Considerable variations exist in the content of glucose, fructose, sucrose, starch and protein and in the activities of enzymes involved in carbohydrate metabolism between different parts of the banana plant (Musa paradisiaca). Sucrose synthetase is present in the highest concentration in rootstock and fruit pulp, and sucrose phosphate synthetase in the pseudostem. The highest ratio of the activity of sucrose phosphate synthetase to sucrose synthetase is found in leaves. Acid invertase is present in leaves, leaf-sheath and fruit pulp and is not demonstrable in rootstock and pseudostem. Neutral invertase activity is high in pseudostem and leaf-sheath. Starch phosphorylase is largely concentrated in fruit pulp and rootstock. The maximum activity of ATP:d-phosphoglucose (ADPG) pyrophosphorylase is found in rootstock. β-Amylase is not demonstrable in rootstock and is largely concentrated in leaf-sheath. Hexokinase is most active in rootstock and the lowest in leaves. Acid phosphatase and alkaline phosphatase activity is highest in fruit pulp and pseudostem. Glucosephosphate isomerase is most active in the rootstock and lowest in the leaves.  相似文献

The possible involvement of acid invertase (sucrose hydrolysis) as a prerequisite for sucrose mobilization from the vacuole of storage cells was investigated. Sugarcane ( Saccharum officinarum ) stalks, carrot ( Daucus carota ) roots and red beet ( Beta vulgaris ) hypocotyls were planted under greenhouse conditions and allowed to resume growth. The plants, however, were not permitted to become photosynthetically autotrophic by removing the new expanded leaves. Sucrose levels declined significantly in all three tissues without the development of acid invertase (EC 3.2.1.26) during the 21‐day experimental period. Acid invertase and thus sucrose hydrolysis within the vacuole was, therefore, not required for sucrose mobilization.  相似文献

The free sugar in the mature pollen grains of Camellia japonica is almost all sucrose and the sucrose content decreases rapidly during pollen growth. The activity of soluble invertase increases during culturing and a high constant activity was found at the later stages of pollen tube growth. By contrast, the level of sucrose synthetase activity remains constant during pollen growth and that of wall-bound invertase activity is very low. Cycloheximide has little effect on the activity of these enzymes. Exogenous sucrose or glucose was simultaneously incorporated into the pollen grains when they absorbed water and swelled. The free sugar levels in growing pollen depend on the nature of the exogenous sugar. The sugar metabolism in the pollen at the stage of germination differs from that during tube growth, the latter being particularly influenced by exogenous sugar.  相似文献

The organ topography of sucrose synthase and soluble acid invertase in pea seedlings at heterotrophic stage (3–14 days) was studied. Sucrose synthase was most active in the roots, with the highest activity on the 6–8th days. In the leaves, its activity decreased from day 3 to day 14. In the stems, sucrose synthase activity was at an invariantly low level. The patterns of sucrose synthase activity in etiolated and green plants were similar. As distinct from sucrose synthase, invertase activity was the highest in the stem, especially in etiolated plants. The peak of its activity was observed on the 6-8th days. In the leaves, invertase activity was lower but its pattern was the same. In the roots, acid invertase activity decreased from the 3rd day and did not depend on illumination. The conclusion is that differences in sucrose synthase and acid invertase activities in roots, leaves, and stem are determined by differences in the import of hydrolytic products of stored compound from the cotyledons as well as by different demands of these organs for these products for the processes of organ expansion and for the maintenance of organ metabolism.  相似文献

The aim of this study was to investigate carbohydrate metabolism in rice seedlings subjected to salt-alkaline stress. Two relatively salt-alkaline tolerant (Changbai 9) and sensitive (Jinongda 138) rice cultivars, grown hydroponically, were subjected to salt-alkaline stress via 50 mM of salt-alkaline solution. The carbohydrate content and the activities of metabolism-related enzymes in the leaves and roots were investigated. The results showed that the contents of sucrose, fructose, and glucose in the leaves and roots increased under salt-alkaline stress. Starch content increased in the leaves but decreased in the roots under salt-alkaline stress. The activities of sucrose-phosphate synthase, sucrose synthase, amylase, and ADP-glucose pyrophosphorylase increased whereas the activities of neutral invertase and acid invertase decreased in the leaves under salt-alkaline stress. The activities of sucrose-phosphate synthase, sucrose synthase, amylase, neutral invertase, and acid invertase increased in the roots under salt-alkaline stress. In conclusion, salt-alkaline stress caused the accumulation of photosynthetic assimilates in the leaves and decreased assimilation export to the roots.  相似文献

The aim of this work was to relate [14C]-sucrose metabolism to the activities of sucrose synthetase and acid and alkaline invertases in roots of Pisum sativum. We fed [U-14C]-sucrose to 5-day-old plants and then excised the apical 6 mm of the roots and dissected the regions 6–24 mm from the root apices into stele and cortex. The detailed distribution of 14C in these parts of the roots was determined at the end of the feeding period and after a chase. The data indicate that sucrose arriving in the stele is divided between storage, conversion to polysaccharide, and consumption in respiration, whereas sucrose arriving in the rest of the root is used in respiration or converted to polysaccharide or hexose so rapidly that little is stored. Fractionation of carefully prepared extracts of pea roots, tubers of Solanum tuberosum, and spadices of Arum maculatum showed that sucrose synthetase was recovered in the soluble fraction. The results are discussed in relation to the roles of the aforementioned enzymes.  相似文献

Activities of acid and alkaline invertases and sucrose synthase were determined in roots and nodules of lentil at various stages of development. Alkaline invertase and sucrose synthase were both involved in sucrose metabolism in the nodule cytosol, but there was only a small amount of acid invertase present. Activity of sucrose metabolizing enzymes in roots was significantly less than that observed in the nodules. Amongst sugars, sucrose was found to be the main component in the host cytosol. Lentil neutral invertase (LNI) was partially purified from nodules at 50 days after sowing (DAS). Two forms of invertase were identified, i.e., a major form of 71 kDa which was taken for enzyme characterization and a minor form of 270 kDa which was not used for further studies. The purified enzyme exhibited typical hyperbolic saturation kinetics for sucrose hydrolysis. It had a Km of 11.0 to 14.0 mM for sucrose depending upon the temperature, a pH optimum of 6.8 and an optimum temperature of 40 °C. Compared with raffinose and stachyose, sucrose was better substrate for LNI. The enzyme showed no significant hydrolysis of maltose and p-nitrophenyl--D-glucopyranoside, showing its true -fructosidase nature. LNI is completely inhibited by HgCl2, MnCl2 and iodoacetamide but not by CaCl2, MgCl2 or BaCl2.  相似文献

Stele and cortex were separated from the region 6–24 mm from the tip of roots of seedlings of Pisum sativum L. that had been grown in the dark for 5 days. The activities of sucrose synthetase (E.C. 2.4.1.13) and sucrose phosphate synthetase (E.C. 2.4.1.14) in extracts of stele were 34 and 5·9 nmol product formed/min per mg protein, respectively. The corresponding figures for extracts of cortex were 17 and 5·2. Appreciable labelling of sucrose occurred when samples of either stele or cortex were incubated in [14C]glucose for 90 min. The labelling of sucrose after incubation of the tissues for 45 min in [14C]glucose followed by 45 min in glucose suggested some turnover of sucrose in the cortex but none in the stele. These results are discussed in relation to the control of sucrose metabolism in the root.  相似文献

Hydroponic culture was used to comparatively investigate the copper (Cu)-induced alteration to sucrose metabolism and biomass allocation in two Elsholtzia haichowensis Sun populations with one from a Cu-contaminated site (CS) and the other from a non-contaminated site (NCS). Experimental results revealed that biomass allocation preferred roots over shoots in CS population, and shoots over roots in NCS population under Cu exposure. The difference in biomass allocation was correlated with the difference in sucrose partitioning between the two populations. Cu treatment (45 μM) significantly decreased leaf sucrose content and increased root sucrose content in CS population as a result of the increased activities of leaf sucrose synthesis enzymes (sucrose phosphate synthetase and sucrose synthase) and root sucrose cleavage enzyme (vacuolar invertase), which led to increased sucrose transport from leaves to roots. In contrast, higher Cu treatment increased sucrose content in leaves and decreased sucrose content in roots in NCS population as a result of the decreased activities of root sucrose cleavage enzymes (vacuolar and cell wall invertases) that led to less sucrose transport from leaves to roots. These results provide important insights into carbon resource partitioning and biomass allocation strategies in metallophytes and are beneficial for the implementation of phytoremediation techniques.  相似文献

Accumulation of 60–70 % of biomass in turnip root takes place between 49–56 days after sowing. To understand the phenomenon of rapid sink filling, the activities of sucrose metabolising enzymes and carbohydrate composition in leaf blades, petiole and root of turnip from 42–66 days of growth were determined. An increase (2–3 folds) in glucose and fructose contents of roots accompanied by an increase in activities of acid and alkaline invertases was observed during rapid biomass accumulating phase of roots. The observed decrease in the activities of acid and alkaline invertases along with sucrose synthase (cleavage) in petiole during this period could facilitate unrestricted transport of sucrose from leaves to the roots. During active root filling period, a decrease in sucrose synthase (cleavage) and alkaline invertase activities was also observed in leaf blades. A rapid decline in the starch content of leaf blades was observed during the phase of rapid sink filling. These metabolic changes in the turnip plant led to increase in hexose content (35–37 %) of total dry biomass of roots at maturity. High hexose content of the roots appears to be due to high acid invertase activity of the root.  相似文献

Zinc deficiency caused an accumulation of 14C into malic acid, sugar phosphates, sugar nucleotides, glucose, fructose, phosphoenolpyruvate, glycine and alanine, whereas the 14C labelling in sucrose decreased. The activity of sucrose synthetase (EC 2.4.1.13) was unaffected up to the 15th day and thereafter it declined. Severe Zn deficiency reduced the biosynthesis of total protein and sucrose synthetase by 50 and 20%, respectively.  相似文献

Leaf sucrose, starch, hexose and maximum extractable soluble acid invertase activity were compared throughout the day in source leaves of 13 plant species chosen for their putative phloem-loading type (apoplastic or symplastic). Four species which represent the different phloem-loading types (tomato, barley, maize and Fuchsia ) were studied in detail. Using this information we wished to determine whether a positive correlation between foliar carbohydrates and acid invertase activity exists in leaves from different species and, furthermore, whether this relationship is determined by phloem-loading type. Acid invertase activity was relatively constant throughout the day in all species. The extent of sucrose, hexose and starch accumulation and the sucrose: starch ratio measured at a given time were species-dependent. No correlations were found between foliar soluble acid invertase activity and the hexose, sucrose or starch content of the leaves in any of the species, regardless of phloem-loading type. The species examined could be divided into three distinct groups: (1) high sucrose, low invertase; (2) low sucrose, low invertase; and (3) low sucrose, high invertase. The absence of an inverse relationship between leaf sucrose, hexose or starch contents and endogenous soluble acid invertase suggests that this enzyme is not directly involved in carbon partitioning in leaves but serves an auxiliary function.  相似文献

Cadmium (Cd) uptake effects on sucrose content, invertase activities, and plasma membrane functionality were investigated in Rangpur lime roots ( CITRUS LIMONIA L. Osbeck). Cadmium accumulation was significant in roots but not in shoots and leaves. Cadmium produced significant reduction in roots DW and increment in WC. Leaves and shoots did not show significant differences on both parameters. Sucrose content was higher in control roots than in Cd-exposed ones. Apoplastic sucrose content was much higher in Cd-exposed roots than in control ones. Cd-exposed roots showed a significant decrease in both cell wall-bound and cytoplasmic (neutral) invertase activities; while the vacuolar isoform did not show any change. Alterations in lipid composition and membrane fluidity of Cd-exposed roots were also observed. In Cd-exposed roots phospholipid and glycolipid contents decreased about 50 %, while sterols content was reduced about 22 %. Proton extrusion was inhibited by Cd. Lipid peroxidation and proton extrusion inhibition were also detected by histochemical analysis. This work's findings demonstrate that Cd affects sucrose partitioning and invertase activities in apoplastic and symplastic regions in Rangpur lime roots as well as the plasma membrane functionality and H (+)-ATPase activity.  相似文献

The reducing sugar content of sugar beet (Beta vulgaris L.) roots increased during 30 days of storage at 21 C and 160 days at 5 C as a result of an increase in acid invertase activity. Sucrose synthetase and neutral invertase activities were high at harvest but declined during storage, thus showing no relationship with postharvest reducing sugar accumulation in sugar beet roots. Acid α-glucosidase activity was detected in fresh roots but showed no activity with sucrose as a substrate.  相似文献

The major carbohydrates stored in carrots are sucrose, glucose and fructose. The ratio of sucrose to reducing sugars varies between cultivars, with early forcing types generally having a higher level of reducing sugars while storage types have a greater proportion of sucrose.In an early forcing cultivar, Super Sprite, high acid invertase activity was correlated with low levels of stored sucrose. As acid invertase activity decreased, the levels of reducing and non-reducing sugars appeared to be related to a balance between alkaline invertase and sucrose synthetase activities.Foliar applications of gibberellic acid at 35 and 42 days after sowing reduced the root/shoot ratio while similar applications of chlormequat chloride marginally increased the ratio. Both growth regulators temporarily increased sucrose stograge, but only gibberellic acid consistently reduced hexose accumulation.Gibberellic acid reduced acid invertase activity following both applications while only reducing the activities of sucrose synthetase after the first application and alkaline invertase after the second application, respectively. Chloremequat chloride increased acid invertase activity after the first application but otherwise has no effect on the activities of the enzymes studied. The significance of changes in assimilate partitioning are discussed in relation to published schemes on carbohydrate storage in root vegetables.  相似文献

Enhanced amylase activity was observed during a 7-day-growth period in the cotyledons of PEG imposed water stressed chickpea seedlings grown in the presence of GA3 and kinetin, when compared with stressed seedlings. During the first 5 days of seedling growth, the seedlings growing under water deficit conditions as well as those growing in the presence of PGRs had a higher amylase activity in shoots than that of control seedlings. Neither GA3 nor kinetin increased the amylase activity of roots whereas IAA reduced root amylase activity. Activity of acid and alkaline invertases was maximum in shoots and at a minimum in cotyledons. Compared with alkaline invertase, acid invertase activity was higher in all the tissues. The reduced acid and alkaline invertase activities in shoots of stressed seedlings were enhanced by GA3 and kinetin. Roots of stressed seedlings had higher alkaline invertase activity and GA3 and IAA helped in bringing the level near to those in the controls. GA3 and kinetin increased the sucrose synthase (SS) and sucrose phosphate synthase (SPS) activities in cotyledons of stressed seedlings, whereas they brought the elevated level of SPS of stressed roots to near normal level. The higher level of reducing sugars in the shoots of GA3 and kinetin treated stressed seedlings could be due to the high acid invertase activity observed in the shoots, and the high level of bound fructose in the cotyledons of stressed seedlings could be due to the high activity of SPS in this tissue.  相似文献

A simple method of growing plants in agar was exploited to investigate the effect of long-term nitrogen (N) and phosphorus (P) deficiencies on respiratory metabolism and growth in shoots and roots of Nicotiana tabacum seedlings, and their interaction with exogenously supplied sucrose. Levels of hexose phosphates and 3-phosphoglyceric acid (3-PGA) were low in P-deficient shoots and roots and high in N-deficient shoots and roots. The ratio of hexose phosphates to 3-PGA and levels of fructose-2,6-bisphosphate were high in P-deficient plants and low in N-deficient plants. These data reflect differences in the way metabolism was perturbed, yet both deficiencies were associated with increased root growth relative to shoot growth, starch accumulation in the shoots, and soluble carbohydrate accumulation, especially hexoses, in the roots. Enzymes for sucrose degradation (sucrose synthase, acid and alkaline invertase) and glycolysis (phosphofructokinase, pyrophosphate-dependent phospho-fructokinase and pyruvate kinase) remained unaltered or declined in the shoots and roots. The accumulation of hexoses in roots of N- and P-deficient plants may result from maintenance of high invertase activities relative to sucrose synthase and glycolytic enzymes in the roots. The possibility that hexose accumulation may drive preferential root growth osmotically in N and P deficiencies is discussed. The addition of sucrose to roots to further investigate the interaction of carbohydrates with growth and allocation in low N and low P produced clear effects even though endogenous levels of soluble carbohydrate were already high in the nutrient-deficient plants. In complete nutrition, growth was stimulated, protein content particularly of the roots was increased and there was a preferential increase in activity of sucrose synthase in roots. At low P, enzyme activities in roots were increased, including sucrose synthase, and protein content increased, particularly in the roots, but there was no increase in growth. In N-deficient plants, exogenous sucrose led to decreased protein, Rubisco and chlorophyll content in shoots, in contrast to the other conditions, and a higher protein content and a general increase of catabolic enzyme activities and growth in the roots.  相似文献
