Tuber yield in open-pollinated (OP) offspring of potato cultivars appears to be correlated with the proportion of hybrids within each population. OP true potato seed (TPS) from ten selected clones was tested in an experiment at two Peruvian locations, San Ramón (mid-altitude warm tropics) and Huancayo (highland cool tropics), to investigate the early selection effect on OP populations. Another experiment in La Molina (dry coast) was included to confirm the results from these other two locations. The results clearly showed that plant survival, tuber yield and berry number can be increased by a strong early selection. Likewise, the results suggested that the most-promising OP offspring for TPS production could be derived from tetraploid clones with a high rate of outcrossing. This experiment demonstrates that high tuber yield may be obtained in OP TPS cultivars with little effort, using early selection for seedling vigor in the nursery. Genetic interpretation of this response to early selection indicates that both the intensity of selection and non-additive genetic variation for tuber yield account for these observed gains.  True potato seed (TPS) may be an alternative method of potato production in developing countries. A breeding method for the sexual propagation of this vegetatively propagated crop should consider the development of parental lines and the type of cultivar to be released. Open-pollinated (OP) cultivars seem to be an inexpensive procedure to produce potato from true seed. However, OP progenies are the result of selfing and outcrossing in male-fertile tetraploid potatoes. The aim of the present research was to establish the effect of inbreeding and open pollination in TPS. Ten Andigena clones were used as parental material to derive hybrid (S0), inbred (S1 and S2), and open-pollinated (OP1 and OP2) generations. Significant differences among generations were found for pollen production, pollen viability (as determined by its stainability with aceto-carmine glycerol), number of flowers and berries plant-1, number of seeds berry-1, weight of 1000 seeds, and tuber yield plant-1. The parental populations were significantly different for most of the traits, but not for flower production and berry weight. The interaction of population ×generation was significant for pollen and seed production as well as for weight for 1000 seeds. All the traits evaluated except seed weight showed a strong inbreeding depression, while the OP progenies had intermediate values between the S0 and the S1. Summary The potential of tuber disc culture for chromosome doubling was investigated in somaclonal populations of four dihaploid genotypes and one tetraploid cultivar of potato (Solanum tuberosum). Laser scanning confocal microscopy (LSCM) was used for rapid determination of the ploidy level based on the number of chloroplasts in stomatal guard cells of leaves. Factorial analysis of chloroplast number in 58 clones and two leaf types showed that somaclones were clearly divided in two groups. Clones with 5–7 chloroplasts per cell as observed in tuber derived diploid controls were classified as 2X (not doubled), while those with 9–14 chloroplasts resembled the tuber derived tetraploid controls and were considered 4X (doubled). A high frequency of spontaneous chromosome doubling, 42% – 50%, was detected in 3 dihaploid genotypes, whereas no doubling was observed in one of the dihaploids as well as the tetraploid cultivar Yukon Gold. Effects of leaf type on chloroplast number was also significant. The middle leaf showed significantly higher chloroplast number than the younger leaves.  相似文献

Three doubled dihaploid potatoes were hybridised with six tetraploid cultivars and the offspring examined for commercially important characters. Doubled dihaploids used as parents did not reduce the within progeny phenotypic variation for most characters below that found in material produced by intercrossing heterozygous tetraploids. They could however prove useful for improving some characters in a breeding programme. The products of the hybridisations were used to examine the genetic control of discrete variables such as tuber skin pigmentation, tuber flesh colour, and skin russetting. Doubled dihaploids were found to be useful for such studies because they have only three out of the five possible tetraploid genotypes. Presence or absence of red tuber skin in the progenies of cv. Désirée was concluded to be due to the segregation of a dominant major gene which was simplex in the parental cultivar. The yellow flesh of Desiree was found to be recessive. On the hypothesis that russet skin was due to three complementary dominant genes, possible genotypes of the doubled dihaploids and cultivars could also be deduced.  相似文献

Somatic hybrids of Solanum nigrum (+) 2× potato were successfully crossed with S. nigrum and with potato. First and second backcross progeny with S. nigrum could easily be obtained. One of the BC1 genotypes was already self-fertile. Backcrosses with potato had a much lower success rate. Only pollinations with tetraploid potato resulted in seed-containing berries. Two BC1 genotypes were obtained after 4362 pollinations from which 505 ovules were cultured. The first BC1 genotype grew vigorously in vitro and in the greenhouse and flowered abundantly. The second BC1 showed many abnormalities and dropped its flowers before anthesis. The first BC1 was again crossed with tetraploid potato and in this generation also the success rate was low. Over 5000 pollinations resulted in 1750 berries from which over 3000 ovules were obtained. Twelve plants germinated from these ovules and they were not as vigorous in vitro and in vivo as the BC1 parent. Some of the BC2 genotypes were used for further backcrosses but no BC3 plants were obtained. BC1 and BC2 genotypes that resulted from the backcross program with potato were tested for resistance to Phytophthora infestans. The BC1 genotype was as resistant as the S. nigrum fusion parent, but among the eight BC2 genotypes scored six were resistant, whereas two genotypes showing lesions were susceptible. Broadening the genetic base for resistance to Russian wheat aphid, Diuraphis noxia (Mordvilko) (Homoptera: Aphididae), in bread wheat, Triticum aestivum L., is desirable. To date, identified Russian wheat aphid resistance genes are either located to the D chromosomes or to rye translocation of wheat, and resistance derived from the A or B genomes of tetraploid Triticum spp. would therefore be highly beneficial. Fifty-eight synthetic hexaploid wheat, derived from interspecific crosses of Triticum dicoccum Schrank. and Aegilops tauschii (Coss.) Schmal. and their parents were evaluated for resistance to Russian wheat aphid under field conditions. Plots infested with aphids were compared with plots protected with insecticides. The T. dicoccum parents were highly resistant to Russian wheat aphids, whereas the Ae. tauschii parents were susceptible. Resistance levels observed in the synthetic hexaploids were slightly below the levels of their T. dicoccum parents when a visual damage scale was used. but no major resistance suppression was observed among the synthetics. Russian wheat aphid infestation on average reduced plant height and kernel weight at harvest in the synthetic hexaploids and the T. dicoccum parents by 3-4%, whereas the susceptible control 'Seri M82' suffered losses of above 20%. Because resistance in the synthetic hexaploid wheat is derived from their T. dicoccum parent, resistance gene(s) must be located on the A and/or B genomes. They must therefore be different from previously identified Russian wheat aphid resistance genes, which have all been located on the D genome of wheat or on translocated segments.  相似文献

Differences in tuber initiation, tuber growth and partitionof assimilate during development were investigated in six localsweet potato cultivars. Study of the quantitative morphogenesisof root types in the sweet potato root system indicated thattuber initiation was completed by 8 weeks after planting inmost cultivars and frustrated thereafter. Subsequent differencesin patterns of tuber development were not obviously relatedto final tuber yield. Thus, high yield resulted either froma short period of rapid tuber growth or a longer period of slowertuber growth. Maximum shoot growth was achieved by week 12 inall except one cultivar, and the cessation of shoot growth resultedin either increases or decreases in tuber growth rate or hadno affect on the rate of tuber growth (cv. A28/7). The partitionof assimilate to tubers at final harvest was the parameter mostclosely related to the yield of the six cultivars studied, butthere was some evidence that such partition was related to totaldry weight. It is suggested that limiting factors in sweet potatotuber yield, e.g. assimilate production and transport or capacityfor tuber growth varied with the cultivar studied.  相似文献

Interval mapping of quantitative trait loci (QTL) for 16 yield, agronomic and quality traits in potato was performed on a tetraploid full-sib family comprising 227 clones from a cross between processing clone 12601ab1 and table cultivar Stirling. Thirty-eight AFLP primer combinations and six SSRs provided 514 informative markers which formed a molecular marker map comprising 12 linkage groups (LGs) in 12601ab1 (nine with four homologous chromosomes) which were aligned with 12 in Stirling (11 with four homologous chromosomes), with four partial groups remaining in 12601ab1. Two LGs were identified unequivocally as chromosomes IV and V and eight others were tentatively assigned with chromosomes VII and X unidentified. All of the traits scored had moderately high heritabilities with 54–92% of the variation in clone means over 3 years and two replicates being due to genetic differences. A total of 39 QTLs were identified. A QTL for maturity was identified on chromosome V which explained 56% of the phenotypic variance, whereas the other QTLs individually explained between 5.4 and 16.5%. However, six QTLs were detected for after-cooking blackening and four for each of regularity of tuber shape, fry colour both after storage at 4 and 10°C and sprouting. Just two QTLs were found for each of yield, the two ‘overall’ scores, crop emergence, tuber size and common scab and just one QTL was detected for each of dry matter content, keeping quality, growth cracks and internal condition. The implications for practical potato breeding and for practical linkage and QTL analysis in autotetraploids are discussed.  相似文献

 Using RFLP markers, QTLs for tuber starch-content and tuber yield were mapped in two F1 populations derived from crossing non-inbred di-haploid potato breeding lines. QTLs were identified and mapped, based on both single-marker tests and interval analyses. A model specifically developed for interval QTL analysis in non-inbred plant species was successfully applied for the first time to experimental data. Results of both methods of QTL analysis were similar but not identical. QTLs for tuber starch-content and tuber yield were analysed in segregating populations K31 and LH in five and two environments, respectively. Population K31 was fully genotyped whereas population LH was selectively genotyped according to high and low tuber-starch content. Eighteen putative QTLs for tuber starch-content were identified on all 12 potato linkage groups and eight putative QTLs for tuber yield were identified on eight linkage groups. Twenty of twenty six putative QTLs were reproducibly detected in at least two environments and/or mapping populations. Few major QTLs for tuber starch-content were highly stable across environments but were detected in only one of the two mapping populations analysed. Most QTLs for tuber yield were linked with QTLs for tuber starch-content suggesting that the effects on both traits are controlled by the same genetic factors. The potato tuberworm, Phthorimaea operculella (Zeller), is a major pest of potatoes in fields and traditional storage. A common method of nonrefrigerated storage is to pile potatoes in straw-covered heaps in the field. Tubers may be stored up to 3-4 mo in this manner, until the next harvest. We studied the dynamics of potato tuber moth infestation associated with such field storage in a 12-wk experiment in Israel. We set up six potato heaps, and sampled them for potato tuber moth at different locations at weekly intervals. Potato tuber moth infestation was significantly higher at the perimeter of the heaps than at their center, but it did not differ between bottom, mid-height, and top of the heaps. The proportion of potato tuber moth-infested potato tubers increased from 10 to 65% over the course of the experiment, and the mean number of potato tuber moth larvae per tuber increased from 0.25 to 2.50. Potato tuberworm populations increased sharply after 3, 6, and 9 wk of study, possibly corresponding to successive generations that developed within the heaps. This interpretation is supported by calculations of potato tuberworm generation length based on temperature data. Catches in pheromone traps that were placed near the heaps were not correlated (spatially and temporally) with potato tuberworm densities within heaps, hinting that migration among heaps did not considerably affect within-heap population dynamics. Potato tuberworm levels were not correlated with ambient temperatures, perhaps because of the warm, humid, and constant microclimate within the heaps. We discuss the significance of our findings for control efforts of the potato tuberworm.  相似文献

Summary Variations in the ploidy level of 69 transgenic potato (Solanum tuberosum L.) plants regenerated from the tuber discs of 17 diploid lines were studied: 24 plants (35%) were diploid, the other 45 plants (65%) were tetraploid. Seventy-eight control regenerants obtained without Agrobacterium inoculation showed a relatively low tendency to tetraploidization (35%). The results obtained suggested that chromosome doubling occurred frequently in diploid potato lines during the tissue culture process for regeneration. Putative somaclonal changes in in vitro-formed tuber proteins were detected in three out of six transformants by electrophoretic analysis.  相似文献

Tuber yield, starch content, starch yield and chip color are complex traits that are important for industrial uses and food processing of potato. Chip color depends on the quantity of reducing sugars glucose and fructose in the tubers, which are generated by starch degradation. Reducing sugars accumulate when tubers are stored at low temperatures. Early and efficient selection of cultivars with superior yield, starch yield and chip color is hampered by the fact that reliable phenotypic selection requires multiple year and location trials. Application of DNA-based markers early in the breeding cycle, which are diagnostic for superior alleles of genes that control natural variation of tuber quality, will reduce the number of clones to be evaluated in field trials. Association mapping using genes functional in carbohydrate metabolism as markers has discovered alleles of invertases and starch phosphorylases that are associated with tuber quality traits. Here, we report on new DNA variants at loci encoding ADP-glucose pyrophosphorylase and the invertase Pain-1, which are associated with positive or negative effect with chip color, tuber starch content and starch yield. Marker-assisted selection (MAS) and marker validation were performed in tetraploid breeding populations, using various combinations of 11 allele-specific markers associated with tuber quality traits. To facilitate MAS, user-friendly PCR assays were developed for specific candidate gene alleles. In a multi-parental population of advanced breeding clones, genotypes were selected for having different combinations of five positive and the corresponding negative marker alleles. Genotypes combining five positive marker alleles performed on average better than genotypes with four negative alleles and one positive allele. When tested individually, seven of eight markers showed an effect on at least one quality trait. The direction of effect was as expected. Combinations of two to three marker alleles were identified that significantly improved average chip quality after cold storage and tuber starch content. In F1 progeny of a single-cross combination, MAS with six markers did not give the expected result. Reasons and implications for MAS in potato are discussed.  相似文献

Dihaploids obtained from a somatically chromosome-doubled dihaploid potato were crossed with Solanum phureja clones. To test the effect of inbreeding, measurements were made of their seed production and the tuber yield, tuber number and mean tuber weight of their offspring. On average, seed production of the second generation dihaploids was higher than that of the original dihaploid progenitor. Progeny tuber yield and its components were little different from those of the original dihaploid's progeny. Tuber flesh quality, as measured lack of blemishes, was better in the offspring of second generation dihaploids.
It is suggested that the negative effects of producing second generation dihaploids are minor compared with producing the first generation dihaploid from a tetraploid, because most deleterious recessives have already been unmasked. The results indicated residual variation within the original dihaploid which could be exploited for plant breeding purposes.
An indicator of inbred status, alternative to the inbreeding coefficient, is suggested which could be applied to both diploids and tetraploids.  相似文献

The ideal marker system for tetraploid potato would be dosage-sensitive and have the ability to distinguish heterozygous genotypes with multiple haplotypes within the genomic region targeted by the marker. The objective of this study was to evaluate the utility of high-resolution DNA melting (HRM) for genotyping and polymorphism detection in diploid and tetraploid potato. Amplicon scanning, unlabelled probe, and short amplicon assays were developed for four candidate genes affecting tuber skin and flesh colour, and starch, and a marker linked to nematode resistance. Genotyping a set of 95 potato clones revealed several examples of clones with three distinct haplotypes. Combined probe and amplicon analysis identified between 29 and 44 unique genotypes for the same assays. Assays developed for four of the five target genes are suitable for marker-assisted selection in potato breeding programs. This study illustrates the use of HRM in potato genetics. Further advances in the technology and associated data analysis should make HRM a useful tool for basic and applied studies of potato.  相似文献

Potato is one of the most important food crops all over the world. Breeding activities for this crop are mainly aimed to improve the quality and yield of tuber. However, genetic architecture of various traits contributing to the quality and yield of potato are not yet completely understood. Genome wide association studies provides a broader way to identify the genomic regions associated with various traits. Panels of 237 tetraploid potato genotypes from different countries were grown for two consecutive years 2016 and 2017 at experimental research area of Potato research center Niğde, Turkey. A genome wide association study using SolCAP 12K array was performed for various morpho-agronomic traits. Structure algorithm and neighborhood joining analysis clearly divided all genotypes into 4 clusters on the basis of their origin. For the marker trait association, Mixed Linear Model in TASSEL was performed and 36 genomic regions were found for the traits under study. The mean r2 value was found to be 0.92 and mean significant LD was 47.5% in the populations. LD patterns reflected the breeding history of potato. The findings of present study provide a framework which could be useful for future potato breeding programs to enhance the production and to reduce the challenges in the coming years to feed world’s population.  相似文献

Summary Environmental influence on physiological factors that determine seed quality is discussed in relation to true potato seed (TPS). This review has been motivated by the need for high quality seed for the proper evaluation of TPS technology. The objectives of tuber production may not be in harmony with those required for the production of high quality TPS. The production of high quality TPS may be influenced by the stronger sink strength ability of fast developing tubers for assimilation of available nutrients. Earliness of tuber formation, which is needed in TPS progenitors, may tend to result in incomplete TPS development. Limiting conditions during seed development decrease the potential of the seed for field establishment. Seed vigor is the most important attribute of seed quality since it is essential for seedling performance under the adverse conditions commonly encountered during field development. The attractiveness of TPS technology for tropical areas, where potato production is limited by low-quality seed tuber availability, is decreased by the lack of seedling vigor and uniformity of seedlings derived from TPS. Current research indicates that considerable genetic improvement of TPS vigor and uniformity is possible. It is suggested that significant immediate improvement of TPS quality would result from agronomic techniques that reflect understanding of the physiological factors that influence the production and maintenance of high quality seed. Three somaclonal populations of potato (Solanum tuberosum L.), each comprised of at least 1,000 plants, were regenerated from the cultivars Kennebec, Russet Burbank, and Superior. The frequency of formation of adventitious meristems from tuber disc explants varied significantly between these potato genotypes. Only 1.0–1.3% of each somaclonal population exhibited morphological aberrations. Regenerated populations of Kennebec and Superior, when compared to respective control populations over three asexual generations, were similarly enriched with somaclones having more elongated tubers, a higher total tuber number and weight, a higher cull tuber number and weight, and earlier maturity. Somaclones of Russet Burbank also produced more elongated tubers, a higher total tuber number, and a higher cull tuber number and weight but, in contrast, these somaclones were lower in total tuber weight, lower in U.S. 1 tuber number and weight, shorter in stem length, and lower in vigor. Of the three cultivars, Russet Burbank somaclones possessed the greatest variability for most traits. Besides this significant genotype effect, quantitative traits differed amongst each other in respect of relative changes resulting from somaclonal variation.  Selfed and backcross progenies developed from tetraploid somatic hybrids between Solanum tuberosum (tbr) and S. commersonii (cmm) were characterized for nonacclimated freezing tolerance (NA) and acclimation capacity (ACC) (two independent genetic components of freezing tolerance) under controlled environments. The segregation covered 28% and 71% of the parental range for NA and ACC, respectively, with the distribution skewed toward the tbr parent. Therefore, ACC appeared to be relatively easier to recover in the segregating generation. Some first backcross progeny had greater freezing tolerance than the cultivated parent primarily through the increase in ACC. When grown in the field, the improved freezing tolerance observed in the selfed progeny under controlled conditions was confirmed. Among NA, ACC, and freezing tolerance after acclimation (AA, which is the cumulative performance of NA and ACC), AA exhibited the highest correlation coefficient with field frost tolerance. In addition to freezing tolerance, vine maturity and tuber traits including tuber yield, tuber number per plant, mean tuber weight, and specific gravity were also segregating. No significant correlation between undesirable tuber traits and freezing tolerance was detected. Vine maturity and freezing tolerance were significantly correlated, so more careful selection for earliness was necessary in incorporating freezing tolerance. Yield comparable or superior to the backcross parent Wis AG 231 and an early Canadian cultivar, ‘Sable’, was found in many backcross progeny and some selfed progeny. The observed high yield can be attributed to the increase in mean tuber weight as well as tuber number. Moreover, a high portion of progeny had a specific gravity higher than 1.085, and some greater than 1.1. A high amount of intravarietal variation in satellites and C-banded chromosomes was observed in the hexaploid wheatgrass synthetic cultivar 'Greenleaf' (Agropyron intermedium ssp. trichophorum (Link) A. &Gr., 2n = 6x = 42, genome E1E1E2E2SS). The cultivar is an open-pollinated perennial that shows extensive interplant polymorphism for many biological characters. Maximum number of satellites detected varied among plants from zero to six. In 61% of the plants, we observed two large satellites in association with zero, one, or two small ones. Chromosome constitution differed significantly among plants as revealed by analysis of variance based on the total number of banded chromosomes and the number of banded chromosomes with telomeric bands at either one or both ends. Heteromorphism in C-banding patterns between homologues was found in most of the chromosomes and was classified into four types: (i) difference in band size, (ii) difference in presence/absence of one or two bands, (iii) completely different banding patterns, and (iv) banded versus unbanded. Homologous chromosomes having types iii and iv heteromorphism could only be matched by their relative length and arm ratio instead of C-banding patterns. Deletions were detected in two chromosomes. Overall, C-banded chromosomes of this cultivar were characterized by the presence of large telomeric bands and were quite different from the previously reported karyotypes of the supposed diploid ancestor Agropyron elongatum (Host) P. Beauv. (genome EE) and an Ag. intermedium (Host) P. Beauv. accession (E1E1E2E2SS) The results suggest that dramatic chromosome modifications have occurred in this species during the course of evolution. The study sheds light on the extent of intrapopulation polymorphism present in the karyotypes of outcrossing polyploids and synthetic cultivars and has implications regarding strategies for chromosomal manipulation involving open-pollinated species.  相似文献

The production of attractive, uniform true potato seed (TPS) progenies was investigated. Four breeding schemes were compared: intercrossing tetraploid cultivars (cv x cv); doubled dihaploids x cultivars (ddh x cv); cultivars x diploid unreduced-gamete producers (cv x FDR) and doubled dihaploids x diploid unreduced-gamete producers (ddh x FDR). Fifty three progenies and five clones were grown in a glasshouse in a randomised complete block design with three replicates of 25 plants per progeny and clone. Each plant's tubers were counted and the colour, shape, quality of skin finish, flesh colour, and commencai attractiveness (which includes yield) recorded. The most uniform progenies were also selected by visual comparison with the clones. For mean attractiveness, differences (P < 0.001) between breeding schemes and between progenies within breeding schemes were detected. The cv x cv and cv x FDR progenies were more attractive than clonal controls. There were significant additive and non-additive effects for attractiveness in all breeding schemes except cv x FDR. There were between-progeny differences (P < 0.001) for uniformity for all characters. Progenies uniform for one character could be variable for other traits. Breeding schemes gave different levels of uniformity (P < 0.001) for all characters except shape and flesh colour, but none gave low levels of variation for all traits. Doubled-dihaploid parents increased the variation in progenies. There were uniformity differences (P < 0.001) between progenies within breeding schemes for all characters. Evidence of additive and nonadditive genetic variation for uniformity in all traits was detected. In each breeding scheme, parents with good general combining ability (GCA) for uniformity in several characters were identified. Visually selected uniform progenies had parents with good GCAs for uniformity in a range of traits and high specific combining abilities (SCAs) for several traits. A desynaptic first-division restitution (FDR) clone and a male-sterile doubled-dihaploid clone had the best GCAs for tuber uniformity in TPS progenies. Achieving multitrait uniformity in TPS is problematic but may be aided by the selection of parents with GCAs for uniformity coupled with progeny testing to allow for non-additive effects.  相似文献
