From callus cultures of Cinchona ledgeriana seven known anthraquinones, purpurin, anthragallol-1,2-dimethylether, anthragallol-1,3-dimethylether, rubiadin, 1-hydroxy-2-hydroxymethylanthraquinone, 1-hydroxy-2-methylanthraquinone and morindone-5-methylether (or 1,7-dihydroxy-8-methoxy-2-methylanthraquinone), and eight new anthraquinones, 5,6-dimethoxy-1-(or -4-)hydroxy-2-(or -3-)hydroxymethylanthraquinone, 5-methoxy-2-(or -3-)methyl-1,4,6-trihydroxyanthraquinone, 2-hydroxy-1,3,4-trimethoxyanthraquinone, 4-methoxy-1,3,5-trihydroxyanthraquinone, 1,4-dimethoxy-2,3-methylenedioxyanthraquinone, 1,3-dihydroxy-4-methoxyanthraquinone, 1,3-dihydroxy-2,5-dimethoxyanthraquinone and 2,5-(or 3,5-)dihydroxy-1,3,4-(or -1,2,4-)trimethoxyanthraquinone have been isolated.  相似文献

The addition of autoclaved mycelia of Aspergillus niger and the known phytopathogenic fungus Phytophtora cinnamomi to cultured cells of Cinchona ledgeriana Moens. caused a marked increase in the anthraquinone content of the plant cells. The growth and alkaloid production of a liquid suspension culture of Cinchona pubescens has been studied, particularly with attention to the effect on the alkaloid spectrum of feeding cultures with L-tryptophan. This treatment did not enhance the production of any of the known alkaloids of Cinchona. Above 2mmM, however, the presence of the amino acid was toxic, causing extreme acidification of the medium and cell death. Under these conditions a number of indole and quinoline derivatives accumulated. The principal component of the alkaloid fraction proved to be norharman; indole-3-aldehyde was also isolated. Both these products probably occur by uncharacteristic metabolism of L-tryptophan. Furthermore, evidence for the degradation of endogenous alkaloids was obtained, as 4-hydroxymethylquinoline was also isolated. None of the known quinoline alkaloids of Cinchona, which were present in untreated cells, could be detected after L-tryptophan treatment, even when large amounts of culture were analysed. Rheum ribes L. (Polygonaceae) is the source of one of the most important crude drugs in Asiatic regions .The medicinal character of rhubarb is due to its anthraquinone content . Different parts of sterile seedlings were cultured on MS medium to study the generation of callus. The explants were cultured with different ranges of plant growth regulators and the best range of plant growth regulators for generation of callus was IBA (1 mg l–1) and BA (1 mg l–1). The content of anthraquinones were determined by HPLC . The concentration of sucrose, vitamins and Myo-inositol and ratio of NO3 to NH4 in the medium was changed and growth rate and content of 2 anthraquinones was determined . The growth rate of callus declined with increased rate of secondary metabolites production. Myo-inositol 100 mg l–1 in the medium increased anthraquinone content and in medium that had NO3/NH4:1/1 the maximum content of anthraquinone was obtained.  相似文献

The uptake of l-[methylene-14C]-tryptophan from culture medium into root organs of Cinchona ledgeriana and the subsequent incorporation of the radiolabel into quinine and quinidine is reported. In addition, feeding unlabelled l-tryptophan at levels of 500mg/l to the cultures results in a 5-fold increase in the yields of both quinoline alkaloids.  相似文献

From cell suspension cultures of Morinda citrifolia five known anthraquinones, rubiadin, lucidin, morindone, lucidin-3-β-primeveroside and morindone-6-β-primeveroside, and seven new anthraquinones were isolated. Six of the seven new quinones were characterized as 2-methyl-3,5,6-trihydroxyanthraquinone, 3-hydroxymorindone, 5,6-dihydroxylucidin, 2-methyl-3,5,6-trihydroxyanthraquinone-6-β-primeveroside, 3-hydroxymorindone-6-β-primeveroside and 5,6-dihydroxylucidin-3-β-primeveroside, respectively.  相似文献

Summary The earliest histological event observed in light-grown shoot-forming tobacco (Nicotiana tabacum L. cv. Wisconsin 38) callus was the deposition of safranin-stainable substances (probably suberin) on cut, exposed cell surfaces. This was followed by the initiation of cell files and the appearance of starch granules. Nodules with lignified tracheary elements also were observed in the upper part of the callus. Pronounced starch accumulation occurred in the lower part of the callus in which protrusions of tissue into the medium occurred. Meristemoids were found in these protrusions as well as elsewhere. In between meristemoids, parenchyma cells with starch granules of varying sizes were observed. Cell strands that connected with the meristemoids also were observed. These strands often terminated at the surface of the protrusion at which point shoot apices originated. The earliest shoots were formed in these protrusions. With time, additiional shoots were formed in other parts of the bottom of the callus and finally in the top part of the callus on prolonged culture. The determination of the loci at which shoot primordia were formed sequentially, was interpreted in relation to the physiological gradient concept. This work was carried out while E. Factors affecting the regeneration of shoots from Nicotiana plumbaginifolia colonies were studied. As opposed to results obtained with tobacco, phytohormone pretreatments of those colonies did not appreciably affect their subsequent aptitude to develop shoots. Hexoses generally inhibited shoot morphogenesis at concentrations higher than 20 mM, but ribose sustained efficient shoot induction at higher concentration. Shoot induction was promoted by a high nitrogen supply. Nitrate promoted greater shoot induction than ammonium succinate or glutamine. Among the micronutrients tested, zinc was stringently required for efficient shoot induction, concentrations ranging between 10 and 100 M being optimal. Under optimized conditions of regeneration, wild type as well as nitrate reductase deficient mutants could be efficiently regenerated.  相似文献

Cell suspension cultures of Cinchona succirubra were cultivated in shake cultures and for the first time in airlift fermenters. Under both conditions L-tryptophan exerts a stimulatory effect on alkaloid formation. In this context the regulatory pattern of some shikimate pathway enzymes was investigated in non-supplemented and tryptophan supplemented Cinchona cell cultures. A remarkable increase of tryptophan decarboxylase (TDC) activity was observed in Cinchona cells under the influence of tryptophan. Apparently, like in some other indole alkaloid producing cell cultures, a high TDC activity is a prerequisite for alkaloid formation. Growth pattern and some enzyme activities of C. succirubra fermenter cultures at controlled and non-regulated pH levels were followed. Licorice plants, Glycyrrhiza glabra, G. uralensis, and G. inflata, were investigated for callus induction using Murashige and Skoog (MS) medium combined with auxins and cytokinins. After 4 weeks of culture, 33-100% of leaf or stem explants formed calli. Maximum of shoot induction from callus cultures was achieved by G. inflata stem explants cultured on MS medium supplemented with 1 mg/l alpha-naphthaleneacetic acid (NAA) and 0.5 mg/l 6-benzyladenine (BA) (67%) which also gave maximum shoot formation per explant (two shoots per explant). These results indicated that all three Glycyrrhiza species regenerated shoots from callus cultures on MS medium combined with NAA and BA or only thidiazuron (TDZ; 0.1 and 0.5 mg/l). Glycyrrhizin contents of G. uralensis calli induced using MS medium in combination with NAA and BA [(27.60 +/- 8.47) microg/g DW] or TDZ alone [(36.52 +/- 2.45) microg/ g DW] were higher than those found in other combinations.  相似文献

St-20 and St-7 lines were isolated from the stem callus of Scutellaria baicalensis Georgi on indole-3-acetic acid and 2,4-dichlorophenoxyacetic acid media, respectively. The flavonoid content of St-20 line was superior to that of St-7 line. The growth and flavonoid (baicalin, baicalein, wogonin and wogonin-7-0-glucuronide) content in St-20 line were best on Linsmaier-Skoog's basal medium containing 10-7 M–10-5 M kinetin. St-20 line showed the same flavonoid content and pattern as the root of parent plant after the culture period of 70 days.  相似文献

Summary The contents of nitrogen-containing compounds and the activities of nitrate assimilating enzymes were determined in tobacco callus cultured under shoot-forming and non-shoot-forming conditions. Whole tissue and tissues cut into top and bottom portions were examined. Highes levels of total-N, protein-N, nitrate and ammonium-N, as well as higher activities of nitrate and nitrite reductases were found in shoot-forming whole tissue and in the shoot-forming bottom portion of tobacco callus in comparison to the non-shoot-forming proliferating tissues throughout the culture period. Legume callus cultures were examined to assess whether regulation of phytoalexin biosynthetic pathways is retained in cultured tissues. Callus tissue cultures ofCanavalia ensiformis (jackbean),Medicago sativa (alfalfa), and nine species ofTrifolium (clover) were established (six clover species for the first time) and maintained on modified Gamborg's B5 medium. Phytoalexins educed in cultures incubated for 48 h with an abiotic elicitor (3.15 mM HgCl2) were detected by their antifungal activity and were purified by column chromatography and high-performance liquid chromatography. Following crystallization, phytoalexins were identified by ultraviolet and proton nuclear magnetic resonance spectroscopy. None of the treated cultures yielded the same complement of phytoalexins reported for fungal-inoculated leaves of the corresponding plants. Callus from all species exceptT. pratense yielded medicarpin, the only phytoalexin reported in treated leaves of all the corresponding plants. A second phytoalexin, maackiain, was found in treatedT. pratense andT. medium calli; maackiain has been reported in fungal-inoculated leaves of those plant species as well asT. hybridum. The phytoalexins sativan and vestitol were not found in treated callus tissues even though they were reported to be present in fungal-inoculated leaves of the same species. These results suggest that (a) the pathway for medicarpin biosynthesis is of central importance for this group of legumes, (b) some phytoalexin anabolic pathways contain metabolic blocks in cells of cultured tissue, and (c) the mechanism for regulating phytoalexin accumulation in tissues is not lost in culture. Initiation and culture of callus and cell suspensions of Cinchona ledgeriana and C. succirubra as well as the successful isolation and selection of a high-yielding alkaloid-forming strain derived from the leaf rachis of a C. succirubra plant are described. Results of feeding experiments with L-tryptophan using two different culture procedures are presented and discussed. Maximum alkaloid yields of up to 0.9% (based on dry weight) or 6.35 mg/l have been obtained.  相似文献

Plants ofNicotiana tabacum L. cv. Burley 21 which showed no difference in nicotine content were used to establish callus cultures. Cultures were initiated from different plants and from different leaves within each plant. The nicotine content of the calli was determined, and the results subjected to an analysis of variance. Differences between plants and differences within plants significantly affected the nicotine content of the cultures. The differences between plants were transmitted sexually and asexually, providing evidence that they are genetically determined. No such differences in nicotine content were found between the plants from which the cultures were established, indicating that nicotine production in vitro involves additional genes to those which are needed for nicotine production in the plant. The differences within plants were further investigated by establishing callus cultures from pith explants taken from different parts of the stem. Explants from apical pith tissue gave calli having far more nicotine and more roots than cultures derived from basal pith explants. This results may reflect the proximity of the apical pith explants to the site of auxin synthesis in the stem apex. Callus cultures derived from pith explants showed greater growth and nicotine production than those derived from leaf explants when the calli were induced on Murashige-Skoog medium containing -naphthalene acetic acid. This result is in conflict with the widely held belief that explants from different parts of the plant give cultures with similar yields of species-specific compounds.Abbreviations HN High nicotine - LN low nicotine  相似文献
