The antibacterial activity of photocatalytic titanium dioxide (TiO2) substrates is induced primarily by UV light irradiation. Recently, nitrogen- and carbon-doped TiO2 substrates were shown to exhibit photocatalytic activities under visible-light illumination. Their antibacterial activity, however, remains to be quantified. In this study, we demonstrated that nitrogen-doped TiO2 substrates have superior visible-light-induced bactericidal activity against Escherichia coli compared to pure TiO2 and carbon-doped TiO2 substrates. We also found that protein- and light-absorbing contaminants partially reduce the bactericidal activity of nitrogen-doped TiO2 substrates due to their light-shielding effects. In the pathogen-killing experiment, a significantly higher proportion of all tested pathogens, including Shigella flexneri, Listeria monocytogenes, Vibrio parahaemolyticus, Staphylococcus aureus, Streptococcus pyogenes, and Acinetobacter baumannii, were killed by visible-light-illuminated nitrogen-doped TiO2 substrates than by pure TiO2 substrates. These findings suggest that nitrogen-doped TiO2 has potential application in the development of alternative disinfectants for environmental and medical usages.  相似文献

There is an increasing interest in the application of photocatalytic properties for disinfection of surfaces, air, and water. Titanium dioxide is widely used as a photocatalyst, and the addition of silver reportedly enhances its bactericidal action. However, the synergy of silver nanoparticles and TiO(2) is not well understood. The photocatalytic elimination of Bacillus atrophaeus was examined under different calcination temperatures, dip-coating speeds, and ratios of TiO(2), SiO(2), and Ag to identify optimal production conditions for the production of TiO(2)- and/or TiO(2)/Ag-coated glass for surface disinfection. Photocatalytic disinfection of pure TiO(2) or TiO(2) plus Ag nanoparticles was dependent primarily on the calcination temperature. The antibacterial activity of TiO(2) films was optimal with a high dip-coating speed and high calcination temperature (600°C). Maximal bacterial inactivation using TiO(2)/Ag-coated glass was also observed following high-speed dip coating but with a low calcination temperature (250°C). Scanning electron microscopy (SEM) showed that the Ag nanoparticles combined together at a high calcination temperature, leading to decreased antibacterial activity of TiO(2)/Ag films due to a smaller surface area of Ag nanoparticles. The presence of Ag enhanced the photocatalytic inactivation rate of TiO(2), producing a more pronounced effect with increasing levels of catalyst loading.  相似文献

Exposure to the soil-borne pathogens Burkholderia pseudomallei and Burkholderia cenocepacia can lead to severe infections and even mortality. These pathogens exhibit a high resistance to antibiotic treatments. In addition, no licensed vaccine is currently available. A nanoscale platinum-containing titania photocatalyst (TiO(2)-Pt) has been shown to have a superior visible light-responsive photocatalytic ability to degrade chemical contaminants like nitrogen oxides. The antibacterial activity of the catalyst and its potential use in soil pathogen control were evaluated. Using the plating method, we found that TiO(2)-Pt exerts superior antibacterial performance against Escherichia coli compared to other commercially available and laboratory prepared ultraviolet/visible light-responsive titania photocatalysts. TiO(2)-Pt-mediated photocatalysis also affectively eliminates the soil-borne bacteria B. pseudomallei and B. cenocepacia. An air pouch infection mouse model further revealed that TiO(2)-Pt-mediated photocatalysis could reduce the pathogenicity of both strains of bacteria. Unexpectedly, water containing up to 10% w/v dissolved soil particles did not reduce the antibacterial potency of TiO(2)-Pt, suggesting that the TiO(2)-Pt photocatalyst is suitable for use in soil-contaminated environments. The TiO(2)-Pt photocatalyst exerted superior antibacterial activity against a broad spectrum of human pathogens, including B. pseudomallei and B. cenocepacia. Soil particles (<10% w/v) did not significantly reduce the antibacterial activity of TiO(2)-Pt in water. These findings suggest that the TiO(2)-Pt photocatalyst may have potential applications in the development of bactericides for soil-borne pathogens.  相似文献

Titanium dioxide (TiO(2)) under ultraviolet (UV) light produces a strong oxidative effect and may therefore be used as a photocatalytic disinfectant. Although many studies on the photocatalytic inactivation of bacteria have been reported, few studies have addressed virus inactivation. In the present study, we demonstrated the inactivation of influenza virus through TiO(2) photocatalysis using TiO(2) nanoparticles immobilized on a glass plate. The influences of the UV intensity, UV irradiation time and bovine serum albumin (BSA) concentration in the viral suspensions on the inactivation kinetics were investigated. Additionally, we also determined whether the International Organization for Standardization (ISO) methodology for the evaluation of antibacterial activity of TiO(2) photocatalysis could be applied to the evaluation of antiviral activity. The viral titers were dramatically reduced by the photocatalytic reaction. Even with a low intensity of UV-A (0.01 mW cm(-2)), a viral reduction of approximately 4-log(10) was observed within a short irradiation time. The viral inactivation kinetics were associated with the exposure time, the UV intensity and the BSA concentration in virus suspensions. These results show that TiO(2) photocatalysis could be used to inactivate the influenza virus. Furthermore, a minor modification of the ISO test method for anti-bacterial effects of TiO(2) photocatalysis could be useful for the evaluation of antiviral activity.  相似文献

The International Organization for Standardization (ISO) was used to evaluate antibacterial activity by titanium dioxide (TiO(2)) photocatalysis since 2006. We evaluated photocatalytic inactivation of Qβ and T4 bacteriophages induced by low-intensity, long-wavelength ultraviolet A (UVA; 0.1 mW cm(-2) and 0.001 mW cm(-2)) irradiation on a TiO(2)-coated glass plate using the ISO methodology. The results indicated that both bacteriophages were inactivated at 0.001 mW cm(-2) UVA. The intensity of UV light, including long-wavelength light (UVA), is very low in an actual indoor environment. Thus, TiO(2) photocatalysis can be beneficial for inactivating viruses in an indoor environment. Experiments using qPCR and bovine serum albumin degradation assume that viral inactivation is caused by outer viral protein disorder and not by viral RNA reduction by reactive oxygen species produced during TiO(2) photocatalysis. Furthermore, we showed that the ISO methodology for standard testing of antibacterial activity by TiO(2) photocatalysis can be applied to assess antiviral activity.  相似文献

A photocatalyst was applied to a lancet for pricking the finger to obtain an antibacterial property. A photocatalytic and uniform nano-layer of titanium dioxide (TiO2) on the surface of the lancet (0.36 mm x 24.5 mm) was formed by sputtering and annealed for crystallization of the TiO2 layer. By elementary analysis of the TiO2 layer, titanium and oxygen were detected. Next, for the estimation of the antibacterial properties resulting from the photocatalytic effect, the lancet was packed into a capillary tube filled with a suspension of Escherichia coli K-12 (non-spore-forming bacterium), and was continuously rolled in a continuous UV-irradiation system under black-light irradiation. Distinct antibacterial effects after irradiation at 0.5 mW cm(-2) for 45 min were observed in the crystallized TiO2 layer on the lancet. Finally, lancing resistances obtained by pricking an artificial skin sheet were examined using control lancets, and lancets with an unannealed TiO2 layer or an annealed TiO2 layer. The results showed almost the same lancing resistances for the control (0.53+/-0 N, n=3) and the lancet with an annealed TiO2 layer (0.51+/-0.018 N), while the lancet with an unannealed TiO2 layer showed a high lancing resistance compared with the other lancets (0.62+/-0.05 N). In conclusion, the lancet coated with a crystallized, velvety nano-layer of TiO2 obtained by annealing had antibacterial properties and a similar lancing resistance compared with the bare lancet, and showed potential for application in monitoring blood glucose in diabetes.  相似文献

This work focuses on the photocatalytic performances and antibacterial activity of nitrogen doped TiO2 nanosystems with three and five layers obtained by a sol-gel route, followed by thermal treatment in oxygen or ammonia atmosphere at temperatures between 400 and 1000°C. Subsequently, the antibacterial activity of the obtained nanosystems on the Escherichia coli cells are determined and discussed. The obtained results show a significant dependence of the functional performances on the system’s composition. In particular, the antimicrobial activity of nitrogen-doped TiO2 films is correlated with the temperature of thermal treatment and illumination time with visible artificial light.  相似文献

Three microplate-based viability assays for assessing the antibacterial effects of photocatalytic coatings were compared to the conventional colony count method. In the experimental design, cultured Escherichia coli were exposed to photocatalysis on various TiO2 films in the presence of either UVA or visible light. The photocatalytic effects on the bacterial physiology were determined by real-time measurements of metabolic activity (XTT assay), biomass formation in the liquid medium (growth assay), and by assessing membrane integrity (with propidium iodide and SYTO 9 fluorescent nucleic acid binding dyes—BacLight assay). All three methods proved to be more sensitive and reproducible than colony count for the evaluation of the bactericidal effect of photocatalysis, XTT, and growth assay succeeded in detecting differences in both UVA and visible light-activated photocatalytic coatings. BacLight could efficiently detect the visible light-dependent photocatalytic effect on bacteria and identify membrane damage, but resulted inadequate for evaluating the UVA-dependent antibacterial effects. The described microplate-based evaluation methods proved being more effective and rapid than the colony count assay for assessing the antibacterial effect of various photocatalytic coatings.  相似文献

Quaternized poly(vinylpyridine) is known to kill up to 99% of drug-resistant gram-positive and -negative bacteria but shows minimal biocompatibility. We report enhanced bactericidal activity of vinylpyridine through copolymerization with hydroxyethyl methacrylate and poly(ethylene gycol) methyl ether methacrylate. Copolymers with increasing comonomer content were synthesized by radical polymerization and quaternized with hexylbromide. We assessed the effects of the changes in polymer composition on the bactericidal activity of the surface activity using a bioluminescent pathogenic strain of Escherichia coli (O157:H7). By recording the photoluminescence emitted by these bacteria in contact with the copolymers, it was shown that several of the copolymers possess better antibacterial efficiency than quaternized poly(vinylpyridine). Results indicate that several of the copolymers synthesized possess antibacterial activity approximately 20 times greater than the pure quaternized poly(vinylpyridine) homopolymer, while only containing 1 wt % hexylated pyridinium. This behavior is explained by the increased surface wettability of the copolymers containing lesser amounts of poly(vinylpyridine), as bactericidal behavior correlates to the hydrophilicity of the system as measured by contact angles. A hydrophilicity based design-paradigm can significantly improve both the efficacy and the biocompatibility of antibacterial materials.  相似文献

A decline in CFU of gram-positive and gram-negative bacteria on the surface of UV illuminated TiO2 films (wavelength of 380 nm) is shown. A 29, 45, and 47% decrease in bacterial viability of Staphylococcus aureus, Staphylococcus epidermidis, and Escherichia coli, respectively, was seen following a 12-min exposition. It was first discovered that the reuse of TiO2 films to test a bacterial suspension for viability removes UV-induced bactericidal activity. However, annealing of TiO2 at a temperature above 400 degrees C restores the photoinduced bactericidal activity to its initial state.  相似文献

Cu (II) and Ag(I) together with TiO2 powder were deposited on conducting support substrates to enhance the photocatalytic ability. The catalytic efficiency was tested by monitoring the photocatalytic degradation and detriment of methylene blue (MB) and bovine serum albumin (BSA). The conformational change of BSA induced by catalysts was also observed by circular dichroism spectroscopy.The antibacterial activities were studied by Escherichia coli. Both MB and BSA could be degraded more efficiently than pure TiO2. After treatment with catalyst, the morphology of cells became twisted and rougher. Regular wrinkles were damaged and groove-like rift appeared on the surface. The fluorescence polarization has shown a significant decrease in membrane fluidity and the increase of permeability of cell membrane. Changes of the spectral profile of E. coli were observed, which suggested the damages of surface groups on the cell membrane.  相似文献

Background

Titania dioxide (TiO2) photocatalyst is primarily induced by ultraviolet light irradiation. Visible-light responsive anion-doped TiO2 photocatalysts contain higher quantum efficiency under sunlight and can be used safely in indoor settings without exposing to biohazardous ultraviolet light. The antibacterial efficiency, however, remains to be further improved.

Methodology/Principal Findings

Using thermal reduction method, here we synthesized silver-nanostructures coated TiO2 thin films that contain a high visible-light responsive antibacterial property. Among our tested titania substrates including TiO2, carbon-doped TiO2 [TiO2 (C)] and nitrogen-doped TiO2 [TiO2 (N)], TiO2 (N) showed the best performance after silver coating. The synergistic antibacterial effect results approximately 5 log reductions of surviving bacteria of Escherichia coli, Streptococcus pyogenes, Staphylococcus aureus and Acinetobacter baumannii. Scanning electron microscope analysis indicated that crystalline silver formed unique wire-like nanostructures on TiO2 (N) substrates, while formed relatively straight and thicker rod-shaped precipitates on the other two titania materials.

Conclusion/Significance

Our results suggested that proper forms of silver on various titania materials could further influence the bactericidal property.  相似文献

Mammals have four peptidoglycan recognition proteins (PGRPs or PGLYRPs), which are secreted innate immunity pattern recognition molecules with effector functions. In this study, we demonstrate that human PGLYRP-1, PGLYRP-3, PGLYRP-4, and PGLYRP-3:4 have Zn(2+)-dependent bactericidal activity against both Gram-positive and Gram-negative bacteria at physiologic Zn(2+) concentrations found in serum, sweat, saliva, and other body fluids. The requirement for Zn(2+) can only be partially replaced by Ca(2+) for killing of Gram-positive bacteria but not for killing of Gram-negative bacteria. The bactericidal activity of PGLYRPs is salt insensitive and requires N-glycosylation of PGLYRPs. The LD(99) of PGLYRPs for Gram-positive and Gram-negative bacteria is 0.3-1.7 muM, and killing of bacteria by PGLYRPs, in contrast to killing by antibacterial peptides, does not involve permeabilization of cytoplasmic membrane. PGLYRPs and antibacterial peptides (phospholipase A(2), alpha- and beta-defensins, and bactericidal permeability-increasing protein), at subbactericidal concentrations, synergistically kill Gram-positive and Gram-negative bacteria. These results demonstrate that PGLYRPs are a novel class of recognition and effector molecules with broad Zn(2+)-dependent bactericidal activity against both Gram-positive and Gram-negative bacteria that are synergistic with antibacterial peptides.  相似文献

用超声驱动方法合成了CdSe/TiO_2复合纳米粒子,并通过扫描电镜(SEM)、透射电镜(TEM)、X射线衍射(XRD)、紫外可见光(UV-Vis)吸收光谱和荧光(FS)光谱对CdSe/TiO_2复合纳米粒子进行表征。使用CCK-8法测定CdSe/TiO_2对白血病细胞的可见光光催化活性并通过SEM研究HL60细胞的表面超微结构形态。实验结果表明,用CdSe/TiO_2复合纳米粒子处理的组中观察到明显的HL60细胞生长抑制,并且HL60细胞在CdSe掺杂TiO_2复合纳米粒子作用下的PDT效率显著高于TiO_2,表明可以通过CdSe的修饰有效增强TiO_2的可见光光催化活性。此外,CdSe/TiO_2在可见光辐照下在4μg/m L的终值浓度下显示非常高的光动力效率,达76%。荧光光谱分析表明,CdSe/TiO_2复合纳米粒子可能通过分离光生空穴电子对提高此复合纳米粒子的光催化活性,从而提高CdSe/TiO_2对HL60细胞的PDT灭活效率。  相似文献

Etimicin is a novel fourth generation semisynthetic aminoglycoside. It has good antimicrobial activity against both gram-positive and gram-negative bacterial infections and also against aminoglycoside resistant strains. In the present study, in vitro antibacterial activity of etimicin was determined by minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and time kill curve tests against type strains and 407 clinical isolates (obtained in a surviellance study), in comparison to other aminoglycosides. Test results revealed that etimicin has potential antimicrobial activity and MIC, MBC values for etimicin were low compared to other aminoglycosides. In MBC test etimicin has exhibited potential bactericidal effect ranging from 0.25 to 2?mg/L. The time kill-curve study further demonstrated the rapid, concentration dependent killing and comparative study showed etimicin to exhibit long and effective bactericidal activity over amikacin. The interesting fact is that most of the tested aminoglycoside resistant clinical isolates were susceptible to etimicin. In view of its potent in vitro antibacterial activity and efficacy profiles, it can be concluded that etimicin can be a potent injectable agent for the treatment of severe bacterial infections.  相似文献

Mango ginger (Curcuma amada Roxb.) rhizome is used in the manufacture of pickles and other food preparations due to its unique raw mango flavour. The chloroform extract of mango ginger rhizome was subjected to antibacterial activity-guided purification by repeated silica gel column chromatography to obtain a pure compound. The structure of the isolated compound was deduced by analysing UV, IR, LC-MS and 2D-HMQCT NMR spectral data, and named it as amadaldehyde, a novel compound. It exhibited a wide range of antibacterial activity with potential bactericidal activity against several bacteria. The purified compound also exhibited antioxidant activity, cytotoxicity and platelet aggregation inhibitory activities.  相似文献

TiO2纳米管阵列由于其优异的生物相容性及光催化效应,在生物医学领域引起了广泛关注.但能否将肾小管上皮细胞较好地黏附于TiO2纳米管材料并使其发挥肾小管的功能,目前还未见报道.为研究TiO2纳米管材料的表面特性对猪肾小管上皮细胞株(LLC-PK1)的黏附及增殖影响,采用阳极氧化法制备新型高强度的TiO2纳米管阵列,利用荧光显微镜考察了TiO2纳米管阵列光照特性、晶型结构及几何形貌参数对LLC-PK1细胞黏附的影响,采用MTT方法检测了黏附细胞的活性;同时使用扫描电子显微镜观察了4种不同管径上细胞生长的形态,并与纯钛片上细胞的生长形态进行对照.结果表明,管径为70nm的TiO2纳米管阵列膜最有利于LLC-PK1细胞的黏附及增殖,且细胞的活性最高;未经紫外光照射时锐钛矿型的TiO2比无定型更有利于细胞的黏附,然而锐钛矿型TiO2经紫外光照射后会导致细胞凋亡.扫描电子显微镜观察显示,细胞在纳米管上延伸为长条状,而在钛片上则呈堆积平板状.证实TiO2纳米管阵列膜具有良好的生物相容性,有助于改善细胞与材料的黏附.  相似文献

Naturally occurring cationic antimicrobial peptides (CAPs) are an essential component of the innate immune system of multicellular organisms. At concentrations generally higher than those found in vivo, most CAPs exhibit strong antibacterial properties in vitro, but their activity may be inhibited by body fluids, a fact that could limit their future use as antimicrobial and/or immunomodulatory agents. In the present study, we evaluated the effects of human serum components on bactericidal activity of the human beta-defensin 3 (hBD-3), a CAP considered particularly promising for future therapeutic employment. Human serum diluted to 20% strongly inhibited the bactericidal activity of the peptide against both the Gram-positive species Staphylococcus aureus and the Gram-negative species Acinetobacter baumannii. Such activity was not restored in serum devoid of salts (dialyzed), pre-treated with protease inhibitors, or subjected to both of these treatments. The addition of physiological concentrations of NaCl, CaCl2, and human albumin in the bactericidal assay abolished bactericidal activity of hBD-3 against S. aureus, while it only partially inhibited the activity of the peptide against A. baumannii. Although a proteolytic activity of serum on hBD-3 was demonstrated at the protein level by Western blot, addition of physiological concentrations of trypsin to the bactericidal assay only partially affected the antibacterial properties of the peptide. Altogether, these results demonstrate a major role of mono-divalent cations and serum proteins on inhibition of hBD-3 antibacterial properties and indicate a relative lack in sensitivity of the bactericidal activity of this peptide to trypsin and trypsin-like proteases.  相似文献

A bactericidal domain, P(18-39), of the proteinase inhibitor aprotinin, possesses the structural feature of two antiparallel beta-sheets connected by a short turn. In order to understand the structural requirements for antibacterial activity, two peptides, each having the sequence corresponding to a single beta-sheet structure of P(18-39), were synthesized and their antibacterial properties investigated. One peptide, P(18-28), with the sequence IIRYFYNAKAG, was active against almost all the bacterial strains investigated. However, the bactericidal activity of P(18-28) was reduced compared to the parent molecule, P(18-39). The other peptide, P(29-39), with the sequence LCQTFVYGGCR, was only weakly bactericidal against Pseudomonas aeruginosa. A peptide, P(18-26), devoid of the C-terminus dipeptide Ala-Gly of P(18-28), retained the bactericidal activity of P(18-28) against most of the bacterial strains investigated. Only Klebsiella pneumoniae, P. aeruginosa and Staphylococcus aureus were resistant to P(18-26). Replacement of lysine 26 by arginine in P(18-26) (IIRYFYNAR) improved the bactericidal activity. The retropeptide, RANYFYRII, retained the antibacterial activity of IIRYFYNAR toward Gram-negative bacteria, but it was less active against Gram-positive bacteria. The random peptide, IANRIYRYF, was as bactericidal as IIRYFYNAR. Moreover, the random peptide possessed, in contrast to IIRYFYNAR, a strong antifungal activity against Candida albicans. Elimination of the N-hydrophobic terminal Ile-Ile from P(18-26) (RYFYNAK) strongly reduced the bactericidal potency of the peptide. Attaching the hydrophobic peptide, FFVAP, to the C-terminal of P(18-26) (IIRYFYNAKFFVAP) increased the bactericidal potency of the peptides considerably. We concluded that the order of the amino acids in the sequence of the peptides is not, per se, a critical feature for bactericidal activity. Hydrophobic interaction between peptide and bacterial membrane is probably the most important feature involved in the bactericidal mechanism of the antibiotic peptides.  相似文献

When titanium dioxide (TiO(2)) is irradiated with near-UV light, this semiconductor exhibits strong bactericidal activity. In this paper, we present the first evidence that the lipid peroxidation reaction is the underlying mechanism of death of Escherichia coli K-12 cells that are irradiated in the presence of the TiO(2) photocatalyst. Using production of malondialdehyde (MDA) as an index to assess cell membrane damage by lipid peroxidation, we observed that there was an exponential increase in the production of MDA, whose concentration reached 1.1 to 2.4 nmol. mg (dry weight) of cells(-1) after 30 min of illumination, and that the kinetics of this process paralleled cell death. Under these conditions, concomitant losses of 77 to 93% of the cell respiratory activity were also detected, as measured by both oxygen uptake and reduction of 2,3,5-triphenyltetrazolium chloride from succinate as the electron donor. The occurrence of lipid peroxidation and the simultaneous losses of both membrane-dependent respiratory activity and cell viability depended strictly on the presence of both light and TiO(2). We concluded that TiO(2) photocatalysis promoted peroxidation of the polyunsaturated phospholipid component of the lipid membrane initially and induced major disorder in the E. coli cell membrane. Subsequently, essential functions that rely on intact cell membrane architecture, such as respiratory activity, were lost, and cell death was inevitable.  相似文献
