Using quartz optical fibres, penetration of both monochromatic (310 nm) and polychromatic UV-B (280–320 nm) radiation in leaves of Brassica napus L. (cv. Ceres) was measured. Plants were grown under either visible light (750 μmol m−2 s−1 photosynthetically active radiation) or with the addition of 8. 9 KJ m−2 day−1 biologically effective UV-B (UV-BBE) radiation. Results showed that of the 310 nm radiation that penetreated the leaf, 90% was within the intial one third of the leaf with high attenuation in the leaf epidermis, especially in UV-treated plants. Polychromatic UV-B radiation, relative to incident radiation, showed a relatively uniform spectral distribution within the leaf, except for collimated radiation. Over 30% of the UV-screening pigments in the leaf, including flavonoids, were found in the adaxial epidermal layer, making this layer less transparent to UV-B radiation than the abaxial epidermis, which contained less than 12% of the UV-screening pigments. UV-screening pigments increased by 20% in UV-treated leaves relative to control leaves. Densely arranged epicuticular wax on the adaxial leaf surface of UV-treated plants may have further decreased penetration of UV-B radiation by reflectance. An increased leaf thickness, and decreases in leaf area and leaf dry weight were also found for UV-treated plants.  相似文献

Although the oil body is known to be an important membrane enclosed compartment for oil storage in seeds, we have little understanding about its biogenesis during embryogenesis. In the present study we investigated the oil body emergence and variations in Brassica napus cv. Topas. The results demonstrate that the oil bodies could be detected already at the heart stage, at the same time as the embryos began to turn green, and the starch grains accumulated in the chloroplast stroma. In comparison, we have studied the development of oil bodies between Arabidopsis thaliana wild type (Col) and the low-seed-oil mutant wrinkled1-3. We observed that the oil body development in the embryos of Col is similar to that of B. napus cv. Topas, and that the size of the oil bodies was obviously smaller in the embryos of wrinkled1-3. Our results suggest that the oil body biogenesis might be coupled with the embryo chloroplast.  相似文献

以甘蓝型油菜新鲜嫩叶为实验材料提取其总DNA,以其为模板,根据拟南芥Toc33基因编码区序列设计引物,PCR扩增甘蓝型油菜叶绿体外膜蛋白转运机器的构件蛋白基因Toc33,得到两条扩增带,测序结果显示克隆到的两个片段分别长1370bp、1490bp,将这两个片段分别命名为Bn Tpc33-1,Bn Toc33-2,序列比较发现它们之间的同源性为78%,其中外显子的同源性为96%,而内含子的同源性仅为60%。为研究Toc33与同一基因家族的Toc34基因功能间的关系,对拟南芥、油菜、诸葛菜等植物的Toc33、Toc34蛋白序列进行比较分析并构建了分子系统进化树。  相似文献

We conducted three experiments to examine the influence of ultraviolet-B radiation (UV-B; 280–320 nm) exposure on reproduction in Brassica rapa (Brassicaceae). Plants were grown in a greenhouse under three biologically effective UV-B levels that simulated either an ambient stratospheric ozone level (control), 16% (“low enhanced”), or 32% (“high enhanced”) ozone depletion levels at Morgantown, WV, USA in mid-March. In the first experiment, we examined whether UV-B level during plant growth influenced in vivo pollen production and viability, and flower production. Pollen production and viability per flower were reduced by ≈50% under both enhanced UV-B levels relative to ambient controls. While plants under high-enhanced UV-B produced over 40% more flowers than plants under the two lower UV-B treatments, whole-plant production of viable pollen was reduced under high-enhanced UV-B to 17% of that of ambient controls. Whole-plant production of viable pollen was reduced under low-enhanced UV-B to 34% of ambient controls. In the second experiment, we collected pollen from plants under the three UV-B levels and examined whether source-plant UV-B exposure influenced in vitro pollen germination and viability. Pollen from plants under both enhanced-UV-B treatments had initially lower germination and viability than pollen from the ambient level. After in vitro exposure to the high-enhanced UV-B levels for 6 h, viability of the pollen from plants grown under ambient UV-B was reduced from 65 to 18%. In contrast, viability of the pollen from plants grown under both enhanced UV-B treatments was reduced to a much lesser extent: only from ≈43 to 22%. Thus, ambient source-plant pollen was more sensitive to enhanced UV-B exposure. In the third experiment, we used pollen collected from source plants under the three UV-B levels to fertilize plants growing under ambient-UV-B levels, and assessed subsequent seed production and germination. Seed abortion rates were higher in plants pollinated with pollen from the enhanced UV-B treatments, than from ambient UV-B. Despite this, seed yield (number and mass) per plant was similar, regardless of the UV-B exposure of their pollen source. Our findings demonstrate that enhanced UV-B levels associated with springtime ozone depletion events have the capacity to substantially reduce viable pollen production, and could ultimately reduce reproductive success of B. rapa.  相似文献

Peroxidase activity in isolated thylakoids from degreening canola ( Brassica napus cv. Westar) seeds was demonstrated. The enzyme catalyzes the degradation of thylakoid-bound pigments in the presence of H2O2 and 2,4-dichlorophenol. Peroxidase activity is related to degreening, with periods of rapid degreening associated with high enzyme activity. Both de novo synthesis and substrate availability appear to control enzyme activity. Peroxidase is initially inhibited and then stimulated by sublethal freezing. Therefore, inhibition of peroxidase activity following sublethal freezing may be responsible, in part, for a failure of the seed to degreen.  相似文献

根据已知序列设计引物,通过PCR扩增获得质体定位的乙酰辅酶A羧化酶的4个亚基的基因序列。先将该酶4个亚基的基因进行拼接,然后将这4个拼接好的片段,克隆到pMD18-T载体上,得到质粒pH BM714。再以质粒pHBM714 DNA为模板,用分别带有CpoI和Asc I酶切位点的引物进行PCR扩增,PCR产物在dTTP的保护下经T4 DNA聚合酶处理,与将质粒pHBM720DNA纯化后经CpoI和AscI双酶切后得到的大片段连接,连接产物转化大肠杆菌Xl_(10)-gold,得到正确的重组子命名为pHBM726。此质粒pH BM726,即为带有壮观霉素抗性基因(aadA)筛选标记的质体定位的乙酰辅酶A羧化酶基因油菜叶绿体单交换表达载体;在此载体中壮观霉素抗性基因(aadA)、乙酰辅酶A羧化酶的4个亚基的基因(ACC)和绿色荧光蛋白基因(gfp)共6个基因串联在一起,共用一个启动子序列,一起来进行表达;通过酶切检测、PCR验证和测序验证,均表明该表达载体构建成功。最后此载体在大肠杆菌中表达时,发现重组菌能够在含壮观霉素的培养基上生长,且在可见光下,能看到绿色荧光,表明壮观霉素抗性基因和绿色荧光蛋白基因均在大肠杆菌中成功表达;表达产物通过Western印迹验证表明组成乙酰辅酶A羧化酶的4个亚基的基因在大肠杆菌中成功表达。以上结果表明,该表达载体中串联排列的这6个基因均在大肠杆菌中成功表达。该研究结果可为质体定位的乙酰辅酶A羧化酶转叶绿体的研究奠定基础,为油菜油脂代谢研究提供参考。  相似文献

Influences of UV-B radiation on Rhizophora apiculata were studied in terms of chlorophylls, their presence in protein complexes of the chloroplast, PS I and PS II photochemical activities, in vitro absorption spectrum of the chloroplast, in vivo leaf fluorescence and UV absorbing compounds. The seedlings were exposed to the various levels of UV-B radiations, equivalent to 0 (control), 10, 20, 30 and 40% stratospheric ozone depletion of the study area. The low doses of UV-B (10 and 20%) increased the reaction centre chlorophyll (10 and 8%) and activities of PS-I (98 and 39%) and PS-II (77 and 38%) respectively; whereas, 30 and 40% UV-B treatments decreased the reaction centre chlorophylls by 11 and 33% and PS II activity by 0 and 20%; while PS I activity did not show any inhibitory effect. Chloroplasts isolated from control and 10% UV-B treated plants exhibited the same level of absorption at 676 nm. In vivo leaf fluorescence was found to be diminished with UV-B radiation and at the 10% UV-B, variable fluorescence was promoted significantly by 10%. The content of UV-absorbing compounds was progressively enhanced with doses of UV-B radiation along with higher absorption at 276 and 330 nm.  相似文献

Abstract

A study was made on the changes observed in the protein, starch and soluble sugar content during development of siliquas and seeds of rapeseed grown in central Italy.

Concentration of starch and soluble sugars in the seed increases to 75 per cent dry matter during the first few weeks of pod development and then drops to minimum values. The protein increases steadily until maturity, when a level of 0.85 mg per seed is reached, equivalent to 18 per cent dry matter. The protein and starch in the hull? decrease continuously during development, while in the initial stages the soluble sugars are accumulated until they account for 33 per cent dry matter, after which they decline towards maturity.  相似文献

以生长在非控温控光下的4个冬性甘蓝型油菜(BrassicanapusL.)品种为供体,进行游离小孢子培养。研究发现,多数品种在开花3-7天取材最为适宜。在甘蓝型油菜小孢子培养中只有单核晚期的小孢子才可能发育成胚状体,而花药培养时处于单核早期的小孢子易于发育成胚状体。在适当花期选取发育比较一致的单核晚期小孢子培养,经数小时后,部分小孢子便开始膨大,这是小孢子发育成胚的最早标志,膨大的小孢子中,有部分形成多细胞球并进一步发育成胚。用春性甘蓝型油菜为材料进行蔗糖浓度的实验结果表明:培养3天后,在16%蔗糖培养基中存活的小孢子最多,达16.13%;培养30天后,胚状体诱导频率则以13%蔗糖浓度为最高,每花蕾可达144个胚状体。如果在16%蔗糖培养基中培养3天后,添加等体积的13%蔗糖培养基,能够大大提高胚状体的诱导频率,为仅用13%蔗糖培养基培养的3.7倍。这一实验体系正在用于抗菌核病的诱变与筛选,并作为外源基因导入的实验体系。  相似文献

Cultured microspores of Brassica napus L. cvs Topas and Reston initiated cell divisions within 3 to 4 days, and globular, heart and torpedo shaped embryos were prevalent after approximately 6, 8, and 10 days, respectively. Embryos with rudimentary cotyledons were evident within 2 weeks, but those that reached this stage of development represented only 1–5% of the original microspore population. The fresh weight of microspore-derived embryos at all stages of development was significantly greater than that for zygotic embryos, but the pattern of change in fresh weight and fatty acid accumulation was similar in developing zygotic and microspore embryos. In freshly isolated microspores of both Topas (low erucic acid) and Reston (high erucic acid), the predominant fatty acid was 18:3, while 18:1 comprised less than 15% of total fatty acids. During development in both zygotic and microspore embryos, the level of 18:3 declined markedly while 18:1 rapidly increased. Erucic acid (22:1) was not detected in the early stages of embryogenesis in Reston. However, small amounts of 22:1 appeared by early cotyledonary stage and the level gradually increased in both zygotic and microspore embryos through the later stages of development. The fatty acid compositions of mature embryos was nearly identical to that of dry seed, except the level of 22:1 in Reston embryos was consistently less than in the seed. Triacylglycerols comprised only 15% of total lipids in freshly isolated microspores, but increased to more than 90% by 4 weeks. The fatty acid composition of the triacylglycerol fraction was generally similar to that of total lipids at all stages of development of microspore-derived embryos.  相似文献

In order to study Brassica napus fatty acid (FA) metabolism and relevant regulatory networks, a systematic identification of fatty acid (FA) biosynthesis-related genes was conducted. Following gene identification, gene expression profiles during B. napus seed development and FA metabolism were performed by cDNA chip hybridization (〉8000 EST clones from seed). The results showed that FA biosynthesis and regulation, and carbon flux, were conserved between B. napus and Arabidopsis. However, a more critical role of starch metabolism was detected for B. napus seed FA metabolism and storage-component accumulation when compared with Arabidopsis. In addition, a crucial stage for the transition of seed-to-sink tissue was 17-21 d after flowering (DAF), whereas FA biosynthesis-related genes were highly expressed pri- marily at 21 DAF. Hormone (auxin and jasmonate) signaling is found to be important for FA metabolism. This study helps to reveal the global regulatory network of FA metabolism in developing B. napus seeds.  相似文献

Saplings of pedunculate oak (Quercus robur L.) were exposed at an outdoor facility to modulated levels of elevated UV-B radiation (280–315 nm) under treatment arrays of cellulose diacetate-filtered fluorescent lamps which also produced UV-A radiation (315–400 nm). Saplings were also exposed to UV-A radiation alone under control arrays of polyester-filtered lamps and to ambient levels of solar radiation under arrays of unenergized lamps. The UV-B treatment corresponded to a 30% elevation above the ambient level of erythemally weighted UV-B radiation. Sapling growth and the occurrence of associated organisms were examined over two years. In both years, leaves of saplings exposed to UV-B treatment were thicker and smaller in area relative to leaves exposed to ambient and control levels of radiation. UV-B treatment also retarded bud burst at one sampling in the first year of the study. Some responses were recorded which were common to both treatment and control arrays, implying that UV-A radiation, or some other factor associated with energized lamps, was responsible for the observed effects. Saplings under treatment and control arrays were taller in the first year of the study, suffered greater herbivory from chewing insects, and had lower root dry weights and greater insertion heights of secondary branches than saplings exposed to ambient levels of radiation. Exposure of saplings to elevated UV-A radiation alone under control arrays increased estimated leaf volumes in the second year of the study and reduced the number of secondary branches and the total number of branches per sapling after two years, relative to both treatment and ambient arrays. There were no effects of elevated ultraviolet radiation on shoot or total plant weight, root/shoot ratios, stem diameter, the numbers or insertion heights of primary or tertiary branches, total leaf number, timing of leaf fall or frequency of ectomycorrhizas. Our study suggests that any increases in UV-B radiation as a result of stratospheric ozone depletion will influence the growth of Q. robur primarily through effects on leaf morphology.  相似文献

Our knowledge of the effects of increased levels of ultraviolet-B radiation (UV-B) on plant fitness is limited mainly to yield studies in a few crop species. Previous greenhouse and garden studies of Brassica have found greater detrimental effects of UV-B on fitness in gardens than in the greenhouse, suggesting the possibility that additional stresses in the field decrease the ability of Brassica to cope with UV-B. Possible interactions between UV-B and water/nutrient stress in determining plant fitness have rarely, if ever, been studied experimentally. Here we report measurements of female fitness in two species of Brassica in an experiment in which both UV-B and levels of water and nutrients were varied in a 2 x 2 factorial design. Water and nutrient stress reduced female fitness in both species, while UV-B caused fitness reductions in only one of the species. There was evidence for interactions between UV-B and water/nutrient stress for only a few of the traits measured; most traits, including those closely related to fitness, showed no evidence of an interaction.  相似文献

本研究在NLN-16和NLN-13的培养基中分别加入0.1mg/L 6-BA和0.05%的活性碳,结果表明6-BA对小孢子再生胚有明显地促进作用,再生胚的频率比对照增加26枚/皿,经分析达到显著水平;而0.05%的活性碳对小孢子再生胚促进作用不显著。对甘蓝型油菜小孢子培养再生植株的染色体加倍及移栽的研究结果表明,在小孢子培养初期加50mg/L秋水仙碱加倍效率最佳,加倍率达到67.6%。小孢子培养的再生苗移栽至大田后,采用遮阳网覆盖小苗,移栽成活率达到87.6%。 Abstract:The application of microspore culture technique was restricted because of its low frequency of embryogenesis and chromosome doubling.Two methods of enhancing the frequency of embryogenesis were employed in the study,namely,activated charcoal treatment in NLN-13 media and 6-BA treatment in NLN-16 media.The treatment with 0.05% activated charcoal produced 24 embryos per plate,which increased 1.7 embryos per plate,as compared with the treatment without activated charcoal.However,the analysis of T-test showed that it was not significant.After adding 0.1mg/L 6-BA in NLN-16 media,the frequency of embryogeny was 38.3 embryos per plate,and it was 26 embryos more per plate than that of CK.Analysis of T-test is significant.This indicates that 6-BA promotes embryogeny in microspore culture.Adding 50mg/L colchicines in NLN-16 media,the doubling frequency was 67.6%.The plantlets transplanted into field with two methods of light-covered net and plastic films were investigated.A survival rate of 87.6% was obtained using light-covered method whereas 57.7% survived using plastic film method.  相似文献

从甘蓝型油菜 (Brassicanapuscv .H1 65)叶绿体基因组克隆得到了编码核糖体蛋白的基因rps7。经序列分析得知 ,该基因编码区包含 468个核苷酸 ,编码一个分子量为 2 0 1 0 9D、由 1 55个氨基酸组成的蛋白质。该基因的核苷酸和编码的氨基酸序列与烟草对应基因的同源性皆高达 97% ;而与水稻对应基因的同源性分别为 90 %和 84%。该基因不含内含子 ,没有典型的SD序列 ,但在 5’端 - 2 5~- 2 2位发现一个与烟草psbA基因的顺式作用元件RBS2完全相同的TGAT框。与烟草和水稻的同源序列比较 ,该基因在 3’端非编码区变异较大 ,发生了多次插入和缺失。构建了包含该基因在内的一个 1 .0kbDNA的限制性内切酶图谱。所报告的基因序列已登录GenBank。  相似文献

Endogenous cytokinin levels were analyzed in four different organs viz., root, stem, leaf and mature flower, of the wild type (cv. Westar) and a genic male sterile (GMS) line of Brassica napus . Various cytokinins viz., bases (Z, DZ), ribosides (ZR, DZR), glucosides (OGZ + OGZR, OGDZ + OGDZR) and nucleotides (ZNT, DZNT) were quantified using Amaranthus betacyanin bioassay and ELISA. The major cytokinin in all the organs was DZ. In general, leaves had the highest levels of cytokinins as compared to the other organs. Root, stem and mature flowers of the wild type plants had higher levels of cytokinins and their metabolites than the GMS line. However. levaves of the GMS line had greater amount of various cytokinins as compared to the wild type. The major cytokinin quantitatively different between the two lines was DZ. The results suggest the possible involvement of cytokinins in the expression of genic male sterility in Brassica napus .  相似文献
