为解决土壤中残留的菊酯类农药问题,通过富集筛选法从70余份土壤中获得一株高效分解利用菊酯类农药的微生物菌株SSCL-5,该菌株可在含1 000 mg/L的氯氰菊酯无机盐培养基中正常生长。经形态学及ITS测序鉴定,确定该菌为草酸青霉(Penicillium oxalicum),对多种高浓度菊酯类农药耐受。经紫外分光光度法及HPLC验证该菌株草酸青霉SSCL-5在无机盐培养基、28℃、180 r/min摇瓶培养24 h的条件下,对400 mg/L氯氰菊酯的降解率为97%。土壤室内试验证明该菌在土壤中,温度20-34℃、水分含量保藏40%-60%条件下,30 d可将土壤中400 mg/L的氯氰菊酯降解67.6%。  相似文献

应用气升式反应器培养草酸青霉菌菌丝球的研究   总被引:4,自引:0,他引:4  
应用0.5L气升式反应器成功实现了对高效染料吸附丝状真菌——草酸青霉菌(Penicillium oxalicum)的颗粒化,初步解决了菌体与培养液分离的问题。以OD600为0.23的孢子悬浊液接种时,菌丝成球的最佳接种量为2.5%,最佳通气量为0.5L/min。菌球直径及沉降速度随时间均呈线性增长。在最佳接种量(2.5%)和最佳通气量(0.5L/min)下,30℃培养72h后,菌球的直径和沉降速度分别为(4.0±0.8)mm和(14±3)mm/s。  相似文献

A beta-glucosidase from the medium of an autolyzed culture of Penicillium oxalicum has been purified by tannic acid precipitation, sephacryl S-200, DEAE-Biogel, CM-Biogel and Mono Q successively. The purification process produced a homogeneous band in the SDS-PAGE that correspond to a Mr of 133,500. The enzyme had a pl of 4, and the active optima were found at pH 5.5 and 55 degrees C. The enzyme hydrolyzed different substrates showing maximum affinity against p-nitrophenyl-beta-D-glucoside with a Km value of 0.37 mM. The beta-glucosidase was inhibited by Glucono-D-lactone but not by glucose in the concentration range of 1 to 10 mM. The enzyme was adsorbed by Concanavalin-A-Sepharose.  相似文献

High 1,3-beta-glucanase activity was detected during autolysis in a culture medium containing Penicillium oxalicum. It was due to the combined action of four enzymes. The purification process for the major enzyme produced a homogeneous band in the SDS polyacrylamide gel that corresponded to a molecular weight of 79,400 daltons. The enzyme pI was 6.3 and it was only active against 1,3-beta-glucans, with a S0.5 of 0.23 mg ml-1 against laminarin. The enzymatic optima were found at pH 4 and 55 degrees C, and instability was evident when pH and temperature were altered. The enzyme was not active against oxidated laminarin and was barely inhibited by glucono-D-lactone. Hg2+, Ag+ and Fe2+ were effective inhibitors. The enzyme was adsorbed by concanavalin-A-sepharose.  相似文献

【目的】探讨含钾硅酸盐矿物在草酸青霉(Penicillium oxalicum)作用下的风化及钾溶出情况。【方法】利用等离子体发射光谱、X-射线能谱、X-射线光电子能谱分析了3种常见含钾硅酸盐矿物(钾长石、白云母和黑云母)在草酸青霉作用后浸出液和矿物表面元素含量的变化;通过X-射线衍射分析矿物晶相结构的变化;采用共聚焦激光扫描显微镜表征了草酸青霉在矿物表面形成的生物膜;测量了培养液中不同碳源与氮源组分对草酸青霉解钾的影响。【结果】草酸青霉对结构稳定的钾长石和白云母风化速率较低,相比而言黑云母容易被风化并释放可溶性钾元素;草酸青霉在矿物表面形成了网状结构的生物膜,有利于微环境的生成及有机酸在其内的富集,促进微环境内钾的释放,强化微生物对矿物的风化作用;草酸青霉对多种碳源及氮源都表现出较好的适用性。【结论】草酸青霉是一种能够促进多种含钾矿物风化和钾溶出的真菌,在堆肥和生物肥料领域具有广泛的应用前景。  相似文献

An enzyme producing micro-organism, which can directly saccharify rice straw that has only been crushed without undergoing the current acid or alkaline pretreatment, was found. From the homology with the ITS, 28S rDNA sequence, the strain named A592-4B was identified as Penicillium oxalicum. Activities of the A592-4B enzymes and commercial enzyme preparations were compared by Novozymes Cellic CTec2 and Genencore GC220. In the present experimental condition, activity of A592-4B enzymes was 2.6 times higher than that of CTec2 for degrading milled rice straw. Furthermore, even when a quarter amount of A592-4B enzyme was applied to the rice straw, the conversion rate was still higher than that by CTec2. By utilizing A592-4B enzymes, improved lignocellulose degradation yields can be achieved without pre-treatment of the substrates; thus, contributing to cost reduction as well as reducing environmental burden.  相似文献

【背景】柑橘是我国南方农村地区的一种重要经济作物,每年因采后真菌感染导致大量柑橘腐烂和劣变,造成了巨大的经济损失,严重影响柑农的积极性。【目的】丝状真菌SG-4对柑橘采后致腐菌橘青霉(Penicillium citrinum)和指状青霉(Penicillium digitatum)具有很强的拮抗作用,为明确其抑菌的物质基础,对该菌进行了生物学鉴定和活性成分分析。【方法】将SG-4菌株的核糖体DNA的内转录间隔区进行测序,结合菌株形态进行鉴定;对其菌体相和培养液进行抑菌活性追踪,利用醇沉淀和反相高效液相色谱分离得到主效物质,通过高分辨质谱和核磁共振等波谱分析方法确定结构;采用离体的牛津杯抑菌圈法和柑橘的体内接种试验来评价其抑菌活性;通过液相分析测定了活性物质在发酵液中的含量。【结果】分子和形态鉴定表明SG-4是一株草酸青霉(Penicillium oxalicum),其抑制柑橘采后致腐菌的活性物质位于培养液的水相提取物中,且具有强极性。进一步研究表明其活性成分为一种新型线性醇溶五肽,含3个具羟基的氨基酸,并且富含甲基化修饰,构成寡肽的第一个氨基酸为β-氨基丁酸(beta-aminobut...  相似文献

草酸青霉能产生完整的纤维素酶和木聚糖酶酶系,其纤维素酶基因的表达主要受转录因子的调控。前期工作中,通过对草酸青霉菌株HP7-1在不同碳源培养基培养条件下转录组的比较分析,获得了调控纤维素酶和木聚糖酶产量的候选调控基因集。本研究以草酸青霉ΔPoxKu70为出发菌株,通过同源重组法,构建并获得了其中一个候选调控基因POX05145的缺失突变株ΔPOX05145。在微结晶纤维素Avicel诱导培养条件下,与出发菌株ΔPoxKu70相比,ΔPOX05145的纤维素酶产量和木聚糖酶产量发生了显著改变。其中,在诱导第2天时,ΔPOX05145对硝基苯-β-D-纤维二糖苷酶产量和木聚糖酶产量分别上升43.4%和164.7%,对硝基苯-β-D-半乳糖吡喃葡萄糖苷酶产量下降92.8%,但是,滤纸酶产量和羧甲基纤维素酶产量没有显著变化。然而,在诱导第4天时,所有纤维素酶产量和木聚糖酶产量上升100.4%~294.0%。实时荧光定量PCR检测表明POX05145在不同的时间不同程度的调控主要的纤维素酶基因和木聚糖酶基因的表达。序列分析表明POX05145含有一个GAL4类锌指结构的DNA结合功能域和一个保守的真菌特有的转录因子结构域(Fungal_TF_MHR)。  相似文献

A phytase from Penicillium oxalicum PJ3, PhyA, was purified near to homogeneity with 427-fold increase in specific phytase activity by ammonium sulfate precipitation, gel filtration, and ion-exchange chromatographies. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and zymogram analysis of the purified enzyme indicated an estimated molecular mass of 65 kD. The optimal pH and temperature of the purified enzyme were pH 4.5 and 55°C, respectively. The enzyme activity was strongly inhibited by Ca2+, Cu2+, Zn2+, and phenylmethylsulfonyl fluoride (PMSF). The Km value for sodium phytate was 0.545 mM with a Vmax of 600 U/mg of protein. The phyA gene was cloned, and it contains an open reading frame of 1,383 with a single intron (118 bp), and encodes a protein of 461 amino acids.  相似文献

采用响应面分析法对草酸青霉(Penicillium oxalicum)L5菌株液体发酵产果胶酶条件进行了优化。结果表明:桔皮粉、米糠及硫酸铵的添加量分别为4.85%、5.89%、0.97%,摇瓶初始pH为6.0~8.0,接种量为9%时,优化后的果胶酶活达54 391.70 U,是初始酶活18 148.00 U的3倍。另外,对其果胶酶性质进行了初步探讨,结果表明:该酶较适反应温度和pH分别为50℃和5.0;30~50℃时有较好的热稳定性,pH值为5.0时稳定性最佳。  相似文献

A Penicillium oxalicum strain was capable of the phosphate-sensitive utilization of phosphonoacetic acid as the sole source of phosphorus. A carbon-to-phosphorus bond-cleavage enzyme yielding acetic acid and inorganic phosphate was detected and characterized in extracts from cells grown on this phosphonate. 为筛选具抗疟疾活性的的内生真菌,采用平板分离法对海南金鸡那树内生真菌进行分离纯化。利用"4日抑制疟疾"测试内生真菌发酵液的抗疟活性,采用TLC、HPLC等分析技术对内生菌发酵液等成分进行分析。从金鸡那树叶片分离到31种内生真菌,通过筛选找到了1株具有抗疟疾活性的产奎宁或奎宁类似物的内生真菌。从金鸡那树叶片中分离得到1株产奎宁的结实串孢霉属内生真菌,为奎宁的资源开发开辟了新途径。  相似文献

为探究蕨类植物内生真菌的多样性,本研究采用可培养方法分离江西龙虎山和广东丹霞山的岩壁常见蕨类植物——卷柏的内生真菌,结合形态特征和ITS序列进行菌株鉴定、研究多样性。结果表明:从1 080个组织块中,共分离出234株菌株,卷柏内生真菌的总定殖率为18.98%,总分离率为21.67%。根据分离频率,毛壳菌属Chaetomium(26.50%)、青霉属Penicillium(18.38%)、棒束孢属Isaria(11.97%)、曲霉属Aspergillus(9.42%)为优势属。不同居群地春季卷柏的内生真菌多样性存在差异,龙虎山的分离率为34.72%,而丹霞山仅为6.39%,其优势属也不尽相同。对龙虎山不同季节的卷柏内生真菌多样性研究表明,春季的分离率为34.72%,高于冬季的23.89%,春、冬两季之间的真菌类群相似性系数仅为0.39,差异显著。研究结果将有助于进一步揭示丹霞地貌地区蕨类植物内生真菌物种组成及其影响因素。  相似文献

研究内生青霉菌(Penicillium sp.B-4)胞外纤维素酶在槐米总黄酮提取中的辅助应用。内生青霉菌在起始pH4,5的综合马铃薯培养基中,150r/min,40℃下摇瓶,培养7d,具有较高的纤维素酶比活力(3.57U/mL)。槐米干粉投入青霉菌发酵液中进行酶解处理,比较酶料比、酶解温度、酶解时间和酶解液pH对槐米总黄酮提取率的影响,发现槐米干粉以酶料比40:1(mL/g)加入粗酶液中,在pH4.5、温度40℃下酶解处理1h后,黄酮提取率可达12.2%,比常规提取率增加了38.7%。内生菌纤维素酶辅助提取法为槐米黄酮提取的可行新方法。  相似文献

喜树内生真菌抗水稻纹枯病菌活性的研究   总被引:2,自引:0,他引:2  
从喜树健康组织叶、茎以及果实中经过分离、纯化共得到26株内生菌株。利用生长速率法测定这26株菌株对植物病原菌——水稻纹枯病病原菌的抑制作用。抑菌试验结果显示:26株菌株中有22株喜树内生真菌的发酵液对植物病原菌菌丝生长均有不同程度的抑制作用;其中,菌株XSY10的发酵液抑菌效果最强,达到了74.97%。水稻盆栽试验结果表明:喷施菌株XSY10次生代谢产物7d后,对水稻纹枯病的防治效果为34.77%。以上试验结果说明喜树内生真菌XSY10对植物病害的防治具有一定的功效。  相似文献
