This study presents the results of the first genetic analysis of ancient chestnut trees (Castanea sativa Mill.) in Italy and in the Iberian Peninsula to better understand the effect of grafting on the domestication process of chestnut and to investigate the impacts of early selection and improvement on the genetic diversity retained. We evaluated 105 giant ancient trees from Italy, Spain and Portugal and compared them with the European Union (EU) database of chestnut cultivars by using a set of 24 simple sequence repeats (SSRs; microsatellite markers). We measured the perimeter (girth) at the diameter at breast height (DBH). Samples from both the canopy and the roots of each tree were analysed to distinguish which trees were self‐rooted and which were grafted. Diversity was compared using standard metrics and model‐based approaches based on the expected heterozygosity (He) at equilibrium. We could differentiate 91 new genotypes; 9.6% matched known chestnut cultivars. We found the first evidences of cultivation, that is, grafting to produce “instant domestication” in Galicia and in the Douro Valley in trees of 14‐m perimeter (15th century) and in the Basque Country (first report in that area) in a tree of 11.5‐m perimeter (16th century). In Italy, the cultivar “Marrone Fiorentino” was found in some giant trees with perimeters of 8 and 9 m (17th‐18th centuries) in the Toscana and Umbria. Those findings matched with written references in Portugal from the 16th century and from the 18th century in Spain. “Instant domestication” could be dated back to the 15th century and was related to the wild populations existing in the same areas where cultivars are being propagated, without a different genetic structure for wild chestnut trees and with a high diversity maintained through the initiation of domestication. 相似文献

Thirteen polymorphic microsatellite loci were isolated from Castanea sativa (Mill.). Six contained dinucleotide repeats, six contained trinucleotide repeats, and one contained a compound microsatellite of a trinucleotide and a tetranucleotide repeat. The loci were characterized using C. sativa trees from three populations in the UK and the parents and six seedlings from a Turkish mapping population. The number of alleles revealed varied from two to 14 (mean = 5.15) per loci. Eight loci were found to be useful in the mapping family. 相似文献

The tortricid moth Lobesia botrana is considered to be an important pest of the cultivated grapevine (Vitis vinifera, Vitaceae). It feeds on young flower buds and young fruits, and is the primary vector of the ‘noble mould’ fungus (Botrytis cinerea). In order to study the population dynamics and genetic structure of this species in space and time, we developed seven polymorphic microsatellite markers (three to 12 alleles). Although the presence of null alleles is suspected, these polymorphic loci are likely to provide information on the population genetics of L. botrana, and could help in the development of an efficient control strategy against this pest. 相似文献

Analysis of population genetic structure is a key aspect to understand insect pest population dynamics in agricultural scenarios. Here the role of geography, hosts and time on the population genetic structure of codling moth Cydia pomonella (Linnaeus) (Lep., Tortricidae) populations is described. Temporal variation was examined in two French orchards among each of three adult flights during two successive years. Analyses were conducted using two insecticide resistance markers (variation at the sodium channel gene and enzymatic activity of cytochrome P450 oxidases) and three microsatellite loci. Levels of genetic variation among temporal populations were not significant based on variation in the sodium channel gene and microsatellite loci. However, P450 oxidase activity differed significantly during both flights and years, decreasing during the three flights of the first year and increasing during the second. These results suggest that phytosanitary measures are among the factors shaping the genetic structure of C. pomonella populations over temporal and geographical scales. We discuss the relative importance of natural and passive dispersal related to anthropogenic activities affecting C. pomonella population genetics and highlight population genetic research needs in order to design more efficient pest management practices. 相似文献

Deriving useful microsatellite markers in lepidopterans has been challenging when relying on scans of genomic DNA libraries, presumably due to repetitiveness in their genomes. We assayed 96 of 320 microsatellites identified in silico from a collection of Bicyclus anynana ESTs, in 11 independent individuals from a laboratory population. From the 68 successful assays, we identified 40 polymorphic markers including 22 with BLAST-based annotation. Nine of 12 selected polymorphic markers tested in a panel of 24 wild-caught individuals converted to successful assays and were all polymorphic. We discuss how microsatellite discovery in ESTs is an efficient strategy with important attendant advantages. 相似文献

Thirteen polymorphic microsatellite loci suitable for population genetic studies of Helicoverpa zea were discovered by screening partial genomic libraries enriched for microsatellite sequences. Insects collected (N = 96) in Stoneville, Mississippi were used to characterize these markers. The observed and effective number of alleles per locus ranged from two to nine (average 4.46) and from 1.07 to 2.45 (average 1.81), respectively. Fisher exact tests detected significant deviations from Hardy–Weinberg equilibrium at three loci, probably due to inbreeding, null alleles, or Wahlund's effect. Significant genotypic disequilibrium was not detected between any pair of loci. 相似文献

应用70个微卫星标记分析了3个籼稻测验种和3个粳稻测验种的多态性,发现其中36个标记可以区分籼粳测验种。再以18个籼粳品种进一步筛选,找到了分布于12条染色体的21个籼粳特异性微卫星标记。在这21个标记中,20个在籼粳亚种间带型相异,其中7个在亚种内带型一致,13个在亚种内带型不一致;1个标记在12个籼稻品种和1个粳稻品种检测到相同的带型,其余11个粳稻品种具有另一种带型。微卫星标记和RFLP标记检测籼粳亚种不仅具有一致性,而且还有互补性。Abstract:Six indica and japonica testers were assayed using 70 microsatellite markers.Thirty-six markers distinguishing indicas from japonicas were detected.By further-screening among 18 indica and japonica varieties,21 markers distributed on 12 rice chromosomes were found to be indica-japonica differentiated.No indica varieties shared same patterns with any japonica varieties at 20 marker loci,of which identical patterns were observed within subspecies at 7 loci while within-subspecies variations were observed at 13 loci.At the remaining locus,12 indica and 1 japonica varieties had the same allele,while other 11 japonica varieties had another allele.It also showed that SSLP was not only consistent,but also complementary,to RFLP for the subspecies identification. 相似文献

Pheromone biosynthesis in the redbanded leafroller moth, Argyrotaenia velutinana, was stimulated by homogenates of the bursa copulatrix. Although pheromonotropic activity was also extractable from the ovary, the activity of pheromone biosynthesis activating neuropeptide (PBAN) or bursa extracts was not impaired in isolated abdomens by removal of the ovary. Response to the bursa extracts was dependent on the dose administered and the time of incubation. Amounts of pheromone present in adult females of different ages appeared to be correlated with the extractable amount of pheromonotropic activity from their bursa copulatrix. Decapitation did not result in the suppression of burse factor production. Homogenates of the bursa elicited similar effects in both isolated gland and isolated abdomen incubations, but the brain neuropeptide, PBAN, was less active in the former than in the latter. Bursa extracts stimulated pheromone production in isolated abdomen incubations deprived of the bursa copulatrix, but PBAN did not. Loss of activity of bursa homogenates after treatment with either pronase E or carboxypeptidase Y indicated that the pheromonotropic factor is a proteinaceous substance. The mechanism through which pheromone production is regulated in redbanded leafroller moths is discussed. © 1992 Wiley-Liss, Inc. 相似文献

Campuloclinium macrocephalum (Less.) DC. (Asteraceae) (pompom weed), an invader in South Africa and Swaziland, threatens biodiversity conservation, agriculture and tourism in the region. We report on the host range and impact of the flower-feeding moth, Cochylis campuloclinium Brown (Lepidoptera: Tortricidae), the second insect biological control agent to be considered for C. macrocephalum in South Africa. Laboratory host-specificity tests were conducted on 31 Asteraceae species. Field host range studies included 17 non-target Asteraceae species. Results of both C. campuloclinium laboratory and field host-range trials indicated that it is suitably host specific. In laboratory host-range trials, only C. macrocephalum and the closely related native, Adenostemma caffrum DC. (Asteraceae), received feeding damage, while in field host-range trials, the moth was only recorded on the target. Laboratory impact studies showed that C. campuloclinium destroyed a significant number of florets in flower buds (76%) and seeds in mature flowers (54%). Based on evidence from the native range, there appears to be no competitive interactions between C. campuloclinium and the already established stem- and leaf-deforming thrips, Liothrips tractabilis Mound & Pereyra (Thysanoptera: Phlaethripinae). The two insect agents should perform a complementary role of reducing flowering (L. tractabilis) and seed production (C. campuloclinium). The impact of low levels of spinosad on the obliquebanded leafroller, Choristoneura rosaceana Harris (Lepidoptera: Tortricidae), and the koinobiont endoparasitoid, Apophua simplicipes Cresson (Hymenoptera: Ichneumonidae), was assessed when the parasitoid was in the larval stage within second‐ and fourth‐instar hosts. These are developmental stages that would be exposed to spring orchard treatments of the insecticide. Oral spinosad LC50 levels for unparasitized obliquebanded leafroller hosts were <1% of the recommended orchard treatment levels. Apophua simplicipes survival was significantly reduced within parasitized spinosad‐treated second‐ and fourth‐instar larval hosts. Both the leafroller host and parasitoid were much more susceptible (ca. 65‐fold) to spinosad when larval hosts fed on spinosad‐treated leaf material as opposed to being treated topically. When hosts were exposed to extremely low doses of spinosad, a small percentage of parasitoids was able to survive to emerge as adults. These laboratory trials predict that applications of spinosad may reduce biological control of C. rosaceana populations by ichneumonid endoparasitoids developing within treated hosts. 相似文献

We appraised mating disruption (MD) to control pea moth, Cydia nigricana (Fabricius) (Lepidoptera: Tortricidae), by assessing male attraction to monitor traps, larval pod infestation, and larval age structure in pheromone‐treated and untreated grain pea fields [Pisum sativum L. (Fabaceae)], over a 5‐year period. Cellulose pheromone dispensers were manually attached to the top shoots of pea plants and released 540 mg ha?1 day?1 synthetic pheromone E8,E10‐dodecadien‐1‐yl acetate in a first test series (2000–2001) and ca. 4 200 mg pheromone ha?1 day?1 in a second series (2004–2006). The dispensers had a half‐life of about 30 days. Although male attraction to pheromone monitoring traps was largely suppressed at the edges and within MD fields in both test series, MD treatments did not reduce pod infestation in the open field in 2000 and 2001. In the 2004–2006 series, larval damage reduction was achieved in the majority of the trials but overall MD efficacy in the open field was only 61% and not significant. In contrast, in field cages placed within the experimental sites and supplied with unmated pea moths, MD control was consistently high and significant. There were no obvious differences in the larval age distribution in all MD and control treatments, suggesting that infestations started and developed further similarly. As a univoltine species, C. nigricana larvae stay in the soil of pea fields for hibernation and pupate. The following year, emerging adults disperse and fly to the closest pea crop. Combined emergence site and pea crop treatments were conducted over 2 years to include this early migration phase of C. nigricana adults. However, the emergence site treatments did not enhance MD‐control efficacy. We conclude that mating activity was only prevented in cage tests, whereas substantial mating occurred during the transit phase outside the pheromone‐treated fields either within non‐crop vegetation and/or at the edges of pheromone‐treated pea fields orientated upwind. Thus, resulting gravid female entry can be regarded as the major constraint to reliable MD control. 相似文献

Inundative releases of Trichogramma minutum Riley were made in a commercialblueberry field in British Columbia, Canada, todetermine the extent of dispersal of adult wasps, andto determine whether complete parasitism of eggswithin egg masses of the target host [oblique-bandedleafroller (OBLR), Choristoneura rosaceana(Harris)] occurred. Three weekly releases of waspswere made from a single release point in the center ofeach field during the flight of first generation OBLRadults. Sticky trap, sentinel egg mass andtarget-host egg mass data all indicated only a limitedrange of dispersal of T. minutum within thefield. In addition, only partial parasitism of eggmasses of the target host occurred even in closeproximity to the release point. We discuss theimportance of determining the response of Trichogramma females to previously-parasitized eggswhen using Trichogramma wasps for biologicalcontrol of lepidopterous species that oviposit largeegg masses. 相似文献

Mating competitiveness and pheromone trap catches of mass‐reared, male codling moth, Cydia pomonella (L.) (Lepidoptera: Tortricidae), from the Osoyoos, British Columbia, Canada, mass‐rearing facility operated by the Okanagan‐Kootenay Sterile Insect Release Board, were compared to wild males using mark–release–recapture field experiments in spring, summer, and autumn at Summerland, British Columbia. In spring, significantly more wild diapause males mated with tethered, wild females than did non‐irradiated (0 Gy) or irradiated (100 or 250 Gy) non‐diapause, mass‐reared males. A lower dose of radiation did not improve mating competitiveness, nor catches of mass‐reared males released in spring. Median mating time for wild males was approximately 45 min earlier than mass‐reared males with most wild males (70.5%) mating before sunset and mass‐reared males mating at or shortly after sunset in spring. Superior mating competitiveness of wild males in spring was mirrored by greater recapture rates in pheromone‐baited traps. In summer, mating competitiveness of mass‐reared moths improved relative to wild males and there was a significant inverse relationship between radiation dose (0, 100, and 250 Gy) and competitiveness of mass‐reared males. In autumn, untreated, wild males were significantly more responsive to pheromone traps than non‐diapausing mass‐reared males receiving 250 Gy of radiation. Mass‐reared males, subjected to diapause‐inducing conditions as larvae and emerged from diapause before this irradiation treatment, were recaptured significantly more often than similarly irradiated, non‐diapause, mass‐reared males, but not more than untreated, diapause wild males. We hypothesize that differences between wild and mass‐reared males in daily timing or speed of responses to natural or synthetic pheromone sources under montane weather patterns typical of spring in British Columbia may partially explain poor activity of sterile males, and low sterile : wild overflooding ratios during spring when measured using pheromone traps by the sterile insect release programme in British Columbia. 相似文献

- 1 A method for area‐wide risk assessment of pea moth infestation in commercial pea‐growing areas based on spatial and temporal analyses of pea moth abundance and the phenological distribution of pea fields was investigated.
- 2 In a commercial pea‐growing region in Saxony, Germany, all pea fields were identified, mapped and characterized, recording the pea plant phenology, pea moth flight and larval infestation of each field in the years 2006–2008.
- 3 The relationship between pea moth flight and pea plant phenology was studied in detail in small‐scale field experiments in Hesse, Germany, using different pea cultivars and sowing dates.
- 4 In the study area, the abundance of Cydia nigricana Fabricius (Lepidoptera: Tortricidae) in organic green peas increased linearly with the pea‐cropping area of the previous year in the surroundings of the current fields according to the continuous abundance index.
- 5 Considering solely the early flowering period (= early pea sowing dates) of the organic green peas, we calculated that a minimum distance of the current pea field to the nearest pea field of the previous year of 500 m was necessary to significantly reduce pea moth flight and larval infestation.
- 6 In small‐scale field experiments, a correlation between pea moth flight and larval investation, as well as the importance of the pea flower for the pea moth occurrence, was demonstrated.

The search for mates and food is mediated by volatile chemicals. Insects sense food odorants and sex pheromones through odorant receptors (ORs) and pheromone receptors (PRs), which are expressed in olfactory sensory neurons. Molecular phylogenetics of ORs, informed by behavioral and functional data, generates sound hypotheses for the identification of semiochemicals driving olfactory behavior. Studying orthologous receptors and their ligands across taxa affords insights into the role of chemical communication in reproductive isolation and phylogenetic divergence. The female sex pheromone of green budworm moth Hedya nubiferana (Lepidoptera, Totricidae) is a blend of two unsaturated acetates, only a blend of both elicits male attraction. Females produce in addition codlemone, which is the sex pheromone of another tortricid, codling moth Cydia pomonella. Codlemone also attracts green budworm moth males. Concomitantly, green budworm and codling moth males are attracted to the host plant volatile pear ester. A congruent behavioral response to the same pheromone and plant volatile in two tortricid species suggests co‐occurrence of dedicated olfactory channels. In codling moth, one PR is tuned to both compounds, the sex pheromone codlemone and the plant volatile pear ester. Our phylogenetic analysis finds that green budworm moth expresses an orthologous PR gene. Shared ancestry, and high levels of amino acid identity and sequence similarity, in codling and green budworm moth PRs offer an explanation for parallel attraction of both species to the same compounds. A conserved olfactory channel for a sex pheromone and a host plant volatile substantiates the alliance of social and habitat signals in insect chemical communication. Field attraction assays confirm that in silico investigations of ORs afford powerful predictions for an efficient identification of behavior‐modifying semiochemicals, for an improved understanding of the mechanisms of host plant attraction in insect herbivores and for the further development of sustainable insect control. 相似文献

Since the discovery of Lobesia botrana Denis & Schiffermüller (Lepidoptera: Tortricidae) sex pheromone, it has played an important role in the control and detection of this pest, for example, through the use of pheromone‐baited traps and mating disruption techniques. Rubber septa are the most common pheromone dispensers used in monitoring traps, but often dispenser performance is not optimized. The key to improve methods based on pheromones as attractants (monitoring, mass trapping, or ‘attract and kill’) is to know the optimum emission interval, because release rates can strongly affect the attraction. In this work, five levels of pheromone load with different release rates were compared in traps using mesoporous pheromone dispensers to investigate the optimum release rate maximizing L. botrana catches. Residual pheromone loads of the dispensers were extracted and quantified by gas chromatography, to study release profiles and to estimate the various emission levels. The efficacy of pheromone emission was measured in field trials as number of moths caught. A quadratic model was fitted to relate the numbers caught vs. the daily emission rates. The resulting quadratic term was statistically significant, confirming the existence of a relative maximum for L. botrana catches. Taking into account that the trial was carried out only in one location, an optimum emission value of ca. 400 μg per day could be considered to enhance the attraction of L. botrana under West‐Mediterranean weather conditions. 相似文献

Abstract: Host plant patch size plays an important role in the distribution and densities of many insect species. Understanding how the host plant patch size influences the population of a biological control agent is necessary to monitor the success of a biological control programme. Urophora quadrifasciata (Dipt. Tephritidae) was released in the early 1970s to control the spotted knapweed, Centaurea biebersteinii (Asteraceae) in North America. The studies reported here investigated the influences of spotted knapweed patch size and stem density on U. quadrifasciata larval populations overwintering within capitula in the Upper Peninsula of Michigan, USA. An inverse relationship was found between the percentage of capitula with U. quadrifasciata larvae and spotted knapweed patch size in 2002 from Houghton county, MI. In 2003, an inverse relationship was also found in the south region of the study area in Houghton and Keweenaw counties, Michigan. The percentage of capitula with U. quadrifasciata larvae and spotted knapweed patch size were positively related for the central region. No relationship was found for the North region in 2003. Dispersal of U. quadrifasciata is directed north-east along the Keweenaw Peninsula, Michigan. With this directed spread, areas with well-established populations of U. quadrifasciata would provide source individuals for new colonization. Spotted knapweed patch size may play an important role in the overwintering populations of U. quadrifasciata as a result of dispersal. 相似文献
