In several surveys in the tropical forests in Thailand, specimens that looked morphologically similar to Metarhizium martiale and Cordyceps variegata, as well as other Metarhizium species were collected and cultured in vitro. A combined phylogeny of several genes including the small (18S) and large (28S) subunits of the ribosomal DNA, elongation factor 1-α (TEF), RNA polymerase II subunits 1 and 2 (RPB1, RPB2) genes has shown these to be new taxa in the Clavicipitaceae. Nigelia is described as a new genus closely related to Metarhizium, to the scale insect pathogens Aschersonia (Hypocrella), Samuelsia and Moelleriella, and to plant pathogens in Claviceps and Balansia, and other relatives. Nigelia comprises M. martiale and a new species Nigelia aurantiaca, which has been found infecting lepidopteran larvae and which produces pseudoimmersed, obliquely arranged, obpyriform perithecia with curved or bent ostioles and with whole (non-separating) cylindric ascospores. Metarhizium chaiyaphumense, M. kalasinense, M. prachinense, M. samlanense, and M. takense are described as new species of Metarhizium. Metarhizium martiale is transferred to Nigelia, and Paecilomyces reniformis is transferred to Metarhizium.  相似文献

Palm fungi are highly diverse in the tropical regions of Asia. Recent investigations on these palmicolous fungi have led to the collection of astrosphaeriella-like taxa from Calamus, Caryota, and Licuala species in Thailand (Chiang Rai and Narathiwat provinces) and southwest China (Yunnan Province). This study characterizes fungal taxa, which are new to science, based on morphological examination and concatenated DNA sequence data, to infer their familial relationships. Morphological comparisons reveal six new species, viz. Astrosphaeriellopsis caryotae, Fissuroma calami, F. caryotae, Neoastrosphaeriella sribooniensis, Pithomyces caryotae, and P. licualae.Their similarities and differences to other extant species are discussed. The phylogenetic results indicate that all of these new taxa belong to Aigialaceae and Astrosphaeriellaceae (Pleosporales) and support their establishment. Astrosphaeriellopsis is assigned to Astrosphaeriellaceae and the family is amended in order to accommodate both coelomycetous and hyphomycetous asexual morphs. A generic key is presented for Astrosphaeriellaceae to delimit Astrosphaeriella, Astrosphaeriellopsis, Pteridiospora, and Pithomyces. Asexual morph connections of Pithomyces caryotae and P. licualae are established from axenic cultures derived from single ascospores. DNA-based sequence data supports the establishment of our new species; however, the affinities of Astrosphaeriella tornata to other Astrosphaeriella and Pithomyces species are unclear and warrant further investigations with increased taxon sampling.  相似文献

The muscles of the male genitalia were studied for the first time in two species endemic to the Oriental zoogeographical region, namely Curetis bulis from the subfamily Curetinae (Lycaenidae) and Paralaxita damajanti from the tribe Abisarini (Riodinidae). Both taxa possess a common plan of musculature reflected in the positions of muscles m1, m2(10), m5(7), m7(6), m21, and m28. Two new autapomorphies of Curetis bulis were discovered: the splitting of m4 into two muscles and a shift of the attachment site of one of these muscles onto the dorsal area of the anellus. Apomorphic differences in the position of the genital muscles were found between Paralaxita damajanti and the previously studied Polycaena tamerlana from the family Riodinidae. A new synapomorphy between the latter two species, namely splitting of the aedeagus protractors m6(5), was also found.  相似文献

Six new natural compounds were isolated from two Far Eastern starfish species, Henricia aspera and H. tumida, collected in the Sea of Okhotsk. Two new glycosylated steroid polyols were obtained from H. aspera: asperoside A and asperoside B, which were shown to be (20R,24R, 25S)-3-O-(2,3-di-O-methyl-β -D-xylopyranosyl)-24-methyl-5α-cholest-4-ene-3β, 6β,8,15α,16β,26-hexaol and (20R, 24R,25S,22E)-3-O-(2,4-di-O-methyl-β-D-xylopyranosyl)-24-methyl-5α-cholest-22-ene-3β,4β,6β,8,15α,26-hexaol, respectively. Two other glycosylated polyols, tumidoside A, with the structure elucidated as (20R, 22E)-3-O-(2,4-di-O-methyl-β -D-xylopyranosyl)-26,27-dinor-24-methyl-5α-cholest-22-ene-3β,4β,6β,8,15α,25-hexaol, and tumidoside B, whose structure was elucidated as (20R,24S)-3-O-(2,3-di-O-methyl-β-D-xylopyranosyl)-5α-cholestan-3β,4β,6β,8,15α,24-hexaol, were isolated from the two starfish species. (20R, 24S)-5α-Cholestan-3β,6β,15α,24-tetraol and (20R, 24S)-5α-cholestan-3β,6β,8,15α,24-pentaol were identified only in H. tumida. The known monoglycosides henricioside H1 and laeviuscolosides H and G were also identified in both species.  相似文献

Phylogenetic analyses of a combined DNA data matrix containing ITS, LSU, rpb2 and tub2 sequences of representative Xylariales revealed that the genus Barrmaelia is a well-defined monophylum, as based on four of its described species (B. macrospora, B. moravica, B. oxyacanthae, B. rhamnicola) and the new species B. rappazii. The generic type of Entosordaria, E. perfidiosa, is revealed as the closest relative of Barrmaelia, being phylogenetically distant from the generic type of Clypeosphaeria, C. mamillana, which belongs to Xylariaceae sensu stricto. Entosordaria and Barrmaelia are highly supported and form a distinct lineage, which is recognised as the new family Barrmaeliaceae. The new species E. quercina is described. Barrmaelia macrospora, B. moravica and B. rhamnicola are epitypified and E. perfidiosa is lecto- and epitypified. Published sequences of Anthostomella and several Anthostomella-like species from the genera Alloanthostomella, Anthostomelloides, Neoanthostomella, Pseudoanthostomella and Pyriformiascoma are evaluated, demonstrating the necessity of critical inspection of published sequence data before inclusion in phylogenies. Verified isolates of several species from these genera should be re-sequenced to affirm their phylogenetic affinities. In addition, the generic type of Anthostomella should be sequenced before additional generic re-arrangements are proposed.  相似文献

An overview of the literature changes in the systematics of algae in the order Laminariales (Phaeophyta) based on molecular phylogentic data is given. In a recent taxonomic revision by Lane et al., [45], the number and status of the families traditionally included in the order have been revised. One family was transferred to the order Tilopteridales; a new family, the Costariaceae, was described; and the genus Laminaria was split into 2 genera, Laminaria and a newly resurrected genus Saccharina. These innovations have necessitated a systematic revision of the Far Eastern species of the Laminariales. Our genetic studies indicate that 2 species of Laminaria and 12 intraspecific taxa (1 subspecies and 11 forms) from the Russian Pacific coasts should be transferred to the genus Saccharina. The following new nomenclatural combinations are proposed: Saccharina bongardiana, comb. nov. (including 4 forms: f. bifurcata, f. subsessilis, f. subsimplex, f. taeniata) and Saccharina gurjanovae, comb. nov. (including f. lanciformis). In addition, new nomenclature combinations are proposed for intraspecific taxa of the Laminaria species (L. angustata, L. cichorioides, L. japonica) that have already been transferred to the genus Saccharina [45]. These include S. angustata subsp. sibirica, comb. nov., 4 new combinations for the forms of S. cichorioides (f. coriacea, f. sacchalinensis, f. sikotanensis, and f. sinuicola), and 2 new combinations for the forms of S. japonica (f. diabolica, and f. longipes). The taxonomic status of the rest of the members of the Laminariales known from the seas of the Russian Far East is discussed. Laminariaceous algae in this area represent 6 of the 8 known families currently included in the Laminariales (Chordaceae, Pseudochordaceae, Alariaceae, Arthrothamnaceae, Laminariaceae, and Costariaceae).  相似文献

Helicosporous hyphomycetes are a morphologically allied group of Tubeufiales. We introduce a new helicosporous genus, Neohelicosporium, with five new species, Neohelicosporium aquaticum, N. guangxiense, N. hyalosporum, N. parvisporum, and N. thailandicum, based on morphological and phylogenetic evidence. The RPB2 protein gene data are provided to analyze their phylogeny in Tubeufiales. Phylogenetic analyses of combined ITS, LSU, RPB2, and TEF1α sequence data from 13 new isolates of Neohelicosporium provided evidence to support the establishment of the new taxa. The morphological characters of Neohelicosporium that differentiate it from other helicosporous species are compared and discussed.  相似文献

Endophytic fungi (EPF) are an important contributor to fungal diversity. It is surmised that EPF colonizing plant roots have high diversity. This study aimed to alleviate the scarcity of information regarding EPF in tropical forests, by isolationg and identifying EPF from a tropical forests in Indonesia. Soils were collected from five forests: (1) Tectona grandis monoculture; (2) Swietenia macrophylla monoculture; (3) Gmelina sp., Artocarpus champeden, Dipterocarp mixed; (4) Dipterocarp primary; (5) Macaranga sp. secondary. Four trees (Calliandra calothyrsus, Paraserianthes falcataria, Sesbania grandiflora, and Cassia siamea) and three crops (Sorghum bicolor, Allium fistulosum, and Trifolium repens) were grown in the forest soils to trap EPF. EPF were isolated from roots and isolation rates were calculated. Based on the isolation rates, P. falcataria and S. bicolor were chosen and grown again in forest soils. EPF were isolated and identified by their rDNA ITS1 region. Twelve and 21 EPF were isolated from 250 roots of P. falcataria and 300 roots of S. bicolor, respectively. Identified EPF were from genera Acrocalymma, Fusarium, Tolypocladium, Penicillium, Talaromyces, Exophiala, Dictyosporium, Pseudochaetosphaeronema, Mariannaea, Trichoderma, and Mycoleptodiscus. Acrocalymma, Tolypocladium, Penicillium, Exophiala, Pseudochaetosphaeronema, Mariannaea, and Mycoleptodiscus spp. were isolated from only one forest. Fusarium, Talaromyces, and Trichoderma spp. were isolated from more than one forest. The numbers of EPF isolated from Gmelina sp., Artocarpus champeden, Dipterocarp mixed forest, and Macaranga sp. secondary forest were higher than those from other forests, suggesting that different plant species in forests affect the root EPF community.  相似文献

The challenge posed by rapidly changing wheat rust pathogens, both in virulence and in environmental adaptation, calls for the development and application of new techniques to accelerate the process of breeding for durable resistance. To expand the resistance gene pool available for germplasm improvement, a panel of 159 landraces plus old cultivars was evaluated for seedling and adult plant resistance (APR) to over 35 Australian pathotypes of Puccinia triticina, Puccinia graminis f. sp. tritici, and Puccinia striiformis f. sp. tritici. Known seedling resistance (SR) genes for leaf rust (Lr2a, Lr3a, Lr13, Lr23, Lr16, and Lr20), stem rust (Sr12, Sr13, Sr23, Sr30, and Sr36), and stripe rust (Yr3, Yr4, Yr5, Yr9, Yr10, Yr17, and Yr27) were postulated. The APR genes identified via field assessments and marker analyses included the pleiotropic genes (Lr34/Yr18/Sr57, Lr46/Yr29/Sr58, Lr67/Yr46/Sr55, and Sr2/Lr27/Yr30), Lr68, Lr74, and uncharacterized APR. A genome-wide association analysis using linear mixed models detected 79 single nucleotide polymorphism (SNP) markers significantly associated with rust resistance, which were mapped on chromosomes 1A, 1B, 1D, 2A, 2B, 3A, 3B, 3D, 4A, 5A, 5B, 6A, 6B, 6D, 7A, 7B and 7D. SNPs associated with multiple rust resistances probably indicate the presence of new pleiotropic or closely linked genes. SNPs were mapped on chromosome positions (1AL, 1DS, 2AL, 4AS, 5BS, 6DL, and 7AL) that have not been known to carry APR genes. This study revealed the presence of a range of possibly unidentified effective seedling and APRs among the landraces, which might represent new sources of rust resistance for the ongoing effort to develop improved wheat cultivars.  相似文献

Radiotherapy is an effective form of therapy for most thoracic malignant tumors. However, myocardial injury resulting from the high doses of radiation is a severe complication. Here we aimed to study the possibility of reducing radiation-induced myocardial injury with mesenchymal stem cell (MSC) transplantation. We used MSCs extracted from bone marrow (BMSCs) to transplant via the tail vein into a radiation-induced heart injury (RIHI) rat model. The rats were divided into six groups: a Sham group, an IRR (irradiation) group, and four IRR + BMSCs transplantation groups obtained at different time points. After irradiation, BMSC transplantation significantly enhanced the cardiac function in rats. By analyzing the expression of PPAR-α, PPAR-γ, TGF-β, IL-6, and IL-8, we found that BMSC transplantation alleviated radiation-induced myocardial fibrosis and decreased the inflammatory reaction. Furthermore, we found that expression of γ-H2AX, XRCC4, DNA ligase4, and TP53BP1, which are associated with DNA repair, was up-regulated, along with increased secretion of growth factors SDF-1, CXCR4, VEGF, and IGF in rat myocardium in the IRR + BMSCs transplantation groups compared with the IRR group. Thus, BMSC transplantation has the potential to improve RIHI via DNA repair and be a new therapeutic approach for patients with myocardial injury.  相似文献

A study of Russian cystic fibrosis (CF) patient DNA was conducted to assess the incidence frequency of 19 mutations, namely CFTRdele2,3(21kb), F508del, I507del, 1677delTA, 2143delT, 2184insA, 394delTT, 3821delT, L138ins, 604insA, 3944delGT, G542X, W1282X, N1303K, R334W, and 3849 + 10kbC > T, S1196X, 621 + 1g > t, and E92K of the CFTR gene. We also sought to determine the estimated CF frequency in Russian Federation. In addition, we determined the total information content of the approach for 19 common mutations registration in the CFTR gene, 84.6%, and the allelic frequencies of the examined mutations: three mutations were observed with a frequency exceeding 5% (F508del, 53.98%, E92K, 6.47%, CFTRdele2,3(21kb), 5.35%); other mutations were observed with frequencies ranging from 0.13 to 3.0%. The CF population carrier frequency was 1 in 38 subjects, while the predicted CF frequency was 1 in 5776 newborns.  相似文献

This study focuses on the former Verticillium section Albo-erecta that included white or whitish species with verticillium-like, erect conidiophores. Its type species was Verticillium fungicola, which was transferred to Lecanicillium, a genus that otherwise contains species with prostrate conidiophores. Phylogenetic analysis of ITS, 18S and 28S rDNA sequences revealed that this group is heterogeneous and its members belong to at least three families. The present study is a first step to its resolution. Supported by analyses of 68 sequences of ITS, 23 sequences of 18S and 43 sequences of 28S ribosomal DNA, the species are distributed over three families. In the Cordycipitaceae, Leptobacillium gen. nov. is described with L. leptobactrum comb. nov. and its new variety, calidius. In the Hypocreaceae, Hypomyces is treated with one new combination from former Nectriopsis, H. tubariicola, and two new anamorphic species, H. ellipsosporus and H. subglobosus; in Sphaerostilbella, one new anamorphic species, S. parabroomeana, is described. In the Bionectriaceae, anamorphic isolates of Nectriopsis requiring a new combination, N. lindauiana, and a new species, N. fuliginicola, are described. Parsimony analysis of the LSU suggests that Ovicillium gen. nov. with four new species, O. attenuatum, O. oosporum, O. subglobosum and O. napiforme, is a member of the Bionectriaceae by 93 % bootstrap support. Of uncertain family classification are Chlamydocillium gen. nov. with one new soil-borne species, C. cyanophilum, and Chlorocillium gen. nov. based on Acremonium griseum Petch. A few further white verticillium-like anamorphs, particularly V. biguttatum, could not yet be reclassified.  相似文献

Molecular breeding for pathogen resistance faces two major problems that delay its widespread adoption, resistance breakdown and difficulties in unambiguously identifying the alleles of the markers associated with specific resistance genes. Since the breakdown of the Rvi6 (Vf) gene in the Northern part of Europe breeders have intensified the search for new resistance sources to be introduced into their breeding programs. Alternative major genes to Rvi6 are available (e.g. Rvi2, Rvi4, Rvi5, Rvi10; Rvi11, Rvi12, Rvi13, and Rvi15, respectively Vh2, Vh4, Vm, Va, Vbj, Vb, Vd, Vr2 according to the old apple scab resistance gene nomenclature) but, with few exceptions (i.e., Rvi4, Rvi5 and, Rvi13), they have so far not been incorporated in commercial varieties. Pyramiding, i.e., combining several of these major resistance genes (R-genes) in individual plants, is one of the most promising strategies currently available to develop apple cultivars with durable apple scab resistance. But, which genes are the best suited to produce such new cultivars? Although the most interesting genes are surely those whose resistance so far has not been broken by the pathogen, genes with resistance that has been overcome coupled with only limited spread of the virulence may also be used in the pyramiding process. However, obtaining information on whether an R-gene is overcome and if so, the extent of the spread of the virulence is difficult and time consuming. Furthermore, often such reports are not up-to-date and the correctness of the data is difficult to verify. To solve these problems, the initiative “Monitoring of Venturia inaequalis virulences” has been proposed. The monitoring is based on a network of orchards of selected differential hosts. Incidence and severity of scab on these genotypes will be collected yearly; and after validation, the data will be published through the homepage of the project (www.vinquest.ch). Here, we present an outline of this initiative. A second major obstacle for broad adoption of marker assisted selection is the lack of tools to align marker analyzes performed in different laboratories to unambiguously identify the alleles linked to specific resistances. The identification of the alleles of the markers in coupling with the resistance genes is often very difficult, if the same genotype used to develop the markers is not simultaneously analyzed. In this paper we present an approach to standardize the size of the alleles in coupling with the resistance genes, using easily accessible cultivars. The proposed procedure has been applied to selected markers for the apple scab resistance genes Rvi2, Rvi4, Rvi5, Rvi6, Rvi11, Rvi12, Rvi13, Rvi14 and Rvi15 (respectively Vh2, Vh4, Vm, Vf, Vbj, Vb, Vd, Rvi14 and Vr2 according to the old nomenclature).  相似文献

Chemical investigation of the freshwater microalga Chlorella sorokiniana led to the isolation of a monogalactosyldiacylglycerol (MGDG)-rich fraction possessing dose-dependent inhibitory activity against pancreatic lipase activity. The MGDG-rich fraction contains 12 MGDGs identified by LC/HRMS analysis. Among them, three MGDGs were new compounds, namely, (2S)-1-O-(7Z,10Z-hexadecadienoyl)-2-O-(7Z,10Z,13Z-hexadecatrienoyl)-3-O-β-D-galactopyranosylglycerol (1), (2S)-1-O-linoleoyl-2-O-(7Z,10Z-hexadecadienoyl)-3-O-β-D-galactopyranosylglycerol (6), and (2S)-1-O-oleoyl-2-O-(7Z,10Z-hexadecadienoyl)-3-O-β-D-galactopyranosylglycerol (8). The major galactolipids were isolated by semipreparative HPLC and tested for their effect toward pancreatic lipase inhibitory activity. All the tested MGDGs showed significant reduction of pancreatic lipase activity indicating possible beneficial use for management of lipase-related disorders such as obesity.  相似文献
