Karyoplasts obtained from full-grown oocytes of the starfish Aphelasterias japonica have practically no cytoplams and are incapable of maturation. Karyoplasts of oocytes of starfishes Marthasterias glacialis and Acanthaster planci have the cytoplasm (10%–15% of the total karyoplast volume) and are often capable of maturation, fertilization and one or several cleavage divisions. The embryoskaryoplasts completely lose supersensitivity and retain usual sensitivity to cytostatic antagonists of neurotransmitters. The assumption is made that the incapability or limited capability of this embryos for development might be due to a deficiency of certain components of the "prenervous" neurotransmitter systems.  相似文献

The red half embryos and related quarter embryos (yolk and pigment) of Arbacia lixula, obtained by means of centrifugation of the eggs in sucrose gradient, retain the normal level of sensitivity and supersensitivity to cytotoxic neuropharmaca, antagonists of biogenic monoamines. The white half embryos and clear quarter embryos practically lack supersensitivity whereas the granular quarter embryos restore it to the initial level. The non-pigmented blastomers of stratified embryos are characterized by somewhat weakened supersensitivity. A suggestion is put forward that the supersensitive embryos of A. lixula possess a sensibilizing factor which couples the supersensitivity receptors with the processes of cell division and moves together with the yolk granules upon centrifugation. This factor is not observed in the Strongylocentrotus granularis embryos lacking evident supersensitivity.  相似文献

Immature oocytes or mature eggs of starfish were centrifuged in a sucrose density gradient. They were then separated into two fractions of fragments, nucleate light fragments and anucleate heavy fragments. Vital-staining experiments showed that the oocytes were elongated along the animal-vegetal (AV) axis during the centrifugation in a contrast to centrifuged eggs whose centrifugal axis was not related to the AV axis. The light and heavy oocyte fragments were comprised of animal and vegetal halves of oocytes, respectively. When matured and fertilized, most of the light oocyte fragment-derived embryos failed gastrulation and developed into Dauerblastulae. Two-dimensional gel electrophoretic analysis of fragments revealed that three basic proteins were predominantly enriched in the heavy oocyte fragments but scarcely detected in the light oocyte fragments. One of these proteins, App20, was identified as a homologue of cyclophilin (peptidyl-prolyl cis-trans isomerase). The present study provides a simple means of separating a population of starfish oocytes into animal and vegetal halves, thereby enabling us to analyze any difference of components between animal and vegetal cytoplasm of the oocytes.  相似文献

Early embryos of Arbacia lixula sea urchin, obtained from eggs pretreated with KYR-12 serotonin analog or A-83 dopamine analog, develop quite normally. At the same time they have a sharply decreased supersensitivity to cytostatic analogs of "prenervous" transmitters; usual sensitivity to this analogs does not change. Besides, both the supersensitivity and the usual sensitivity stop decreasing upon an increase in the density of density of experimental embryos in the incubation medium.  相似文献

1. P. plebeius, a trichopteran with marine intertidal larvae, oviposits in the coelom of a starfish, Patiriella exigua. Oviposition occurs mainly in the spring and autumn months. 2. In spite of the intracoelomic location of the embryos, the development of P. plebeius follows an unmodified trichopteran mode, including the characteristic blastokinesis. Nutrients are not supplied to the caddis embryos by the host starfish. 3. Hatching takes place in the starfish coelum after 17-18 days. The newly hatched caddis larvae quickly escape to their rock pool habitat. 4. The form of the female ovipositor indicates that other species of Chathamidae utilize starfish species as oviposition hosts. 5. This mode of oviposition offers protection to the caddis embryos in the intertidal habitat.  相似文献

The consequences of the chronic continuous administration of Na+-phenobarbital in an average dose of 9.8 mg/kg e.w./24 h were studied in chick embryos. Administration was always started on the 4th day of incubation and lasted 4-12 days. Spontaneous motility was tested in 13- and 17-day-old embryos. The continuous, chronic administration of phenobarbital produced the following significant changes in the embryos' spontaneous motor activity and in the reactivity of their central motor output: 1) In 17-day-old embryos it reduced spontaneous motility in direct correlation to the time of administration. In 13-day-old embryos the effect was not yet significant. 2) It significantly inhibited strychnine, bicuculline and metrazol activation of motor output. 3) It raised the sensitivity to the acute administration of oxazepam (by 18-29.4%), but weakened the inhibitory effect of gamma-aminobutyric acid (by 17.6-33.2%). From the results of these observations it is deduced that spontaneous central motor output activity is not just a developmental epiphenomenon, but that it is an important prerequisite for normal development of the embryonic CNS.  相似文献

Research from many laboratories over the past several decades indicates that invertebrate oocytes and eggs are extraordinarily difficult to freeze. Since starfish oocytes, eggs, and embryos are an important cell and developmental biology model system, there is great interest to cryopreserve these cells. Previous starfish oocyte cryopreservation studies using slow cooling protocols revealed that these cells are highly sensitive to osmotic stress and form intracellular ice at very high sub-zero temperatures, suggesting that common freezing methodologies may not prove useful. We report here that a short exposure to 1.5 M Me2SO/1 M trehalose in hypotonic salt solution followed by ultra-rapid cooling to cryogenic temperatures allows starfish oocytes to be cryopreserved with the average survival rate of 34% when normalized to control oocytes that were exposed to CPA, but not frozen. On average, 51% of the oocytes in 77% of the batches of frozen oocytes underwent meiotic maturation in response to the starfish maturation hormone, 1-methyladenine. In one experiment, eggs developing from thawed oocytes were capable of being fertilized and two developed into embryos. These data suggests that successful cryopreservation of starfish oocytes is possible, but will need further refinement to increase the numbers of fully competent embryos.  相似文献

There is a lag period in the growth of excised wheat (Triticum vulgare Host.) embryos extending from 0.5 hour to 4.5 hours of their imbibition in water. During this time there is a sharp increase in the embryos' capability to retain several amino and organic acids, to synthesize cell wall components and starch, and to take up glucose. Their capability to metabolize glucose also increases by 30%. Elevation of the ATP content of 1-hour embryos by incubation in 3 mm adenosine is not sufficient to bring about these changes. These changes may be a part of a metabolic adjustment in the embryos which increases their growth potential.  相似文献

Involvement of calmodulin-dependent processes in preimplantation development of mouse embryos was studied with the use of N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide (W-7), a specific antagonist of calmodulin. At 25 microM, W-7 interfered with compaction of eight-cell embryos, caused decompaction of compacted eight-cell embryos, inhibited cavitation of late morulae, and caused collapse and degeneration of blastocysts. These effects of W-7 appear to be due to specific inhibition of calmodulin-dependent processes, because W-5, a less active analogue of W-7, was less effective in interfering with development; at 25 microM, W-5 had only a slight effect on compaction and had no effect on blastocyst formation, maintenance of blastocoels, or post-blastocyst development. In addition to the developmental effects just described, W-7 inhibited cell proliferation in four-cell embryos and reduced cell numbers of morulae after treatment at the two- to eight-cell stages. There was a marked increase in embryos' sensitivity to W-7 at the late morula stage, and the sensitivity increased further as embryos developed into blastocysts; the effects of W-7 were largely reversible after treatment at the two-cell through the compacted eight-cell stages, but not after treatment at the late morula or blastocyst stage. At the blastocyst stage, inner cell mass cells appeared to be slightly more resistant to W-7 than trophectoderm cells. This differential sensitivity became more pronounced at the late blastocyst stage: after 3.5-4-h exposure of late blastocysts to 25 microM W-7, all trophectoderm cells degenerated but most of the inner cell masses survived. From these results it appears that calmodulin-dependent processes are involved in development of mouse embryos at all of the preimplantation stages examined.  相似文献

The influence of increased and decreased salinity on early larvae of the mussel Mytilus edulisL. and the starfish Asterias rubensfrom the White Sea is studied. Fertilized egg cells, embryos, larvae, and yearlings of the two species were compared. Salinity tolerance changes with the stage of development of mussels and starfish. One-cell parthenogenetic haploid embryos and blastomeres of the 2- and 4-cell diploid mouse embryos were observed in vitro for the occurrence of two cytoplasmic activities: the cortical activity and the chromatin condensation activity. For this purpose anucleated halves (AHs) and nucleated halves (NHs) were produced by bisection of one-cell embryos and of blastomeres. The cortical activity (manifested by surface deformations) was observed only during the first cleavage cycle. In AHs the surface activity began at the same time as in NHs and disappeared before the time of the cleavage division of nucleated halves. Anucleate fragments of blastomeres from 2- and 4-cell embryos did not exhibit any cortical activity. In the absence of the native nucleus the chromatin condensation activity (assayed by premature chromatin condensation of interphase thymocyte nuclei introduced into cytoplasts by cell fusion) could also have been detected only in the first cleavage cycle. In AHs this activity appeared at the time when NHs started to cleave and disappeared after the NHs finished the first cleavage division. AHs obtained from 2-cell and 4-cell stage blastomeres did not reveal condensation activity. © 1995 Wiley-Liss, Inc.  相似文献

Mating with attractive or dominant males is often predicted to offer indirect genetic benefits to females, but it is still largely unclear how important such non-random mating can be with regard to embryo viability. We sampled a natural population of adult migratory brown trout (Salmo trutta), bred them in vitro in a half-sib breeding design to separate genetic from maternal environmental effects, raised 2098 embryos singly until hatching, and exposed them experimentally to different levels of pathogen stress at a late embryonic stage. We found that the embryos' tolerance to the induced pathogen stress was linked to the major histocompatibility complex (MHC) of their parents, i.e. certain MHC genotypes appeared to provide better protection against infection than others. We also found significant additive genetic variance for stress tolerance. Melanin-based dark skin patterns revealed males with 'good genes', i.e. embryos fathered by dark coloured males had a high tolerance to infection. Mating with large and dominant males would, however, not improve embryo viability when compared to random mating. We used simulations to provide estimates of how mate choice based on MHC or melanin-based skin patterns would influence embryos' tolerance to the experimentally induced pathogen stress.  相似文献

The activation of M-phase promoting factor (MPF) in one-cell mouse embryo is independent from the nucleus. Other autonomous phenomena include the cortical activity observed at the end of the first cell cycle and the reorganization of the microtubule network. Here, we observed that the autonomous control of MPF activation is present also in two-cell mouse embryos (H1 kinase activity being higher in the first than in the second cell cycle). Moreover, the disappearance of the cortical activity in anucleated halves is observed when MPF activation takes place. The rounding up of the cytoplast and the mitotic reorganization of the microtubule network correlates with the maximum activity of H1 kinase in anucleated halves from one-cell embryos. In anucleated halves of two-cell stage blastomeres neither the cortical activity nor the microtubule reorganization were observed. The degree of activation of histone H1 kinase, and, as a consequence, the cortical activity and the microtubule reorganization, does not depend on the distribution of cyclin B. Finally, the level of cyclin B synthesis is similar in anucleated and nucleated halves derived from both one- and two-cell embryos.  相似文献

Effects of hydroxyurea, an inhibitor of ribonucleotide reductase, on cleavage of starfish embryos were studied. In the presence of 1 mM hydroxyurea, fertilized eggs of the starfish, Asterina pectinifera, cleaved up to the 256-cell stage and decomposed before blastulation. Before the 16-cell stage, each blastomere contained a normal nucleus or chromosomes with mitotic apparatus. The cleavage after the 16-cell stage was slow compared to the control embryos, and not all blastomeres contained a nucleus or normal chromosomes. During the fifth cell division (between 16-cell- and 32-cell-stage embryos), chromatin mass unassociated with the mitotic apparatus remained near the cleavage furrow. When hydroxyurea was removed before the 16-cell stage, the embryos developed to normal bipinnalia larvae via normal blastulae. However, the embryos were disintegrated before blastulation when hydroxyurea was removed after the 32-cell stage. DNA synthesis was normally observed before the 16-cell stage but not after the 16-cell stage, but dNTP contents in the embryos remained low throughout development in the presence of hydroxyurea. The achromosomal cleavage observed in the presence of hydroxyurea was reversed by the combination of extracellular dAR and dCR. Therefore, it is assumed that the synthesis of dNTPs required for DNA synthesis in the presence of hydroxyurea occurs via the salvage pathway using deoxynucleosides (dNR) (dNR to dNTP via dNMP and dNDP).  相似文献

Neutral DNA-ase acts as a general teratogen when given to young chicken blastoderms. The rate of the differents abnormalities varies according to the dose given and the embryos' stage. The frequency of the different defects reaches a maximum value with increasing doses it falls generally down with more important doses. Peanut agglutinin (PNA) receptors are expressed in the caudal halves of sclerotomes in chick embryos after 3 days of incubation (stages 19–20 of Hamburger & Hamilton). The neural crest cells forming dorsal root ganglia (DRG) and motor nerves appear to avoid PNA positive regions and concentrate into rostral halves of sclerotomes. To investigate the role of PNA receptors in gangliogenesis and nerve growth, we examined PNA binding ability in quail sclerotomes and in chick-quail chimeric embryos made by transplanting quail somites to chick embryos, comparing the development of DRG, motor nerves and sclerotomes. PNA did not bind to any part of the somites of 4.5-day quail embryos, although dorsal root ganglia and motor nerves appeared only in the rostral halves of sclerotomes as in chick embryos. Moreover, in spite of no PNA binding ability of the transplanted quail somite in 4.5-day chick-quail chimeric embryos, DRG and motor nerves derived from chick tissues appeared only in the rostral halves of the sclerotomes derived from these somites. Thus, both quail and chick neural crest cells and motor nerves recognized the difference between the rostral and caudal halves of sclerotomes of quail embryos in the absence of PNA binding ability, indicating that PNA binding site on somite cells does not support the selective neural crest migration and nerve growth.  相似文献

In Smittia and other chironomid embryos, both anterior and posterior egg halves can give rise to either anterior or posterior segments. Upon various types of experimental interference, eggs may develop one of four basic body patterns: normal embryos, double cephalons, double abdomens, or inverted embryos. From a previous model of anteroposterior determination, we derive four sets of predictions for the results of combined ultraviolet irradiation and centrifugation experiments. While most of the actual results are in agreement with the predictions, some are not. Most of the discrepancies are resolved in a modified version of the model. According to the new model, anterior and posterior egg halves contain both anterior and posterior cytoplasmic determinants. These are thought to be mutually repressive, and to control an overall determination for either anterior or posterior development. Centrifugation and ultraviolet irradiation appear to affect the relative strength of anterior determinants in one or both of the egg halves, thus modifying the probabilities for the four basic body patterns to develop. Different frequencies of these patterns, which have been obtained after similar experimental treatment of different chironomid species, can be ascribed to species-specific variation in the ultraviolet sensitivity of anterior and posterior determinants.  相似文献

A fate map has been constructed for the embryo of Crania. The animal half of the egg forms the ectodermal epithelium of the larva's apical lobe. The vegetal half of the egg forms endoderm, mesoderm, and the ectoderm of the mantle lobe. The vegetal pole is the site of gastrulation; this site becomes the posterior ventral region of the mantle lobe of the larva. The plane of the first cleavage goes through the animal-vegetal axis of the egg; it bears no relationship to the future plane of bilateral symmetry of the larva. The timing of regional specification was examined by isolating animal, vegetal, or meridional halves from oocytes, eggs, or embryos from prior to germinal vesicle breakdown through gastrulation. Animal halves isolated from oocytes formed either the epithelium of the apical lobe or a larva with all three germ layers. Animal halves isolated from unfertilized eggs and eight-cell embryos formed only apical lobe epithelium. Beginning at the blastula stage, animal halves formed mantle in addition to apical lobe epithelium. In animal halves isolated after gastrulation, the mantle lobe was always truncated. Vegetal halves isolated at all stages prior to gastrulation gastrulated and formed apical and mantle lobes with endoderm and mesoderm; however, the relative size of the apical lobe that formed decreased substantially when vegetal halves were isolated at later developmental stages. When meridional halves were isolated from unfertilized eggs and two- to four-cell embryos, both halves frequently formed normally proportioned larvae. Beginning at the blastula stage, a number of pairs frequently had a member that lacked dorsal setae on its mantle lobe while the other member of the pair formed setae, indicating that the dorsoventral axis had been set up. The process of regional specification in Crania is compared to those of Discinisca and Glottidia in the brachiopod subphylum Linguliformea and Phoronis in the phylum Phoronida.  相似文献

小麦成熟胚作为转化受体可克服小麦幼胚存在的受季节和幼胚发育阶段限制的缺点。以湖北省小麦品种‘鄂麦12’和模式品种‘Bobwhite’为材料,成熟胚为转化受体,优化基因枪转化法的轰击压力、轰击距离、选择剂等因素,建立以小麦成熟胚为转化受体的高效转化系统。结果表明:小麦成熟胚作为转化受体时,适宜轰击压力和轰击距离组合是900 psi、6 cm;成熟胚对选择剂G418的敏感性强于幼胚,轰击后需要延长恢复时间,选择剂G418的适合浓度为20~40 mg/L。在以上优化条件下小麦成熟胚转化频率达0.3%~0.9%,已初步建立基因枪介导的小麦成熟胚遗传转化系统。  相似文献
