The localization of sympathetic fibers on the floor of the cranium was studied in rats using amine fluorescence histochemistry, neuropeptide-Y (NPY) immunohistochemistry, and electron microscopy. The vast majority of amine fluorescent fibers joined the abducent nerve and were localized in the peripheral zone under the perineurium. After advancing along this nerve for some distance, the fibers diverged into many bundles that converged to form the cavernous plexus at a rostral end of the trigeminal ganglion. On the dorsal surface of the trigeminal ganglion, one or two medium-calibered fluorescent bundles ran inside or in close proximity to the trochlear nerve, while many small-calibered, brightly fluorescent bundles also extended longitudinally in the epidural connective tissue. In rats that had undergone nerve severance, NPY-immunoreactive fibers were detected at the cut ends of the abducent and trochlear nerve. The differing amounts of NPY accumulated at the rostral and the caudal stumps indicated the direction of the NPY-bearing fibers. Electron microscopy confirmed the presence of unmyelinated fibers in both the abducent and trochlear nerves.  相似文献

Summary It is demonstrated that 5-hydroxydopamine (5HDA) is capable of forming highly fluorescent products with formaldehyde under conditions suitable for histochemical analyses. Prom absorptiometric and fluorometric studies it is reasonable to assume that the fluorophores formed are isoquinolines, in analogy with the reaction between, e.g., dopamine and formaldehyde. The reaction proceeds readily in protein layers and actual nerve terminals in tissues, allowing demonstration and identification of 5HDA in tissue structures. Summary Nine cultures of fibroblast cell types and 13 epithelial-like cell types were maintained for 1 week in media supplemented with L-asborbic acid (50 μg per ml). All fibroblast-like cultures produced extracellular fibers that stained positively by a silver-impregnation reticulin stain. Nine of the 13 epithelial-like cultures produced fibers that stained positively for reticulin. Nearly all cultures not supplemented with ascorbic acid showed no fiber staining. Those few lines that stained positively for reticulin in the absence of ascorbic-acid supplementation demonstrated only slight reticulin formation. Reticulin from one fibroblast culture and one epithelial culture was examined by electron microscopy, and the silver-impregnated fibrils were morphologically identical to collagen. The reticulin was digestible with collagenase, providing further evidence that the silver-impregnation reticulin stain identifies collagen in culture. The demonstration of collagen can be performed easily in histology laboratories using Formalin-fixed cells, and provides a means of assaying a functional property of cells in culture which is characteristic of connective tissue fibroblasts in general as well as certain specialized epithelia.  相似文献

Summary An electron microscopic study has been made of the sympathetic ganglia of a 15 and a 17 week old male human fetus. The fetal sympathetic neurons were densely packed in a scanty connective tissue matrix which also contained blood vessels. The fetal sympathetic neurons had a large, electron-light nucleus with one or two nucleoli, and was of a somewhat mottled appearance due to irregularly dispersed aggregates of fine and coarse granules. The perikaryon usually formed a thin envelope around the nucleus and contained, except for large pigment granules, all intracytoplasmic structures which were also found in mature sympathetic neurons. Adjacent sympathetic cells were either in immediate contact, or slightly separated by a wedge of electron-light satellite expansions, or lined by primitive axons. The satellite cells were in the early state of development. Electron-dense axons either stood side by side with, or were slightly engulfed by light Schwann cell expansions and formed distinct bundles surrounded by a common basement membrane. Neuropeptide Y (NPY)-containing nerve structures were detected in the Syrian hamster gallbladder by using peroxidase-antiperoxidase immunohistochemical staining and immunoelectron microscopic techniques. In addition, Alcian blue-P.A.S. was used to differentiate two types of surface cells in the epithelium of the gallbladder: the columnar cells and the tuft cells. NPY-immunoreactive varicose nerve fibers were detected in the adventitial layer of the arterioles, precapillary arterioles and around axial venules of mucosal folds; they were not observed in the capillary beds. Since NPY is associated with norepinephrine (NE) in the sympathetic nerves, it could prolong the adrenergic vasoconstrictive effects. We propose that NPY and NE are simultaneously released to restrict the blood flow in the subepithelial capillary beds by increasing vascular resistance thereby perturbing the absorption of water and electrolytes.  相似文献

In the convoluted proximal tubules of the kidney of pregnant rats alkaline phosphatase activity decreases from 21 days gestation till the 2nd day after birth. During the same time the reaction product of the histochemical succinate dehydrogenase assay exhibits an atypical granular character in some of the proximal tubules, which sometimes additionally contain sudanophilic structures. Electron microscopy reveals dilated mitochondria, an increased number of autophagosomes and big lipid vacuoles of low density.  相似文献

Summary The localization of serotonin (5-HT)-immunoreactive nerve fibers in the cerebellum of the rat and cat was investigated by means of the peroxidase-anti-peroxidase (PAP) method using highly specific antibodies to 5-HT.Serotonin-containing nerve fibers were distributed throughout the entire cerebellum including the deep cerebellar nuclei, while 5-HT-positive neuronal somata were not detected in the cerebellum of either species. A different pattern of 5-HT innervation was found among the three layers of the cerebellar cortex. There were also interspecific differences in the pattern of distribution of 5-HT. In the rat, the pool of 5-HT nerve fibers mainly consisted of tangential elements, which were predominant in the molecular layer, while in the cat only a few 5-HT fibers were found in the molecular layer of the cerebellar cortex; dense networks of 5-HT nerve fibers were present in the granular layer. The intermediate nerve (IN) in embryogenesis of man, cat and white rat is formed similarly. From the common with the VIII cranial nerve anlage the geniculum node, the vestibular and cochlear ganglia are emerged. A separated root of the IN connects the geniculum node with the nuclei, situating in the CNS. From the geniculum node main branches of the IN get off: the greater petrosal nerve and the cord of the tympanum, its fibers in the temporal bone canal run in the facial nerve trunk. In the periphery the IN branches unite with other cranial nerves, make connections with branches of vegetative (parasympathetic) ganglia and plexuses. They spread along a large territory, forming a system of parasympathetic innervation in the area of the head and ensuring with specific nervous apparatuses the gustatory organ. It is expedient to select the IN into an independent cranial nerve and confirm it the number of the regular pair.  相似文献

The purpose of the present works was to clarify whether the cranial nerves III, IV and VI carry proprioceptive afferent fibres from the extrinsic ocular muscles. In sheep the picture is now clear. The cranial nerves III, IV and VI carry many large proprioceptive fibres (12-16 micrometer) to the central nervous system. These nerves also contain many small fibres of the y-range (2-6 micrometer) which innervate the intrafusal muscle fibres in the spindles. In man the picture is still vague: most of the spindles are not typical, the large proprioceptive fibres (12-16 micrometer) and the small y-fibres (2-6 micrometer) are very few in the cranial nerves III, IV and VI. It is to be concluded that in sheep the cranial nerves III, IV and VI are not purely motor nerves to the extrinsic ocular muscles, but they also carry many of the large fibres of the proprioceptive function. In man, such large fibres are not found and the pathway of proprioceptive afferents from the orbital muscles is still not certain.  相似文献

In anaesthetised Wistar rats, electrical sympathetic activity and a somatosympathetic reflex in the cervical sympathetic trunk elicited by a single electrical shock to forelimb or hindlimb afferent nerves, were recorded. The spontaneous activity was shown to conform with the pulse and respiratory waves of arterial pressure. Somatosympathetic reflex consists of early and late discharges evoked by somatic myelinated afferent fibres stimulation, and C-response elicited by stimulation of unmyelinated afferent fibres in spinal nerves.  相似文献

Summary Distribution of the serotonin nerve fibers in the neurohypophysis of adult cats was demonstrated using the peroxidase-antiperoxidase method. The serotonin nerve fibers were distributed in the internal and external layers of the infundibulum, terminating on the wall of the capillary loops and the outer surface of the external layer. Ultrastructural and histochemical changes of skeletal muscle were studied in three patients affected with gas gangrene. There was complete lack of the phosphorylase, succinate dehydrogenase and adenosine triphosphatase activities in the affected muscles of all the patients. In unaffected muscles these enzymes showed weaker activities than in norm. The lactate dehydrogenase activity, especially the heart type isozyme (LDH-1 or H4) proved less sensitive to the effect of clostridial toxin. A general increase in the acid phosphatase activity was found both in affected and in unaffected muscles. On electron microscopic examination damage to sarcolemmal membrane and disintegration of myofilaments was seen. The mitochondria were swollen and their cristae distorted and fragmented.  相似文献
