Effects of sodium tungstate on the nuclear uptake of rat liver cytosolic glucocorticoidreceptor complex were examined at pH 7. The nuclear uptake of heat-activated [3H]triamcinolone acetonide-receptor complex was blocked completely in the presence of 1 mm tungstate. A preincubation of nuclear preparation with tungstate (>0.1 mm) blocked the subsequent uptake of [3H]triamcinolone acetonide-receptor complex. When the tungstate-treated nuclear preparation was washed with 0.3 M KCl, its [3H]triamcinolone acetonide-receptor complex binding capacity recovered to 50% of that of control samples with no tungstate treatment. A preincubation of chromatin with tungstate yielded similar results. The nuclear-bound [3H]triamcinolone acetonide-receptor complex, formed either by an in vivo administration of [3H]triamcinolone acetonide or by an in vitro incubation of glucocorticoid-receptor complex with isolated nuclei, was extracted by tungstate in a concentration-dependent manner. The majority of nuclear-bound [3H]triamcinolone acetonide could be extracted with 0.1 and 1 mm tungstate from in vitro- and in vivo-labeled nuclei, respectively. The tungstate-extracted steroid-receptor complexes sedimented in 4–5 S and 3.3–3.5 S region in 10 mm KCl- and 0.3 mm KCl-containing sucrose gradients, respectively. Tungstate treatment caused an irreversible loss of the nuclear binding capacity of [3H]triamcinolone acetonide-receptor complex which could not be recovered after dialysis. These studies indicate that tungstate affects both glucocorticoidreceptor complex and certain nuclear or chromatin proteins.  相似文献

The feeding habits of Helicolenus hilgendorfii were studied using 93 specimens (35–135 mm in standard length) collected from the southeastern coast of Korea during the summer season (June–August) from 2005 to 2007. Helicolenus hilgendorfii is a carnivore that mainly consumes shrimps and fishes. Its diet also includes small quantities of mysids, amphipods, euphausiids, copepods, and isopods. Our graphical method for feeding habits revealed that H. hilgendorfii is an opportunistic and specialized predator on shrimps, especially Crangon hakodatei, although H. hilgendorfii showed an ontogenetic dietary shift, the proportion of shrimp decreasing and the consumption of fishes gradually increasing with the increase of fish size.  相似文献

The present study reports weight-length (W-L) relationships and discusses the relative growth of the banded murex (Hexaplex trunculus) from intertidal and offshore areas of the Gulf of Gabès (southern Tunisia). Overall, 9634 H. trunculus were sampled (6608 from intertidal and 3026 from offshore), with both populations comprising broad ranges in shell length (intertidal: 13.6–78.5 mm; offshore: 22.9–91.1 mm) and total weight (intertidal: 0.2–63.8 g; offshore: 0.6–92.5 g). In both populations, W-L relationships were highly significant and the morphometric parameters were highly correlated (intertidal: r?=?0.967; offshore: r?=?0.973). Relative growth was significantly different between populations, with intertidal H. trunculus displaying a negative allometry (b?=?2.962) and offshore H. trunculus exhibiting isometric growth (b?=?3.013). Factors influencing total weight (soft-body weight and shell weight) and potentially responsible for differences in W-L relationships and relative growth between populations were discussed. In addition, a useful comparative analysis for fisheries biology, assessment and management purposes was performed with analogous information from other populations of H. trunculus from the Mediterranean Sea and the Atlantic Ocean.  相似文献

Taxonomic and phylogenetic studies of Haploporus were carried out. Three species in Haploporus, H. cylindrosporus, H. septatus and H. subpapyraceus, are described as new based on morphological differences and molecular phylogenetic analyses inferred from the internal transcribed spacer (ITS), the large subunit nuclear ribosomal RNA gene (nLSU), the small subunit mitochondrial rRNA gene (mtSSU), the second subunit of RNA polymerase II (rpb2) and the translation elongation factor 1-α gene (TEF1) sequences. Haploporus cylindrosporus is characterized by big irregular crystals occasionally present in the subiculum, an abundant oily substance among hyphae and typically cylindrical basidiospores 10–11.5?×?4.5–5 μm; H. septatus differs from other species in the genus by its leathery to corky basidiomata when dry, small round pores (5–6 per mm), simple septate skeletal hyphae at the edge of the dissepiments, and oblong to ellipsoid basidiospores 8.5–11?×?5–6 μm; H. subpapyraceus is separated by white to cream basidiomata, numerous apically simple septate cystidioles and ellipsoid basidiospores 9–12?×?5.5–8 μm. The effect of sodium butyrate on Tipula iridescent virus (TIV) synthesis in suspension-cultured cells of Estigmene acrea was investigated. Sodium butyrate reduces viral-induced cell fusion but this is reversible with the removal of butyrate. At 7 mM sodium butyrate, TIV replicates in cells within 8 hr, but does not replicate in this time with 10–20 mm butyrate in the cell medium; cells so treated contain large vesicles with inoculum. Upon removal of the inhibitor, TIV replication appears normal, but large inoculum vesicles can still be found in the cytoplasm, and many infected cells have highly condensed chromatin in their nuclei. Sodium butyrate causes a lag of at least 2 hr in viral DNA synthesis as detected by [3H]thymidine incorporation into viroplasmic centres and at 7 mm butyrate viral DNA synthesis is reduced by 50–60%. In comparison, butyrate at 7 and 10 mm concentration does not inhibit host DNA synthesis, but at 15 and 20 mm, nuclear DNA synthesis is markedly reduced.  相似文献

A new species, Huernia humpatana Bruyns (Apocynaceae–Ceropegieae), closely related to H. similis N.E.Br., is described from the Chela Mountains of Huila Province in southern Angola. The two species are distinguished by the 5-angled and erect stems with more prominent tubercles up to 6 mm long joined into clear angles and separated by V-shaped grooves in H. humpatana as opposed to very obtusely 4-angled stems with tubercles only 2 mm long and only indistinct grooves between the angles in H. similis. Furthermore, in H. similis the nodding corolla is ± 9 mm in diameter with sepals ± 2 mm long, while in H. humpatana the horizontally facing corolla is 18–20 mm in diameter with sepals 4–6 mm long.  相似文献

Helminthosporium oryzae, a fungal pathogen causing Brown Spot of Rice is one of the disastrous biotic factors responsible for 16–43% yield in rice. To control in vitro fungal growth of this pathogen, four fungicides viz. Mencozeb, Thiophanate Methyl, Iprovalicarb + Propineb and Propineb were used with four different concentrations: 50, 100, 150 and 200 ppm. Plant extracts of five species as Azadirachta indica, Calotropisprocera, Allium sativum, Datura stramonium and Zingiber officinale were also tested with three doses (5, 10 and 15 ml). While, 24 isolates of Bacterial isolates were investigated for their antagonistic effect. The in vitro results of the four fungicides contributed that Mancozeb and Thiophanate Methyl inhibited the colony growth at the higher doses (150 and 200 ppm) with no linear colony growth. However, Propineb was found moderately effective at (5.00 mm) and Iprovalicarb + Propineb was less effective (7.25 mm). Similarly, the in-vitro efficacy of the five plant extracts against H. oryzae at different doses revealed that Zingiber officinale and Allium sativum were more effective at high doses (00.00 mm, 2. mm). Also, Datura stramonium (4.62 mm) Azadirachta indica (18.00 mm) and Calotropisprocera (12.00 mm) performed better as compared to the control (40.00 mm). The results revealed that out of 24 isolates, 4 isolates of bacteria Agrobacterium spp, Xanthomonas, Erwinia, Streptomyces has inhibited the linear colony growth of H. oryzae up to 1-2 mm. This study can be useful for controlling the fungal disease as the two fungicides Mancozeb and Thiophanate Methyl were more effective and the Zingiber officinale and Allium sativum plant extracts were highly effective against the brown spot of rice. Biocontrol also reduced leaner colony Helminthosporium oryzae.  相似文献

A new species,Heliotropim gossypii Ponert, is described from N.E. Iran. Observations made in the type locality suggest that this new species may be a facultative cotton weed. The following diagnostic characters separateH. gossypii fromH. noëanum Boiss.,H. sultanense Bunge andH. chorassanicum Bunge: branching of inflorescence, calyx persistent, up to 3.5 mm long corolla less than 2 mm in diameter, anthers 0.85 mm long, superstigmatic part of style erecto-pilose throughout, nutlets 2 mm long, shortly pilose.  相似文献

The effect of media supplements on total and polyglutamylfolate concentrations has been examined in Neurospora crassa wild type (FGSC 853), an ethionine-resistant mutant (FGSC 1212), and a methionine auxotroph (FGSC 1330) which lacks folylpolyglutamate synthetase. When the culture medium contained 1 mm glycine, folate concentrations in the wild type were increased by over 90% and more p-[3H]aminobenzoate was incorporated into folates. Growth in l-methionine-supplemented media (1–5 mm) decreased folate levels and labeling in all three strains. In the wild type, this effect of l-methionine was reversed on transfer to unsupplemented media but p-[3H]aminobenzoate pulse-chase experiments suggested that exogenous methionine did not increase the turnover of labeled folates. At 1 mm, d-methionine did not affect polyglutamylfolate labeling but l-methionine reduced 3H incorporation by 65% in the wild type. Ion-exchange chromatography showed that p-[3H]aminobenzoate was incorporated in formyl- and methyltetrahydrofolates which in the wild type, were principally hexaglutamyl derivatives. Glycine-supplemented growth yielded labeled folates that were 24% heptaglutamates but these and pentaglutamates were lacking when l-methionine was supplied. The specific activity of GTP cyclohydrolase was not significantly affected by culture in l-methionine-containing media. Dialysis and gel filtration both lowered enzyme activities and product formation was not changed when up to 10 μmol of l-methionine was added to the reaction system. The data suggest that methionine or its metabolic products exerts some control over folate production which is distinct from the established inhibition of methylenetetrahydrofolate reductase by AdoMet.  相似文献

Taxonomic studies including morphological observations and phylogenetic analyses were conducted on Japanese “uragin-take”, an unidentified species from Amazonia, Brazil and their allies. Phylogenetic analyses using ITS, nrLSU and RPB2 regions revealed that “uragin-take”, Neofomitella polyzonata and the unidentified species formed a monophyletic clade separate from the clade including the other four Neofomitella spp. and that “uragin-take” is conspecific with N. polyzonata. Morphological investigations on authentic specimens revealed that Polyporus subradiatus is a prior name for N. polyzonata. We propose Hirticrusta gen. nov. typified by H. subradiata segregated from Neofomitella, and we erected H. amazonica sp. nov. for the unidentified species. Hirticrusta is characterized by annual to biennial and sessile basidiocarps, semicircular to dimidiate pileus, velutinous to tomentose hairs on pileus surface, buff to brown context with a crustose layer indicated by a dark brown line forming a longitudinal section below the superficial hairs, a trimitic hyphal system, crustose layer composed of parallel and densely arranged brown hyphae and cylindrical basidiospores. The new species, H. amazonica is distinguishable from other polypores by downy and long tomentum on the pileus surface (up to 20 mm thick), brown context with a dark brown layer below the tomentum and round pores (5–7/mm).  相似文献

The cooperativity effects between the O/N–H???F anionic hydrogen-bonding and O/N–H???O hydrogen-bonding interactions and electrostatic potentials in the 1:2 (F:N-(Hydroxymethyl)acetamide (signed as “ha”)) ternary systems are investigated at the B3LYP/6-311++G** and MP2/6-311++G** levels. A comparison of the cooperativity effect in the “F???ha???ha” and “FH???ha???ha” systems is also carried out. The result shows that the increase of the H???O interaction energy in the O–H???O–H, N–H???O–H or N–H???O?=?C link is more notable than that in the O–H???O?=?C contact upon ternary-system formation. The cooperativity effect is found in the complex formed by the O/N–H???F and O/N–H???O interactions, while the anti-cooperativity effect is present in the system with only the O/N–H???F H-bond or the “FH???ha???ha” complex by the N???H–F contact. Atoms in molecules (AIM) analysis and shift of electron density confirm the existence of cooperativity. The most negative surface electrostatic potential (V S,min ) correlates well with the interaction energy E int.(ha???F–) and synergetic energy E syn., respectively. The relationship between the change of V S,min (i.e., ΔV S,min ) and E syn. is also found.

The stinging catfish, Heteropneustes fossilis (Bloch, 1974) is a commercially important fish species in Asia. This is an important food fish as is enriched with high amounts of protein, iron and calcium. The current research demonstrates the population structure, size at sexual maturity, spawning- and peak-spawning season and fecundity of H. fossilis in an important wetland ecosystem - Gajner Beel in northwestern Bangladesh with an aim of its sustainable conservation through induced breeding and aquaculture practices. A total of 426 stinging catfish captured from the Gajner Beel through monthly sampling from January to December 2019 used in the study. Total length (TL), standard length (SL) and body weight (BW) of individual fishes were measured. The size ranges were with 6.70–24.10 cm TL, 1.37–83.94 g BW. Gonads were removed carefully through ventral dissection and weighted. Lm was 14.02, 13.5, 13.0 and 15.0 cm based on maximum length (Lmax), TL vs. GSI (%), TL vs. SL and logistic model, respectively. Monthly variations of GSI and maturation stages were confirmed in April to August as spawning season and June as peak spawning month. Fulton’s condition factor (KF) was found to be with significant relations with GSI values. Fecundity was 1,730 to 23,870 and significantly correlated with both TL and BW. Temperature has been increasing 0.029 °C/year with the falling of rainfall at 2.96 mm/year in the study area. Environmental factors -Temperature, rainfall, dissolved oxygen and pH were found to be significantly correlated with GSI. We found the optimal range of temperature (29–31 °C), rainfall (350–380 mm), dissolved oxygen (5.0–6.0 mg/l) and pH (7.1–7.5) for spawning of H. fossilis. The paper recommended the policy guidelines to pave the ways of the aquaculture, conservation and management of H. fossilis in the changing eco-climatic events through specific management measures.  相似文献

Hysterothylacium simile n. sp., collected from the Japanese seabass Lateolabrax japonicus (Cuvier) (Perciformes: Lateolabracidae) in the Bohai Sea off China, is described using both light and scanning electron microscopy. The new species differs from its congeners in the presence of narrow lateral alae originating a short distance posterior to the base of the ventrolateral lips, a long intestinal caecum (60.4–79.1% of oesophageal length) and a relatively short ventricular appendix (intestinal caecum to ventricular appendix ratio 1:0.58–0.85), long spicules (2.11–2.99 mm, 4.25–7.83% of body length), the number and arrangement of the caudal papillae (32–36 pairs arranged as follows: 27–31 pairs precloacal, 1 pair paracloacal, and 4–5 pairs postcloacal with the second or third pair double) and the presence of a particular midventral precloacal papilla. Specimens originally identified as Contracaecum paralichthydis Yamaguti, 1941 [now H. paralichthydis (Yamaguti, 1941)] by Xü (1957), collected from the yellow striped flounder Pseudopleuronectes herzensteini (Jordan & Snyder) (Pleuronectiformes: Pleuronectidae) in the Yellow Sea off China, were also re-examined. Their morphology clearly revealed they belong to H. aduncum (Rudolphi, 1802), which is also redescribed based on Xü’s material. In addition, the morphological variation of caudal papillae in H. aduncum from P. herzensteini was compared, using scanning electron microscopy, with specimens collected from another three fish hosts, Lophius litulon (Jordan) (Lophiiformes: Lophiidae), Scomberomorus niphonius (Cuvier) (Perciformes: Scombridae) and Cleisthenes herzensteini (Schmidt) (Pleuronectiformes: Pleuronectidae), from the Yellow Sea off China.  相似文献

We used computer-generated stimuli to assess the stimulus parameters eliciting prey-directed behaviors in four mantis species: Rhombodera basalis (60 mm body length), Deroplatys lobata (70 mm), Hierodula membranacea (80 mm), Miomantis sp (38 mm). All responded appetitively to erratically moving disks subtending 10° or more. R. basalis responded more consistently to checkered (versus solid black) disks; only D. lobata displayed a size preference (14–23 deg disks); Miomantis sp. struck at disks as large as did the largest species; only Miomantis sp. displayed approaching behavior. H. membranacea and R. basalis preferred black disks on a white background (versus the reverse), and red disks (versus blue or green) on a brightness-matched grey background. All preferred elongated stimuli moving parallel (versus perpendicular) to their long axes.  相似文献

Psidium guayaquilense from western Ecuador is described and illustrated. It is most similar to P. acidum, having in common with it non-angular, rounded seeds, anthers with a few subequal glands and peduncles with wings. Psidium guayaquilense differs from P. acidum in various ways, most notably in the principal marginal veins mainly 1–3 mm from margin (versus 3–10 mm or more from margin in P. acidum); closed calyx with a prominent apiculum (versus without an apiculum or with only a weak apiculum); ovules per locule 12–40 (versus 50–70); style 8–10 mm long (versus 12–16 mm); fruit 1.5–3 cm in diameter (versus 2–5 cm); fruit wall 0.5–2.5 mm thick (versus 4–13 mm); seeds usually 4–6 (versus 20–60 or more).  相似文献

Protamine sulfate was found to precipitate completely the nonactivated [3H]-dexamethasone-receptor complex of rat liver. This observation was then used as the basis of a method to separate activated from nonactivated complex. Thus, addition of 10 mg/ml of protamine sulfate to the rat hepatic cytosol [3H]dexamethasone-receptor complex, incubated at 0–4°C for 2 hr, resulted in the complete precipitation of [3H]dexamethasone-receptor complex. The remaining supernatant obtained on centrifugation at 800g was unable to bind either to nuclei or to DNA-cellulose. An increase in temperature to 25°C or the addition of 10 mm CaCl2 to the cytosol resulted in the appearance of activated [3H]dexamethasone-receptor complex in the supernatant obtained by addition of protamine sulfate. This was determined by characteristic binding to nuclei or DNA cellulose and by pI. Protamine sulfate could not affect the separation of activated [3H]dexamethasone-receptor complex at salt concentrations above 100 mm NaCl. This procedure therefore had to be carried out under conditions of relatively low ionic strength. Finally, a one-step rapid method is described for the separation of activated [3H]dexamethasone-receptor complex from nonactivated receptor complex. The homogeneous population of activated complex thus obtained should have considerable applicability in studies of the mechanisms of in vitro glucocorticoid-receptor activation.  相似文献

Halichoeres bleekeri (Steindachner &; Döderlein, 1887), previously regarded as a junior synonym ofH. tenuispinis (Günther), is a valid species. It is distinct fromH. tenuispinis in nearly always having 13 instead of 14 pectoral rays. 11–15 instead of 7–11 suborbital pores, longer dorsal soft rays (1.95–2.3 in head length vs 2.25–2.4 fortenuispinis). and some features of color such as the absence of a dark spot at the upper base of the caudal fin of the female.H. tremebundus Jordan and Snyder, 1902 is a synonym ofH. bleekeri based on the female form, andArtisia festiva de Beaufort is a synonym ofH. tenuispinis, also described from the female phase,H. bleekeri is known from Korea and in Japan from Tokyo to the Izu Islands.H. tenuispinis is recorded from Hong Kong and Xiamen. The effect of high (H) or low (L) levels of food intake, during the preceding 4 weeks, on ovarian follicle numbers and steroidogenic capacity were investigated in groups of 12 adult Scottish Blackface ewes. Ewes of the two treatments had similar levels of body condition at the time of study but there was a twofold difference in levels of food intake. Ovaries were surgically removed on day 11 or 12 of the oestrous cycle (luteal phase; n=6 per nutritional treatment) or at 30 h after injection (i.m.) of prostaglandin F analogue on day 11 or 12 of the cycle (follicular phase; n=6 per nutritional treatment). Ovarian follicles >1 mm diameter were dissected out and incubated individually for 2 h at 37°C, in 1 ml of medium 199 which was then assayed to determine concentrations of oestradiol and testosterone. There were significantly more small follicles (1–2.5 mm diameter) in (H) than (L) ewes (P<0.05) but no treatment difference in the numbers of large follicles (>2.5 mm diameter) during either phase of the cycle and no difference in the mean diameters (mm) of the two largest follicles in each animal. However, although there were higher rates of synthesis of both oestrogen (P<0.05) and testosterone (P<0.01) in the large follicles of (L) ewes as compared with (H) ewes, there was a lower oestrogen/testosterone ratio in (L) than (H) follicles which may indicate a lower level of aromatase activity in (L) follicles. It is concluded that the effects of level of food intake on ovulation rate are expressed through differences in late stages of follicle development, probably through effects on the intrafollicular steroid milieux.  相似文献

Ninety percent of rat pulmonary angiotensin-converting enzyme was solubilized in a single step using the nonionic detergent 1-O-n-octyl-β-d-glucopyranoside. This detergent has low absorbance at 228 nm and, thus, is compatible with the commonly used spectrophotometric assay of D. W. Cushman and H. S. Cheung (1971, Biochem. Pharmacol.20, 1637–1648). The maximum solubilization occurred with 30 mm 1-O-n-octyl-β-d-glucopyranoside and at this concentration of detergent a four-fold increase in specific activity was observed.  相似文献

Pathogenicity ofHeliothis nuclear polyhedrosis virus (HSNPV) to the corn earworm,Heliothis armigera, was studied using 3 different inoculative methods. The LD50 values of 4th-instar larvae inoculated with corn-fed, diet-fed and inoculum-imbiding method were 1.85×106, 2.55×105 and 1,22×103 PIBs/larva, respectively. The inoculum-imbiding is more sensitive and convenient for inoculatingH. armigera with HSNPV. The HSNPV product, Elcar®, was highly pathogenic toH. armigera, the LD50 values of 2nd-, 3rd- and 4th-instar larvae being 27, 83 and 1,221 PIBs/larva, respectively, as measured by the inoculum-imbiding method. The mortality of 4th-instar larvae caused by HSNPV was increased, but the incubation period was shortened with higher incubation temperatures. However, the high temperature at 35°C caused a lower mortality, and a prolongation of the median lethal time (LT50). Stability and persistence of HSNPV preparations were better in January–February and April–May than in June–July and October–November periods when sprayed on corn silks under field conditions. The HSNPV was inactivated by weak alkaline dew (pH 8.1) collected from soybean leaves, but it remained active on those from corn, tomato and asparagus with pH 7.2–7.3. The artificial heavy rainfall of 242 mm/h for 30 min did not wash off HSNPV preparations sprayed on the corn silks.  相似文献
