Spore discharge in Entomophthora grylli   总被引:1,自引:0,他引:1  
Entomophthora grylli Fres. has been found on larvae of Bradysia (Sciaridae, Diptera) on wood in Kansas State University greenhouses since November 1967. Infected larvae crawled to an exposed site in the night and by morning spores of E. grylli were being discharged. In the greenhouse and under controlled environments spore discharge showed a marked periodicity; spore discharge occurred in the light with peaks in the first 1–4 h; discharge then gradually declined but extended into the dark. On the second and third days peaks occurred in the light but were progressively smaller. In a greenhouse under fluctuating conditions twin peaks occurred at 10·00 and 22·00; in a growth chamber, at constant 21° C. 90% r.h., alternating 12 h light and dark periods, spore discharge was similar, with maximum numbers about 21/2 h after the onset of the photo-period. In continuous dark and continuous light no endogenous pattern was evident. Temperature range for spore discharge was 2–28·, with optimum at 15·. Decreasing humidities resulted in decreased spore production and cessation of discharge below 40% r.h.  相似文献

Summary Entrainment of the circadian rhythm in the pineal N-acetyltranferase activity by prolonged periods of light was studied in rats synchronized with a light:dark regime of 1212 h by observing phase-shifts in rhythm after delays in switching off the light in the evening or after bringing forward of the morning onset of light. When rats were subjected to delays in switching off the light of up to 10 h and then were released into darkness, phase-delays of the evening N-acetyltransferase rise during the same night corresponded roughly to delays in the light switch off. However, phasedelays of the morning decline were much smaller. After a delay in the evening switch off of 11 h, no N-acetyltransferase rhythm was found in the subsequent darkness. The evening N-acetyltransferase rise was phase-delayed by 6.2 h at most 1 day after delays. Phase-delays of the morning Nacetyltransferase decline were shorter than phasedelays of the N-acetyltransferase rise by only 0.7 h to 0.9 h at most. Hence, 1 day after delays in the evening switch off, the period of the high night N-acetyltransferase activity may be shortened only slightly. The N-acetyltransferase rhythm was abolished only after a 12 h delay in switching off the light.Rats were subjected to a bringing forward of the morning light onset and then were released into darkness 4 h before the usual switch off of light. In the following night, the morning N-acetyltransferase decline, but not the evening rise, was phase advanced considerably. Moreover, when the onset of light was brought forward to before midnight, the N-acetyltransferase rise was even phase-delayed. Hence, 1 day after bringing forward the morning onset of light, the period of the high night N-acetyltransferase activity may be drastically reduced. When rats were subjected to a 4 h light pulse around midnight and then released into darkness, the N-acetyltransferase rhythm in the next night was abolished.The data are discussed in terms of a two-component pacemaker controlling the N-acetyltransferase rhythm. It is suggested that delays in the evening switch off of light may disturb the N-acetyltransferase rhythm the next day only a little, as the morning component may adjust to phasedelays of the evening component almost within one cycle. A circadian rhythm in growth was detected by computer-aided image analysis in 3–4-cm-long, juvenile sporophytes of the kelp species Pterygophora California Rupr. and in seven Laminaria spp. In P. californica, the free-running rhythm occurred in continuous white fluorescent light, had a period of 26 h at 10°or 15°C, and persisted for at least 2 weeks in white or blue light. The rhythm became insignificant in continuous green or red light after 3 cycles. Synchronization by white light-dark regimes, e.g. by 16 h light per day, resulted in an entrained period of 24 h and in a shift of the circadian growth minimum into the middle of the light phase. A morning growth peak represented the decreasing portion of the circadian growth curve, and an evening peak the increasing portion. The circadian growth peak was not visible during the dark phase, because growth rate decreased immediately after the onset of darkness. At night, some growth still occurred at 16 or 12 h light per day, whereas growth stopped completely at 8 h light per day, as in continuous darkness. During 11 days of darkness, the thallus area became reduced by 3.5%, but growth rate recovered in subsequent light–dark cycles, and the circadian growth rhythm reappeared in subsequent continuous light.  相似文献

We examined the effects of pinealectomy and blinding (bilateral ocular enucleation) on the circadian locomotor activity rhythm in the Japanese newt, Cynops pyrrhogaster. The pinealectomized newts were entrained to a light-dark cycle of 12 h light and 12 h darkness. After transfer to constant darkness they showed residual rhythmicity for at least several days which was gradually disrupted in prolonged constant darkness. Blinded newts were also entrained to a 12 h light/12 h dark cycle. In subsequent constant darkness they showed free-running rhythms of locomotor activity. However, the freerunning periods noticeably increased compared with those observed in the previous period of constant darkness before blinding. In blinded newts entrained to the light/dark cycle the activity rhythms were gradually disrupted after pinealectomy even in the presence of the light/dark cycle. These results suggest that both the pineal and the eyes are involved in the newt's circadian system, and also suggest that the pineal of the newt acts as an extraretinal photoreceptor which mediates the entrainment of the locomotor activity rhythm.Abbreviations circadian period - DD constant darkness - LD cycle, light-dark cycle - LD 12:12 light-dark cycle of 12 h light and 12 h darkness  相似文献

Gametophytes of Laminaria saccharina cultivated from zoospores in a light-dark regime (16:8), release eggs exclusively during the dark cycles, 8–10 days after seeding of the zoospores, and mainly during the first 30 min of darkness. The inhibiting effects of light during the light cycle of 16 h per day is also apparent in gametophytes which have experienced only two dark cycles prior to day 8, when egg release begins. Egg release can be shifted to any time during the light cycle by prolonging the irradiation with white fluorescent light and by subsequent darkening for 1 h. In gametophytes cultivated in continuous white fluorescent light eggs are also released from day 8–10, so in this case no inhibiting activity of light is apparent. Egg release is inhibited by blue light and u.v., with peak wavelengths for inhibition at 372, 413, 438 and 481 nm. No inhibition occurs at wavelengths above 513 nm. The light requirement for inhibition is very low. A photon fluence rate of 1·4 μE m-2s-1, given for 45 min at 449 nm, inhibits egg release in 50% of the mature gametophytes. There is some evidence that a circadian rhythm is involved, primarily since in gametophytes which are transferred at the beginning of day 8 from 16:8 to constant conditions (darkness, continuous red or green light) the diel rhythm of egg release persists until day 11.  相似文献

Conidiation in Botrytis fabae was stimulated by irradiating 1 to 3 day old, but not 4 to 5 day old mycelium. Three cycles of 12 h irradiation + 12 h darkness stimulated the production of about twice as many spores compared with only 12 h irradiation. At 18°C all the spores had been produced within 3 days but not within 2 days from the start of irradiation. Near-u.v. irradiation at wavelengths of 375–400 nm induced most sporulation. Red light at 600–650 nm also stimulated conidiation but irradiation at other wavelengths from 300 to 700 nm was ineffective. Fewer conidia were produced when the fungus was kept in darkness at 4°C between periods of irradiaton at 18°C compared with continuous 18°C. The optimum osmotic potential of the culture, medium for conidiation was about-27 bar although more mycelium grew at even lower osmotic potentials. Abundant spore production occurred when the fungus was grown in media with a wide range of pH values.  相似文献

Growth rates in terms of area increase per 30 min were measured in flat thalli of several seaweed, species by means of computer-assisted image analysis, at 12 h light per day and a photon fluence rate of 20 μmol · m-2· s?1. Light fields included white fluorescent, imitated underwater, blue, green, and red light. In the green alga Ulva pseudocurvata Koeman et Hoek, blue light caused an immediate reduction of thallus area and growth rate after the onset of light, whereas green light and red light resulted in an initial peak in growth rate followed by inhibition 60 min after the onset of light. More growth was observed in darkness than in blue light in U. pseudocurvata. All brown and red algae tested, with Laminaria saccharina (L.) Lamour. and Palmaria palmata Stackh. as the main investigated species, grew faster during the day than during the night, irrespective of light quality during the main light phase. The upper intertidal red alga Porphyra umbilicalis (L.) J. Ag. achieved most of its thallus expansion per 24 h during the first 3 h of the light phase, with maximum growth rates of 2–3% increase in area per hour. Maximal growth rates were 0.7% for juvenile laminarian sporophytes and were lower than this in Palmaria palmata and other perennial red algae. The temporary growth inhibition by light in Ulva pseudocurvata suggests photomorphogenetic events, similar to the kinetics of stem elongation in higher plant seedlings after blue or red light pulses in darkness.  相似文献

In Daldinia concentrica spore discharge under natural conditionsis periodic, most spores being discharged during the night andfew in the daytime. An experimental study has been made of dischargeunder controlled conditions of light and temperature. In continuousdarkness periodic discharge was maintained for 12 days, butthen, although spore output continued, it ceased to be periodic.Return to alternating light (12 hrs., 100 f.c.) and darkness(12 hrs.) at once re-established the periodicity. In continuouslight (100 f.c.) periodic discharge ceased after 2 to 3 days,but was immediately re-established in alternating light (12hrs.) and darkness (12 hrs.). When the fungus was placed underconditions of alternating light and darkness each of 6 hours'duration, two peaks of spore-output were soon developed in the24-hour period. The experiments suggest that the natural periodicityis determined by the alternation of day and night.  相似文献

We examined the well-known spontaneous discharge (SD) and lightevoked (PD) discharge of the crayfish caudal photoreceptor for the possible existence of a daily rhythm in spike frequency. To do this, we isolated the abdominal nerve cord in vitro and studied its discharge frequency in constant darkness. Single cosinor analysis revealed significant SD and PD circadian rhythms (P <. 05) with periods τ = 24.4h and 24.2h, respectively. These oscillations correspond to an endogenous circadian discharge of the caudal photoreceptor that is enhanced by light. Abstract The influence of air temperature on the release pattern of Venturia inaequalis ascospores was studied by volumetric spore samplers in two spore sampling periods. In the first period (1991–1996; Passo Segni, Ferrara), 15 ascospore dispersal events were considered occurring in daylight, with high spore counts (168–5892 ascospores per m3 air per event), at an average temperature between 8.4 and 20.3°C. Both the length of the ascospore release period and distribution of airborne spores over time were significantly influenced by temperature. A logistic regression model was used to fit the proportion of ascospores trapped from the orchard air as a function of time after the beginning of the discharge event and air temperature. The accuracy of this equation was tested against data collected in the second spore sampling period (1997–2000; Sala Bolognese, Bologna, and Castelfranco, Modena); 16 dispersal events were considered, triggered by rainfall that occurred both in the dark and in daylight, with low to high spore counts (29–458 ascospores per m3 air per event), at an average temperature between 2.8 and 14.3°C. There was a general agreement between the proportion of ascospores trapped from the orchard air during these events and that estimated by using the logistic equation – in most cases, actual and estimated values showed a high coincidence. Statistical comparison showed a significant correlation (r=0.93, P < 0.01) between observed and estimated data.  相似文献

Spore discharge in Ascobolus crenulatus occurs both in the lightand in the dark. In a 12 h light: 12 h dark daily regime discharge-ratehas peaks in the dark periods, due apparently to light stimulationwith about half a day's interval between stimulus and response. Using a ‘spore clock’ the course of discharge hasbeen followed for a single apothecium on changing from darknessto light. Exposure to light (500 lux) of wave-lengths around400, 440 and 460 mµ immediately causes ’puffing‘,whilst light of longer wave-length (504 and 580 mµ) hasno effect. Change from darkness to white light has no immediateeffect, but there is a delayed stimulation with a marked increasein discharge-rate 10–14 h later. Simultaneous illuminationof an apothecium, which has been in darkness, by blue light(420 mµ, or 440 mµ, 500lux) and yellow light (580mµ, 500 lux) does not result in puffing. Plodia interpunctella Hübner (Lepidoptera: Pyralidae) comprises a model insect to analyse the photoperiodic time‐measuring system controlling its larval diapause. In the present study, the effective length of light pulse in night interruption experiments is determined at 25 °C. Various lengths of light pulse are tested by inserting them at the midnight of an LD 12 : 12 h photoperiod. When the light pulse is 15 or 30 min, the incidence of diapause is 86%. To inhibit the induction of diapause effectively, a light pulse of 1.75–2 h is needed. The incidence of diapause is 12% under an LD 12 : 5 : 2 : 5 h photoperiod. To determine the precise role of the light pulse, 2‐h light pulses placed at the midnight of an LD 12 : 12 h photoperiod are disrupted systematically by darkness. When a 2‐h light pulse is disrupted by 15 min of darkness, diapause is generally prevented (< 29%) regardless of the temporal position of darkness. Longer disruption by darkness induces diapause moderately (37–67%). A Bünsow experiment is also conducted at 25 and 20 °C, in which the main photophase of 12 h of light is combined with 24–72‐h scotophases scanned by a 2‐h light pulse. The photoperiodic cycle length tested, therefore, varies in the range 36–84 h. In each cycle length, the incidence of diapause fluctuates as the light pulse moves toward dawn. However, no regular and circadian changes in the percentage diapause are observed in relation to diapause determination.  相似文献

The effects of light on the spore germination of a hornwort species,Anthoceros miyabeanus Steph., were investigated. Spores of this species were photoblastic, but their sensitivities to light quality were different. Under either continuous white, red or diffused daylight, more than 80% of the spores germinated, but under blue light none or a few of them germinated. Under continuous far-red light or in total darkness, the spores did not germinate at all.Anthoceros spores required red light irradiation for a very long duration, i.e., over 12–24 hr of red light for saturated germination. However, the spore germination showed clear photo-reversibility by repeated irradiation of red and far-red light. The germination pattern clearly varied with the light quality. There were two fundamental patterns; (1) cell mass type in white or blue light: spores divide before germination, and the sporelings divide frequently and form 1–2 rhizoids soon after germination, and (2) germ tube type in red light: spores germinate without cell division, and the single-cell sporelings elongate without cell division and rhizoid formation.  相似文献

ABSTRACT. Domestic turkeys naturally infected with Leucocytozoon smithi were blinded by bilateral ocular enucleation, pinealectomized, sham-pinealectomized, pinealectomized plus enucleated, or maintained as controls. Groups of turkeys were acclimated to either light-dark periods of 14L:10D or “darkness” with intermittent periods (10–20 min) of red light at irregular hours approximately every three days as required for maintenance of turkeys. Peripheral gametocyte numbers of L. smithi in all groups were determined every 2 h over a 36 h period. Under 14L: 10D photoperiod, no observable difference in the pattern of gametocyte circadian rhythmicity between pinealectomized, enucleated, pinealectomized plus enucleated, and control turkeys was noted. Although mean parasitemias differed among groups, peak gametocyte numbers occurred between 1000 and 1800 h; how parasitemias occurred between 2000 and 0400 h. However, the phase of gametocyte rhythmicity in pinealectomized plus enucleated turkey hosts did exhibit a lag with reference to other hosts when examined by least squares fits of simple harmonics. Under conditions of “darkness” with intermittent, irregular periods of red light, L. smithi gametocyte numbers of individual turkeys, pinealectomized, sham-pinealectomized, or maintained as controls, exhibited a circadian periodicity though parasite cycles were out of phase with the natural photoperiod to which the turkeys previously had been exposed. A slight drift out of phase of L. smithi gametocyte periodicity occurred among turkeys in the sham-pinealectomized and the control groups while a considerably more prominent drift out of phase was seen among the parasite rhythmicity patterns of the pinealectomized birds. Data indicate that the pineal gland of the turkey did not directly mediate L. smithi gametocyte circadian periodicity, although an indirect involvement in regulating the timing of parasite rhythmicity is suggested.  相似文献

Summary The retinal rods, cones and epithelial pigment of most lower vertebrates display rhythmic photomechanical (retinomotor) migrations in response to changes in ambient lighting conditions. This study examines the extent of these migrations in the absence of the daily changes in illumination (constant darkness and constant light) in three species of teleosts. Salmo trutta, a crepuscularly active fish, showed two peaks of light adaptation occurring around dawn and dusk when kept in constant darkness. Tinca tinca, a nocturnal species, also showed an endogenous rhythm during extended periods of darkness, but, unlike Salmo trutta, it was light-adapted throughout what would normally have been day. At the maximal extent of migration under conditions of continual darkness, the pigment migrated 59% as much as it did during a normal light/dark cycle. Nannacara anomala, a tropical diurnally active species, showed a similar but more pronounced rhythm than Tinea tinea for all 3 days of experimental darkness, behaving essentially identically to fish exposed to a light/dark cycle. Nannacara anomala also displayed a weak rhythm when kept in constant light.It is concluded from these and previous results that the pattern of endogenous photomechanical movement depends both on the activity pattern of a species and on the constancy of the lighting conditions to which it has been exposed during its lifetime.  相似文献

Seasonal and diurnal changes in the spore liberation of Gracilaria corticata J. Agardh and the effect of exposure to air and light and dark periods on spore output have been studied. Tetraspore and carpospore shedding was found in all months of the year with a maximum output in March–April and again between September and December. Spore output varied at different times of the day and the highest values were recorded at night. Exposure to air and photoperiod affected spore production and the maximum liberation was observed in submerged fronds kept in complete darkness. Viability of tetraspores was low under laboratory conditions and only 26% of germlings were found in a healthy condition at the end of one month.  相似文献

The impacts of unnatural every day cycles (circadian) for 60 days on the histological structure of kidneys and ATPase activities in MF1 mice were studied. The exposure times were 16 h dark, 16 h light, 24 h dark, and 24 h light, and control exposure times were 12 h dark followed by 12 h light. Our results showed an increase in the total ATPase activity of mice in all groups. Additionally, the activity of the enzyme Na+/K+-ATPase was increased after 24 h darkness, 24 h light, and 16 h light exposures compared to control. The enzyme Mg+2-ATPase activities of the groups were higher when exposed to 16 h light, 24 h light, 24 h darkness and 16 h darkness. The activities of total ATPase, Na+/K+-ATPase and Mg+2-ATPase in kidneys were increased in all groups after 24 h light, 24 h darkness, 16 h darkness and 16 h light exposures. Interestingly, the activity of V-type ATPase was reduced after 16 h darkness, 24 h darkness and 16 h light. Taking everything into account, changes in the day by day cycle prompt neurotic changes, enzymatic and histological changes in the kidneys of mice. More studies should be directed to explore the impacts of light and darkness that can prompt these progressions.  相似文献

Summary The cross-sectional area of rhabdomeres in the compound eye of the blowfly, Lucilia, was found to remain constant under 12 h light/12 h dark cyclic lighting, and 10 days constant light or darkness. It was reduced only slightly during 3 h light after 10 days darkness (by 21%), or on exposure to 2h darkness + 1.5 h light after 10 days light (by 10%). Morphological evidence indicates that shedding of photoreceptor membrane during turnover is achieved by an extracellular route, and by pinocytosis from the bases of the microvilli. The photoreceptor membrane shed by both mechanisms appears to accumulate in multivesicular bodies. The amount of photoreceptor membrane shedding, as indicated by numbers of multivesicular bodies, is constant throughout the day and night on cyclic lighting, decreases in constant darkness, but returns to its normal level after an exposure to 3 h light subsequent to 10 days darkness.  相似文献

Apical root segments of Zea mays L. cv. Orla 264 undergo some geotropic curvature in complete darkness but the curvature increases considerably if prior to geostimulation the segments are given a light pretreatment. If the light treatment is follwed by a dark treatment before the root is geostimulated the light-induced response is not changed by dark periods up to 2 h but declines with longer ones, and disappears completely after 5 h of darkness.  相似文献
