This work describes the isolation of seven (17) secondary metabolites from Espeletia barclayana (Asteraceae, Espeletiinae) and their identification by spectroscopic (NMR) and spectrometric (MS) techniques. Ten (817) additional compounds were identified based on their retention times, high-resolution mass spectrometry data, and comparison with reference substances or data from literature. The systematic significance of some of the identified substances – the sesquiterpene lactone longipilin acetate, four caffeoylquinic acids and two tri-caffeoylaltraric acids – is discussed with the aim of providing insights into the complex relationships among Espeletiinae taxa and its closest relatives. Five of the isolated metabolites [5-O-(E)-caffeoylquinic acid (1), 1,3-di-O-(E)-caffeoylquinic acid (2), 1,5-di-O-(E)-caffeoylquinic acid (3), 3,4-di-O-(E)-caffeoylquinic acid (4) and 3-O-methylquercetin 7-O-β-glucopyranoside (7)] constitute new reports for the genus Espeletia and for the subtribe Espeletiinae. Chemical data suggest that Espeletiinae might have a closer relationship with Smallanthus than with Ichthyothere, i.e., the two genera suggested to be the sister groups of Espeletiinae based on molecular markers.  相似文献

Apomictic complexes in flowering plants often harbor multiple sexual taxa, which must be studied to interpret accurately the origin and diversity of apomictic derivatives. The daisy fleabane group in North America (Erigeron sect. Phalacroloma) includes the widespread polyploid apomictic taxa E. annuus, E. strigosus, and E. tenuis, as well as recently discovered sexual diploid populations in the southeastern United States (E. strigosus var. calcicola, E. strigosus var. dolomiticola, sexual E. strigosus var. strigosus). Phylogenetic analysis for 35 sexual populations was conducted using sequences from the internal and external transcribed spacers (ITS and ETS) of ribosomal DNA. Results indicate that the three groups of sexual plants form separate monophyletic clades and that edaphic specialization is ancestral in the group. Sequence analysis for sexual E. strigosus var. strigosus was confounded by the fact that 13 of 31 samples (42%) possessed nucleotide polymorphisms at 1.0-1.8% of sites. However, analysis of the genomic sequence for plants with low polymorphism levels in conjunction with analysis of cloned PCR products for plants with high levels of intra-individual polymorphism indicated that three combinations of haplotypes (N I + N II, two plants; N I + N III, five plants; N II + N III, six plants) accounted for the majority of sequence polymorphisms encountered. These data are consistent with historical diversification and subsequent reticulate evolution at the diploid level in sexual E. strigosus var. strigosus.  相似文献

The present study considers genetic diversity of 38 populations in 4 Cirsium species of the genus Cirsium Mill. (Asteraceae), occurring in different ecological regions and tries to compare degree of genetic variability among the species with wide geographical distribution versus endemic C. pyramidale showing confined geographical distribution. The results showed that the endemic species has similar value of genetic diversity parameters as the species with wider distribution. We also studied the possible admixture nature of these populations and tried to understand the relation between genetic changes, geographical distribution and polyploidy level and chromosome pairing in these species. ISSR analysis showed population difference in allele composition and frequency. Clustering and PcoA ordination produced different groupings in each species, while STRUCTURE and reticulation analyses revealed high degree of genetic admixture and gene exchange among populations as well as allelic rearrangement. No significant correlation was observed between geographical distance and genetic distance of the populations and AMOVA test revealed no significant difference among populations in each species studied. However, high amount of within population variation occurred in all 4 species indicating their cross-pollination nature and high genetic admixture. The populations also varied in chiasma frequency and chromosome pairing as well as the occurrence of heterozygote translocations all creating more variability to be used by plants for local adaptation.  相似文献

Random Amplified Polymorphic DNA (RAPD) markers were used to measure genetic diversity within and divergence among species of Dendroseris (Asteraceae: Lactuceae), a genus endemic to the Juan Fernandez Islands, Chile. Results were compared to previous studies employing allozymes. For five of the species, RAPD banding patterns distinguished all individuals examined, and different mutilocus genotypes were found even in species exhibiting no allozyme diversity. RAPD band diversities ranged from 0.003 to 0.022 within species; >90% of total diversity was among species and <10% within them. Relative levels of allozyme and RAPD diversity were similar for some species, particularly those with highest and lowest diversities, but overall there was no significant correlation. Relationships inferred from a neighbor-joining tree generated from RAPD bands were similar to results obtained from morphology, chloroplast DNA (cpDNA) restriction site mutations, and sequences from the internal transcribed spacer regions of nuclear ribosomal DNA (ITS), but somewhat better resolution was achieved. Relationships shown by allozymes differed from trees based on other data; this ostensibly is a result of the sharing of ancestral alleles and the absence of alleles generated subsequent to speciation. Dendroseris represents an example where RAPD markers, because of their greater variability, provide a useful alternative to allozymes for assessing diversity in rare species endemic to oceanic islands and for resolving relationships among the species.  相似文献

From March 2001 to December 2002, we studied the reproductive phenology, pollination ecology, and growth rates of Espeletia grandiflora Humb. and Bonpl. (Asteraceae), a giant caulescent rosette from the Páramos of the Eastern Andes of Colombia. Espeletia grandiflora was found to be predominantly allogamous and strongly self-incompatible. Bumblebees (Bombus rubicundus and B. funebris) were the major pollinators of E. grandiflora, although moths, hummingbirds, flies, and beetles also visited flowers. Inflorescence development began in March and continued through August to September. Plants flowered for 30 - 96 days with a peak from the beginning of October through November. The percentage of flowering plants strongly differed among size classes and between both years. Seed dispersal occurred as early as September through May of the following year. The average absolute growth rate for juveniles and adults rate was 7.6 cm/year. Given the scarcity of floral visitors at high altitudes due to climatic conditions, we suggest that even small contributions from a wide range of pollinators might be advantageous for pollination of E. grandiflora. Long-term studies on different populations of E. grandiflora are required to determine if the high growth rates are representative, to quantify the variation in the flowering behavior within and among populations, and to establish if nocturnal pollination is a trait that is exclusive to our population of E. grandiflora.  相似文献

Weeds with broad distributions and large morphological variation are challenging for systematists and evolutionarily intriguing because their intensive dispersal would likely prevent local morphological differentiation. Study on weeds will help to understand divergence in plants unlikely to be affected by geographical and ecological barriers. We studied Youngia japonica based on nrDNA and cpDNA sequences. This is a widespread native in Asia and invasive worldwide; nevertheless, three subspecies (japonica, longiflora, and formosana) and an undescribed variant occur in Taiwan. Bayesian and the most parsimonious phylogenies revealed that subspecies longiflora is a different linage and independently arrived in Taiwan during the Pleistocene via land connection to the Asian Continent. Bayesian time estimation suggested that Youngia in Taiwan diverged in the lower Pleistocene or more recently. Extreme habitats that emerged in the Pleistocene, i.e., cold mountain ranges for subspecies formosana and xeric, raised coral reefs for the undescribed Youngia variant probably had triggered the divergence. Components of Youngia in Taiwan are not monophyletic; a coalescent-based test suggested incomplete lineage sorting. Nevertheless, the samples within each taxon share unique morphological features suggesting a common gene pool and each taxon has different dominant ITS and/or cpDNA types; these conditions suggest ongoing process toward monophyly via coalescent processes and support the delimitation of intraspecific taxa.  相似文献

Molecular studies of apomictic plant species often detect more genotypic variation than predicted from their assumed reproductive mode. The two most commonly invoked mechanisms to explain these high levels of variation are recombination, via facultative sexuality, and mutation. The potential for sexual reproduction in the facultative apomict Hieracium pilosella (Asteraceae) was determined at three field sites by artificially pollinating with the closely related, but morphologically distinct, H. aurantiacum. The level of genotypic variation at the three sites was recorded using inter-simple sequence repeats (ISSRs). There was a significant, positive relationship between the measured potential for sexual reproduction and population genotypic variability, indicating that sex has played a role in the structuring of these populations; however, a causal relationship cannot be stated because of the use of regression. We also applied the recently developed method of compatibility analysis. Compatibility analysis can determine, using the occurrence of "character incompatibilities," whether patterns of variation observed in populations are most parsimoniously explained by mutation or recombination. Compatibility analysis also indicated that sexual reproduction had played a role in generating genotypic diversity in these populations. Combining these different types of data may give a greater understanding of the potential for the generation of genotypic diversity in facultative apomictic populations.  相似文献

 Assessing the relative contributions to seed set for each of a plant species’ floral visitors provides an indication of the relative influence of these visitors on the plant’s reproductive success. This study examined pollinator activity and seed set in a population of Heterotheca subaxillaris, a species that exhibits a floret dimorphism (heads bearing disk and ray florets), and that is visited by both generalist foragers and specialist bees. Visits by nine bee genera and one genus of skipper were recorded in the study population. During the period of study, these insects varied in their relative abundance, in their foraging activity on a head, and in their pollination effectiveness. The pattern of pollination effectiveness shown by the different pollinators was similar for both floret types, although seed set was higher overall for ray florets. Pollinator importance, calculated as the product of pollination effectiveness and relative abundance, was dictated by a pollinator’s relative abundance. The single specialist bee species observed in the study population proved to have neither higher pollination effectiveness nor higher pollinator abundance than other pollinators. This would suggest that H. subaxillaris is not under strong selective pressure to co-specialize with its specialist visitor. Received: 21 December 1995 / Accepted: 20 June 1996  相似文献

Ecological speciation, driven by adaptation to contrasting environments, provides an attractive opportunity to study the formation of distinct species, and the role of selection and genomic divergence in this process. Here, we focus on a particularly clear‐cut case of ecological speciation to reveal the genomic bases of reproductive isolation and morphological differences between closely related Senecio species, whose recent divergence within the last ~200 000 years was likely driven by the uplift of Mt. Etna (Sicily). These species form a hybrid zone, yet remain morphologically and ecologically distinct, despite active gene exchange. Here, we report a high‐density genetic map of the Senecio genome and map hybrid breakdown to one large and several small quantitative trait loci (QTL). Loci under diversifying selection cluster in three 5 cM regions which are characterized by a significant increase in relative (FST), but not absolute (dXY), interspecific differentiation. They also correspond to some of the regions of greatest marker density, possibly corresponding to ‘cold‐spots’ of recombination, such as centromeres or chromosomal inversions. Morphological QTL for leaf and floral traits overlap these clusters. We also detected three genomic regions with significant transmission ratio distortion (TRD), possibly indicating accumulation of intrinsic genetic incompatibilities between these recently diverged species. One of the TRD regions overlapped with a cluster of high species differentiation, and another overlaps the large QTL for hybrid breakdown, indicating that divergence of these species may have occurred due to a complex interplay of ecological divergence and accumulation of intrinsic genetic incompatibilities.  相似文献

The subtribe Espeletiinae (Asteraceae, Heliantheae) comprises morphologically and ecologically diverse plants endemic to the tropical montane paramos of the Andes of Venezuela, Colombia, and Ecuador. Though the ecophysiology and ecology of this adaptive radiation have been well studied, relationships among taxa in the subtribe and between the subtribe and other taxa in the Heliantheae are poorly known. In this study, sequences from the internal transcribed spacer (ITS) region of nuclear ribosomal DNA are used to test previous hypotheses about the phylogenetic position of the Espeletiinae within the Heliantheae and to determine which taxa are the subtribe's closest relatives. Gene phylogenies based on maximum parsimony analyses reveal that the Espeletiinae clade is nested well within the subtribe Melampodiinae and thus should be considered a monophyletic complex of species, not a separate subtribe. The most parsimonious gene trees suggest that the genus Ichthyothere may be the sister taxon to the Espeletia complex and that the genus Smallanthus and a species of Rumfordia are likely among the complex's other closest living relatives. These data offer preliminary insights into the origins of this adaptive radiation and the broader phylogenetic context in which it occurred.  相似文献

Despite long‐standing interest in the evolutionary role of plant hormones, relatively few studies have used hormonally manipulated phenotypes to address questions about phenotypic evolution in plants. In the present investigation, I subjected plants of Crepis tectorum subsp. pumila to simple gibberellin (GA) treatments under both field and greenhouse conditions to assess developmental lability and correlational constraints of phenotypic traits that distinguish this dwarf ecotype from conspecific populations of the much taller weed ecotype (subsp. tectorum). The hormonally manipulated plants largely phenocopied the weed ecotype in leaf shape, plant stature and branching habit, indicative of both high lability and tight integration of traits reflecting gross morphology. Floral size traits sometimes declined after GA application, especially under field conditions. The latter result conflicts with the positive correlations between floral and vegetative size traits seen in previous comparative analyses and point to a tradeoff that could act as a constraint on ecotype divergence. The response to GA was consistent in direction for most traits, as opposed to the magnitude of response, which varied depending on the trait, the population, the growing environment and the timing and level of hormone application. Taken together, the results highlight the potential for simple hormonal changes to cause large, plastic shifts in phenotype, but also illustrate the constrained nature of such influences.  相似文献

Determining the genetic structure of an in situ conserved population can provide insight into the dynamics of population genetic processes associated with successful plant conservation. We used 21 microsatellite loci to analyse the genetic relationships among individuals (n = 813) collected from a small Oryza rufipogon population conserved since 1993 in Hunan Province of China. The analysis revealed four distinct genetic subpopulations (F(ST) = 0.145) without geographic isolation. One subpopulation was composed of possible introgressed individuals, two subpopulations were composed of seed recruits and their descendants, and the fourth subpopulation consisted of reintroduced individuals, seed recruits and their descendants. Positive spatial genetic structures were detected by spatial autocorrelation statistics at the population (c. 63 m) and subpopulation levels (11-30 m), but the degree of autocorrelation was stronger at the population level. These results showed that prejudging the cryptic structure is important before autocorrelation analysis for the entire population. Our study suggests that population history can be a significant determinant on population structure for plant restoration projects.  相似文献

The russet mite, Aceria anthocoptes (Nalepa), is the only eriophyoid that has been recorded on Cirsium arvense (L.) Scop. It has been noted in several European countries and recently in the USA. In this study we explored the geographic and host-related variability of Aceria spp. inhabiting different Cirsium spp. We applied landmark-based geometric morphometric methods to study morphological variability of three body regions (ventral, coxigenital and prodorsal) of 13 Aceria spp. populations inhabiting five Cirsium spp. in Serbia (Europe) and four Cirsium spp. in Colorado (North America). Analyses of size and shape variation revealed statistically significant differences between Aceria spp. living on European native and North American native Cirsium spp., as well as between A. anthocoptes s.s. inhabiting European C. arvense and North American C. arvense. The coxigenital region was the most informative when considering inter-population shape differences. European Aceria spp. dwelling on Cirsium spp., including A. anthocoptes s.s. from C. arvense, are characterized by higher inter-population size and shape variability than their North American counterparts. This finding supports a Eurasian origin of A. anthocoptes, presumed to consist of a complex of cryptic taxa probably coevolved with host plants in the native environment. Morphological similarity among Aceria spp. inhabiting North American native Cirsium spp. may indicate that speciation of A. anthocoptes started relatively soon after the host shift to plants different from C. arvense in the invaded region.  相似文献

A multivariate morphometric study based on 22 characters was carried out on samples of the 26 accepted species of the Eurasian and north-African genusDoronicum, to assess whether quantitative, mainly continuous, characters were of use in suggesting, delimiting and structuring natural groups. The study concentrated on three-medium sized to small groups: Mediterranean (D. plantagineum group), European (D. grandiflorum group), and central Asian. Two questions were addressed for each of these groups: species delimitation within them and the inclusion/exclusion of species of uncertain, but putative close affinities. Principal components analysis and discriminant analysis (canonical variates analysis), gave moderately satisfactory results. The addition of doubtfully-related species to presumably natural groups increased cohesiveness of the groups because the between-species differences were minimized when placed in a broader more diverse context. The relative status of the uncertain species, either as outliers or true members, was appropriately depicted by the scatterplots, but more convincingly solved when the species turned out to be outliers. Discrimination between species, which belong to cohesive groups was effective in some cases at least when using discriminant analysis, and the exceptions involved particularly problematic taxa. It was concluded that morphometric characters are a useful complementary source of information in groups where qualitative (in particular, shared) characters are scarce as is the case inDoronicum.  相似文献

By influencing the proliferation of different genotypes, clonal growth can affect the maintenance of genetic variability and magnitude of genetic drift within plant populations. However, estimates of effective population size rarely incorporate the contribution of both asexual and sexual reproduction. We estimated effective size (Ne) for two populations of the clonal, self-incompatible plant, Hymenoxys herbacea, using a stage-structured demographic model for organisms with asexual and sexual recruitment and then examined the impact of reproductive strategy using an elasticity analysis. Plant rosettes monitored in two successive years had high survival rates in both populations (mean 0.94). The mean number of sexually derived recruits per initial ramet was 0.041 (SE 0.039), whereas the mean number of clonal recruits was 0.61 (SE 0.90). Effective size was 1642 and 5769 in the two populations and the Ne/N ratio averaged 0.34, comparable to values for other clonal species. Elasticity analysis indicated that increases in both clonal and sexual recruitment cause an increase in Ne while increasing the variance reduced Ne. However, Ne was more sensitive to changes in the mean and variance of asexual recruitment than sexual recruitment. These results highlight the importance of considering asexual modes of reproduction when examining the role of genetic stochasticity in populations.  相似文献

Solidago altissima and S. gigantea were introduced from North America to Europe ~250 yr ago and are now considered aggressive weeds in abandoned fields and conservation areas. We studied patterns of genetic differentiation in these two species along their present latitudinal range in Europe (44-61 degrees N). Two generations of clonally propagated ramets from randomly selected genets of 24 populations of each species were grown in a common-garden experiment at latitude 47 degrees N from 1991 to 1992. Both species showed significant variation among populations in morphological and life-history characters: at this southern location, plants from northern populations were smaller and flowered earlier than plants from southern populations. The gradient of clinal variation was more pronounced in the second year of cultivation than in the first and was steeper in S. altissima than in S. gigantea. Within-population variation among genotypes was significant tot most characters in the case of S. altissima. Phenological rate (reciprocal of days to flowering) and size at maturity showed a significant negative correlation among populations bot not among genotypes within populations, indicating that genetic trade-offs may occur at one but not another infraspecific level. We suggest that the pattern of among-population variation reflects rapid adaptive population differentiation after introduction of the species to Europe.  相似文献

Cytogenetic investigations of 13 natural populations of Coptis teeta , an endemic and endangered medicinal plant from the Eastern Himalaya, were carried out. About 70–80% individuals in some of the populations were synaptic mutants. The predominantly sexually breeding plant populations were confined to higher cooler latitudes, whereas the mutant apomict inhabited lower warmer latitudes. The breeding systems were observed to be associated with morphological differentiation and ecological preference of some populations. Individuals of subsp. teeta reproduced sexually as well as vegetatively, but those of subsp. lohitensis have adapted almost entirely to a vegetative mode of reproduction. Though no variation was observed in somatic chromosome number and karyotypes, the two subspecies differed significantly with respect to their male meiotic behaviour and rhizome morphology. The mutants either produced little fertile pollen or were completely sterile, leading to low seed production or none at all. This has resulted in low reproductive fitness, localized distribution and small population sizes of the taxon.  相似文献

Quantitative genetic variation was assessed in a population of Crepis tectorum subsp. pumila , a winter annual confined to calcareous grassland on the Baltic island of öland (SE Sweden). Plants from 40 maternal sibships were grown in a greenhouse and scored for a large number of traits representing all stages of the life cycle. The study included several characters that have been subject to ecotypic differentiation as well as traits known to be under current selection. Highly significant family differences were found for all but one character, suggesting that past selection was too weak to eliminate the genetic variability of characters presumed to be adaptive and there is a potential for further adaptive change in most traits. Two additional traits treated as qualitative were also found to differ significantly among families. A parallel cultivation experiment showed that the extent of population divergence in a quantitative trait was positively correlated with the amount of inter- family variation, implying stability of the relative variability for substantial periods of time, a possible reflection of phenotypic constraints being expressed both within and between populations. Additional data indicated that genetic trade-offs among traits under simultaneous selection, year-to- year variation in selection in combination with a long-lived seed bank, and genotype × environment interactions, could prevent the erosion of genetic variability in some characters connected with fitness.  相似文献
