The giant panda is an example of a species that has faced extensive historical habitat fragmentation, and anthropogenic disturbance and is assumed to be isolated in numerous subpopulations with limited gene flow between them. To investigate the population size, health, and connectivity of pandas in a key habitat area, we noninvasively collected a total of 539 fresh wild giant panda fecal samples for DNA extraction within Wolong Nature Reserve, Sichuan, China. Seven validated tetra‐microsatellite markers were used to analyze each sample, and a total of 142 unique genotypes were identified. Nonspatial and spatial capture–recapture models estimated the population size of the reserve at 164 and 137 individuals (95% confidence intervals 153–175 and 115–163), respectively. Relatively high levels of genetic variation and low levels of inbreeding were estimated, indicating adequate genetic diversity. Surprisingly, no significant genetic boundaries were found within the population despite the national road G350 that bisects the reserve, which is also bordered with patches of development and agricultural land. We attribute this to high rates of migration, with four giant panda road‐crossing events confirmed within a year based on repeated captures of individuals. This likely means that giant panda populations within mountain ranges are better connected than previously thought. Increased development and tourism traffic in the area and throughout the current panda distribution pose a threat of increasing population isolation, however. Maintaining and restoring adequate habitat corridors for dispersal is thus a vital step for preserving the levels of gene flow seen in our analysis and the continued conservation of the giant panda meta‐population in both Wolong and throughout their current range.  相似文献

Sequence variation and genetic diversity in the giant panda   总被引:3,自引:0,他引:3  
About 336–444 bp mitochondrial D-loop region and tRNA gene were sequenced for 40 individuals of the giant panda which were collected from Mabian, Meigu, Yuexi, Baoxing, Pingwu, Qingchuan, Nanping and Baishuijiang, respectively. 9 haplotypes were found in 21 founders. The results showed that the giant panda has low genetic variations, and that there is no notable genetic isolation among geographical populations. The ancestor of the living giant panda population perhaps appeared in the late Pleistocene, and unfortunately, might have suffered bottleneck attacks. Afterwards, its genetic diversity seemed to recover to some extent. 2007年10月和2008年4月,对佛坪自然保护区和观音山自然保护区内大熊猫的活动痕迹进行了样线调查,研究了该区内大熊猫种群及其同域主要伴生动物的分布情况,并分析了影响大熊猫活动分布格局的环境因素.结果表明:观音山和佛坪自然保护区的大熊猫及同域主要伴生野生动物的分布格局基本一致,大熊猫在观音山自然保护区的活动痕迹密度、范围均小于佛坪自然保护区;研究区内2个大熊猫高密度等级的活动中心均分布在佛坪自然保护区内;观音山自然保护区内的大部分地区无大熊猫活动;羚牛、斑羚、野猪等大熊猫主要伴生动物在观音山自然保护区内的活动痕迹数量高于佛坪保护区;人类干扰可能对大熊猫种群活动的分布格局产生影响.  相似文献

气候变化对邛崃山系大熊猫主食竹和栖息地分布的影响   总被引:1,自引:0,他引:1  
气候变化对生物多样性的影响,特别是珍稀濒危物种的影响是当前的研究热点。全球气候变化对大熊猫的影响一直受到广泛关注。根据野外调查的大熊猫活动痕迹点、竹类分布点和主食竹扩散距离数据,采用Maxent模型,利用植被、地形、气候等因素,在RCP8.5下分析了2050年和2070年邛崃山系大熊猫主食竹分布及栖息地变化趋势。结果显示:(1)未来大熊猫适宜生境及主食竹气候适宜区面积均有所减少,到2070年分别减少37.2%和4.7%;(2)未来主食竹分布范围总体向高海拔扩展,但面积持续减少,到2070年分布面积比当前减少8.3%;(3)大熊猫栖息地未来有向高海拔扩张的趋势,在低海拔地区退缩明显,到2070年较当前减少27.2%;但到2070年大熊猫栖息地面积加上非栖息地有主食竹分布的面积,较现有大熊猫栖息地面积大1.5%;(4)受气候变化影响较严重的区域是邛崃山系南部以及低海拔地区,其余区域所受影响相对较小;(5)未来需要加强对受气候变化影响严重区域的监测与保护,特别是邛崃山系中部的大熊猫集中分布区。  相似文献

种群扩散格局是研究种群扩散规律和机制的关键信息,也是制定物种保护对策的重要基础。大型动物种群扩散格局研究方法为扩散生态学研究的薄弱领域,并制约扩散生态学的发展。以秦岭大熊猫为研究对象,根据2000年以来的种群调查数据,基于大熊猫领域的特性,利用GIS的扩展区分析功能和景观分析方法研究了大熊猫种群分布区及动态;基于聚集的特性,利用GIS的核密度分析功能对大熊猫种群多度和聚集状况及空间变化进行了分析。发现2012年的秦岭大熊猫种群分布区较2000年增加5.5%(即15307.8hm~2),高密度种群聚集区从2处变成1处,种群聚集程度进一步增加、聚集格局的完整性大大提升,尤以中密度聚集区增长最显著,种群格局呈明显的分布区扩张、聚集度增加的态势。表明基于物种的生物学特性,立足于种群分布和多度格局变化,通过长期调查和监测可以有效掌握物种的种群扩散格局;大型动物可根据其生物学特性探索可行的方法与量化种群扩散的参数来研究其扩散格局,从而促进大型动物种群扩散研究的开展。  相似文献

大熊猫肠道纤维素分解菌的分离鉴定及产酶性质   总被引:3,自引:0,他引:3  
【目的】从健康大熊猫新鲜粪便中分离具有纤维素酶活性的菌株,并对其进行菌种鉴定及产酶性质研究。【方法】利用羧甲基纤维素钠培养基分离纯化具有较高纤维素酶活性的菌株,根据形态学特征、生理生化特性以及16S rDNA分析对其进行分类鉴定,研究影响该菌株纤维素酶的产酶条件,以及对不同纤维素底物的降解情况。【结果】分离得到一株纤维素酶产生菌株P2,该菌株为好氧的革兰氏阳性细菌,生长温度范围20-50℃(最适温度37℃),pH范围6.0-9.0(最适pH7.0),NaCl浓度范围0%-15%(最适2%NaCl),培养24h达到产酶高峰。16S rDNA基因序列分析显示,菌株P2与解淀粉芽胞杆菌(Bacillusamyloliquefaciens)NBRC15535相似性为99.66%。该菌株对四种纤维素底物(滤纸、脱脂棉、秸秆、竹纤维)均有不同程度的降解,内切葡聚糖酶、外切葡聚糖酶、β-葡萄糖苷酶和总酶活具有不同的酶活变化。【结论】本研究首次从大熊猫粪便中分离出了好氧纤维素分解菌,并鉴定为解淀粉芽胞杆菌,对上述四种纤维结构均有一定的破坏和分解作用,为进一步研究大熊猫竹纤维消化机制提供了菌源。  相似文献

理清自然保护地的空间关系与分布格局是加强空间管控、整合优化自然保护地体系的基础。以大熊猫国家公园四川片区内的自然保护地为案例,基于ArcGIS空间数据的处理、分析与可视化表达等功能,结合韦恩(Venn)图在空间层面上量化分析了公园范围内各类自然保护地的空间关系,并进一步揭示了不同保护情景下大熊猫(Ailuropoda melanoleuca)的分布格局。研究结果表明:(1)研究区内含有6类自然保护地,占研究区总面积的75.13%,其中40.68%为交叉重叠区域。(2)各类自然保护地皆存在大面积的交叉重叠。自然保护区为研究区面积最大的自然保护地类型,占自然保护地总面积的72.53%,其中45.89%为交叉重叠区域;其他类自然保护地占自然保护地总面积的60.87%,其中66.48%为交叉重叠区域。(3)猫点密度与自然保护地的交叉重叠程度呈现逆向增长趋势,区域的重叠水平越高,猫点密度越低。(4)自然保护地整体非重叠区的猫点密度高于重叠区。自然保护区是整体猫点密度最高的自然保护地类型,其非重叠区密度明显高于重叠区;森林公园非重叠区与水利风景区重叠区呈现较高的猫点密度。(5)与自然保护区交叉重叠...  相似文献

Nonlactating female giant pandas experience a single estrus during the spring mating season. Although both hormonal and behavioral aspects of estrus have been reported on captive females, little is known about the relationship between them. Data were collected on a single captive female during three successive seasons, revealing a high degree of regularity in both estrogen profiles and the temporal pattern of associated behaviors. The birth of a live cub in the last of the three seasons, using artificial insemination, confirmed the time of ovulation as occurring shortly after a peak in urinary estrogen values. In this study, we examined the occurrence of female scent marking, rear presentations, chirp and bleat vocalizations, and the tail‐up display, and plotted their occurrence relative to the underlying estrogen values. We conclude that the role of estrogen in the expression of these behaviors is neither simple nor directly causal. Temporal associations indicate that a triggering rather than a maintenance function for estrogen is implicated. This analysis provides a clear identification of estrous behaviors, but in both the time and form of occurrence these behaviors most likely vary between females and seasons. 综合运用大熊猫生物学与行为生态学研究成果、遥感数据分析与地理信息系统技术 ,在系统研究岷山地区大熊猫生境分布、生境质量与空间格局的基础上 ,明确岷山地区保护大熊猫的关键区域 ,分析岷山地区大熊猫保护与自然保护区建设的对策 ,以期为岷山地区大熊猫保护及其与岷山地区资源开发与发展的协调提供依据。遥感数据分析结果表明 ,岷山地区以森林为主 ,各类森林面积 1 91 790 3.4 4 hm2 ,占 5 5 .4 2 % ,将与大熊猫生境密切相关的森林植被分为常绿阔叶林、落叶阔叶林、针阔混交林、亚高山针叶林等 ,其中亚高山针叶林面积 5 380 4 9.6 4 hm2 ,占全部森林的占 2 8.0 5 % ,落叶阔叶林面积 4 6 1 35 5 .6 7hm2 ,占2 4 .0 6 % ,针阔混交林面积 4 0 30 36 .2 6 hm2 ,占 2 1 .0 1 %。结果表明岷山地区有大熊猫潜在生境 1 32 3789.1 5 hm2 ,由于森林资源利用、交通、农业活动影响、居民薪柴砍伐 ,以及生境破碎化与生境隔离等导致的生境丧失 5 5 341 3.4 5 hm2 ,目前尚存的大熊猫适宜生境 770 375 .7hm2 ,由于交通等隔离而成为至少 5个相互分隔的生境单元 ,大熊猫种群交流受到严重阻碍。为了有效地保护岷山大熊猫 ,首先应充分注意到各种人类活动 ,尤其交通的建设对大熊猫生境的影响 ;其次要扩大自然保护区范围 ,  相似文献

When Chu‐Lin, a male giant panda (studbook #249), died at Madrid Zoo, his reproductive tract was removed 4 hr postmortem and the epididymal spermatozoa were collected. Extended sperm were kept at 5°C for 4 hr, loaded into straws, and frozen for 7 min in liquid nitrogen vapor before the straws were plunged into liquid nitrogen. Two straws were thawed and evaluated. Sperm motility was assessed in fresh, refrigerated, and thawed spermatozoa (75%, 60%, 35%, respectively). Sperm viability and acrosome status were estimated using a triple‐stain technique (TST). The results showed 33% live sperm with intact acrosomes after thawing. A hypoosmotic swelling (HOS) test demonstrated the retention of membrane integrity in 72% of thawed sperm. To evaluate the in vitro fertilizing ability of thawed sperm, a sperm penetration assay (SPA) was performed. The values obtained for the percentage of penetration and the penetration index were 62% and 1.78 sperm/oocyte, respectively. The results obtained demonstrate that epididymal sperm recovered from a giant panda postmortem can be successfully cryopreserved. The sperm fertilizing ability demonstrated in vitro after thawing may provide a final opportunity for this male to contribute to the currently small germplasm reserves of this endangered species, and to reproduce in the future through assisted reproductive technology. Chronic pain is a classic example of gene × environment interaction: inflammatory and/or nerve injuries are known or suspected to be the etiology of most chronic pain syndromes, but only a small minority of those subjected to such injuries actually develop chronic pain. Once chronic pain has developed, pain severity and analgesic response are also highly variable among individuals. Although animal genetics studies have been ongoing for over two decades, only recently have comprehensive human twin studies and large-scale association studies been performed. Here, I review recent and accelerating progress in, and continuing challenges to, the identification of genes contributing to such variability. Success in this endeavor will hopefully lead to both better management of pain using currently available therapies and the development and/or prioritizing of new ones.  相似文献

大熊猫是生态保护的旗舰物种,保护大熊猫及其栖息地具有重要的示范作用和科学价值。中国致力于建设以国家公园为主体的自然保护地体系,分析大熊猫保护地生态安全,为建立大熊猫国家公园提供理论依据,具有重要意义。以四川省为例,选取被划入或将来可能被划入大熊猫保护地的典型县域,构建大熊猫保护地生态安全评价指标体系,采用熵权法、层次分析法进行综合评价,并结合空间相关性分析,从时间和空间两个维度对大熊猫保护地生态安全状况进行时空动态分析。结果表明:(1)2003-2017年四川省大熊猫保护地生态安全综合指数呈波动性上升趋势,生态环境改善显著,但仍有部分县域出现不同程度的环境恶化现象。(2)各县域生态安全状况表现出较强的差异性,并且相邻县域生态安全指数存在显著的空间正相关性和空间集聚性。(3)大熊猫保护地生态安全与周边社会经济发展之间矛盾依然尖锐,相邻县域之间的经济发展和生态保护状况相互影响的作用明显,需联合治理,共同促进生态平衡,提高大熊猫及其栖息地保护效果。  相似文献

Understanding the links between genetic variation and fitness in natural populations is a central goal of evolutionary genetics. This monumental task spans the fields of classical and molecular genetics, population genetics, biochemistry, physiology, developmental biology, and ecology. Advances to our molecular and developmental toolkits are facilitating integrative approaches across these traditionally separate fields, providing a more complete picture of the genotype‐phenotype map in natural and non‐model systems. Here, we summarize research presented at the first annual symposium of the UNVEIL Network, an NSF‐funded collaboration between the University of Montana and the University of Nebraska, Lincoln, which took place from the 1st to the 3rd of June, 2018. We discuss how this body of work advances basic evolutionary science, what it implies for our ability to predict evolutionary change, and how it might inform novel conservation strategies.  相似文献

We describe 10 polymorphic microsatellite loci for the giant panda, Ailuropoda melanoleuca. Microsatellite sequences were isolated from three partial genomic libraries of giant panda DNA that were enriched for (i) (GT), (ii) (GAA) & (CAA), and (iii) (GATA) repeat sequences. The markers were tested for polymorphism in up to 82 pandas. Number of alleles at each locus varied between four and 11, and the observed and expected heterozygosities varied between 0.267 and 0.732, and between 0.601 and 0.799, respectively.  相似文献

These BACs could be used as a validation resource for the de novo assembly accuracy of the whole genome shotgun sequencing reads of giant panda newly generated by the Illumina GA sequencing technology. Complete sanger sequencing, assembly, annotation and comparative analysis were carried out on the selected BACs of a joint length 878 kb. Homologue search and de novo prediction methods were used to annotate genes and repeats. Twelve protein coding genes were predicted, seven of which could be functionally annotated. The seven genes have an average gene size of about 41 kb, an average coding size of about 1.2 kb and an average exon number of 6 per gene. Besides, seven tRNA genes were found. About 27 percent of the BAC sequence is composed of repeats. A phylogenetic tree was constructed using neighbor-join algorithm across five species, including giant panda, human, dog, cat and mouse, which reconfirms dog as the most related species to giant panda. Our results provide detailed sequence and structure information for new genes and repeats of giant panda, which will be helpful for further studies on the giant panda.  相似文献

Understanding the impacts of meteorological factors on giant pandas is necessary for future conservation measures in response to global climate change. We integrated temperature data with three main habitat parameters (elevation, vegetation type, and bamboo species) to evaluate the influence of climate change on giant panda habitat in the northern Minshan Mountains using a habitat assessment model. Our study shows that temperature (relative importance = 25.1%) was the second most important variable influencing giant panda habitat excepting the elevation. There was a significant negative correlation between temperature and panda presence (ρ = −0.133, P < 0.05), and the temperature range preferred by giant pandas within the study area was 18–21°C, followed by 15–17°C and 22–24°C. The overall suitability of giant panda habitats will increase by 2.7%, however, it showed a opposite variation patterns between the eastern and northwestern region of the study area. Suitable and subsuitable habitats in the northwestern region of the study area, which is characterized by higher elevation and latitude, will increase by 18007.8 hm2 (9.8% habitat suitability), while the eastern region will suffer a decrease of 9543.5 hm2 (7.1% habitat suitability). Our results suggest that increasing areas of suitable giant panda habitat will support future giant panda expansion, and food shortage and insufficient living space will not arise as problems in the northwest Minshan Mountains, which means that giant pandas can adapt to climate change, and therefore may be resilient to climate change. Thus, for the safety and survival of giant pandas in the Baishuijiang Reserve, we propose strengthening the giant panda monitoring program in the west and improving the integrity of habitats to promote population dispersal with adjacent populations in the east.  相似文献

大熊猫作为国家保护动物,其健康问题备受瞩目。为了维护大熊猫的肠道健康,本研究从大熊猫肠道内分离出适宜于大熊猫肠道环境的乳酸菌菌株,有望将其制成熊猫肠道微生物制剂,从而改善大熊猫肠道菌群环境。从雅安市宝兴县蜂桶寨自然保护区选取圈养与野生大熊猫的粪便,通过体外培养分离出9个菌株。分离菌株经过革兰氏染色镜检、过氧化氢产气、菌落形态观察等方法与技术初步鉴定为乳酸菌。对这9株乳酸菌进行耐酸试验、耐胆盐试验、抑菌能力试验和产酸能力等测试,筛选出了3个适应性较强,有望制成调节大熊猫肠道内环境平衡作用的微生态菌剂的菌株。16S rRNA基因序列分析表明:分离菌株J1、J2和J4分别为融合魏斯氏菌(Weissella confusa),海氏肠球菌(Enterococcus heynei)和非解乳糖链球菌(Streptococcus alactolyticus),有望被应用于大熊猫肠道微生态制剂的研究。  相似文献
