Summary Changes in the expression pattern and intracellular localization of Forssman glycolipid (FA) and GM1 ganglioside (GM1) in fetal mouse gonads were examined during germ cell differentiation by immunofluorescence microscopy and immunoelectron microscopy.In male germ cells from the 12th to 14th day p.c., anti-FA binding was localized in granular structures aggregated on one side of the cytoplasm and/or in the plasma membrane. On day 16 p.c., some germ cells still showed patch-like positive reactions in the plasma membrane, but by day 18 p.c., positive reactions for FA had completely disappeared. The female germ cells showed granular bindings of anti-FA scattered throughout their cytoplasm during the 13th to 16th day p.c., although the positive reactions in female germ cells on day 12 p.c. tended to be found in one side of cytoplasm and/or plasma membrane similar to those in male germ cells from 12th to 14th day p.c. On day 18 p.c., positive reactions remained in the plasma membrane of some germ cells, but these positive reactions disappeared before birth. Immunoelectron microscopic observation showed that the sites of anti-FA bindings were equivalent to the small dense bodies (SDB) and the Golgi lamellae both in male and female germ cells. On the other hand, GM1 was not detected in male germ cells at any time during fetal testicular development, whereas an anti-GM1 reaction was detected in the plasma membrane of female germ cells from the 16th to 18th day p.c. (oocytes in the first meiotic prophase).Therefore, these results indicate that kinetic translocation of FA between the plasma membrane and the SDB and Golgi lamellae takes place during germ cell differentiation in fetal gonads. Moreover, the ganglioside composition containing GM1 seems to change in association with the first meiotic prophase.  相似文献

Proteins homologous to the Xenopus oocyte mRNA binding proteins mRNP3+4 and designated p48/52 have been identified in male mouse germ cells (1993: Dev Biol 158:90–100). Western and Northwestern blots of extracts from testes and isolated germ cells indicate that p48/52 are present during meiosis but reach their highest levels postmeiotically at a time when many mRNAs are stored. Here we analyze the cellular and subcellular distribution of p48/52 in rat and mouse testes by LM and EM immunocytochemistry using an anti-mRNP3+4 antibody. Immunolabeling was found to be predominantly cytoplasmic and specific to germ cells at certain periods during their development. p48/52 were first detected in early pachytene spermatocytes at stage V of the seminiferous cycle and progressively increased during the remainder of meiotic prophase to a post-meiotic peak in steps 1–8 round spermatids; thereafter, labeling gradually declined as elongated spermatids underwent nuclear condensation and elongation. A proportionally higher concentration of cytoplasmic immunolabeling was found within the lacunae of the anastomotic granulofilamentous network of the chromatoid body. The pattern of synthesis of these mRNA binding proteins together with their association with the chromatoid body suggests a role as germ cell-specific mRNA stabilizing and/or storage proteins. © 1996 Wiley-Liss, Inc.  相似文献

Meiosis and the meiotic spindle pole body cycle were studied electron microscopically in basidia of the heterobasidiomycetePachnocybe ferruginea. Spindle pole body splitting in prometaphase I and II, and intermeiotic and postmeiotic duplication were investigated in particular detail. During prophase, the spindle pole body consists of two three-layered discs connected by a middle piece. At late prophase I and again in prometaphase II, the discs contact the nuclear envelope. Then, the nuclear membrane at the contact area is separated from the non-contacted part of the nuclear envelope and finally disappears. Each disc nests into the nuclear opening of the otherwise intact nuclear envelope. The disc remains in the gap and generates a half spindle. At late metaphase I, a co-disc develops eccentrically within the parent disc. The co-disc detaches from the parent disc during interphase I and becomes one of the metaphase II spindle pole bodies. Co-discs are absent during the second division. A cap of endoplasmic reticulum encloses each disc during prophase I through anaphase I. In the second meiotic division, the caps covering the spindle pole bodies of one nucleus of the pair, are developed from the neighbouring nucleus. Spindle pole bodies ofP. ferruginea are similar to those of the rusts, and especially to those ofEocronartium muscicola andHelicobasidium mompa. The germ-line cells of many animals possess a characteristic cytoplasmic structure termed nuage or germinal granules. In mice, nuage that is prominent in postnatal male germ cells is also called intermitochondrial cement or chromatoid bodies. TDRD1/MTR-1, which contains Tudor domain repeats, is a specific component of the mouse nuage, analogously to Drosophila Tudor, a constituent of polar granules/nuage in oocytes and embryos. We show that TDRD6 and TDRD7/TRAP, which also contain multiple Tudor domains, specifically localize to nuage and form a ribonucleoprotein complex together with TDRD1/MTR-1. The characteristic co-localization of TDRD1, 6 and 7 was disrupted in a mutant of mouse vasa homologue/DEAD box polypeptide 4 (Mvh/Ddx4), which encodes another evolutionarily conserved component of nuage. In vivo over-expression experiments of the TDRD proteins and truncated forms during male germ cell differentiation showed that a single Tudor domain is a structural unit that localizes or accumulates to nuage, but the expression of the truncated, putative dominant negative forms is detrimental to meiotic spermatocytes. These results indicate that the Tudor-related proteins, which contain multiple repeats of the Tudor domain, constitute an evolutionarily conserved class of nuage components in the germ-line, and their localization or accumulation to nuage is likely conferred by a Tudor domain structure and downstream of Mvh, while the characteristic repeated architecture of the domain is functionally essential for the differentiation of germ cells.  相似文献

In vivo casein phosphorylation was analysed in Xenopus full-grown oocytes arrested in the prophase of the meiotic cell division. The phosphorylation was inhibited by the protein kinase inhibitor (PKI) and also by heparin (3 g/ml; final concentration). casein phosphorylation was increased by spermine (2 mM). Therefore, protein kinase A and casein kinase II are both activein vivo in full-grown oocytes and may be involved in the prophase arrest of meiotic cell division.  相似文献

Microsporocytes sometimes undergo an achiasmatic meiosis when placed into culture early in the season at a time after premeiotic S but prior to leptonema. Trillium meiocytes were examined by light and electron microscopy to analyze the frequency of cells in various stages of meiotic prophase and the occurrence of the synaptonemal complex at different times of culture. On the basis of the results, a hypothesis is proposed that suggests there is a tripartite sensitive period that occurs between S phase and leptonema. Where the cells are in this sensitive period at the time of transplantation into culture determines whether the cells do not enter meiotic prophase, enter but produce achiasmatic division figures, or enter and develop normally.This work was supported in part by grants from the National Science Foundation (GB 5173X and GB 6476) and the National Institute of Health (GM 16882)  相似文献

Summary Cytoplasmic structures ultrastructurally similar to the nuage are present in the cytoplasm of all spermatogenic cells in adult rats. The nuage is a discrete organelle which should not be confused with the chromatoid body. In step 7–8 spermatids transient contact is established between the nuage and the chromatoid body. This indicates a very specific recognition of the nuage by the chromatoid body. It is suggested that the nuage and the chromatoid body are separate cell organelles the functions of which are somehow related to each other.  相似文献

In male hybrids of the cross Chironomus thummi thummi (stock Hl) x Ch. th. piger (stock E) , but not the reciprocal cross, rudimentary testes develop at a growth temperature of 21 ° C. Within the dysgenic testes of these hybrids a number of abnormalities are observed which are restricted to the germ line. Approximately 60% of the hybrid males show allocyclic chromosome behaviour in spermatogonia and spermatocyte I nuclei. Within these nuclei two groups of four chromosomes are formed which differ from one another in their state of condensation. Each chromosome group consists of three long and one short chromosome. In cases where allocycly is very pronounced, the chromosomes of both groups disintegrate into numerous unequally sized fragments at meiotic prometaphase I, and gametes are not produced. In individuals, in which the allocycly is less pronounced or absent the nuclear divisions appear to be normal but chromosome and chromatid aberrations are frequent, and the number of viable sperm is reduced. In these males, the chiasma frequency is also decreased more than 12-fold in comparison with the reciprocal, unaffected piger x thummi hybrids.  相似文献

Fetal mouse testes and ovaries with their urogenital connections were cultured singly or in pairs on Nuclepore filters. When a testis in which the sex was not yet morphologically detectable was cultured together with older ovaries containing germ cells which were progressing through the meiotic prophase, the male germ cells were triggered to enter meiosis. When older fetal testes in which the testicular cords have developed were cultured together with ovaries of the same age with germ cells in meiosis, the oocytes were prevented from reaching diplotene stage. It was concluded that the fetal male and female gonads secrete diffusable substances which influence germ cell differentiation. The male gonad secretes a "meiosis-preventing substance" (MPS) which can arrest the female germ cells within the meiotic prophase. The female gonad secretes a "meiosis-inducing substance" (MIS) which can trigger the nondifferentiated male germ cells to enter meiosis.  相似文献

During the meiotic prophase the nuclei of the spermatocytes and oocytes of Panorpa communis contain a paracrystalloid body (PB). This is built up of about 100 filaments of 80 Å thickness all running parallel to one another. The filaments are connected by a network of many thin fibers. At pachytene and diplotene the PB is connected with the nucleolus. In spermatocytes at late diplotene the PB is enveloped by RNP-material, which is produced by certain chromosomes at the diffuse diplotene stage. Thereafter the PB is gradually removed. During this process the RNP-material coils up to form three or four spherical RNP-bodies. It is suggested that the PB has a function in assembling and connecting the RNP-material to form long ribbons which build up the RNP-bodies.  相似文献

Summary Apical application of silver nitrate (AgNO3; 50 and 100 g per plant) and silver thiosulphate anionic complex (Ag(S2O3) 2 3– ; STS; 25, 50 and 100 g per plant) to female plants of Cannabis sativa induced the formation of reduced male, intersexual and fully altered male flowers on the newly formed primary lateral branches (PLBs); 10 g per plant of AgNO3 was ineffective and 150 g treatment proved inhibitory. A maximum number of fully altered male flowers were formed in response to 100 g STS. The induced male flowers produced pollen grains that germinated on stigmas and effected seed set. Silver ion applied as STS was more effective than AgNO3 in inducing flowers of altered sex. The induction of male flowers on female plants demonstrated in this work is useful for producing seeds that give rise to only female plants. This technique is also useful for maintaining gynoecious lines.  相似文献

Prophase chromosomes of growing oocytes from thelytokous, viviparous females of Amphorophora tuberculata Brown and Blackman (n=2) were studied using a modified propionic acid squash technique with Feulgen staining. In early prophase, prior to the growth phase of the oocyte, the X chromosomes are partially condensed and looped together so that all four ends appear to be associated. Later in prophase the X chromosomes separate in oocytes destined to be female, but remain associated in presumptive male oocytes. The autosomes condense gradually throughout prophase. The nucleus of the presumptive male oocyte is further characterised by the formation of a spherical Feulgen-positive body, which attains a large size (7 m diameter) in late prophase. At this stage, the X chromosomes are no longer visible as separate entities, and are apparently included in the spherical body. At metaphase this disappears, leaving the X chromosomes still united as a condensed bivalent. The spherical body seems to have nucleolar as well as chromatin constituents; nucleolar organisers are present at the ends of the X chromosomes where it first arises. It may function in maintaining the cohesion between the X chromosomes through prophase, and could also facilitate correct orientation of the X bivalent on the spindle of the maturation division. As sex determination in aphids is controlled by juvenile hormone concentration, it appears that the hormone may interact with the X chromosomes during prophase, bringing about their separation in female oocytes, perhaps by inhibiting the formation of the spherical body.  相似文献

Summary Plants of the w4-mutable line of soybean [Glycine max (L.) Merr.] are chimeral for anthocyanin pigmentation. Mutable plants produce both near-white and purple flowers, as well as flowers of mutable phenotype with purple sectors on near-white petals. It is established here that the mutable trait is conditioned by an unstable recessive allele of the w4 locus that conditions anthocyanin biosynthesis. The gene symbol w4-m is assigned to the mutable allele. Allele w4-m was derived from a stable, wild-type W4 progenitor allele and reverts at high frequency to a stable, wild-type W4 allele. Reversion occurs both early and late during the development of the germ line. Several experiments give estimates of germinal reversion frequency, indicating that approximately 6% of mutable alleles revert to wild-type from one generation to the next. Allele w4-m exhibits many features typical of an allele controlled by a transposable element.  相似文献

A period of chromosome contraction between premeiotic interphase and leptotene was regularly observed in three samples of Lilium longiflorum Croft. Extensive preleptotene chromosome contraction was also observed in L. longiflorum Ace and in the Lilium hybrid Enchantment. Although the stage resembles late mitotic prophase, microsporocytes never develop to metaphase, but despiralize to leptotene, and regular alignment, pairing and chiasma formation follow. As preleptotene chromosome contraction is discovered in an increasing number of organisms it becomes less likely that it represents a true reversion to mitosis. However its absence in many organisms and its extreme variability in others do not support the concept of preleptotene chromosome contraction as a regular meiotic stage. It is suggested that the line of demarcation between mitosis and meiosis is often imprecise, and meiocytes may fluctuate to some extent between these states before a final transition to meiosis is made. The occurrence and extent of this fluctuation may possibly be related to some externally produced substances required for the orderly development of meiotic prophase.  相似文献

Several experiments had indicated that in planta transformation of Arabidopsis thaliana by Agrobacterium involves the female germ line. In order to identify the precise stage at which transformation occurs we have monitored expression of a gusA reporter gene in the two products of the double fertilization of infiltrated plants. The plantlets and the remaining endosperm of seeds were separately tested after germination. It appeared that in the majority of cases only the plantlet or the endosperm were transformed. Based on transformation with two vectors borne by two different Agrobacterium strains, the minority of co-transformed plantlets and endosperm can be explained by simultaneous but independent transformation events. These results indicate that mature female gametes could be the targets of T-DNA.  相似文献

A systematic search for chromosome pairing defects in foetal mouse oocytes has been carried out in two different strains (Swiss and CBA/Ca) over days 15–19 of gestation and on day 1 post-partum. The aim was to seek direct cytological evidence for a production line of oocyte development, or the occurrence of pairing anomalies at meiotic prophase that might lead, in the adult female, to nondisjunction at anaphase I. No evidence for either was found. The data argue against the production line hypothesis as the basis for maternal age-related increases in aneuploidy in the mouse. Attempts to analyse chiasmata in oocytes at diplotene were unsuccessful.  相似文献

The multiple sex chromosome system, X1X2Y /X1X1X2X2, in the small Indian mongoose, Herpestes auropunctatus, results from a translocation of a part of Y chromosome to an autosome. It is not possible to distinguish the autosome which harbours the Y chromosome element in the somatic complement. By employing the surface-spreading technique to prophase I meiocytes we have identified the region to which the Y chromosome has been translocated as the short arm of chromosome 9 which is a subtelocentric chromosome. This Y chromosome component lacks heterochromatin and no sex vesicle is organised during meiotic prophase. This suggests to us that Y heterochromatin in mammals may be required for the production of a sex vesicle.We take great pleasure in dedicating this paper to our revered teacher Prof. S.P.
