A study on nutrient regeneration processes and a measure of their fluxes at the sediment-water interface was carried out in two different stations of a shallow lagoon of the Po delta river (Italy). A few parameters on the solid fraction (grain-size, porosity, C, N) and pore water profiles of o-P, NH3, NO inf3 sup– , SiO2, Tot-CO2, SO inf4 sup2– , Fe, Mn, Ca, Mg, pH, Eh were determined. At both stations the results were typical for fine sediments rich in organic matter. The ratio of variations of sulphate (SO inf4 sup2– ) to total carbonate demonstrates the main role sulphate reduction plays on the organic matter decay. The use of the ratios of variations of sulphate (SO inf4 sup2– ) to ammonia (NH3) and of sulphate (SO inf4 sup2– ) to phosphate (o-P) in pore waters enabled us to calculate the C/N/P of the decomposing organic matter. Obtained C/N/P indicated an enrichment of N and P with regard to C/N/P ratios of the solid fraction, due to the selective stripping of N and P during organic matter mineralization. This phenomenon decreases with depth, where organic matter becomes more refractory. Calculations on saturation degrees have shown the possibility of authigenic calcite, apatite and rhodochrosite precipitation in sediments. Nutrient fluxes were estimated for SiO2, NH3 and o-P by means of benthic chambers and modelling the pore water profiles. The model used for the calculation of fluxes allowed us to account for the bioturbation-irrigation influence near the interface, by means of a cumulative diffusion coefficient. Directly measured fluxes proved to be always significantly greater than the theoretical ones. These differences seem to be due to surface processes which do not affect pore water concentrations (degradation of fresh materials at the interface; micro-bioturbation by small gasteropoda such as Hydrobia ventrosa) and/or to the different concept of the two methods in time and space. Number, size and biomass of macrobenthic species living in the sediment underneath the benthic chambers were determined. The comparison between data on macrobenthic populations and flux values showed a good relationship between the number of organisms and benthic fluxes within each station. However, flux variations between stations are to be attributed mainly to the different arrangement of the tubes of the polychaetes Polydora ciliata in the sediment.  相似文献

The aim of this work was to assess whether agroforestry species have the ability to acquire P from pools unavailable to maize. Tithonia diversifolia(Hemsley) A. Gray, Tephrosia vogelii Hook f., Zea mays and Lupinus albusL. were grown in rhizopots and pH change and depletion of inorganic and organic P pools measured in the rhizosphere. Plants were harvested at the same growth stage, after 56 days for maize and white lupin and 70 days for tithonia and tephrosia, and the rhizosphere sampled. The rhizosphere was acidified by tithonia (pH change –0.3 units to pH 4.8) and lupins (–0.2 units to 4.9), alkalinised by tephrosia (+0.4 units to pH 5.4), and remained unchanged with maize growth. Concurrent with acidification in the rhizosphere of tithonia there was a decline in resin-P (0.8 g P g–1). However, there was also a decline in NaOH extractable inorganic P (NaOH-Pi) (5.6 g P g–1 at the root surface) and organic P pools (NaOH-Po) (15.4 g P g–1 at 1.5 mm from the root), which would not be expected without specific P acquisition mechanisms. Alkalinisation of tephrosia rhizosphere was accompanied by changes in all measured pools, although the large depletion of organic P (21.6 g P g–1 at 5 mm from the root) suggests that mineralisation, as well as desorption of organic P, was stimulated. The size of changes of both pH and P pools varied with distance away from the rhizoplane. Decline of more recalcitrant P pools with the growth of the agroforestry species contrasted with the effect of maize growth, which was negligible on resin-P and NaOH-Pi, but led to an accumulation of P as NaOH-Po (14.2 g P g–1 at 5 mm from the root). Overall the depletion of recalcitrant P pools, particularly Po, suggests that the growth of tithonia and tephrosia enhance desorption and dissolution of P, while also enhancing organic P mineralisation. Both species appear to have potential for agroforestry technologies designed to enhance the availability of P to crops, at least in the short term.  相似文献

Effects of N source and media-N and P levels were examined on growth, N uptake, and N2 fixation ofAzolla pinnata withAnabaena azollae association (azolla) at two inoculum-P concentrations. Each expeiment was conducted for 7 days in a growth chamber using azolla at a predetermined inoculum-P concentration and the growth media containing a combination of four levels of P (0, 15, 75, and 200 M) and three levels (0, 1, and 5 mM) of either15N-enriched NH 4 + as ammonium sulfate or15N-enriched NO 3 as potassium nitrate. Nitrogen uptake and N2 fixation were measured by15N isotopic dilution method. Tissue P and N, N uptake, and N2-fixation increased with increasing P concentration in the media regardless of the inoculum-P level of azolla. Increasing P concentration in the media increased growth of azolla at low inoculum P, but the effect on high inoculum-P azolla was either small or absen. High inoculum-P concentration resulted in increased growth, tissue-N and P concentrations, N uptake, and N2 fixation by azolla. Ammonium in the growth media caused larger increase in tissue-N and greater repression of N2 fixation than equimolar concentration of NO 3 . In the presence of NH 4 + or NO 3 , in the growth media, N uptake by azolla exceeded the corresponding decrease in N2 fixation, resulting in an overall increase in tissue-N concentration. Phosphorus in the media tended to negate the inhibitory effect of NH 4 + or NO 3 on N2 fixation. A multiple regression model showed that the effect of tissue-N on N2 fixation was negative while that of tissue-P was positive. Therefore, a relative change in tissue-N and P appeared to regulate N2 fixation. Tissue-N and P had similar effects on relative growth rate of azolla also. The effects of ficoll in liquid culture media have been contradictory in previous reports. The objective of this study was to determine the functional properties of ficoll in potato 4 (P4) liquid induction medium and their influence on anther culture responses of wheat. Ficoll addition significantly (p0.01) reduced callus production from the anthers of spring wheat cv. Pavon 76. The reduction was directly related to the concentration of ficoll added within the range of 50 to 200 g l-1 medium. Although the addition of ficoll significantly (p0.01) increased the percentage of regenerable calli and the ratio of green vs. albino plants, the final yield of green plants per 100 anthers was significantly lower. Consistent results also were obtained with four other spring wheat genotypes (Chris, Butte 86, WA 6916, and Edwall). Ficoll concentration affected the density, viscosity, and osmolality of the liquid media. The higher medium density caused by ficoll addition increased the percentage of floating calli, as well as the percentage of regenerable calli and the ratio of green vs. albino plants. However, the increased medium viscosity by ficoll addition significantly (p0.01) reduced callus production. Ficoll addition also increased medium osmolality, which affected callus production by interacting with the sugar concentration of the induction media. Using response functions, the estimated maltose concentration for maximum callus production was 105 g l-1 for the standard P4 media, compared with 68 g l-1 for the ficoll-containing P4 media. These results clearly demonstrate that ficoll addition to the liquid P4 induction medium containing high sucrose concentration (90 g l-1) is deleterious to the maximum production of green plants from wheat anther culture.  相似文献

Three freshwater and one marine algal species were grown under inorganic carbon limitation in laboratory continuous cultures. Comparisons were made between HCO3? alkalinity and bubbled CO2 as carbon sources. HCO3? alkalinity was an excellent source of inorganic carbon below specific pH levels, but chemical precipitation at high pH placed an upper limit on productivity that was far lower than potential light-limiting levels. With bubbled CO2 it was possible to achieve light limitation. The main factor controlling productivity was the mass flux of inorganic carbon added to the culture, which is the product of gas flow rate and influent P level. Small bubbles were more efficient than large bubbles at low gas flow rates and P levels, but led to froth flotation of algal cells and concomitant reductions in productivity at high bubble rates. At 1% CO2 productivity was still dependent on mass fluxes of added carbon, but was independent of bubble size. At high bubble rates with 1% CO2 narcosis was evident. Maximum yields occurred at intermediate dilution rates when inorganic carbon was supplied via bubbled gas.  相似文献

Using primary cultures of gill pavement cells from freshwater rainbow trout, a method is described for achieving confluent monolayers of the cells on glass coverslips. A continuous record of intracellular pH was obtained by loading the cells with the pH-sensitive flourescent dye 2,7-bis(2-carboxyethyl)-5(6)-carboxyfluorescein and mounting the coverslips in the flowthrough cuvette of a spectrofluorimeter. Experiments were performed in HEPES-buffered media nominally free of HCO3. Resting intracellular pH (7.43 at extracellular pH=7.70) was insensitive to the removal of Cl or the application of 4-acetamido-4-isothiocyanatostilbene-2,2-disulfonic acid (0.1 mmol·l–1), but fell by about 0.3 units when Na+ was removed or in the presence of amiloride (0.2 mmol·l–1). Exposure to elevated ammonia (ammonia prepulse; 30 mmol·l–1 as NH4Cl for 6–9 min) produced an increase in intracellular pH (to about 8.1) followed by a slow decay, and washout of the pulse caused intracellular pH to fall to about 6.5. Intracellular non-HCO 3 buffer capacity was about 13.4 slykes. Rapid recovery of intracellular pH from intracellular acidosis induced by ammonia prepulse was inhibited more than 80% in Na+-free conditions or in the presence of amiloride (0.2 mmol·l–1). Neither bafilomycin A1 (3 mol·l–1) nor Cl removal altered the intracellular pH recovery rate. The K m for Na+ of the intracellular pH recovery mechanism was 8.3 mmol·l–1, and the rate constant at V max was 0.008·s–1 (equivalent to 5.60 mmol H+·l–1 cell water·min–1), which was achieved at external Na+ levels from 25 to 140 mmol·l–1. Methanosarcina barkeri was grown by acetate fermentation in complex medium (N2 gas phase). The molar growth yield was 1.6–1.9 g cells/mol methane formed. Under these conditions 63–82% of the methane produced byMethanosarcina strains was derived from the methyl carbon of acetate, indicating that some methane was derived from other media components. Growth was not demonstrated in complex media lacking acetate or mineral acetate medium containing acetate but lacking H2/CO2, methanol, or trypticase and yeast extract. Acetate metabolism byM. barkeri strain MS was further exmined in mineral acetate medium containing H2/CO2 and/or methanol, but lacking cysteine. Under these conditions, more methane was derived from the methyl carbon of acetate than from the carboxyl carbon. Methanogenesis from the methyl group increased with increasing acetate concentration. The methyl carbon contributed up to 42% of the methane formed with H2/CO2 and up to 5% with methanol. Methanol stimulated the oxidation of the methyl group of acetate to CO2. The average rates of methane formation from acetate were 1.3 nomol/min ·ml/culture (0.04mg2 cell dry weight) in defined media (gas phase H2/CO2) and complex media (gas phase N2). Acetate contributed up to 60% of cell carbon formed under the growth conditions examined. Similar quantities of cell carbon were derived from the methyl and carboxyl carbons of acetate, suggesting incorporation of this compound as a two-carbon unit. Incorporated acetate was not preferentially localized in lipid material, as 70% of the incorporated acetate was found in the wall and protein cell fractions. Acetate catabolism was stimulated by pregrowing of cultures in media containing acetate, while acetate anabolism was not influenced. Nitrate uptake and nitrite efflux patterns in Nostoc MAC showed a rapid phase followed by their saturation. Nitrite efflux was maximum in nitrate medium whereas the cells incubated in N2 and NH 4 + media exhibited a decreased nitrite efflux activity. The simultaneous presence of NH 4 + and nitrate significantly decreased nitrite efflux. L-Methionine-Dl-sulphoximine (MSX) prevented inhibition of nitrite efflux by NH 4 + . In the dark there was negligible nitrite efflux, whereas illumination increased the rate of nitrite efflux significantly. The nitrite efflux system was maximally operative at pH 8.0, 30°C and a photon fluence rate of 50 mol m-2. s-1. These results confirm that (i) the nitrite efflux system in Nostoc MAC is dependent upon nitrate uptake and assimilation and is repressible by NH 4 + ; (ii) NH 4 + itself is not the actual repressor of nitrite efflux; a product of NH 4 + assimilation via glutamine synthetase (GS) is required for repression to occur; (iii) the catalytic function of GS does not appear to be involved in nitrate assimilation-dependent nitrite efflux, and (iv) the optimum pH, temperature and illumination for maximum nitrite efflux were found to be 8.0, 30°C and 50mol m-2. s respectively.B.B. Summary At pH 4.0, >10–7 m nigericin was found capable of conducting net charge transfer across bimolecular lecithin membranes, with a stoichiometry of three uncharged ionophore moieties per cation. At neutral or alkaline pH, nigericin catalyzed the transfer of net charge through dimer forms. In agreement with these results, quantitative analysis of nigericin-potassium complexes formed at pH 4.0 showed a 31 ratio, and a 21 ratio at neutral or alkaline pH. A 11 stoichiometry was observed when the ionophore complex was not transferred from methanol-water to chloroform. Moreover,1H-NMR spectra of nigericin-cation complexes formed at pH 4.0, displayed clear-cut chemical shift variations different to those observed at neutral or alkaline pH. Thus, it is apparent that acid pH causes a transition from dimeric to trimeric forms of nigericin-cation complexes. The membrane conductance increased up to ten times when negatively charged phosphatidyl glycerol was used, while the conductance decreased in positively charged cetylpyridinium containing membranes at pH 4.0. These results suggest that the nigericin-K+ oligomeric complex is positively charged. In this respect, pK a values around 8.0 were obtained for the nigericin carboxylate group in media of different dielectric constant, indicating that this chemical group is undissociated under these conditions. Moreover, the values for the complex formation constants as well as the G values calculated for the dimers and trimers indicated that such ionophore cation oligomeric complexes are thermodynamically stable.  相似文献

Whole cells of the methanogen Methanosarcina barkeri were immobilized in an alginate network which was crosslinked with Ca2+ ions. The rates of methanol conversion to methane of entrapped cells were found to be in the same range as the corresponding rates of free cells. Furthermore, immobilized cells were active for a longer period than free cells. The particle size of the spherical alginate beads (1.2 mm-3.7 mm ?) and thus diffusion had no obvious influence on the turnover of methanol. The half-value period for methanol conversion activity determined in a buffer medium was approximately 4 days at 37°C for entrapped cells. The apparent Km value K for such cells was nearly 140mM and the Vmax value was about 1.2 μmol methanol/min/mg entrapped protein. Therefore the high rates of methanol degradation measured, e.g., 0.5 μmol methanol/min/mg entrapped protein, indicated that the immobilization technique preserved the cellular functions of this methanogenic bacterium.  相似文献

The effects of solution acidity and form of nitrogen on net nutrient uptake rates in Acer pseudoplatanus and Calamagrostis villosa seedlings were examined as part of a complex ecological study. Uptake rates were measured by the depletion method under controlled conditions (temperature 20 °C, irradiance 400 mol m–2 s–1 PAR) from a nutrient solution containing 1.5 mM nitrogen in the form of nitrate or ammonium or an equimolar mixture of both. The solution acidity was kept constant at pH 5.5 (control treatment), 4.5 or 3.5 (low pH treatments). Strongly acid pH decreased or stopped the uptake rates of NO3 , Mg2+ and Ca2+, but the uptake of NH4 + was not changed in both species. Ammonium ions reduced the uptake rate of NO3 in Acer but increased the uptake rate in Calamagrostis. Ammonium as the sole source of nitrogen had a strong negative impact on the uptake rates of calcium and magnesium and this effect was independent of the media acidification usually connected with NH4 + uptake and assimilation. However, the negative effect of ammonium ions on the base cation uptake was more pronounced at low pH values.  相似文献

Spectrophotometric techniques have been employed to study the binding of bromophenol red (BPR) to hen egg white lysozyme and the consequent inhibition of enzyme activity. Experimental evidence is given from the dye binding studies in the presence of hexasaccharide and from the studies on activity that BPR binds at a site outside the proposed cleft region (A–F) in such a way that it inhibits the lytic activity towards cell walls but does not inhibit the activity towards hexasaccharide. These observations are consistent with the kinetics of binding [studied using temperature-jump (T-jump)] in the presence of Co++ or chitotriose in large concentrations and the experiments with acetylated lysozyme which suggest that the binding site of BPR is closer to a lysine residue near the cleft. It is suggested that the binding site of BPR could be important in positioning the peptide segment of the cell walls, which are cleaved in the cleft. Evidence for the statement that this binding takes place at least by a two-step process, in which the bimolecular step is followed by a slower monomolecular step, is given from the observations of two types of 1:1 complexes at 24°C in equilibrium studies and from the concentration dependence of the relaxation observed at 605 nm in the T-jump experiments. The binding process is examined by analyzing the T-jump data obtained between 18 and 33°C in the pH range 5.2–9.2 and ionic strength 0.01–01. The ionic strength and pH dependences of the equilibrium constant associated with the bimolecular step k2/k1 and the forward rate constant associated with monomolecular step k3 have been given as evidence for the suggestion that a Coulombic interaction is involved in the first step of binding. However, the final state of binding is hydrophobic in nature. The enthalpy of activation ΔH and the entropy of activation ΔS associated with kf[= k3(k1/k2)] showed compensation behavior with pH variation, with maxima around pH ~ 7.5 in H2O. This has been interpreted as a maximal disordering of water structure in a region of the enzyme at this pH during the monomolecular step. However, the binding of chitotriose or Co++ in the cleft reduces the ΔH and ΔS associated with the monomolecular step of BPR binding, probably by disordering the structured water during their binding in the cleft. The differences in the kinetic parameters obtained in H2O and in D2O probably arise due to subtle differences in the conformation of the enzyme in the two solvents and apart from isotope effects. The correlation between the pH (or pD) dependence of the “intrinsic activity” towards cell walls and ΔH or ΔS indicates that ordered water structure could be playing a role in controlling the catalytic activity. It is also suggested that this factor is associated with the rate constant k3s of the monomolecular step leading to the formation of the final bound state of the substrate in cell lysis, which is also a factor controlling kcat.  相似文献

The decline of Cirsium dissectum in dessicatingwetlands is attributed to acidification and eutrophication. Experimentalevidence was obtained for the first time on ammonium toxicity under low pH. Inahydroculture experiment, interactive effects of nitrogen forms (250mol NH4 + or 250molNO3 ) and pH (4, 5 or 6) were studied with regardtothe vitality of C. dissectum seedlings. The results showthat 250 mol l–1 ammonium as sole nitrogensource only had negative effects on C. dissectum incombination with a low pH. Ammonium uptake at a rhizosphere pH of 4, resultedinlower nitrogen contents of both roots and shoots, lower internal pH of rootsandshoots and increased contents of basic amino-acids, resulting indecreased survival rate and biomass development. At higher pH, or when nitratewas the nitrogen source, these processes do not take place. This phenomenonstresses the importance of periodic influence of base rich groundwater duringthe winter in wet species-rich heathlands and grasslands, necessary torestore the acid neutralising capacity of the soil. Anthropogenic lowering ofthe groundwater table will lead to acidification enabling ammonium to becometoxic to herbaceous plant species such as C. dissectum.  相似文献

Summary The energetics, enzyme activities and end-product synthesis of Zymomonas mobilis 113 in continuous culture were studied after the shift from an anaerobic to an aerobic environment. Aeration diminished ethanol yield and lactic acid concentration, but increased glucose consumption rate and production of acetic acid. After the shift to aerobic conditions reduced nicotinamide adenine dinucleotide (phosphate) [NAD(P)H]-oxidase activity was stimulated. Washed cell suspensions consumed oxygen with glucose, lactate and ethanol as substrates. The aerobic Z. mobilis 113 regulated their intracellular redox balance by production and reoxidation of the end products, coupled with the formation of NAD(P)H. An increase in transmembrane pH gradient (pH) and a decrease in intracellular ATP concentration were observed after the shift to aerobic conditions. At low medium redox potential (Eh) values the H+ balance was regulated in an energy-independent way via end-product excretion. Summary An electric field-mediated transformation (i.e. electroporation) was performed to determine optimal conditions for P. putida transformation. The effects of culture age, electroporation buffer composition, electric field strength, pulse time constant and DNA concentration on transformation efficiency were examined. When plasmid DNA of 8 to 11 kb in size was used with an electroporation buffer containing 1 mM HEPES (pH 7.0), maximum transformation efficiency of 1.0 × 107 transformants/g DNA was obtained at field strength of 12 kV/cm with pulse time of 2.5 millisecond. A linear increase in the number of transformants was observed as DNA concentration was increased over 4 orders of magnitude. A linear relationship was observed between growth phase and transformation efficiency up to OD600 = 2.0. This reliable and simple method should be useful for introduction of plasmid DNA into intact P. putida cells.  相似文献

Three solution experiments were performed to test the importance of NH 4 + versus NO 3 - +NH 4 + to growth of 23 wild-forest and open-land species, using field-relevant soil solution concentrations at pH 4.5. At N concentrations of 1–200 M growth increased with increasing N supply in Carex pilulifera, Deschampsia flexuosa, Elymus caninus and Bromus benekenii. Geum urbanum was the most N demanding species and had little growth below 200 M. The preference for NH 4 + or NO 3 - +NH 4 + was tested also at pH 4.0; no antagonism was found between NH 4 + and H+, as indicated by similar relative growth in both of the N treatments at both pH levels. Growth in solution with NH 4 + relative to NO 3 - +NH 4 + , 200 M, was negatively related to the mean pH of the field occurrence of the species tested; acid-tolerant species grew equally well with only NH 4 + as with NO 3 - +NH 4 + (Oxalis acetosella, Carex pilulifera, Festuca gigantea, Poa nemoralis, Deschampsia flexuosa, Stellaria holostea, Rumex acetosella), while species of less acid soils were favoured by NO 3 - +NH 4 + (Urtica dioica, Ficaria verna, Melandrium rubrum, Aegopodium podagraria, Geum urbanum, Bromus benekenii, Sanguisorba minor, Melica ciliata, Silene rupestris, Viscaria vulgaris, Plantago lanceolata). Intermediate species were Convallaria majalis, Elymus caninus, Hordelymus europaeus and Milium effusum. The transformed root culture of Polygonum tinctorium Lour. was established by infecting leaf explants with Agrobacterium rhizogenes A4. These cultures were examined for their growth and indigo content under various culture conditions. Among the four different culture media tested, SH medium showed the highest yield for root growth (28 mg dry wt/30 ml) and indigo production (152 g/dry wt). In SH medium, 30 g sucrose l–1, 2500 mg KNO3 l–1, 300 mg NH4H2PO4 l–1 were the best conditions for indigo production at pH 5.7. The production of indigo in hairy roots slightly increased with the addition of 200 mg chitosan l–1 (186 g/dry wt) and 20 U pectinase l–1 (181 g/dry wt).  相似文献

Summary The experiments reported here evaluate the capability of isolated intestinal epithelial cells to accomplish net H+ transport in response to imposed ion gradients. In most cases, the membrane potential was kept constant by means of a K+ plus valinomycin voltage clamp in order to prevent electrical coupling of ion fluxes. Net H+ flux across the cellular membrane was examined at pH 6.0 (the physiological lumenal pH) and at pH 7.4 using methylamine distribution or recordings of changes in media pH. Results from both techniques suggest that the cells have an Na+/H+ exchange system in the plasma membrane that is capable of rapid and sustained changes in intracellular pH in response to an imposed Na+ gradient. The kinetics of the Na+/H+ exchange reaction at pH 6.0 [K t for Na+=57mm,V max=42 mmol H+/liter 3OMG (3-O-methylglucose) space/min] are dramatically different from those at pH 7.4 (K t for Na+=15mm,V max=1.7 mmol H+/liter 3OMG space/min). Experiments involving imposed K+ gradients suggest that these cells have negligible K+/H+ exchange capability. They exhibit limited but measurable H+ conductance. Anion exchange for base equivalents was not detected in experiments performed in media nominally free of bicarbonate.  相似文献

The exudation of certain organic anions and protons by roots which may affect solubility of metals and P and uptake by plants, is affected by nitrogen form and pH. The objective of this work was to study exudation of carboxylates and H+/OH by tomato plants in response to NH4/NO3 ratio and pH in nutrient solution. Four NH4/(NH4+NO3) ratios (R= 0, 0.33, 0.67 and 1) and constant vs. variable solution pH treatments were investigated. The sum of the exudation rates of all carboxylates tended to decline with increasing R, particularly tri- and dicarboxylates. The molar fraction of the exuded tri- and dicarboxylates, averaged over all treatments and plant ages, increased in the order tartarate 2%), malate (6%), succinate (15%), citrate (26%) and fumarate (46%). At R=1 the solution pH dropped from 5.2 to 3 and at R=0 increased to 8. The R corresponding to the pH stat of tomato plant was 0.3. For the constant solution pH treatment, the effect of solution pH on carboxylate exudation rate was small as compared to the effect of R. The exudation of citrate and H+ efflux which were initiated when NO3 and NH4 uptake rates per plant exceeded certain threshold values, increased with plant age.  相似文献

Summary The present study describes a new perfusion technique—based on the use of a routine spectrofluorometer—which enables fluorometric evaluation of polarity, regulation and kinetics of Na+/H+ exchange at the level of an intact monolayer. Na+/ H+ exchange was evaluated in bicarbonate-free solutions in OK (opossum kidney) cells, a renal epithelial cell line. Na+/H+ exchange activity was measured by monitoring changes in intracellular pH (pH i ) after an acid load, using the pH-sensitive dye 27-bis (carboxyethyl) 5–6-carboxy-fluorescein (BCECF). Initial experiments indicated that OK cells grown on a permeable support had access to apical and basolateral perfusion media. They also demonstrate that OK cells express an apical pH i , recovery mechanism, which is Na+ dependent, ethylisopropylamiloride (EIPA) sensitive and regulated by PTH. Compared to resting conditions (pH i =7.68; pH o =7.4) where Na+/H+ exchange is not detectable, transport rate increased as pH i decreased. A positive cooperativity characterized the interaction of internal H+ with the exchanger, and suggests multiple H+ binding sites. In contrast, extracellular [Na+] increased transport with simple Michaelis-Menten kinetics. The apparent affinity of the exchanger for Na+ was 19mM at an intracellular pH of 7.1 and 60mM at an intracellular pH of 6.6. Inhibition of Na+/H+ exchange activity by EIPA was competitive with respect to extracellular [Na+] and theK i was 3.4 M. In conclusion, the technique used in the present study is well suited for determination of mechanisms involved in control of epithelial cell pH i and processes associated with their polarized expression and regulation.  相似文献
