采用PEG法诱导普能小麦的原生质体与新麦草、无芒雀麦及韦氏雀麦3种近缘或缘间禾草的原生质体进行对称融合。对融合再生细胞系进行形态比较、同工酶分析及5SrDNA间隔序列差异分析,以鉴定其杂种性质。对部分杂种细胞系进行了染色体组成的分析。就体细胞杂种中小染色体及染色体断片的产生与异源核质的互作及亲缘远近的关系等问题进行了讨论。  相似文献

普通小麦(TriticumaestivumL.)99P原生质体与经紫外线照射的无芒雀麦(BromusinermisLeys)愈伤组织来源的原生质体用PEG法诱导融合。融合再生的3个单细胞克隆(No.1~No.3)经染色体、同工酶和RAPD分析表明,它们均为杂种细胞系。No.1克隆形成的愈伤组织分化出大量白化苗,其叶片同工酶和RAPD分析均出现双亲特征谱带及新带,白化苗的染色体数目为42~54,大于亲本小麦,并具有无芒雀麦的小染色体和染色体断片,确认它们为不对称体细胞杂种。  相似文献

在成功培养原生质体的基础上, 用改进的PEG-高pH高钙法诱导草木樨状黄芪(Astragalus melilotoides)和木本霸王(Zygophyllum xanthoxylum)原生质体融合, 得到了科间体细胞杂种融合细胞。采用罗丹明-6G预处理草木樨状黄芪原生质体以及UV-B辐照霸王原生质体, 使双亲原生质体及其同源融合产物均不能持续分裂而死亡, 融合后的杂种细胞由于生理互补可恢复持续分裂能力而被筛选出来。融合产物经培养分裂获得了2个杂种细胞系, 其中1个分化出芽。染色体计数和分子鉴定证明了杂种的真实性。初步比较了杂种细胞系及亲本对盐分和水分胁迫的耐受性, 结果表明杂种细胞系对盐分和水分胁迫的耐受性介于两个亲本之间。  相似文献

通过PEG诱导红豆草抗羟脯氨酸细胞系原生质体与苜蓿根癌农杆菌702转化系原生质体融合,首次选择得到非对称属间体细胞杂种愈伤组织,并分化出17株小植株.PEG可诱导原生质体紧密粘连,使相连膜环化形成泡囊状结构,导致原生质体相互融合.融合诱导溶液中附加 5%~ 10%甘油可以明显提高异源融合频率.采用后滴原生质体法具有较好的结果.杂种再生芽的染色体数目处于30~60条之间,并包含红豆草的小染色体和苜蓿的2条具有2个明显缢痕的染色体.同工酶分析和胭脂碱合成酶活性检测,均证实其杂种特征.  相似文献

采用电融合法获得了骆驼刺和鹰嘴紫云英属间体细胞杂种。骆驼刺原生质体来自发根农杆菌A4转化系细胞,并经碘乙酰胺处理;鹰嘴紫云英原生质体从甲硫氨酸抗性系细胞分离。双亲及同源融合产物均不能在无激素的筛选培养基上持续分裂,融合后的杂种细胞由于双亲生理互补效应可恢复持续分裂能力而被筛选出来。本实验优化了电融合参数,如直流脉冲、交流脉冲和脉冲次数。融合产物经培养获得的杂种细胞系经形态学、染色体数目检查、生化及随机扩增多态性DNA分析,鉴定出10个杂种克隆,并从3个杂种克隆再生了小植株。  相似文献

以聚乙二醇(PEG)为诱导剂,试验用4种方法进行青菜幼嫩花粉原生质体与芥菜下胚轴原生质体的融合,结果以修改的Terada等的方法最好。融合后原生质体的培养形成了愈伤组织,但未分化出芽,个别愈伤组织分化了根。用RAPD技术对任选的5个细胞系进行鉴定,发现1个细胞系含双亲的DNA片段,可确认是青菜幼嫩花粉原生质体与芥菜下胚轴原生质体融合后产生的杂种细胞系。  相似文献

本文进行了小麦和裸燕麦悬浮细胞原生质体的电融合,并基于双亲失活(用IOA处理受体小麦原生质体,用γ-射线照射供体裸燕麦细胞系),获得可能的杂种愈伤组织。对7块愈伤组织进行了乙醇脱氢酶(Adh)同工酶筛选,发现5块表现出双亲特征酶带。对3个杂种细胞系进行5种同工酶分析,证实它们均为稳定的不对称体细胞核杂种细胞系;它们表现出小麦的完整谱带和裸燕麦的部分谱带。对2个杂种细胞系及亲本的核糖体DNA Southern分析结果表明只有一个杂种细胞系(HB 95)含有双亲的全部谱带。细胞学观察表明,2个杂种细胞系的染色体数目均显著高于双亲。从杂种细胞系HB 94中分化出叶原基等分化结构。Adh同工酶分析表明,这些分化结构具有和母体细胞系完全相同的杂种谱带。  相似文献

普通小麦与簇毛麦对称及不对称体细胞杂交的比较   总被引:4,自引:3,他引:1  
由长期继代的小麦品种济南177悬浮系制备原生质体为受体,取继代一年的簇毛麦愈伤组织不经照射或经60Co-g射线照射,剂量分别为40,60,80Gy(1.3Gy/min),然后分离原生质体为供体,在PEG诱导下分别进行了对称及不对称融合(g融合).经形态学、细胞学、同工酶及5SrDNA间隔序列分析鉴定,对称及不对称融合均高频率地获得了体细胞杂种细胞系,并在对称融合及低剂量g辐照组合得到再生杂种植株.基因组原位杂交的结果证实,对称及不对称融合杂种中异源染色体间不同方式的易位及重组均普遍存在.g辐照存在剂量效应.结果表明:对称及不对称体细胞杂交均可有效地实现核基因的转移与重组,而不对称体细胞杂交还可直接导致染色体小片段易位,显示出此方法在小麦育种上的独特优势.  相似文献

以小麦品种济南177悬浮细胞系来源的原生质体与同品系胚性愈伤组织制备的原生质体混合后作为受体;以经过380μW/cm2紫外线照射1min、2min的新麦草原生质体分别作为供体,用PEG法诱导融合。组合Ⅰ(176+cha9+新麦草UV 1min)获得16个再生克隆。经过形态学、同工酶、染色体和RAPD分析,确定其全部为属间体细胞杂种。其中的5个克隆再生杂种植株。用7对小麦SSR引物对杂种克隆的叶绿体基因组进行了分析;组合Ⅱ(176+cha9+新麦草UV 2min)只获得3个克隆,且逐渐褐化死亡。表明以小麦济南177的两种培养细胞混合作受体的融合体系有利于杂种的获得及再生;紫外线对融合产物的生长发育有明显的剂量效应。  相似文献

Asymmetric hybridization was conducted between wheat and Brorrats inermis keyss which is a distanfiy related intergeneric plant (belonging to different tribe) of wheat and possesses some favorable traits, such as resistant to cold, drought and disease. Protoplasts isolated from young embryo-derived calli of common wheat ( Triticura aestivum L., tv. 99P, (AABBDD), 2n = 42) were fused with UV-treated protoplasts isolated from young embryo-derived calli of Bromus inermis by PEG method. Three clones (No. 1 ~ No. 3) were regenerated from the fusion products and differentiated into albino seedlings. The clones and the seedlings were all verified as hybrids by chromosome counting, isozyme and RAPD analysis. Their isozyme and RAPD pattern contained the characteristic bands of both parents as well as new band(s). The chromosome numbers of albino were in the range of 42~54 with small chromosomes of Bromus inerm/s and chromosome fragments. The above results confirmed that hybrid albinos were obtained.  相似文献

Two types of protoplasts of wheat (Triticum aestivum L. cv. Jinan 177) were used in fusion experiments—cha9, with a high division frequency, and 176, with a high regeneration frequency. The fusion combination of either cha9 or 176 protoplasts with Russian wildrye protoplasts failed to produce regenerated calli. When a mixture of cha9 and 176 protoplasts were fused with those of Russian wildrye, 14 fusion-derived calli were produced, of which seven differentiated into green plants and two differentiated into albinos. The morphology of all hybrid plants strongly resembled that of the parental wheat type. The hybrid nature of the cell lines was confirmed by cytological, isozyme, random amplified polymorphic DNA (RAPD) and genomic in situ hybridization (GISH) analyses. GISH analysis revealed that only chromosome fragments of Russian wildrye were transferred to the wheat chromosomes of hybrid calli and plants. Simple sequence repeat (SSR) analysis of the chloroplast genome of the hybrids with seven pairs of wheat-specific chloroplast microsatellite primers indicated that all of the cell lines had band patterns identical to wheat. Our results show that highly asymmetric somatic hybrid calli and plants can be produced via symmetric fusion in a triparental fusion system. Morphological changes in interphase nuclei were cytologically studied in heterophasic dinucleate cells formed by the fusion of mitotic and interphase plant protoplasts. Mitotic protoplasts were isolated from a partially synchronized suspension culture of wheat (Triticum monococcum). The mitotic cells were accumulated by colchicine after release of hydroxyurea block. Treatment of protoplast populations with polyethylene glycol-dimethyl sulphoxide solution resulted in metaphase-interphase fusion. Three hours after fusion, the appearance of chromosomes with single chromatid as well as of fragmented, pulverized chromatin in heterophasic cells indicated the induction of premature chromosome condensation (PCC) in somatic wheat cells. Condensation in interphase nuclei of mitotically inactive rice protoplasts was also detected after fusion with mitotic wheat protoplasts.  相似文献

The suspension derived protoplasts of wheat (Triticum aestivum) cv. Jinan 177 were used as a recipient to fuse with the protoplasts of the 60Co gamma-ray irradiated calli of Legmus chinensis. The wheat suspension cells and their protoplasts were not capable of differentiating to whole plants. The irradiated calli of L. chinensis were also the same. The protoplasts originated from the treated or untreated calli were both unable to divide under the conditions of this experiment. However, the fusion products grew and developed to whole plants which were identified as hybrids according to the analysis of chromosome, isozyme and morphology. The above result revealed that the lost regeneration capacity of both parents could be complementarily restored through somatic hybridization. This phenomenon also occurred with our work on Triticum aestivum (+) Haynaldia villosa, T. aestivum (+) Agropyron elongatum and T. aestivum (+) Psathyrostachys juncea.  相似文献

以失去植株再生能力的小麦单倍体愈伤组织和羊草二倍体愈伤组织为材料,游离原生质体,并用紫外线自理2羊草原生质体,用PEG法融合,对融合克隆进行染色体和同工酶分析,在已贩26个克隆中有21个是杂种,其中有一个克隆再生出短命小植株,结果 体小麦与二倍体草的不对称融合虽然再生互补效应不如二倍体小麦,然而杂种优先生长的现象仍然比较明显。如果改善实验条件和双亲原始的再生能力,这种融合方式仍然可以利用。  相似文献

Suspension cell-derived protoplasts of wheat, inactivated with different concentrations (0-2.5mol/L) of IOA, were fused by PEG method with the Haynaldia villosa protoplasts which originated from the calli 4-5d after subculture and were irradiated with 60Co-γ ray. Cell colonies, calli or regenerated plants were obtained from different combinations of fusion. The calli and plants were verified to be hybrids by chromosome counting, isozyme analysis and morphological inspection.  相似文献

To explore the feasibility of constructing a whole genome radiation hybrid (WGRH) map in plant species with large genomes, asymmetric somatic hybridization between wheat (Triticum aestivum L.) and Bupleurum scorzonerifolium Willd. was performed. The protoplasts of wheat were irradiated with ultraviolet light (UV) and gamma-ray and rescued by protoplast fusion using B. scorzonerifolium as the recipient. Assessment of SSR markers showed that the radiation hybrids have the average marker retention frequency of 15.5%. Two RH panels (RHPWI and RHPWII) that contained 92 and 184 radiation hybrids, respectively, were developed and used for mapping of 68 SSR markers in chromosome 5A of wheat. A total of 1557 and 2034 breaks were detected in each panel. The RH map of chromosome 5A based on RHPWII was constructed. The distance of the comprehensive map was 2103 cR and the approximate resolution was estimated to be ~501.6 kb/break. The RH panels evaluated in this study enabled us to order the ESTs in a single deletion bin or in the multiple bins cross the chromosome. These results demonstrated that RH mapping via protoplast fusion is feasible at the whole genome level for mapping purposes in wheat and the potential value of this mapping approach for the plant species with large genomes.  相似文献

Summary The cytology of bread wheat (Triticum aestivum) suspension lines, recycled lines (selected for high division frequency) and their dividing protoplasts, have been examined. Extensive numerical and structural chromosome variation was present in all the lines. The most frequently observed chromosome numbers were around 2n=32, indicating that considerable chromosome loss from the normal wheat complement (2n=6x=42) had occurred during selection of the lines. Chromosome aberrations also indicated loss of chromosome arms and chromosome segments. The implications of this variation for studies on transformation and for the potential regeneration of whole plants from protoplasts of bread wheat are discussed.  相似文献

Forty-five Iranian isolates of Xanthomonas campestris obtained from wheat, barley and grasses were compared with reference strains using polyacrylamide gel electrophoresis (PAGE) of the whole-cell and membrane proteins.
The PAGE profiles of the whole-cell and membrane proteins of the Iranian isolates obtained from barley, with the exception of IBLS 11 and IBLS 12, were identical and clearly distinguishable from those of the other isolates. The barley group isolates, which were pathogenic only to barley, were similar to UPB 458 (NCPPB 2389), the reference strain of pathovar hordei. The isolates obtained from wheat and grasses, as well as IBLS 11 and IBLS 12, which can infect wheat, barley and some wild grasses, had similar banding patterns: only IBLS 40 isolated from Hordeum sp. displayed the same profile as the barley group. Reference strains UPB 443 (NCPPB 2821) and UPB 513, which correspond to pathovars undulosa and translucens , respectively, were related to the wheat group. IBLS45, isolated from Bromus sp., had a banding pattern that differed from those observed for strains of the barley and wheat groups.
The results suggest that this method can be useful for discriminating different pathovars of X. campestris attacking cereals and grasses, and sodium dodecyl sulphate (SDS)-PAGE of membrane proteins was not more sensitive than SDS-PAGE of total proteins for differentiating the isolates.  相似文献
