Differences in seed vigour of zero- and high-tannin faba beans were investigated using 25 seed lots of 12 cultivars following earlier reports of poor emergence in the zero-tannin types. Field emergence ranged from 54–96% indicating differences in seed vigour between cultivars all having high laboratory germination (>91%). Seed from zero-tannin accessions with poor emergence had a higher incidence of testa and cotyledon cracking, a smaller percentage of hard seeds, more rapid water uptake, a lower percentage of vital staining of cotyledons and a greater leaching of solutes than high-tannin types. Nevertheless, variation in these characteristics existed between cultivars and lines of both types. Seeds with more cracks in the seed coat and fewer hard seeds imbibed water more rapidly and consequently showed lower levels of vital staining and more cracks in the abaxial surface of the cotyledons. Slower imbibition in polyethylene glycol lessened the incidence of these deleterious characteristics and may provide a practical resolution to the problem of poor field emergence in zero-tannin lines of faba bean with low seed vigour.  相似文献

The laboratory germination (criterion radicle emergence) of seven seed lots of winter wheat cv. Slejpner was similar. However, they differed in vigour as demonstrated by differences in germination after controlled deterioration carried out at a range of seed moisture contents, at two temperatures and for different times. A vigour assessment for each lot was quantified by calculating a value for the seed lot constant Ki, of the viability equation. Germination in lower water potentials reduced the uptake of water, radicle and coleoptile emergence and radicle and coleoptile extension. There was no difference in the water uptake of seed lots differing in vigour. However, seed lots of lower vigour showed less radicle emergence, less coleoptile emergence and shorter radicles than higher vigour seed lots in low water potentials. Similarly, controlled deterioration resulted in reduced radicle and coleoptile emergence and growth compared to unaged seed, and also to a greater sensitivity to low water potentials. The implications for field establishment are discussed.  相似文献

By means of a machine vision facility, the process of water-imbibitionin a small seeded brassica species was recorded as the visibleincrease in seed volume. Dry cauliflower seed (Brassica oleracea)showed an immediate rapid phase of imbibition upon the additionof water. This initial phase was associated with the reductionin seedling root growth resulting from the imbibition of coldwater, and the rate of uptake of water at 20?C was negativelycorrelated with subsequent seedling growth. Damage to the testaof dry seed resulted in an increased rate of imbibition anda corresponding decrease in seedling root growth measured onslant boards. This showed that the intact testa of cauliflowerseed is capable of acting as a barrier to water influx and thata high rate of water uptake is damaging to the embryo. Testadamage reduced percentage soil emergence of seeds, as did raisingthe soil moisture content during the imbibition period by wateringimmediately after sowing. Both these treatments were believedto increase imbibition rate. Conditions which encouraged a lowrate of water uptake also improved the rate and uniformity ofemergence. Correlation of mean imbibition rates (measured ina laboratory test) with soil-emergence indicated that the sensitivityto imbibition damage varied between seed lots and interactedwith the absolute rate of water influx to determine the finalpercentage emergence. This factor prevented reliable predictionof seedling performance from the ranked-order of measured imbibitionrates. Six different levels of vigour of sorghum seeds from an identicallot were obtained through accelerated aging for various periodsof time. Vigour was estimated according to yielding abilityof the seed in the field. After 6 d of aging, seed invigoration,to the extent of a 20% increase in yield, was observed. However,this was followed by a gradual decrease in yield following agingof up to 48 d. All seed lots showed 88–92% germinationor emergence capacity when tested in the laboratory, which correlatespoorly with vigour. However, vigour correlated well with therates of germination and root emergence when tested in the laboratory,as well as with emergence rate in the field.  相似文献

Seeds of Sesbania punicea (Cav.) Benth. are largely impermeable,and the natural, low, percentage which germinate are permeableat the lens. Scarifying seed in acid stimulates germinationand causes various types of damage to the testa, including thelens. Placing seeds in boiling water stimulates germinationby rendering the seeds permeable at the lens. Seeds were mechanicallyscarified at precise sites on the testa to assess influenceof the site and degree of damage on germination. Results indicatethat both the site of initial water entry and the rate of waterentry are important in determining seed and seedling vigour.Damaging the lens is more effective in stimulating germinationand reducing seedling abnormalities than complete excision ofthe lens. This effect can be reversed if seeds from which thelens was excised are allowed to imbibe gradually in inert osmotica.The leguminous lens thus acts as the site of initial water entryand regulates the rate of water uptake, thereby increasing seedand seedling vigour. The effects of damaging the testa and the application of seed dressings were examined in field trials on several short-and long-season cultivars of groundnut (Arachis hypogea) with differently-pigmented testas at Samaru, Nigeria, in 1967. There was a high correlation between the number of seedlings which emerged in these trials and the resistance or susceptibility of the seed to invasion by the fungus Aspergillus flavus, as assessed by laboratory tests. When the testa was undamaged the emergence of white (susceptible) seed was only 50% while that of coloured (resistant) seed was between 95 and 98%. Damage to the testa by scratching greatly decreased emergence. The application of seed dressing increased the emergence of susceptible seed and also restored the emergence of scratched seeds to the level of undamaged seeds. The effect of the complete removal of the testa was not counteracted by seed dressing; naked seeds, with and without dressing, gave c. 10% emergence. The importance of pigment in the testa, the condition of the seed and the effects of seed dressing are discussed.  相似文献

  • The facultative root hemi‐parasite Rhinanthus minor is often used in grassland habitat restoration projects to regulate ecosystem structure and function. Its impact on community productivity and diversity as a function of resource supply, sward composition and management has been widely investigated. However, there is a lack of information about the possible influence of seed quality on the efficacy of the hemi‐parasite.
  • Ten seed lots from commercial sources were sown in the field and their germination characteristics investigated in the laboratory. Seeds from four lots were also germinated and sown in pots alongside plants of two host species, Lotus corniculatus and Holcus lanatus. Plant establishment, height and flowering density were evaluated for the hemi‐parasite, while plant biomass was measured for both R. minor and its host.
  • Two aspects of seed quality influenced the field emergence of seed lots of R. minor, the radicle emergence (%) and the length of the lag period from the beginning of imbibition to germination (mean germination time), which indicates seed vigour. A longer lag period (lower vigour) was associated with higher levels of seedling mortality and lower plant vigour, in terms of plant height and biomass accumulation and was also reflected in the parasitic impact of the seed lots.
  • Seed quality, specifically germination and vigour, can influence the establishment, survival, subsequent plant productivity and parasitic impact of R. minor in vegetation restoration projects. Seed quality is discussed as a key factor to consider when predicting the impact of the hemi‐parasite on community productivity and diversity.

Seedling emergence and subsequent growth from untreated seeds and selected-germinated seeds which had been dried before sowing (low-moisture-content germinated (LMCG) seeds) were compared in cellular trays in a glasshouse and in the field following transplanting. Three seed lots of different quality of the same cultivar were used for cabbage and cauliflower. In all cases, seedling emergence was greater, earlier and more uniform from LMCG seed than that from untreated seeds. LMCG seed effectively eliminated the differences due to seed quality shown by untreated seeds at emergence suggesting an improvement in seed vigour as well as viability from the selection treatment. Higher quality seed and LMCG cauliflower seed led to larger curd weights and therefore an increase in marketable yield at harvest compared to that from lower quality seeds. However, differences in transplant uniformity due to seed quality and seed treatment had little impact of agronomic significance following transplanting.  相似文献

Background and Aims

Imbibition of Japanese soybean (Glycine max) cultivars was studied using micro-magnetic resonance imaging (MRI) in order to elucidate the mechanism of soaking injury and the protective role of the seed coat.

Methods

Time-lapse images during water uptake were acquired by the single-point imaging (SPI) method at 15-min intervals, for 20 h in the dry seed with seed coat, and for 2 h in seeds with the seed coat removed. The technique visualized water migration within the testa and demonstrated the distortion associated with cotyledon swelling during the very early stages of water uptake.

Key Results

Water soon appeared in the testa and went around the dorsal surface of the seed from near the raphe, then migrated to the hilum region. An obvious protrusion was noted when water reached the hypocotyl and the radicle, followed by swelling of the cotyledons. A convex area was observed around the raphe with the enlargement of the seed. Water was always incorporated into the cotyledons from the abaxial surfaces, leading to swelling and generating a large air space between the adaxial surfaces. Water uptake greatly slowed, and the internal structures, veins and oil-accumulating tissues in the cotyledons developed after the seed stopped expanding. When the testa was removed from the dry seeds before imbibition, the cotyledons were severely damaged within 1·5 h of water uptake.

Conclusions

The activation of the water channel seemed unnecessary for water entry into soybean seeds, and the testa rapidly swelled with steeping in water. However, the testa did not regulate the water incorporation in itself, but rather the rate at which water encountered the hypocotyl, the radicle, and the cotyledons through the inner layer of the seed coat, and thus prevented the destruction of the seed tissues at the beginning of imbibition.Key words: Dry seeds, Glycine max, MRI, seed coat, soaking injury, soybean, testa, role of inner layer of seed coat, water uptake  相似文献

These experiments suggest a number of causes for the poor establishment of direct reseeded grass. An adequate supply of moisture to the seed is of prime importance and burial of trash in drill slits may impair this supply by preventing seed/soil contact directly or by preventing closure of the drill slit. Management techniques which aim to reduce the burden of trash in areas to be sown by direct drilling, usually result in improvements in emergence. This is achieved by improved seed/soil contact, reduced evaporation of water from an open drill slit and lowered probability of phytotoxic leachates being produced. It is possible that benefits might be achieved were it possible to select for sowing seed of proven high vigour, i.e. seed that establishes well in poor conditions.  相似文献

The effect of field weathering on oxyradical accumulation and subsequent changes were studied in the seeds of soybean [Glycine max (L.) Merr.] cv. JS 71-05. Electron spin resonance (ESR) quantification of oxyradical revealed that field weathering plays an important role in acceleration of their accumulation. One week of weathering increased the accumulation of oxyradicals to almost 2-fold and triggered the deteriorative cascade, by enhancing the lipid peroxidation and membrane perturbation, leading to cell death in seed tissues and poor germinability and vigour of soybean seeds. Thus, the weather conditions at the time of physiological maturity to harvesting of crop are very crucial and the field weathering plays a critical role for the maintenance of seed quality.  相似文献

The germination of seeds of Xanthium pensylvanicum Wallr. occurs in 2 phases, an initial passive phase of water uptake followed by an active phase of growth. These 2 phases have been separated experimentally, and shown to occur similarly in isolated cotyledons and embryonic axes. Measurements of the physical thrust generated by the entire seed and its separate components of cotyledon and axis reveal that non-dormant Xanthium seeds develop more than twice the thrust of dormant seeds, and that this difference develops principally in the second phase of enlargement of the axis. Measurement of the forces required for piercing the testa of these seeds establishes that whereas the thrust developed by non-dormant seed is adequate to cause testa rupture, that developed by dormant seeds is not. It is concluded that the dormancy of Xanthium involves an inadequacy in the embryo for rupture of the testa.  相似文献

Extensive and kinetically well-defined water exchanges occur during germination of seeds. A putative role for aquaporins in this process was investigated in Arabidopsis. Macro-arrays carrying aquaporin gene-specific tags and antibodies raised against aquaporin subclasses revealed two distinct aquaporin expression programs between dry seeds and young seedlings. High expression levels of a restricted number of tonoplast intrinsic protein (TIP) isoforms (TIP3;1 and/or TIP3;2, and TIP5;1) together with a low expression of all 13 plasma membrane aquaporin (PIP) isoforms was observed in dry and germinating materials. In contrast, prevalent expression of aquaporins of the TIP1, TIP2 and PIP subgroups was induced during seedling establishment. Mercury (5 microM HgCl(2)), a general blocker of aquaporins in various organisms, reduced the speed of seed germination and induced a true delay in maternal seed coat (testa) rupture and radicle emergence, by 8-9 and 25-30 h, respectively. Most importantly, mercury did not alter seed lot homogeneity nor the seed germination developmental sequence, and its effects were largely reversed by addition of 2 mM dithiothreitol, suggesting that these effects were primarily due to oxidation of cell components, possibly aquaporins, without irreversible alteration of cell integrity. Measurements of water uptake in control and mercury-treated seeds suggested that aquaporin functions are not involved in early seed imbibition (phase I) but would rather be associated with a delayed initiation of phase III, i.e. water uptake accompanying expansion and growth of the embryo. A possible role for aquaporins in germinating seeds and more generally in plant tissue growth is discussed.  相似文献

Myrigalone A (MyA) is a rare flavonoid in fruit leachates of Myrica gale, a deciduous shrub adapted to flood-prone habitats. As a putative allelochemical it inhibits seed germination and seedling growth. Using Lepidium sativum as a model target species, experiments were conducted to investigate how environmental cues modulate MyA's interference with key processes of seed germination. Time course analyses of L. sativum testa and endosperm rupture under different light conditions and water potentials were combined with quantifying testa permeability, endosperm weakening, tissue-specific gibberellin (GA) and abscisic acid (ABA) contents, as well as embryo growth and apoplastic superoxide production important for cell expansion growth. Lepidium sativum testa permeability and early water uptake by imbibition is enhanced by MyA. During late germination, MyA inhibits endosperm weakening and embryo growth, both processes required for endosperm rupture. Inhibition of embryo cell expansion by MyA depends on environmental cues, which is evident from the light-modulated severity of the MyA-mediated inhibition of apoplastic superoxide accumulation. Several important key weakening and growth processes during early and late germination are targets for MyA. These effects are modulated by light conditions and ambient water potential. It is speculated that MyA is a soil seed bank-destroying allelochemical that secures the persistence of M. gale in its flood-prone environment.  相似文献

At an optimal germination temperature loss of viability in wheatseed is reflected in reduced levels of nucleoside triphosphatesand nucleotide sugars in the embryo compared to the levels foundin high viability embryos during the early hours of imbibition.These differences are magnified on germination at 10° Cand are accompanied by a greatly reduced rate of protein synthesisin embryos of low viability compared with the correspondingrate in embryos of high viability. Loss of vigour precedes lossof viability in seed but differences in biochemical parametersbetween high and medium vigour seed do not become apparent unlessgermination under stress conditions, e. g. low temperature,occurs. Under these stress conditions, wheat seed of reducedvigour can be distinguished from high vigour seed lots of similarviability by the decreased levels of nucleotides and nucleotidesugars found in embryos from reduced vigour seed lots duringthe early hours of imbibition and by reduced rates of proteinsynthesis in these embryos during this same period. Two tomato (Lycopersicon esculentum) mutants with dark testae displaying poor germination rate and percentage on both water and 100 microM gibberellin(4 + 7) were recovered. The mutants were allelic (black seed1-1; bks1-1 and bks1-2), inherited in Mendelian fashion as a recessive gene residing on chromosome 11. They are not allelic to bs (brown seed) -1, -2, or -4, which impair seed germination and possess dark testae. The bks/bs mutants accumulated dark pigment in the cell layers of the testa above the endothelium, which itself accumulated proanthocyanidins similar to wild type. The poor germination performance of bks mutant seeds was because of impediment of the mutant testae to radicle egress. Imbibition on gibberellin(4 + 7) did not ameliorate germination percentage or rate. The toughening of the bks testa and associated poor germination were partially overcome when seeds were not dried before germination or were dried under N(2). The seeds of the bks mutant have elevated activity of at least one enzyme responsible for the detoxification of reactive oxygen species. The bks mutant is epistatic to 12 anthocyaninless mutants of tomato. Bio- and physicochemical analysis of the bks testa determined that it accumulated a melanic substance. Inheritance of bks/bs mutations contrasts with that of the anthocyaninless mutants, which are inherited according to the genotype of the maternally derived testa. This suggests that the testa manufactures components before its demise that can maximize testa strength, whereas the endosperm/embryo produces factors that are conveyed to the testa, mitigating this process.  相似文献

BACKGROUND AND AIMS: Water uptake by dry kidney beans (Phaseolus vulgaris 'Rajma') and adzuki beans (Vigna angularis) was traced using micro-magnetic resonance imaging in order to elucidate the channel of water entry, the manner of water delivery and the timing of swelling of the seeds. METHODS: Magnetic resonance images of beans absorbing water were continuously measured with the single-point imaging method for 16 h or 20 h at 15-min intervals. With this technique, it was possible to detect and visualize the location of water in the beans, at a low water content, in the initial stages of water entry. KEY RESULTS: Water was taken up through a specified tissue, the lens, near the hilum, and distributed primarily to the testa. When water reached the radicle, it began to be incorporated into cotyledons with considerable swelling of the seeds. Water uptake took place within a short time for kidney beans. The initial process of water entry was associated with mechanical vibration of the seed. Rapid hydration of the testa and the swelling of the cotyledons were then observed. Water was supplied to cotyledons through the adaxial epidermis. In contrast, it took a long time, approx. 7 h, to activate the water channel of the lens for adzuki beans which have a tightly fitting testa. Steeping of the testa was not uniform, which induced temporary slanting before enlargement of the seed. CONCLUSIONS: The activation of the lens as the sole water channel, the delivery of water to the radicle within the testa, the swelling of the cotyledons, and the further increment of water are physiologically different processes during imbibition, and were separated by locating water in various tissues and by analysing the time course of water uptake using magnetic resonance imaging with the single-point imaging method.  相似文献

Seeds with low vigour as a result of ageing showed a large improvementin seed quality following a 2 h soak in water, reflected inan increased rate of germination, higher emergence in soil,and the retention of high germination after 24 h rapid ageingin a controlled deterioration vigour test. Similar, but smallerimprovements were seen after a priming treatment using polyethyleneglycol. Seeds dried to their initial moisture content aftertreatment retained these beneficial effects. The treatmentshad little effect on high vigour seed. These findings suggestthat seed invigoration by hydration treatments results fromthe repair of previously sustained deterioration. Comparisons of five pairs of isogenk lines of peas, differingonly in the A gene for seed coat colour showed that white seeds(genotype aa) imbibed more rapidly than coloured seeds (AA),suffered greater imbibition damage revealed by dead tissue onthe cotyledons, and higher solute leakage. Seed-coat pigmentationwas closely associated with slow water uptake, since when expressionof the A gene was suppressed by the recessive pollens gene,the resulting white seeds {palpal AA) imbibed rapidly. The slowwater uptake by coloured seeds was not due to the restrictionof water entry by the seed coat since the differences in imbibitionrate were maintained when a portion of the seed coat was removedand seeds were imbibed with the exposed cotyledon in contactwith moist filter paper. Imbibition of similarly treated seedsby immersion in polyethylene glycol solutions (1–4%) whichincreased the seed/solution wettability, had little effect onthe water uptake of coloured seeds compared to imbibition inwater whereas that of white seeds increased in the first 10mins imbibition. Poor wettability of the inner surface of colouredseed coats did not therefore explain the slow imbibition ofthese seeds. The white seed coats loosened rapidly during imbibitionwhilst the coloured seed coats remained closely associated withthe cotyledons suggesting that the adherence of the seed coatto the cotyledons and therefore the ease of access of waterbetween the testa and cotyledons determines the rate of imbibition.The rapid water uptake by white-coated seeds and the subsequentimbibition damage may explain the high incidence of infectionof these seeds by the soil-bome fungus Pythhan after 2 d insoil.
