The role of tyrosine residues in the biological activity of cytotoxin-1 was evaluated using N-bromo succinimide. N-bromo succinimide effected the oxidation of tyrosine residues in cytotoxin-1 with an increase in absorption at 260 nm. N-chloro succinimide was ineffective in the oxidation of tyrosine residues in the toxin. Oxidation of a single tyrosine residue (at 3.50 equivalents of N-bromo succinimide/mole of the toxin) resulted in complete loss of lethal activity of the toxin. The lytic activity of the toxin (lysis of erythrocytes) remained uneffected even after three of the four tyrosine residues in the toxin were oxidised.  相似文献

Summary The distribution and cytoarchitectonic pattern of the magno- and parvocellular hypothalamic nuclei of the cobra, Naja naja, are described at the light-microscopic level. With respect to their tinctorial affinity to paraldehyde fuchsin (AF) as a representative of the Gomori-type of stains, the magnocellular neurons belong to the AF-positive and the parvocellular neurons to the AF-negative elements. In addition to the supraoptic and paraventricular nuclei proper, two accessory aggregations of magnocellular neurons, the nucleus retrochiasmaticus and nucleus circularis, can be identified. Although in a peculiar location, they may be regarded as subunits of the supraopticoparaventricular neurosecretory complex. As many as 22 AF-negative nuclear areas are identified in the hypothalamus of the cobra. The nucleus periventricularis hypothalami of earlier authors is subdivided into several circumscribed neuronal complexes. The nucleus arcuatus, nucleus hypothalamicus lateralis and nucleus lateralis recessus infundibuli are well developed. Indian cobra (Naja naja naja) venom obtained from three different geographical regions was studied in terms of electrophoretic pattern, biochemical and pharmacological activities. SDS-PAGE banding pattern revealed significant variation in the protein constituents of the three regional venoms. The eastern venom showed highest indirect hemolysis and hyaluronidase activity. In contrast, western and southern venoms were rich in proteolytic activity. All the three regional venoms were devoid of p-tosyl-L-arginine methyl ester hydrolysing activity. The eastern venom was found to be most lethal among the three regional venoms. The lethal potency varied as eastern > western > southern regional venoms. In addition, all the three regional venoms showed marked variations in their ability to induce symptoms/signs of neurotoxicity, myotoxicity, edema and effect on plasma coagulation process. Polyclonal antiserum prepared against the venom of eastern region cross-reacted with both southern and western regional venoms, but varied in the extent of cross-reactivity by ouchterlony immunodiffusion and ELISA.  相似文献

Morphological changes of the ovary of the Chinese cobra, Naja naja, throughout the annual reproductive cycle are described. A single clutch of between 6 and 22 eggs is produced in late June. From July to the following April the ovary remains quiescent and contains small previtellogenic, hydration stage follicles. The growth of an ovarian follicle from a primary oocyte to maturation and ovulation is estimated to take three years. The histology of the germinal epithelium and the follicular granulosa shows seasonal changes correlated with the growth of the oocyte. During the quiescent period, the germinal epithelium lacks mitotic activity, but during April, when yolk deposition and rapid growth of the preovulatory follicles take place, the germinal epithelium shows intense mitotic activity. The growth of the smallest hydration stage follicles, and the occurrence of cytoplasmic bridges between the pyriform cells of the granulosa and the developing oocyte, also appear to increase during this period. The possible function of the pyriform cell is discussed and the literature on the origin and fate of these cells in the squamate ovary is reviewed. Postovulatory follicles (corpora lutea) and two types of atresia are described and compared with what is known of these structures in other reptiles.  相似文献

目的 探讨眼镜蛇毒对成年大鼠下托巢蛋白(Nestin)阳性神经细胞表达的影响.方法 采用免疫组织化学方法,观察并比较Nestin阳性细胞在眼镜蛇毒组、生理盐水组、正常对照组大鼠下托的表达.结果 眼镜蛇毒组大鼠下托Nestin阳性神经细胞比生理盐水组、正常对照组表达明显增强(P〈0.01).结论眼镜蛇毒对大鼠下托Nestin表达有上调作用.  相似文献

Reaction of phospholipase A2 (Naja naja naja) with p-bromophenacyl bromidine leads to almost complete loss of enzymatic activity. The rate of inactivation is pH-dependent with pKa equals 6.9 for the ionizing residue. p-Bromophenacyl bromide modifies 0.5 mol of histidine/mol of enzyme as judged by amino acid analysis and incorporation studies with 14C-labeled reagent. The rate of inactivation is affected by various cations; a saturating concentration of Ca2+ decreases the rate 5-fold, while Mn2+ increases the rate by a factor of 2. Triton X-100, which by itself has little affinity for the enzyme, protects against inactivation, presumably by sequestering p-bromophenacyl bromide into the apolar micellar core. The mixed micelle system of Triton X-100, dipalmitoyl phosphatidylcholine, and Ba2+ offers the best protection, lowering the inactivation rate by at least 50-fold. This suggests an active site role for the histidine residue. Ethoxyformic anhydride also modifies phospholipase A2, by acylation of the two amino groups, a tyrosine, and 0.5 mol of histidine/mol of enzyme without totally inactivating the enzyme. Removal of the ethoxyformyl group from the histidine does not reactivate the enzyme. Thus, modification of 0.5 mol of histidine with this reagent is not responsible for the 85% loss of activity seen. Ethoxyformylated enzyme, with 0.5 mol of acylated histidine/mol of enzyme, can be further inactivated by treatment with p-bromophenacyl bromide. The resulting derivative contains 0.4 mol of the 14C-labeled p-bromophenacyl group. Other modifiable groups do not show this half-residue reactivity. For example, oxidation of phospholipase A2 with N-bromosuccinimide leads to rapid destruction of 1.0 tryptophan residue and 5% residual activity. The results of these chemical modification experiments can be interpreted in terms of a model in which the active species of enzyme interacting with mixed micelles is a dimer (or possibly higher order aggregate). The dimer, though composed of identical subunits, is asymmetric; the histidine of one subunit is accessible to ethoxyformic anhydride, while the other histidine is near a hydrophobic region of the enzyme and is chemically reactive toward p-bromophenacyl bromide.  相似文献

Hyaluronidase is present virtually in all snake venoms and has been known as a "spreading factor." The enzyme damages the extracellular matrix at the site of the bite, leading to severe morbidity. In this study, the benefits of inhibiting the hyaluronidase activity of Indian cobra (Naja naja) venom have been investigated. Anti-NNH1 and aristolochic acid both inhibited the in vitro activity of the purified hyaluronidase, (NNH1) and the hyaluronidase activity of whole venom in a dose-dependent manner. Both anti-NNH1 and aristolochic acid abolished the degradation of hyaluronan in human skin tissue sections by NNH1 and by whole venom. Aristolochic acid quenched the fluorescent emission of NNH1. A non-competitive mechanism of NNH1 inhibition was observed with aristolochic acid. NNH1 potentiates the toxicity of Daboia russellii VRV-PL-VIII myotoxin and hemorrhagic complex-I. However, the potentiation of toxicity was inhibited dose-dependently by anti-NNH1 and aristolochic acid. Further, mice injected with whole venom which had been preincubated with anti-NNH1/aristolochic acid, showed more than a two-fold increase in survival time, compared to mice injected with venom alone. A more moderate increase in survival time was observed when mice were injected with anti-NNH1/aristolochic acid 10 min after whole venom injection. This study illustrates the significance of venom hyaluronidase in the pathophysiology of snake venom poisoning and the therapeutic value of its inhibition.  相似文献

Nerve growth factor was isolated from the venom of Naja naja atra by ion exchange and gel permeation chromatography and was found to be homogeneous by disc gel electrophoresis. The molecular weight was estimated to be approximately 20,000 by gel filtration and 22,000 by ultracentrifugation. This protein, which showed an isoelectric point of pH 7.02, probably consists of two subunits of equal molecular weight which are held together or interact with each other noncovalently. The biological activity survives treatment by a number of proteolytic enzymes, such as trypsin [EC 3.4.21.4], chymotrypsin [EC 3.4.21.1], and pepsin [EC 3.4.23.1].  相似文献

The cobras Naja naja and N. kaouthia are abundant in eastern and north-eastern India, accounting for maximum snakebite deaths. Here we report on variation in the composition of Naja kaouthia and N. naja venom from eastern India on corresponding differences in the severity of pathogenesis. These two venoms differ in chromatographic elution profile through Sephadex G-50 and enzyme activity, protein and carbohydrate contents associated with each fraction. The presence of greater amounts of basic phospholipase A2, L-amino acid oxidase and low molecular weight membrane active polypeptides in the N. naja venom makes it more toxic than N. kaouthia venom. A commercial polyvalent antivenom raised against N. naja venom inactivates lethality and variety of toxic effects of homologous venom more effectively than N. kaouthia venom.  相似文献

Summary Due to a decided lack in the literature of reports on the microangioarchitecture of the pancreas of snakes, an analysis was performed in three different species of two different ophidian families with the use of cast preparations and complementary scanning electron microscopy. The vascular architecture reflects the lobular substructure of the pancreas; the organ is supplied by branches of the superior mesentric artery. Coiled lobular arteries and arterioles continue into a meshwork of capillaries. Dilated capillaries of the endocrine portion of the pancreas are an integral component of this fine capillary network. A few very small capillaries establish a connection between the endocrine and the exocrine pancreas. The other capillaries drain into venules, which ultimately join their respective veins. No interspecific differences were noted in the vascularization of the pancreas of the three ophidian species examined. The present results suggest the existence of arterio-venous anastomoses and speak against the presence of a portal system, but establish a feed-forward capillary connection from the endocrine to the exocrine component of the ophidian pancreas.  相似文献

Cortisol, testosterone, dehydroepiandrosterone (DHEA), and aldosterone were measured in the blood serum of healthy subjects who adopted the cobra posture (bhujangasana), a key posture of hatha yoga. The subjects were trained in yoga; blood samples were taken before and after the exercise at an interval of no more than 5 min. As revealed with a new confidence interval–evaluating method developed by A.I. Ivanov, cortisol decreased by 11% with a reliability of 0.99 in all subjects, while testosterone increased by 16% with a reliability of 0.95. Changes in DHEA and aldosterone varied. Proceeding from ideas about motor–visceral interactions, the authors hypothesize that, when the subjects adopted the cobra posture, the production of steroid hormones was controlled by neural mechanisms.  相似文献
