The portal appearance rates and net rates of amino acids' absorption were studied in rats fed semi-synthetic diets containing either casein or lactalbumin (CAS and LA, respectively) as the only protein sources. Rats were pre-adapted to the experimental diets for 5 days prior to the absorption studies. Rats fed the LA diet had higher (p < 0.05) portal vein concentrations of free essential amino acids than those fed the CAS diet at 0, 60, 105 and 150 min after feeding. Portal and arterial concentrations of arginine, leucine, tryptophan, lysine and methionine were higher (p < 0.05) in rats fed LA at most time points tested, while concentrations of tyrosine were higher (p < 0.05) in CAS fed rats. When portal flow rates were compared, values for arginine, threonine, alanine, leucine, tryptophan and lysine were higher (p < 0.05) in LA at most time points tested, while proline, tyrosine and valine were higher (p < 0.05) for CAS fed rats after 60 and 105 min feeding. Portal blood flow varied (p < 0.05) with time in rats fed protein-free or LA diets, and was higher (p < 0.05) than that of CAS at 105 min. Intestinal net rates of absorption of tyrosine, valine, leucine and lysine were higher (p < 0.05) for LA fed rats as compared to those fed CAS at most time points tested, while alanine and proline net rates were higher (p < 0.05) for CAS fed rats at 60, 105 and 150 min. Amounts of protein in stomach contents of rats fed the CAS diet were significantly higher (p < 0.05) than those in LA fed rats at 60, 105 and 150 min after feeding. The relative liver weight of the rats fed the CAS diet was lower (p < 0.05) than that of animals fed the LA diet. Lower (p < 0.05) liver glycogen and lipid contents were determined in rats fed CAS diet respect to LA or protein-free fed rats. Results indicate that dietary and plasma amino acids profile are only partially related, and that under normal feeding conditions amino acids from CAS and LA are absorbed at different rates, which is likely to affect liver composition and metabolism.  相似文献

异育银鲫幼鱼对饲料中赖氨酸的利用及需要量研究   总被引:8,自引:3,他引:5  
以添加晶体氨基酸的半精制饲料饲喂异育银鲫幼鱼,通过69d的生长实验来确定其赖氨酸需要量。饲料以白鱼粉为主要蛋白源,饲料中的总赖氨酸含量分别为1.82%、2.32%、2.82%、3.32%3、.82%、4.32%和4.82%7个水平。实验在室内循环水养殖系统中进行,每种饲料随机3个重复。实验结果表明,异育银鲫能够利用饲料中的晶体赖氨酸、蛋氨酸。在投喂后3h,其血浆中的游离赖氨酸、蛋氨酸含量最高。当饲料中赖氨酸含量为3.32%时,异育银鲫的终末尾均重、特定生长率和鱼空壳占体重的百分比最高,肝体指数最低。当饲料中赖氨酸含量为3.82%时,异育银鲫的干物质表观消化率显著高于其他组(PPP>0.05)。血红蛋白含量以赖氨酸含量为2.82%的饲料组最高,4.82%组最低;随着饲料中赖氨酸含量的升高,异育银鲫红细胞数下降,血清脲氮含量升高,且血清脲氮含量具有组间显著性差异(P<0.05)。根据折线法,由异育银鲫的特定生长率同饲料中赖氨酸水平的相关性得出其赖氨酸需要量为3.27%,占饲料蛋白的8.52%。    相似文献

The absorption of lysine is facilitated by leucine, but there is no information regarding the effect of crude protein, lysine and leucine levels on the expression of cationic amino acid transporters in pigs. Therefore, an experiment was conducted with 20 pigs (14.9 +/- 0.62 kg initial body weight) to evaluate the effect of two protein levels, and the content of lysine, threonine, methionine and leucine in low crude protein diets on the expression of b(0,+) and CAT-1 mRNA in jejunum, Longissimus dorsi and Semitendinosus muscles and serum concentration of amino acids. Treatments were as follows: (i) wheat-soybean meal diet, 20% crude protein (Control); (ii) wheat diet deficient in lysine, threonine and methionine (Basal diet); (iii) Basal diet plus 0.70% L-lysine, 0.27% L-threonine, 0.10% DL-methionine (Diet LTM); (iv) Diet LTM plus 0.80% L-leucine (Diet LTM + Leu). Despite the Basal diet, all diets were formulated to meet the requirements of lysine, threonine and methionine; Diet LTM + Leu supplied 60% excess of leucine. The addition of lysine, threonine and methionine in Diet LTM increased the expression of b(0,+) in jejunum and CAT-1 in the Semitendinosus and Longissiums muscles and decreased CAT-1 in jejunum; the serum concentration of lysine was also increased (p < 0.01). Further addition of L-leucine (Diet LTM + Leu) decreased the b(0,+) expression in jejunum and CAT-1 in the Longissimus dorsi muscle (p < 0.05), increased the serum concentration ofleucine and arginine and decreased the concentration of isoleucine (p < 0.05). Pigs fed the Control diet expressed less b(0,+) in jejunum, and CAT-1 in the Semitendinosus and Longissiums muscles expressed more CAT-1 in jejunum (p < 0.05) and had lower serum concentration ofisoleucine, leucine and valine (p < 0.05), but higher lysine concentrations (p < 0.01) than those fed Diet LTM. These results indicated that both, the level and the source of dietary amino acids, affect the expression of cationic amino acid transporters in pigs fed wheat-based diets.  相似文献

Two experiments were conducted with 72 pigs between 28 and 56 days of age to study the effect of tryptophane supplementation on their performance when fed on diets containing wheat and meat meal.In the first experiment, pigs were fed on a basal diet (Diet 1) or on the same diet supplemented with calcium dihydrogen phosphate (Diet 2), bone meal (Diet 3) or bone meal plus tryptophane (Diet 4), all to 3.1% calcium. The weight gains of the pigs (315 g day?1) fed on Diet 3 were significantly lower than that of the pigs fed on the other three diets (363 g day?1). The feed conversion ratios showed a similar trend. Diet 3 contained 0.16% tryptophane while the other diets contained 0.18–0.19% tryptophane. The crude protein, lysine and methionine contents of all diets were similar.In the second experiment, a basal diet containing meat meal and bone meal was supplemented with tryptophane, lysine plus methionine or all three amino acids. Feed intake was increased by all amino acid supplements. Weight gains were improved significantly (57%) by the addition of all three amino acids to the diets, but the improvements due to tryptophane alone (28%) or methionine plus lysine (35%) were not significant. Tryptophane supplementation alone or with lysine plus methionine increased the nitrogen retention of the pigs.It was concluded that the requirement for tryptophane of pigs between 28 and 56 days of age was greater than 0.16% of diets containing wheat and meat meal.  相似文献

采用化学分析和动物实验的方法对冷冻干燥、微波干燥和低温干燥的蝇蛆粉进行了营养评价。结果表明,3种干燥方法不影响蝇蛆粉的粗蛋白和灰分含量,但微波干燥蝇蛆粉的脂肪含量显著降低;氨基酸分析显示,冷冻干燥蝇蛆粉的氨基酸总含量>微波干燥的蝇蛆粉>低温干燥的蝇蛆粉,而3种方法干燥的蝇蛆粉EAA/TAA均等于或大于41%,说明干燥所得的蝇蛆粉均属优质蛋白。另外,与FAO/WHO参考模式值相比,冷冻干燥蝇蛆粉的必需和半必需氨基酸总量明显高于FAO/WHO参考模式值,微波干燥的蝇蛆粉与FAO/WHO参考模式值相当,而低温干燥的蝇蛆粉中必需与半必需氨基酸含量显著低于FAO/WHO参考模式值,说明微波干燥及低温干燥会破坏蝇蛆中的某些必需或半必需氨基酸。动物实验结果表明,冷冻干燥的蝇蛆粉在动物体内被吸收利用的程度最高,其次是酪蛋白,再次是低温干燥的蝇蛆粉,而微波干燥的蝇蛆粉被动物吸收利用的程度最低。  相似文献

This study was conducted to compare the safety of soybean meal prepared from genetically modified (GM) glyphosate-tolerant (Roundup Ready; RR) soybeans and conventional soybeans. Eighty Sprague-Dawley rats (40 males and 40 females) were randomly allotted to one of four groups according to sex and body weight for a 13-week feeding experiment. The rats were fed corn-based diets containing 60% conventional soybean meal, a mixture of 30% conventional and 30% RR soybean meal, 60% or 90% RR soybean meal. All diets were adjusted to an identical nutrient level except the 90% RR diet. The two soybean meals were similar in chemical analysis and amino acid composition. During the 13-week growth trial, body weight (P < 0.05) and feed intake (P < 0.05) decreased only in rats fed with 90% RR soybean meal at the first week. No treatment-related deaths occurred during the experiment. Gross necropsy findings, haematological or urinalysis values and clinical serum parameters showed no meaningful differences between rats fed the control and RR soybean meals. A 145 bp of cp4 epsps gene specific for the GM constructs from RR soybean meal or a 407 bp of lec gene from endogenous soybean DNA could not be detected in investigated masseter muscle samples. No adverse effects of glyphosate-tolerant soybean meal on rats were seen even at levels as high as 90% of the diet.  相似文献

A 10-week feeding trial was conducted to study the effect of feeding level and dietary lysine concentration on growth, protein and lysine retention, and body composition in juvenile turbot. Maintenance requirement for lysine and the efficiency of lysine utilisation were determined as well. Two experimental diets were formulated based on fishmeal or wheat gluten as main protein sources, containing 6.4 g (Diet A, control) and 4.5 g lysine per 100 g CP (Diet B), respectively. Diets were fed once daily at six feeding levels (per day 0.3%, 0.6%, 0.9%, 1.2%, and 1.5% of body weight [BW] and ad libitum) to a total of 432 fish of 48 g initial BW. No differences in the growth parameters were observed between diets at the same feeding level, except a lower feed to gain ratio (p < 0.05) at the highest feeding level at Diet B. Whole-body composition was not affected by diet, whereas muscle protein concentration was significantly lower for fish fed Diet B. Amino acid concentration in whole-body protein was affected by dietary treatment and fish fed Diet B showed lower concentrations of all essential amino acids. In fish muscle protein, lysine, methionine, leucine, isoleucine, and valine concentrations were significantly lower in Diet B. Efficiency of lysine utilisation for growth (klys) was determined by linear regression analysis and amounted for 0.69 for Diet B. The maintenance lysine requirement defined at zero lysine retention was 6.5 mg · kg?0.8 · d?1. Lysine intakes at zero protein retention were 13.0 mg and 12.9 mg · kg?0.8 · d?1 for Diet A and B, respectively. Growth and nutrient retention were similar for both diets and, therefore, a lysine deficiency in Diet B did not occur. In conclusion, a proportion of 330 g wheat gluten per kg feed did not influence growth performance and maintenance requirement for lysine in juvenile turbot. However, the effect of diet composition on the amino acid profile of body protein might be relevant for the derivation of the amino acid requirement from protein retention.  相似文献

The effects of adding lysine and/or methionine to a ration of calculated deficiency in these amino acids of 10% and 20%, respectively, were studied in 24 Brown Swiss cows. The mixed rations (27% grass silage, 19% maize silage, 5% hay and 49% concentrate on DM basis) contained 14.5% CP on average. Lysine supply was selectively elevated by adding fish meal in exchange for other concentrate ingredients. Methionine was supplied in a rumen‐protected form. Milk protein content was elevated whereas fat amount decreased by adding both amino acids. Lactose content increased without additional lysine from fish meal. Live weight, milk yield, milk fat content and protein amount remained unaffected by any variation of amino acids supply. Also nutrient digestibility and nitrogen balance were not changed by the treatments. Blood plasma concentrations confirmed the assumed variation in metabolic lysine and, less clear, methionine supply. Effects on plasma concentrations of other amino acids were relatively small. Most plasma hormones and enzymes, and metabolites in plasma, urine and milk did not respond to the variation in amino acid supply. Lysine addition via fish meal increased aspartate amino transferase and decreased urinary allantoin concentration. Additional methionine elevated plasma ornithine. Overall lysine and methionine appear to have been only marginally deficient in the unsupplemented ration fed for 3 weeks despite the deficiency of 10% to 20% as calculated by the I.N.R.A. method.  相似文献

Energy budgets were calculated for the southern catfish, Silurus meridionalis, fed diets replacing 0%, 13%, 26%, 39%, 52% and 65% fish meal protein with soybean meal (SBM) protein with or without methionine supplementation to apparent satiation at 27.5 °C. With increasing dietary soybean protein levels (SPL), the feed energy lost in feces, excretion and metabolism increased, while that available for growth decreased (P < 0.05). When 0.12% or 0.26% methionine at 39% SPL was added to reach that in body carcass or the control group (0% SPL), no significant differences were found in each component of energy budgets. When 0.21% or 0.33% methionine at 52% SPL was added to reach the content of methionine in body carcass or the control group, energy spent on growth increased, but that on excretion and metabolism decreased (P < 0.05). These results suggested that the differences in growth rate among the southern catfish fed the diets with different SPL were due to decreasing absorption rate, increasing excretion and metabolism with increasing dietary SPL. The most important factor limiting the use of soybean protein was the imbalance of essential amino acids, which resulted in more energy spent on metabolism and excretion, less energy on growth. Supplementation of methionine produced a relatively better amino acid profile and subsequently improved the utilization of soybean protein at high SPL, which resulted in less energy used for metabolism or lost in excretion and more energy available for growth.  相似文献

The ideal protein concept has allowed progress in defining requirements as well as the limiting order of amino acids in corn, soybean meal, and a corn–soybean meal mixture for growth of young chicks. Recent evidence suggests that glycine (or serine) is a key limiting amino acid in reduced protein [23% crude protein (CP) reduced to 16% CP] corn–soybean meal diets for broiler chicks. Research with sulfur amino acids has revealed that small excesses of cysteine are growth depressing in chicks fed methionine-deficient diets. Moreover, high ratios of cysteine:methionine impair utilization of the hydroxy analog of methionine, but not of methionine itself. A high level of dietary l-cysteine (2.5% or higher) is lethal for young chicks, but a similar level of dl-methionine, l-cystine or N-acetyl-l-cysteine causes no mortality. A supplemental dietary level of 3.0% l-cysteine (7× requirement) causes acute metabolic acidosis that is characterized by a striking increase in plasma sulfate and decrease in plasma bicarbonate. S-Methylmethionine, an analog of S-adenosylmethionine, has been shown to have choline-sparing activity, but it only spares methionine when diets are deficient in choline and(or) betaine. Creatine, or its precursor guanidinoacetic acid, can spare dietary arginine in chicks.  相似文献

研究和评价了蝇蛆几丁低聚糖咀嚼片的调节血脂作用及其安全性。将50只实验动物(大鼠)随机分为普通饲料对照组、高脂饲料对照组和3个实验组,实验组分别喂以不同剂量蝇蛆几丁低聚糖咀嚼片和高脂饲料。饲养28天后,分别测定大鼠血清总胆固醇(total cholesterol, TC)、血清总甘油三酯(triglyceride, TG)、血清高密度脂蛋白胆固醇(high density lipoprotein cholesterol, HDLC)的含量。用Horn's法对蝇蛆几丁低聚糖咀嚼片的急性毒性进行研究,连续观察7天,记录各组动物的中毒反应情况和死亡只数,计算咀嚼片对小鼠的半数致死剂量。同时对几丁低聚糖咀嚼片作了调节血脂的动物试验,研究结果表明,蝇蛆几丁低聚糖咀嚼片对大鼠血清总胆固醇、总甘油三酯有明显降低和对血清高密度脂蛋白胆固醇有明显稳定作用,具有辅助降血脂作用;对咀嚼片进行急性毒性实验表明,蝇蛆几丁低聚糖咀嚼片对小鼠的半数致死剂量大于10 g/kg·bw,提示其基本无毒。

The effects of the addition of individual amino acids on methionine-induced hypercholesterolemia (experiment 1), and the interacting effects of dietary protein level and sulfur-containing amino acids and glycine on plasma cholesterol concentration (experiment 2) were studied in growing rats fed on a high cholesterol diet. In experiment 1, rats were fed on a 25% casein-0.75% methionine (25CM) diet containing 2.5% of individual amino acids for 2 weeks. Methionine-induced hypercholesterolemia was prevented by the concurrent addition of glycine or serine, but the other amino acids tested (alanine, threonine, leucine, phenylalanine, lysine, arginine, and glutamic acid) had no effect. Histidine rather enhanced the hypercholesterolemia. In experiment 2, rats were fed on a 10%, 25%, or 50% casein diet containing 0.75% methionine, 0.60% cystine, 0.63% taurine, 2.5% glycine, or 0.75% methionine +2.5% glycine for 3 weeks. Dietary addition of 0.75% methionine increased the plasma cholesterol concentration for the 25% and 50% casein diets, but it decreased the plasma cholesterol for the 10% casein diet. When the addition level of methionine was doubled in the 10% casein diet, the plasma cholesterol concentration was significantly higher for the 1.5% methionine-added diet than for the 0.75% methionine-added diet. Cystine and taurine lowered plasma cholesterol for all dietary casein levels. Methionine-induced hypercholesterolemia with 25% and 50% casein diets was prevented by the glycine supplementation. These data suggest that sulfur-containing amino acids and glycine are important in plasma cholesterol regulation.  相似文献

The objective of this study was to evaluate the effect of supplementing a CP-reduced diet with rumen-protected methionine on growth performance of Fleckvieh bulls. A total of 69 bulls (367 ± 25 kg BW) were assigned to three feeding groups (n = 23 per group). The control (CON) diet contained 13.7% CP and 2.11 g methionine/kg diet (both DM basis) and was set as positive control. The diet reduced in CP (nitrogen) (RED) diet as negative control and the experimental RED + rumen-protected methionine (MET) diet were characterised by deficient CP concentrations (both 9.04% CP). The RED + MET diet differed from the RED diet in methionine concentration (2.54 g/kg DM vs. 1.56 g/kg DM, respectively) due to supplementation of rumen-protected methionine. Rumen-protected lysine was added to both RED and RED + MET at 2.7 g/kg DM to ensure a sufficient lysine supply relative to total and metabolisable protein intake. Metabolisable energy (ME) and nutrient composition were similar for CON, RED, and RED + MET. Bulls were fed for 105 days (d) on average. Individual feed intake was recorded daily; individual BW was recorded at the beginning of the experiment, once per month, and directly before slaughter. At slaughter, blood samples were collected and carcass traits were assessed. Reduction in dietary CP concentration reduced feed intake, and in combination with lower dietary CP concentration, daily intake of CP for RED and RED + MET was lower compared with CON (P < 0.01). Daily ME intake was reduced in RED and RED + MET compared with CON (P < 0.01). Consequently growth performance and carcass weights were reduced (both P < 0.01) in both RED and RED + MET compared with CON. Supplemental rumen-protected methionine was reflected in increased serum methionine concentration in RED + MET (P < 0.05) as compared to RED but it did not affect growth performance, carcass traits and serum amino acid (AA) concentrations, except for lysine which was reduced (P < 0.01) compared to CON and RED. In conclusion, bulls fed RED or RED + MET diets were exposed to a ruminal CP deficit and subsequently a deficit of prececal digestible protein, but methionine did not appear to be the first-limiting essential AA for growth under the respective experimental conditions.  相似文献

AimsIt has been suggested that variations in the proportions of some dietary amino acids can slow down aging. In this study, the influence of amino acids other than methionine on aging was investigated.Main methodsRats were fed either with normal (ND) or except methionine, protein restricted diet (PREMD) for 4 months and oxygen radical production, oxidative protein and DNA damage along with telomere length and telomerase activity were evaluated in the liver.Key findingsExcept mitochondrial superoxide production rate, feeding with PREMD significantly decreased the oxygen radical production rate and protein carbonyl levels in the homogenate and mitochondria of 16-month-old rats. Feeding with PREMD prevented 8-OHdG formation in mitochondrial DNA but not in the genomic DNA. Although liver telomerase activities of rats receiving either ND or PREMD seemed to have some variations, these did not reach a statistical significance. Feeding with PREMD conserved the telomere length in the liver. The telomere length of 8- and 16-month-old rats fed PREMD was similar, 16-month-old rats fed ND had telomeres shortened by 36% (p < 0.05).SignificanceLong-term restriction of the amino acids other than methionine may decrease oxygen radical generation and oxidative damage of cellular constituents, and may also prevent telomere shortening in rat liver.  相似文献

Sarcosine and methionine sulfoxide were investigated in several wild or laboratory-reared symbiotic and aposymbiotic strains of Sitophilus oryzae and S. zeamais. The amino acid composition of fourth-instar larvae indicated that a high level in sarcosine found together with a low level of methionine sulfoxide were biochemical characteristics of the aposymbiotic state in this genus. Nutritional experiments demonstrated that the synthesis of these two amino acids depended on dietary precursors. Since sarcosine and methionine sulfoxide are both methionine derivatives, it is therefore suggested that methionine metabolism in Sitophilus larvae might differ according to the presence or the absence of the symbiotic bacteria.  相似文献

Commercially produced haem fraction of abattoir blood with a dry matter content of 35% was treated with formic acid of 80–85% purity at the rate of 3:100 (v/v) which transformed the “haem” from a liquid to a solid state and reduced pH from 7.2 to 3.6. The product was included at approximately 5% of the dietary dry matter for pigs over the live weight range 33–80 kg. In one experiment the diet containing the haem was compared with diets containing either 7 or 10% herring meal on an air dry basis. The pigs on the 10% herring meal diet grew slightly, but non-significantly, faster and more efficiently than pigs on the other two diets. The carcases of the pigs on the haem diet contained less lean and more fat than those on the herring meal diets, apparently because of dietary deficiencies of isoleucine and methionine + cystine. In the second experiment, diets with and without the haem fraction were formulated by linear programme taking ingredient costs into account. The performance and carcase composition of pigs on the haem diet were equal to or better than those on the control diet.The total lysine content of the haem fraction of abattoir blood treated with formic acid was 115 g/kg dry matter with an availability (1-fluoro-2,4-dinitrobenzene-reactive lysine) of 86%. In relation to the requirements of the finishing pig the limiting amino acids appear to be isoleucine and methionine + cystine.  相似文献

Fishmeal has long been a staple protein feedstuff for fish, but its global shortage and high price have prompted its replacement with alternative sustainable sources. In this experiment involving largemouth bass (a carnivorous fish), a new mixture of feedstuffs (45% poultry byproduct meal, 30% soybean meal, 15% blood meal, and 10% krill shrimp meal) was added to low (14.5%) fishmeal diets along with 0.0%, 0.5% taurine, 0.5% methionine, or 0.5% taurine plus 0.5% methionine (dry matter basis). The positive control diet [65.3% fishmeal (46% crude protein on dry matter basis)] and all low-fishmeal diets contained 40% true protein and 10% lipids. There were 3 tanks per treatment group (20 fish/tank). Fish with the mean initial body weight of 16.6 g were fed to satiety twice daily. Compared with the unsupplemented low-fishmeal group, supplementing either 0.5% methionine or 0.5% methionine plus 0.5% taurine to the low-fishmeal diet improved (P < 0.05) the growth, feed utilization, retention of dietary protein and lipids, and health of largemouth bass, reduced (P < 0.05) the occurrence of black skin syndrome from ~ 40 to ~ 10%. Histological sections of tissues from the fish with black skin syndrome showed retina degeneration, liver damage, and enteritis in the intestine. Compared with methionine supplementation, supplementing 0.5% taurine alone to the low-fishmeal diet did not affect the growth or feed efficiency of fish and had less beneficial effects (P < 0.05) on ameliorating the black skin syndrome. These results indicated that: (a) the basal low-fishmeal diet was inadequate in methionine or taurine; and (b) dietary supplementation with methionine was an effective method to improve the growth performance, feed efficiency, and health of largemouth bass. Further studies are warranted to understand the pathogenesis of the black skin syndrome in largemouth bass.

Abstract— Adult mice were fed standard diets that were enriched with selected amino acids, i.e. 3% methionine, 6% valine, or 8% lysine. These diets caused the following changes in the amino acid pool of the brain measured at 7 and 21 days. The high methionine diet resulted in 50-fold higher levels of methionine and cysteine and somewhat lower levels of serine and glutamine. The valine and lysine-enriched diets also caused 2- to 4-fold increases in valine and lysine contents of brain, respectively. In spite of the large changes in amino acid levels, however, there were essentially no changes in aspartate: α-ketoglutarate, alanine: α-ketoglutarate, ornithine: α-ketoglutarate, methionine: α-ketoglutarate, and the branched chain aminotransferase activities of brain 3, 10, and 21 days after the onset of the dietary regimen. In contrast, these diets produced significant changes in some of these enzyme activities in liver. Changes in liver included a 2-fold increase in ornithine and alanine aminotransferase activities with the methionine-enriched diet. Liver ornithine aminotransferase activity also increased slightly in animals fed the valine-enriched or lysine-enriched diet.  相似文献
