On the basis of the recent discovery that precisely replicating triplets of impulses present in All-Interval histograms of spike trains generated by visual cortex cells of Rhesus monkeys are surrounded by multiple copies of ghost doublets of such triplets, we have examined and compared in detail, the spike trains generated by four complex cells in the striate cortex of curarized monkeys with respect to: (1) The number of precisely replicating triplet patterns embedded in trains of discharges generated in response to specific Hubel-Wiesel stimulation; (2) The effect of time separating the occurrence of such replicating triplets on the number and time distribution of their ghost doublets; (3) The effect of decreasing the precision criterion for the detection of replicating (parent) triplets (from the standard 0.14 ms criterion to 0.5 ms) on the relationships between triplets and their ghosts and (4) The comparison of the distributions in time of ghost doublets around the first and second copies of triplets when the time intervals separating them were greater than or less than 0.5 s. We found that the precision of replication of triplets varies somewhat from one cell to another, and that ghosts doublets are more copiously associated with replicating triplets emitted near in time to each other than with triplets emitted after larger time intervals, except in the case of one cell. In order to assess the statistical significance of our findings, we systematically shuffled the order of occurrence of intervals in every burst of all the records of one of the studied cells and repeated the analysis. Both the number of replicating triplets and of associated ghost doublets is significantly depressed (but not totally obliterated) by the above shuffling procedure. Finally, further implications based on a model of neural information transmission in the form of temporal correlations between spikes are discussed.  相似文献

We analysed evoked responses recorded from 97 cells in the visual cortex of 4 adult cats and 8 kittens, stimulated by a drifting sinusoidal grating. A Fourier analysis of the responses allowed us to select 30 cells showing a clear modulating response (relative modulation index > 1). The 162 records from these selected cells were scanned to detect precise temporal correlations in the form of replicating triplets and associated ghost doublets. Temporal correlations of this nature were observed in these cells. They are about 10 times more abundant in adult cats than in kittens, and mostly observed in infragranular cortical layer cells. The possible role of these precise temporal patterns in information processing in the brain is examined, as well as the relation between this type of temporal correlation with coherent oscillations and principal components waveforms.  相似文献

In response to a visual stimulation, "replicated triplets" of impulses may appear in many spike trains recorded from the cat dorsal lateral geniculate nucleus (dLGN). The number and the temporal structure of these triplets depend upon the general organization of the geniculate impulse trains. In this study, we show that a pharmacological blockade of the corticothalamic activity, obtained through microinjection of GABA into area 17, affects the replicated triplet production and leads to an increase in the dispersal of their structure. These results suggest that the corticothalamic pathway closely influences the fine temporal organization of the geniculate messages.  相似文献

We have examined thin sections and replicas of freeze-fractured cilia of Tetrahymena pyriformis. The ciliary necklace located at the base of all freeze-fractured oral and somatic cilia has been studied in thin sections. Since electron-dense linkers have been found to connect both microtubule doublets and triplets to the ciliary membrane at the level of the necklace, the linkers and the associated necklace seem to be related to the transition region between the doublets and triplets of a cilium. Plaque structures, consisting of small rectangular patches of particles located distal to the ciliary necklace, are found in strain GL, but are absent in other strains examined in this study. In freeze-cleaved material, additional structural differentiations are observed in the distal region of the ciliary membranes of somatic and oral cilia. Somatic cilia contain many randomly distributed particles within their membrane. Oral cilia can be divided into three categories on the basis of the morphology of their freeze-fractured membranes: (a) undifferentiated cilia with very few randomly distributed particles: (b) cilia with particles arranged in parallel longitudinal rows spaced at intervals of 810–1080 Å that are located on one side of the cilium; and (c) cilia with patches of particles arranged in short rows oriented obliquely to the main axis of the cilium. The latter particles, found on one side of the cilium, seem to serve as attachment sites for bristles 375–750 Å long and 100 Å wide which extend into the surrounding medium. The particles with bristles are located at the tips of cilia in the outermost membranelle and may be used to detect food particles and/or to modify currents in the oral region so that food particles are propelled more efficiently into the buccal cavity. Examination of thin-sectioned material indicates that the particles in oral cilia which form the longitudinal rows could be linked to microtubule doublets. Linkage between microtubule doublets and adjacent membrane areas on one side of the cilium could modify the form of ciliary beat by restricting the sliding of the microtubules. It is suggested that membrane-microtubule interactions may form the basis for the various forms of ciliary beat observed in different organisms.  相似文献

Determining which traits may allow some introduced plant species to become invasive in their new environment continues to be a key question in invasion biology. Vincetoxicum rossicum is an invasive, perennial vine colonizing natural and seminatural habitats primarily in the northeastern United States and southeastern Canada. More than half its seeds exhibit polyembryony, a relatively uncommon condition in which a single seed produces multiple seedlings. For evaluating the potential consequences of polyembryony on invasiveness, V. rossicum plants derived from seeds of three embryonic classes-singlets, doublets, and triplets (one, two, and three seedlings per seed, respectively)-were paired in all combinations intraspecifically and with the co-occurring native herbs Solidago canadensis and Asclepias syriaca in a greenhouse study. Vincetoxicum rossicum biomass was 25-55% greater and follicle production 55-100% greater under intraspecific competition compared with interspecific competition. However, within a competitive environment, follicle production varied little. Regardless of competitive environment, V. rossicum originating from seeds with a greater number of embryos typically performed no better than plants arising from seed with fewer embryos (singlets = doublets = triplets)-except intraspecifically where doublets outperformed singlets, and with S. canadensis where triplets outperformed singlets. Our findings suggest that overall performance and fitness of V. rossicum is higher in monocultures than in mixed stands and that its ability to invade new habitats may not be attributable to the production of polyembryonic seeds.  相似文献

Coevolving residues in a multiple sequence alignment provide evolutionary clues of biophysical interactions in 3D structure. Despite a rich literature describing amino acid coevolution within or between proteins and nucleic acid coevolution within RNA, to date there has been no direct evidence of coevolution between protein and RNA. The ribosome, a structurally conserved macromolecular machine composed of over 50 interacting protein and RNA chains, provides a natural example of RNA/protein interactions that likely coevolved. We provide the first direct evidence of RNA/protein coevolution by characterizing the mutual information in residue triplets from a multiple sequence alignment of ribosomal protein L22 and neighboring 23S RNA. We define residue triplets as three positions in the multiple sequence alignment, where one position is from the 23S RNA and two positions are from the L22 protein. We show that residue triplets with high mutual information are more likely than residue doublets to be proximal in 3D space. Some high mutual information residue triplets cluster in a connected series across the L22 protein structure, similar to patterns seen in protein coevolution. We also describe RNA nucleotides for which switching from one nucleotide to another (or between purines and pyrimidines) results in a change in amino acid distribution for proximal amino acid positions. Multiple crystal structures for evolutionarily distinct ribosome species can provide structural evidence for these differences. For one residue triplet, a pyrimidine in one species is a purine in another, and RNA/protein hydrogen bonds are present in one species but not the other. The results provide the first direct evidence of RNA/protein coevolution by using higher order mutual information, suggesting that biophysical constraints on interacting RNA and protein chains are indeed a driving force in their evolution.  相似文献

Abstract: Codas (patterned click sequences) produced by sperm whales ( Physeter catodon ) were recorded during four research cruises in the southeast Caribbean. Two coda patterns comprised more than 50% of the codas analyzed from 46 h of recording. These two patterns were called "shared" codas because they were produced by numerous whales throughout the area, both as repeated sequences by individual whales, at times simultaneously by two or more whales, and occasionally in overlapping series. Analyses of both the absolute timing and the relative timing of click intervals indicate equivalent variability in the temporal patterns of similar shared codas, whether produced by the same or different whales. The shared codas appear to have a function that is different from individual identification as hypothesized previously for some coda patterns.  相似文献

In the presence of specific inhibitors of beat. 20 microM VO4(3-) or pCa 4, mussel gill lateral (L) cilia can be arrested in two positions--"hands down" or "hands up"--at opposite ends of the stroke cycle. Cilia move to these positions by doublet microtubule sliding. Axonemes of arrested cilia, still tethered to the cell, are intact after demembranation and protease treatment. When reactivated by 4 mM ATP with inhibitors present, about 40% split apart. Splits are not random but occur preferentially between different specific doublets in the two opposite arrest positions. Several different related patterns of splitting are observed; for every pattern in "hands down" axonemes, there is a corresponding complementary split pattern in "hands up" axonemes. In some split patterns two doublets remain firmly attached to the central pair; these also differ depending on axonemal position. Although some of the patterns seen may be artifactual or difficult to explain, the complementary splitting patterns are predictable with simple assumptions by a "switch point" hypothesis of ciliary activity where, during each recovery stroke, doublets 6-8 have active dynein arms, while during each effective stroke, arms on doublets 1-4 become active, and arms 6-8 are turned off. Because of a difference between the patterns seen and the predictions, the status of the arms on doublet 9 is unresolved. The patterns also suggest that a spoke-central sheath attachment cycle may correlate with switching of arm activity during the generation of an asymmetric beat.  相似文献

Analysis of serial cross-sections of the Chlamydomonas flagellum reveals several structural asymmetries in the axoneme. One doublet lacks the outer dynein arm, has a beak-like projection in its B-tubule, and bears a two-part bridge that extends from the A-tubule of this doublet to the B-tubule of the adjacent doublet. The two doublets directly opposite the doublet lacking the arm have beak-like projections in their B-tubules. These asymmetries always occur in the same doublets from section to section, indicating that certain doublets have consistent morphological specializations. These unique doublets give the axoneme an inherent structural polarity. All three specializations are present in the proximal portion of the axoneme; based on their frequency in random cross-sections of isolated axonemes, the two-part bridge and the beak-like projections are present in the proximal one quarter and one half of the axoneme, respectively, and the outer arm is absent from the one doublet greater than 90% of the axoneme's length. The outer arm-less doublet of each flagellum faces the other flagellum, indicating that each axoneme has the same rotational orientation relative to the direction of its effective stroke. This strongly suggests that the direction of the effective stroke is controlled by a structural component within the axoneme. The striated fibers are associated with specific triplets in a manner suggesting that they play a role in setting up or maintaining the 180 degrees rotational symmetry of the two flagella.  相似文献

Responses of auditory neurons in the torus semicircularis (TS) of Pleurodema thaul, a leptodactylid from Chile, to synthetic stimuli having diverse temporal patterns and to digitized advertisement calls of P. thaul and three sympatric species, were recorded to investigate their temporal response selectivities. The advertisement call of this species consists of a long sequence of sound pulses (a pulse-amplitude-modulated, or PAM, signal) having a dominant frequency of about 2000 Hz. Each of the sound pulses contains intra-pulse sinusoidal-amplitude-modulations (SAMs). Synthetic stimuli consisted of six series in which the following acoustic parameters were systematically modified, one at a time: PAM rate, pulse duration, number of pulses, and intra-pulse SAM rate. The carrier frequency of these stimuli was set at the characteristic frequency (CF) of the isolated units (n = 47). Response patterns of TS units to synthetic call variants reveal different degrees of selectivities for each of the temporal variables, with populations of neurons responding maximally to specific values found in the advertisement call of this species. These selectivities are mainly shaped by neuronal responsiveness to the overall sound energy of the stimulus and by the inability of neurons to discharge to short inter-pulse gaps. The conduction velocity (CV) of a muscle fiber is affected by the fiber's discharge history going back ~1 s. We investigated this dependence by measuring CV fluctuations during voluntary isometric contractions of the human brachioradialis muscle. We recorded electromyogram (EMG) signals simultaneously from multiple intramuscular electrodes, identified potentials belonging to the same motor unit using EMG decomposition, and estimated the CV of each discharge from the interpotential interval. In 12 of 14 subjects, CV increased by ~10% during the first second after recruitment and then fluctuated by about ±2% in a way that mirrored the fluctuations in the instantaneous firing rate. The CV profile could be precisely described in terms of the discharge history by a simple mathematical model. In the other two subjects, and one subject retested after cooling the arm, the CV fluctuations were inversely correlated with instantaneous firing rate. In all subjects, CV was additionally affected by very short interdischarge intervals (<25 ms): it was increased in doublets at recruitment, but decreased in doublets during continuous firing and after short interdischarge intervals in doubly innervated fibers. CV also exhibited a slow trend of about -0.05%/s that did not depend on the immediate discharge history. We suggest that measurements of CV fluctuations during voluntary contractions, or during stimulation protocols that involve longer and more complex stimulation patterns than are currently being used, may provide a sensitive approach for estimating the dynamic characteristics of ion channels in the human muscle-fiber membrane.  相似文献

In an elevated maze consisting of three reconvergent radial arms, golden hamsters were tested with the same experimental rule: to choose each path without repeating any choice. However, variations of procedure concerning (a) the location of the reward in the maze, and (b) reinforcement contingencies, were introduced in order to define several problems involving variable levels of difficulty. The relationship between response strategies and difficulty of the task was then studied. The common learning criterion was the achievement of three consecutive correct daily sessions, each session corresponding to a particular sequence (pattern) of choices of paths. Response strategies were studied by analyzing the patterns obtained over the three final sessions in which an animal reached the learning criterion. Such a set of patterns (triplet) could be heterogeneous (patterns all different), mixed (two identical patterns, one different) or stereotyped (identical patterns). No relationship was found between the mean level of difficulty presented by each learning problem and the occurrence of a particular type of triplet. However, in each situation, mixed triplets were the most frequently recorded and corresponded to the medium individual speeds of learning whereas heterogeneous triplets corresponded to rapid successes and stereotyped triplets to delayed successes. These findings indicate that, whatever the problem designed to be tested in a three-arm maze, the various forms of solutions reflect different individual adaptative mechanisms.  相似文献

The axoneme "9 + 2" is basically a system constituted of a cylinder of 9 microtubule doublets surrounding a central pair of microtubules. These bi-tubular structures are considered as the support system of the active molecular complexes that generate and regulate the axonemal movement. Schoutens has calculated their moments of inertia [Schoutens, 1994: Journal of Theoretical Biology 171:163-177]. The results obtained allowed us to assume that these bi-tubular systems are endowed with dynamic properties that could be involved in the regulation of the axonemal machinery. For the first time, using the finite elements methods and the resistance of material principles, we have now calculated that the curvature of the axoneme induces the deviated-bending of the bi-tubular structures of the axoneme, because of their geometry only; they behave as beams in a framework. This approach is similar to the one used to measure the deflection of a single microtubule [Kasas et al., 2004: Chem Phys Chem 5:252-257]. These behaviors induce internal movement or constraints of either couples or triplets of doublets within the axonemal cylinder that could be directly involved in a constrained or a spontaneous "convergence/divergence" equilibrium of the cylindrical generatrices that they draw along the axonemal cylinder, which could apparently regulate the activity of the axonemal motors (the dynein arms). These results are discussed here, taking into consideration the dynamic propagation of the wave train along the flagellar axoneme, and the regulated balance between the activities of the two opposite sides of the axoneme during the beat. This study raises a few questions about the architecture-activity duo of the axonemal doublets.  相似文献

Summary Transmission electron microscopy was used to study the development of the flagellar base and the flagellar necklace during spermatogenesis in a moth (Ephestia kuehniella Z.). Until mid-pachytene, two basal body pairs without flagella occur per cell. The basal bodies, which contain a cartwheel complex, give rise to four flagella in late prophase I. The cartwheel complex appears to be involved in the nucleation of the central pair of axonemal microtubules. In spermatids, there is one basal body; this is attached to a flagellum. At this stage, the nine microtubular triplets of the basal body do not terminate at the same proximal level. The juxtanuclear triplets are shifted distally relative to the triplets distant from the nuclear envelope. Transition fibrils and a flagellar necklace are formed at the onset of axoneme elongation. The flagellar necklace includes Y-shaped elements that connect the flagellar membrane and the axonemal doublets. In spindle-containing spermatocytes, the flagellar necklace is no longer detectable. During spermatid differentiation, the transition fibrils move distally along the axoneme and a prominent middle piece appears. Our observations and those in the literature indicate certain trends in sperm structure. In sperms with a short middle piece, we expect the presence of a flagellar necklace. The distal movement of the transition fibrils or equivalent structures is prevented by the presence of radial linkers between the flagellar membrane and the axonemal doublets. On the other hand, the absence of a flagellar necklace at the initiation of spermiogenesis enables the formation of a long middle piece. Thus, in spermatozoa possessing an extended middle piece, a flagellar necklace may be missing.  相似文献

Recent work on the insect olfactory system has shown that its mushroom bodies (one of its major components) are involved in the fine discrimination of odours and that the temporal organisation of spike discharges plays a fundamental role. We propose here a model of a network that is able to decode the temporal patterns which characterise an odour. This model has three fundamental properties that seem to exist in all mushroom bodies of insects studied so far: a) long lasting inhibitions with rebounds, able to facilitate delayed spike generation; b) synaptic plasticity, which allows the network to learn to recognise temporal patterns; c) above all a large interconnection, which allows this network to recognise intervals of various duration. This model thus appears suited to identify combinations of temporal patterns in the dendrites of Kenyon cells (the principal cells in the calyces of the mushroom bodies). Moreover, the mushroom bodies integrate multimodal inputs, suggesting that the detection of temporal patterns may be extended to the detection of a complex environment, combining in particular olfactive and visual inputs.  相似文献

Nucleotide sequences were converted into purine (R)-pyrimidine (Y) series and divided into several groups, embracing higher and lower organisms. The frequencies of R-Y doublets, triplets and quartets in each were calculated. Whereas eukaryotes uniformly show RR + YY greater than RY + YR, in bacteria and phage no such relationship is observed. The triplet and quartet patterns in higher organisms differ from those seen in prokaryotes. In the higher organisms a correlation is observed between the frequencies of triplets and quartets and some DNA structural parameters. Specifically, the most frequent triplets are those with minimal torsion angle deviations from a regular B-DNA. The most frequent quartets are those with minimal roll angle deviations. No such correlations are observed in prokaryotes. We therefore propose that in eukaryotic DNA, tight, smooth packaging imposes sequence constraints.  相似文献

Multiple pulses of light administered to humans have been reported to result in type 0 phase responses. These results suggest the underlying pacemaker to be nonsimple. At present, results with this type of protocol have only been reported for humans. Therefore, multiple pulses of light were administered to rats. Rats were exposed to one, two, three, or four pulses of light for 5 h (1000 lux) at successive 24-h intervals. Results did not suggest a type 0 phase response. Nonetheless, results with a second, third, or fourth light exposure were not fully predictable from a phase response curve derived from a single light pulse.  相似文献

Both chemical and physical effects of red cells have been implicated in the spontaneous aggregation of platelets in sheared whole blood (WB). To determine whether the chemical effect is due to ADP leaking from the red cells, a previously described technique for measuring the concentration and size of single platelets and aggregates was used to study the shear-induced aggregation of platelets in WB flowing through 1.19-mm-diameter polyethylene tubing in the presence and absence of the ADP scavenger enzyme system phosphocreatine-creatine phosphokinase (CP-CPK). Significant spontaneous aggregation was observed at mean tube shear rates, (G) = 41.9 and 335 s-1 (42% and 13% decrease in single platelets after a mean transit time (t) = 43 s, compared to 89 and 95% decrease with 0.2 microM ADP). The addition of CP-CPK, either at the time of, or 30 min before each run, completely abolished aggregation. In the presence of 0.2 microM ADP, CP-CPK caused a reversal of aggregation at (t) = 17 s after 30% of single cells had aggregated. To determine whether red cells exert a physical effect by increasing the time of interaction of two colliding platelets (thereby increasing the proportion of collisions resulting in the formation of aggregates), an optically transparent suspension of 40% reconstituted red cell ghosts in serum containing 2.5-micron-diameter latex spheres (3 x 10(5)/microliters) flowing through 100-microns-diameter tubes was used as a model of platelets in blood, and the results were compared with those obtained in a control suspension of latex spheres in serum alone. Two-body collisions between microspheres in the interior of the flowing ghost cell or serum suspensions at shear rates from 5 to 90 s-1 were recorded on cine film. The films were subsequently analyzed, and the measured doublet lifetime, tau meas, was compared with that predicted by theory in the absence of interactions with other particles, tau theor. The mean (tau meas/tau theor) for doublets in ghost cell suspensions was 1.614 +/- 1.795 (SD; n = 320), compared to a value of 1.001 +/- 0.312 (n = 90) for doublets in serum. Whereas 11% of doublets in ghost cell suspensions had lifetimes from 2.5 to 5 times greater than predicted, in serum, no doublets had lifetimes greater than 1.91 times that predicted. There was no statistically significant correlation between tau meas/tau theor and shear rate, but the values of tau meas/tau theor for low-angle collisions in ghost cell suspensions were significantly greater than for high-angle collisions.  相似文献
