Bulk segregant analysis was used to obtain a random amplified polymorphic DNA (RAPD) marker specific for the rye chromosome arm of the 1BL.1RS translocation, which is common in many high-yielding bread wheat varieties. The RAPD-generated band was cloned and end-sequenced to allow the construction of a pair of oligonucleotide primers that PCR-amplify a DNA sequence only in the presence of rye chromatin. The amplified sequence shares a low level of homology to wheat and barley, as judged by the low strength of hybridization of the sequence to restriction digests of genomic DNA. Genetic analysis showed that the amplified sequence was present on every rye chromosome and not restricted to either the proximal or distal part of the 1RS arm. In situ hybridization studies using the amplified product as probe also showed that the sequence was dispersed throughout the rye genome, but that the copy number was greatly reduced, or the sequence was absent at both the centromere and the major sites of heterochromatin (telomere and nucleolar organizing region). The probe, using both Southern blot and in situ hybridization analyses, hybridized at a low level to wheat chromosomes, and no hybridizing restriction fragments could be located to individual wheat chromosomes from the restriction fragment length polymorphism (RFLP) profiles of wheat aneuploids. The disomic addition lines of rye chromosomes to wheat shared a similar RFLP profile to one another. The amplified sequence does not contain the RIS 1 sequence and therefore represents an as yet undescribed dispersed repetitive sequence. The specificity of the amplification primers is such that they will provide a useful tool for the rapid detection of rye chromatin in a wheat background. Additionally, the relatively low level of cross-hybridization to wheat chromatin should allow the sequence to be used to analyse the organization of rye euchromatin in interphase nuclei of wheat lines carrying chromosomes, chromosome segments or whole genomes derived from rye.  相似文献

Chromosome 1R of rye is a useful source of genes for disease resistance and enhanced agronomic performance in wheat. One of the most prevalent genes transferred to wheat from rye is the stem rust resistance gene Sr31. The recent emergence and spread of a stem rust pathotype virulent to this gene has refocused efforts to find and utilize alternative sources of resistance. There has been considerable effort to transfer a stem rust resistance gene, SrR, from Imperial rye, believed to be allelic to Sr31, into commercial wheat cultivars. However, the simultaneous transfer of genes at the Sec-1 locus encoding secalin seed storage proteins and their association with quality defects preclude the deployment of SrR in some commercial wheat breeding programs. Previous attempts to induce homoeologous recombination between wheat and rye chromosomes to break the linkage between SrR and Sec-1 whilst retaining the tightly linked major loci for wheat seed storage proteins, Gli-D1 and Glu-D3, and recover good dough quality characteristics, have been unsuccessful. We produced novel tertiary wheat-rye recombinant lines carrying different lengths of rye chromosome arm 1RS by inducing homoeologous recombination between the wheat 1D chromosome and a previously described secondary wheat-rye recombinant, DRA-1. Tertiary recombinant T6-1 (SrR+ Sec-1-) carries the target gene for stem rust resistance from rye and retains Gli-D1 but lacks the secalin locus. The tertiary recombinant T49-7 (SrR- Sec-1+) contains the secalin locus but lacks the stem rust resistance gene. T6-1 is expected to contribute to wheat breeding programs in Australia, whereas T49-7 provides opportunities to investigate whether the presence of secalins is responsible for the previously documented dough quality defects.  相似文献

用改良的Giemsa C-带技术、DNA原位杂交和酸性聚丙烯酰胺凝胶电泳(A-PAGE)对来源于小麦品种绵阳11与不同黑麦自交系远缘杂交获得的高代株系(BC1F7)的染色体结构和醇溶蛋白进行了研究。结果发现,在鉴定的200个株系中,有45个株系经C-带和A-PAGE检测均一致地发现它们含有一对1RS /1BL易位染色体,而一个株系843-1-1,C-带鉴定、原位杂交结果均证明它含有一对1RS/1BL易位染色体,但A-PAGE醇溶蛋白图谱却不具有黑麦1RS染色体臂的黑麦碱特征带,而表达出既不同于黑麦碱又不同于亲本绵阳11的醇溶蛋白带型。这一结果表明,利用不同的黑麦亲本资源,可以获得黑麦碱基因Sec-1表达缺失的新的1RS/1BL易位系。这种新的1RS/1BL易位系缺失了影响小麦品质的黑麦碱蛋白,因此是进一步研究1RS/1BL 易位对小麦品质影响的珍贵材料。研究指出,在利用外源基因的植物育种中,外源种供体材料的遗传多样性是值得重视的基因资源。  相似文献

Summary Chromosome pairing between rye chromosome arm 1RS, present in two wheat-rye translocation stocks, and its wheat homoeologues was induced by introducing the translocations into either a ph1bph1b or a nullisomic 5B background. This rye arm carries a gene conferring resistance to wheat stem rust, but lines carrying the translocation produce a poor quality dough unsuitable for breadmaking. Storage protein markers were utilised along with stem rust reaction to screen for allosyndetic recombinants. From a 1DL-1RS translocation, three lines involving wheat-rye recombination were recovered, along with thirteen lines derived from wheat-wheat homoeologous recombination. From a 1BL-1RS translocation, an additional three allosyndetic recombinants were recovered. Nullisomy for chromosome 5B was as efficacious as the ph1b mutant for induction of allosyndesis, and the former stock is easier to manipulate due to the presence of a 5BL-encoded endosperm protein. The novel wheat-rye chromosomes present in the recombinant lines may enable the rye disease resistance to be exploited without the associated dough quality defect.  相似文献

Summary Linkage relationships were established between the secalin loci, Sec 1 (40-K gamma and omega secalins, homologous to the wheat gliadins) and Sec 3 (HMW = high-molecular-weight secalins, homologous to the wheat HMW glutenin subunits), and five chromosomal rearrangements involving chromosome 1R of rye (Secale cereale L.). These were: interchanges T273W (1RL/5RS), T306W (1RS/5RL), and T850W (1RS/ 4RL), Robertsonian centromere split Rb1RW and the interchanged Robertsonian split Rb2R/248W. The analysis established the linkage relationships between the secalin loci and the breakpoints of the rearrangements, in addition to the quantitative effects of the rearrangements on the linkage. Sec-1 is located in the satellite at a position at least 2.5 cMorgan from the proximal border of the terminal C-band, and about 30 cMorgan from the nucleolar organizing region (NOR). The locus is also physically closer to the terminal C-band than to the NOR, but not as much as corresponds with the map distances. Similarly, the physical distance between Sec-3 and the centromere is greater than corresponds with the recombination frequency (0%–9%). Although overall recombination in 1RL remains the same, recombination between the centromere and Sec-3 is greatly reduced in the Robertsonian split combined with the interchange. Oligonucleotide primers were developed to detect the presence of four rye sequences using a PCR assay. These assays give a rye-specific signal from wheat DNA template which contains various rye chromosomes or chromosome segments. The sequences identified were associated with the nucleolar organiser region, the 5S-Rrna-R1 locus, the telomere, and a widely dispersed, rye-specific repetitive element Ris-1. The primers amplified from the well-established loci Nor-R1 and 5S-Rrna-R1 on rye chromosome arm 1RS, and also located a 5s-Rrna locus on chromosome 3R. The telomere-associated sequence was present on every rye chromosome, and was also present, at a low copy number, in both wheat and barley. These assays will be particularly useful for introgression programmes aimed at reducing the rye content of the 1BL.1RS wheat-rye translocation. When multiplexed, the primers will enable a rapid, simultaneous assay for a number of distinct rye loci, which can be derived from a small portion of mature endosperm tissue.  相似文献

Analysis of alcohol-soluble protein (gliadin) patterns of a group of winter common wheat varieties from different breeding centers of Ukraine resulted in identification of six varieties with the rye 1AL/1RS translocation. The line 7086 AR also carries this translocation. The identified 1AL/1RS translocation is similar to that of the variety Amigo in the secalin pattern. The appearance of this translocation in varieties developed in different breeding centers of the forest-steppe of Ukraine indicates its involvement in coadaptive gene associations.  相似文献

Bread wheat (Triticum aestivum L.) is one of the major cereal crops utilized worldwide for bread making. The presence of secalin locus on 1RS leads to the sticky dough and poor bread-making quality of wheat. In the present study, two donor parents, one with distal rye chromatin (1RS44:38) and another with distal wheat chromatin (Pavon MA1) without secalin, and one recipient elite wheat cultivar HD2967 were used. In 1RS44:38, the distal rye region has the Pm8 gene to which the QTL for superior root traits is linked, while in Pavon MA1 with Glu-B3/Gli-B1, the Pm8 gene was found to be absent. This distal rye region having root trait QTL was introgressed into the HD2967 derivatives using marker-assisted backcross selection. The derivatives with distal rye region introgression had higher root biomass, drought resistance, and 6–8% higher yield than the recipient parent cultivar. HD2967 is highly susceptible to yellow rust. Therefore, in the second backcross, the rust-resistant version of HD2967 (Lr57?+?Yr40) was used to introgress rust resistance in the derivatives. Background selection was done using polymorphic wheat anchored SSR markers of A, B, and D genomes of wheat which led to the selection of derivatives with?> 90% background of the recipient cultivar. The significant findings in this study include higher root biomass, improved yield, rust resistance in the derivatives, and retaining the alleles of Glu-B3/Gli-B1 along with Pm8 and the absence of secalin.

Two new T1BL.1RS translocation lines, 48112 and 89121, derived from cross between common wheat (Triticum aestivum L.) cultivar “Xiaoyan No. 6” and rye (Secale cereale L.) cultivar “German White”, were developed and identified by using of molecular markers and cytogenetical methods, GISH and FISH. PCR results of primers NOR-R1 specific for rye and Glu-B3 for 1BS detected the presence of 1RS chromatin and absence of 1BS, and primer for gene 1Bx14 in 1BL indicated the existence of chromosome arm 1BL in the two lines. GISH and FISH methods confirmed the replacement of chromosome arm 1BS with 1RS. Further stripe rust resistant test and quality analysis demonstrated that the new 1BL.1RS translocation lines were higher resistant to mixed races of P. striiformis Westend and observed considerable better quality than other popularized T1BL.1RS cultivars in China. The two lines have been used in wheat breeding for high-yield potential and rust resistance.  相似文献

Summary The heat shock protein profiles from Secale cereale L. cv Imperial, Triticum aestivum L. cv Chinese Spring, S. cereale x T. aestivum amphiploid, and the seven disomic S. cereale addition lines to T. aestivum were used to compare the wheat, rye, and triticale Heat Shock Protein profiles and to study the influence of the rye genome on heat shock protein expression in triticale. Three-day-old seedlings were heat shocked for 2 h at 40 °C in the presence of 35S-methionine, and polypeptides from root tissues were subjected to one- or two-dimensional gel electrophoresis. The wheat and rye heat shock protein profiles each consisted of > 150 heat shock proteins, of which 94 were sufficiently reproducible to construct a standard map. There were 11 unique rye heat shock proteins compared to 22 unique wheat heat shock proteins. The triticale heat shock protein profile resembled the rye parent more than the wheat parent. There were 22 heat shock proteins expressed uniquely by wheat that were not expressed in triticale. Rye chromosomes 1 and 3 exhibited a substantial repressive influence on the expression of 95% of the unique wheat heat shock proteins in triticale, while rye chromosome 4 appeared to have the least repressive influence on expression of the unique wheat heat shock proteins in triticale.Mention of a trade name or proprietary product does not constitute a guarantee, warranty, or recommendation of the product by the United States Department of Agriculture or the University of Missouri and does not imply its approval to the exclusion of other products that may be suitable  相似文献

Photosynthetic parameters were measured in triticale and its parents wheat and rye. Soluble protein content in leaves, ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) content per fresh mass, total chlorophyll content, biomass yield, leaf area, leaf mass and specific leaf mass were higher but Rubisco content expressed as percentage of soluble protein, carboxylase activity, photosynthetic rate and stomatal conductance were significantly lower in rye than in wheat. Native-PAGE of Rubisco revealed that rye carboxylase was different from that of wheat. The difference was not related to either the small or large subunit of Rubisco but, may be, to the ionic and/or other properties of the Rubisco protein moiety. Triticale Rubisco was similar to wheat. For most of the studied physiological parameters, triticale showed much more similarity with wheat than with rye.  相似文献

The gluten proteins document the genotypic identity of a wheat variety, in addition to providing valuable clues about its ancestry and technological properties. In this study, an Indian durum wheat genotype B662 was identified to carry 1BL/1RS translocation and characterized further for its effect on end use quality traits. Comparison of the end use quality traits of B662 with five other durum cultivars without 1BL/1RS, showed decreased gluten content, lower swelling index of glutenins and low MSDS-SV indicating that, B662 with 1BL/1RS is not good for pasta making. In F2:3 seeds from a durum wheat cross between the 1BL/1RS cultivar B662 and HI8498 without the translocation, the secalin Sec-1 loci segregated in theoretically expected 3:1 proportion and were inherited as a block of the rye chromosome arm. The analysis of F2:3 harvests for the two most important durum wheat quality tests showed that the presence of 1BL/1RS translocation did not alter the grain protein content values, but was associated with significant reduction of micro SDS-sedimentation volume indicating inferior quality, thus limiting the commercial exploitation of durum wheat genotypes with 1BL/1RS translocation. The cautious use of rye translocation in Indian durum wheat breeding is suggested.  相似文献

Summary Thirty-five hexaploid and twenty octoploid primary triticales (xTriticosecale Wittmack) derived from homozygous wheat and rye inbred lines were used (1) to investigate the parental wheat, rye, and interaction effects and (2) to estimate quantitative genetic parameters for agronomic traits. The winter triticales were tested in four environments in a three-replicate split-plot design with drilled 1 m2 plots. Superior performance of hexaploid triticales as compared to the octoploids was revealed. Substantial genetic variation and high heritability estimates were found for nearly all of the characters investigated. Estimates of wheat, rye, and wheat×rye interaction variance components disclosed parental main effects to be the most important source of genetic variation in primary triticales. The rye parent was dominant for all characters affecting fertility, and the wheat parent was more important for vegetative development. Character correlations were very similar for triticales of both ploidy levels. The lack of association between grain yield and tillering and the positive correlation between kernels per spike and thousand kernel weight indicated physiological disorders specific for primary triticales.  相似文献

Chromosome painting using multicolor fluorescence in situ hybridization showed that, in addition to the T1AL·1RS translocation derived from rye, a segment from chromosome 3Ae#1 of Agropyron elongatum (2n=10x =70), is present in Amigo wheat. The Agropyron chromosome segment is located on the satellite of chromosome 1B and the translocation chromosome is designated as T1BL·1BS-3Ae#1L. T1BL·1BS-3Ae#1L was inherited from Teewon wheat and carries resistance genes to stem rust (Sr24) and leaf rust (Lr24). The Agropyron chromosome segments in different Sr24/Lr24 carrier wheat lines, including Agent, TAP 48, TAP 67, Teewon, and Amigo, showed a diagnostic C-band, and were derived from the same chromosome, 3Ae#1.  相似文献

A genetic map of six chromosomes of rye, (all of the rye chromosomes except for 2R), was constructed using 77 RFLP and 12 RAPD markers. The map was developed using an F2 population of 54 plants from a cross between two inbred lines. A rye genomic library was constructed as a source of clones for RFLP mapping. Comparisons were made between the rye map and other rye and wheat maps by including additional probes previously mapped in those species. These comparisons allowed (1) chromosome arm orientation to the linkage groups to be given, (2) the corroboration of several evolutionary translocations between rye chromosomes and homoeologous chromosomes of wheat; (3) an increase in the number of available markers for target regions of rye that show colinearity with wheat. Inconsistencies in the location of markers between the wheat and rye maps were mostly detected by multi-copy probes.  相似文献

The detection and analysis of DNA polymorphisms in crops is an essential component of marker-assisted selection and cultivar identification in plant breeding. We have explored the direct amplification of minisatellite DNA by PCR (DAMD-PCR) as a means for generating DNA probes that are useful for detecting DNA polymorphisms and DNA fingerprinting in wheat. This technique was facilitated by high-stringency PCR with known plant and animal minisatellite core sequences as primers on wheat genomic DNA. The products of DAMD-PCR from Triticum aestivum, T. durum, T. monococcum, T. speltoides and T. tauschii showed a high degree of polymorphism and the various genomes could be identified. Cloning of the DAMD-PCR products and subsequent Southern hybridization frequently revealed polymorphic probes showing a good degree of genome specificity. In addition, polymorphic, single locus, and moderately dispersed PCR products were cloned that may have a potential for DNA fingerprinting. Our experiments were limited primarily to diploid wheats and the results indicated that DAMD-PCR may isolate genome-specific probes from wild diploid wheat species that could be used to monitor genome introgression into hexaploid wheat.This paper reports the results of research only. Summary The patterns of chloroplastic and cytosolic isoenzymes of triosephosphate isomerase were analysed by immunoblotting in leaves of rye, wheat, and some species of Aegilops or Agropyrum. While rye contained solely one chloroplastic and one cytosolic isoenzyme, wheat had a much more complex pattern which can be explained by the presence of three genomes in 6 x wheats (AABBDD) with distinct triosephosphate isomerase genes that provided different subunit species for the dimeric isoenzyme molecules. The 6 × wheats contained five, the 4 × wheats three, and the 2 × wheats only one chloroplastic isoenzyme band. The isoenzyme patterns were in accordance with a potential origin of one of the three chloroplastic triosephosphate isomerase genes of 6 × wheats from an Aegilops ancestor. The descent of the other two genes was, however, not in accordance with common contentions on the general evolution of cultural wheats. In the reciprocal intergeneric hybrids Secalotricum and Triticale both the chloroplastic and the cytosolic isoenzyme patterns of rye and wheat were biparentally inherited, indicating that both isoenzymes were controlled by nuclear genes. When monitored by immunoblotting the chloroplastic triosephosphate isomerase isoenzymes may provide useful genetic markers.  相似文献

A 371 base pair segment (bordered by Hind III and Eco RI cutting sites) of wheat embryo nuclear DNA has been cloned and sequenced. It is AT-rich (68%), shares some sequence features with autonomously replicating sequence (ARS) elements, and occurs in approximately 7600 copies per haploid genome. When used as probe for blot hybridization to Hind III-digested wheat DNA, it gives an irregular series of hybridization bands. Essentially the same hybridization pattern was observed for rye DNA. It is concluded that this segment is distributed irregularly but, apparently, according to the same rule in both wheat and rye genomes.  相似文献

利用APAGE、荧光原位杂交技术和RFLP标记,对导入黑麦(SecalecerealeL.)多小穗等性状创制的小麦新种质10_A进行了分子标记检测。APAGE分析发现,10_A与其他1RS/1BL易位系一样,含有1RS的醇溶蛋白标记位点Gld1B3。以黑麦基因组总DNA作探针,用中国春(Triticumaestivumcv.ChineseSpring)基因组DNA作封阻,与10_A根尖细胞有丝分裂染色体进行荧光原位杂交。结果表明,黑麦的1RS易位到10_A中。用25个RFLP探针进行Southern分析,进一步发现10_A的1BS特异限制性片段发生丢失,代之以黑麦1RS的特异限制性片段,而位于其他染色体上的特异限制性片段未发生缺失。据此认为,多小穗小麦新种质10_A属于1RS/1BL易位系。同时还讨论了10_A在小麦遗传改良中的利用情况。  相似文献
