In the presence of 2 × 10?6 M Ca2+ in Tris-buffered medium 0.5 × 10?6 M, oestradiol-17β or corticosterone significantly increased the head-to-head association of washed bull spermatozoa; in the same concentration, testosterone and 5α-dihydrotestosterone had no significant effects, whereas progesterone significantly dissociated the associated spermatozoa. At 8 × 10?6 M Ca2+ in the same medium, all five hormones increased the association to about the same level. In Tyrode solution with a Ca2+ concentration of 1.4 × 10?3 M, oestradiol-17β and corticosterone acted as above, whereas progesterone and the two testosterones effected dissociation. In Tyrode solution each of the dissociating hormones was combined with oestradiol-17β. In each case a sum of the effects of the two hormones was obtained without any stimulation or inhibition. All five hormones still produced significant effects at 5 × 10?7 M in Tyrode solution. A corresponding value for ATP was found at 1 × 10?5 M.  相似文献

The effects of various pharmacological agents on neurally evoked contractions of the visceral muscles of the oviduct of Locusta migratoria have been examined. The pentapeptide, proctolin, at low concentrations (10?11 M?10?10 M), induced an increase in the amplitude of neurally evoked contractions and basal tonus, and induced the appearance and increased the frequency of myogenic contractions. Glutamate, at 10?4 M, produced a small transient contraction which in some preparations was accompanied by a reduction in amplitude of neurally evoked contractions. Octopamine, at 10?6 M, reduced the amplitude of neurally evoked contractions and also resulted in a relaxation of the muscles. The octopaminergic effects were inhibited by the α-aminergic antagonist phentolamine. Neurally evoked contractions were unaffected by dopamine, 5-HT or the acetylcholine receptor antagonists atropine and hexamethonium. Acetylcholine increased the amplitude of neurally evoked contractions, but only at the high concentration of 10?3 M. The possible role of proctolin and glutamate as excitatory neuro-transmitters and the inhibitory action of octopamine is discussed.  相似文献

A high basal rate of prolactin (PRL) secretion (.16±.03 μg/well/hr) was produced for over four weeks by pre-confluent male rat pituitary monolayer cell cultures. When the media was changed, a rapid release of microgram quantities of PRL occurred followed by a return to the basal PRL secretory rate by seven hours. Theophylline (3.8×10?3M), but not dibutyrl cAMP (1×10?3M), produced a significant (p<.02) increase in PRL secretion, and simultaneous addition of these agents potentiated the PRL secretory rate. TRH (2×10?8M) had no effect on PRL release by six hours, whereas dopamine (4.9×10?5M) produced a significant suppression (p<.002) of PRL secretion. In addition, the effects of theophylline, TRH, and dopamine on PRL secretion were similar in cultures of various ages. Ovine prolactin in concentrations up to 50 μg per ml produced no change in PRL secretion during 72 hours of incubation suggesting that PRL feedback control of its own secretion may be transmitted via the hypothalamus. These studies show that a high rate of PRL secretion can be maintained by pre-confluent monolayer cultures for extended periods of time, permitting repeated experimentation on the same wells.  相似文献

Inhibition of bovine erythrocyte acetylcholinesterase (free and immobilized on controlled pore glass) by separate and simultaneous exposure to malathion and malathion transformation products which are generally formed during storage or through natural or photochemical degradation was investigated. Increasing concentrations of malathion, its oxidation product malaoxon, and its isomerisation product isomalathion inhibited free and immobilized AChE in a concentration-dependent manner. KI, the dissociation constant for the initial reversible enzyme inhibitor-complex, and k3, the first order rate constant for the conversion of the reversible complex into the irreversibly inhibited enzyme, were determined from the progressive development of inhibition produced by reaction of native AChE with malathion, malaoxon and isomalathion. KI values of 1.3 × 10? 4 M? 1, 5.6 × 10? 6 M? 1 and 7.2 × 10? 6 M? 1 were obtained for malathion, malaoxon and isomalathion, respectively. The IC50 values for free/immobilized AChE, (3.7 ± 0.2) × 10? 4 M/(1.6 ± 0.1) × 10? 4, (2.4 ± 0.3) × 10? 6/(3.4 ± 0.1) × 10? 6 M and (3.2 ± 0.3) × 10? 6 M/(2.7 ± 0.2) × 10? 6 M, were obtained from the inhibition curves induced by malathion, malaoxon and isomalathion, respectively. However, the products formed due to photoinduced degradation, phosphorodithioic O,O,S-trimethyl ester and O,O-dimethyl thiophosphate, did not noticeably affect enzymatic activity, while diethyl maleate inhibited AChE activity at concentrations > 10 mM. Inhibition of acetylcholinesterase increased with the time of exposure to malathion and its inhibiting by-products within the interval from 0 to 5 minutes. Through simultaneous exposure of the enzyme to malaoxon and isomalathion, an additive effect was achieved for lower concentrations of the inhibitors (in the presence of malaoxon/isomalathion at concentrations 2 × 10? 7 M/2 × 10? 7 M, 2 × 10? 7 M/3 × 10? 7 M and 2 × 10? 7 M/4.5 × 10? 7 M), while an antagonistic effect was obtained for all higher concentrations of inhibitors. The presence of a non-inhibitory degradation product (phosphorodithioic O,O,S-trimethyl ester) did not affect the inhibition efficiencies of the malathion by-products, malaoxon and isomalathion.  相似文献

The bundle of tonic fibres situated at the proximal end of the locust metathoracic extensor tibialis muscle is innervated by the dorsal unpaired median neurone (DUMETi) as well as by the slow excitatory (SETi)) and common inhibitor (CI) neurones. It is not innervated by the fast excitatory neurone (FETi).These fibres contract spontaneously and rhythmically. The myogenic rhythm can be modified by neural stimulation.Spontaneous slow depolarizing potentials resembling the pacemaker potentials of insect cardiac muscle were demonstrated in these fibres.The actions of glutamate on the tonic muscle fibres are not compatible with its being a specific excitatory transmitter. Glutamate can stimulate weak contractions of the muscle, but this action is inhibited when chloride ions are removed from the saline.10?6 M Octapamine hyperpolarizes the tonic fibre membrane. Octopamine, GABA and glutamate all inhibit the myogenic contractions and reduce the force of the neurally evoked contractions.The tonic muscle is very responsive to proctolin. At 5 × 10?11 M proctolin enhances the force and increases the frequency of myogenic contractions. At 10?9 M it depolarizes the muscle membrane potential, and at that and higher concentrations it causes the muscle to contract. At 2 × 10?7 M proctolin induces contractures which resemble those evoked by sustained high-frequency neural stimulation. Iontophoretic experiments show that proctolin receptors occur at localized sites on the tonic fibre membrane.  相似文献

Cerebral microvessels isolated from cat forebrain contain a specific β-adrenergic-sensitive adenylate cyclase. Among various compounds tested, the most potent activator of enzyme activity is isoproterenol (ka = 1.4 × 10?7M), followed in order by epinephrine (ka= 1.5 × 10?6M), norepinephrine (ka= 1.4 × 10?5M) and phenylephrine (ka> 3 × 10?4M). Isoproterenol-stimulated enzyme activity is blocked by propranolol (ki= 2.4 × 10?9M, IPS 339 (ki= 4 × 10?9M), H35/25 (ki = 1.2 × 10?7M), atenolol (ki= 5.9 × 10?6M) and practolol (ki= 1.8 × 10?5M). These agonist and antagonist properties are quite similar to those demonstrated by β2-adrenergic receptors and β2-stimulated adenylate cyclase present in other tissues and indicate that the majority of adenylate cyclase-associated adrenergic receptors in cerebral microvessels are β2. The findings are relevant to physiological studies of cerebral blood flow and vascular permeability.  相似文献

The action of chlordimeform on the nerve-muscle preparation of the larvae of the waxmoth Galleria mellonella has been studied by means of microelectrodes. Exitatory junction potential evoked by nerve stimulation is reversibly suppressed by 2 × 10?3 M chlordimeform, and spike-like component is abolished. The resting membrane potential of the muscle fibre and the action potential from the nerve terminal are not affected at 5 × 10?3 M chlordimeform. The depolarizing membrane response caused by outward current and the effective membrane resistance are not appreciably affected. It appears that chlordimeform exerts its blocking action on the neuromuscular junction rather than the conductance mechanism of muscle fibre membrane.  相似文献

The fungicide triarimol was tested for its effect on abscisic acid (ABA) accumulation in growing culturesof Cercospora rosicola. ABA accumulation was reduced by approximately 50% with 10?8 M triarimol. Growth ofC. rosicola, as measured by dry weight accumulation, was inhibited by triarimol concentrations at or greater than 10?7 M. These results are compared with those obtained with clomazone, ancymidol, and paclobutrazol, which inhibit ABA accumulation by 50% at concentrations of 5 × 10?5, 5 × 10?6, and 5 × 10?7 M, respectively. Triarimol, therefore, is among the most potent inhibitors of ABA biosynthesis reported to date. Feeding studies with [14C]mevalonic acid confirmed the inhibition of ABA biosynthesis by 5 × 10?8 M triarimol. These results support previous suggestions that one or more of the steps in the ABA biosynthetic pathway from mevalonic acid is catalyzed by cytochrome P-450. Feeding studies with 1′-deoxy-[2H]-ABA in resuspended cultures ofC. rosicola show that the conversion of this substrate is not inhibited by triarimol.  相似文献

Human leucocyte cell cultures were stimulated to initiate DNA synthesis by phytohemagglutinin and mercuric chloride. Both mitogens enhanced the accumulation of β2-microglobulin in the medium, which was synthesized by lymphocytes. Mercuric chloride promoted the accumulation of this protein optimaly with a concentration (1 × 10?5M) to produce the maximum stimulation of DNA synthesis. Combined use of phytohemagglutinin (50 μg/ml) and mercuric chloride (1 × 10?5M) produced additive effect on both DNA synthesis and β2-microglobulin accumulation. These findings suggest that mercuric ion causes the proliferative response of lymphocytes by a mechanism different from that for the stimulation by phytohemagglutinin.  相似文献

InEquisetum arvense, apogamous sporophytes were produced on medium containing 5×10?6–5×10?8 g/ml kinetin. NAA, IAA, GA3, glucose and saccharose were ineffective for the induction of apogamy. On medium containing 5×10?7–5×10?8 g/ml kinetin, the gametophytes passed into sporophytic structures directly. On medium containing 5×10?6 g/ml kinetin, some gametophytes passed into sporophytic structures directly, and others became a callus-like cell mass from which an apogamoun shoot arose. The results of the morphological observations on them were reported and compared with the sexually produced sporophyes. The apogamous sporophytes induced by 5×10?7 g/ml kinetin were haploid in their nuclear phase and some of those induced by 5×10?6 g/ml kinetin had a tendency to become diploid.  相似文献

The short term uptake of phosphate involving 10 min absorption followed by 5 min desorption, both at 30 °C, in the concentration range 1.0×10?9 to 7.5×10?2 M KH2PO4 by fresh and washed maize (Zea mays L. cv. Ganga Safed-2) roots can be described by a single isotherm having five phases (0 and I–IV) with regularly spaced kinetic constants. Almost identical kinetics were observed in both fresh and washed maize roots. The kinetics of phase 0 in the concentration range 1.0×10?9–3.0×10?5 M. was sigmoidal in fresh maize roots, however, in washed tissue exhibited 2 phases termed here as 0a and 0b. 0a covered the concentration range 1.0×10?9–5.0×10?6 M and 0b 6.0×10?6–3.0×10?5 M. In the concentration range 1.0×10?4–7.5×10?2 M four distinct phases, termed as I, II, III and IV were evident in both fresh and washed maize roots. Each phase obeyed Michaelis—Menten kinetics. The values of Km and Vmax have been estimated for each phase. The uptake isotherm was accompanied by discontinuous transitions.  相似文献

The effect of hormone-like compounds at different concentrations: 2,4-D (2 × 10?6; 2 × 10?5; and 2 × 10?4M) and 1-NAA (2 × 10?7; 2 × 10?6; 2 × 10?5; 4 × 10?5, and 6 × 10?5 M) on the growth and production of phenolic compounds, including flavans and lignin, was investigated in callus culture of tea plant (Camellia sinensis L., a highly productive strain IFR ChS-2). The growth of the culture was vigorous, and production of phenolic compounds therein was efficient in the medium containing 2 × 10?5 M 2,4-D. Substitution of 1-NAA for 2,4-D in all the cases decelerated the growth of the culture. These changes were more pronounced when 2 × 10?7 and 2 × 10?6 M 1-NAA was used; in this case, biomass accumulation decreased by 1.5–2.0 times as compared with control material growing on the medium with 2 × 10?5 M 2,4-D. In the presence of 1-NAA, the content of total soluble phenolic compounds and flavans in the calli rose by 30% on the average as compared with control material. Accumulation of lignin remained essentially the same. Therefore, the replacement of 2,4-D with 1-NAA in the nutrient medium used for the growing of highly productive strain of tea plant callus did not induce considerable changes in its ability to produce phenolic compounds.  相似文献

Each of a series of seven monohydroxyl alcohols caused an increase in the force of the neurally evoked contractions of the innervated retractor unguis muscle isolated from the metathoracic femurs of the locust, Schistocerca gregaria. The negative logarithm of equipotent concentrations of the alcohols was proportional to the logarithm of the partition coefficient (1-octanol/water) of the alcohols: log1C = 1·282 log P+1·684, where C is concentration, and P is the partition coefficient.Ethylene glycol and glycerol did not potentiate the contractions, possibly due to their very low lipophilic character. Acetone (10?2 M) and benzene (10?5 M) also potentiated the neurally evoked contractions of the insect muscle.  相似文献

Rat submandibular gland was dissociated by enzymatic digestion with collagenase and hyaluronidase, followed by mild mechanical shearing and filtration through a nylon mesh. The dissociated cell populations contained predominantly groups of acinar cells which maintained their acinar arrangement. The morphological and functional viability of the cells was confirmed by electron microscopic examination and a normal secretory response to β-adrenergic or cholinergic stimulation was observed. Both isoproterenol (IPR) and carbachol caused the fusion of secretory granules into large vacuoles which were also continuous with the lumen, and into which the secretory product was released. Secretion was assessed quantitatively from the incorporation of 14C-glucosamine into the acinar cells and its subsequent release into the culture medium as labelled glycoprotein. IPR stimulated secretion to 125% of untreated controls in the concentration range 5 × 10?5 to 5 × 10?7 M, and to 110% of controls at 5 × 10?8 M, after 40 min incubation. Carbachol stimulated secretion to 131% of controls at 5 × 10?5 M and to 115% at 5 × 10?6 M but had no effect at 5 × 10?7 or 5 × 10?8 M. The secretory response was blocked by the respective β-adrenergic and cholinergic antagonists, propranolol and atropine. These findings show that dissociated rat submandibular acinar cells provide a useful in vitro model for the study of mucus synthesis and secretion.  相似文献

The activity of concentrated exsheathing fluid of Haemonchus contortus against isolated sheaths was not inhibited by ethylenediamine tetra-acetic acid (EDTA), 10?2 M, even when the concentrations of Mg and Mn were < 4 × 10?4 M and < 0·9 × 10?6 M respectively. Purified or diluted solutions of exsheathing fluid, even in the presence of Mg2+, 10?3 M, were inhibited. Leucine aminopeptidase (LAP) in exsheathing fluid was active even at concentrations of Mg < 1·3 × 10?5M. Concentrated solutions were partially inhibited by EDTA, 10?2 M, at low concentrations of Mg; inhibition was increased in diluted and purified preparations.1,10-phenanthroline (Ophen) strongly inhibited exsheathing activity (Zn < 1 × 10?6 M). When Zn2+, 10?3 M was added, the inhibition was abolished. The hydrolysis of l-leucinamide was greatly increased in the presence of Ophen, 10?4 M; this effect was abolished by adding Zn2+, 10?3 M.It is suggested that exsheathing fluid from at least some ‘strains’ of H. contortus contains a Zn metallo-enzyme, probably LAP, which is involved in the process of exsheathment.  相似文献

Nitrilase was purified ca 28-fold from Chinese cabbage seedlings. Km values of 5.2 × 10?4 and 2.6 × 10?3 M were obtained for indoleacetonitrile (IAN) and 3-cyanopyridine (3-CP) as substrates. For hydrolysis of 3-CP, the maximal velocity was 44 times higher than for the natural substrate IAN. The pH optimum is at 7.5. IAA concentrations from 10?6 to 10?3 M did not inhibit the partially purified enzyme. Nitrilase activity was investigated during development of seedlings grown under continuous light. Roots with hypocotyls exhibited only slightly lower activity than cotyledons based on fresh weight, although their specific activity was ca 5 times higher. Etiolated seedlings showed a very similar distribution of nitrilase activity. The significance of the results for IAA biosynthesis is discussed.  相似文献

Acetylcholine (ACh) was found to markedly enhance the nerve stimulation induced twitch response of isolated, field-stimulated rat vas deferens (RVD). The ED200 (concentration which enhances the twitch response to 200% of control) for this potentiation was 6 × 10?6M with the maximum twitch response being increased by more than 3 fold (325 ± 30%). Carbachol (ED200 = 8.5 × 10?7M) showed identical results. With each drug the potentiation was competitively antagonized by atropine (10?7?10?5M). Physostigmine 10?8?10?6M) both enhanced the basal twitch response (215 ± 8% of control at 10?5M) and the sensitivity of the RVD to ACh (ED200 = 3.3 × 10?7M) but not to carbachol. Atropine, on the other hand reduced the basal twitch response by 18 ± 3% at 10?5M. Hemicholinium (10?4M) also reduced the basal twitch responses by 23 ± 5%. ACh (10?7M?10?5M) did not modify the responses of unstimulated RVD to norepinephrine or KCl suggesting a pre-synaptic site of action. Taken together these results are compatible with the presence of a pre-junctional, excitatory muscarinic mechanism in the field stimulated RVD. That this cholinergic system may be of physiological significance is supported by the observations that atropine and hemicholinium depress while physostigmine enhances the twitch response in the absence of exogenous ACh.  相似文献

The effects of varying concentrations of Fe2+ (5 × 10?5 ?5 × 10?1 M) on benzylglucosinolate degradation in Lepidium sativum seed autolysates were investigated. Increased glucosinolate decomposition was observed over the whole range with a maximum effect at ca 6 × 10?3 M Fe2+, at which point glucosinolate degradation was more than three times that obtained in the absence of added Fe2+ . Nitrile formation was especially enhanced in the presence of all concentrations of Fe2+ studied, and maximum amounts were obtained at ca 6 × 10?3 M Fe2+ when a more than four-fold increase over quantities produced in the absence of Fe2+ was observed. Thiocyanate formation was also promoted with a maximum at ca 4 × 10?3 M Fe2+, but isothiocyanate production was considerably reduced in allcases. It is suggested that Fe2+ inhibits isothiocyanate formation by interfering with the availability of ascorbic acid which is a proven co-factor for most thioglucosidase isoenzymes, but that an Fe2+-ascorbate complex might then be responsible for promoting enzymic production of nitrile. The effects of a limited range of concentrations of Fe3+ and Cu+ were also studied, and results related to those for Fe2+. The relevance of the findings to natural systems and to glucosinolate-containing foods is briefly discussed.  相似文献

β-Bungarotoxin and taipoxin are snake venoms that have been reported to induce neuromuscular blockade exclusively in somatic motor neurons by a presynaptic mechanism. Taipoxin (1 μg/ml), but not β-bungarotoxin (1 μg/ml), depressed the contractile response of canine airway smooth muscle to electrical stimulation. Taipoxin (1 μg/ml) also slightly depressed the contractile activity of canine airways to two concentrations (5 × 10?6M and 10?5M) of exogenously administered acetylcholine. We conclude that taipoxin, but not β-bungarotoxin, induces a weak neuromuscular blockade in the parasympathetic fibers innervating canine airways. The sites of this inhibition are believed to be both presynaptic and postsynaptic.  相似文献

Morphine, met-enkephalin, and leu-enkephalin in a concentration of 1×10?5 M depress rapidly and reversibly the amplitude of depolarization induced by dopamine application toHelix pomatia neurons; the effect is naloxone-dependent. The amplitudes of dopamine-induced hyperpolarization and also of the depolarization and hyperpolarization responses to acetylcholine application are unchanged under these circumstances. The hypothesis of blocking of chemosensitive sodium channels by enkephalins is discussed. It is suggested that this hypothesis is true for high concentrations of morphine and enkephalins (1×10?4 to 1×10?3 M). In lower concentrations (1×10?5 M) morphine and enkephalins lead to modulation of the reponses to the action of neurotransmitters, evidently through their influence on the cyclic nucleotide system.  相似文献
