In this work the methods for the passive control and automatic regulation of the concentration of vaccinal aerosols in static chambers are analyzed. For such analysis, a method for the calculation of the parameters of vaccinal aerosols, taking account of the law of the distribution of particles by their sizes and of the processes of sedimentation and inactivation of microbial aerosols, has been developed. To make empirically established coefficients more precise, the necessary experimental studies have been carried out. High effectiveness of the method providing for the regulation of aerosol mass concentration has been demonstrated, and conditions necessary for effective aerosol immunization have been established.  相似文献

Aerosols have been suspected to transport food pathogens and contaminate fruits and vegetables grown in close proximity to concentrated animal feeding operations, but studies are lacking that substantiate such transport. To monitor the potential transport of bacteria originated from fresh or dry manure through aerosols on a dairy, we identified by 16S rRNA sequencing, bacteria in aerosols collected within 2 to 3 meters from dairy cows at two dairies. Gram-positive Firmicutes were predominant in aerosols from a dairy in Sonoma, California, and surrounded by vineyards, in contrast to sequences of Gram-negative Proteobacteria predominant in aerosols from a dairy in Modesto, California, also surrounded by other dairies. Although Firmicutes represented approximately 50% of the 10 most abundant sequences, aerosols from the Sonoma dairy also contained sequences of Bacteriodetes and Actinobacteria, identified previously with animal feces. While none of the top 10 sequences from fresh or dry manure from Modesto dairy were detected in aerosols, two of the sequences from the phylum Bacteriodetes and one from class Clostridia from fresh manure were detected in aerosols from Sonoma. Interestingly, none of the sequences from dry manure were in the top 10 sequences in aerosols from both dairies. The 10 most abundant sequences in aerosols from the Modesto dairy were all from Proteobacteria and nearly half of them were from genus Massilia, which have been isolated previously from immune-compromised people and aerosols. We conclude that the predominant bacteria in aerosols are diverse among locations and that they do not reflect the predominant species of bacteria present in cow feces and/or in close proximity to cows. These results suggest that the aerosol sequences did not originate from manure. Large volumes of aerosols would be required to determine if bacterial sequences from aerosols could be used to track bacteria in manure to crops grown in proximity.  相似文献

AIM: To quantify microbial aerosols generated by a series of laboratory accidents and to use these data in risk assessment. METHODS AND RESULTS: A series of laboratory accident scenarios have been devised and the microbial aerosol generated by them has been measured using a range of microbial air samplers. The accident scenarios generating the highest aerosol concentrations were, dropping a fungal plate, dropping a large bottle, centrifuge rotor leaks and a blocked syringe filter. Many of these accidents generated low particle size aerosols, which would be inhaled into the lungs of any exposed laboratory staff. Spray factors (SFs) have been calculated using the results of these experiments as an indicator of the potential for accidents to generate microbial aerosols. Model risk assessments have been described using the SF data. CONCLUSIONS: Quantitative risk assessment of laboratory accidents can provide data that can aid the design of containment laboratories and the response to laboratory accidents. SIGNIFICANCE AND IMPACT OF THE STUDY: A methodology has been described and supporting data provided to allow microbiological safety officers to carry out quantitative risk assessment of laboratory accidents.  相似文献

In the domain of gene-gene network analysis, construction of co-expression networks and extraction of network modules have opened up enormous possibilities for exploring the role of genes in biological processes. Through such analysis, one can extract interesting behaviour of genes and would help in the discovery of genes participating in a common biological process. However, such network analysis methods in sequential processing mode often have been found time-consuming even for a moderately sized dataset.It is observed that most existing network construction techniques are capable of handling only positive correlations in gene-expression data whereas biologically-significant genes exhibit both positive and negative correlations. To address these problems, we propose a faster method for construction and analysis of gene-gene network and extraction of modules using a similarity measure which can identify both negatively and positively correlated co-expressed patterns. Our method utilizes General-purpose computing on graphics processing units (GPGPU) to provide fast, efficient and parallel extraction of biologically relevant network modules to support biomarker identification for breast cancer. The modules extracted are validated using p-value and q-value for both metastasis and non-metastasis stages of breast cancer. PNME has been found capable of identifying interesting biomarkers for this critical disease. We identified six genes with the interesting behaviours which have been found to cause breast cancer in homo-sapiens.  相似文献

Efforts to evaluate the roles and overall levels of allergens carried by microaerosols remain preliminary. It is not clear what range of allergen specificities may yet be recovered from small particle fractions under suitable analysis. However, it now appears that airborne units sized below 10 μm may contribute, perhaps substantially, to inhaled allergen burdens. Innovative application of filters and high speed impingers, as well as of immunoassays, should increasingly clarify the clinical roles of such previously overlooked, small aerosols.  相似文献

Many techniques to inactivate or remove biofilms in a wide variety of applications have been developed. Most of these techniques have been applied to biofilms at their initial stage of growth, since they are generally difficult to eradicate once established. The removal of established biofilms has received relatively little attention. In this paper, we report the effectiveness of periodic jets of carbon dioxide aerosols (a mixture of solid and gaseous CO2) to remove Escherichia coli (XL1-blue) biofilms of different ages (up to 3 weeks) on silicon surfaces. The biofilms were not immersed in liquids after growth/rinsing and were treated with the CO2 aerosols. The CO2 aerosols were generated by the adiabatic expansion of high-pressure CO2 gas through a nozzle. The surface area of the biofilms was measured from fluorescent images before and after applying the aerosols for 11, 20, and 30 cycles (each cycle: 8 sec), to compute the removal efficiency. The removal efficiencies decreased with increasing growth time and for the 3-week-old biofilms, they ranged from 91.5 to 99.6% within 4 min. This technique was highly effective for removing both fresh and old biofilms, but some of the biofilm debris such as growth media remained. Further, this CO2 aerosol technique was compared with other removal techniques.  相似文献

Hominins are smaller, slower, and weaker than most large mammals, yet they have been eating meat from freshly killed large mammals since before the invention of sophisticated weaponry. It is thought that they could have achieved this seemingly impossible feat through persistence hunting, a practice powered by endurance running. Essentially, one or more hunters pursue a prey animal in the heat of the day until it reaches the point of hyperthermia. This allows a hunter to safely kill the weakened animal at close range using methods such as beating, strangling, or spearing. The energy balance of this approach to getting food is controversial and has not been calculated previously. We examined the energy costs and gains of persistence hunting through several energy returned on investment (EROI) calculations based on synthesizing available field and laboratory data on the energy used by the hunters and the energy returned from the greater kudu (Tragelaphus strepsiceros). We estimate that the EROI of these hunter-gatherers hunting a kudu ranged from 26:1 to 69:1. The net energy gained from such an effort would sustain an average sized !Kung family for 6.7 to 11.2 days. The “profit” energy within these ranges would have supported the early human societies that practiced persistence hunting, contributing to the often-noted “leisure” characterizing many foraging societies (Sahlins 1974).  相似文献

Oxygen transfer rate and the corresponding power requirement to operate the rotor are vital for design and scale-up of surface aerators. Present study develops simulation or scale-up criterion correlating the oxygen transfer coefficient and power number along with a parameter governing theoretical power per unit volume (X, which is defined as equal to F(4/3)R(1/3), where F and R are impellers' Froude and Reynolds number, respectively). Based on such scale-up criteria, design considerations are developed to save energy requirements while designing square tank surface aerators. It has been demonstrated that energy can be saved substantially if the aeration tanks are run at relatively higher input powers. It is also demonstrated that smaller sized tanks are more energy conservative and economical when compared to big sized tanks, while aerating the same volume of water, and at the same time by maintaining a constant input power in all the tanks irrespective of their size. An example illustrating how energy can be reduced while designing different sized aerators is given. The results presented have a wide application in biotechnology and bioengineering areas with a particular emphasis on the design of appropriate surface aeration systems.  相似文献

利用离子色谱分析北京市大气气溶胶中的水溶性无机离子,结果表明,北京城乡结合部TSP中水溶性离子的年均浓度为54.97μg.m-3,其中SO42-、NO3-、C a2 、NH4 4种组分占总离子浓度的85%。TSP中总离子浓度冬季最高、夏季最低。K 浓度夏季最高,C a2 和M g2 秋季浓度明显比其他季节高。NH4 和SO42-的浓度变化趋势相似,相关系数为0.97。大气气溶胶中水溶性离子的粒径分布各不相同,F-、M g2 和C a2 呈粗模态分布,NH4 是细模态分布,其余离子呈双模态分布。F-、NH4 、M g2 和C a2 4种离子粒径分布的季节变化不明显,冬季NO3-在细颗粒中的比例最大,春季N a 在粗颗粒中的比例最大,采暖期前后SO42-的粒径分布有明显变化。  相似文献

Pharmaceutical inhalation aerosols have been playing a crucial role in the health and well being of millions of people throughout the world for many years. The technology's continual advancement, the ease of use and the more desirable pulmonary-rather-than-needle delivery for systemic drugs has increased the attraction for the pharmaceutical aerosol in recent years. But administration of drugs by the pulmonary route is technically challenging because oral deposition can be high, and variations in inhalation technique can affect the quantity of drug delivered to the lungs. Recent advances in nanotechnology, particularly drug delivery field have encouraged formulation scientists to expand their reach in solving tricky problems related to drug delivery. Moreover, application of nanotechnology to aerosol science has opened up a new category of pharmaceutical aerosols (collectively known as nanoenabled-aerosols) with added advantages and effectiveness. In this review, some of the latest approaches of nano-enabled aerosol drug delivery system (including nano-suspension, trojan particles, bioadhesive nanoparticles and smart particle aerosols) that can be employed successfully to overcome problems of conventional aerosol systems have been introduced.  相似文献

Aerosol particle size influences airway drug deposition. Current inhaler devices are inefficient, delivering a heterodisperse distribution of drug particle sizes where, at best, 20% reaches the lungs. Monodisperse aerosols are the appropriate research tools to investigate basic aerosol science concepts within the human airways. We hypothesized that engineering such aerosols of albuterol would identify the ideal bronchodilator particle size, thereby optimizing inhaled therapeutic drug delivery. Eighteen stable mildly to moderately asthmatic patients [mean forced expiratory volume in 1 s (FEV1) 74.3% of predicted] participated in a randomized, double-blind, crossover study design. A spinning-top aerosol generator was used to produce monodisperse albuterol aerosols that were 1.5, 3, and 6 microm in size, and also a placebo, which were inhaled at cumulative doses of 10, 20, 40, and 100 microg. Lung function changes and tolerability effects were determined. The larger particles, 6 and 3 microm, were significantly more potent bronchodilators than the 1.5-microm and placebo aerosols for FEV1 and for the forced expiratory flow between exhalation of 25 and 75% of forced vital capacity. A 20-microg dose of the 6- and 3-microm aerosols produced FEV1 bronchodilation comparable to that produced by 200 microg from a metered-dose inhaler. No adverse effects were observed in heart rate and plasma potassium. The data suggest that in mildly to moderately asthmatic patients there is more than one optimal beta2-agonist bronchodilator particle size and that these are larger particles in the higher part of the respirable range. Aerosols delivered in monodisperse form can enable large reductions of the inhaled dose without loss of clinical efficacy.  相似文献

Chacma baboons (Papio ursinus) intentionally overturn rocks to feed on the invertebrates beneath. However, baboons do not move all the rocks they encounter, with this presumably reflecting cost–benefit (or effort–reward) trade‐offs in their foraging behavior. We ask, how do “clever baboons” choose rock sizes and shapes and move these rocks? Using optimal foraging theory, we predicted that baboons would prefer to move medium‐sized rocks, a trade‐off between moving larger rocks that might require more effort to move, and smaller rocks that likely do not provide enough prey (the reward) to make the effort worthwhile. We also expected baboons to prefer rounded rocks as these will require less energy to move by rolling (rather than being flipped as for flat rocks) and that the effort of rock movement might be offset by moving rocks along the shortest axis. We show that baboons have clear preferences for specific rock sizes (medium‐sized) and shapes (angular and flat when these were medium‐sized), and the way in which rocks are moved (along the shortest axis). Prey occurred infrequently under rocks. The low predictability of prey beneath rocks suggests that such prey, when encountered, is of considerable value to baboons for them to expend the search effort, and also explains the extensive nature of rock movement by baboons in the landscape. Our study provides a novel application of the optimal foraging theory concept and has important implications for understanding and predicting how animals choose to move rocks.  相似文献

We and others have recently reported that prions can be transmitted to mice via aerosols. These reports spurred a lively public discussion on the possible public-health threats represented by prion-containing aerosols. Here we offer our view on the context in which these findings should be placed. On the one hand, the fact that nebulized prions can transmit disease cannot be taken to signify that prions are airborne under natural circumstances. On the other hand, it appears important to underscore the fact that aerosols can originate very easily in a broad variety of experimental and natural environmental conditions. Aerosols are a virtually unavoidable consequence of the handling of fluids; complete prevention of the generation of aerosols is very difficult. While prions have never been found to be transmissible via aerosols under natural conditions, it appears prudent to strive to minimize exposure to potentially prion-infected aerosols whenever the latter may arise – for example in scientific and diagnostic laboratories handling brain matter, cerebrospinal fluids, and other potentially contaminated materials, as well as abattoirs. Equally important is that prion biosafety training be focused on the control of, and protection from, prion-infected aerosols.  相似文献

The influence of diesel exhaust particles (DEP) on the lungs and heart is currently a topic of great interest in inhalation toxicology. Epidemiological data and animal studies have implicated airborne particulate matter and DEP in increased morbidity and mortality due to a number of cardiopulmonary diseases including asthma, chronic obstructive pulmonary disorder, and lung cancer. The pathogeneses of these diseases are being studied using animal models and cell culture techniques. Real-time exposures to freshly combusted diesel fuel are complex and require significant infrastructure including engine operations, dilution air, and monitoring and control of gases. A method of generating DEP aerosols from a bulk source in an aerodynamic size range similar to atmospheric DEP would be a desirable and useful alternative. Metered dose inhaler technology was adopted to generate aerosols from suspensions of DEP in the propellant hydrofluoroalkane 134a. Inertial impaction data indicated that the particle size distributions of the generated aerosols were trimodal, with count median aerodynamic diameters less than 100 nm. Scanning electron microscopy of deposited particles showed tightly aggregated particles, as would be expected from an evaporative process. Chemical analysis indicated that there were no major changes in the mass proportion of 2 specific aromatic hydrocarbons (benzo[a]pyrene and benzo[k]fluoranthene) in the particles resulting from the aerosolization process.  相似文献

Despite the recognition of potential aerosol hazards associated with cell sorting by the flow cytometry community, there has been no previous study that has thoroughly characterized the aerosols that can be produced by cell sorters. In this study, an aerodynamic particle sizer was used to determine the concentration and aerodynamic diameter (AD) of aerosols produced by a FACS Aria II cell sorter under various conditions. Aerosol containment and evacuation were also evaluated using this novel methodology. The results showed that high concentrations of aerosols in the range of 1-3 μm can be produced in fail mode and that with decreased sheath pressure, aerosol concentration decreased and AD increased. Although the engineering controls of the FACS Aria II for containment were effective, sort chamber evacuation of aerosols following a simulated nozzle obstruction was ineffective. However, simple modifications to the FACS Aria II are described that greatly improved sort chamber aerosol evacuation. The results of this study will facilitate the risk assessment of cell sorting potentially biohazardous samples by providing much needed data regarding aerosol production and containment.  相似文献

BACKGROUND: Containment of potentially biohazardous aerosols that result from high-speed sorting of human cells has been an increasingly important problem in analytical cytometry. The current method for assessing the efficiency of aerosol containment involves detection of aerosols containing sorted T4 bacteriophage on lawns of T4-susceptible Escherichia coli on plates that are placed in and around the sort area. Although this method is sensitive, it is time consuming and involves maintenance and handling of bacteria and sorting of bacteriophage that may themselves serve as sources of contamination for sorted viable human cells. METHODS: Glo Germ (5-microm melamine copolymer resin beads), which are fluorescent under black light illumination, were sorted on a Beckman-Coulter Elite ESP sorter in order to visualize deposition of aerosols under normal and mock failure modes. RESULTS: Glo Germ was successfully used under both normal sorting conditions, as well as mock failure mode, to visualize aerosol formation. CONCLUSIONS: We have developed a method to examine aerosol containment using modified Glo Germ, a product used for teaching aseptic technique in hospitals, industry, restaurants, and schools. Use of this technique represents a rapid, inexpensive, qualitative analysis of the extent and location of aerosol contamination from cell sorters.  相似文献

Abstract There has been debate over the cause of the extinction of ‘megafauna’ species during the late Pleistocene of Australia. One view is that environmental change, either natural or human‐induced, was the main factor in the extinctions. Some support for this comes from the observation that, among herbivores, most of the species that went extinct were apparently browsers rather than grazers. Browsers would presumably have been more dependent on shrubland and woodland habitats than grazers, and it has been argued that such habitats might have contracted in response to aridity or changed fire regimes in the late Pleistocene. Here, we test this idea by comparing extinction rates of browsers and grazers in the late Pleistocene, controlling for body mass in both groups. We show that in both browsers and grazers the probability of extinction was very strongly related to body mass, and the body mass at which extinction became likely was similar in the two groups. It is true that more browsers than grazers went extinct, but this is largely because most very large herbivores in the late Pleistocene were browsers, not because large browsers were more likely to go extinct than similarly sized grazers. This result provides evidence against some forms of environmental change as a cause of the extinctions.  相似文献

Trisomies 18 and 21 are genetic disorders in which cells possess an extra copy of each of the relevant chromosomes. Individuals with these disorders who survive birth generally have a shortened life expectancy. As telomeres are known to play an important role in the maintenance of genomic integrity by protecting the chromosomal ends, we conducted a study to determine whether there are differences in telomere length at birth between individuals with trisomy and diploidy, and between trisomic chromosomes and normal chromosomes. We examined samples of peripheral blood lymphocytes (PBLs) from 31 live neonates (diploidy: 10, trisomy 18: 10, trisomy 21: 11) and estimated the telomere length of each chromosome arm using Q-FISH. We observed that the telomeres of trisomic chromosomes were neither shorter nor longer than the mean telomere length of chromosomes as a whole among subjects with trisomies 18 and 21 (intra-cell comparison), and we were unable to conclude that there were differences in telomere length between 18 trisomy and diploid subjects, or between 21 trisomy and diploid subjects (inter-individual comparison). Although it has been reported that telomeres are shorter in older individuals with trisomy 21 and show accelerated telomere shortening with age, our data suggest that patients with trisomies 18 and 21 may have comparably sized telomeres. Therefore, it would be advisable for them to avoid lifestyle habits and characteristics such as obesity, cigarette smoking, chronic stress, and alcohol intake, which lead to marked telomere shortening.  相似文献

Bats tend to have less intestinal tissue than comparably sized nonflying mammals. The corresponding reduction in intestinal volume and hence mass of digesta carried is advantageous because the costs of flight increase with load carried and because take-off and maneuverability are diminished at heavier masses. Water soluble compounds, such as glucose and amino acids, are absorbed in the small intestine mainly via two pathways, the transporter-mediated transcellular and the passive, paracellular pathways. Using the microchiropteran bat Artibeus literatus (mean mass 80.6+/-3.7 g), we tested the predictions that absorption of water-soluble compounds that are not actively transported would be extensive as a compensatory mechanism for relatively less intestinal tissue, and would decline with increasing molecular mass in accord with sieve-like paracellular absorption. Using a standard pharmacokinetic technique, we fed, or injected intraperitoneally the metabolically inert carbohydrates L-rhamnose (molecular mass = 164 Da) and cellobiose (molecular mass = 342 Da) which are absorbed only by paracellular transport, and 3-O-methyl-D-glucose (3OMD-glucose) which is absorbed via both mediated (active) and paracellular transport. As predicted, the bioavailability of paracellular probes declined with increasing molecular mass (rhamnose, 90+/-11%; cellobiose, 10+/-3%, n = 8) and was significantly higher in bats than has been reported for laboratory rats and other mammals. In addition, absorption of 3OMD-glucose was high (96+/-11%). We estimated that the bats rely on passive, paracellular absorption for more than 70% of their total glucose absorption, much more than in non-flying mammals. Although possibly compensating for less intestinal tissue, a high intestinal permeability that permits passive absorption might be less selective than a carrier-mediated system for nutrient absorption and might permit toxins to be absorbed from plant and animal material in the intestinal lumen.  相似文献

Transfer of condensed viral DNA into eukaryotic cells using proteoliposomes   总被引:1,自引:0,他引:1  
High-molecular-weight viral DNAs have been packed into proteoliposomes prepared by reverse-phase evaporation followed by phospholipid membrane targeting by influenza virus glycoprotein bound to hydrophobic 'anchors'. DNA has been encapsulated in the form of spermine condensates--toroidal structures sized approx. 0.1 micron, resistant to ultrasound. The efficiency of entrapping into liposomes reached 30% for condensed DNA of Mr up to 3 X 10(7). Specific infectivity of simian virus 40 DNA and simian adenovirus DNA packed into such proteoliposomes was 50- to 100-fold higher than that shown by free DNA preparations under Ca.phosphate-precipitation conditions.  相似文献
