The species status of Dermacentor marginatus and Dermacentor reticulatus was evaluated by scanning electron microscope (SEM) examination of adult ticks, cross-breeding experiments and molecular biological analysis of eggs derived from transspecific pairings. The SEM investigations including the morphometric quantification of phenotypic features resulted in an unequivocal differentiation of adult D. marginatus and D. reticulatus ticks. The cross-breeding experiments demonstrated that irrespective of whether female ticks of both species were applied with con- or transspecific male ticks or without males to sheep, they engorged and laid eggs. The larvae, however, developed only in eggs which originated from conspecific matings. A nested polymerase chain reaction (PCR) of the second internal transcribed spacer (ITS2) using the DNA of eggs from transspecific pairings and sequencing of the PCR products revealed two different genotypes. The genotypes of eggs originating from D. marginatus and D. reticulatus females of these pairings differed. However, the eggs deposited by D. marginatus always possessed the same two genotypes as did the eggs produced by D. reticulatus. These results argue for a strict reproductive isolation of D. marginatus and D. reticulatus and, therefore, for a separate species status.  相似文献

The water content, the survival time at various relative humidities(r.h.) and the critical equilibrium activity of unfed adultDermacentor marginatus and D.reticulatus ticks were investigated at a constant temperature of20 °C. It was also examined whether these ticks use liquidwaterto compensate water loss. Both Dermacentor spp. showed nosignificant differences in water content in relation to body mass. The meanwater content of D. marginatus and D.reticulatus was 54.6% and 54.7%, respectively, in females and 56.3%and 57.0%, respectively, in males. The survival time of unfed adults prolongedwith decreasing saturation deficits. On average, males survived longer thanfemales and D. marginatus ticks survived mostly longerthanD. reticulatus ticks. The 50% mortality period rangedbetween 40 d at 33% r.h. and 420 d at 95% r.h. in D.marginatus, and between 43 d at 33 r.h. and 366 d at 95% r.h. inD. reticulatus. The critical equilibrium activity of unfedadults was estimated to be 0.84 for both species and was independent of sex.When dehydrated adult D. marginatus and D.reticulatus ticks were offered liquid water, only a few slightlygained weight while most further lost weight. Liquid water was not attractivefor dehydrated or non-dehydrated ticks and drinking was not observed. Aftersubmerging in water for 2 d, most of the dehydrated ticks had gained weight.  相似文献

Dermacentor reticulatus ticks are recognized as the most important vectors of Babesia canis, the aetiological agent of canine babesiosis occurring throughout Europe. Vector competence of D. reticulatus for B. canis is well described and experimentally determined; however, by using molecular analysis it was proven so by one recent study in Russia. Herein, the additional molecular evidence of B. canis infection in D. reticulatus ticks collected in Slovakia is provided. Using PCR followed by sequencing of distinctive amplicons we determined the presence of Babesia canis canis in one of 100 tested adult ticks. Two zoonotic pathogens, Francisella tularensis and Coxiella burnetii, were previously isolated from D. reticulatus ticks in Slovakia. In our samples, we detected only the presence of F. tularensis.  相似文献

Abstract. A total of 924 questing Dermacentor reticulatus (Fabricius), 504 Ixodes ricinus (L.), sixty Haemaphysalis concinna Koch and 718 mosquitoes (Aedes spp.) were examined in a floodplain forest ecosystem during the 1994-95 outbreak of tularaemia in South Moravia, Czech Republic. Francisella tularensis was not isolated from H.concinna ticks or Aedes spp. mosquitoes, whereas twenty-one isolates were recovered from the other haematophagous arthropods. Dermacentor reticulatus revealed a significantly higher infection rate (2.6%) than I.ricinus (0.2%). This tick species acts as principal vector for tularaemia in the enzootic focus. Monitoring of D.reticulatus for F.tularensis thus seems to be a very efficient approach in the surveillance of tularaemia in the flood-plain forest ecosystems of Europe.  相似文献

In unfed adult Dermacentor marginatus and D. reticulatusticks survival and capability to restore water balance after loss of high percentages of exchangeable body water were investigated. Furthermore, it was examined how frequently dehydrated ticks of these species were able to rehydrate by uptake of atmospheric water vapour. The critical water mass, defined as the water mass remaining in a tick at the nonambulatory state, differed between light and heavy weight groups and averaged 62.4 and 55.8%, respectively, of the total body water of fully hydrated ticks in females, and 54.4 and 51.1% respectively, in males of D. marginatus. In D. reticulatus, the corresponding figures were 55.9 and 54.7% in females and 52.1 and 52.7% in males. All ticks survived dehydration to 50, 75 or 100% of the critical water mass, and 96.7% of the D. marginatus ticks and 95.8% of the D. reticulatus ticks compensated water losses during subsequent incubation at 95% relative humidity (r.h.) and 20°C. Unfed females and males of both Dermacentor spp. were capable to balance water loss very frequently over a period of several months. When ticks were repeatedly dehydrated at 0% r.h. for 7 days and rehydrated at 95% r.h. and 20°C, females and males of D. marginatus reached the 50% mortality after 22 and 29 cycles of de- and rehydration, respectively, during 211 and 285 days, respectively. In D. reticulatus, 50% of females and males survived 23 and 17 cycles, respectively, during 248 and 186 days, respectively. Rehydration weights were as high or even higher as those of ticks kept at permanent 95% r.h.  相似文献

Nosema slovaca (Microsporidiae) was isolated from a maleDermacentor reticulatus tick collected in the Mélykút locality, Bács-Kiskun province, Hungary. This parasitic protozoan caused an acute infection in partly engorgedD. reticulatus females when inoculated intracoelomically resulting in death of the ticks within 5–15 days post-infection. This strain ofNosema can be maintained in the laboratory by passaging between partly engorgedD. reticulatus females and is currently being studied for its potential as a biological control agent of ixodid ticks.  相似文献

A total of 7778 host‐seeking adult Dermacentor reticulatus (Ixodida: Ixodidae) ticks were examined for the prevalence of Francisella tularensis holarctica (Thiotrichales: Francisellaceae) in a natural focus of tularaemia in the floodplain forest–meadow ecosystem along the lower reaches of the Dyje (Thaya) river in South Moravia (Czech Republic) between 1995 and 2013. Ticks were pooled (10 specimens per pool) and their homogenates inoculated subcutaneously in 4‐week‐old specific pathogen‐free mice. Dead mice were sectioned, their spleens cultivated on thioglycollate–glucose–blood agar and impression smears from the spleen, liver and heart blood were Giemsa‐stained. Sixty‐four pools were positive for F. tularensis: the overall minimum infection rate (MIR) was 0.82%. Overall MIRs for the 4714 female and 3064 male D. reticulatus examined were 0.89 and 0.72%, respectively; MIRs fluctuated across years between 0.0 and 2.43%. The estimated bacterial load in infected ticks varied from 0.84 to 5.34 log10 infectious F. tularensis cells per tick (i.e. from about seven to 220 000 cells). Ticks with low loads were more prevalent; more than 1000 infectious cells were detected in 24 ticks (0.3% of all ticks and 37.5% of infected ticks). Monitoring of D. reticulatus for the presence and cell numbers of F. tularensis may be a valuable tool in the surveillance of tularaemia.  相似文献

Tick-borne encephalitis virus (TBEV) (Flaviviridae, Flavivirus) is an arthropod-borne virus, an etiologic agent of tick-borne encephalitis (TBE), a human infection involving the central nervous system. The disease is endemic in a large region in Eurasia, where it is transmitted mainly by Ixodes ricinus and Ixodes persulcatus ticks. It is known that also Dermacentor reticulatus is involved in circulation of TBEV. However, the current knowledge of D. reticulatus importance in TBE epidemiology is still insufficient. A total of 471 adult D. reticulatus ticks were collected by flagging vegetation in the Bia?owie?a Primeval Forest, Biebrza National Park, Masurian Landscape Park (North-Eastern Poland) and in the city of Warsaw in the years 2007–2010. All collected ticks were examined individually for the presence of RNA of TBEV using nested RT-PCR assay. Positive results were noted in all investigated localities with the infection rate ranging from 0.99 to 12.5 % with a total mean of 2.12 %. The difference in the percentage of infective males and females was not statistically significant.  相似文献

Mixed infection of Rickettsiella phytoseiuli and Coxiella burnetii was investigated in hemolymph and organs of experimentally infected females of Dermacentor reticulatus ticks. Following intracoelomic infection, both agents, with the exception of Gene's organ, multiplied well in the cells of the tick host's organs. Two out of six developmental stages of R. phytoseiuli, i.e., crystal-forming and small dark particles, in dual infection with C. burnetii revealed marked morphological alterations. C. burnetii in the presence of R. phytoseiuli penetrated into the cortical layer of the synganglion and into the alveoli of the second and third type of salivary glands, but did not occur in the single infection.  相似文献

The ultrastructural aspects of the interaction of R. sibirica and R. slovaca with cells of mites of the species Dermacentor reticulatus, D. marginatus and Ixodes ricinus after their parenteral infection, as well as in the organs of D. marginatus infected naturally in the environment, have been studied. Both rickettsial species have similar morphology in different organs of the vector. These rickettsiae not only multiply, their populations are also partly destroyed in phagolysosomes. The natural mixed infection of R. sibirica and orbivirus in cells of D. reticulatus is described. As shown in this study, both associates pass through the complete ontogenetic cycle of development on the level of the host body and also on the level of an individual cell.  相似文献

In recent years, the distribution of Dermacentor reticulatus ticks has expanded into new territories in many European countries, including Poland, with increased population densities in areas of their regular occurrence. The spread of D. reticulatus enhances the risk of exposure of domestic animals and their owners to tick-borne diseases. The objective of this study was to assess the prevalence of infection of D. reticulatus ticks feeding on dogs with the pathogens Borrelia burgdorferi sensu lato and Anaplasma phagocytophilum. The study material comprised 152 D. reticulatus ticks collected from dogs in the northeastern part of Lublin Province (eastern Poland). A ready-made AmpliSens® TBEV, B.burgdorferi sl, A.phagocytophilum, E.chaffeensis/E.muris-FRT PCR kit was used for qualitative detection and differentiation of tick-borne infections. The assessment of the degree of infection of the analyzed ticks with the two pathogens revealed that 9.2% (14/152) of the examined ticks were infected with one of the pathogens. No co-infections with the pathogens were detected in any of the ticks. The highest specific percentage of infections (8.6%, 13/152) was associated with A. phagocytophilum. The presence of B. burgdorferi s.l. was detected in only one of the examined ticks (0.7%). The spread of D. reticulatus to new territories and the increase in population density in areas of their regular occurrence implies the need for further studies of the prevalence of pathogens with medical and veterinary importance in order to assess the risk of tick-borne diseases.

We collected a total of 169 adult hard ticks and 120 nymphs from under the leaves of plants located along tourist nature trails in ten localities. The results present data examining the vector competence of ticks of different genera and the presence of Rickettsia and Anaplasma species. The ticks belonged to three genera, Amblyomma, Dermacentor, and Haemaphysalis, comprising 11 species. Rickettsia bacteria were detected at three collection sites, while Anaplasma bacteria were detected at only one site. Phylogenetic analysis revealed new rickettsia genotypes from Thailand that were closely related to Rickettsia tamurae, Rickettsia monacensis, and Rickettsia montana. This study was also the first to show that Anaplasma bacteria are found in Haemaphysalis shimoga ticks and are closely related evolutionarily to Anaplasma bovis. These results provide additional information for the geographical distribution of tick species and tick‐borne bacteria in Thailand and can therefore be applied for ecotourism management.  相似文献

The recent implication of Dermacentor reticulatus (Ixodida: Ixodidae) in the transmission of canine babesiosis in the U.K. has highlighted the lack of accurate published data on its distribution in this country. This paper aims to collate and appraise historical data for D. reticulatus, to supplement such data with more recent surveillance data and to report on field sampling conducted during 2009–2016. These updated data facilitate better understanding of the current distribution of this tick in the U.K., which will better inform disease risk assessments. There appear to be four known regions of the U.K. in which D. reticulatus currently occurs, including western Wales, North and South Devon, and Essex. The majority of foci are located in coastal sand dunes and maritime grasslands, including grazing marsh. However, more recently the tick has been detected in urban greenspace in Essex. The emergence of this tick as a vector of babesiosis in the U.K. and its recent apparent spread in Essex into urban greenspace highlight the need for continued surveillance and for further research into its status as a vector of human and veterinary pathogens.  相似文献

In this paper, activity concentrations of radiocesium in mushrooms from various locations in the Czech Republic and the Slovak Republic in the years 2000–2004 are presented. Summary reports of Czech regulatory bodies have judged the average activity concentrations in mushrooms to be remarkably below the maximum permissible level of contamination. However, there are certain regions where radiocesium activities have approached the maximum permissible level for contamination of basic foodstuff. For example, the highest activity concentration of 137Cs measured by gamma-spectrometry was 2,263 Bq kg−1 (on a dry mass basis), in Xerocomus badius from Staré Ransko (a locality in the Czech-Moravian Highlands, Czech Republic). The highest activity concentration of 137Cs measured in Slovakia was 966 Bq kg−1 (on a dry mass basis), for Suillus luteus from Senica. For comparison, the corresponding activity concentration in a sample taken within the 30 km zone around Chernobyl was measured to be 6,000 Bq kg−1 (on a dry mass basis). Our results have also demonstrated remarkably lower activity concentrations of 137Cs in Slovakia compared to those in the Czech Republic.  相似文献

Worldwide, ticks are important vectors of human and animal pathogens. Besides Lyme Borreliosis, a variety of other bacterial and protozoal tick-borne infections are of medical interest in Europe. In this study, 553 questing and feeding Ixodes ricinus (n = 327) and Dermacentor reticulatus ticks (n = 226) were analysed by PCR for Borrelia, Rickettsia, Anaplasma, Coxiella, Francisella and Babesia species. Overall, the pathogen prevalence in ticks was 30.6% for I. ricinus and 45.6% for D. reticulatus. The majority of infections were caused by members of the spotted-fever group rickettsiae (24.4%), 9.4% of ticks were positive for Borrelia burgdorferi sensu lato, with Borrelia afzelii being the most frequently detected species (40.4%). Pathogens with low prevalence rates in ticks were Anaplasma phagocytophilum (2.2%), Coxiella burnetii (0.9%), Francisella tularensis subspecies (0.7%), Bartonella henselae (0.7%), Babesia microti (0.5%) and Babesia venatorum (0.4%). On a regional level, hotspots of pathogens were identified for A. phagocytophilum (12.5–17.2%), F. tularensis ssp. (5.5%) and C. burnetii (9.1%), suggesting established zoonotic cycles of these pathogens at least at these sites. Our survey revealed a high burden of tick-borne pathogens in questing and feeding I. ricinus and D. reticulatus ticks collected in different regions in Belarus, indicating a potential risk for humans and animals. Identified hotspots of infected ticks should be included in future surveillance studies, especially when F. tularensis ssp. and C. burnetii are involved.  相似文献
