Coturnix coturnix japonica eggs were exposed to 2.45 GHz continuous wave microwave radiation at an incident power density of 5 mW/cm2 (SAR = 4 mW/g) during the first 12 days of embryogeny. After hatching, hematologic changes in response to an acute hemorrhage were measured in exposed and nonexposed (control) juveniles and adults of both sexes. Reticulocyte numbers and percentages were depressed below control numbers at 24 hr postphlebotomy in exposed adult females. Lymphocyte numbers were depressed below control levels at 24 hr postphlebotomy in exposed juvenile and adult males. At 72 hr heterophil numbers were depressed in exposed juvenile and adult males. These data suggest that microwave irradiation during embryogeny affects the ability of Japanese quail to recover from an acute and voluminous hemorrhage and that these radiation effects are small.

1. Hematological responses to hemorrhage by phlebotomy with and without replacement of blood volume with saline were measured in juvenile and adult male Coturnix coturnix japonica. 2. Recovery of total peripheral erythrocyte numbers and total peripheral leukocyte numbers occurred within 72 hr postphlebotomy in both treatment groups. 3. Saline replacement of blood volume following hemorrhage increased the total numbers and differential percentages of circulating reticulocytes at 72 hr postphlebotomy above the reticulocyte values of phlebotomized quail receiving no saline in both adult and juvenile Japanese quail.

Exposure of rats to simulated altitude (15,000 ft) for 1 day and 3 and 9 wk produced progressive polycythemia, elevated 2,3-diphosphoglycerate levels and raised P50 values; the latter two parameters decreased toward control values after 9 wk. Carbon monoxide (38-43% HbCO) exposure produced polycythemia after 3- and 9-wk exposure, no change in 2,3-DPG and a fall in P50 value. Ten days' treatment with sodium cyanate produced a large decrease in 2,3-DPG and P50. Survival during 90 min of acute hypobaria (0.3 atm) under Nembutal anesthesia was highest with NaOCN (75%), intermediate with 3- and 9-wk exposure to altitude and CO (56-58%) lower in 1-day altitude exposure (44%) and lowest in controls (5%). Heart and ventilation rate was monitored during this hypobaric test and response patterns established for each exposure/treatment. In states of extreme oxygen deprivation the results suggest, in order of importance, the survival value of 1) increased oxygen-hemoglobin affinity, and 2) polycythemia.

Water deprivation (WD) resulted in increased serum osmotic pressure (OP) and decreased body weight (WB); adrenal aldosterone content did not change. Adrenal corticosterone content tended to be elevated during early WD, indicating a stress response, but tended to decrease after seven days of WD, suggesting adrenal fatigue. During water restriction (WR), after the period of weight loss, adrenal corticosterone content and serum OP were elevated. As the birds began to gain weight, aldosterone levels did not change but adrenal corticosterone content and serum OP approached control values, suggesting that the birds were beginning to adapt to the WR. Adrenal sensitivity to ACTH was indicated by the elevated adrenal aldosterone and corticosterone content after ACTH injection.

The conditioning effect of cyclic photoperiods was studied in 9 experiments. Quail after 1 day of age were exposed to repeated cycles of 6 hr of light and 18 hr of darkness during their first 4 weeks.Afterwards the groups were established to study the effect of (a) ahemeral (unnatural daylength); (b) phase shifted, and (c) abbreviated photoperiods. The quail were kept on such regimens for 3 weeks. The results based on the 7-week testicular weights indicated that quail did not respond to a phase shift in the light period but responded to an abbreviation of a previously established photoperiod.Quail responded to unnatural daylength even when the actual and relative length of the dark period increased.

1. Juvenile Coturnix coturnix japonica males were injected intravenously with 2, 20 or 200 mg ethyl methanesulfonate (EMS)/kg body wt; 0.2, 2 or 20 mg paraquat (PARA)/kg body wt; or 0.6, 6 or 60 mg phenylhydrazine (PHZ)/kg body wt; and hematologic variables were measured at 0 (non-injected), 24 and 72 hr post-injection. 2. EMS, PARA and PHZ-induced hemolytic anemia began within 24 hr post-injection. 3. Recovery from anemia began within 72 hr post-injection of EMS or PARA, but PHZ injected quail continued to show a marked anemia at that time. 4. EMS and PARA induced lymphocytopenia, monocytopenia and heterophilia, while PHZ induced lymphocytosis, monocytopenia and heteropenia after injection. 5. These results suggest that the anemia induced by EMS and PARA was dissimilar from that induced by PHZ, that all chemicals affected leukopoiesis and that Japanese quail can mount a marked recovery from the hematologic affects of PARA, a widely used herbicide, in a short interval after intoxication.

Experiments were conducted to determine the effect of injection of lipopolysaccharide (LPS, from S. typhimurium) or muramyl dipeptide (MDP, N-acetylmuramyl-L-ala-isoglutamine) in Japanese quail. Doses of MDP between 0.3 and 10 mg/kg body wt. had no effect on body temperature. In contrast, doses of 1.0-22.5 mg LPS/kg body wt. caused significant increases in body temperature. None of the doses of LPS or MDP resulted in mortality. The febrile response to LPS was diminished following a second injection 48 h after the first, and was absent following a third injection. Plasma zinc, an indicator of the acute phase response, was significantly reduced by either LPS or MDP after the first injection (P<0.001), but not after the second or third injection. Splenic interleukin 1-beta (IL-1beta) mRNA expression was increased after the first and last injection of LPS (P<0.001), but only after the first injection of MDP (P<0.005). Hepatic IL-1beta mRNA expression was increased after the first, but not the third injection of LPS (P<0.001), while MDP had no effect. These data indicate that Japanese quail are less sensitive to MDP than LPS, and that quail demonstrate tolerance to LPS following repeated injections.

Physiological manifestations of social stress in stable and unstable (resident and visitor) pairs of adult male Japanese quail (Coturnix coturnix japonica) were compared after 7, 14, 21 and 28 days of regrouping. Unstable pairs had reduced body and relative testes (mg/100 g) weights. Plasma cholesterol was increased significantly in unstable pairs, and adrenal cholesterol was reduced significantly by daily regrouping of unstable pairs. Twelve consecutive days of ACTH treatment (2, 4 or 8 IU/100 g body wt given intramuscularly) to birds in stable pairs induced a decrease in body and relative testes weights. The results were similar to those found in birds subjected to daily regrouping.

Summary This study was undertaken to examine the effect of electrical stimulation of the hypothalamus at different times of day on luteinizing hormone (LH) secretion in male castrated quail on short days (8L16D). The posterior hypothalamus was stimulated with square-wave pulses of 80 A for 2 min through chronically-implanted platinum microelectrodes. Stimulation was carried out on each quail at 4 (treatment A), 10 (B), or 14 h (C) after dawn. Plasma LH levels were increased markedly within 2 min of ending the stimulation but reached basal levels again over the next 20 min or so. The absolute increase was significantly greater in treatment B (10 h after lights on) than at the other times tested. This is consistent with a rhythm in hypothalamic responsivity. The results are discussed in the context of the rhythm of photoinducibility which occurs early in the night and which is used by quail as a photoperiodic clock to regulate seasonal reproduction.Abbreviations LH luteinizing hormone - LH-RH luteinizing hormone-releasing hormone  相似文献

Behavioral and adrenocortical reactivity to stressful stimulation was examined in 12- and 13-day-old chicks of two lines of Japanese quail selected over several generations for exaggerated (HS: high stress) or reduced (LS: low stress) plasma corticosterone (B) response to brief immobilization stress. Plasma B concentrations and tonic immobility (TI) fear reactions were measured in unstressed (control) and stressed (overnight cooping) chicks of both lines. The stress treatment was applied over a period of 12-20 hr and it involved capture by the experimenter, inescapable exposure to an unfamiliar environment and to strange conspecifics, reductions in ambient temperature and floor space, and the deprivation of food and water. Chicks of the HS line were more susceptible to the induction of TI and they remained immobile longer than did LS chicks. Therefore HS chicks were considered to be more fearful than their LS counterparts. Stress treatment elicited a marked adrenocortical response that was more pronounced in HS than in LS chicks. Stress treatment also increased susceptibility to TI but did not significantly affect the duration of immobility. These findings suggest that selecting the quail for differential corticosterone response to a particular stressor had exerted an unconscious and concomitant effect on underlying fearfulness as well as on their adrenocortical reactivity to several other stressful situations. The results are further discussed in terms of a putative relationship between adrenocortical activation and fearfulness.

Heat stress is one of the major causes of a decreased performance of laying quail in tropical and subtropical countries. The aim of this study was to investigate the impact of temperature humidity index (THI) on fertility aspects, external and internal egg quality parameters in Japanese quail. One hundred and forty four (144) Japanese quail, 12 of weeks age, were used. Birds were divided randomly into three equal groups, control (at low THI, lower than 70), H1 (at moderate THI, 70–75) and H2 (at high THI, 76–80). Quail in the control and H1 groups had significant greater fertility (p=0.021) and hatchability % (p=0.037), compared with H2 group. Quail in the control group (at low THI) laid heavier egg weight with a higher external (egg weight (p=0.03), shell thickness, shell weight, eggshell ratio and eggshell density (p=0.001)) and internal egg quality score (albumin weight (p=0.026), yolk height (p=0.003), yolk index (p=0.039) and Haugh unit (p=0.001)). Otherwise, such quality traits were compromised in heat-stressed quail. At the high THI level, egg weight had a significant positive correlation with albumin weight (r=0.58, p<0.01), yolk weight (r=0.22, p<0.05), albumen ratio (r=0.17, p<0.05), yolk height (r=0.14, p<0.05) and yolk index (r=0.18, p<0.05), but was negatively correlated with yolk ratio (r=−0.15, p<0.05). Japanese quail exposed to heat stress (THI over 75) revealed drop in fertility indices and egg quality traits, indicating a detrimental policy of economic income.

Summary Japanese quail,Coturnix coturnix japonica, which were acclimated to a simulated altitude of 6100 m for six weeks were compared with control quail maintained at sea level. Body weight initially decreased in both groups, and by the end of six weeks the altitude quail weighed an average of 8% less than the control quail (Fig. 1). Compared with the controls, the altitude birds exhibited mean increases in hematocrit ratio and blood hemoglobin concentration of 31 and 37% respectively (Fig. 2). Blood volume was 36% higher in the altitude acclimated quail, while plasma volume was unchanged (Fig. 3). Exposure to chronic hypoxia resulted in hypertrophy of the right ventricle, while left ventricular mass was unchanged (Fig. 4). Rates of O2 consumption (VO2) at 5°C were continuously recorded as ambient PO2 was slowly reduced from 155 to 30 torr. VO2 was significantly higher at any given PO2 in the altitude quail (Fig. 5). Exposure to reduced PO2 resulted in a significantly greater decrease in body temperature in the control quail than in the altitude acclimated birds (Fig. 6).

1. Femurs taken from Japanese quail (Coturnix coturnix japonica) genetically selected for either exaggerated (HS; high-stress) or reduced (LS; low-stress) blood corticosterone (B) response to unfamiliar environments were examined to determine their cortical bone porosity (CBP; degree of osteoporosis) and medullary bone (MB) status following treatment with or without stress. 2. Femur CBP was elevated in both sexes of stressed, HS-line quail and was attributed directly to the presumably exaggerated blood B responses known to occur in this line as a consequence of the stress treatments used. 3. The increase in CBP in stressed, HS-line birds occurred independently of changes in the numbers of osteoclast cells (CLASTS) associated with cortical bone. 4. Stress treatment was associated with lower CLAST numbers/mm2 of MB and with increased MB accretion. 5. These stress-induced effects were considered to be the consequence of a reduced rate of lay found in stressed quail hens. 6. There was only marginal evidence for a direct effect of B in stimulating MB resorption.

Starvation effects for five weeks on energy reserves, oxidative stress and hematological indices in Nile catfish Clarias gariepinus was studied. The low protein level in starved fish may result from the lowering effect of prolonged starvation on protein synthesis rather than due to its degenerating protein. Moreover, the elevated level of serum amino acids may promote gluconeogenesis in liver. In addition, the lipid depletion in starved fish may be related to the preferential uses of lipids as an energy to starve fish. Also, unchanged glycemic level may introduce a potent evidence for the presence of active gluconeogenesis, depending on both amino and fatty acids precursors. Also, kidney and liver showed disturbances in metabolites associated with oxidative damage such as elevations in total peroxide, carbonyl protein and DNA fragmentation; these may cause dysfunction to these organs after five weeks of starvation. Total peroxide, carbonyl protein and DNA fragmentation were significantly increased in gills, liver and kidney by 29.9, 30.9 and 30.5; 83.6, 84.6 and 53.7; 82.4, 43.3 and 75.7%, respectively. Starvation induced severe anemia and loss of body weight in the fish. However, white muscle did not show any oxidative damage after five weeks of starvation.

Animals perceiving repeated aversive events can become chronically stressed. Chronic activation of the hypothalamic–pituitary–adrenal (HPA) axis can have deleterious consequences on physiological parameters (e.g. BW, blood chemistry) and behaviour (e.g. emotional reactivity, stereotypies, cognition). Environmental enrichment (EE) can be a mean to reduce animal stress and to improve welfare. The aim of this study was first, to assess the effects of EE in battery cages on the behaviour of young Japanese quail and second, to evaluate the impact of EE on quail exposed to chronic stress. The experiment involved quail housed in EE cages and submitted or not to a chronic stress procedure (CSP) (EE cages, control quail: n=16, CSP quail: n=14) and quail housed in standard cages and exposed or not to the CSP (standard non-EE cages, control quail: n=12, CSP quail: n=16). Our procedure consisted of repeated aversive events (e.g. ventilators, delaying access to food, physical restraint, noise) presented two to five times per 24 h, randomly, for 15 days. During CSP, EE improved quail’s welfare as their stereotypic pacing decreased and they rested more. CSP decreased exploration in all quail. After the end of CSP, quail presented increased emotional reactivity in emergence test. However, the effect of EE varied with test. Finally, chronic stress effects on comfort behaviours in the emergence test were alleviated by EE. These results indicate that EE can alleviate some aspects of behavioural alterations induced by CSP.

Common responses to hypoxia include decreased body temperature (Tb) and decreased energy metabolism. In this study, the effects of hypoxia and hypercapnia on Tb and metabolic oxygen consumption (V.O2) were investigated in Japanese quail (Coturnix japonica). When exposed to hypoxia (15, 13, 11 and 9% O2), Tb decreased only at 11% and 9% O2 compared to normoxia; quail were better able to maintain Tb during acute hypoxia after a one-week acclimation to 10% O2. V.O2 also decreased during hypoxia, but at 9% O2 this was partially offset by increased anaerobic metabolism. Tb and V.O2 responses to 9% O2 were exaggerated at lower ambient temperature (Ta), reflecting a decreased lower critical temperature during hypoxia. Conversely, hypoxia had little effect on Tb or V.O2 at higher Ta (36 °C). We conclude that Japanese quail respond to hypoxia in much the same way as mammals, by reducing both Tb and V.O2. No relationship was found between the magnitudes of decreases in Tb and V.O2 during 9% O2, however. Since metabolism is the source of heat generation, this suggests that Japanese quail increase thermolysis to reduce Tb. During hypercapnia (3, 6 and 9% CO2), Tb was reduced only at 9% CO2 while V.O2 was unchanged.
