocial. The European honeybee (Apis mellifera), which was introduced to the islands for apiculture in the 1880s, became naturalized in a few islands shortly after introduction.
To detect the impact of the honeybees upon native bee diversity, we analyzed pollen harvest by honeybees and surveyed the
relative abundance of honeybees and native bees on flowers on several islands. Both hived and feral honeybee colonies were
active throughout the year, harvesting pollen of both native and alien flowers and from both entomophilous and anemophilous
flowers. Honeybees strongly depended on the alien plants, especially during winter to spring when native melittophilous flowers
were rare. From June to November, honeybees exhaustively utilized native flowers, which had originally been utilized and pollinated
by native bees. On Chichi and Haha Islands, where human disturbance of forests has been severe, both native and alien flowers
were dominated by honeybees, and native bees were rare or extinct even in well-conserved forests. In contrast, on Ani Island
and Haha's satellite islands where primary forests were well conserved and honeybees were still uncommon or absent, native
bees remained dominant. These results suggest that competition for nectar and pollen of the native flowers between honeybees
and native bees favors honeybees on the disturbed islands, which are thoroughly invaded by alien nectariferous, sometimes
aggressive, weedy plants.
