Transgenic white poplar (Populus alba L.) plants expressing a novel Arabidopsis thaliana cysteine proteinase inhibitor (Atcys) gene have been produced using Agrobacterium tumefaciens-mediated gene transfer. Internodal stem segments of cv. Villafranca were co-cultivated with the EHA105 pBI-Atcys A. tumefaciens strain. Sixteen putative transgenic plant lines were regenerated from different calli with a transformation efficiency of 11%. The integration and expression of the cysteine proteinase inhibitor (Atcys) gene into the plant genome was confirmed by Southern and northern blot analyses. Papain inhibitory activity was detected in poplar transgenic tissues by means of a specific in vitro assay. Such activity was sufficient to inhibit most of the digestive proteinase activity of chrysomelid beetle (Chrysomela populi L.) and confer resistance to C. populi larvae on selected transgenic plants. A close correspondence between the inhibition of papain and resistance to poplar leaf beetle was observed in all tested transgenic lines. Our results indicate that Atcys could be succesfully employed in breeding programmes aimed at the selection of new poplar genotypes resistant to major insect pests.  相似文献

1 The defoliator beetle, Mesoplatys ochroptera Stål, is a serious pest of the legume tree sesbania (Sesbania sesban (L.) Merrill) in agroforestry systems in southern Africa. The survival of the overwintering stage of M. ochroptera and post‐ emergence longevity and fecundity of adults in sesbania fallows in eastern Zambia were quantified. 2 Only adult M. ochroptera survived during the winter (May–August) and the dry season (September–October) hidden under weeds, plant litter, in soil cracks and under rocks. During these periods, some beetles were occasionally found feeding on S. sesban, particularly in natural stands in humid areas. The adults were parasitized by the braconid Perilitus larvicida van Achterberg at a rate of 8.5–16.4%. The beetles stayed in the winter refuges for up to 210 days and emerged with the first heavy rains in October–November. The effective survival of beetles overwintering in an insectary was 18.1% in 1998 and 37.7% in 1999, and that of beetles overwintering in the field was estimated at 0.4 and 2.8% in 1998 and 1999, respectively. 3 The post‐emergence longevity and oviposition period of females in the insectary was 14–31 days and 5–29 days, respectively. The post‐emergence fecundity varied from 87 to 783. 4 It is concluded that the serious M. ochroptera infestation observed every year following the rains is due to synchronous emergence of overwintered resident adult populations and their high capacity for reproduction on many species of Sesbania. Recommendations for pest management in sesbania planted‐fallows are given.  相似文献

Field parasitism of two lady beetles, a recently introduced species, Harmonia axyridis Pallas, and an indigenous species, Coleomegilla maculata lengi Timberlake, by Dinocampus coccinellae Schrank (Hymenoptera: Braconidae) was investigated in southwestern Québec. Adult coccinellids were sampled from June to September 2001 in alfalfa and corn fields. Parasitism rates were recorded by dissection and rearing of coccinellids in controlled condition. The average proportions of coccinellids with a parasitoid larva were 4.6%for H. axyridis and 32%for C. maculata but 0%of the H. axyridis and 5.9%of theC. maculatawere successfully parasitized. In July, more C. maculata than H. axyridiswere found to be parasitized and among all individuals dissected, C. maculata were more frequently found superparasitized than H. axyridis. Our results suggest that D. coccinellae is not well adapted to H. axyridis in Quebec. This is the first mention of H. axyridis being attacked by D. coccinellae in Canada.  相似文献

In China, an unintentionally introduced beetle Ophraella communa shows good control efficiency against invasive common ragweed, Ambrosia artemisiifolia. With the aim of understanding the capacity of O. communa to overcome sub-zero low temperature rapidly, the longevity, survival and fecundity of the beetle adults were studied at conditions of different rapid cold-hardening in the laboratory. The results showed that approximately 20% female or male individuals still survive at ?12°C for 2 h, thus the discriminating temperature for rapid cold-hardening of both female and male beetles is ?12°C. The survival, longevity and fecundity of adult beetles were significantly affected by rapid cold-hardening treatments. Compared to the control, the survival rates of adult beetles experiencing 2 h of rapid cold-hardening at 2, 5 and 8°C prior to exposure to its discriminating temperature (?12°C) were significantly increased. Adult longevity and fecundity were significantly higher after pre-treatment at 2, 5 and 8°C for 2 h than those of the control. Both longest longevity and maximum fecundity of adult beetles were observed at the rapid cold-hardening of 2°C. Our experiment implies that the cold-tolerance ability of O. communa can be rapidly increased after rapid cold-hardening treatments, which reveals an important ecological significance for population establishment and expansion of O. communa in the invaded areas of common ragweed.  相似文献

Studies on the reproduction, longevity and life table parameters of Iphiseius degenerans (Berlese) were carried out under laboratory conditions of 25 ± 1 °C, 75 ± 5% RH and 16L:8D h. As food sources for the predatory mite, Ricinus communis L. pollen, all stages of the spider mite Tetranychus urticae Koch, Frankliniella occidentalis (Pergande) larvae, and Ephestia kuehniella Zeller eggs were selected. All diets were accepted as food by the adult mites. Female longevity ranged from 29.5 to 42.4 days, the highest value was recorded on a diet of Ephestia eggs. The highest percentage of females escaping the experimental arena was observed on the diet consisting of thrips larvae. The highest oviposition rate (1.9 eggs/female.day) was recorded when the predator was fed on spider mites on an artificial substrate. For other diets, oviposition rates ranged from 1.0 to 1.3 eggs/female.day. The intrinsic rate of natural increase (r m) of I. degenerans varied between 0.015 and 0.142 females/female.day. The diet consisting of castor bean pollen resulted in the highest population growth whereas the diet on spider mites brushed off onto a bean leaf arena resulted in the slowest population growth. This can be explained by the inability of the predator to cope with the webbing of T. urticae, and the high escape rate of the progeny when reared on spider mites. Coccinella septempunctata Linnaeus, 1758 (Coleoptera: Coccinellidae) is an aphidophagous ladybird beetle species with cosmopolitan distribution. However, it may also thrive on arthropods other than aphids, when the latter are not readily available. Certain life history traits and demographic attributes of C. septempunctata were determined using seven different diets with different alternations and proportions of an aphid, Lipaphis erysimi (Kaltenbach,1843) and a mite, Tetranychus urticae Koch, 1836, in the laboratory (at 25±0.5°C, 65±5% RH and a photoperiod of 16 h L:8 h D).The larval duration was longest (12.61±6.17 days) when fed an alternating diet of two days aphid/two days mite. Adult longevity was longest (78.65±1.09 days) when fed aphids only and shortest (3.17±1.25 days) when fed mites only. Life expectancy (ex) was highest (111 days) when adults were fed only aphids and lowest (11.3 days) when fed only mites. Survival rates of ladybird beetles were positively related to an increasing ratio of aphids in their diet. Their ability to prey on both aphids and mites indicates the effectiveness of C. septempunctata as a biological control agent on plants infested with these two pests.  相似文献

Short‐rotation coppice (SRC) has great potential for supplying biomass‐based heat and energy, but little is known about SRC's ecological footprint, particularly its impact on the water cycle. To this end, we quantified the water use of a commercial scale poplar (Populus) SRC plantation in East Flanders (Belgium) at tree and stand level, focusing primarily on the transpiration component. First, we used the AquaCrop model and eddy covariance flux data to analyse the different components of the stand‐level water balance for one entire growing season. Transpiration represented 59% of evapotranspiration (ET) at stand scale over the whole year. Measured ET and modelled ET were lower as compared to the ET of reference grassland, suggesting that the SRC only used a limited amount of water. Secondly, we compared leaf area scaled and sapwood area scaled sap flow (Fs) measurements on individual plants vs. stand scale eddy covariance flux data during a 39‐day intensive field campaign in late summer 2011. Daily stem diameter variation (?D) was monitored simultaneously with Fs to understand water use strategies for three poplar genotypes. Canopy transpiration based on sapwood area or leaf area scaling was 43.5 and 50.3 mm, respectively, and accounted for 74%, respectively, 86%, of total ecosystem ET measured during the intensive field campaign. Besides differences in growth, the significant intergenotypic differences in daily ?D (due to stem shrinkage and swelling) suggested different water use strategies among the three genotypes which were confirmed by the sap flow measurements. Future studies on the prediction of SRC water use, or efforts to enhance the biomass yield of SRC genotypes, should consider intergenotypic differences in transpiration water losses at tree level as well as the SRC water balance at stand level.  相似文献

Insects form the most species‐rich lineage of Eukaryotes and each is a potential host for organisms from multiple phyla, including fungi, protozoa, mites, bacteria and nematodes. In particular, beetles are known to be associated with distinct bacterial communities and entomophilic nematodes. While entomopathogenic nematodes require symbiotic bacteria to kill and reproduce inside their insect hosts, the microbial ecology that facilitates other types of nematode–insect associations is largely unknown. To illuminate detailed patterns of the tritrophic beetle–nematode–bacteria relationship, we surveyed the nematode infestation profiles of scarab beetles in the greater Los Angeles area over a five‐year period and found distinct nematode infestation patterns for certain beetle hosts. Over a single season, we characterized the bacterial communities of beetles and their associated nematodes using high‐throughput sequencing of the 16S rRNA gene. We found significant differences in bacterial community composition among the five prevalent beetle host species, independent of geographical origin. Anaerobes Synergistaceae and sulphate‐reducing Desulfovibrionaceae were most abundant in Amblonoxia beetles, while Enterobacteriaceae and Lachnospiraceae were common in Cyclocephala beetles. Unlike entomopathogenic nematodes that carry bacterial symbionts, insect‐associated nematodes do not alter the beetles' native bacterial communities, nor do their microbiomes differ according to nematode or beetle host species. The conservation of Diplogastrid nematodes associations with Melolonthinae beetles and sulphate‐reducing bacteria suggests a possible link between beetle–bacterial communities and their associated nematodes. Our results establish a starting point towards understanding the dynamic interactions between soil macroinvertebrates and their microbiota in a highly accessible urban environment.  相似文献

Hemisarcoptes coccophagus (Acari: Astigmata: Hemisarcoptidae) is an obligate parasite of armored-scale insects (Homoptera: Diaspididae). Hypopodes (non-feeding heteromorphic deutonymphs) of this mite cannot complete their development without a phoretic sojourn on the coccinellid beetle Chilocorus bipustulatus. We tested whether the feeding history of the beetles affects the probability of the hypopodes completing their development. Two diets were offered: armored-scale insects (Homoptera: Diaspididae) (suitable for both beetles and mites) and soft-scale insects (Homoptera: Coccidae) (suitable for the beetles but not for the mites). The hypopodial survival was similar for mites which had been on beetles reared on the different scales. However, female hypopodes which had stayed on beetles reared on soft scales suffered higher mortality than those from Chilocorus reared on armored scales. Male survival was not affected. A tri-trophic-level effect on sex-specific survival in a phoretic association was thus demonstrated.  相似文献

To develop a risk-assessment system for small organisms accidentally introduced with imported organisms, we investigated as a first case study parasitic canestriniid mites, which have been imported into Japan via pet lucanid beetles from Southeast Asia. We collected mites from pinned specimens of Japanese lucanids collected before 1999—when the Japanese government lifted a ban on the import of the beetles—and living mites from imported and native lucanid beetles collected after that. No foreign canestriniid was found on any of the native Japanese beetles. Because the mites collected from imported beetles were different from Japanese species, we conclude that the foreign mites have not yet established wild populations in Japan. However, because the Japanese mites migrate between hosts without host physical contact, introduced mites are assumed to be able to migrate from a foreign to Japanese host. In fact, possible contamination was observed in pet shops. We observed host switching in only one direction: Southeast Asian Canestrinia nr spectanda switched to Japanese Dorcus rectus, but Japanese Coleopterophagus berlesei never switched to Indonesian D. titanus. The foreign mites reproduced between 15°C and 25°C, suggesting that the mites could survive in mountainous sites in southern Japan and at low elevations in northern Japan. The ability of foreign parasitic canestriniids to infect and survive on Japanese hosts at temperatures characteristic of much of Japan leads us to conclude that these mites present a potential risk to Japanese endemic canestriniids as well as to native Japanese lucanids.  相似文献

The establishment, spread and increase of the invasive coccinellid Harmonia axyridis Pallas in North America and Britain have coincided with declines in native ladybirds. In Britain, this pattern was predicted soon after H. axyridis was first recorded. However, predictions of the impact that H. axyridis may have on the parasites, parasitoids and pathogens of native coccinellids have been less certain, largely because of uncertainty over whether H. axyridis would become an alternate host for such agents. Dinocampus coccinellae (Schrank) is a braconid parasitoid of ladybirds of the sub-family Coccinellinae. In Japan, this wasp uses both Coccinella septempunctata brucki Mulsant and H. axyridis as hosts, but successfully parasitizes a higher proportion of the former species. Data are here presented that indicate the same is true in Britain, but to a greater extent. This study investigates the interactions of D. coccinellae with C. septempunctata L. and H. axyridis from Japan and Britain. We show that coccinellid activity affects encounter rates with the wasp, with H. axyridis being more active than C. septempunctata in the tests. Escalation rates from investigation to ovipositional attack were higher for C. septempunctata than H. axyridis for both Japanese and British wasps. Wasp emergence was higher, relative to ovipositional attack, from C. septempunctata than from H. axyridis. Wasps did not discriminate between Japanese and British ladybirds of the same species. British wasp eclosion rates were higher from Japanese than from British H. axyridis. We consider both the effect D. coccinellae may have on H. axyridis population demography, and the effect H. axyridis may have on D. coccinellae in Europe.  相似文献

  1. Extensive boring damage into trunks of living poplar trees in the urban forest of Shanghai, eastern China, was investigated in 2017.
  2. In order to determine the taxonomic identity of the pest, we used morphological identification and sequenced mitochondrial COI gene. Based on both morphology and molecular data, it was concluded that the pest boring into the poplars was the ambrosia beetle Euwallacea interjectus (Blandford).
  3. Its occurrence and damage on poplar across Shanghai were also investigated. In the Pudong District, a total of 104.52 ha of urban forest were investigated thoroughly, and 16.22% of surveyed forests were found to be infested. This insect species is an important native pest capable of causing damage across the Shanghai area.
  4. Observations in the field suggested that the damage may be related to distance to water sources. Our statistical analysis confirmed this hypothesis, as the close distance to water bodies correlated with higher levels of damage. We conclude that E. interjectus prefers host trees stressed by an occasional excessive abundance of water, and acts as a parasite on living trees without killing them.

The aim of this study was to evaluate the biomass production potential for the Spanish Iberian Peninsula using the Populus spp. ‘I‐214’ clone under several management regimes and land availability scenarios, and to determine its future contribution to Spanish energy demands. Empirical models were fitted to the data from a network of 144 plots located at 12 sites in the continental Mediterranean climatic regions of the Iberian Peninsula, in which yield was related to climate and soil, as well as to plantation management variables. Four models were developed considering average maximum temperature of the hottest month (TMAXH, °C), length of drought (A, months), intensity of drought (K, unitless) and soil pH. Predictions were made for the irrigated agricultural land (IAL), where the value of the independent variables were within the validity range, and for two management scenarios. Energy production capacity was evaluated by considering the alternatives for transforming poplar SRC biomass: heat, bio‐ethanol and electricity. The results indicated a mean productivity for the Spanish Iberian peninsula of between 15.3 and 10.9 Mg ha?1 yr?1 for the standard management scenario and the poorly irrigated and weeded management scenario respectively. Two IAL scenarios were considered for the calculation of biomass production potential: all IAL for which it was possible to make predictions is made available for poplar SRC (TP, maximum hypothetical production capacity), and another in which only unproductive IAL is available for poplar SRC (RP, production capacity without constricting agricultural production). The TP scenario contributes up to 6.8–9.6% of total energy demands, and the RP scenario 0.7–0.9%, depending on plantation management.  相似文献

Life history and demographic parameters of Psyllaephagus zdeneki Noyes and Fallahzadeh (Hymenoptera: Encyrtidae) were studied on its host the olive psyllid, Euphyllura pakistanica Loginova (Hemiptera: Psyllidae). Experiments were conducted in a growth chamber at 20±1 °C, relative humidity of 60±5%, and a photoperiod of 16:8 (L: D) hours. Four different olive cultivars (Fishomi, Shenge, Oil and Yellow) were used to test possible host plant influence on parasitoid performance. The pre-imaginal developmental period of female P. zdeneki varied from 24.96 (on Fishomi) to 26.34 (on Shenge) days, and for males from 21.63 (on Fishomi) to 24.44 (on Yellow) days. Adult female longevity differed significantly among the four cultivars, ranging from 12.46 (on Fishomi) to 14.97 (on Shenge) days. For each cultivar, adult female longevity was significantly greater than male longevity. Life table parameters showed survival rates (l x ) in newly emerged females were 84.61, 82.25, 85.71 and 78.12% on Fishomi, Yellow, Shenge and Oil, respectively. Female egg deposition was highest on Yellow (138.4 eggs per female) and lowest on Fishomi (116.3 eggs per female). The highest and lowest intrinsic rate of increase were 0.28 (on Shenge) and 0.24 (on Oil), respectively. The mean generation time ranged from 14.6 (on Shenge) to 16.7 (on Oil) days. These results are discussed with respect to the potential impact P. zdeneki as a natural enemy of E. pakistanica, the most important pest of olive in the Fars province of Iran, as well as the influence of olive cultivar on parasitoid life table parameters.  相似文献

Tree root activity, including fine-root production, turnover and metabolic activity are significant components of forest productivity and nutrient cycling. Differences in root activity among forest types are not well known. A 3-year study was undertaken in red pine (Pinus resinosa Ait.) and hybrid poplar (Populus tristis X P. balsamifera cv `Tristis no. 1') plantations to compare belowground root dynamics. We measured fine-root production, mortality and standing crop, as well as soil CO2 efflux. Pine fine-root production was only 2.9% of that of poplar during three years; 85 pine roots were observed in minirhizotron tubes compared with 4088 poplar roots. Live-root density oscillated seasonally for both species with late winter minimum and autumn maximum. Poplar reached constant maximum live-root length within the first growing season, but pine continued to increase observed fine-root length for three growing seasons. Within the first 100 days following initial appearance, 22% of the pine roots disappeared and 38% of the poplar roots disappeared. Median fine-root longevity of pine was 291 days compared with 149 days for poplar roots. Fine-root longevity increased with depth in the soil, and was greater for roots with initial diameter >0.5 mm. The probability of poplar root death from late February to May was more than three times that in any other season, regardless of root age. Despite the greater poplar root production and live-root length, fine-root biomass and soil CO2 efflux was greater in pine. Greater metabolic activity in the pine stand may be due to greater fine-root biomass or greater heterotrophic respiration. During the period 1986–1988 the parasitoids of smaller poplar borer,Saperda populnea L. (Coleoptera, Cerambycidae) along the Danube in Bulgaria were studied. A total of 898 smaller poplar borer galls were investigated—289 in 1986, 404 in 1987, and 205 in 1988. In the particular years the parasitoids of smaller poplar borer namelyBillaea irrorata Meig.(Diptera, Tachinidae), Dolichomitus populneus Ratz. (Hym.: Ichneumonidae), andIphiaulax impostor Scop. (Hym.: Braconidae) destroyed from 13.1 up to 28.0% of the pest larvae, thus decreasing their frequency together with the other limiting factors by 56.1% up to 81.2%. The tachinid flyB. irrorata decreased the smaller poplar borer larvae frequency by 9.3% up to 18.9%, while the ichneumonoidD. populneus, and the braconoidI. impostor destroyed insignificant percentages of these larvae—0.5% up to 1.7% and 0.5% up to 2.5% resp. The flight period of the pest preceded or almost coincided with the ones ofD. populneus, andI. impostor. The flyB. irrorata always appeared last and its flight period was expanded.   相似文献

Parasitism ofCoccinella septempunctata L. byPerilitus coccinellae (Schrank) varies seasonally with an 11% overall rate among beetles overwintering in the Hackensack Meadowlands, which is less than that reported in Europe.P. coccinellae may parasitizeC. 7-punctata twice yearly. FemaleC. 7-punctata were parasitized more heavily than males. Although parasitism byP. coccinellae varied significantly among host species, it was not significantly different among 3 habitats. High host density favored greater parasitism in 3 coccinellid species. Rearing for parasite cocoons, consistently showed lower parasitism rates than did dissection for parasitized larvae. Overwintering survival of cagedC. 7-punctata near Stillwater, OK, varied between years averaging 53,5%. The relationship between the development ofCoccinella septempunctata brucki Mulsant (Coleoptera: Coccinellidae) and its parasitoid,Perilitus coccinellae (Schrank) (Hymenoptera: Braconidae) was studied at two photoperiods (L 16:D8 and L 12: D12) at 26°C. The development ofP. coccinellae is well synchronized with the physiological state of the host,C. septempunctata, which can be parasitized not only as adult but also as larva or pupa. The parasitoid larva completed larval development within 19 days in a non-diapausing host, while in diapausing adults as well as in pupae held at diapause-averting conditions, the parasitoid larva ceased growth at the first instar. Growth was resumed when diapause of the host terminated or by the emergence of the adult host from the pupa. About 550 spheric cells, teratocytes, were liberated into the host hemocoel when the parasitoid egg hatched. The teratocytes increased in size in the active host, while their development was arrested in the diapausing host. Application of methoprene caused diapause termination of both host and parasitoid larva. The results indicate that the development of the larva ofP. coccinellae depends on the physiological conditions of the host,C. septempunctata brucki. The host-parasite relation thus represents an ‘endogenous synchronization’ in the sense of Schoonhoven's definition.  相似文献
