Abstract Five different bacterial communities were enriched in substrate-amended slurries of sediment from the Tay Estuary, Scotland. During incubation of the slurries, concentrations of volatile fatty acids, sulphate, sulphide and methane were monitored to clearly define the activity of the stimulated populations. An aerobic population, a ‘microaerophilic’ population and three anaerobic populations (fermentative heterotrophs, sulphate-reducing bacteria and methanogens plus acetogens) were established to reflect community growth and metabolism both in surface oxic and deeper anoxic layers. Similar numbers of cells involved in division were observed in all five slurries, demonstrating the potential for bacterial production. Thymidine incorporation rates in glucose-stimulated slurries under both aerobic and fully anaerobic conditions were similar, confirming the ability of fermentative anaerobic heterotrophs to incorporate [ methyl -3H]thymidine into DNA during growth. Although anaerobic communities of sulphate-reducing, acetogenic plus methanogenic bacteria were stimulated and actively growing, they did not incorporate [ methyl -3H]thymidine into DNA. Since the thymidine technique does not measure the growth of these important groups, calculated productivity values based upon thymidine incorporation within anoxic sediment systems will be substantially underestimated, even if growth substrates are not limiting.  相似文献

The purpose of this study was to determine the depth distribution of bacterial biomass and production in a stratified lake and to test techniques to measure bacterial production in anaerobic waters. Bacterial abundance and incorporation of both [3H]thymidine and [3H]leucine into protein were highest in the metalimnion, at the depth at which oxygen first became unmeasurable. In contrast, [3H]thymidine incorporation into DNA was highest in the epilimnion. The ratios of incorporation into DNA/protein averaged 2.2, 0.49, and 0.95 for the epilimnion, metalimnion, and hypolimnion, respectively. Low incorporation into DNA was not due to artifacts associated with the DNA isolation procedure. Recovery of added [3H]DNA was about 90% in waters in which the portion of [3H]thymidine incorporation into DNA was about 40%. At least some obligate anaerobic bacteria were capable of assimilating thymidine since aeration of anaerobic hypolimnion waters substantially inhibited thymidine incorporation. The depth profile of bacterial production estimated from total thymidine and leucine incorporation and the frequency of dividing cells were all similar, with maximal rates in the metalimnion. However, estimates of bacterial production based on frequency of dividing cells and leucine incorporation were usually significantly higher than estimates based on thymidine incorporation (using conversion factors from the literature), especially in anaerobic hypolimnion waters. These data indicate that the thymidine approach must be examined carefully if it is to be applied to aquatic systems with low oxygen concentrations. Our results also indicate that the interface between the aerobic epilimnion and anaerobic hypolimnion is the site of intense bacterial mineralization and biomass production which deserves further study.  相似文献

Conceptual models and some empirical studies in plant systems show that species interactions can shift from competitive under low stress and high productivity conditions to positive under high stress and low productivity conditions. In this study, we explore the relative strength and direction of marsh plant interactions at early life stages across a stress and productivity gradient in South Slough, Coos Bay, Oregon, USA. Germination and survival of five plant species (Atriplex patula L., Distichlis spicata (L.), Plantago maritima (Lam.) A. Gray, Sarcocornia pacifica (Standl.) A. J. Scott, Triglochin maritimum L.) were examined in the presence and absence of neighboring vegetation at three intertidal levels in each of three marsh sites along an estuarine gradient. We found that many of the interactions measured across these gradients were negative or neutral. The direction of the interactions depended on life stage, with neighboring plants having mostly negative and neutral effects on seed germination, and mostly neutral and rarely positive effects on seedling survival especially at lower intertidal locations. The exception was in the high intertidal at all sites, where competition was common. We found that the intensity of the interactions varied depending on marsh site, intertidal elevation, and plant species identity and that life stage and regional climate may be responsible for the general lack of positive interactions. We suggest that measuring species interactions across complex gradients of physical stress at different life stages can help refine our conceptual models and lead to better predictions of the factors controlling community structure.  相似文献

Microbial communities in water from Baltimore Harbor and from the mainstem of Chesapeake Bay were examined for sensitivity to mercuric chloride, monomethyl mercury, stannic chloride, and tributyltin chloride. Acute toxicity was determined by measuring the effects of [3H]thymidine incorporation, [14C]glutamate incorporation and respiration, and viability as compared with those of controls. Minimum inhibitory concentrations were low for all metals (monomethyl mercury, less than 0.05 microgram liter-1; mercuric chloride, less than 1 microgram liter-1; tributyltin chloride, less than 5 micrograms liter-1) except stannic chloride (5 mg liter-1). In some cases, mercuric chloride and monomethyl mercury were equally toxic at comparable concentrations. The Chesapeake Bay community appeared to be slightly more sensitive to metal stress than the Baltimore Harbor community, but this was not true for all treatments or assays. For culturable bacteria the opposite result was found. Thymidine incorporation and glutamate metabolism were much more sensitive indicators of metal toxicity than was viability. To our knowledge, this is the first use of the thymidine incorporation method for ecotoxicology studies. We found it the easiest and fastest of the three methods; it is at least equal in sensitivity to metabolic measurements, and it likely measures the effects on greater portion of the natural community.  相似文献

Effects of sediment properties on benthic primary production were investigated along transects in the intertidal regions of the Columbia River estuary. On the tidal flats, benthic diatoms were the most abundant plants, while microalgae and submergent vascular macrophytes were relatively rare. At five intensive study sites, mean rates of benthic primary production varied between 5 mg C m−2 h−1 at Clatsop Spit and 84 mg C m−2 h−1 at Youngs Bay. A regional analysis of data from these sites and from 31 less intensively studied survey sites indicated that Youngs Bay and Baker Bay were more productive than Grays Bay, Cathlamet Bay and the tidal flats in the upper estuary above these bays. This pattern corresponded to the stability and particle size distribution of the sediment in these areas. Benthic gross primary production for the total estuarine area of 410 km2 was estimated to be 4895 t carbon per year and represents a mean rate of 72 g C m−2 year−1. Of the total net primary production in the estuary, benthic plants accounted for about 7%, while phytoplankton and emergent vascular plants in the marshlands contributed approximately 56% and 37%, respectively. Data from this study also suggested that physical processes related to estuarine circulation can ultimately determine the productivity of benthic plant assemblages, and minimize the regulatory and limiting effects of temperature, nutrient supply and consumption by benthic animals.  相似文献

We studied the composition, density, size distribution and biomass of the food supply for waders in an estuarine area in the Bay of Cádiz (SW Iberian Peninsula), in winter (January-February) and in the pre-migratory period (late March). The estuarine area comprises an intertidal mudflat and an adjacent salina or salt-pan. On the intertidal mudflat, the biomass was 53 and 37 g  AFDW.m-2in winter and the pre-migratory period, respectively. The main food source on mudflat was the polychaete Nereis diversicolor (44–54 % of the total biomass). On the other hand, the biomass in the salina was comparatively very poor, ranging from 0.008 to 0.079 g  AFDW.m-2in winter and ranging from 0.011 to 0.09 g AFDW in late March. The main source of food in the salina was the crustacean Artemia. The total biomass on the mudflat during the pre-migratory period was 1.4 times lower than in February. This depletion could be caused by wader predation, mainly by Nereis diversicolor consumption. Although the potential food on the mudflats could allow high intertidal densities of waders, the availability of high tide foraging areas in the salina seems to contribute to the maintenance of these high intertidal densities.  相似文献

Spartina anglica is an exotic perennial grass that can rapidly colonise the intertidal zone of temperate estuaries and lagoons. Consequently, there is considerable concern about its impact on estuarine flora and fauna. This study provides the first investigation of ecological impacts by S. anglica in Australia. The objective was to investigate the impacts of S. anglica on benthic macroinvertebrate communities inhabiting mudflat and native saltmarsh habitats at Little Swanport estuary, Tasmania. The null hypothesis that species richness and species abundance of benthic macroinvertebrates in exotic S. anglica marsh does not differ from adjacent native saltmarsh and mudflat habitats was tested. Eighteen species and 3716 macroinvertebrates were collected from 60 intertidal core samples in three habitats. Species richness, total abundance of invertebrates, crustacean abundance and mollusc abundance of mudflat communities were significantly (P < 0.05) lower when compared to those inhabiting adjacent S. anglica marsh and native saltmarsh. However, species richness and total abundance of invertebrates of native saltmarsh and S. anglica marsh did not differ significantly. Ordination of macroinvertebrate data clearly separated mudflat sites from vegetated sites but showed remarkable similarity between exotic and native vegetated sites.  相似文献

Abstract Incorporation of [3H]methyl thymidine into bacterial DNA was measured using samples of bacterioplankton from Lake Constance and pure cultures of CO, H2 and CH4-oxidizing bacteria. Thymidine was incorporated by Pseudomonas carboxydovorans, Paracoccus denitrificans, Methylosinus trichosporium, Methylomonas agile , and by various chemolithotropic or methylotrophic isolates from Lake Constance. Thymidine incorporation by bacterial cultures was stimulated by increasing concentrations of CO or H2. Increased CH4 concentrations stimulated thymidine incorporation by Ms. trichosporium only if the cells had been starved. In stratified Lake Vechten, The Netherlands, protozoan grazing was estimated on the basis of uptake of fluorescently labeled bacteria and compared with bacterial production estimated on the basis of thymidine incorporation. By using a grazer-free mixed bacterial population from the lake in continuous culture, an empirical relationship between cell production and thymidine incorporation was established. Thymidine incorporation into total cold-trichloroacetic-acid-insoluble macromolecules yielded a relatively constant empirical conversion factor of ca. 1018 (range, 0.38 × 1018 to 1.42 × 1018) bacteria mol of thymidine−1 at specific growth rates (μ) ranging from 0.007 to 0.116 h−1. Although thymidine incorporation has been assumed to measure DNA synthesis thymidine incorporation appeared to underestimate the independently measured bacterial DNA synthesis by at least 1.5- to 13-fold, even if all incorporated label was assumed to be in DNA. However, incorporation into DNA was found to be insignificant as measured by conventional acid-base hydrolysis. Methodological problems of the thymidine technique are discussed. Like the cultures, Lake Vechten bacteria showed considerable thymidine incorporation into total macromolecules, but no significant incorporation into DNA was found by acid-base hydrolysis. This applied not only to the low-oxygen hypo- and metalimnion but also to the aerobic epilimnion. Thus, the established empirical conversion factor for thymidine incorporation into total macromolecules was used to estimate bacterial production. Maximum production rates (141 × 106 bacteria liter−1 h−1; μ, 0.012 h−1) were found in the metalimnion and were 1 order of magnitude higher than in the epi- and hypolimnion. In all three strata, the estimated bacterial production was roughly balanced by the estimated protozoan grazing. Heterotrophic nanoflagellates were the major consumers of the bacterial production and showed maximum numbers (up to 40 × 106 heterotrophic nanoflagellates liter−1) in the microaerobic metalimnion.  相似文献

The production and turnover of dissolved DNA in subtropical estuarine and oligotrophic oceanic environments were investigated. Actively growing heterotrophic bacterioplankton (i.e., those capable of [3H]thymidine incorporation) were found to produce dissolved DNA, presumably through the processes of death and lysis, grazing by bacteriovores, and excretion. Production of dissolved DNA as determined by [3H]thymidine incorporation was less than or equal to 4% of the ambient dissolved DNA concentration per day. In turnover studies, the addition of [3H]DNA (Escherichia coli chromosomal) to seawater resulted in rapid hydrolysis and uptake or radioactivity by microbial populations. DNA was hydrolyzed by both cell-associated and extracellular nucleases, in both estuarine and offshore environments. Kinetic analysis performed for a eutrophic estuary indicated a turnover time for dissolved DNA as short as 6.5 h. Microautoradiographic studies of bacterial populations in Tampa Bay indicated that filamentous and attached bacteria took up most of the radioactivity from [3H]DNA. Dissolved DNA is therefore a dynamic component of the dissolved organic matter in the marine environment, and bacterioplankton play a key role in the cycling of this material.  相似文献

Bacterial production and amino acid metabolism in aquatic systems can be estimated by simultaneous incubation of water samples with both tritiated methyl-thymidine and 14C-labeled amino acids. This dual-label method not only saves time, labor, and materials, but also allows determination of these two parameters in the same microbial subcommunity. Both organic carbon incorporation and respiration can be estimated. The results obtained with the dual-label technique are not significantly different from single-radiolabel methods over a wide range of bacterial activity. The method is particularly suitable for large-scale field programs and has been used successfully with eutrophic estuarine samples as well as with oligotrophic oceanic water. In the mesohaline portion of Chesapeake Bay, thymidine incorporation ranged seasonally from 2 to 635 pmol liter−1 h−1 and amino acid turnover rates ranged from 0.01 to 28.4% h−1. Comparison of thymidine incorporation with amino acid turnover measurements made at a deep, midbay station in 1985 suggested a close coupling between bacterial production and amino acid metabolism during most of the year. However, production-specific amino acid turnover rates increased dramatically in deep bay waters during the spring phytoplankton bloom, indicating transient decoupling of bacterial production from metabolism. Ecological features such as this are readily detectable with the dual-label method.  相似文献

The incorporation of [H]thymidine by axenic cultures of anaerobic bacteria was investigated as a means to measure growth. The three fermentative strains and one of the methanogenic strains tested incorporated [H]thymidine, whereas the sulfate-reducing bacterium and two of the methanogenic bacteria were unable to incorporate [H]thymidine during growth. It is concluded that the [H]thymidine incorporation method underestimates bacterial growth in anaerobic environments.  相似文献

Abstract: The bacterial community response to pH was studied for 16 soils with pH(H2O) ranging between 4 and 8 by measuring thymidine incorporation into bacteria extracted from the soil into a solution using homogenization-centrifugation. The pH of the bacterial solution was altered to six different values with dilute sulfuric acid or different buffers before measuring incorporation. The resulting pH response curve for thymidine incorporation was used to compare bacterial communities from the different soils. There was a correlation between optimum pH for thymidine incorporation and the soil pH(H2O). Even bacterial communities from acid soils had optima corresponding to the soil pH, indicating that they were adapted to these conditions. Thymidine incorporation was also compared with leucine incorporation for some soils. The leucine to thymidine incorporation ratio was constant over the tested pH interval when incorporation values were adjusted for isotope dilution. A good correlation was found between the scores along the first component (explaining 80% of the variation) and soil pH ( r 2 = 0.85), if principal component analysis of the pH response curves for thymidine incorporation was used. The pH response curves differed most for the extreme pH values used, and a linear relationship was found between the logarithm of the ratio of thymidine incorporation at pH 4.3 to incorporation at pH 8.2 and the soil pH ( r 2 = 0.86). Thus, a simplified technique using only two pH values, when measuring the thymidine incorporation, could be used to compare the response to pH of bacterial communities.  相似文献

Heterotrophic marine bacterium isolates from Tampa Bay, Fla., Chesapeake Bay, Md., and a coral surface microlayer were examined for thymidine uptake (transport), thymidine incorporation, the presence of thymidine kinase genes, and thymidine kinase enzyme activity. Of the 41 isolates tested, 37 were capable of thymidine incorporation into DNA. The four organisms that could not incorporate thymidine also transported thymidine poorly and lacked thymidine kinase activity. Attempts to detect thymidine kinase genes in the marine isolates by molecular probing with gene probes made from Escherichia coli and herpes simplex virus thymidine kinase genes proved unsuccessful. To determine if the inability to incorporate thymidine was due to the lack of thymidine kinase, one organism, Vibrio sp. strain D19, was transformed with a plasmid (pGQ3) that contained an E. coli thymidine kinase gene. Although enzyme assays indicated high levels of thymidine kinase activity in transformants, these cells still failed to incorporate exogenous thymidine into DNA or to transport thymidine into the cells. These results indicate that the inability of certain marine bacteria to incorporate thymidine may not be solely due to the lack of thymidine kinase activity but may also be due to the absence of thymidine transport systems.  相似文献

Abstract Incorporation of [ methyl -3H]thymidine into bacterial DNA was determined for a range of axenic anaerobic bacterial cultures: fermentative heterotrophs, sulphate-reducing bacteria, purple sulphur bacteria, acetogens and methanogens. Anaerobically growing Bacillus sp. and the obligate aerobe Thiobacillus ferrooxidans were also investigated. Actively growing cultures of sulphate-reducing bacteria belonging to the genera Desulfovibrio, Desulfotomaculum, Desulfobacter, Desulfobotulus and Desulfobulbus , purple sulphur bacteria ( Chromatium vinosum OP2 and Thiocapsa roseopersicina OP1), methanogens ( Methanococcus GS16 and Methanosarcina barkeri ) and an acetogen ( Acetobacterium woodii ) did not incorporate [ methyl -3H]thymidine into DNA. The only obligate anaerobes in which thymidine incorporation into DNA could be unequivocally demonstrated were members of the genus Clostridium . Anaerobically growing Bacillus sp. also incorporated thymidine. These data demonstrate that pure culture representatives of major groups of anaerobic bacteria involved in the terminal oxidation of organic carbon and anoxygenic phototrophs within sediments are unable to incorporate [ methyl -3H]thymidine into DNA, although some obligate and facultative anaerobes can. Variability in thymidine incorporation amongst pure culture isolates indicates that unless existing techniques can be calibrated to take this into consideration then productivity estimates in both aerobic and anaerobic environments may be greatly underestimated using the [ methyl -3H]thymidine technique.  相似文献

The coupling of primary and bacterial production over depthwas examined in three lakes which differed greatly in verticalpatterns of primary productivity. We measured bacterial production,chlorophyll and light, and estimated primary production in PaulLake (Gogebic County, Michigan) and Crystal and Trout lakes(Vilas County, Wisconsin) during the summer stratification period(May–September 1991). Bacterial productivity was measuredusing the [3H]leucine incorporation method and primary productivityestimated from measured photosynthesis–irradiance relationships.Three distinct vertical patterns were observed. In Paul Lake,bacterial production was highest at the interface between theaerobic and anaerobic layers, well below the depth of maximumprimary production. In Crystal Lake, bacterial production wasuniform with depth, although primary productivity was highestin the hypolimnion. In the largest lake. Trout Lake, primaryand bacterial production tended to co-vary with maximum ratesof both processes occurring in the metalimnion. Overall, bacterialproductivity was poorly related to contemporaneous primary productionin the three lakes, suggesting that other factors, such as nutrientrecycling, phytoplankton loss rates and allocthonous loading,determine patterns in the depth distribution of bacterial productivity.  相似文献

We examined the simultaneous incorporation of [3H]thymidine and [14C]leucine to obtain two independent indices of bacterial production (DNA and protein syntheses) in a single incubation. Incorporation rates of leucine estimated by the dual-label method were generally higher than those obtained by the single-label method, but the differences were small (dual/single = 1.1 ± 0.2 [mean ± standard deviation]) and were probably due to the presence of labeled leucyl-tRNA in the cold trichloroacetic acid-insoluble fraction. There were no significant differences in thymidine incorporation between dual- and single-label incubations (dual/ single = 1.03 ± 0.13). Addition of the two substrates in relatively large amounts (25 nM) did not apparently increase bacterial activity during short incubations (<5 h). With the dual-label method we found that thymidine and leucine incorporation rates covaried over depth profiles of the Chesapeake Bay. Estimates of bacterial production based on thymidine and leucine differed by less than 25%. Although the need for appropriate conversion factors has not been eliminated, the dual-label approach can be used to examine the variation in bacterial production while ensuring that the observed variation in incorporation rates is due to real changes in bacterial production rather than changes in conversion factors or introduction of other artifacts.  相似文献
