Development of dark-grown “Clark” soybean (Glycine max [L.] Merr.) seedlings is abnormal at 25 C but normal at 20 and 30 C. At 25 C, hypocotyls swell and fail to elongate normally; lateral root formation and seedling ethylene evolution are enhanced.

Co2+ promoted hypocotyl elongation of etiolated “Clark” soybean seedlings by 28% when grown at 25 C. The same growth-promoting concentration reduced hypocotyl thickness and primary root elongation by 28 and 43%, respectively. Co2+ inhibited ethylene production both of intact seedlings and of apical 1-centimeter hypocotyl segments with attached epicotyls and cotyledons by 65 and 60%, respectively. These results suggest that Co2+ exerts its effects on the hypocotyl growth by inhibiting ethylene production, and also confirm our previous conclusion that abnormal ethylene production at 25 C is responsible for the inhibition of hypocotyl elongation and for its swelling.

In Clark and Shelby soybean (Glycine max [L.] Merr.) seedlings, hypocotyl elongation was inhibited and hypocotyl swelling and root dry weight were increased by a temperature of 25 C. At 20 and 30 C, development was normal, as was development of Hawkeye and Mandarin soybean seedlings at all three temperatures. Dry matter distribution at 9 days indicates that inhibition of hypocotyl elongation is not due to a lack of translocation from cotyledons, but to a diversion of dry matter from hypocotyl to root. Ethylene evolution by Clark seedlings at 25 C exceeds that at 20 and 30 C. At all three temperatures, Mandarin seedlings' ethylene evolution is at the same low rates as those of Clark at 20 and 30 C. Clark's enhanced rate at 25 C precedes the deceleration of hypocotyl elongation occurring at 5 days. The abnormal effects of a temperature of 25 C on Clark seedlings' development is partially reversed by CO2 and GA3 and can be duplicated in Mandarin by applying ethylene. In Clark, effects of the temperature can be further accentuated by indoleacetic acid, which stimulates ethylene evolution, and by applying ethylene to the seedlings. It is concluded that the temperature-induced symptoms, similar in most respects to the well known “triple response” of legume seedlings, are caused by abnormally high levels of ethylene in tissues of the anomalous cultivars.  相似文献

Summary Ethylene and 2.4-dichlorophenoxyacetic acid (2.4-D) inhibited the growth of etiolated soybean (Glycine max cv. Hawkeye) seedlings causing tissue swelling and an increase in RNA, DNA and protein content in the subapical hypocotyl tissue. 2.4-D increased ethylene evolution from soybean seedlings and it was found that some of the effect of this herbicide on soybeans was due to the increased ethylene production.Ethylene is responsible in part for the inhibition of elongation and of increase in weight that occurs at supraoptimal concentrations of 2.4-D applied to excised hypocotyl sections. Abscisic acid inhibits 2.4-D-induced tissue swelling and ethylene production in the excised, elongating section. The cotyledons of the soybean seedlings appear to regulate the 2.4-D-induced production of ethylene and the roots are necessary for the 2.4-D-induced tissue swelling.  相似文献

The apical 1-cm hypocotyl of dark-grown `Clark' soybean (Glycine max [L.] Merr.) seedlings produced ethylene at rates of 7 to 11 nanoliters per hour per gram when attached to the cotyledons. Such physiologically active rates occurred prior to the deceleration of hypocotyl elongation caused by the temperature of 25 C.

Daily exposure of the etiolated seedlings to red light promoted hypocotyl elongation and prevented its lateral swelling. Red light treatment also caused a 45% decrease in ethylene production. Far red irradiation following the red treatment reversed the red effects, suggesting that the ethylene intervenes as a regulator in the phytochrome control of `Clark' soybean hypocotyl growth at 25 C.

Cucumber (Cucumis sativus L. cv. Poinsett 76) seedlings withfully expanded cotyledons, and excised cotyledons, first trueleaves, hypocotyl segments and fruit mesocarp discs were exposedto vapours from a series of aqueous alcohol solutions of 0 to320 mM methanol, ethanol, n-propanol, n-butanol, and n-pentanolduring chilling at 2.5C for 5 d. Certain concentrations ofeach alcohol reduced subsequent chilling-induced ion leakagefrom the cotyledons and leaves. Exposure of cotyledons to certainmethanol or ethanol solutions also reduced chilling-inducedethylene production, but not carbon dioxide production. In contrast,exposing cucumber seedlings with fully expanded cotyledons tothe same series of alcohol concentrations that resulted in reducedchilling-induced ion leakage and ethylene production of excisedcotyledons actually increased chilling injury of the seedlings.The hypocotyl region directly below the cotyledons was the siteof chilling-induced injury and contained the most chilling-sensitivehypocotyl tissue. Exposing hypocotyl segments excised from thissensitive region to alcohol solutions did not significantlyreduce chilling-inducedion leakage. Exposing excised cucumbercotyledons or hypocotyl segments to equivalent osmotic nonvolatilesolutions of mannitol and glycerol at 2.5C or to alcohol solutionsat 12.5C had no significant effect on the rate of ion leakage.For the series of alcohols used, the relationship between thelog of the alcohol concentration that minimized chilling-inducedion leakage from cucumber cotyledon discs held at 2.5C for5 d and the log of the partition coefficient of the alcoholinto olive oil or the log of the molecular weight of the alcoholswas highly significant. The same concentrations of alcoholsthat reduced chilling-inducedion leakage also reduced stomatalaperture as measured as decreased porosity of excised cotyledons.The correlation between reduced chilling injury and stomatalconductance of cotyledons exposed to a series of ethanol solutionswas highly significant. It appears that alcohols may reducechilling injury of cucumber cotyledons by inducing stomata closure.Sufficient endogenously synthesized ethanol accumulated in discsheld in N2 at 10C for 1 d to confer tolerance to chilling at2.5C for 5 d. Two important environmental signals, shade light, where the red/far-red (R/FR) light ratio is reduced, and elevated temperatures can each promote shoot growth. We examined their interactions using hypocotyl elongation of young sunflower (Helianthus annuus) seedlings, and we did this in the context of a possible hormonal mechanism for the growth increases that were induced by each environmental signal. Seedlings were subjected to combinations of six different temperatures (10, 15, 20, 25, 30 and 35°C) and four R/FR ratios (normal at 1.2 and reduced at 0.9, 0.6 and 0.3). Hypocotyl length was significantly increased by each of elevated temperature and FR enrichment. The magnitude of elongation induced by FR enrichment (low R/FR ratios) was dependent on temperature, with maximal effects of FR enrichment being seen at 20°C. Hypocotyl tissue concentrations of four endogenous gibberellins (GAs) and abscisic acid (ABA) were measured using the stable isotope dilution method. Hypocotyl ethylene evolution was also assessed. Thus, hypocotyl growth in both normal and shade light is highly dependent on temperature, with the most significant increases in FR-induced growth occurring at 20 and 25°C. A causal involvement of endogenous hormones, especially the GAs, in the growth that is induced by elevated temperatures, as well as in FR-induced growth, is strongly implied, with temperature being the stronger signal.  相似文献

Auxin induced an increase in the rate of ethylene productionby hypocotyl segments of etiolated mungbean seedlings aftera 1 hr lag period. The increase in the production rate was greatestwith segments immediately below the cotyledonary hook. Effectiveconcentrations of indoleacetic acid ranged from 1 µm to0.5 mM. Length of the lag period was the same regardless ofthe interval between segment excision and the addition of auxin.Auxin-induced ethylene production was inhibited by cycloheximidebut not by chloramphenicol. Light-grown sunflower seedlings contain 10–7–3 x10–7 M indoleacetic acid (IAA). The even distributionof this endogenous IAA in straight hypocotyls does not changeduring phototropic curvature. The diffusate from hypocotylscontains substance(s) influencing the elongation rate of Avenacoleoptile segments but hardly any IAA. Phototropic curvature of the hypocotyl requires the presenceof illuminated cotyledons. Illumination of cotyledons inhibitshypocotyl growth. It is concluded that the phototropic response of the sunflowerhypocotyl is regulated by factors promoting and inhibiting cellelongation other than IAA.  相似文献

Exposure of gibberellic acid-treated seedlings of a monoecious cucumber cultivar `Chipper' (Cucumis sativus L.) to ethylene caused thickening of the hypocotyl, inhibited longitudinal growth, and had no effect on fresh weight. Downward curvature of cotyledons was increased by the presence of ethylene. A gynoecious breeding line, `Gy 3,' had thicker hypocotyls and displayed its cotyledons in a more downward position than `Chipper'. Excised hypocotyls of the gynoecious seedlings produced three times as much ethylene as did the monoecious Chipper hypocotyls. Thus, ethylene may play a role in the regulation of cucumber seedling morphology.  相似文献

Co2+ promoted elongation of hypocotyl segments of light-grown cucumber (Cucumis sativus) seedlings. Time course and dose response data are presented and interactions with IAA, gibberellin, cyclohexanol, and cotyledons described. Segments without cotyledons responded to Co2+ only if grown in gas-tight vessels with IAA added. When bases of cotyledons were ringed with an inhibitor of auxin transport, Co2+ caused no growth promotion in the hypocotyl. Co2+ prevented lateral swelling of hypocotyls treated with supraoptimal IAA. Removal of ethylene from the atmosphere reduced the Co2+ response, but Co2+ did not counteract the inhibitory effect of increased ethylene levels. These results are consistent with the hypothesis that Co2+ promotes hypocotyl elongation by inhibiting ethylene production. The hypothesis was confirmed by a direct demonstration that Co2+, at growth-promoting concentrations, powerfully inhibited ethylene production in the cucumber hypocotyl.  相似文献

Hypocotyl elongation under white fluorescent light was aboutdoubled in dwarf watermelon (Citrullus lanatus0 (Thunb.) Matsu.and Nakai) seedlings treated with 0.1 to 0.3 µg apicaland 3 x 10–6 to 10.3 M root applications of 6-benzyladenine(BA). BA-enhancement of growth occurred primarily during thefirst 48 h after treatment. Increased hypocotyl length in BA-treatedseedlings was attributed more to longer cells than to an increasein cell number. Early hypocotyl growth of normal seedlings wasalso significantly enhanced by BA although final hypocotyl lengthwas not substantially affected. Benzyladenine caused expansion of cotyledons and, at higherdoses, lateral expansion of hypocotyls. BA-induced increasesin fresh weight of cotyledons and hypocotyls were accompaniedby an increase in dry weight of hypocotyls at the expense ofroots which had less dry matter than untreated seedlings.  相似文献

The role of cotyledons in hypocotyl elongation caused by gibberellicacid was studied using young seedlings of lettuce, Lactuca saliva,var. ‘Grand Rapids’. Removal of cotyledons fromintact seedlings resulted in a depression of hypocotyl elongationcaused by gibberellic acid. Gibberellic acid-induced hypocotylelongation in decotylized seedlings, was however, substantiallyenhanced by incubating the seedlings together with excised cotyledons.The exudate from excised cotyledons also enhanced the effectof gibberellic acid on hypocotyl elongation in decotylized seedlings.This active principle (named the cotyledon factor) in the cotyledonexudate was stable against heating at 100?C for 15 min, permeatedthe dialysis membrane, and was extractable with ethyl acetate.Biological activity of the cotyledon factor was not replacedby indole-3-acetic acid, kinetin, cyclic AMP, vitamins, sucroseor inorganic nutrients. Polar transport of kinetin-8-14C occurred in segments of petioles, hypocotyls, and roots of radish (Raphanus sativus L.). The polarity was basipetal in petioles and hypocotyls and acropetal in roots. In segments excised from seedlings with fully expanded cotyledons, indole-3-acetic acid was required for polarity to develop. In hypocotyl segments isolated at this stage, basipetal and acropetal movements were equal during the first 12 hours of auxin treatment after which time acropetal movement declined. Pretreatment with auxin eliminated this delay in the appearance of polarity. In hypocotyl segments excised from seedlings with expanding cotyledons, exogenous auxin was unnecessary for polarity. Potassium cyanide abolished polarity at both stages of growth by allowing increased acropetal movement. The rate of accumulation of kinetin in receiver blocks was greater than the in vivo increase in cytokinin content of developing radish roots.  相似文献

Hypocotyl-derived callus from the Helianthus annuus L. inbred line SS415B regenerated significantly more plants if the seedlings were grown in the light. The difference between light- and dark-grown seedlings was not correlated with differences in seedling ethylene production, but seemed to be due to a difference in sensitivity to ethylene at a specific time during seedling growth. Treating 3-day-old dark-grown seedlings with 10 μ M aminoethoxyvinylglycine (AVG) effectively inhibited ethylene production for at least 7 days. Hypocotyl callus derived from AVG-treated seedlings gave the same amount of regeneration as callus from light-grown seedlings. Promotion of regeneration by AVG was not seen unless the 3-day-old seedlings were grown for 4 additional days prior to culturing hypocotyl explants. The effects of AVG could be reversed by treatment with 1-aminocyclopropane-1-carboxylic acid (ACC) during these 4 days. After the 4 days, ACC was no longer effective.  相似文献

Hypocotyl hooks have been shown to influence greening in excised cucumber (Cucumis sativus) cotyledons. The properties of the lag phase are greatly affected by the presence or absence of the hook tissue. A 45-second light pretreatment followed by 4 hours of darkness is sufficient to remove the lag phase from cotyledons with hooks, while hookless cotyledons require 2 hours of continuous illumination followed by 1 hour of dark incubation to break the lag phase. The effect of hooks on cotyledon greening is enhanced if the hooks are shielded from light. Cutting off the hooks after lag phase removal caused a marked decrease in chlorophyll accumulation in the cotyledons. These observations may indicate that the hypocotyl hooks produce a substance or substances needed in the greening process, which are translocated to the cotyledons. Indoleacetic acid, abscisic acid, gibberellin A3, 6-benzylamino purine and δ-aminolevulinic acid do not show any activity; on the other hand, ethylene appears to replace partially the hypocotyl hooks.  相似文献

The effect of the herbicide chlorsulfuron (2-chloro-N-[(4-methoxy - 6 - methyl -1, 3,5 - triazin - 2 - yl)aminocarbonyl]benzenesulfonamide) on ethylene production in light-grown sunflower (Helianthus annuus L.) seedlings was examined. Application of chlorsulfuron to the apex stimulated ethylene production in all tissues examined: cotyledons, hypocotyls, and roots. The greatest stimulation occurred in the upper portion of the hypocotyl adjacent to, and including, the cotyledonary node. Ethylene evolution from hypocotyls excised from treated seedlings was stimulated over control levels 1 day after herbicide application and reached a maximum (approx. 75 x control or 17 nl/g f wt/h) 2 to 3 days after treatment. Labeling and inhibitor studies indicated that the ethylene produced was derived primarily from methionine. Chlorsulfuron treatment stimulated the rate of accumulation of the ethylene precursor, 1-aminocyclopropane-1-carboxylic acid (ACC), as well as the ability of the tissue to convert exogenous ACC to ethylene. Chlorsulfuron had little effect on ethylene production when administered to the hypocotylsin vitro. Removal of the cotyledons from treated seedlings reduced the rate of ethylene evolution from the hypocotyls. These results suggest that stimulation of ethylene production in sunflower hypocotyls by chlorsulfuron is not a wound response but rather is dependent on factors derived from the cotyledons.  相似文献

The photosynthetic rate of cucumber cotyledons (Cucumis sativuscv. Dahla) reached a maximum value of 12 mg dm–2 h–1,10 d after emergence. In 12-d-old seedlings removal of one cotyledondoubled the CO2 fixation rate of the other, as observed 3 dafter treatment. When the primary leaf was removed, the photosyntheticrate of the cotyledons was decreased by 33%. At this stage ofgrowth elimination of the roots as a sink for assimilates bygirdling the hypocotyl affected neither the photosynthetic ratenor the carbohydrate content of the cotyledons. By contrast,in 18-d-old seedlings removal of the first leaf brought abouta 42% increase in the photosynthetic rate of the cotyledons.The simultaneous removal of the first leaf and one cotyledondoubled the rate of CO2 fixation of the remaining cotyledon.Girdling the hypocotyl lowered the photosynthetic rate of thecotyledons by 73%. In both 12- and 18-d-old seedlings a decreaseor increase in the sink-source ratio was correlated with anincrease or a decrease respectively in the carbohydrate contentof the cotyledons. The stomatal resistance of the cotyledonswas not affected by any of the treatments. Hypocotyl sections with and without the cotyledons were cutfrom bean seedlings and incubated under white light of 6000lux. The cotyledons had an inhibitory effect as well as a promotiveeffect on hypocotyl growth. The former effect was more strikingin the dwarf variety, and the latter in the tall variety. Whenthe hypocotyl units were exposed to light for shorter times(6 hr or less) or incubated under weaker light (1600 and 50lux), the inhibitory effect of the cotyledons decreased greatly,and in the tall variety the presence of cotyledons producedno inhibition, but a promotion of hypocotyl growth. GA treatmentenhanced hypocotyl growth and counteracted the growth inhibitioncaused by the cotyledons. On the whole, the GA effect was moremarked in the tall variety than in the dwarf. The elongation of bean hypocotyls may be controlled by a balancebetween the inhibitory and promotive effects of cotyledons,and the predominance of the former over the latter may be oneof the causes for expressing dwarfing. The amount of IAA-C14 transported basipetally through excised hypocotyl sections was strongly affected by the pH of the donor blocks, less so by the pH of the receivers. The effect of donor pH was mostly on uptake. The small amount of acropetal movement was not noticeably affected by pH. Sucrose added to the donor resulted in increased basipetal transport. The time-course of C14 movement into basal receivers followed a linear course from 1.5 to 3 hr as expected, but there was no net loss from the donors until after 30-45 min. The usual type of velocity calculation, which assumes uptake starting from zero time, would therefore be lower than the true value. Basipetal transport through segments cut from various positions in the hypocotyl and from seedlings of various ages was maximal in 6-8-day-old hypocotyl segments cut 25-30 mm below the cotyledons. Acropetal movement was minimal at all positions of all ages tested.  相似文献

myo-[2-3H]Inositol was fed to bean seeds by imbibition and itsmetabolic fate was studied during germination and seedling growth.The largest amount of myo-inositol was taken up from a 500 HIMsupply (8 mg/seed) and the highest percentage was from 1 HIM(29%). myo-Inositol was incorporated to new cell wall polysaccharidesof hypocotyl and roots, mostly as uronic acid and pentose residues.In the 80% ethanolinsoluble cell walls of hypocotyls at 3, 4and 5 days after imbibition, 47 to 52% of 3H was detected asuronic acids, 20 to 24% as arabinose and 11 to 19% as xylose.Glucogenesis from myo-inositol was low: less than 6% was recoveredas hexoses. The 3H in uronic acid and arabinose residues decreasedwith increasing age (i.e. 0 to 6 cm from cotyledons) and increasedin older segments (further than 6 cm from cotyledons). In theoldest segment of 5-day-old hypocotyl (> 10 cm), 3H in thesugar residues was more than that in the youngest part (0–2cm). On the other hand, 3H in xylose residues increased steadilyin the older part, but did not exceed that in arabinose. The results show that the myo-inositol oxidation pathway functionsin growing hypocotyls and roots of bean seedlings to provideexclusively uronic acid and pentose units for cell wall synthesis.Results also show that incorporation of arabinose and uronicacids derived from myo-[2-3H]inositol to cell wall polysaccharidesis active in two regions of the hypocotyl; first, for the constructionof the primary walls in the young, growing region of the hypocotyl,and second, for thickening of the walls after completion ofelongation growth. 1Supported by NSERC of Canada.
