The genetic diversity of 158 individuals from eight semi-wild populations from Yunnan Province was estimated using ISSR method (8 primers). The results revealed an extraordinarily high level of genetic diversity ( at species level,percentage of polymorphic loci PPB = 91.04% , effective number of alleles Ne = 1.5244 , Nei′s (1973 ) gene diversity He= 0.3070, and Shannon′s information index Ho = 0 . 4618 ; at population level, PPB = 55. 04% , Ne = 1.3826, Nei′s (1973) gene diversity He = 0.2171, and Shannon′s information index Ho = 0.3178). The level of genetic differentiation between populations is lower than that among populations . The low level of genetic differentiation among populations was detected, based on Nei′s genetic diversity analysis (29.44%), and AMOVA (36.50%). There is no associations between geographical distance and genetic identity.We suggest that Jatropha curcas of Yunnan Province might not be introduced from the same place.  相似文献

应用ISSR 分子标记方法对采自云南的8 个居群的小桐子( Jatropha curcas) 共158 个个体进行遗传多样性分析。8 个ISSR 引物共扩增到了67 个位点, 其中61 个是多态性位点。分析结果表明: (1) 云南小桐子的遗传多样性水平很高。在物种水平上, 平均每个位点的多态位点百分率PPB = 91.04% , 有效等位基因数Ne = 1.5244, Nei′s 基因多样性指数He= 0.3070, Shannon 多样性信息指数Ho = 0.4618; 在居群水平上, PPB = 55.04%, Ne = 1.3826, He = 0.2171, Shannon 多样性信息指数Ho = 0.3178。(2) 居群间的遗传分化低于居群内的遗传分化。基于Nei''s 遗传多样性分析得出的居群间遗传多样性分化系数Gst = 0.2944。AMOVA分析显示: 云南小桐子的遗传变异主要存在于居群内, 占总变异的63.50%, 居群间的遗传变异占36.50%。(3) 居群间的地理距离及遗传一致度并不存在相关性。鉴于以上指标, 我们推测云南小桐子可能来自不同的地区。  相似文献

Xylella fastidiosa is a xylem-limited bacterium that causes various diseases, among them Pierce's disease of grapevine (PD) and almond leaf scorch (ALS). PD and ALS have long been considered to be caused by the same strain of this pathogen, but recent genetic studies have revealed differences among X. fastidiosa isolated from these host plants. We tested the hypothesis that ALS is caused by PD and ALS strains in the field and found that both groups of X. fastidiosa caused ALS and overwintered within almonds after mechanical inoculation. Under greenhouse conditions, all isolates caused ALS and all isolates from grapes caused PD. However, isolates belonging to almond genetic groupings did not cause PD in inoculated grapes but systemically infected grapes with lower frequency and populations than those belonging to grape strains. Isolates able to cause both PD and ALS developed 10-fold-higher concentrations of X. fastidiosa in grapes than in almonds. In the laboratory, isolates from grapes overwintered with higher efficiency in grapes than in almonds and isolates from almonds overwintered better in almonds than in grapes. We assigned strains from almonds into groups I and II on the basis of their genetic characteristics, growth on PD3 solid medium, and bacterial populations within inoculated grapevines. Our results show that genetically distinct strains from grapes and almonds differ in population behavior and pathogenicity in grapes and in the ability to grow on two different media.  相似文献

 利用AFLP分子标记技术, 运用EcoRⅠ/MseⅠ双酶切组合, 选用多态性高、分辨力强的4对选择性扩增引物组合E32/M48、E33/M61、E35/M61、E33/M62分别对四川省3个野生核桃(Juglans regia)种群和1个野生铁核桃(J. sigillata)种群共46个样品进行遗传多样性分析、居群遗传结构分析及种属关系探讨。结果表明: 1)共扩增出244个遗传位点, 其中146个多态位点, 多态率为59.84%; 核桃群体组和铁核桃群体的多态性百分率分别为55.33%和52.05%, 两个物种遗传多态性水平相当; 核桃群体组所检出的位点平均有效等位基因数Ae、Nei’s基因多样度H、平均Shannon信息指数I分别为1.322 9、0.190 8和0.286 3, 而铁核桃群体分别为1.339 9、0.196 1和0.289 8, 铁核桃群体遗传多样性水平略高于核桃群体。2)群体间特异带及群体间共有带占总扩增带数的15.16%, 其中铁核桃群体特异谱带最多, 群体特异谱带揭示了群体间的遗传差异及相似性。3) Shannon信息指数(I)、Nei’s基因多样性指数(H)和分子方差分析(AMOVA)表明核桃遗传多样性在群体间和群体内的分布分别为14.36%和85.64%、12.6%和87.4%、11.07%和88.93%, 表明群体内的遗传多样性大于群体间的遗传多样性; 核桃群体组与铁核桃群体的变异主要存在于群体组内, 组间的遗传变异仅占总变异的9.35%, 两者间的遗传分化系数Gst为0.093 5, 与AMOVA分析结果一致。4) 4个群体的Nei’s遗传距离在0.038 2~0.069 2之间, 遗传一致度在0.933 2~0.962 5之间, 表现出较高的遗传相似性; 运用Nei’s遗传一致度对供试种群进行了UPGMA聚类, 结果表明核桃的3个群体优先聚类, 大渡河流域群体与甘南地区群体聚类最近。AFLP所检测出的结果既是核桃与铁核桃生物学特性的反映, 又是其各自生态学特性的反映, 该研究结果对核桃种质资源的保护和育种提供一定的理论依据。  相似文献

利用 RAPD技术分析了分布于浙江省天台山 3个不同海拔高度的天然大血藤群体的遗传多样性、遗传分化以及与环境因子的相关性。 13种随机引物在 3 6株个体中共检测到 88个可重复的位点 ,其中多态位点 74个 ,总多态位点百分率为84.0 9% ,大血藤具有丰富的遗传多样性。 Shannon信息指数显示的遗传多样性以海拔 950 m的群体为最高 ,其次是海拔 73 0 m的群体 ,最低的是海拔 52 0 m的群体 ;群体内的遗传多样性占总遗传多样性的 43 .68% ,群体间的遗传多样性占 56.3 2 %。 Nei指数估计大血藤群体间的遗传分化系数为 0 .540 6,大血藤群体间的基因流很低。大血藤海拔 73 0 m群体与海拔 52 0 m群体的遗传相似度较高 ,海拔 950 m群体与其它两群体的遗传相似度较低。大血藤群体内的遗传多样性与土壤总氮呈极显著的正相关。  相似文献

为对比显性标记应用于居群遗传结构研究时不同统计参数的适用性,利用RAPD技术对中国5个居群的100个疣粒野生稻个体进行了遗传结构分析。在衡量居群遗传多样性水平时,多态位点比率(PPB)会低估遗传变异的量,其价值不如Shannon多样性指数和Nei基因多样性指数,而采用Nei指数时不必进行Lynch-Milligan矫正。对个体间遗传关系进行分析时,17种遗传相似性指数矩阵两两之间的Mantel检测都表现出极显著的相关性(r>0.95,t>t  相似文献

利用SRAP分子标记技术,对7个居群79株金毛狗进行遗传多样性分析。结果表明:10对SRAP引物组合共得到107条扩增条带,多态性条带比率为85.98%,Nei’s基因多样性指数为0.229 6,Shannon’s多样性指数为0.358 6,表明金毛狗居群水平具有较高的遗传多样性;金毛狗7个居群的总基因多样度为0.229 6,居群内遗传多样度为0.135 4,居群间的遗传分化指数为0.410 6,表明有41.06%的变异存在于居群间,有58.94%的变异存在于居群内;居群间基因流为0.717 8,表明居群间基因交流频率较低;遗传一致度和UPGMA聚类分析结果显示,生境条件相似的居群优先聚集,说明金毛狗种质亲缘关系与地理分布相关性不显著。  相似文献

Quercus acutissima (Fagaceae), a deciduous broad-leaved tree, is an important forest element in hillsides of South Korea. We used allozyme loci, Wright's F statistics, and multilocus spatial autocorrelation statistics to examine the distribution of genetic diversity within and among three local populations and the spatial genetic structure at a landscape scale (15 ha, 250 × 600 m) on Oenaro Island, South Korea. Levels of genetic diversity in Q. acutissima populations were comparable to mean values for other oak species. A moderate but significant deficit of heterozygotes (mean F(IS) = 0.069) was detected within local populations and low but significant differentiation was observed among populations (F(ST) = 0.010). Spatial autocorrelation analyses revealed little evidence of significant genetic structure at spatial scales of 100-120 m. The failure to detect genetic structure within populations may be due to intraspecific competition or random mortality among saplings, resulting in extensive thinning within maternal half-sib groups. Alternatively, low genetic differentiation at the landscape scale indicates substantial gene flow among local populations. Although wind-borne pollen may be the primary source of gene flow in Q. acutissima, these results suggest that acorn movement by animals may be more extensive than previously anticipated. Comparison of these genetic data for Oenaro Island with a disturbed isolated inland population suggests that population-to-population differences in internal genetic structure may be influenced by local variation in regeneration environment (e.g., disturbance).  相似文献

利用SRAP分子标记技术,对湖北省河岸带植物中华蚊母树的4个自然居群和1个迁地居群的遗传多样性和遗传结构进行了分析。结果表明,中华蚊母树物种具有较高水平的遗传多样性,7对SRAP引物进行PCR扩增的多态性位点百分率(PPF)为80.43%,每个位点的等位基因数(A)为2,有效等位基因数(Ae)为1.34,总遗传多样性Nei’s基因多样性指数(Hp)为0.215 9,Shannon信息多样性指数(I)为0.350 9。在居群水平上,5个居群总的遗传变异Ht为0.218 8,居群内的遗传变异Hs为0.193 4,居群间的遗传分化系数Gst为0.116 1,表明在总的遗传变异中有88.39%变异存在于居群内,仅11.61%存在于居群间,居群间的基因流Nm为3.807 2,表明居群间有较大程度的基因交流。UPGMA聚类分析和主成分分析显示中华蚊母树主要分为两个居群组,在长江三峡沿岸香溪和乐天溪由于遗传距离比较近聚为一小类再与高家堰聚为一大类,而沿渡河和三峡植物园居群聚为另一大类,表明迁地居群三峡植物园的中华蚊母树与来自巴东沿渡河居群的样本亲缘关系最近,且三峡植物园迁地保护居群基本保育了其遗传多样性总水平。同时在分析讨论了中华蚊母树遗传多样性与其繁育系统、生境及其起源进化的关系的基础上,评价了中华蚊母树的保护策略,并在评价保护成果的基础上,提出了今后进一步保育的策略。结果还表明SRAP标记是分析中华蚊母树遗传多样性和遗传结构非常可靠的一种标记,而且这是使用SRAP标记研究中华蚊母树的首次报道。  相似文献

采用ISSR分子标记对分布于山东省长岛县南部的北长山、南长山、大黑山和小黑山4个岛屿上的藜天然种群共81个个体进行了遗传多样性及遗传结构的研究。13个引物共检测到157个可重复的位点。遗传多样性研究结果表明:种内的多态位点比率为66.24%,具有较高的遗传多样性(Shannon(I)指数在物种水平上为0.332 0);种群间有一定的遗传分化,根据Gst值,种群间的遗传多样性占总群体的9.27%。遗传距离分析表明,XHS种群和NCS种群的遗传一致度最高,与地理距离无相关性。  相似文献

利用微卫星(SSR)标记对来自山西和陕西两省的7个翅果油树(Elaeagnus mollisDiels)种群进行遗传多样性和遗传结构研究。10对SSR标记共检测到126个位点,其中多态位点114个。在物种水平上,平均多态位点百分率为90.79%,有效等位基因数(Ne)、Nei基因多样性指数(H)和Shannon多样性指数(I)分别为1.6072、0.3166、0.4603;在种群水平上,多态位点百分率为61.99%,有效等位基因数(Ne)、Nei’s基因多样性指数(H)、Shannon多样性指数(I)分别为1.5445、0.2683、0.3815。遗传分化系数GST为0.2074,表明了翅果油树种群的遗传变异主要存在于种群内。基因流Nm为1.9111〉1,说明种群间基因交流可以阻止由于遗传漂变导致的遗传分化。聚类结果表明,翅果油树种群间的遗传距离与地理距离有一定的相关性,经Mantel检验,种群的地理距离与遗传距离之间呈正相关,但未达到显著水平(p〉0.05)。结果表明,遗传多样性水平与物种本身特性和不同干扰生境有关,濒危植物并不一定表现为遗传变异水平的降低。  相似文献

The genetic diversity within and among populations of Hepatacodium miconioides collected at three different altitudes in Tiantai Mountain,Zhejiang Province and its relationships to environmental factors were analyzed by random amplified polymorphic DNA(RAPD)technique.Amplification using 12 random primers of 60 plants and 122 repetitive loci were produced.The percentage of polymorphic loci of three populations ranged from 18.85% to 23.77% with an average of 21.86%,indicating the relatively low genetic diversity of H.miconioides.The average Shannon index of phenotypic diversity(0.1329)and Nei index(0.0925)within populations were relatively low.A distinct genetic differentiation existed among populations Of H miconioides in spite of the relatively small geographical distribufion.The average genetic diversity within populations of H.miconioides accounted for 33.58% of the total genetic diversity while the genetic diversity among populations accounted for 66.42% as estimated by the Shannon index of phenotypic diversity,The genetic differentiation among populations of H.miconioides was 0.6546,as estimated by Nei index.The gene flow estimated from Gsr was only 0.2656 and it indicated that gene flow among populations of H.miconioides was relatively low.The mean value of the genetic identity among populations of H.miconioides was 0.7126 and the average of genetic distance of H.miconioides was 0.3412.The genetic identity between populations at the elevation of 990m and at the elevation of 780 m was the highest.The genetic identity between population at the elevation 500 m and other two populations was relatively low.The correlation analysis showed that the genetic diversity within populations was significantly related with the soil total nitrogen.  相似文献

利用相关序列扩增多态性SRAP(Sequence-related amplified polymorphism)分子标记首次对4个暗纹东方鲀(Takifugu obscurus)群体(1个长江捕捞群体、1个放流群体和2个养殖群体)进行了遗传多样性分析。从49对引物组合中筛选出18对扩增条带清晰、稳定的引物组合对4个群体进行扩增,共获得231个位点,其中多态位点数为156个,总多态位点百分率为67.53%。4个群体内多态位点比例为54.98%—58.87%,群体的Nei’s基因多样性指数(H)为0.1992—0.2005,群体Shannon多样性指数(I)为0.2953—0.3016,群体间基因流(Nm)为4.1291。长江捕捞群体的多态位点比例、基因多样性指数、Shannon多样性指数均略高于其他三个群体。采用UPGMA法对4个群体进行聚类分析显示,上海养殖群体单独聚为一类,其余3个群体聚为另一类。AMOVA结果表明遗传变异主要来源于群体内,占总遗传变异的87.40%。以上结果表明,4个暗纹东方鲀群体具有较高的遗传多样性,群体间相似性较大,并且存在一定的基因交流。  相似文献

利用AFLP和SSAP分子标记,研究了松嫩平原4个以耐盐突变体为主要组成的短芒大麦(Hordeum brevisubulatium(Trin.)Link)种群的遗传多样性和种群遗传结构。由于2种分子标记原理稍有不同,获得分析结果显现出微小差异,但2种方法的结果仍保持了极显著的相关性(r=0.88,P<0.01)。AFLP和SSAP检测结果表明,短芒大麦种群多态位点比率P分别为23.3%和37.7%;Shannon多样性指数分别为0.10和0.14;Nei基因多样性指数分别为0.07和0.09;基因分化系数Gst分别为0.49和0.44。表明,各短芒大麦种群虽以单一突变种为主要组成,但经多年的种植,种群内已具有较高的遗传多样性。AMOVA和聚类分析结果也表明,虽然种群间遗传差异较大,但种群遗传变异仍主要存在于种群内。  相似文献

利用4对AFLP引物对来自安徽石台、湖北大悟和河南桐柏的3个省沽油群体进行遗传多样性分析,共扩增出489条带,其中多态性条带460条,多态性百分比为93.99%。不同群体Nei’s基因多样性指数(H)和Shannon’s信息指数(I)变化范围分别为0.1920~0.2046和0.2937~0.3151,其中湖北大悟群体遗传多样性最高。物种水平和种群水平的H分别为0.2190和0.1964,群体内变异占总变异的89.68%,表明省沽油遗传变异主要存在于各群体内部。3个群体的平均遗传距离为0.0292,UPGMA聚类分析结果说明省沽油各群体间亲缘关系较近并和地域具有相关性。建议省沽油的遗传资源保护应以种内遗传多样性的保护为主。  相似文献

利用RAPD技术对浙江省境内的七子花9个天然种群遗传多样性和遗传分化进行研究.结果表明,12种随机引物对180棵植物进行检测,共得到164个可重复的位点.多态位点百分率在14.60%~27.44%(平均为20.73%),以括苍山种群最高,其次是四明山种群,最低是观音坪种群.Shannon指数和Nei指数均反映出七子花各种群具有较低的遗传多样性,但遗传分化明显.Shannon指数显示种群内遗传多样性只占总遗传多样性的27.28%,而种群间遗传多样性却占72.72%;Nei指数表明种群内的遗传变异较小,种群间的遗传变异较大,种群间的遗传分化系数为0.7157.七子花种群间的基因流为0.1987,遗传相似度平均为0.7306,遗传距离平均为0.3150,各种群间的遗传分化明显.根据遗传距离聚类分析,大致可以将9个七子花种群分为东部和西部两大类群.  相似文献

明确珍稀濒危小种群阜康阿魏(Ferula fukanensis)的遗传多样性和遗传结构,是对其制定有效的保护和管理策略的基础和前提。本研究基于10对多态性好且可以稳定扩增的SSR引物对来源于3个居群87个珍稀濒危植物阜康阿魏的遗传多样性和遗传结构进行分析。结果显示:小种群的阜康阿魏具有相对较高的遗传多样性,居群间Nei’s基因多样性指数(hS)为0.514,总的Nei’s基因多样性指数(hT)为0.516,观测杂合度(Ho)为0.881,期望杂合度(He)为0.512,香农信息指数(I)为0.836,多态位点百分率(PPB)为100%,遗传分化程度较低(Fst=0.007),95.9%的变异发生在居群内,遗传距离与地理距离无显著相关性,66.7%的居群遭遇了遗传瓶颈。结果表明,阜康阿魏遗传变异丰富,有较高的进化潜力。结合该种野外种群现状,建议建立保护区,开展就地保护,并加强引种和人工繁育等迁地保护措施,辅助阜康阿魏保育。本研究可为阜康阿魏植物资源保护提供理...  相似文献

The genetic diversity within and among populations of Hepatacodium miconioides collected at three different altitudes in Tiantai Mountain, Zhejiang Province and its relationships to environmental factors were analyzed by random amplified polymorphic DNA (RAPD) technique. The gene flow estimated from G ST was only 0.2656 and it indicated that gene flow among populations of H. miconioides was relatively low. The mean value of the genetic identity among populations of H. miconioides was 0.7126 and the average of genetic distance of H. miconioides was 0.3412. The genetic identity between populations at the elevation of 990 m and at the elevation of 780 m was the highest. The genetic identity between population at the elevation 500 m and other two populations was relatively low. 采用10对SSR引物对8个山荆子[Malus baccata (L.) Borkh.]居群140个单株的基因组总DNA进行PCR扩增,并据此对8个居群的遗传多样性和遗传关系进行了分析.结果表明:用10对SSR引物共扩增出91条带,多态性条带百分率达100.00%.8个居群的遗传多样性参数差异较大,有效等位基因数为1.437 9~1.535 0,Nei's基因多样性指数为0.256 0~0.309 2,Shannon信息指数为0.376 7~0.459 2,多态性条带百分率为64.84%~85.71%.居群间的有效等位基因数为1.616 9,Nei's基因多样性指数为0.355 1,Shannon信息指数为0.528 5,均明显高于居群内;8个居群间的基因流为1.739 5,基因分化系数为0.223 3,显示居群间的基因交换较多.UPGMA聚类分析结果表明:在Nei's遗传距离0.148 6处,8个居群被分为3组,河北塞罕坝居群单独为一组,山西五台山居群和北京东灵山居群为一组,其余5个居群为一组.据此推测:山荆子起源于中国华北和东北地区,山西灵空山、黑龙江小兴安岭、吉林长白山和山西中条山居群可能是其遗传多样性的核心居群.  相似文献

卵叶海桑 (Sonneratiaovata)是海桑科濒危红树植物 ,在我国仅分布于海南文昌清澜自然保护区内。采用简单序列重复区间扩增 (ISSR)分子标记技术对该天然居群和东寨港红树林自然保护区引种的人工居群共 3个居群 3 9个个体进行了遗传变异分析。 1 1个引物共扩增出 1 85条带 ,其中 1 2 7条具多态性 ,多态位点百分率为 68.65 %。在居群水平上相对较低 ,多态位点百分率 3 6.76%~ 5 4.5 9% ,平均值为 47.2 1 %。Nei的基因多样性、Shannon信息指数在物种水平上分别为 0 .1 41 1和 0 .2 2 92 ;在居群水平上平均值分别为 0 .1 2 0 9和0 .1 91 0。Nei的遗传分化系数Gst表明 :87.5 8%遗传变异分布在居群内 ,1 2 .42 %的遗传变异分布在居群间。居群间的遗传一致度达 0 .970 7。东寨港迁地保护的人工居群有效地保护了卵叶海桑的遗传多样性。  相似文献
