Plants of Cucumis sativus were eithe preinoculated with TNV or treated with drops of a 5% solution of CuSO4 on the cotyledons, and 5 days later challenge-inoculated with Sphaerotheca fuliginea on the first true leaf. The induced systemic resistance was assessed by evaluating the percentage of conidial germination, the length of the hyphae derived from single conidia, the number of cells with lignified walls and that of cells containing haustoria. The number of necrotic cells was also recorded. Compared with controls, in TNV-preinoculated plants conidial germination was lower, hyphal length shorter and the number of haustoria much reduced. The majority of haustoria was found in cells with lignified walls. Pretreatment with CuSO4, although inducing a slight increase of cell wall lignification, did not appreciably reduce the infection process. The number of necrotic cells was very low in all cases. These results suggest that, in this host/pathogen interaction, hypersensitive cell death has no part in the induced defence reaction, although a major role is played by the lignification process.  相似文献

In this investigation, the effects of powdery mildew disease [caused by Podosphaera fusca (syn. Sphaerotheca fuliginea)] on specific activities of several defense-related enzymes and phenolic content were studied in cucumber leaves. Spore suspension of the fungus was sprayed on cucumber (cv. Super Dominus) plants in greenhouse and leaves from both inoculated and non-inoculated control plants were sampled at 0, 24, 48, 72 and 144 hours after inoculation (HAI). Spore germination and tissue colonization of P. fusca were microscopically studied on the inoculated surface of leaf samples. Further, Phenolic content (PHE) and specific activities of peroxidase (POX), chitinase (CHI) and phenylalanine ammonia-lyase (PAL) were spectrophotometrically measured in leaf extracts. Time-course of disease progress on the leaf surface showed that maximum spore germination occurred within 24 HAI and host penetration and disease development process began during 24-48 HAI. Evaluation of enzyme activities showed that POX specific activity in inoculated plants significantly increased at 72 HAI onwards and reached 2.5 times of that of control at 144 HAI. CHI specific activity showed a transient reduction in inoculated plants between 48-72 HAI and thereafter increased significantly in relation to control. PAL specific activity in inoculated plants was not significantly different from that of control. PHE in inoculated plants showed a significant increase compared to control at 48 HAI and thereafter. Comparison of time-course of disease progress with changes in enzyme activities indicated that POX activity had an increasing trend during disease progress whereas CHI activity showed a transient decrease at the early stages and then increased during the later stages of infection: PAL activity did not show any changes during the infection and PHE increased at the early stages of infection process and remained constant at rest of the time.  相似文献

1. Leaf area was removed from Solidago altissima in either a dispersed pattern (half of every leaf removed) or a concentrated pattern (every other leaf removed) and effects on leaf gas exchange, vegetative growth and flowering were examined relative to undefoliated controls. Gas exchange was measured for leaves remaining after defoliation and for regrowth leaves that developed post-damage (at 7, 16 and 26 days post-defoliation).
2. Area-based photosynthetic rates of leaves remaining after defoliation were not affected by either dispersed or concentrated damage, but damage of both types enhanced area-based photosynthesis of regrowth leaves at 16 days post-defoliation and to a lesser extent at 26 days post-defoliation.
3. Dispersed damage, but not concentrated damage, stimulated mass-based photosynthesis of undamaged leaves remaining after defoliation. Undamaged leaves remaining after defoliation and regrowth leaves on damaged plants had higher specific leaf area (leaf area/leaf mass) than comparable leaves on control plants. Mass-based photosynthesis was more strongly elevated by defoliation than area-based photosynthesis because of this increase in specific leaf area.
4. Plants with dispersed damage recovered more quickly from defoliation; they had higher relative growth rates in the first week post-defoliation than plants with concentrated damage. Both types of defoliation caused similar reductions in flower production.
5. These results add to accumulating evidence that dispersed damage is generally less detrimental to plants than concentrated damage and suggest that physiological changes in leaves may be part of the reason.  相似文献

An unusual type of interference between two strains of bean common mosaic virus, viz. NY15 and NL3, infecting bean plants, was investigatred. Wdhen a primary leaf was inoculated with NY15 as inducer and 1–8 days later, the opposite leaf with NL3 as challenger, the plant did not develop symptoms characteristic of NL3, i.e. systemic necrosis in the top, stem and wilting and withering of the youngest trifoloclate leaves. A7-h interval between the inoculations with inducer and challenger already sufficed to reduce the number of plants showing NL3 symptoms. Surprisingly, the amount of NL3 in the challenge-inoculated leaf was always than that in the singly infected control, Furthermore, NY15 could not be detected in the opposite leaf until 8 days post-inoculation and appered enen later in that leaf when challenge-inoculated. Histological studies showed that NL3 appered later in the xylem of bothe the petiole and stem between primary leaves and first trifoliolate leaf, as compared with the singly inoculated control.
The results suggest that suppression of Nl3 symptoms by Ny15 is not caused by impeding its multiplication, but by delaying the transport of NL3 to the xylem of petiole and stem.  相似文献

The paper describes the effects on grain development of theremoval of half the flag leaf, the entire flag leaf and allthe leaves from selected main shoots of a field crop of wheat,cultivar Maris Ranger, 7 days after anthesis. None of the treatmentsreduced grain weights in the 14 days following defoliation.Even in the next 14 days only the more severe defoliations reducedgrain weight, with the most severe treatment causing the greatestreduction. Ultimately the rate of grain growth in plants onwhich only a half of the flag leaf had been removed also fellbehind that of the controls. Final grain yields were in theorder, control plants, > plants with half the flag leaf removed,> the entire flag leaf removed, > all the leaves removed.The significance of these results in interpreting the factorslimiting grain growth in wheat is discussed.  相似文献

Abstract. The mobility and deposition of silicon (Si) in cucumber plants grown in hydroponic culture were studied using scanning electron microscopy and energy dispersive X-ray analysis under conditions of powdery mildew fungus infection. Following transfer to Si containing media, plants grown in the absence of Si showed rapid silicification of leafy tissue, primarily in the trichome bases. These plants also exhibited increased disease resistance to Sphaerotheca fuliginea (Schlecht.:Fr.) Poll, and concentration of Si in the leaf epidermis surrounding the invading pathogen. Plants grown in Si-supplemented media and transferred into Si-deficient media contained residual Si in the leaf trichome bases, but failed to display disease resistance or silicification of host tissue surrounding the invading pathogen.  相似文献

Hyphomycete Paecilomyces fumosoroseus that is well known as saprophytic and entomopatogenic fungus was investigated for its mycoparasitism on the cucumber powdery mildew pathogen. Mycoparasitism was documented by using standard bioassay and SEM. Effects of mycoparasitism were evaluated in three types of experiments. Paecilomyces fumosoroseus was applied in the form of graded suspensions into a colony of powdery mildew on a leaf segment. Interaction between both fungi was observed as the percentage of colonized area vs. experimental time. In the second experiment, young cucumber plants were sprayed with a suspension of Paecilomyces fumosoroseus 24 h before inoculation of Sphaerotheca fuliginea. Pre-treatment with P. fumosoroseus reduced development and spreading of powdery mildew infection significantly 15 days post-inoculation in contrast to pre-treatments with sulfur fungicide and distilled water. The development of pure culture powdery mildew under determined experimental conditions was observed and compared with treated variants. In the third experiment, mildewed plants were treated with a suspension of P. fumosoroseus. The control treatments with sulfur fungicide and distilled water were tested. Effects of P. fumosoroseus on the dispersion of powdery mildew during a 21-day period were observed.P. fumosoroseus suppressed the development and spread of cucumber powdery mildew significantly during the time of the experiment. The mechanical and physical damages and disruptions of vegetative and fruiting structures of powdery mildew were recorded under light microscopy and S.E.M.Results were concluded in pursuance to differences between the natural behaviour and development of S. fuliginea on cucumber plants treated with P. fumosoroseus and non-treated plants.  相似文献

The role of polyamine (PA) metabolism in tobacco (Nicotiana tabacum) defense against pathogens with contrasting pathogenic strategies was evaluated. Infection by the necrotrophic fungus Sclerotinia sclerotiorum resulted in increased arginine decarboxylase expression and activity in host tissues, as well as putrescine and spermine accumulation in leaf apoplast. Enhancement of leaf PA levels, either by using transgenic plants or infiltration with exogenous PAs, led to increased necrosis due to infection by S. sclerotiorum. Specific inhibition of diamine and PA oxidases attenuated the PA-induced enhancement of leaf necrosis during fungal infection. When tobacco responses to infection by the biotrophic bacterium Pseudomonas viridiflava were investigated, an increase of apoplastic spermine levels was detected. Enhancement of host PA levels by the above-described experimental approaches strongly decreased in planta bacterial growth, an effect that was blocked by a PA oxidase inhibitor. It can be concluded that accumulation and further oxidation of free PAs in the leaf apoplast of tobacco plants occurs in a similar, although not identical way during tobacco defense against infection by microorganisms with contrasting pathogenesis strategies. This response affects the pathogen's ability to colonize host tissues and results are detrimental for plant defense against necrotrophic pathogens that feed on necrotic tissue; on the contrary, this response plays a beneficial role in defense against biotrophic pathogens that depend on living tissue for successful host colonization. Thus, apoplastic PAs play important roles in plant-pathogen interactions, and modulation of host PA levels, particularly in the leaf apoplast, may lead to significant changes in host susceptibility to different kinds of pathogens.  相似文献

The levels of peroxidase activity in leaves of non-infected melon plants resistant to Sphaerotheca fuliginea (Schl. ex. Fr.) Poll. were considerably higher than those in the leaves of susceptible plants. After infection the ratio of peroxidase activity to that in non-infected plants reached a value of up to 47.5 in susceptible plants whereas in resistant plants it remained between 1 and 2. Changes in isozyme pattern were investigated by polyacrylamide gel electrophoresis. In non-infected resistant leaves slowly migrating isozymes were found. Their intensities increased with time following infection. These isozymes were absent in the susceptible leaves but appeared after the leaves had been infected with S. fuliginea. The role of these isozymes in the resistance mechanism is discussed.  相似文献

The host selection for oviposition by Spodoptera frugiperda (J.E. Smith) among corn, millet, cotton and soybean, and its relationship with the biological characteristics were investigated. Free and non-choice tests for oviposition using plots containing five plants each, from each host in plastic greenhouse, resulted in similar oviposition preference among the host plants. In addition, selected biological characteristics of S. frugiperda were determined in the laboratory with larvae feeding on host leaves, and the combination of leaf and cotton boll. Neonate larvae exhibited low success of colonization on cotton boll compared to the leaves of all other hosts. Spodoptera frugiperda fed only on cotton bolls exhibited longer larval and pupal development, and longer adult life span; however with similar egg production. Larvae fed cotton leaves during six days and then transferred to cotton bolls, however, exhibited development and reproduction similar to those reared on corn or only on cotton leaves. Therefore, the variations on immature stages of S. frugiperda were not related with host selection for oviposition which was similar among the studied hosts. Based on our data, the millet as a winter, rotational, and cover crop is a potential host for S. frugiperda, while leaves and cotton bolls were diets of intermediate suitability as compared to corn and soybean leaves.  相似文献

Cloned hybridization probes have been used to investigate the effect of infection of susceptible Hordeum vulgare cv Prior by Erysiphe graminis f. sp. hordei on the abundance of host mRNAs coding for the large (LSU) and precursor to the small (SSU) subunits of ribulose 1,5-bisphosphate carboxylase/oxygenase and the rapidly metabolized 32kD thylakoid protein (32kDP). In leaf RNA preparations from control (noninoculated) plants the amount of mRNA for the LSU and SSU declined from 7 to 11 days after sowing, whereas that for the 32kDP increased over this period. Following inoculation at 6 days after sowing, the abundance of each of the mRNA species was significantly reduced below that of controls at 1, 3, and 5 days later. Results indicate that infection causes a rapid and extensive reduction in host mRNA species coding for proteins with important photosynthetic functions.  相似文献

Phytochrome A (phyA) in higher plants is known to function as a far-red/shade light-sensing photoreceptor in suppressing shade avoidance responses (SARs) to shade stress. In this paper, the Avena PHYA gene was introduced into creeping bentgrass (Agrostis stolonifera L.) and zoysiagrass (Zoysia japonica Steud.) to improve turf quality by suppressing the SARs. In addition to wild-type PHYA, a hyperactive mutant gene (S599A-PHYA), in which a phosphorylation site involved in light-signal attenuation was removed, was also transformed into the turfgrasses. Phenotypic traits of the transgenic plants were compared to assess the suppression of SARs under a simulated shade condition and outdoor field conditions after three growth seasons. Under the shade condition, the S599A-PhyA transgenic creeping bentgrass plants showed shade avoidance-suppressing phenotypes with a 45?% shorter leaf lengths, 24?% shorter internode lengths, and twofold increases in chlorophyll concentrations when compared with control plants. Transgenic zoysiagrass plants overexpressing S599A-PHYA also showed shade-tolerant phenotypes under the shade condition with reductions in leaf length (15?%), internode length (30?%), leaf length/width ratio (19?%) and leaf area (22?%), as well as increases in chlorophyll contents (19?%) and runner lengths (30?%) compared to control plants. The phenotypes of transgenic zoysiagrass were also investigated in dense field habitats, and the transgenic turfgrass exhibited shade-tolerant phenotypes similar to those observed under laboratory shade conditions. Therefore, the present study suggests that the hyperactive phyA is effective for the development of shade-tolerant plants, and that the shade tolerance nature is sustained under field conditions.  相似文献

I studied the influence of gibberellic acid (GA3) treatment in a field population of common bean on plant tolerance to leaf removal. Individual bean seedlings were treated with a foliar application of 10 μM GA3 on day 7 and day 14 after emergence, which led to a significant increase in height in GA3-treated plants. Twenty-eight days after emergence, either zero, one, two, or three leaflets from each trifoliate leaf were removed from each of 20 GA3-treated and 20 control plants. All pods were harvested from each plant after plants became senescent 6 weeks later. Multivariate analyses revealed that leaf removal produced significant reductions in several yield components in both GA3-treated and control plants, although the effects were not pronounced until at least two leaflets from each trifoliate leaf (67% of the total leaf area) were removed. However, GA3-treated plants suffered greater reductions in total pod wall mass and total seed number than control plants after 33 and 67% leaf area removal. These results indicate that GA3 treatment may have altered the assimilatory capacity or resource allocation pattern of treated plants in such a way as to decrease their ability to tolerate leaf removal, a negative consequence of the hormonal alteration of traits important to plant compensation for biotic stressors. The effect of timing of the application of the mycoparasite , Verticillium lecanii, on cucumber powdery mildew , Sphaerotheca fuliginea, was studied in a rooted cucumber leaf bioassay . The mycoparasite was applied at different times before and after mildew inoculation . At near - to maximum humidity ( 95% relative humidity) , early preventative (9 and 5 days before mildew inoculation) and early curative control treatments (2 days after mildew inoculation) gave considerable reduction in mildewed leaf areas , while late curative treatments resulted in greater mildewed leaf area but ultimately a reduced amount of healthy mildewed leaf area ( 20 % ) . Appropriate timing of biocontrol treatment application of V. lecanii is important to achieve good control .  相似文献

Using antiserum globulins that reacted only weakly with plant materials, potato leafroll virus (PLRV) at 10 ng/ml was detected consistently by enzyme-linked immunosorbent assay (ELISA). The reaction with PLRV particles was slightly impaired in potato leaf extracts that were diluted less than 10-1 but not at greater dilutions. Antiserum globulins that reacted more strongly with plant materials could be used satisfactorily for coating microtitre plates but were unsuitable for conjugating with enzyme. The detection end-point of PLRV, in leaf sap of potato cv. Cara plants grown from infected tubers in the glasshouse, was about 10-2 and the virus was reliably detected in extracts of composite samples of one infected and 15 virus-free leaves. PLRV concentration was much less in extracts of roots or stolons than in leaf extracts. The virus was detected in infected leaves of all 27 cultivars tested. PLRV was readily detectable 2 wk before symptoms of secondary infection developed in field-grown plants of cv. Cara and Maris Piper and remained so for at least 5 wk. Its concentration was slightly greater in old than in young leaves and was similar to that in glasshouse-grown plants. In field-grown plants of cv. Maris Piper with primary infection, PLRV was detected in tip leaves 21–42 days after lower leaves were inoculated by aphids; in some shoots it later reached a concentration, in tip leaves, similar to that in leaves with secondary infection. Symptoms of primary infection developed in the young leaves of some infected shoots but were inconspicuous and were not observed until at least a week after PLRV was detected by ELISA.  相似文献

The chorioallantoic membrane (CAM) of the fertilized egg allows grafting of human melanomas for short-term investigations and offers the opportunity to investigate the behavior of metastasizing cells and the release of S100beta into peripheral blood. Tissue from one primary melanoma as well as cutaneous and subcutaneous metastases of 10 melanoma patients with elevated levels of S100 in the peripheral blood before surgery were transplanted onto the CAM of chick embryos at day 5/6 of development. Grafts were nourished by the host blood supply 2 days after transplantation. Histologically, 3 days after grafting, metastasizing melanoma cells could be found near the vessels of the host membrane, penetrating the endothelial layer and entering the blood system. Growth conditions remained stable for 6 days after transplantation. Blood samples were taken from a larger CAM vessel before collecting the xenografts 5 days after grafting. Measurement of human S100 in peripheral blood was performed in a blinded manner. No negative control showed elevated levels of human S100 protein. Samples deriving from melanoma xenografts contained highly elevated levels of S100 protein in 80% of cases. The data strongly support the concept of graft-host interaction concerning adherence of tumors and extravasation of human melanoma cells.  相似文献

Two endophytic strains of the entomopathogenic fungus Tolypocladium cylindrosporum, originally isolated from the grass Festuca rubra, were artificially inoculated in tomato and bean plants. Strains 11-1L and 11-0BR were isolated from asymptomatic leaf fragments of both plant species at 3, 7, 14, 21, and 35 days after their inoculation. The percentage of leaf fragments infected by the fungus in inoculated leaves decreased at each sampling time, and no systemic colonization of the plants occurred. The two T. cylindrosporum strains tested were isogenic, differing in the infection by the victorivirus TcV1, harboured by strain 11-1L, but not by 11-0BR. The percentage of infected leaf fragments in leaves inoculated with the virus infected strain was greater in bean than in tomato plants, while the virus-free strain was more successful in tomato than in bean plants. This result suggests that the mycovirus infection can affect the adaptation of T. cylindrosporum to particular host plants.  相似文献

The present study was designed to elucidate the role of gammadelta T cells in the host defense against pulmonary infection with Cryptococcus neoformans. The gammadelta T cells in lungs commenced to increase on day 1, reached a peak level on day 3 or 6, and then decreased on day 10 after intratracheal infection. The increase of these cells was similar in monocyte chemoattractant protein (MCP)-1-deficient mice, although that of NK and NKT cells was significantly reduced. The number of live microorganisms in lungs on days 14 and 21 was significantly reduced in mice depleted of gammadelta T cells by a specific mAb compared with mice treated with control IgG. Similarly, elimination of this fungal pathogen was promoted in gammadelta T cell-deficient (TCR-delta(-/-)) mice compared with control littermate mice. Finally, lung and serum levels of IFN-gamma on days 7 and 14 and on day 7 postinfection, respectively, were significantly higher in TCR-delta(-/-) mice than in littermate mice, whereas levels of TGF-beta showed the opposite results. IL-4 and IL-10 were not different between these mice. IFN-gamma production by draining lymph node cells upon restimulation with cryptococcal Ags was significantly higher in the infected TCR-delta(-/-) mice than in control mice. Our results demonstrated that gammadelta T cells accumulated in the lungs in a manner different from NK and NKT cells after cryptococcal infection and played a down-modulatory role in the development of Th1 response and host resistance against this fungal pathogen.  相似文献
