Leptospira interrogans serotype pomona was isolated from the kidneys of a normal striped skunk (Mephitis mephitis hudsonicus) collected near Kindersley, Saskatchewan. Although there were no gross lesions, small foci of chronic interstitial nephritis were observed microscopically. This is the first reported isolation of serotype pomona for the prairie provinces.  相似文献

The sensitivity and specificity of the microscopic agglutination test (MAT) as a method for detection of exposure to Leptospira spp. in California sea lions (Zalophus californianus) were determined. Sera came from individuals that demonstrated clinical signs of renal disease, had lesions suggestive of leptospirosis at necropsy, and had visible leptospires in silver stained kidney sections as positive controls. Sera from unexposed captive individuals were used as negative controls. The test was 100% sensitive at 1:3,200 for confirming renal infection and 100% specific at negative < 1:100 for detection of Leptospira interrogans scrovar pomona antibodies by MAT in California sea lions. Leptospira interrogans serovar pomona was used as a screening serovar because it has been isolated previously from the kidneys and placentas of California sea lions, and there appears to be cross-reactivity between serovar pomona and other serovars. Sera from 225 free-ranging California sea lions presented to one of three participating California (USA) coastal marine mammal rehabilitation centers in 1996 were then evaluated for antibodies to serovar pomona using the MAT. The overall seroprevalence was 38.2% (86/225), although the prevalence varied among locations from 100% (38/38) in animals at the Marine Mammal Care Center (Fort MacArthur, California, USA) to 0% (0/14) at SeaWorld California (San Diego, California). At The Marine Mammal Center (Sausalito, California) [prevalence 27.8% (48/173)], the majority of seropositive animals were subadults and adults, and males were 4.7 times more likely to be seropositive to serovar pomona than females. When combining results from all three centers, subadult and adult animals were more likely to be seropositive than pups and juvenile sea lions, and the highest proportion of seropositive animals presented during the autumn months. Serum elevations of blood urea nitrogen, creatinine, phosphorus, and/or calcium were associated with seropositivity to serovar pomona. We found no association between potassium or sodium levels and seropositivity.  相似文献

Serum from volunteer subjects vaccinated with a bivalent whole cell vaccine of Leptospira interrogans serovar hardjo/serovar pomona grown in protein-free medium, was tested by the microscopic agglutination test (MAT), enzyme-immunoassay (EIA) and immunoblotting. Specific IgM antibodies to either serovars hardjo or pomona were detected in some subjects as early as 6 days after vaccination with peak antibody levels occurring 13-68 days after vaccination. Whereas all subjects produced specific IgM to both serovars, not all produced specific IgG to both serovars. Immunoblotting with hardjo sonicate revealed that all subjects produced IgM antibodies reacting with the 15, 23 and 28 kDa components of hardjo lipopolysaccharide (LPS), and most produced IgM antibodies that reacted with the 34.5 kDa flagellar doublet. In contrast, not all sera immunoblotted against pomona sonicate reacted with the 29 and 35 kDa components of pomona LPS. However all subjects produced antibodies reacting with a diffuse 14.4-27 kDa band. These antibodies appeared early in the immune response. Serum from the one vaccinated subject tested protected hamsters from acute lethal infection with serovar pomona.  相似文献

The role of the red fox (Vulpes vulpes) in the epizootiology of leptospirosis in southwestern Ontario was investigated in 1973-1974. Leptospira interrogans serovar pomona (kennewicki by DNA analysis) was isolated from the kidneys of three of eight foxes tested. Severe hemorrhagic nephritis and interstitial nephritis were common to these foxes and to five others out of nine foxes examined. Autumnalis antibodies were detected at titers 10(-2) to 10(-5) in 12 of 100 fox sera. Pomona antibodies occurred in 6% of the sera, always accompanied by autumnalis antibodies, and at titers never exceeding the autumnalis titers. Cultural, serological, and pathological findings together indicated that the red fox could have been acting as an amplifier host, but not as a maintenance host, for pomona.  相似文献

ABSTRACT Little information on foraging habitats of sympatric species of skunks in Texas, USA, is available. We compared 11 western spotted skunks (Spilogale gracilis) and 10 striped skunks (Mephitis mephitis) using radiotelemetry data to assess habitat use during foraging at broad levels of selection in a fragmented habitat. Western spotted skunks used areas with more large mesquite (Prosopis glandulosa) trees than did striped skunks and randomly selected points. Striped skunk habitat use was not different from randomly chosen locations. Contrary to previous research, both species appear to avoid agricultural habitat. A habitat management plan may be difficult to implement for striped skunks in Texas because they did not favor any available habitat. Conservation of western spotted skunks in west-central Texas should focus on areas with older mesquite trees, areas that are now often brush controlled for management of livestock.  相似文献

We assessed patterns and energetic consequences of different overwintering strategies, torpor, and social thermoregulation in the striped skunk (Mephitis mephitis) under natural ambient temperature and photoperiod. Striped skunks entered spontaneous daily torpor, with the lowest torpid body temperature (T(b)) reaching 26.0 degrees C, the lowest recorded T(b) for a carnivore. Patterns of daily torpor differed between solitary and grouped skunks: all solitary skunks regularly entered daily torpor, but only some individuals in communal dens employed torpor. When they did, it was shallow and infrequent. Solitary skunks entered torpor on average 50 times (in 120 d) compared with 6 times for grouped skunks. During torpor, solitary skunks had average minimum T(b) of 26.8 degrees C and bout duration of 7.8 h, whereas grouped skunks had average minimum T(b) of 30.9 degrees C and bout duration of 5.4 h. Torpor by solitary skunks occurred during their activity phase, but grouped skunks' shallow torpor bouts were restricted to their diurnal resting phase. On average, grouped skunks experienced lower percent daily fat loss, and they emerged in spring with higher percent body fat of 25.5%. In contrast, solitary skunks emerged in spring with only 9.3% body fat. In conclusion, the use of daily torpor and social thermoregulation in northern populations of striped skunks represent two strikingly different mechanisms to minimize energetic costs and increase individual fitness in response to unfavorable environmental conditions.  相似文献

Twenty nine skunks (Mephitis mephitis) were vaccinated orally with raccoon poxvirus (RCN) recombinants: 10 with a recombinant expressing the rabies virus glycoprotein (RCNRG), 10 with RCNRG mixed with a recombinant expressing the rabies virus nucleoprotein (RCNRN) and nine with RCN alone. Rabies virus neutralizing antibodies were detected in six of the 20 skunks; five skunks (three given RCNRG, two given a mixture of recombinants) survived a rabies challenge that was lethal for nine skunks vaccinated with RCN alone.  相似文献

Twenty-three striped skunks (Mephitis mephitis) without demonstrable antibodies in 1:25 serum dilution in the modified agglutination test (MAT) were fed sporulated Toxoplasma gondii oocysts (9 skunks) or tissue cysts (10 skunks), and 4 skunks (controls) were not fed T. gondii. Skunks were bled before feeding T. gondii, 10 and 23- 25 days postinoculation (PI). All 9 seronegative skunks fed oocysts died of acute toxoplasmosis between 7 and 19 days PI; T. gondii tachyzoites were found in histological sections of many tissues. One of the 10 skunks fed tissue cysts and 1 of the 4 controls also died of acute toxoplasmosis days 19 and 20 PI; these animals probably became infected by ingestion of unexcysted oocysts passed in feces of skunks fed oocysts that were housed in the same room that skunks fed tissue cysts were housed. The remaining 9 skunks fed tissue cysts and the 3 controls developed only a mild illness and were killed in good health on days 23-25 PI. Antibodies to T. gondii were not found in 1:25 serum dilution of any of the 19 of 23 skunks that were alive on day 10 PI; 12 of 13 skunks had antibodies (MAT 1:80 or higher) on the day they were killed. Antibodies were not found in 1 skunk. Results indicate that skunks can develop IgG antibodies to T. gondii within 3 wk PI, and primary toxoplasmosis can be fatal in skunks.  相似文献

Twenty-eight hog-nosed skunks (Conepatus leuconotus), 23 striped skunks (Mephitis mephitis), and nine spotted skunks (Spilogale gracilis) from an area of sympatry in west-central Texas were examined for helminth parasites. Shared helminth species among all three host species were one nematode (Physaloptera maxillaris), two acanthocephalans (Pachysentis canicola, Macracanthorhynchus ingens), and one cestode (Mathevotaenia mephitis). Two nematodes (Gongylonema sp. and Filaria taxidaea) occurred in both the striped and hog-nosed skunks. One nematode (Filaroides milksi) and one acanthocephalan (Oncicola canis) were collected only from C. leuconotus. The most common helminth infections for striped and hog-nosed skunks were P. maxillaris and P. canicola. Helminth species richness was highest in hog-nosed skunks, but striped skunks had the highest prevalences and intensities of all the common helminth species. The helminth fauna of spotted skunks was markedly depauperate in terms of species richness and helminth abundance compared to the other two host species. Differences in helminth communities across these three sympatric skunks may be related to differences in their relative abundance, behavior, food habits, and geographic range.  相似文献

Twenty-four adult striped skunks (Mephitis mephitis) were administered the raccoon product formulation of Rabies Vaccine, Live Vaccinia-Vectored (Raboral V-RG, Merial Limited, Athens, Georgia, USA), either by oral instillation or in vaccine-filled coated sachets either as single or multiple doses. A control group remained unvaccinated. Twenty-three of the skunks were challenged 116 days postvaccination with rabies virus (skunk isolate). Six of six naive skunks succumbed to challenge. Four of six skunks that received the vaccine by oral instillation survived challenge. The skunks that did not survive failed to seroconvert following vaccination. None of the skunks that accepted multiple doses of the vaccine offered in coated sachets survived challenge, nor were rabies virus-neutralizing antibodies (VNAs) detected in the sera. Likewise, none of the five skunks ingesting a single sachet developed VNA against rabies. However, in this group one skunk did survive rabies challenge. This preliminary study showed that the vaccinia-vectored oral rabies vaccine Raboral V-RG, as formulated for use in raccoons, is capable of protecting a percentage of skunks against rabies. However, although the fishmeal-coated sachets were readily consumed, subsequent challenge of these animals revealed poor vaccine delivery efficiency.  相似文献

Live-captured striped skunks (Mephitis mephitis) and raccoons (Procyon lotor) were immunized with inactivated rabies vaccine by intramuscular injection and released at the point of capture during a rabies control program in Metropolitan Toronto (Ontario, Canada). Serum samples collected prior to and following vaccination revealed that 100% of the skunks and 98% of the raccoons seroconverted. Rabies antibody was still detectable 314 to 757 days postvaccination. Five of six skunks vaccinated in the laboratory survived challenge with rabies virus 90 days postvaccination. To our knowledge, this is the first documentation of the successful seroconversion of skunks and raccoons vaccinated against rabies in the field.  相似文献

The Arctic variant of rabies virus has been maintained in striped skunks in small foci in southwestern Ontario, Canada, despite the control of the disease in red foxes. To control the disease in skunks, high-density baiting with ONRAB(?) oral rabies vaccine baits was conducted by air and by hand distribution of baits in the vicinity of skunk cases. During 2009, antibody prevalences in skunks were higher in areas baited at a density of 300 baits/km(2) and flight-line spacing of 0.25 km than at 0.5-km spacing. Once an area containing Arctic-variant cases was treated with high densities of ONRAB baits, the disease did not reoccur in skunks in those areas. During 2009, only eight skunks were diagnosed with the Arctic variant of rabies virus in Ontario.  相似文献

12 female skunks were hypophysectomized during the 7-month preimplantation period. Sham operations were performed on 7 skunks by drilling through the basisphenoid but without exposing the pituitary. 22 skunks remained as controls. Complete hypophysectomy significantly reduced peripheral plasma progesterone levels within 40 days while there was a tendency for values to increase in sham hypophysectomized skunks. The only obvious effect hypophysectomy had on ovarian histology during the preimplantation period was the abolition of all antral follicles; corpora lutea remained unchanged for up to 90 days. Blastocysts from hypophysectomized skunks were undergoing a slow attrition of cells after Day 34, and there was some indication that hypophysectomy may have a deleterious effect as early as Day 22. Hypophysectomy significantly prolonged the preimplantation period in 6 out of 7 skunks. It was concluded that the pituitary is essential for initiation of nidation.  相似文献

Safety of the modified live rabies virus vaccine, SAD B19, was studied in striped skunks (Mephitis mephitis). Seven skunks received 10(7.9) foci formatting units by direct oral administration. In four cages, a vaccinated animal was placed with a control animal, the other three vaccinated skunks were housed individually. Saliva and nasal swabs were collected 1, 2, 4, 24, 48, and 72 hr post-vaccination. From all vaccinated and control animals (n = 11) blood samples were collected 0, 28, 56, 84, and 296 days post-vaccination. Three of seven vaccinated skunks seroconverted. None of the control animals had detectable levels of rabies virus neutralizing antibodies. Also no vaccine virus was isolated from the nasal and saliva swabs collected from any animal. Thus, SAD B19 was innocuous for skunks in our study after direct oral administration at field concentration.  相似文献

Lipids were extracted from virulent Leptospira pomona and were purified. These lipids fixed complement in the presence of antiserum to L. pomona but did not stimulate the production of homologous agglutinins in rabbits, mice, or hamsters. When subsequently challenged, all of the mice and hamsters were fully susceptible to L. pomona. The lipid material was neither dermonecrotic nor lethal for mice or hamsters, but 382 mug of lipid from virulent or avirulent leptospires inhibited the growth of normal mice. Leptospiral lipids were toxic for peritoneal macrophages maintained in vitro, and when administered simultaneously with a million lethal doses of L. pomona, the lipids hastened death of the hamsters, presumably by inhibiting phagocytosis early in the course of the infection.  相似文献

The number and geographic distribution of rabies cases in striped skunks (Mephitis mephitis) from Saskatchewan (n = 2,506 cases), Montana (n = 1,142), and Alberta (n = 199) since 1963 were reviewed. In Saskatchewan the number of cases increased steadily for 5 yr and then fluctuated consistently in a 4 yr cyclic pattern. Similarly an initial sweep across the province was followed by a cyclic pattern of geographic expansion (3 to 4 yr) and reduction (1 to 2 yr). No organized control efforts were conducted in Saskatchewan. Similar cyclic pattern were not seen in data from Montana or Alberta. In the latter areas, the number and distribution of rabies cases in skunks appeared to reflect efforts to reduce the population of skunks. An integrated program of skunk removal using poison and live-traps in association with research and public education successfully contributed to limiting the spread and establishment of rabies in striped skunks within prairie habitats. Rabies did not persist in skunks in other habitats.  相似文献

Nine percent of 198 serum samples from striped skunks, Mephitis mephitis (Schreber) from five areas of Alberta were positive for rabies neutralizing antibody. Positive samples were minimal (2%) from specimens sampled in an area enzootic for rabies and occurred at greater rates in areas negative for skunk rabies. Transmission of rabies virus to skunks may have been from a source other than skunks in those areas, most probably from bats.  相似文献
