A decline in mitochondrial function occurs in many conditions. A report in this issue of Cell Metabolism (Larsen et al., 2011) shows that dietary inorganic nitrates enhance muscle mitochondrial efficiency. It is an attractive hypothesis that dietary changes enhance energy efficiency, but its potential application depends on long-term studies investigating net benefits versus adverse effects.  相似文献

Native plant individuals often persist within communities dominated by exotics but the influence of this exposure on native populations is poorly understood. Selection for traits contributing to competitive ability may lead to native plant populations that are more tolerant of the presence of exotic invaders. In this way, long‐term coexistence with an exotic may confer competitive advantages to remnant (experienced) native populations and be potentially beneficial to restoration. In past studies we have documented genetic differentiation within native grass populations exposed to the exotic invader Russian knapweed (Acroptilon repens). Here, we examine populations of a cool‐season grass, needle‐and‐thread (Hesperostipa comata [Trin. & Rupr.]) and a warm season, alkali sacaton (Sporobolus airoides [Torr.]) collected from Russian knapweed‐invaded sites and adjacent noninvaded sites to assess their relative competitive ability against a novel exotic neighbor, Canada thistle (Cirsium arvense). Experienced S. airoides (from within A. repens invasions) appear to better tolerate (accumulate biomass, leaf nitrogen content, and to initiate new tillers) the presence of a novel competitor (C. arvense). Experienced and inexperienced H. comata genets differ in their response to the presence of C. arvense. Relative neighbor effects of native grasses on C. arvense were generally greater from experienced grasses. The ability to compete with novel neighbors may be driven by general competitive traits rather than species‐specific coevolutionary trajectories. Irrespective of competitive mechanisms, the conservation of native species populations within weed invasions may provide an important restoration tool by retaining unique components of native gene pools selected by competitive interactions with exotics.  相似文献

Low efficiency is a key problem confronting the development and application of phytoremediation technology. Based on political pressure to reduce CO2 emissions in China and the fact that CO2 is necessary for plant photosynthesis, the effects of captured CO2 fertilization on phytoremediation of soil di-(2-ethylhexyl) phthalate (DEHP) pollution by C3 plant (mung bean, Vigna radiata L.) and C4 plant (maize, Zea mays L.) were investigated. Results showed that DEHP pollution negatively affected the growth and rhizosphere environments of both plants. After CO2 fertilization, both plants had more biomass (aboveground, belowground, and total dry weight), higher alkaline phosphatase activity, and more microbes with DEHP tolerance in their rhizospheres. Superoxide dismutase activity in leaves of both plants decreased significantly. Microbial community composition in both rhizospheres changed. CO2 fertilization also increased plant uptake of DEHP, particularly in the roots, and decreased residual DEHP concentrations in the rhizospheres. These effects were more evident in the C3 than in the C4 plant. This study indicated that CO2 fertilization can enhance the phytoremediation process of polluted soil through promoting plant growth, improving the rhizosphere environment, and increasing plant uptake of DEHP, particular in a C3 plant. CO2 fertilization could be considered as a measure to enhance phytoremediation.  相似文献

As well as being phytohormones, gibberellins (GAs) are present in some fungi and bacteria. Indeed, GAs were first discovered in the fungus Gibberella fujikuroi, from which gibberellic acid (GA3) and other GAs are produced commercially. Although higher plants and the fungus produce structurally identical GAs, there are important differences in the pathways and enzymes involved. This has become particularly apparent with the identification of almost all of the genes for GA-biosynthesis in Arabidopsis thaliana and G. fujikuroi, following the sequencing of the Arabidopsis genome and the detection of a GA-biosynthesis gene cluster in the fungus. For example, 3b-hydroxylation occurs early in the pathway in G. fujikuroi and is catalyzed by a cytochrome P450 monooxygenase, whereas it is usually the final step in plants and is catalyzed by 2-oxoglutarate-dependent dioxygenases. Similarly, 20-oxidation is catalyzed by dioxygenases in plants and a cytochrome P450 in the fungus. Even where cytochrome P450s have equivalent functions in plants and Gibberella, they are unrelated in terms of amino acid sequence. These profound differences indicate that higher plants and fungi have evolved their complex biosynthetic pathways to GAs independently and not by horizontal gene transfer.  相似文献

Site-specific risk assessment of contaminated areas indicates prior areas for intervention, and provides helpful information for risk managers. This study was conducted in the Ervedosa mine area (Bragança, Portugal), where both underground and open pit exploration of tin and arsenic minerals were performed for about one century (1857 – 1969). We aimed at obtaining ecotoxicological information with terrestrial and aquatic plant species to integrate in the risk assessment of this mine area. Further we also intended to evaluate if the assessment of other parameters, in standard assays with terrestrial plants, can improve the identification of phytotoxic soils. For this purpose, soil samples were collected on 16 sampling sites distributed along four transects, defined within the mine area, and in one reference site. General soil physical and chemical parameters, total and extractable metal contents were analyzed. Assays were performed for soil elutriates and for the whole soil matrix following standard guidelines for growth inhibition assay with Lemna minor and emergence and seedling growth assay with Zea mays. At the end of the Z. mays assay, relative water content, membrane permeability, leaf area, content of photosynthetic pigments (chlorophylls and carotenoids), malondialdehyde levels, proline content, and chlorophyll fluorescence (Fv/Fm and ΦPSII) parameters were evaluated. In general, the soils near the exploration area revealed high levels of Al, Mn, Fe and Cu. Almost all the soils from transepts C, D and F presented total concentrations of arsenic well above soils screening benchmark values available. Elutriates of several soils from sampling sites near the exploration and ore treatment areas were toxic to L. minor, suggesting that the retention function of these soils was seriously compromised. In Z. mays assay, plant performance parameters (other than those recommended by standard protocols), allowed the identification of more phytotoxic soils. The results suggest that these parameters could improve the sensitivity of the standard assays.  相似文献

Studies of human-mediated rates of introduction of organisms to islands are few, results fall into two models (linear and exponential), and relationships with human population and activities are obscure. Owing to their late settlement and good scientific record, data from Galapagos may be exceptionally informative. The rate of introduction of vascular plant species to Galapagos has been suggested to be exponential, paralleling the rise in human population. However, detailed reconstruction of growth in numbers of introduced plant species, based on historical and recent records, reveals changes in study criteria over the last two centuries, which obscure the true introduction rate. At first, cultivated species were deliberately excluded from most studies. From the 1960s, naturalized cultivated species were included more consistently, but non-naturalized species were still excluded. From the mid-1980s, the latter were deliberately included. Accidental introductions increased linearly from 1807 (the first records) to the present. Escapes from cultivation show increases in rate around 1906 and in the period 1970–1990, the latter coinciding with the first studies directed at areas affected by human activities. Non-naturalized cultivated species rose abruptly from the late 1980s, as they became deliberately studied. There seems to be no direct link with human population size. Data represent rate of discovery rather than true introduction rate, and the changing overall rate reflects changing botanical interests and research effort. Data from other islands also suggest that linear increases in naturalized plants are the norm. Galapagos data do not permit confident statements about the introduction rate of cultivated species, but suggest that this may depend more on human activities than human population size.  相似文献

Top-predator (fourth-trophic-level) controlled trophic cascades are thought to be uncommon in terrestrial systems, but actual quantitative tests and comparisons of bottom-up and top-down forces in systems with more than three linear trophic levels are rare. Here, we describe the density patterns of the arthropod community associated with Piper ant-plants in Costa Rican wet forests. Consumers in this community comprise a complex, interacting web of herbivores, predaceous ants, and predators of ants. Although the hollow stems and petioles of the Piper plants provide some protection to resident ants from predation, specialized Dipoena spiders and Phyllobaenus beetles exploit Pheidole ants inhabiting Piper plants. We report abundance patterns of plants, ants and predators in four forests. These patterns of abundance are consistent with predictions of top-down cascades across four trophic levels when the top predators are effective (beetles). We discuss how top-down and bottom-up forces may interact in systems with less effective top predators (spiders).  相似文献

In rain forest, the large numbers of species of fleshy-fruited plants and frugivorous animals result in a large number of potential fruit–frugivore interactions, which are challenging to survey in the field. Yet, knowledge of these relationships is needed to predict consequences of changes in the frugivore assemblage for seed dispersal. In the absence of comprehensive dietary information, it may be possible to delineate between frugivores that disperse different plants using ‘functional traits,’ or morphological and behavioral attributes of frugivores that interact with differences in salient characteristics of plant species. Here we use data on the consumption of 244 Australian rain forest plant species by 38 bird species to test for associations between patterns of frugivory and birds': (1) degree of frugivory, (2) gape width, and (3) seed treatment (seed crushing or seed dispersing). Degree of frugivory and gape width explain 74 percent of the variation in the sizes of fruits consumed by frugivorous birds. Among birds that consume a substantial dietary proportion of fruit, birds with wider gapes consume larger fruits. In contrast, this relationship was not shown by birds for which fruit is only a minor dietary component. Degree of frugivory and gape width, together with seed treatment, also strongly predict the overall taxonomic composition and diversity of plants consumed by bird species. Functional classifications of frugivore species may prove useful in developing a predictive understanding of fruit–frugivore interactions in other rain forest regions where detailed dietary information is not available for most frugivores.  相似文献

Invasive alien plants are a problem for conservation management, and control of these species can be combined with habitat restoration. Subsoil burial of uprooted plants is a new method of mechanical control, which might be suitable in disturbed habitats. The method was tested in Rosa rugosa (Japanese Rose), an invasive shrub in north‐western Europe with negative effects on coastal biodiversity. Two months after uprooting and burial in dunes of north‐eastern Denmark, 89% of the 58 shrubs resprouted from roots and rhizomes; on average 41 resprouts per shrub. Resprout density was twice as high at former shrub margins compared with the center; resprouts were taller and originated from more superficial soil layers at the margin than in the center. Resprouting was negatively correlated with fragment depth, and no resprouts were observed from greater than 15 cm depth. The number of resprouts increased with fragment dry mass (0.5–168.5 g). After 18 months with harrowing the species was still resprouting, flowering, and fruiting, albeit with no difference between shrub margin and center. Resprouts were taller (26 cm) and coverage was higher (0–4%) after two compared with three times harrowing, whereas no difference was found in cover of native dune species (1–5%). The results show that even small fragments of R. rugosa resprout, and that resprouting persists despite repeated harrowing. Thus, careful subsoil burial of all fragments is necessary, special attention should be paid to the shrub margin, and follow‐up treatments are needed. The effectiveness of the burial method is discussed for restoration of coastal dunes.  相似文献

Objective:  This review highlights the role of cytopathology in cancer management within UK Head and Neck Cancer Networks and informs on the issues raised by recent UK Department of Health documents and other UK professional guidance. UK guidance requires the formal involvement of cytopathologists within multidisciplinary cancer teams, with medical and non-medical cytopathology staff setting up and running rapid access lump clinics, and support for image-guided fine needle aspiration cytology (FNAC) services. UK guidance also makes recommendations for training, resources and quality control. This review also highlights the resource gap between best practice evidence-based guidance for head and neck (HN) cancer services and existing UK provision for cytopathology, as evidenced by lack of availability of experienced staff and adequacy of training and quality control (QC). Finally, it stresses the importance in the UK of the Royal College of Pathologists' guidance, which defines the need for training, the experience needed for new consultants, the requirements for audit and QC. The implications for the additional resources required for HN cancer cytopathology services are discussed. Recent professional guidance specifying the provision of HN cancer services in the UK includes a cytopathology service for cancer networks, such as rapid access FNAC clinics. Although these clinics already operate in some institutions, there are many institutions where they do not and where the provision of cytopathology services would have to be restructured. This would need the support of local cancer networks and their acceptance of the detailed requirements for cytopathology, including resources, training and QC. The standards are not defined locally, as Strategic Health Authorities and Primary Care Trusts have been instructed by the Department of Health to support, invest and implement them.  相似文献

Histones package DNA in all eukaryotes and play key roles in regulating gene expression. Approximately 150 base pairs of DNA wraps around an octamer of core histones to form the nucleosome, the basic unit of chromatin. Linker histones compact chromatin further by binding to and neutralizing the charge of the DNA between nucleosomes. It is well established that chromatin packing is regulated by a complex pattern of posttranslational modifications (PTMs) to core histones, but linker histone function is less well understood. In this review, we describe the current understand- ing of the many roles that linker histones play in cellular processes, including gene regulation, cell division, and devel- opment, while putting the linker histone in the context of other nuclear proteins. Although intriguing roles for plant linker histones are beginning to emerge, much of our current understanding comes from work in animal systems. Many unanswered questions remain and additional work is required to fully elucidate the complex processes mediated by linker histones in plants.  相似文献

Maize (Zea mays L.) and sunflower (Helianthus annuus L.) plantswere grown in large volumes of soil and leaf growth rate wasmonitored on a daily basis. Half the plants were given a soildrying treatment and when they showed a significant restrictionof growth rate (compared to both their daily growth rate beforedrying and the average growth rate of well-watered plants onthe same day), leaf water relations were measured and xylemsap was extracted using several techniques. There was a significant negative log-linear relationship betweenthe rate of leaf growth and the concentration of ABA in thexylem for both species. There was no clear relationship betweenleaf growth rate and leaf water potential or turgor for eitherspecies. Assessment of different methods for sampling xylemsap suggests that exudates collected from stem stumps or samplescollected by pressurizing the whole root system are suitablefor estimating ABA concentration in xylem, at least with largeplants of maize or sunflower, provided the first few hundredcubic millimetres of collected sap are used for the assay. Centrifugationof sections of stems resulted in dilution of ABA in the xylemsap with sap squeezed from parenchyma tissue. This is because,at least in plants subjected to mild soil drying, the concentrationof the ABA in the xylem is far higher than that in the cellsap of stem tissue. Results support the proposal that ABA plays a major role asa chemical signal involved in the root-to-shoot communicationof the effects of soil drying. The non-hydraulic restrictionof leaf growth by a chemical signal can be explained by theextra root-sourced ABA in the xylem and may be an importantcomponent of the modification of growth and development whichresults from prolonged soil drought. Accumulation and ethylene-dependent translocation of free polyamines was studied in various organs, the phloem and xylem exudates of common ice plants (Mesembryanthemum crystallinum L.). Under normal conditions (23–25°C), spermidine predominated among polyamines. Cadaverine was found in old leaves, stems, and, in large quantities, in roots. The heat shock treatment (HS; 47°C, 2 h) of intact plant shoots induced intense evolution of ethylene from leaves but reduced the leaf content of polyamines. Under these conditions, the concentration of polyamines in roots, particularly of cadaverine, increased many times. The HS treatment of roots (40°C, 2 h) induced translocation of cadaverine to stems and putrescine to leaves. An enhanced polyamine content after HS treatment was also found in the xylem and phloem exudates. The exposure of detached leaves to ethylene led to a reduction in their putrescine and spermidine and accumulation of cadaverine, which implies the ethylene-dependent formation of cadaverine and a possible relation between the HS-induced translocation of this diamine to roots and the transient ethylene evolution by leaves. To validate this hypothesis, we compared the ethylene evolution rate and interorgan partitioning of cadaverine and other polyamines for two lines of Arabidopsis thaliana: the wild type (Col-0) and ein4 mutant with impaired ethylene reception. In plants grown in light at 20–21°C, the rate of ethylene evolution by rosetted leaves was higher in the mutant than in the wild type. The content of putrescine and spermidine was reduced in mutant leaves, whereas cadaverine concentration increased almost threefold compared with the wild type. In roots, cadaverine was found only in the wild type and not in the mutant line. Our data indicate the ethylene-dependent formation of cadaverine in leaves and possible involvement of cadaverine and ethylene in the long-distance translocation of stress (HS) signal in plants.  相似文献

A Century of B Chromosomes in Plants: So What?   总被引:3,自引:0,他引:3  
BACKGROUND: Supernumerary B chromosomes (Bs) are a major source of intraspecific variation in nuclear DNA amounts in numerous species of plants. They favour large genomes, and create polymorphisms for DNA variation in natural populations. By studying Bs we can gain useful knowledge about the organization, function and evolution of genomes. There are also significant biological questions concerning the origin and structural organization of Bs, and the way in which these selfish elements can establish themselves by exploiting the replicative machinery of their host genome nucleus. SCOPE: It is a sine qua non that Bs originate from the A chromosomes, in a variety of ways. We can study their modes of drive and ask how it is that chromosomes which apparently lack genes can have control over their own drive process which leads to their survival in natural populations. Molecular cytogenetic studies are opening up new avenues of investigation. Population equilibria for B frequencies are determined by a balance between accumulation and harmful effects. Bs are also subject to meiotic loss due to polysomy and to elimination at meiosis as univalents. These balancing forces can be seen in the context of host/parasite interaction, based on a dissection of the genetic elements in both As and Bs (in maize) which interact to bring about a stable equilibrium, at least for a snapshot in time. CONCLUSIONS: Aside from their intrinsic enigmatic properties, B chromosomes make useful experimental tools to study genome organization. Thus far they have not been exploited for their applications, other than through the use of A-B translocations used for gene mapping in maize; but there are opportunities to use them to modulate the frequency and distribution of recombination, to diploidize allopolyploids, to study centromeres and to be developed as plant artificial chromosomes; given that they can be structurally modified and their inheritance stabilized.  相似文献
