Mycelial compatibility of 62 isolates of Valsa malicola from different hosts and areas of Iran were investigated on potato dextrose agar (PDA) and oat meal agar (OMA). Four mycelial compatibility groups (MCGs) on PDA, 1–4, including three single membered (1–3) and a 58 membered (4) were identified. However, 8 MCGs, 1–8, consisting of 6 single membered (1–3, 5, 6 and 8), a 6 membered (4) and a 50 membered (7) were identified on OMA. On PDA, the number of groups and the time to achieve results were less than on OMA as well as the barrage zones were clearer on PDA than OMA. There was no correlation between groups and host or geographical origins of the isolates. The low number of identified MCGs on both culture media revealed low genetic diversity of investigated isolates of V. malicola.  相似文献

NineArmillaria isolates obtained from the roots ofGaleola septentrionalis in Hokkaido were identified asA. jezoensis by means of mating tests. Sphaeropsis pyriputrescens, the causal agent of Sphaeropsis rot of pears and apples, is a recently described species. In this study the effects of culture media, temperature, water potential, pH and light on mycelial growth and pycnidial production of S. pyriputrescens were evaluated. Apple juice agar and pear juice agar were most suitable for mycelial growth of all six isolates tested. Cornmeal agar was not suitable for either mycelial growth or pycnidial production. The fungus grew from -3 to 25 C, with optimum growth at 20 C and no growth at 30 C. The fungus grew at water potential as low as -5.6 MPa on potassium chloride-amended potato-dextrose agar (PDA). Hyphal extension was not observed at -7.3 MPa after 10 d incubation, but growth resumed when the inoculum plugs were placed on PDA. The fungus grew at pH 3.3-6.3 and optimum growth was at pH 3.3-4.2. No mycelial growth was observed at pH above 7.2 after 10 d incubation, but growth resumed when the inoculum plugs were transferred onto PDA. Regardless of medium tested, few pycnidia formed at 20 C in the dark. Pycnidial production was enhanced significantly by fluorescent light, but continuous light appeared to reduce pycnidial production, depending on the medium. Oatmeal agar (OMA) was most suitable for production of pycnidia and conidia. Pycnidia that formed on 3 wk old OMA cultures at 20 C under 12 h light/12 h dark produced abundant conidia, and the technique is recommended for inoculum production.  相似文献

The tents of different ant species collected from young cocoa trees in their first or second year of bearing and free from visible pod rot, were tested for the presence of Phytopthtora palmivora by inoculating wounded cocoa pods with tent material. Tents of all species harboured viable P. palmivora but those consisting mainly of soil were more frequently positive than plant debris-type tents. Although Anoplolepis longipes, a dominant, ground nesting, non-tent building species sometimes transported inocula in the laboratory, it did not significantly increase black pod infection in the field. Trees infested with the dominant debris tent building species, Technomyrmex albipes, however, had significantly more black pod than those infested with A. longipes or trees without ants. A. longipes forms dense populations and can exclude other dominant ants and some cocoa pests; its introduction may be a potentially economical method of reducing the transmission of P. palmivora in redeveloped cocoa in Papua New Guinea.  相似文献

A new species of Phacidiopycnis associated with pome fruits is described. The fungus causes fruit rot on apples during storage and is associated with a twig dieback and canker disease of crabapple trees and dead twigs of pear trees. To characterize the biology of the fungus and compare it with Ph. piri, the type species of the genus, effects of nine media and light on mycelial growth and pycnidial production, mycelial growth in response to temperature and mode of conidial germination in response to nutrient were determined. Apple-juice agar, pear-juice agar, prune-juice agar, potato-dextrose agar (PDA) and malt-extract agar, Czapek-Dox agar and oatmeal agar (OMA) favored mycelial growth. Cornmeal agar (CMA) did not favor mycelial growth. Light effect on pycnidial formation was medium dependent. Abundant pycnidia with mature conidia formed in 14 d old PDA and OMA cultures at 20 C, regardless of light, whereas none or very few pycnidia formed on other media in the dark. Fluorescent light stimulated formation of pycnidia except on CMA. The fungus grew at -3-25 C, with optimum growth at 15-20 C. Conidia germinated either by forming germ tubes or less often by budding. Budding of conidia occurred in 1 and 10% pear-juice solutions but not in 100% pear-juice solution. Six isolates of Ph. washingtonensis from different species of pome fruits had identical ITS sequences. The sizes of the ITS region were the same for both Ph. washingtonensis and Ph. piri, and four polymorphic nucleotide sites were found in the ITS region between Ph. washingtonensis and Ph. piri. The similarity in ITS sequences between these two taxa is confirmatory evidence for the erection of the new species of Phacidiopycnis associated with pome fruits we describe here.  相似文献

A laboratory experiment was conducted to study the variability among the eight isolates of Colletotrichum truncatum of greengram collected from different locations on the basis of cultural, morphological and biochemical characteristics by using nine different culture media viz., Potato Dextrose Agar (PDA), Potato Carrot Agar (PCA), Oat Meal Agar (OMA), Corn Meal Agar (CMA), Carrot Agar (CA), Sabouraud’s Agar (SA), Czapek’s Dox Agar (CDA), Richard’s Agar (RA) and V8 Juice Agar. Colony colour varied in different media from white or white with light brown centres which later changed to black or dark to light brown with increase in the age of the fungal cultures. Mostly, the colonies had fluffy or cottony mycelial growth with slight variations and regular to irregular white margin. PDA, PCA, CA and RA produced maximum mycelial growth (90?mm) at 10 DAI (days after inoculation). Minimum growth was observed on SA (69.56?mm) and V8 juice agar media (55.42?mm) and their difference was statistically significant. Morphological variability among the isolates was studied by comparing their conidial length, breadth and length and breadth of setae and their differences were statistically significant. Biochemical variability among the isolates was based on α- and β-esterase and peroxidise profiling. Positive activity was observed for both α- and β-esterase. α-Esterase enzyme showed the highest enzyme activity in terms of maximum numbers of banding loci among the three isozymes tested. The findings of the present study clearly revealed that cultural, morphological and biochemical variability did exist among the different isolates of C. truncatum.  相似文献

Bud rot disease affecting oil palm in South American countries is reported to be caused by Phytophthora palmivora. P. palmivora is a local pathogen affecting various crops in Malaysia, and this finding caused an alarm, which prompted an investigation of pathogenicity using Malaysian P. palmivora to assess the potentials of this Oomycete to infect oil palm in Malaysia. A total of 11 P. palmivora isolates were obtained from cocoa and durian for the study. Leaf bioassays via artificial inoculation using 50,000 zoospores/ml and mycelial agar disc showed severe necrotic lesions on the infection spot of oil palm (DxP) spear leaves. Mild infection was observed in oil palm spear leaves of OxG hybrids indicating lower susceptibility against P. palmivora infection. Phylogenetic analysis using internal transcribed spacer (ITS) data revealed that Malaysian isolates were genetically similar to Colombian isolates supported by significant bootstrap values. The leaf bioassay results revealed that Malaysian oil palm materials are susceptible towards local P. palmivora infection. The Colombian P. palmivora isolates causing bud rot incidence may have evolved over a long period of time, undergone sequential genetic shift to become more virulent towards Colombian oil palm planting materials.  相似文献

Twelve culture media powders prepared from local raw materials were evaluated for their mycological utility using Nigrospora oryzae as the initial test organism. Radial growth of N. oryzae was greater (P < 0.05) on media from pigeon pea, soybean and ripe plantain than on Oxoid potato-dextrose agar (PDA) control. Two other media supported radial growth comparable to potato-dextrose agar. More conidia were produced on the ripe plantain medium (44/microscope field) than on PDA (3.4/microscope field). In an additional test, Fusarium oxysporum f. sp. cubense, Aspergillus niger, Curvularia lunata and Choanephora cucurbitarum grew and sporulated well on selected media. Local media were inexpensive and easy to produce but tended to be characterised by reduced clarity and presence of particles after autoclaving and jelling.  相似文献

Canker disease of Theobroma cacao L. caused by Phytophthora palmivora (Butler) Butler gives rise to serious damage in Fiji and is common in the S.W. Pacific area. Fusarium decemcellulareBiick (imperfect stage of Calonectria rigidiuscula (Berk. & Br.) Sacc.) was constantly associated with P. palmivora in disorders of cocoa but was not found to be a primary pathogen. Most Amelonado cocoa trees were resistant to canker but there were differences in the reactions of other Forastero and of Trinitario types. The F1 generation of a local Trinitario selection, itself resistant, showed segregation resulting in a proportion of very susceptible plants. Isolates of P. palmivora from black pod, canker and green twigs were equally pathogenic, both to pods and seedling stems. Sporangial dimensions, particularly the length/breadth ratio, differed from those reported elsewhere. It is uncertain whether the reason that cocoa canker is more serious in some countries than others is because of differences in the pathogenicity of P. palmivora strains to bark or the genetic differences of the cocoa grown.  相似文献

The nutritional requirements of Phytophthora palmivora in relationto extracellular pectolytic enzyme production were investigated.These were found to be different from those necessary for mycelialgrowth. The best carbon source for enzyme production was maltose.The best inorganic nitrogen source for enzyme production wascalcium nitrate, the effect of which could not be related solelyto the calcium ions; DL-alanine was the best organic nitrogensource for enzyme production. Evidence was obtained to indicatethat polygalacturonase (PG) and a maceration factor (MF) wereconstitutive, but that -L-arabinofuranosidase (AF) secretionwas induced by arabans present in pectin. The extrinsic factors in relation to growth and enzyme secretionwere also investigated. Both mycelial growth and enzyme productioncould be broadly related to the incubation period, temperature,and the pH of the media. Two peaks of PG and AF activities wererecorded corresponding to the phases of optimum growth and autolysisof the mycelium, respectively. Biological and physiological characteristics of Neotyphodium gansuense were compared with Neotyphodium coenophialum and Epichlo? festucae at a range of temperatures and pH values, and on carbon and nitrogen amended media. N. gansuense was able to grow at 10-30 degrees C, but not at 5 degrees C, and slowly at 35 degrees C. The optimal temperature for both N. gansuense and N. coenophialum was 25 degrees C, but that of E. festucae was 20-25 degrees C. The optimal pH ranges for mycelial growth of N. gansuense, N. coenophialum and E. festucae were 5-9, 5-9 and 5-7, respectively. The Neotyphodium and Epichlo? endophytes varied in their ability to grow on media containing different carbon and nitrogen nutrients. The preference of N. gansuense for carbon source was sucrose>glucose, lactose, sorbitol, inulin, maltose, mannitol, starch, fructose>xylose. Growth of all three endophytes tested was significantly improved by peptone, tryptone, casein, yeast extract and l-proline. Yeast extract, peptone, casein, tryptone, l-proline, potassium nitrate, ammonium oxalic acid and l-leucine significantly improved growth of N. gansuense. However, ammonium nitrite was not utilized at all by any tested endophyte. N. gansuense grew significantly better on potato dextrose agar (PDA) and oat meal agar (OMA) than on corn meal agar (CMA) and drunken-horse-grass agar (DA), and most slowly on water agar (WA) and saltwater nutrient agar (SNA).  相似文献

Very few sporangia of Phytophthora palmivora germinated directlyand produced secondary sporangia in distilled water and in solutionsof amino acids and carbohydrates at 30 °C. Although 1.0per cent (w / v) peptone and yeast-extract stimulated a highpercentage of germination by formation of germ tubes, less than1.0 per cent of the germinated sporangia produced secondarysporangia. Secondary sporangium formation was induced by transferringgerminated primary sporangia from a nutrient medium of sufficientlyhigh concentration to either distilled water or dilute solutionsof organic and inorganic compounds immediately after emergenceof the germ tubes. The percentage of germinated sporangia formingsecondary sporangia was influenced by both the nature and concentrationof the medium into which they were transferred. When electrophoretic profiles of native proteins from vegetative mycelia ofPhytophthora palmivora, Phytophthora capsici and Phytophthora citrophthora causing black pod disease of cocoa in India were compared on a single Polyacrylamide gel, the isolates of same species were readily distinguished both qualitatively by visual similarity in banding patterns and quantitatively by calculating similarity coefficients. Similarity coefficients were generally much higher between isolates within a species than between isolates of different species. The dendrograms obtained after unweighted pair grouping with arithmetic averaging cluster analysis, revealed that all the isolates ofPhytophthora capsici were highly homogenous and formed a single cluster. The isolates ofPhytophthora citrophthora were resolved into two electrophoretic types which were clustered into two distinct sub groups.Phytophthora palmivora formed a separate group. Thus, the results reveal that polyacrylamide gel electrophoresis can be used successfully in distinguishing species and sub groups within a species ofPhytophthora encountered on cocoa. The effects of pod age on oviposition, and on egg and larval survival of the cocoa pod borer (CPB), Conopomorpha cramerella (Snellen), were investigated. CPB were allowed to oviposit on a batch of 50 pods for a period of 7 days over several exposure dates. Numbers of CPB eggs/pod and survival to larval emergence from the pods were recorded. Pods were harvested when ripe and analyzed for larval survival within the pod. The oviposition pattern varied with pod age. Older pods less than 7 weeks before ripening (WBR) were preferred, suggesting greater nutritional value or chemical attraction of the older pods. The proportion of eggs parasitized by Trichogrammtoidea bactrae fumata Nagaraja (TBF), the number of eggs lost and the proportion surviving to larval emergence from the pod were independent of pod age. Egg parasitism was low, implying that TBF was not an effective natural enemy. Mass-releases of TBF should be intensified during cropping periods with higher proportions of susceptible pods (<10 WBR). Survival of larvae in the presclerotic layer (preSCL) was high and independent of pod age. Larval mortality at the SCL (sclerotic layer) was dependent on pod age and was high in older pods. Larval density influenced the survival of larvae inside the pod and was dependent on pod age. Larval mortality associated with SCL hardness and thickness is one of the criteria for the selection of CPB resistant cocoa clones.  相似文献

Sporangia of Phytophthora palmivora germinated by either forminggerm tubes or producing zoospores. Two distinct modes of germ-tubedevelopment have been described. Sporangia in distilled waterformed zoospores at 10–34°C with an optimum at 22°Cbut germinated by means of germ tubes at 30 and 34°C only.Zoospore formation was inhibited to varying degrees by cocoapod extract, I.0 per cent (w/v) peptone and yeast extract, 100–500µg1-1 thiamine, and by very low concentrations of severalamino acids, carbohydrates, and inorganic salts. On agar media the maximum conidia production of Exserohilum monoceras occurred on V-8 juice agar (VA) or centrifuged V-8 juice agar, whereas the optimal radial mycelial growth occurred on Czapek-Dox agar. The optimal temperatures for radial mycelial growth and conidia production were 28 and 27°C respectively. Light prohibited E. monoceras conidia production. The best sporulation occurred under continuous dark conditions. Echinochloa leaf decoction significantly increased conidia production on potato dextrose agar (PDA) and VA, and significantly increased germ tube length on PDA, lima bean agar and VA, but did not affect conidia germination. No conidia were produced in liquid media. Of 22 agricultural-based products evaluated as solid substrates, the most abundant sporulation (1.8 × 106 conidia g-1 of dry weight) occurred on corn leaves. The conidia production of E. monoceras on corn leaves was affected by incubation period, moisture content and substrate quantity. There were no differences in germination rate, germ tube length and virulence of conidia produced on agar media or corn leaves.  相似文献

Potato dextrose agar amended with salicylic acid (SA) suppressed growth of all Ganoderma boninense isolates. However, mycelial growth inhibitory effects varied with times, SA concentrations and G. boninense isolates. At 150 ppm of SA or higher, growth suppression diminished with time and mycelial growth recovery was observed. In contrast, jasmonic acid (JA) unexpectedly improved growth of G. boninense. At 21 day-after-inoculation, pH changes and hydrophobicity for the melanised mycelia were lower compared to non-melanised mycelia. The degree of inhibitory or promotional effects on the growth of G. boninense and the level of G. boninense mycelial hydrophobicity by SA and JA were isolate-dependent.  相似文献

Leaf spot of pawpaw is hereby reported for the first time in Nigeria. The symptom is characterized by a papery center surrounded by a yellow halo. The causal organism is Corynespora cassiicola. Ripe fruits and abaxial surfaces of the leaves were significantly more susceptible to infection than unripe fruits and adaxial surfaces of leaves. Growth and sporulation of the fungus on several media was investigated. The organism grew faster on malt-extract agar (MEA) derived media and slowest on potato-dextrose agar (PDA) supplemented with thiamine. Sporulation was highest on Czapek-dox agar (CDA) plus biotin and lowest on PDA and PDA + thiamine. The fungus Sclerotinia minor (IMI 344141) is being developed as a biological control for dandelion and other broadleaf weeds in turfgrass environments. Being a microbial pest control agent (MPCA), the S. minor strain must be characterized to show relatedness to like organisms and to distinguish the MPCA from related microorganisms. Phenotypic variation among 30 isolates of S. minor, collected from different regions and hosts, was studied on potato dextrose agar (PDA) and oatmeal agar (OMA). Isolates varied significantly in sclerotia shape (length/width ratio) and number, but did not vary in colony morphology or growth rates. There was high diversity (0.6) among the mycelial compatibility groups (MCG) as seven multi-member and 11 single member groups were recognized. Isolates were categorized into highly virulent, virulent, moderately virulent, and hypo virulent based on 48 h post mycelial growth on detached dandelion leaves. When assessed on dandelion plants in the greenhouse, isolate IMI 344141 ranked the highest in biocontrol efficacy, reduction of above- and below-ground biomass, and reduction in dandelion survival. Oxalic acid production was not correlated with isolate aggressiveness or growth rate and did not vary among isolates of the same MCG. IMI 344141 can be phenotypically distinguished from the other tested S. minor isolates by performing vegetative compatibility testing and counting sclerotia produced on standard 9-cm diameter PDA plates. IMI 344141 produces <100 sclerotia/plate.  相似文献
