The effect of various chemicals on induction of dehiscence of nonsterile Coelomomyces psorophorae sporangia from Aedes taeniorhynchus larvae was examined. Tests were made with various salts, reducing agents, chelating agents, buffers, alcohols, carbohydrates, fatty acids and derivatives, amino acids and derivatives, peptides, amines, purines and pyrimidines, antibiotics, and plant hormones. The most active compound was Tris [tris(hydroxymethyl)aminomethane]. Tris was most effective at pH 8–9, and a concentration of 1–20 mM. Structure-activity studies indicated that active compounds had a basic requirement for NH2 and either COOH or CH2OH attached to the alpha carbon, but only certain amino acids and amines were highly active. Preparations virtually free of host debris were not responsive to Tris, but addition of bacteria-free homogenates of A. taeniorhynchus larvae restored responsiveness. Dehiscence of C. psorophorae sporangia from Psorophora howardii was also enhanced by exposure to Tris. 相似文献

The narrow-specialized pathogen of blood sucking mosquitoes Coelomomyces psorophorae Couch, which has not been previously found in the territory of the Ukraine, was detected on Aedes cantans and Al. communis larvae in water bodies located in the forest belt of Pushcha-Voditsa (Kiev Polessie, Ukrainian SSR). The germination of sporangia inCoelomomyces psorophorae vartasmaniensis (C. p. tas.) is uncoordinated and thus there is a mixture of developmental stages in any given population. Continuous urografin gradients separated out the critical stages of germinating sporangia giving four bands, each band representing a consecutive stage of germination. These stages were investigated for changes in the sporangial wall using Transmission Electron Microscopy (TEM). The sporangia have a typical two-layered wall, an electron dense outer layer which can be divided into three distinct sub-layers D1, D2, and D3 and an inner electron transparent secondary wall. Stage 3 sporangia have an intact D1 layer on their outer wall. In the subsequent stages (4 & 4b) there is a progressive breakdown of this layer. 相似文献

Effects of camptothecin, a naturally occurring alkaloid, on seed germination varied from promotive to inhibitory, depending on the species used. It markedly inhibited seedling root growth but its inhibition of hypocotyl growth varied among species. Camptothecin inhibited GA3-induced dark germination of lettuce (Lactuca sativa L.) seeds and hypocotyl elongation of seedlings. In contrast to ABA, the camptothecin inhibition of GA3-induced germination could not be overcome by cytokinin. When seeds were germinated at 29C with a 0.5 h light treatment, little or no germination occurred in the camptothecin treatment, but addition of cytokinin overcame this inhibition. 相似文献

The presence of synaptonemal complexes were checked in dividing chromosomes as evidence for meiotic division in germinating sporangia. Continuous urografin gradients were used to separate out the various phases of germinating sporangia, the nuclei were removed and embedded for ultrastructural studies. Meiotic inhibitors were applied to germinating sporangia to retard meiotic division to highlight the synaptonemal complexes. At an early phase of sporangial differentiation dividing nuclei developed with synaptonemal complexes. Meiotic inhibitors and stimulators may be used to control sporangial germination for an induction of a high meiospore count. This may be of crucial importance in the utilization of Coelomomyces spp. as a biological control agent of mosquito species. 相似文献

The electron microscope was used to examine changes which take place in wall, as well as in internal, structure during germination of mature resistant sporangia of Allomyces neo-moniliformis. When the resistant sporangia are first placed in water to initiate germination, nuclei, mitochondria, and endoplasmic reticulum are not evident, though after the sporangia have been in water for more than 30 min all of these structures become visible. At this time no cracks are evident in the resistant sporangial wall and the cell membrane appears highly convoluted. Within the next 30 min the outer wall splits and the inner wall expands considerably as the protoplast increases in volume. At the same time the cell membrane straightens out, apparently in response to the protoplasmic expansion. The “cementing substances” begin to dissolve about this time so that 1 1/2 hr after placement in water the outer wall is completely separated from the inner wall which now acts as the cell wall. Cleavage appears to be initiated by the invagination of the cell membrane and by the appearance of segments of endoplasmic reticulum with filled vesicles at one end. Between 2 1/2 and 3 hr after placement in water zoospores are released. 相似文献

实验采用 p H2 .0、2 .5、3.0、3.5、4 .0、5 .0模拟酸雨处理培养皿中的水稻 (Oryza sativa)、小麦 (Triticum aestivum)、油菜(Brassica chinensis var. oleifera) 3类抗性种子 ,每皿 5 0粒 ,置恒温培养箱内萌发 (2 5℃ ) ,每天更换 1次酸雨 ,处理与对照均 3次重复。定时测定酸雨胁迫强度、胁迫时间对种子发芽率、发芽势、发芽指数、活力指数、异状发芽率 ,吸水值、呼吸速率、贮藏物质运转效率、贮藏物质消耗率、根长抑制指数与芽长抑制指数的影响。结果表明 ,当酸雨胁迫强度 p H=2 .0~ 2 .5时 ,因胁迫强度过高 ,3类抗性种子皆不萌发 ;胁迫强度 p H≥ 3.0时 ,3类抗性种子 5项萌发指标的变幅是水稻 <小麦 <油菜 ;酸雨伤害阈值是水稻 (p H3.0~ 3.5 ) <小麦 (p H3.5~ 4 .0 ) <油菜 (p H4 .0~ 5 .0 ) ;当胁迫强度 (p H)≥ 2 .5时 ,3类抗性种子 6项生理指标的变幅是水稻 <小麦 <油菜 ,其生理反应阈值 (p H≥ 2 .0 ) <萌发反应阈值 (p H≥ 3.0 ) ;3类抗性种子贮藏物质消耗率、贮藏物质运转率 ,根长抑制指数及芽长抑制指数对酸雨胁迫时间的响应 (组间达到差异显著水平 )时间是 :水稻 (7d、7d,3d、3d)≥小麦 (6 d、6 d,3d、3d)≥油菜 (3d、4 d,3d、3d) ;3类抗性种子对酸雨胁迫强度与胁迫时间的耐受性差异 ( 相似文献

Gibberellic acid (GA3 ) and β-indolylacetic acid (IAA), two of the well known growth hormones, induce four fold the activity of ornithine decarboxylase (ODC) during the germination of barley seeds ( Hordeum vulgare L. var. Beca). The optimal concentration for induction of ODC was 10–5 M for GA3 and 10–3 M for IAA. When 10–3 M of a polyamine, putrescine or spermidine, is added to the growth medium, ODC activity is significantly inhibited. This inhibition is due to the induction of a protein inhibitor of ODC (antizyme), whose apparent molecular weight is 16 000 ± 2 000 daltons. Addition of GA3 to cultures which have been grown for 50 or 98 h in the presence of polyamines, abolishes the observed inhibition of ODC activity, while in the reverse experiment, addition of polyamines at 50 or 98 h does not affect the ODC activity induced by GA3 . Cadaverine, a physiological plant diamine, enhances ODC activity; whereas 1,8-diaminooctane (the alkyl analogue of spermidine) does not have any effect. 相似文献

对TDZ和2,4-D等激素在花生成熟胚外植体分化中的影响进行了研究.结果表明,花生成熟胚3~5 d龄实生苗的幼叶和胚轴在低浓度TDZ的诱导下,可分化产生高频不定芽和少量体细胞胚,转到无激素MS培养基或MS BA 0.5 mg/L NAA 0.4 mg/L的培养基后形成丛生苗.丛生苗分离后转入含1/2 MS(大量元素) IBA 0.4 mg/L的培养基中诱导生根,可形成完整的再生植株.幼叶分化率高于胚轴,但胚轴分化成苗速度快.无菌水浸泡16~24 h的胚轴在5~ 30 mg/L 2,4-D的诱导下,分化产生低频不定芽;而胚叶则产生高频体细胞胚,但畸形较严重. 相似文献

3,4-二羟基苯乙酮(DHAP)是天山云杉(Picea schrenkiana ssp. tianschanica)叶和凋落物中存在的主要自毒物质, 是导致天山云杉林天然更新障碍的原因之一。为了解释自毒物质发生作用的生理机制, 该文设计多个浓度梯度的DHAP溶液处理天山云杉种子, 以发芽率和发芽势为种子萌发参数, 运用反相超高效液相色谱(UPLC)分析技术, 检测了种子萌发过程中内源植物激素玉米素(ZT)、赤霉素(GA3)、吲哚乙酸(IAA)和脱落酸(ABA)含量水平的变化。研究结果表明: DHAP处理对天山云杉种子萌发影响具有浓度效应, 表现为5.0 mmol·L-1 > 0.1 mmol·L-1 > 1.0 mmol·L-1 >对照, 即5.0 mmol·L-1 DHAP处理组对种子萌发的抑制作用最强、0.1 mmol·L-1 DHAP处理组次之、1.0 mmol·L-1 DHAP处理组最弱; DHAP处理组的种子内源ZT、GA3浓度水平降低, ABA含量升高, GA3浓度峰值出现时间延迟, IAA浓度在高浓度(5.0 mmol·L-1 DHAP)处理组短时间内(3-6天)过量积累, 1.0和5.0 mmol·L-1 DHAP处理组的种子内源ABA浓度峰值出现时间延迟; DHAP处理种子萌发1-6天时, ZT/(GA3+IAA)比值降低, IAA/ZT、ABA/ZT比值增大; ABA/(ZT + GA3 + IAA)比值在0.1 mmol·L-1 DHAP处理组增大, 在5.0 mmol·L-1 DHAP处理组降低。DHAP处理引发种子内源激素含量水平及激素含量间比值变化, 可能是抑制、延迟天山云杉种子萌发的主要原因。 相似文献

Resistant starch (RS) has been suggested to prolong satiety in adult pigs. The present study investigated RS-induced changes in behaviour, satiety-related hormones and metabolites in catheterized growing pigs to explore possible underlying mechanisms for RS-induced satiety. In a cross-over design with two 14-day periods, 10 pigs (initial BW: 58 kg) were assigned to two treatments comprising diets containing either 35% pregelatinized starch (PS) or 34% retrograded starch (RS). Diets were isoenergetic on gross energy. Pigs were fed at 2.8× maintenance. Postprandial plasma response of satiety-related hormones and metabolites was measured at the end of each period using frequent blood sampling. Faecal and urinary energy losses were measured at the end of each period. Behaviour was scored 24 h from video recordings using scan sampling. Energy digestibility and metabolizability were ~6% lower in RS compared with PS diet (P<0.001), and metabolizable energy (ME) intake was ~3% lower in RS-fed than in PS-fed pigs (P<0.001). RS-fed pigs showed less feeder-directed (P=0.001) and drinking (P=0.10) behaviours than PS-fed pigs throughout the day. Postprandial peripheral short-chain fatty acid (SCFA) levels were higher in RS-fed than in PS-fed pigs (P<0.001). Postprandial glucose and insulin responses were lower in RS-fed than in PS-fed pigs (P<0.001). Triglyceride levels were higher in RS-fed than in PS-fed pigs (P<0.01), and non-esterified fatty acid levels did not differ between diets (P=0.90). Glucagon-like peptide-1 (GLP-1) levels were lower in RS-fed than in PS-fed pigs (P<0.001), and peptide tyrosine tyrosine (PYY) levels did not differ between diets (P=0.90). Blood serotonin levels were lower (P<0.001), whereas monoamine oxidase activity (P<0.05) and tryptophan (P<0.01) levels were higher in RS-fed than in PS-fed pigs. Despite a lower ME intake, RS seemed to prolong satiety, based on behavioural observations. Possible underlying mechanisms for RS-induced satiety include increased 24 h plasma SCFA levels, and decreased postprandial glucose and insulin responses. GLP-1 and PYY seemed not to play a role in RS-induced satiety. Low blood serotonin levels in RS-fed pigs suggested a difference in intestinal serotonin release between treatments. Increased postprandial plasma triglyceride levels corresponded with increased SCFA levels, but it is unclear whether triglycerides may have signalled satiety in RS-fed pigs. 相似文献

在加温、补光条件下,研究赤霉素(GA3)、萘乙酸(NAA)和吲哚乙酸(IAA)处理对锦绣杜鹃花蕾发育、开花期及开花质量等的影响。结果表明:锦绣杜鹃花蕾经0~3000mg.L-1GA3涂抹后,其催花效果随处理浓度增大呈先升后降趋势,并于2000mg.L-1浓度下达到最佳,表现在花蕾发育快、开花期提前及开花质量高等;而0~3000mg.L-1NAA和IAA对锦绣杜鹃催花效果均不明显。锦绣杜鹃花蕾大小(包括长度和宽度)与开花期提前时间和花径之间均呈极显著正相关,与花蕾期则呈极显著负相关;花蕾期与开花期提前时间之间呈极显著负相关(P<0.01)。 相似文献

比较了半干旱草原科尔沁沙地65种植物新采种子和埋藏1年种子的萌发特性.结果表明:埋藏1年后,有38种植物的种子仍具有活力.新采种子萌发率>80%的植物种子埋藏后萌发率下降;而萌发率<80%的植物种子埋藏后萌发率均有较大幅度的提高.埋藏对种子萌发特性的影响因植物生态类群(杂草、草原植物、草甸植物、沙生植物)而异.种子埋藏1年后,多年生植物萌发率没有明显提高,而1年生植物萌发率显著提高(P<0.05).对于新采种子和埋藏1年种子萌发率间相差10%以下的物种,可根据萌发格局将植物种分为3类:埋藏促进型(如差巴嘎蒿)、埋藏抑制型(如大籽蒿)和埋藏不变型(如鹤虱). 相似文献
