The Sendai virus-induced interferon (IF) production by partially purified human leucocyte suspensions of normal donors and of leukaemic patients have been investigated in vitro. (i) An increased production was observed with leucocytes taken from patients suffering from chronic myelogenous leukaemia (CML) during exacerbation, but the production was approximately normal with cells taken during remission. (ii) IF production was not influenced by large doses of cytostatics (DBM, 5-FU, FUDR, 5-HU, 6-MP, cyclophosphamide) irrespective of whether normal or CML leucocytes were used. (iii) In contrast, production was inhibited in both systems by inhibitors of RNA or protein synthesis (actinomycin D, puromycin, cycloheximide). (iv) CML leucocytes produced IF for a prolonged period of time as compared to normal leucocytes. (v) Leucocytes from children with acute blastic leukaemia and those from adults with chronic lymphoid or acute paramyeloblastic leukaemia produce, in contrast to normal leukocytes, approximately as much IF in the absence as in the presence of serum. It is concluded that the Sendai virus-induced IF synthesis in CML leucocytes requires de novo protein synthesis.  相似文献

The effects of interferon (IF) on erythropoietin (Ep) action and production were studied in mice. In comparison to control animals, Ep action in exhypoxic, polycythemic mice was significantly decreased (p less than 0.05) following two low dose injections of IF (2.9-3.5 X 10(4) units). In addition, renal Ep production in normal intact mice was also significantly decreased (p less than 0.01) following a single injection of IF (5.4-6.3 X 10(4) units) and hypoxic exposure.  相似文献

In vitro interferon (IF) production in peritoneal macrophages of normal and Newcastle disease virus (NDV)-immunized mice was studied. Of ascites cells used, 80% were macrophages, 14% lymphocytes, and 6% polymorphonuclear leukocytes. It was indicated that IF was produced mainly in the macrophages after NDV inoculation. IF production in the macrophages derived from immunized mice was more enhanced than that in those from normal mice. It is not clear at present, however, whether this enhancement is based on immunological specificity. The IF production in the culture of macrophages reached its maximum value in 6 to 9 hr after inoculation of the inducer. After 12 hr, the IF titer in the culture fluid decreased gradually. A possible explanation of this fact is that there may be partial inactivation of IF by some cellular components.  相似文献

The production of the virus-inhibiting factor or interferon (IF) was highest in cells incubated at 37 C after inoculation with Newcastle disease (ND) virus and decreased as the incubation temperature was lowered. Shift-down of incubation temperature to 32 C or 34 C after incubation at 37 C for 4–7 hr enhanced IF production in cell cultures stimulated with ND virus, as compared with cultures incubated continuously at 37 C. Shift-down to 32 C after incubation at 37 C for 6 hr. was optimal for this enhancement of IF yield. Enhanced IF production was also observed in cell cultures irradiated by ultraviolet light 4–7 hr after stimulation with ND virus.  相似文献

Several factors can be responsible for the febrile response evoked by interferon (IF) administration in man. Since different cells and inducers are used for production of IF, the pyrogenic substance may not always be the same. Leucocyte IF (LeIF) may contain endogeneous pyrogen (EP), fibroblast IF (FIF) may contain heterogeneous proteins and/or EP, and lymphoblastoid IF (LyIF) may contain a factor released by the transformed lymphocyte able to induce, in vivo, the release of EP from leucocytes. Finally, IF may be intrinsically pyrogenic and, if this is the case, until homogeneous IF is used, it may potentiate the action of the pyrogenic contaminants.  相似文献

Summary Large scale production of human lymphoblastoid (Namalva) interferon (IF) is described. Cell propagation, in up to 50 1 culture volume, was carried out in a low cost medium by a semi-continuous cultivation method. IF was induced by Sendai virus, testing two induction methods. The yield of crude IF varied in the range of 12 – 100 × 103 IF units.ml-1. A weekly production output of 1 – 5 × 108 units crude IF was obtained.  相似文献

Pretreatment of mouse L cells with interferon (IF) enhanced IF production in response to polyinosinic-polycytidylic acid (poly I-poly C). Post-treatment of cells with IF caused no significant enhancement of IF production. The enhancing effect of IF pretreatment (priming) reached a maximum after incubation with IF (10 or 100 units/ml) for 4-6 hr at 37 C, but this effect was absent when the incubation was done at 4 C. Cells which were incubated for additional several hours at 37 C after IF pretreatment at 4 C did not develop the primed state nor the antiviral state. The presence of protein synthesis inhibitors during the IF pretreatment depressed, though not completely, the development of the primed state. The residual priming effect was lost when the cells were incubated with the inhibitors at 37 C for 2 hr before they were exposed to poly I-poly C. There was no significant difference in the binding rate of poly I-poly C to cells between IF-treated and untreated cells. The degradation rate of cell-bound poly I-poly C and its sensitivity to exogenous pancreatic ribonuclease in the pretreated cells were also similar to those in the untreated cells.  相似文献

Induction of interferon (IF) and tumor necrosis factor (TNF) under the action of two polysaccharide preparations of ginseng i.e. panaxan-1 (from ginseng root) and panaxan-2 (from ginseng cell culture) was studied. Both the preparations induced production of TNF and IF in human leukocytes. By its properties and the typing results the induced IF proved to be gamma-IF. The preparation from the ginseng cell culture in the doses used had a higher IF inducing activity which could be explained by the difference in the polysaccharide composition of the preparations.  相似文献

The thymic extract TP-1 significantly enhanced the production of immune interferon (IF) by human peripheral blood mononuclear cells in Con A- or Raji- but not PHA-induced cultures. TP-1 effect was more pronounced under conditions of low IF production. The effect was evident before cell proliferation took place and without necessary concurrent effect of TP-1 on the blastogenic response. TP-1 had no detectable effect on nonactivated cultures. The enhancement of IF production by TP-1 to the degree demonstrated here necessitates consideration of its possible effects in every assay of thymic hormones that involves lectin or allogeneic stimulation of lymphocytes.  相似文献

Human fibroblast and Syrian hamster embryo cells were induced to synthesize interferon (IF) with rIn . rCn and rIn . rCn + DEAE-dextran, respectively. Following induction, these cells synthesized IF for only a short time before entering into a repressed state and shutting off the synthesis of IF. Homologous and heterologous whole cell translational systems were developed to investigate the molecular basis for the shut-off of IF synthesis. These systems allowedd for the introduction of exogenous hamster and human IF-mRNAs into intact normal and repressed hamster and human cells via an improved CaCl2 precipitation technique. Human IF-mRNA was translated in normal human and hamster cells and in repressed hamster cells but not in repressed human cells. In contrast, the hamster IF-mRNA was translated in normal human, normal hamster, and repressed human cells but not in repressed hamster cells. These results indicate that a species-specific mechanism inhibiting translation of IF-mRNA is directly responsible for the shut-off of IF synthesis in human fibroblasts and Syrian hamster embryo cells.  相似文献

L-MS cells, adapted to grow in suspension, were obtained by selection from a high interferon (IF)-producing line of mouse L cell monolayers. A large volume of L-MS cells (20 liters or more; 1–2 × 1010 cells) was readily grown in a spinner culture, retaining their ability to produce high yields of IF in serum-free medium following induction with Newcastle disease virus (NDV). The optimal condition for the production of IF in the suspension culture of L-MS cells was established. The system also proved itself to be susceptible to IF induction by polyinosinic-polycytidylic acid (Poly I · Poly C) and by NDV inactivated with ultraviolet light (NDV-UV). By employing the present system, large quantities of mouse IF of a high titer could be routinely prepared.  相似文献

Lymphotoxin (LT) production, interferon (IF) production, and DNA synthesis were investigated after mitogen stimulation and in the mixed lymphocyte culture (MLC) reaction using human tonsil lymphocytes. Both LT and IF were assayed in parallel and from the same lymphocyte supernatants. An analysis of the PHA, PWM, one-way and two-way MLC reactions showed that the amounts of LT and IF produced could not be correlated. Polyriboinosinic: polyribocytidylic acid (poly(I: C)) failed to induce either LT production or [3H]TdR incorporation but did induce IF production. Removal of glass-adherent cells (GAC) had no effect on mitogen induced LT production but their removal reduced LT production in MLC reactions. GAC were necessary for IF production and optimal [3H]TdR incorporation in both mitogen stimulated cultures and in MLC reactions. IF and LT activities were shown to be the result of different molecules by using a Sephadex G-75 column. These results indicate that mitogen stimulation differs from MLC reactions in the cell type or control mechanisms involved for LT production, and that in mitogen stimulated cultures all three of these in vitro phenomena are probably the results of either different cell types or of different cell to cell interactions.  相似文献

Incubation of murine spleen-cell cultures with lipopolysaccharide (LPS) induces interferon (IF) production. Maximal IF levels are obtained after incubation with 100 g/ml for 10 h. Two inbred mouse strains differing in their ability to generate LPS-induced IF in spleen-cell cultures were used: C3H/eB, which generates high levels of IF (about 60 units/ml), and C3H/HeJ, which fails to generate detectable quantities of IF. In a genetic analysis these strains were hybridized and IF production was determined in spleen-cell cultures from F1 and F2 generations, and from backcrosses of F1 hybrids to parent strains. The results indicate that, in parent strains, a single dominant autosomal gene is responsible for differences in IF production in spleen cultures. LPS-induced IF in spleen-cell cultures resists pH 2 for as long as 48 h, but is labile to heating at 56° C for 30 min. Both macrophages and lymphocytes must be present in cultures for generation of LPS-induced IF. By using mixed cultures of macrophages and lymphocytes from C3H/eB and C3H/HeJ mice, it was shown that macrophages have to interact directly with LPS to enable IF production in the cultures.  相似文献

The addition of macrophages to cultures of PPD-stimulated lymphocytes from tuberculin-sensitive individuals results in a greater than 3-fold increase in the production of interferon over that observed in cultures of lymphocytes or macrophages alone with PPD. Polymorphonuclear leukocytes (PMN) cannot substitute for macrophages in the augmentation of interferon production. In the combined lymphocyte-macrophage cultures a dissociation in time occurs between the time of peak transformation (as measured by the incorporation of thymidine-3H) and the time of peak interferon production; peak transformation occurs at 4–6 days and peak interferon production at 7–8 days. The amount of interferon produced is related to the degree of transformation. The significance and relevance to in vivo events of this in vitro macrophage-lymphocyte interaction in the production of interferon, a mediator of cellular immunity, is discussed.  相似文献

The production of interferon (IF) by human and mouse lymphocytes sensitized to alloantigens in mixed lymphocyte cultures (MLC) was analyzed. During primary MLC, IF appeared in the culture fluid on day 2 and was maximal on day 5. Based on several biologic criteria, the IF produced is of the "immune" type. When lymphocytes sensitized to alloantigens were reestimulated in vitro, IF was produced within a few hours of culture. In all stimulated cultures, cell proliferation was observed in spite of the high concentrations of IF. The IF-producing cells in human MLC were identified as T lymphocytes lacking the receptor for the Fc fragment of IgG molecules (Fc gamma R(-)). Human MLC supernatants containing immune type IF mediate the enhancement of natural killer (NK) cell activity and protect NK target cells from lysis.  相似文献

Addition of interferon (IF) inducers pyran copolymer, poly(I)-poly(C), an ether extract of Brucella abortus (Bru-Pel), or Salmonella typhimurium lipopolysaccharide (LPS) to cultures of peritoneal macrophages in vitro enhanced their cytotoxic activity for MBL-2 lymphoblastic leukemia cells. To evaluate the role of induced IF in the macrophage activation, highly specific rabbit anti-L-cell IF globulin was added to resting macrophage cultures at the same time as the macrophage-activating agents. Macrophage activation by these various biological and synthetic agents was totally neutralized by anti-IF globulin but not by normal rabbit globulin. Similarly, the anti-IF globulin inhibited the ability of chromatography-purified Newcastle disease virus-induced IF to render macrophages cytotoxic, and the degree of neutralization of IF titer corresponded with the inhibition of IF-induced macrophage-mediated cytotoxicity. In contrast, macrophage activation by concanavalin A-induced lymphokine, which contains an antigenically different IF, was not affected by high titers of the anti-L-cell IF antibodies. The results indicate that endogenously generated type I IF may play an important role in control of macrophage function.  相似文献

The effect of mouse serum interferon (IF) in vitro and an inducer in vivo on the proliferation of a pluripotent stem cell population with high turnover rate was studied. Proliferation rate was characterized by the number of CFUs in the S phase of the cell cycle. Increased proliferation of bone marrow stem cell populations was produced either by irradiating the donor mice with 3.36 Gy (336 rad) 60Co-gamma rays 7 days before the experiment or by incubating normal bone marrow cells with 10(-11) M concentration of isoproterenol. IF considerably reduced the number of CFUs in S phase in both cases without reducing the CFUs content of the samples. Injection of IF inducer (4 mg/kg poly I:C) into regenerating mice also inhibited the proliferation of CFUs without decreasing the femoral CFUs level. Regeneration kinetics of CFUs from irradiated poly I:C-treated mice ran parallel with that of irradiated untreated animals but showed a characteristic delay corresponding to approximately one CFUs doubling. A transient, non-cytotoxic proliferation inhibitory effect of IF or IF inducer is, therefore, proposed.  相似文献

Lungs and liver produced thermolabile virus-inhibiting factor (IF) or interferon at earlier stages.
