This study investigates the palaeoecological record (δ18O, δ13C, pollen, plant macrofossils, chironomids and cladocera) at Lake Bled (Slovenia) sedimentary core to better understand the response of terrestrial and aquatic ecosystems to Late-glacial climatic fluctuations. The multi-proxy record suggests that in the Oldest Dryas, the landscape around Lake Bled was rather open, presumably because of the cold and dry climate, with a trend towards wetter conditions, as suggested by an increase in tree pollen as well as chironomid and cladocera faunas typical for well-oxygenated water. Climatic warming at the beginning of the Late-glacial Interstadial at ca. 14,800 cal yr BP is suggested by an increase in the δ18O value, the appearance of Betula and Larix pollen and macrofossils, and a warmth-adapted chironomid fauna. With further warming at ca. 13,800 cal yr BP, broad-leaved tree taxa (Quercus, Tilia, Ulmus), Artemisia, and Picea increase, whereas chironomid data (Cricotopus B) suggest lowering of lake levels. After 12,800 cal yr BP (and throughout the Younger Dryas), the climate was colder and drier, as indicated by lower δ18O values, decline of trees, increase of microscopic charcoal, xerophytes and littoral chironomids. A warmer climate, together with the spread of broad-leaved tree taxa and a deeper, more productive lake, mark the onset of the Late-glacial/Holocene transition. These results suggest that terrestrial and aquatic ecosystems at Lake Bled were very dynamic and sensitive to Late-glacial climatic fluctuations. Extended abstract Lake Grevelingen is a brackish water lake in the SW Netherlands. The lake has an area of 108 km2, a mean depth of 5.3 m (maximum 48 m), a mean chlorinity of 13 to 16%0 Cl, and a hydraulic residence time of about 8 years. Mass budget studies have shown a consistent seasonal pattern in the phosphorus sediment-water exchange in Lake Grevelingen (Kelderman 1980). From May to August a P mobilization from the sediment takes place, estimated atca. 12.5 mg P · m−2 · day−1. The sediment accumulatesca. 5.5 mg P · m−2 · day−1 during the rest of the year. Temperature may be an important factor in establishing this pattern. Sediment-water exchange was studied by means of laboratory experiments under specified conditions. Sediment cores (30 cm depth, 11 cm diameter) were taken at four stations in the lake, with sediment types varying from medium- to muddy sand (Fig. 1). The cores with overlying water (ca. 21) were placed in the dark at 5 °C in thermostatically controlled water baths. After a week's incubation time the temperature was slowly raised, such that after three weeks eight cores (four sediment types, duplicates) were at 5 °C, eight were at 10 °C, eight at 15 °C and eight at 20 °C. The sediment of Lake Balaton (Hungary) provides important information about the lake’s history, particularly with regard to eutrophication. In this study, we used fossil pigment analysis and subfossil Cladocera remains preserved in a dated sediment core to identify trophic stages from ~250 bc to present. Dates of the most recent eutrophic events are in good agreement with previously published data. In general, the abundance and diversity of the Cladocera community increased with eutrophication and decreased with oligotrophication. The sediments of Lake Balaton were characterised by Chydoridae remains, of which Alona species were the most abundant. Of these, Alona quadrangularis and Alona affinis accounted for 40 and 20% of the total Cladocera remains, respectively. The trophic state of Lake Balaton varied between mesotrophic and eutrophic regimes. Seven different trophic periods were identified in Lake Balaton on the basis of Sedimentary Pigment Degradation Unit (SPDU) content of the sediment. Eutrophic states were (1) from ~250 to ~30 bc, (3) between ~300 and ~590 ad, (5) between 1834 and 1944 and (7) from the 1960s until present. Mesotrophic states were (2) ~30 bc to ~300 ad, (4) 590–1834, (6) 1944–1960s. Discriminant analysis of the cladoceran data confirmed these historic events, except for the short mesotrophic episode between 1944 and 1960. The first stage of eutrophication of Lake Balaton (~250 to ~30 bc) was characterised by extensive macrophyte vegetation, as indicated by the increasing abundance of vegetation-associated Cladocera species (Eurycercus lamellatus, Sida crystallina, Pleuroxus sp.). Intensification of eutrophication was identified since the 1980s, reflected by a high abundance of Bosmina species. The most significant planktivorous fish of Lake Balaton was the Sabre carp (Pelecus cultratus), and when its number decreased, the abundance of Bosmina species increased. This study shows that Cladocera are responsive to trophic state changes, underlining their importance as a tool for the assessment of lake eutrophication.  相似文献

Abstract Viable counts and potential activities of different bacteria were determined as a function of depth in the deep profundal sediment of Lake Constance, Germany. The sediment layer at the bottom of the lake had a total depth of about 7 m and was deposited in the time after the last ice age, i.e., over the past 13,000 years. The high clay content of the sediment prevents seepage. Below 25 cm all of the viable heterotrophic bacteria were present as heat-resistant spores. Numbers of viable spores of both aerobic and anaerobic heterotrophic bacteria decreased exponentially with sediment depth and were below the detection limit (5–55 cells ml−1) at 4–6 m, i.e., in about 8,900-year-old sediment. Absence of viable heterotrophic bacteria in deeper sediment layers demonstrated that aseptic sampling conditions were achieved. The decrease of viable spores with depth may be interpreted as time-dependent death of spores resulting in a death rate of about 0.0013–0.0025 year−1. Viable units of specific metabolic groups of bacteria were detected only in the upper sediment layers (0–50 cm). Nitrifying bacteria could not be detected below 30 cm. Methane-oxidizing bacteria were present in the sediment down to >30 cm, but were in a dormant state. Nitrate reduction activity decreased by a factor of 6 within the upper 25 cm of the sediment, but was still detected at 50 cm. Sulfate reduction, on the other hand, could not be detected at depths of 20 cm and below. By contrast, methanogenesis and methanogenic bacteria could be detected down to 50 cm. Sediments from Xuanwu Lake have been dredged in the past 3 years to improve the water quality, but methanogenesis should still exist in the newly settled sediment. Methane production, methanogens, and physiochemical parameters were detected in the surface sediments (0–5 cm) and/or vertical sediments (0–21 cm, segmented at interval of 3 cm). Methane flux at water–air interface varied among five detected sites. Principal component analysis showed that CH4 flux, content of water and the concentration of total nitrogen (TN), CH4 and organic matters (OM) weighed most heavily on the component I in surface sediments while different patterns were observed for vertical sediments. The copy number of the 16S rRNA gene for bacteria was lower in the surface sediment (0–6 cm) than that in deeper sediments (12–21 cm), while 16S rRNA genes of Archaea were almost evenly distributed in the vertical sediments. Representatives belonging to the orders Methanobacteriales, Methanomicrobiales, and Methanosarcinales were detected in all samples of the vertical sediments, except that no members of the Methanococcales were detected in the samples at 0–6 cm. The level of Methanobacteriales reached a highest density at 18.1 × 104 copies g−1 dry weight (dw) at 6–9 cm; for Methanosarcinales (76.89 × 106 copies g−1 dw) and Methanococcales (82.70 × 103 copies g−1 dw) at 12–15 cm, whereas for Methanomicrobiales (43.37 × 106 copies g−1 dw) at 9–12 cm. Methanosarcinaceae and Methanosaetaceae reached to their highest densities at 6–9 and 9–12 cm, respectively. These data provided useful information for better understanding the methanogenesis in the newly settled sediments of a recently dredged lake.  相似文献

Lake Breukeleveen (180 ha, mean depth 1.45 m), a compartment of the eutrophic Loosdrecht lakes system, was selected to study the effects of whole-lake foodweb manipulation on a large scale. In Lake Loosdrecht (dominated by filamentous cyanobacteria), due to water management measures taken from 1970–1984 (sewerage systems, dephosphorization) the external P load has been reduced from 1.2 g m−2 y−1 to 0.35 g m−2 y−1. The water transparency (Secchi-depthca. 30 cm), however, has not improved. The aim of the food-web manipulation in Lake Breukeleveen was not only to improve the light climate of the lake, but also to study if the successfull effects observed in small lakes (a few ha) can be upscaled. In March 1989 the standing crop of planktivorous and bentivorous fish populations was reduced by intensive fishery, fromca. 150 kg ha−1 toca. 57 kg ha−1. The lake was made unaccessible to fish migrating from the other lakes and it was stocked with large-sized daphnids and 0+ pike. However, water transparency did not increase in the following summer and autumn 1989, which is in contrast with great improvement in the light conditions previously observed in smaller lakes. The main explanations for the negative outcome in Lake Breukeleveen are: 1) the rapid increase of the planktivorous fish biomass and carnivorous cladocerans, predating on the zooplankton community; 2) suppression of the large daphnids by the high concentrations of filamentous cyanobacteria; 3) high turbidity of the lake due to resuspension of bottom material induced by wind, unlike in smaller lakes, and thus inability of submerged macrophytes to develop and to stabilize the ecosystem.  相似文献

The recombinant Pichia pastoris harboring an improved methionine adenosyltransferase (MAT) shuffled gene was employed to biosynthesize S-adenosyl-l-methionine (SAM). Two l-methionine (l-Met) addition strategies were used to supply the precursor: the batch addition strategy (l-Met was added separately at three time points) and the continuous feeding strategies (l-Met was fed continuously at the rate of 0.1, 0.2, and 0.5 g l−1 h−1, respectively). SAM accumulation, l-Met conversion rate, and SAM productivity with the continuous feeding strategies were all improved over the batch addition strategy, which reached 8.46 ± 0.31 g l−1, 41.7 ± 1.4%, and 0.18 ± 0.01 g l−1 h−1 with the best continuous feeding strategy (0.2 g l−1 h−1), respectively. The bottleneck for SAM production with the low l-Met feeding rate (0.1 g L−1 h−1) was the insufficient l-Met supply. The analysis of the key enzyme activities indicated that the tricarboxylic acid cycle and glycolytic pathway were reduced with the increasing l-Met feeding rate, which decreased the adenosine triphosphate (ATP) synthesis. The MAT activity also decreased as the l-Met feeding rate rose. The reduced ATP synthesis and MAT activity were probably the reason for the low SAM accumulation when the l-Met feeding rate reached 0.5 g l−1 h−1.  相似文献

A putative ribose-5-phosphate isomerase (RpiB) from Streptococcus pneumoniae was purified with a specific activity of 26.7 U mg−1 by Hi-Trap Q HP anion exchange and Sephacryl S-300 HR 16/60 gel filtration chromatographies. The native enzyme existed as a 96-kDa tetramer with activity maxima at pH 7.5 and 35°C. The RpiB exhibited isomerization activity with l-lyxose, l-talose, d-gulose, d-ribose, l-mannose, d-allose, l-xylulose, l-tagatose, d-sorbose, d-ribulose, l-fructose, and d-psicose and exhibited particularly high activity with l-form monosaccharides such as l-lyxose, l-xylulose, l-talose, and l-tagatose. With l-xylulose (500 g l−1) and l-talose (500 g l−1) substrates, the optimum concentrations of RpiB were 300 and 600 U ml−1, respectively. The enzyme converted 500 g l−1 l-xylulose to 350 g l−1 l-lyxose after 3 h, and yielded 450 g l−1 l-tagatose from 500 g l−1 l-talose after 5 h. These results suggest that RpiB from S. pneumoniae can be employed as a potential producer of l-form monosaccharides.  相似文献

A new yeast, isolated from natural osmophilic sources, produces d-arabitol as the main metabolic product from glucose. According to 18S rRNA analysis, the NH-9 strain belongs to the genus Kodamaea. The optimal culture conditions for inducing production of d-arabitol were 37 °C, neutral pH, 220 rpm shaking, and 5% inoculum. The yeast produced 81.2 ± 0.67 g L−1 d-arabitol from 200 g L−1 d-glucose in 72 h with a yield of 0.406 g g−1 glucose and volumetric productivity Q\textP Q_{\text{P}} of 1.128 g L−1 h−1. Semi-continuous repeated-batch fermentation was performed in shaker-flasks to enhance the process of d-arabitol production by Kodamaea ohmeri NH-9 from d-glucose. Under repeated-batch culture conditions, the highest volumetric productivity was 1.380 g L−1 h−1.  相似文献

An enantio-selective d-Phe imprinted P(AA-co-AN) membrane was prepared using the wet-phase inversion method. The membrane not only selectively adsorbed phenylalanine but also rejected it with a rejection selectivity of 0.82–0.64 and 0.91–0.63 during the filtration of 100 and 10 ppm (g m−3) racemate solutions, respectively. The fluxes of d-Phe and l-Phe during filtration of 10 ppm racemate solution were 0.0077–0.0229 and 0.0064–0.0208 mg m−2 s−1, respectively, and the fluxes of d-Phe and l-Phe during filtration of 100 ppm racemate solution were 0.1287–0.2522 and 0.1174–0.2458 mg m−2 s−1, respectively. The adsorption selectivity was higher at low concentration. The adsorption selectivities varied from 1.11 to 1.65 and from 1.64 to 2.78 during filtration of 100 and 10 ppm racemate solutions, respectively. In respect to desorption, the fractional difference between d-Phe and l-Phe in the recovered solution from 10 ppm was higher than that from 100 ppm.  相似文献

Yeasts that ferment both hexose and pentose are important for cost-effective ethanol production. We found that the soil yeast strain NY7122 isolated from a blueberry field in Tsukuba (East Japan) could ferment both hexose and pentose (d-xylose and l-arabinose). NY7122 was closely related to Candida subhashii on the basis of the results of molecular identification using the sequence in the D1/D2 domains of 26S rDNA and 5.8S-internal transcribed spacer region. NY7122 produced at least 7.40 and 3.86 g l−1 ethanol from 20 g l−1 d-xylose and l-arabinose within 24 h. NY7122 could produce ethanol from pentose and hexose sugars at 37°C. The highest ethanol productivity of NY7122 was achieved under a low pH condition (pH 3.5). Fermentation of mixed sugars (50 g l−1 glucose, 20 g l−1 d-xylose, and 10 g l−1 l-arabinose) resulted in a maximum ethanol concentration of 27.3 g l−1 for the NY7122 strain versus 25.1 g l−1 for Scheffersomyces stipitis. This is the first study to report that Candida sp. NY7122 from a soil environment could produce ethanol from both d-xylose and l-arabinose.  相似文献

l-Ribose is a rare and expensive sugar that can be used as a precursor for the production of l-nucleoside analogues, which are used as antiviral drugs. In this work, we describe a novel way of producing l-ribose from the readily available raw material l-arabinose. This was achieved by introducing l-ribose isomerase activity into l-ribulokinase-deficient Escherichia coli UP1110 and Lactobacillus plantarum BPT197 strains. The process for l-ribose production by resting cells was investigated. The initial l-ribose production rates at 39°C and pH 8 were 0.46 ± 0.01 g g−1 h−1 (1.84 ± 0.03 g l−1 h−1) and 0.27 ± 0.01 g g−1 h−1 (1.91 ± 0.1 g l−1 h−1) for E. coli and for L. plantarum, respectively. Conversions were around 20% at their highest in the experiments. Also partially purified protein precipitates having both l-arabinose isomerase and l-ribose isomerase activity were successfully used for converting l-arabinose to l-ribose.  相似文献

To better understand the bacterial processes in river sediments, it is necessary to investigate the depth-related bacterial communities in the whole sediment profile. Sediment samples were collected to a depth of 25 cm from the Pearl River. Bacterial abundance, activity, cell-specific respiration rate, and diversity were measured, respectively, by 4′, 6-diamidino-2-phenylindole direct count, 5-cyano-2,3-ditolyl tetrazolium chloride (CTC) staining, electron transport system by CTC reduction, and denaturing gradient gel electrophoresis analysis of 16S rRNA amplification fragments. Results showed that the bacterial metabolism activities decreased with the sediment depth. The total bacterial abundance was highest in the surface sediment with 65.1 × 107 cells g−1, and decreased to 11.1 × 107 cells g−1 below 20 cm in the sample location that suffered from heavy sewage inputs. The active bacteria accounted for 7.50–46.7% of the total bacterial number and decreased with the sediment depth. Electron transport system by the CTC reduction showed that bacterial respiration rate declined from 1.093 μmol CTC-formazan h−1 g−1 in the surface sediment to a half in the bottom sediment, while the cell-specific respiration increased significantly with the depth from 3.56 to 93.75 fmol CTC-formazan cell−1. The bacterial diversity also changed with the depth. Beta-Proteobacteria were the dominant species in the surface sediment, whereas Delta-Proteobacteria were the main species below 10 cm. Results of canonical correspondence analysis (CCA) indicated that the distribution of bacteria was affected by the combined effect of various dissolved inorganic matter, while the respiration rate was independent of the nutrient conditions. The specific bacterial distribution contributed to not only the nutrient cycle but also enhanced pollutant decomposition in sediment of the Pearl River. The results showed that some specific bacterial species had a strong activity in the deeper layers. Therefore, the metabolic functions of the deeper bacterial species should not be neglected.  相似文献

Abstract A principle of the cross-scale interaction (CSI) framework is that disturbance-induced landscape changes resulting in coarser-grained spatial structure may non-linearly amplify transfer processes across scales. We studied suspended sediment losses at two spatial scales (0.24 m2 plots and ca. 0.25 ha hillslopes of about 140 m in length) in a semiarid savanna landscape to determine whether the spatial structure of grassy and bare soil areas introduced a non-linear amplification of sediment loss. Sediment loss rates from 0.24 m2 bare plots averaged 1.527 t ha−1 y−1, which was 23 times the loss rate from nearby grassy plots (0.066 t ha−1 y−1). These rates were then extrapolated linearly to two hillslopes separated by only 200 m and having similar total grass cover, slope and soil type but differing in the spatial structure of bare soil patches. The coarse-grained hillslope had a large bare patch on its lower slope, whereas the fine-grained hillslope had no bare soil patches when quantified at a 4 m grid-cell resolution. Measured sediment loss from the fine-grained hillslope averaged 0.050 t ha−1 y−1, whereas the average sediment loss from the coarse-grained hillslope was 2.133 t ha−1 y−1. By linearly extrapolating from the plot scale, the expected sediment loss for the fine-grained hillslope was 0.066 t ha−1 y−1, which is similar to that observed. The expected sediment loss for the coarse-grained hillslope was 0.855 t ha−1 y−1, where linear extrapolation assumed a 46:54 ratio of bare to grassy plots and that the spatial arrangement of plots does not affect sediment loss processes. For the coarse-grained hillslope observed sediment loss is 2.5 times greater than that expected by linear extrapolation from the plot scale. This result indicates a cross-scale interaction related to spatial configuration of patches. We suggest that there were non-linearities in hillslope ecohydrological transfer processes (runoff, erosion) across scales due to a specific patch configuration that greatly amplified sediment loss because the pattern failed to slow runoff and retain sediment before it entered a creek. This example supports the CSI framework and indicates the importance of considering the effect of spatial structure when predicting system dynamics at different scales.  相似文献

In the fed-batch culture of glycerol using a metabolically engineered strain of Escherichia coli, supplementation with glucose as an auxiliary carbon source increased lycopene production due to a significant increase in cell mass, despite a reduction in specific lycopene content. l-Arabinose supplementation increased lycopene production due to increases in cell mass and specific lycopene content. Supplementation with both glucose and l-arabinose increased lycopene production significantly due to the synergistic effect of the two sugars. Cell growth by the consumption of carbon sources was related to endogenous metabolism in the host E. coli. Supplementation with l-arabinose stimulated only the mevalonate pathway for lycopene biosynthesis and supplementation with both glucose and l-arabinose stimulated synergistically only the mevalonate pathway. In the fed-batch culture of glycerol with 10 g l−1 glucose and 7.5 g l−1 l-arabinose, the cell mass, lycopene concentration, specific lycopene content, and lycopene productivity after 34 h were 42 g l−1, 1,350 mg l−1, 32 mg g cells−1, and 40 mg l−1 h−1, respectively. These values were 3.9-, 7.1-, 1.9-, and 11.7-fold higher than those without the auxiliary carbon sources, respectively. This is the highest reported concentration and productivity of lycopene.  相似文献

A simple and rapid method was devised for determination of tryptophan, based on the Belousov-Zhabotinskii (B-Z) oscillating chemical system. Changes in oscillating period and amplitude were linearly proportional to the negative logarithm of l-tryptophan concentration over the range of 6.44 × 10−7–2.55 × 10−4 M, with the regression coefficients of near unity and a lower detection limit of 6.5 × 10−8 M. d-tryptophan was also examined although it is rarely found in most biological fluids, and perhaps not at all in natural proteins. The change of period against to negative logarithm of d-tryptophan concentration over the range of 4.9 × 10−5–8.24 × 10−4 M is linear. Because the optimum conditions for determination of l- and d-tryptophan are not the same, a little amount of d-tryptophan does not affect the determination of l-tryptophan. Various influences were studied and a possible mechanism of perturbation to the B-Z oscillator by tryptophan was also discussed. Spectrophotometry and fluorescence spectrophotofluorimetry were used for comparision and confirmation of the results.  相似文献

The production of l-phenylalanine is conventionally carried out by fermentations that use glucose or sucrose as the carbon source. This work reports on the use of glycerol as an inexpensive and abundant sole carbon source for producing l-phenylalanine using the genetically modified bacterium Escherichia coli BL21(DE3). Fermentations were carried out at 37°C, pH 7.4, using a defined medium in a stirred tank bioreactor at various intensities of impeller agitation speeds (300–500 rpm corresponding to 0.97–1.62 m s−1 impeller tip speed) and aeration rates (2–8 L min−1, or 1–4 vvm). This highly aerobic fermentation required a good supply of oxygen, but intense agitation (impeller tip speed ~1.62 m s−1) reduced the biomass and l-phenylalanine productivity, possibly because of shear sensitivity of the recombinant bacterium. Production of l-phenylalanine was apparently strongly associated with growth. Under the best operating conditions (1.30 m s−1 impeller tip speed, 4 vvm aeration rate), the yield of l-phenylalanine on glycerol was 0.58 g g−1, or more than twice the best yield attainable on sucrose (0.25 g g−1). In the best case, the peak concentration of l-phenylalanine was 5.6 g L−1, or comparable to values attained in batch fermentations that use glucose or sucrose. The use of glycerol for the commercial production of l-phenylalanine with E. coli BL21(DE3) has the potential to substantially reduce the cost of production compared to sucrose- and glucose-based fermentations.  相似文献

In this study, the effects of citrate addition on d-ribose production were investigated in batch culture of a transketolase-deficient strain, Bacillus subtilis EC2, in shake flasks and bioreactors. Batch cultures in shake flasks and a 5-l reactor indicated that supplementation with 0.2–0.5 g l−1 of citrate enhanced d-ribose production. When B. subtilis EC2 was cultivated in a 15-l reactor in a complex medium, the d-ribose concentration was 70.9 g l−1 with a ribose yield of 0.497 mol mol−1. When this strain was grown in the same medium supplemented with 0.3 g l−1 of citrate, 83.4 g l−1 of d-ribose were obtained, and the ribose yield was increased to 0.587 mol mol−1. Addition of citrate reduced the activities of pyruvate kinase and phosphofructokinase, while it increased those of glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase. Metabolic flux distribution in the stationary phase indicated that citrate addition resulted in increased fluxes in the pentose phosphate pathway and TCA cycle, and decreased fluxes in the glycolysis and acetate pathways.  相似文献

A Holocene sediment sequence from Lake Seukokjaure, a subarctic lake at tree-line in northern Sweden, was analyzed to assess major changes in the structure and functioning of the aquatic ecosystem in response to climate change and tree-line dynamics. The compiled multi-proxy data, including sedimentary pigments, diatoms, chironomids, pollen, biogenic silica (BSi), carbon (C), nitrogen (N) elemental and stable-isotope records, and total lake-water organic carbon (TOC) concentration inferred from near-infrared spectroscopy (NIRS), suggest that the Holocene development of Lake Seukokjaure was closely coupled to changes in terrestrial vegetation with associated soil development of the catchment, input of allochthonous organic carbon, and changes in the light regime of the lake. A relatively productive state just after deglaciation around 9700 to 7800 cal years BP was followed by a slight long-term decrease in primary production. The onset of the local tree-line retreat around 3200 cal years BP was accompanied by more diverse and altered chironomid and diatom assemblages and indications of destabilized soils in the catchment by an increase in variability and absolute values of δ13C. An abrupt drop in the C/N ratio around 1750 cal years BP was coupled to changes in the internal lake structure, in combination with changes in light and nutrient conditions, resulting in a shift in the phototrophic community from diatom dominance to increased influence of chlorophytes, likely dominated by an aquatic moss community. Thus, this study emphasizes the importance of indirect effects of climate change on tree-line lake ecosystems and complex interactions of in-lake processes during the Holocene.  相似文献

We examined the hydrologic controls on nitrogen biogeochemistry in the hyporheic zone of the Tanana River, a glacially-fed river, in interior Alaska. We measured hyporheic solute concentrations, gas partial pressures, water table height, and flow rates along subsurface flowpaths on two islands for three summers. Denitrification was quantified using an in situ 15NO3 push–pull technique. Hyporheic water level responded rapidly to change in river stage, with the sites flooding periodically in mid−July to early−August. Nitrate concentration was nearly 3-fold greater in river (ca. 100 μg NO3–N l−1) than hyporheic water (ca. 38 μg NO3–N l−1), but approximately 60–80% of river nitrate was removed during the first 50 m of hyporheic flowpath. Denitrification during high river stage ranged from 1.9 to 29.4 mg N kg sediment−1 day−1. Hotspots of methane partial pressure, averaging 50,000 ppmv, occurred in densely vegetated sites in conjunction with mean oxygen concentration below 0.5 mgOl−1. Hyporheic flow was an important mechanism of nitrogen supply to microbes and plant roots, transporting on average 0.41 gNO3–N m−2 day−1, 0.22 g NH4+–N m−2 day−1, and 3.6 g DON m−2 day−1 through surface sediment (top 2 m). Our results suggest that denitrification can be a major sink for river nitrate in boreal forest floodplain soils, particularly at the river-sediment interface. The stability of the river hydrograph and the resulting duration of soil saturation are key factors regulating the redox environment and anaerobic metabolism in the hyporheic zone.  相似文献
