Summary The sensilla are associated with 6 enveloping cells. The innermost enveloping cell (e 1) secretes the dendritic sheath (=thecogen cell). All other enveloping cells are involved in the formation of the outer cuticular apparatus in secreting the cuticle of a definite region of the new hair shaft.The development of the new sensilla begins when an exuvial space expands between old cuticle and epithelium. The newly forming hair shafts lie folded back in an invagination of the epidermal tissue. Only a distal shaft part projects into the free exuvial space. The cuticle of the distal and middle shaft region is secreted by the three middle enveloping cells (e 2–e 4) (=trichogen cells), which are arranged around the dendritic sheath.The wall of the cylinder, in which the distal shaft is situated, is formed by the cuticle of the future proximal shaft region. It is secreted by the outer enveloping cells (e 5 and e 6). Furthermore, both enveloping cells form the hair socket (=trichogen-tormogen cells).The outer dendritic segments encased within a dendritic sheath run up through the newly formed hair shaft and continue to the old cuticular apparatus. The connection between sensory cells and old hair shaft is maintained until ecdysis. On ecdysis the old cuticle is shed and the newly formed shaft of the sensillum is everted like the invaginated finger of a glove. The dendritic sheath and the outer dendritic segments break off at the tip of the new hair shaft. Morphologically this moulting process ensures that the sensitivity of the receptors is maintained until ecdysis.The internal organization of the sensory cells shows no striking changes during the moulting cycle. An increased number of vesicles is accumulated distally within the inner dendritic segments and distributed throughout the outer segments of the dendrites. The cytoplasmic feature of the enveloping cells indicates that synthesis and release of substances for the cuticular apparatus of the new sensillum take place. 相似文献

Summary The structure and embryonic development of the two types (A, B) of basiconic sensilla on the antennae of Locusta migratoria were studied in material that had been cryofixed and freeze-substituted, or chemically fixed and dehydrated. Both types are single-walled wall-pore sensilla. Type-A sensilla comprise 20–30 sensory and 7 enveloping cells. One enveloping cell (thecogen cell secretes the dendrite sheath); four are trichogen cells, projections of which form the trichogen process during the 2nd embryonic molt. The trichogen cells form two concentric pairs proximally. Two tormogen cells secrete the cuticular socket of the sensillum. The dendritic outer segments of the sensory cells are branched. Bifurcate type-A sensilla have also been observed. Type-B sensilla comprise three sensory and four enveloping cells (one thecogen, two trichogen and one tormogen). The trichogen process is formed by the two trichogen cells, each of which gives rise to two projections. The trichogen cells are concentrically arranged. The dendritic outer segments of the sensory cells are unbranched. In the fully developed sensillum, all trichogen and tormogen cells border on the outer receptor lymph cavity. Summary There is a considerable sexual dimorphism as regards the development of the antenna inNeodiprion sertifer. In the male this is manifested in a larger antennal surface area and the possession of a great number of long single-walled sensilla (SW 1), which are located on the antennal branches. In the female the antennal branches are vestigial, and the single-walled sensilla, of a second type (SW 2), are shorter and present in lower numbers. Both sexes have in addition four types of sensilla in common:viz. a third type of single-walled sensilla (SW 3), terminal pore sensilla, double-walled sensilla, and poreless sensilla. These four sensillar types are characterized not only by their external appearance but also by their internal morphology. 1. Behavioural studies show that larvae of Locusta migratoria (L.) can discriminate between certain simple chemicals and between chemicals obtained from plant sources. 2. Electrophysiological tests show that within each sensillum the same neurones respond to different chemicals. 3. The frequency of occurrence of sensilla with enhanced specificity to certain chemicals is investigated. 4. Statistical tests confirm that, despite the variability of response, across-fibre analysis could allow discrimination between chemicals provided an adequate number of senilla are used. 5. Palpation increases the amount of sensory input reaching the central nervous system compared with that obtained from sustained contact but that increase is not essential to allow discrimination. During a meal on favoured food, phagostimulatory input from the palps is not needed to drive feeding. When less favoured food is taken, phagostimulatory input from the palps may enhance feeding. Four hypotheses about the temporal variation of the number of spermatozoa in the spermatheca of once-mated females were tested in the migratory locust, Locusta migratoria. The best fit provided a regression model that assumed a sudden drop in sperm numbers being indicative of sperm ejection by females. Thereafter, one-fifth of an average ejaculate is stored. Low numbers of sperm stored do not lead to female sperm limitation, as none of the fitness parameters measured, fertilization success, hatching success, and offspring sex ratio, were correlated with the number of sperm present. Fertilization success decreased with successive egg pods. The offspring sex ratio was slightly male biased and tended to increase throughout the laying period. By applying our sperm ejection model to an independent data set of Parker and Smith (1975), we show that the low numbers of sperm retained by the female and subsequent sperm mixing rather than direct replacement may explain the high P2 values found in this species. 相似文献

Summary The fine structure of the terminal sensilla on the maxillary palps of Schistocerca gregaria has been investigated. Most organules include six neurons with dendrites extending to the tip of the cuticular peg, the opening of which is controlled so that the dendrites are not always exposed. The neurons are isolated from each other by a neurilemma cell and two other glial cells, while typical epidermal cells containing dense bundles of microtubules support the whole group of cells. At the poles of the neurons are specialised areas in which the cytoplasm is differentiated from that elsewhere. It contains a large number of mitochondria and small helical structures, while close to it are characteristic spheres of membranes, termed onion bodies, in various stages of development.It is suggested that the fluid bathing the distal parts of the dendrites and exuding from the tip of the peg has a number of specialised functions. It is probably concerned in forcing open the tip of the peg by hydrostatic pressure, it prevents the exposed tips of the dendrites from desiccating and it acts as a transmitter in which chemicals on the surfaces touched by the sensillum must dissolve before reaching the dendrites. This fluid may be produced by the neurilemma cell or by the neurons themselves. Closure of the pegs does not seem to produce any material reduction in the overall loss of water by the insect.Each neuron sends an axon to the brain; there is no peripheral fusion of axons. Possibly one neuron has a mechanoreceptor function, although no specialised terminal at the base of the peg has been observed. The concentration of mitochondria at either end of the neuron may be concerned in the production of action potentials, while the cavity of the peg and tormogen cell perhaps has a role in the conduction of the receptor potential to the perikaryon. Intercellular connections are such as to give mechanical stability to the cells of the organule and permit transport between the cells. Summary The turnip moth Agrotis segetum possesses seven different types of sensilla: four single-walled (SW), one double-walled (DW), one terminal-pore (TP), and one poreless sensilla (NP).The SW 1 and SW 2 sensilla have the same external appearance, being long and slender, but differ in the branching pattern of the sensory processes: unbranched and branched in SW 1 and SW 2, respectively. The SW 3 sensilla are shorter, sickle-shaped, and contain a large number of branches from the sensory processes. These three sensillar types are innervated by 2–3 sensory cells. The SW 4 sensilla are raisin-shaped and possess three profusely branched sensory processes. The DW sensilla are short and have apical slit-like pores. This sensillar type has 5–6 sensory processes. The TP sensilla possess five sensory processes, one of them terminates basally in a tubular body, the others in the apical part of the long cuticular bristle. The NP sensilla are stout and have apical conelike structures. Two of the sensory processes terminate in the apical part, the third proximally. The third sensory process has a lamellar pattern. The association of proctolin with the external ventral protractor muscle of the VIIth abdominal segment (M234) of Locusta migratoria was investigated using immunohistochemistry and RP-HPLC in conjunction with the sensitive locust oviduct bioassay. Immunohistochemistry of whole-mount tissues revealed two proctolin-like immunoreactive axons in N2B2b1 (the nerve branch which innervates M234) as well as immunoreactive processes and varicosities on the surface of M234. Immunogold staining of M234 demonstrated that the proctolin-like immunoreactivity was present in electron-dense granules in its motor terminals. A material indistinguishable from proctolin and with proctolin-like bioactivity co-eluted with authentic proctolin on two different RP-HPLC systems. Bath application of proctolin at concentrations greater than 10-11
M resulted in a dose-dependent increase in the neurally-evoked fast twitch amplitude and duration of M234. Concentrations greater than 10-9
M resulted in a dose-dependent increase in basal tonus of M234. These results indicate that proctolin, or a peptide very similar to proctolin, is present in the motor innervation of M234 and acts as a cotransmitter and/or neuromodulator at this typical fast skeletal muscle.Abbreviations

The innervation of the external ventral protractor muscle of the VIIth abdominal segment (M234) of Locusta migratoria is described using a combination of neurophysiological and neuroanatomical techniques. Cobalt backfills of the nerve innervating M234 revealed two neurons each with soma and fields of arborization in the VIIth abdominal ganglion. In addition, extracellular stimulation of the nerve while gradually increasing the stimulus amplitude resulted in a stepwise increase in both the excitatory junction potential amplitude and twitch amplitude so that two different amplitudes of each were observed indicating the sequential recruitment of two motor neurons. 4-Di-2-ASP stains of M234 revealed pre-synaptic boutons at M234 and a neurohaemal plexus covering the nerve the latter being corroborated by electron microscopic examination of nerve sections. Electron microscopic examination of M234 revealed two axon terminal types, one which is similar to the neurohaemal varicosities over the nerve, containing granules of high electron-density, and one which contains larger granules of medium electron-density. Both terminals types also contained small electron lucent vesicles. Finally, twitch contractions of M234 were modulated by glutamate, proctolin, octopamine, and SchistoF-LRFamide.Abbreviations

Summary The terminals of the fast axon on extensor tibiae muscle fibres of Locusta were examined in untreated nerve-muscle preparations and in preparations stimulated electrically at frequencies varying from 0.5 to 100 Hz. The ultrastructure of the terminals in preparations stimulated at the lower range of these frequencies, which induce twitch contractions of the muscles, is similar to that of the controls. Stimulation at the higher frequencies induced tetanic muscle responses and rapid fatigue of the muscles after which they would not respond again to high frequency stimulation for about 1 h. This loss and recovery of the responses of the muscles is correlated with changes in the ultrastructural appearance of the terminals, in particular in the number and shape of the synaptic vesicles. Summary Coeloconic sensilla on the antenna of Locusta migratoria were investigated electrophysiologically, labeled and then examined under the scanning or transmission electron microscope. They can be categorized into two main morphological types: 1) sensilla with wall pores and two concentric cuticular walls (wp-dw sensilla), and 2) sensilla lacking wall pores (np-sensilla).Hygroreceptors were observed only in np-sensilla. Olfactory receptors were observed in wp-dw sensilla, but not in np-sensilla. Cold-sensitive units were found in both types. Wp-dw sensilla with a cold-sensitive unit also manifest a fatty acid olfactory receptor. This cold-sensitive unit appears to be less sensitive to drops in temperature than the cold receptor combined with hygroreceptors in np-sensilla. Recordings from both types revealed up to three or sometimes four units that could be distinguished on the basis of impulse amplitude.The number of np-sensilla on each antennal segment ranges from 0 to 6. These sensilla are pegs 4–5 m long with a terminal swelling. The dendritic outer segments of two sensory cells fit tightly into these pegs. The dendrite of the third sensory cell ends at its base. All three are up to 1 m thick, unbranched, and densely filled with microtubules. The cuticular wall and socket of the peg are characterized by (1) an electron-dense inner layer surrounding the dendrites and continuous with electron dense clusters at the base of the peg, and (2) clefts in the cuticle at the insertion of the peg.The present observations favor the hypothesis that insect sensilla combining the triad of moist air, dry air and cold receptors generally lack pores. Before moulting, terrestrial isopods resorb calcium carbonate (CaCO3) from the posterior cuticle and store it in sternal deposits. These consist mainly of amorphous calcium carbonate (ACC) spherules
that develop within the ecdysial space between the anterior sternal epithelium and the old cuticle. Ions that occur in the
moulting fluid, including those required for mineral deposition, are transported from the hemolymph into the ecdysial space
by the anterior sternal epithelial cells. The cationic composition of the moulting fluid probably affects mineral deposition
and may provide information on the ion-transport activity of the sternal epithelial cells. This study presents the concentrations
of inorganic cations within the moulting fluid of the anterior sternites during the late premoult and intramoult stages. The
most abundant cation is Na+ followed by Mg2+, Ca2+ and K+. The concentrations of these ions do not change significantly between the stages whereas the mean pH changed from 8.2 to
6.9 units between mineral deposition in late premoult, and resorption in intramoult, respectively. Measurements of the transepithelial
potential show that there is little driving force for passive movements of calcium across the anterior sternal epithelium.
The results suggest a possible role of magnesium ions in ACC formation, and a contribution of pH changes to CaCO3 precipitation and dissolution. 相似文献

Summary The neurosecretory activity of azadirachtin A-treated Locusta migratoria that failed to mature was compared with that of rapidly maturing control females by means of histological techniques. High resolution drymount autoradiography was used to localize [22,23-3H2]dihydroazadirachtin A in the brain and corpus cardiacum (CC). The neurosecretory system of the locusts treated with this insecticide was accompanied by an unusually high accumulation of paraldehyde fuchsin (PAF)-stainable neurosecretory material (NSM) in the brain fibers and in the storage lobes of the CC. The control females never had such an intense accumulation of NSM. The accumulation of NSM, resulting from its poor release, may influence its synthesis which is controlled by feed-back regulation. [22,23-3H2]Dihydroazadirachtin A is localized mainly at the peripheral blood-brain barrier and does not penetrate into the brain effectively, whereas it completely covers the CC. It is concluded that azadirachtin A affects the endocrine activity of the brain in an indirect way. 相似文献

Summary As in other trypanosomatids, the cell membrane of Crithidia fasciculata overlies a single layer of microtubules. Each microtubule possesses a large number of periodically arranged drumstick-like appendages and adjacent microtubules are joined by fibrillar connectives. Anteriorly, the microtubules gradually taper to terminate just before or just after entering the reservoir. An attempt is made to correlate microtubule tapering with maintenance of form of the truncated anterior end of the cell. Smooth and coated vesicles are proliferated from the Golgi saccules and the prominent contractile vacuole lies nearby. The single mitochondrion is extensive and expanded at one point to form a capsule for the kinetoplast. The cristae are predominantly plate-like but other configurations do occur. The cytostome, a shallow invagination of the reservoir membrane, is found between two constrictions in the reservoir wall. Supporting the cytostome are several microtubules which penetrate deeply into the cytoplasm. Ingestion of ferritin occurs by pinocytosis from the cytostome and by coated vesicle formation from the reservoir membrane. Digestion probably occurs in multivesicular bodies which contain acid phosphatase activity. 相似文献
