The assignment of the 13C- and 1H-NMR spectra of eight oligosaccharides of the lacto-N-tetraose and neotetraose series was obtained from homonuclear and heteronuclear correlation spectroscopy. A reduced tetrasaccharide chain was obtained from human von Willebrand factor (vWF) by mild alkaline borohydride treatment. Summary The conformationalcis-trans equilibrium around the peptide bond in model tripeptides has been determined by 2D NMR methods (HOHAHA, ROESY). The study
was limited to three different N-substituted amino acids in position 2, namely Pro (proline), Tic (1,2,3,4-tetrahydroisoquinoline-3-carboxylic
acid), and N-MePhe (N-methylphenylalanine). In all cases the amino acid in position 1 was tyrosine and in position 3, phenylalanine. The results
of our studies show that thecis-trans ratio depends mostly on the configuration of the amino acids forming the peptide bond undergoing thecis-trans isomerisation. The amino acid following the sequence (in position 3) does not have much influence on thecis-trans isomerisation, indicating that there is no interaction of the side chains between these amino acids. The model peptides with
the L-Tyr-L-AA-(L-or D-)Phe (where AA is N-substituted amino acid) chiralities give 80–100% more of thecis form in comparison to the corresponding peptides with the D-Tyr-L-AA-(L-or D-)Phe chiralities. These results indicate that
the incorporation of N-substituted amino acids in small peptides with the same chirality as the precedent amino acid involved
in the peptide bound undergoing thecis/trans isomerisation moves the equilibrium to a significant amount of thecis form. 相似文献

The O-specific polysaccharide of Citrobacter gillenii PCM 1542 from serotype O-12a,12 b is composed of one residue each of D-glucose, D-GlcNAc, 2-deoxy-2-[(R)-3-hydroxybutyramido]-D-glucose (D-GlcNAcyl) and two GalNAc residues. On the basis of sugar and methylation analyses of the intact and Smith degraded polysaccharides, along with 1D and 2D 1H and 13C NMR spectroscopy, the following structure of the branched pentasaccharide repeating unit of the O-specific polysaccharide was established:This structure differs significantly from that of the O-specific polysaccharide of C. gillenii PCM 1544 from the same serotype O-12a,12 b, which has been established earlier (Kübler-Kielz.shtsls;b, J. et al. Carbohydr. Res. 2001, 331, 331-336). One- and two-dimensional1H- and13C-NMR spectra of lipid extracts fromUlva rigida, Gracilaria longa, Fucus virsoides andCodium tomentosum collected in the northern Adriatic Sea allowed screening of the content of fatty acid chains, carotenoids, free and acylated cholesterol and chlorophylls. The carotenoid-to-polyunsaturated fatty acid molar ratio was taken as a comparison parameter in samples ofUlva rigida collected in differentloci and seasons; the value was markedly higher in samples from the Lagoon of Venice than from marine coastal waters. The total cholesterol concentration was evaluated by1H-NMR spectroscopy and similar values were found for all species. The 7α-methyl analog (II) of 3β-hydroxy-5α-cholest-8(14)-en-15-one (I) was prepared by chemical synthesis and evaluated with respect to its effects on HMG-CoA reductase activity in CHO-K1 cells and on serum cholesterol levels in rats. The 7α-methyl substitution had no detectable effect on the potency of I in lowering HMG-CoA reductase activity in the cultured cells. In contrast, the 7α-methyl substitution had a marked effect on the action of I in the suppression of food consumption in rats. Whereas II was less potent than I in lowering serum cholesterol levels in rats, it did so at dosage levels at which only slight or moderate effects on food consumption were observed. Full 1H and 13C-NMR assignments for II and intermediates in its synthesis have been presented. Conformational analysis, based on 1H-1H coupling constants, NMR shieldings and force-field calculations, indicated that the 7α-methyl substitution had virtually no effect on the conformation of the 15-ketosterol apart from minor distortions of ring B. 相似文献

The cell wall of Fusarium oxysporum was digested with commercial Bacillus pumilus chitosanase. The chitosanase produced low molecular weight heterooligosaccharides consisting of GlcN and GlcNAc from the cell wall. A main component of the digestion products was identified as 2-amino-2-deoxy-β-d-glucopyranosyl-(1 →4)-2-acetamido-2-deoxy-d-glucopyranose. The chitosanase appeared to be more effective than Streptomyces griseus chitinase for cell wall digestion. Moreover, maltose was unexpectedly found in the digestion products, indicating that the cell wall contains α-1,4-linked glucan chain as a polysaccharide component. 相似文献

The major pentasaccharides Fuc(1-2)[GalNAc(1-3)]Gal(1-4)[Fuc(1-3)]Glc and Fuc(1-2) [Gal(1-3)]Gal(1-4)[Fuc(1-3)]Glc, which are normally present in the urine of bloodgroup A Leb and B Leb healthy subjects, were each found to be contaminated by a minor component when analysed by1H-NMR. Rat hemopexin is a plasma glycoprotein that contains 18.3% carbohydrate consisting of onlyN-glycosidically-linked oligosaccharide chains. Glycopeptides obtained from hemopexin by Pronase® digestion could be separated on Concanavalin A-Sepharose into three fractions. The lectin-binding fraction has been characterized as a mixture of monosialyl and disialyl di-antennary compounds ending inN-acetylneuraminic acid residues (2-6)-linked to galactose in the respective branches [Bernard N, Lombart C, Strecker G, Montreuil J, Van Halbeek H, Vliegenthart JFG (1983) Biochimie 65:185–92].The structures of the glycans in the Concanavalin A non-binding fractions were determined by a combination of methylation analysis and 500-MHz1H-NMR spectroscopy. This type of structure has been encountered before in some bovine blood coagulation factors as well as in rat -acid glycoprotein, but the1H-NMR parameters for it are first reported here. Furthermore, by methylation analysis, the occurrence of the NeuAc2-8NeuAc disaccharide element was demonstrated in a minor part of the carbohydrate moiety of rat hemopexin. It is demonstrated that the H2BC NMR pulse sequence (J. Am. Chem. Soc.2005, 127, 6154, Magn. Reson. Chem.2005, 43, 971-974) offers unambiguous assignments and significant simplification of NMR spectra of large and complex carbohydrates compared to other techniques for the establishment of correlations over more than one bond. H2BC almost exclusively correlates protons and proton-bearing carbon spins separated by two covalent bonds and is independent of occasionally vanishing (2)J(CH) coupling constants, which alleviates the problem of missing two-bond correlations in HMBC spectra. H2BC also solves the problem of distinguishing two- and three-bond correlations in HSQC-TOCSY or HMBC. It is a further asset of H2BC that the experiment is significantly shorter than HMBC and HSQC-TOCSY, and hence less sensitive to transverse relaxation. The H2BC experiment is demonstrated on an approximately 30-residue oligosaccharide from Francisella victoria. 相似文献

The hydrocarbon and lipid components of four strains of the filamentous cyanobacterium Scytonema sp. isolated from microbial community Black Cover of limestone walls in Jerusalem were identified by gas chromatography–mass spectrometry using serially coupled capillary columns. The dominant compounds were: 1-heptadecyne (1.5-8%), hexadecanoic acid (14-36%), (Z,Z)-9,12-octadecadienoic acid (12-30%), (Z,Z,Z)-9,12,15-octadecatrienoic acid (6-12%), n-heptadecane (4-16%), and 1-heptadecene (1.5-8%). In addition to unsaturated alkanes and fatty acids, the very long-chain (C30-C32) hydrocarbons, squalene (2.4-3.0%), and branched 4,8,12-trimethyl-C13:0 acid were also isolated. A new Bifidobacterium species is described based on the study of ten Gram-positive strains with fructose-6-phosphate phosphoketolase activity. They are part of a phenotypic group comprising 141 strains isolated from raw milk and raw milk cheeses in French raw milk cheese factories. This group was separated by a numerical analysis based on API 50CH, API 32A tests and growth at 46 degrees C. A strong similarity of 16S rRNA sequences (99.8%) was shown between strain FR62/b/3(T) and Bifidobacterium psychraerophilum LMG 21775(T). However, low DNA-DNA relatedness was observed between their DNAs (31%). The new isolates are able to grow at low temperatures (all ten strains up to 5 degrees C) and strain FR62/b/3(T) grows under aerobic conditions, as does B. psychraerophilum. However, contrary to B. psychraerophilum, they do not ferment L-arabinose, D-xylose, arbutin or melezitose, but they do acidify lactose. The DNA G+C content of FR62/b/3(T) is 56.4mol%. Therefore, the name Bifidobacterium crudilactis sp. nov. is proposed, with its type strain being FR62/b/3(T) (=LMG 23609(T)=CNCM I-3342(T)). 相似文献

Application of tree leaves (C3 plants) on maize (Zea mays L.) (C4 plant) fields is an agroforestry management technology to restore or maintain soil fertility. The rate at which the tree
leaves decompose is crucial for the nutrient supply to the crop. We studied the in situ decomposition of Sesbania sesban (L.) Merr. leaves or C3 sugar for 4 – 8 days after application to a maize field in Kenya. By using the difference of around 10‰ in natural abundance
of 13C between the endogenous soil C (mainly C4) and the applied C (C3), we could calculate the contributions of the two C sources to soil respiration. The δ13C value of the basal respiration was from –15.9 to –16.7‰. The microbial response to the additions of leaves and sugar to
this tropical soil was immediate. Application of sesbania leaves gave an initial peak in respiration rates that lasted from
one to less than 6 days, after which it levelled off and remained about 2 – 3 times higher (230–270 mg C m-2 h-1) than the control respiration rates throughout the rest of the experiment (5 – 8 days). In the sugar treatment, there was
no initial peak in respiration rate. The respiration rate was 170 mg C m-2 h-1 after 4 days. At the end of the experiments, after 4–8 days, as much as 14–17% of the added C had been respired and about
60% of the total respiration was from the added sesbania leaves or C3 sugar. This non-destructive method allows repeated measurements of the actual rate of C mineralisation and facilitates decomposition
studies with high temporal resolution in the field.

Ascorbic acid oxidase (E.C.1.10.3.3) from the green zucchini squash (Cucurbita pepo medullosa) is a copper-containing glycoprotein which catalyzes the reaction:l-ascorbic acid +1/2 O2l-dehydroascorbic acid + H2O. The carbohydrate content of the purified plant glycoprotein amounted to 3% (w/w), and monosaccharide analysis revealed the carbohydrate moiety to be of theN-glycosidic type. A novel structure of exopolysaccharide from the lactic acid bacteria (LAB) Lactobacillus rhamnosus KL37B, from the human intestinal flora, is described. During the structural investigation of the exopolysaccharide it was found that the repeating unit is a nonasaccharide, which is the largest repeating unit found in LAB exopolysaccharides to date. The polysaccharide material was prepared by TCA extraction of a bacterial cell mass, purified by anion-exchange and gel permeation chromatography and characterized using chemical and enzymatic methods. On the basis of monosaccharide and methylation analysis and also 1D and 2D 1H and 13C NMR spectroscopy the exopolysaccharide was shown to be composed of the following nonasaccharide repeating unit:The physicochemical cell surface study and adhesive properties indicated distinct surface properties of Lactobacillus rhamnosus strain KL37B with high adhesive abilities to Caco-2 cells, hydrophobicity and slime production, in comparison to other Lactobacillus strains used as controls. 相似文献

Characterization of O-acetyl-(4-O-methylglucurono)xylan isolated from birch and beech 总被引:6,自引:0,他引:6
The structures of water-soluble birch and beech xylans, extracted from holocellulose using dimethyl sulfoxide, were determined employing 1H and 13C NMR spectroscopy together with chemical analysis. These polysaccharides were found to be O-acetyl-(4-O-methylglucurono)xylans containing one 4-O-methylglucuronic acid substituent for approximately every 15 D-xylose residues. The average degree of acetylation of the xylose residues in these polymers was 0.4. The presence of the structural element -->4)[4-O-Me-alpha-D-GlcpA-(1-->2)][3-O-Ac]-beta-D-Xylp-(1--> was demonstrated. Additional acetyl groups were present as substituents at C-2 and/or C-3 of the xylopyranosyl residues. Utilizing size-exclusion chromatography in combination with mass spectroscopy, the weight-average molar masses (and polydispersities) were shown to be 8000 (1.09) and 11,100 (1.08) for birch and beech xylan, respectively. 相似文献
